This study determined the influence of photoperiod on the expression of toll-like receptor 2 and 4 (TLR2 and TLR4), interleukin 1□ (IL1B), IL-1 receptor type I (IL1R1) and II (IL1R2), interleukin 6 (IL6), the IL-6 receptor (IL6R) and signal transducer (IL6ST), tumor necrosis factor α (TNF), and TNF□ receptor type I (TNFRSF1A) and II (TNFRSF1B) in the choroid plexus (ChP) of ewes with lipopolysaccharide (LPS)-induced acute inflammation. Under short-days (SD, n = 12, anestrous) and long-days (LD, n = 12, synchronized follicular phase), ewes were treated with saline or LPS. Compared to LD conditions, the ewes under SD were characterized by a greater (PTNFRSF1B (only under SD), TNF and TNFRSF1A (no stimulation), and IL6R (decreased (PTLR2, TLR4, IL1B and its receptors, IL6 and TNFRSF1B were higher (PTLR4 was positively correlated with IL1B and IL6 in both saline- (r2 = 0.64, P2 = 0.52, P2 = 0.81, P2 = 0.51, PIL1B (r2 = 0.56, P2 = 0.77, PIL6 (r2 = 0.77, P2 = 0.35, PTLR2 in saline- and LPS-treated ewes, respectively. This indicates that in ewes, the ChP response to acute systemic inflammation is dependent upon the photoperiod with stronger effects being observed under SD. Our results also suggest that gonadal hormones altering TLR4 signaling events are involved in the photoperiodic modulation of the ChP response to LPS. Further experiments are required to explain the mechanism involved in this phenomenon.

This study was designed to determine the effect of acute caffeine (CAF) administration, which exerts a broad spectrum of anti-inflammatory activity, on the synthesis of pro-inflammatory cytokines and their receptors in the hypothalamus and choroid plexus (ChP) during acute inflammation caused by the injection of bacterial endotoxin—lipopolysaccharide (LPS). The experiment was performed on 24 female sheep randomly divided into four groups: control; LPS treated (iv.; 400 ng/kg of body mass (bm.)); CAF treated (iv.; 30 mg/kg of bm.); and LPS and CAF treated. The animals were euthanized 3 h after the treatment. It was found that acute administration of CAF suppressed the synthesis of interleukin (IL-1β) and tumor necrosis factor (TNF)α, but did not influence IL-6, in the hypothalamus during LPS-induced inflammation. The injection of CAF reduced the LPS-induced expression of TNF mRNA in the ChP. CAF lowered the gene expression of IL-6 cytokine family signal transducer (IL6ST) and TNF receptor superfamily member 1A (TNFRSF1) in the hypothalamus and IL-1 type II receptor (IL1R2) in the ChP. Our study on the sheep model suggests that CAF may attenuate the inflammatory response at the hypothalamic level and partly influence the inflammatory signal generated by the ChP cells. This suggests the potential of CAF to suppress neuroinflammatory processes induced by peripheral immune/inflammatory challenges.

Delivery of putative compounds of therapeutic value to the brain is limited by brain barriers: the blood–brain barrier located in the endothelium of the brain microvessels (BrMVs) and the blood–cerebrospinal fluid barrier located in the epithelium of the choroid plexus (ChP). Understanding their function and modulation by the circadian clock may enhance the efficacy of brain-targeting therapies. The aim of the present study was to evaluate the stability of 10 reference genes in the BrMV and ChP, isolated from male and female rats at six time points (ZT1, 5, 9, 13, 17, and 21). Gene evaluations were performed by qPCR, analyzed by RefFinder tool, and verified by analyzing the expression of the brain and muscle ARNT-like 1 (Bmal1) using the qPCR and digital PCR methods. We identified as the most stable genes for circadian studies tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein zeta (Ywhaz) and apolipoprotein E (Apoe) for BrMV, and beta actin (Actb) and hypoxanthine-guanine phosphoribosyltransferase (Hprt1) for ChP. After verification, ribosomal protein (Rps18) was also included as a sufficient reference gene. Additionally, the observed gender difference in the Bmal1 oscillations in both BrMV and ChP suggests that separate studies for each gender are recommended.

Quercetin-3-glucuronide (Q3GA), the main phase II metabolite of quercetin (Q) in human plasma, is considered to be a more stable form of Q for transport with the bloodstream to tissues, where it can be potentially deconjugated by β-glucuronidase (β-Gluc) to Q aglycone, which easily enters the brain. This study evaluates the effect of lipopolysaccharide (LPS)-induced acute inflammation on β-Gluc gene expression in the choroid plexus (ChP) and its activity in blood plasma, ChP and cerebrospinal fluid (CSF), and the concentration of Q and its phase II metabolites in blood plasma and CSF. Studies were performed on saline- and LPS-treated adult ewes (n = 40) receiving Q3GA intravenously (n = 16) and on primary rat ChP epithelial cells and human ChP epithelial papilloma cells. We observed that acute inflammation stimulated β-Gluc activity in the ChP and blood plasma, but not in ChP epithelial cells and CSF, and did not affect Q and its phase II metabolite concentrations in plasma and CSF, except Q3GA, for which the plasma concentration was higher 30 min after administration (p &lt, 0.05) in LPS- compared to saline-treated ewes. The lack of Q3GA deconjugation in the ChP observed under physiological and acute inflammatory conditions, however, does not exclude its possible role in the course of neurodegenerative diseases.

The ovine choroid plexus (ChP) expresses the long isoform of the leptin receptor, which makes this structure a potential target for leptin action. In sheep, leptin concentration in plasma is higher during long days (LD) than short days (SD). This study evaluates the influence a of photoperiod on leptin impact on the gene expression of Toll-like receptor 4 (TLR4), proinflammatory cytokines (IL1B, IL6), their receptors (IL1R1, IL1R2, ILRN, IL6R, IL6ST) and inflammasome components necessary for pro-IL-1&beta, activation (NLRP3, PYCARD, CASP1), chemokine (CCL2), leptin receptor isoforms (LEPRa, LEPRb) and a suppressor of cytokine signalling (SOCS3) in the ChP of ewes treated or not with lipopolysaccharide (LPS). Studies were conducted on adult female sheep divided into four groups (n = 6 in each): control, leptin (20 &mu, g/kg), LPS (400 ng/kg), and LPS and leptin injected under SD and LD photoperiods. The leptin alone did not affect the gene expression but in co-treatment with LPS increased (p &lt, 0.05) IL1B but only during SD, and SOCS3, IL1R2, IL1RN, IL6ST and CCL2 only during LD, and decreased (p &lt, 0.05) the IL1R1 expression only during SD photoperiod. This indicates that the immunomodulatory action of leptin on the ChP is manifested only under the LPS challenge and is photoperiodically dependent.

The aim of this study was to determine whether anthocyanins and their phase II metabolites permeate the blood-cerebrospinal fluid barrier (B-CSF-B) of sheep and to profile these compounds in sheep biofluids after chokeberry intraruminal administration. Anthocyanins were analyzed using micro-HPLC-MS/MS. After chokeberry administration, anthocyanins were absorbed and occurred in body fluids mainly in the form of methylated, glucuronidated, and sulfated derivatives (in total, 21 derivatives were identified). The study showed that anthocyanins penetrated the B-CSF-B and their change in profile and concentration in the cerebrospinal fluid (CSF) resulted from fluctuations in concentrations of these compounds in blood plasma, although the presence of various cyanidin derivatives in CSF also depended on their chemical structure. The biological fate of chokeberry anthocyanins, from absorption into blood to penetration into CSF, was tracked to facilitate the design of further experimental procedures to determine the biological properties of these compounds, including potentially neuroprotective activities.

The present study concerns the effect of melatonin from slow-release implants on the expression of genes coding interleukin-1β (Il1B), inerleukin-6 (Il6), tumour necrosis factor α (Tnf) and their receptors: IL-1 receptor type I (Il1r1) and type II (Il1r2), IL-6 receptor (Il6r) and signal transducer (Il6st), TNFα receptor type I (Tnfrsf1a) and II (Tnfrsf1b) and retinoid-related orphan receptor α (RorA) and Rev.-erbα in the ovine choroid plexus (CP) under basal and lipopolysaccharide (LPS)-challenged conditions. Studies were performed on four groups: 1) sham-implanted and placebo-treated, 2) melatonin-implanted (Melovine, 18mg) and placebo-treated, 3) sham-implanted and LPS-treated (400ng/kg of body weight) and 4) melatonin-implanted and LPS-treated. Under basal conditions, we observed weak expression of Tnf, low expression of Il1B, Il6 and Il1r2 and intermediate expression of other cytokines receptors. LPS treatment induced (P≤0.05) expression in all cytokines and their receptors, except Il6r 3h after the administration. Melatonin attenuated (P≤0.05) LPS-induced up-regulation of Il6 but had no effect on other cytokines and their receptors and up-regulated (P≤0.05) Rev.-erbα expression under basal conditions. This indicates that melatonin from slow-release implants suppresses TLR4-mediated Il6 expression in the ovine CP via a mechanism likely involving clock genes.

Induced by a bacterial infection, an immune/inflammatory challenge is a potent negative regulator of the reproduction process in females. The reduction of the synthesis of pro-inflammatory cytokine is considered as an effective strategy in the treatment of inflammatory induced neuroendocrine disorders. Therefore, the effect of direct administration of acetylcholinesterase inhibitor&mdash, neostigmine&mdash, into the third ventricle of the brain on the gonadotropin-releasing hormone (GnRH) and luteinizing hormone (LH) secretions under basal and immune stress conditions was evaluated in this study. In the study, 24 adult, 2-years-old Blackhead ewes during the follicular phase of their estrous cycle were used. Immune stress was induced by the intravenous injection of LPS Escherichia coli in a dose of 400 ng/kg. Animals received an intracerebroventricular injection of neostigmine (1 mg/animal) 0.5 h before LPS/saline treatment. It was shown that central administration of neostigmine might prevent the inflammatory-dependent decrease of GnRH/LH secretion in ewes and it had a stimulatory effect on LH release. This central action of neostigmine is connected with its inhibitory action on local pro-inflammatory cytokines, such as interleukin (IL)-1&beta, IL-6, and tumor necrosis factor (TNF)&alpha, synthesis in the hypothalamus, which indicates the importance of this mediator in the inhibition of GnRH secretion during acute inflammation.

The study was designed to test the hypothesis that the inhibition of acetylcholinesterase (AChE) activity at the periphery by Neostigmine (0.5 mg/animal) will be sufficient to prevent inflammatory dependent suppression of the gonadotropin-releasing hormone (GnRH)/luteinising hormone (LH) secretion in ewes in the follicular phase of the estrous cycle, and this effect will be comparable with the systemic AChE inhibitor, Donepezil (2.5 mg/animal). An immune/inflammatory challenge was induced by peripheral administration of lipopolysaccharide (LPS; 400 ng/kg). Peripheral treatment with Donepezil and Neostigmine prevented the LPS-induced decrease (P<0.05) in LHβgene expression in the anterior pituitary gland (AP) and in LH release. Moreover, Donepezil completely abolished (P<0.05) the suppressory effect of inflammation on GnRH synthesis in the preoptic area, when pretreatment with Neostigmine reduced (P<0.05) the decrease in GnRH content in this hypothalamic structure. Moreover, administration of both AChE inhibitors diminished (P<0.05) the inhibitory effect of LPS treatment on the expression of GnRH receptor in the AP. Our study shows that inflammatory dependent changes in the GnRH/LH secretion may be eliminated or reduced by AChE inhibitors suppressing inflammatory reaction only at the periphery such as Neostigmine, without the need for interfering in the central nervous system.

International audience; This study was designed to determine the effect of melatonin from continuous slow-release implants on (i) the mRNA expression for the lipopolysaccharide (LPS) receptor complex, which includes toll-like receptor 4 (TLR4), myeloid differentiation-2 factor (MD-2) and cluster of differentiation-14 (CD14) in the choroid plexus (CP) and (ii) on the plasma concentration of LPS binding protein (LBP) in ewes subjected or not to a systemic inflammatory stimulus (intravenous injection of LPS). Experiments were conducted in ovariectomized, oestradiol-implanted ewes during seasonal anoestrus (May/June). In experiment 1, the ewes were divided into four groups: sham-implanted and placebo-treated (C/C), melatonin-implanted and placebo-treated (M/C, Melovine 18 mg), sham-implanted and LPS-treated for 3 h (C/LPS, 400 ng/kg of body mass) and melatonin-implanted and LPS-treated for 3 h (M/LPS). In experiment 2, the ovariectomized ewes were melatonin-implanted or sham-implanted and treated with LPS for 6 h (M/LPS and C/LPS, respectively). Plasma melatonin concentrations reached 4.5 ± 1.5 pg/ml in the C/C group, 151.4 ± 56.4 pg/ml in the M/C group, 7.8 ± 4.3 pg/ml in the C/LPS group and 240.6 ± 93.0 pg/ml in the M/LPS group one month after melatonin or sham implantation. Plasma cortisol concentrations remained at basal level throughout the experiment in the control ewes (C/C and M/C), whereas in the LPS-treated ewes, mean cortisol concentrations increased from 13.7 ± 2.4 ng/ml to 92.0 ± 10.2 ng/ml and from 15.4 ± 5.9 ng/ml to 100.0 ± 10.7 ng/ml in the C/LPS and M/LPS groups, respectively. The LPS treatment significantly (p ≤ 0.05) increased the mRNA expression for TLR4 and CD14 but had no effect on the MD-2 mRNA expression in the C/LPS and M/LPS groups. Plasma LBP concentration was not affected within 3 h of the LPS-treatment (experiment 1) but increased significantly at 4–6 h after LPS-treatment in the C/LPS and M/LPS groups (experiment 2). Melatonin from the slow-release implants did not change the mRNA expression of all the examined genes or the plasma LPB concentration. These results allow us to infer that melatonin implantation does not disturb the CP potential to detect the specific microbial components derived from gram-negative bacteria.

Contractile effect of PGF2α and PGE2 on isolated branches of uterine and ovarian artery in different days of estrous cycle and early pregnancy in pigs The contractile effects of PGF2α (3 × 10-6 to 10-4 M) and PGE2 (10-7 to 10-5 M) were examined on isolated branches of ovarian artery (OA) and extramyometrial branches of uterine artery (UA) collected from pigs in the luteal (day 10-12) and follicular phase (day 17-20) of the estrous cycle, and during early pregnancy (day 10-12). Strong contraction was demonstrated in both arteries during all investigated periods in response to PGF2α, which was significantly higher (P 2, being negligible in the follicular phase. In UA, the effective dose of PGF2α (ED50) amounted 7.9 × 10-6 M and 6.3 × 10-6 M in the luteal and follicular phase, and 5.0 × 10-6 M in early pregnancy. ED50 for PGE2 reached 5.0 × 10-7 M in the luteal phase, and 4.1 × 10-7 M in early pregnancy. For both prostaglandins, the contraction was much stronger (P 50 for PGF2α was 1.2 × 10-5 M in the luteal phase and was significantly higher (P -6 M) and early pregnancy (2.7 × 10-6 M). ED50 for PGE2 amounted 7.3 × 10-7 M in the luteal phase and 1.7 × 10-7 M in early pregnancy. Studies showed the influence of the estrous cycle and early pregnancy on OA branches sensitivity to the contractile effect of PGF2α and the lack of this effect on UA branches, and the influence of the estrous cycle on UA and OA branch contraction in response to PGE2.

Ortho-substituted polychlorinated biphenyls (PCBs) preferentially accumulate in the brain and cerebrospinal fluid (CSF) compared with other PCBs. We previously demonstrated in ewes that an identical dose of PCB153, the most environmentally prevalent congener, resulted in a higher plasma concentration during short days (SD: 1200 pg/ml) than during long days (LD: 200 pg/ml). Moreover, PCB153 treatment only reduced the SD tight junction protein content in the choroid plexus (CP), which was followed by a significant increase of the PCB153 concentration in the CSF. The aim of the present study was to evaluate how PCB153 treatment affects the VEGF/VEGFR system that maintains CSF homoeostasis and CP function. Previous work has shown that photoperiod affects the transfer rate of steroids and protein hormones from blood to CSF and modulates choroid plexus (CP) tight junction protein content. We hypothesized that the CSF proteome would also be modified by photoperiod. We tested this hypothesis by comparing CSF obtained from the third ventricle of mature, ovariectomized, estradiol-replaced ewes exposed to long day length (LD) or short day length (SD). Variations in CSF protein expression between SD- or LD-treated ewes were studied in pools of CSF collected for 48 h. Proteins were precipitated, concentrated and included in a polyacrylamide gel without protein fractionation. After in-gel tryptic digestion of total protein samples, we analyzed the resulting peptides by nanoliquid chromatography coupled with high resolution tandem mass spectrometry (GeLC-MS/MS). Quantitative analysis was performed using two methods based on Spectral Counting (SC) and eXtracted Ion Chromatograms (XIC). Among 103 identified proteins, 41 were differentially expressed between LD and SD ewes (with P < 0.05 and at least a 1.5-fold difference). Of the 41 differentially expressed proteins, 22 were identified by both methods and 19 using XIC only. Eighteen proteins were more abundant in LD ewes and 23 were more abundant in SD ewes. These proteins are involved in numerous functions including hormone transport, immune system activity, metabolism, and angiogenesis. To confirm proteomic results, two proteins, pigment epithelium-derived factor (PEDF) and gelsolin, for each individual sample of CSF collected under SD or LD were analyzed with western blots. These results suggest an important photoperiod-dependent change in CSF proteome composition. Nevertheless, additional studies are required to assess the role of each protein in seasonal functions.

The study was designed to determine the effect of proinflammatory cytokine, interleukin- (IL-) 1β, on melatonin release and expression enzymes essential for this hormone synthesis: arylalkylamine-N-acetyltransferase (AA-NAT) and hydroxyindole-O-methyltransferase (HIOMT) in ovine pineal gland, taking into account the immune status of animals before sacrificing. Ewes were injected by lipopolysaccharide (LPS; 400 ng/kg) or saline, two hours after sunset during short day period (December). Animals were euthanized three hours after the injection. Next, the pineal glands were collected and divided into four explants. The explants were incubated with (1) medium 199 (control explants), (2) norepinephrine (NE; 10 µM), (3) IL-1β(75 pg/mL), or (4) NE + IL-1β. It was found that IL-1βabolished (P<0.05) NE-induced increase in melatonin release. Treatment with IL-1βalso reduced (P<0.05) expression of AA-NAT enzyme compared to NE-treated explants. There was no effect of NE or IL-1βtreatment on gene expression of HIOMT; however, the pineal fragments isolated from LPS-treated animals were characterized by elevated (P<0.05) expression of HIOMT mRNA and protein compared to the explants from saline-treated ewes. Our study proves that IL-1βsuppresses melatonin secretion and its action seems to be targeted on the reduction of pineal AA-NAT protein expression.

The blood-cerebrospinal fluid barrier (BCSFB) located in the epithelial cells of the choroid plexus (CP) forms the interface between the cerebrospinal fluid (CSF) and pathogen components circulating in the blood. The CP is also implicated in the passage of peripheral immune signals and circulation of immune cells into the central nervous system. Toll-like receptors (TLRs) are patternrecognition receptors that play a crucial role in the recognition of pathogens and triggering of the innate immune response. In sheep, ten members of the TLR family have been identified and cloned. We used real-time PCR analyses to examine the profiles of TLR mRNA expression in the CP of cerebral ventricles in healthy adult ewes. The transcripts for all ten TLRs except TLR8 were present; however, we observed a high variation in the degree of expression of the TLR5 and TLR1 genes (coefficient of variation: 61% and 46%, respectively) as well as a moderate variation in the expression of the TLR4 (34%), TLR2 (27%) and TLR6 (26%) genes. The TLR9, TLR7, TLR3 and TLR10 genes were the four receptors with relatively invariable expression levels (coefficient of variation: 7%, 8%, 16% and 17%, respectively) across the six adult ewes. The concentration of cortisol in blood collected prior to sacrificing the ewes ranged from 0.18 to 78.9 ng/ml. There was no correlation between cortisol concentration and mRNA expression of any of the examined TLRs. These data suggest that the CP has the potential to sense the presence of many bacterial and viral components and mediate responses for the elimination of invading microorganisms, thereby protecting the brain.

Scope Reports on the protective effect of certain foods on brain functions are numerous; however, the permeability of the brain barriers by food components is still hardly recognised. There have been in vitro studies aimed at demonstrating this possibility, but not much is known about this phenomenon in in vivo systems. The objective of the study was to determine the metabolites of dietary quercetin (Q) in urine, blood plasma and cerebrospinal fluid (CSF) after intra-rumen administration of Q rich onion dry skin in an animal model. Methods and results Eleven sheep had permanently implanted cannulas in the third ventricle of the brain as the means for CSF collection. The animals were administered Q at the dose of 10 mg/kg bwt. For 12 h the concentration of Q metabolites was measured in urine, blood plasma, and CSF. It was demonstrated that while in blood plasma Q and isorhamnetin mono-glucuronides or mono-sulphates were the main metabolites (80%), in CSF their aglycones were the dominating ones (88%). Conclusion Q and IR aglycones are the main Q metabolites present in CSF after dietary Q intake. Their passive transport through blood–CSF barrier or a de-conjugating mechanism within that barrier may be involved.

Signaling and priming pheromones play an important role in intraspecies behavioral and sexual interactions and in the control of reproduction. It is generally accepted that pheromones act by stimulating the dendritic receptors in the mucus-imbedded cilia of olfactory neurons massed in the olfactory epithelium. The boar pheromone androstenol, known to induce sexual behavior in pigs, is 1 of 2 pheromones that have been chemically defined, tritiated and thus made available for use in studies. In Experiment 1, sexually mature cyclic gilts at Days 16 to 21 of the estrous cycle were humanely killed and the heads separated from the bodies. The heads were attached to a perfusion system using heated, oxygenated, heparinized, autologous blood. A total amount of 10(8) dpm (758 ng) of 3H-5 alpha-androstenol (3HA) was either infused into the angularis oculi veins that drain the nasal cavities (n = 7) over a 5-min period or applied through intranasal catheters onto the mucose surface (n = 16) for 2 min. In both groups frequent blood samples were collected from the carotid rete and from venous effluent. Concentration of 3HA in the arterial blood of the carotid rete after direct (into angularis oculi veins) or indirect (onto the nasal mucosa) administration of 3HA into veins draining the nasal cavities was significantly higher than background radioactivity before 3HA administration (P < 0.0001 and P < 0.05, respectively). The 3HA was selectively accumulated (compared with the respective control tissue) in the neurohypophysis (P < 0.001), adenohypophysis (P < 0.01), ventromedial hypothalamus (P < 0.05), corpus mammillare (P < 0.01), and perihypophyseal vascular complex (P < 0.001). In a second in vitro experiment, active uptake of 3HA into the nasal mucosa of the proximal, respiratory segment of the nasal cavity was observed. These results demonstrate a humoral pathway for the transfer of pheromones from the nasal cavity to the hypophysis and brain. Androstenol was taken up by the respiratory part of the nasal mucosa, resorbed into blood, transported to the cavernous sinus and transferred into the arterial blood of the carotid rete (supplying the hypophysis and brain), and then selectively accumulated in the hypophysis and certain brain structures.

The existence of the hormone passage from venous blood into arterial blood in the area of the perihypophyseal vascular complex has been demonstrated in some mammals, but its mechanism has not been defined. To study the regulatory mechanism we infused hCG into perihypophyseal cavernous sinus of ovariectomized, conscious ewes to test if the hCG would affect putative LH/hCG receptors and inhibit counter-current transfer of GnRH from the venous cavernous sinus to the arterial carotid rete. The latter study was done on an isolated head model. Ewes were ovariectomized in mid-anestrus and, after 4 to 5 wk were used in the experiments. On the day of experiment ewes were treated intramuscularly with estradiol benzoate or oil vehicle, and 18 to 20 h later were infused either with a multielectrolyte solution or hCG for 2 h via the venae angularis oculi. Immediately thereafter the ewes were anesthetized and exanguinated, and subsequently decapitated. The isolated head was perfused with Dextran in multielectrolyte. The 125I-GnRH was infused into the cavernous sinus via the venae angularis oculi for 5 min; contemporaneous samples were taken from the carotid rete and both jugular veins at 1-min intervals. Transfer of 125I-GnRH from the cavernous sinus to the carotid rete was inhibited by hCG in ewes pretreated with estradiol benzoate but not with oil (P0.005). We collected tissue samples from the vascular complex of the cavernous sinus and carotid rete of cyclic ewes to determine the presence of LH/hCG receptors. In situ hybridization showed the presence of LH/hCG receptor mRNA transcripts in the walls of both arterial and venous compartments of the cavernous sinus-carotid rete complex, and immunohistochemistry revealed the presence of receptor proteins. These novel findings confirm previously obtained data suggesting that LH is a modulatory factor for the counter-current transfer of neuropeptides from the venous blood of the cavernous sinus to the arterial blood supplying the brain and hypophysis. The LH could modulate 125I-GnRH transfer acting directly on the vascular smooth muscle.

Countercurrent transfer in the ovarian vascular pedicle elevates the concentration of steroid hormones in blood supplying the oviduct and periovarian part of the uterus during the estrous cycle in the pig. This study was conducted to determine whether during early pregnancy the arterial blood supply to the oviduct and uterus carries greater concentration of steroid hormone than systemic blood. The concentration of ovarian steroid hormones (progesterone, estradiol-17 beta, estrone, androstenedione and testosterone) was measured in 40 gilts on Days 12, 18, 25 or 35 of pregnancy. Silastic catheters were inserted: a) into the jugular vein, b) into the branch of uterine artery close to the ovary (proximal to the ovary) and c) into the branch of the uterine artery close to the cervix (distal to the ovary). On the day following surgery simultaneous blood samples from cannulated vessels were collected every 20 min for 3 hours. The concentration of steroid hormones was determined by radioimmunoassay. The mean concentrations of studied hormones in branches of the uterine artery proximal and distal to the ovary were significantly greater than in the jugular vein (P < 0.001) by 18 to 69% and 7 to 31%, respectively. The concentrations of hormones in proximal and distal to the ovary branch of the uterine artery were also significantly different (P < 0.001). The increase in concentrations of the measured hormones did not differ considerably between investigated days of pregnancy. It is concluded that during maternal recognition of pregnancy, formation of the corpus luteum of pregnancy, implantation of the embryo and the placenta elongation the oviduct and uterus are supplied with locally elevated concentration of steroid hormones compared to systemic blood.

The objective of the study was to determine whether the local permeability of luteinizing hormone-releasing hormone (LHRH) from the venous blood of the perihypophyseal cavernous sinus into the arterial blood of the carotid rete, supplying the brain and hypophysis in gilts, depends on the day of the estrous cycle, as well as to determine whether this transfer exists when LH concentration in the blood is reduced (the experimental short-loop negative feedback for LH secretion after estradiol injection in ovariectomized gilts). Experiments were conducted on isolated gilt heads with necks, on chosen days of the estrous cycle (n = 40), and on previously ovariectomized gilts treated with estradiol benzoate (EB) (n = 5) or corn oil (n = 3). After exsanguination, the gilt heads with necks were disarticulated and about 30–45 min later were supplied with autologous, oxygenated, and heated blood at a stable blood flow and pressure through the left carotid artery for 30 min. 125I-LHRH was infused into both cavernous sinuses through the cannulated angularis oculi veins for 5 min. After 125I-LHRH infusion, radiolabeled LHRH was found (P < 0.001) in arterial blood taken from the carotid rete through the open right carotid artery in all animals used in the experiment: on Days 1–2 (six gilts), on Days 12–14 (seven gilts) of the estrous cycle, and in five ovariectomized gilts during negative feedback for LH surge (40 hr after EB). No significant radioactivity of 125I-LHRH was found in the arterial blood on Days 3–5 (n = 6), 9–11 (n = 4), and 15–21 (n = 17) of the estrous cycle. A very low level of radioactivity was found in the ovariectomized control group after the injection of corn oil (n = 3). These results provide evidence for the permeability of LHRH from the venous to the arterial blood and its retrograde transport with the arterial blood to the hypophysis and brain, after the ovulation period (Days 1–2) and on Days 12–14 of the estrous cycle. This suggests that a close relationship exists between the day of the estrous cycle and LHRH permeability from the venous to the arterial blood in the perihypophyseal cavernous sinus—the carotid rete complex in gilts—and that this mechanism may be included in a short-loop feedback for LHRH secretion.

Ortho-substituted polychlorinated biphenyls (PCBs) preferentially accumulate in the brain and cerebrospinal fluid (CSF) compared to other PCBs. We previously reported that the same dose of 2,2',4,4',5,5'-hexachlorobiphenyl (PCB153) induced higher concentrations in the CSF of treated animals compared to controls during short days (SD), while no differences were observed during long days (LD). Similarly, the plasma concentration of PCB153 in treated ewes was higher during SD than LD. To understand the structural and molecular events explaining the photoperiodically different concentration of PCBs in the CSF in sheep, we studied the effect of photoperiod on PCB153 action on tight junction (TJ) protein expression in the choroid plexus (CP) of ewes. For that purpose, we collected CP from ovariectomised, estradiol-treated ewes maintained under artificial LD or SD and orally administered with a low dose (0.33 mg/kg/day, 3 times per week for 3 weeks) of PCB153 or vehicle. Exposure to PCB153 affected TJ proteins only during SD, and the levels of claudin-1, zonula occludens-2 (ZO-2), and afadin (AF-6) were significantly lower when compared to vehicle-treated animals. No differences were observed for occludin, junctional adhesion molecule-1 (JAM-1), claudin-5, ZO-1 and ZO-3. There was no effect of PCB153 treatment on TJ-mRNA levels. These results indicate that PCB153 selectively alters TJ proteins in the ovine CP. These alterations appear to be associated with the level of PCB153 in the blood plasma, which is modulated by the photoperiod. This study emphasises the importance of photoperiod in the susceptibility of adult sheep to PCBs.

Vascular endothelial growth factor (VEGF-A) plays an important role in maintaining cerebrospinal fluid (CSF) homeostasis and the function of the choroid plexuses (CPs). The objective of the study was to determine the expression of vascular endothelial growth factor (VEGF-A), tyrosine kinase receptors Flt-1 and KDR and KDR co-receptor neuropilin 1 (NRP-1) in ovine CPs during different photoperiods. CPs were collected from the lateral brain ventricles from ovariectomized, estradiol-treated ewes during long day (LD; 16L:8D, n = 5) and short day (SD; 8L:16D, n = 5) photoperiods. We analyzed mRNA expression levels of two VEGF-A isoforms, VEGF-A (120) and VEGF-A (164) and our results indicate that VEGF-A (164) was the predominant isoform. Expression levels of VEGF-A and Flt-1 were similar during the SD and LD photoperiods. There were significant increases in KDR mRNA and protein expression (p < 0.05) and NRP-1 mRNA expression (p < 0.05) during SD. These data show that expression of KDR and its co-receptor NRP-1 are up-regulated by short photoperiod and that this effect is not dependent on ovarian steroids. Our results suggest that the VEGF-A-system may be involved in photoperiodic plasticity of CP capillaries and may therefore be responsible for photoperiodic changes in the CSF turnover rate in ewes.

Ortho-substituted polychlorinated biphenyl (PCB) congeners, which constitute a large part of PCB residues found in the environment and in animal tissues, are known to exert potent vascular effects and can activate endothelial cells in the periphery and in the brain. The choroid plexus (CP) is responsible for cerebrospinal fluid (CSF) production and its epithelial cell layer is responsible for structure and functions of the blood-CSF barrier. The aims of this study were: 1) to investigate if environmentally relevant doses of PCB153 and similar doses of PCB104 caused changes in the expression of vascular endothelial growth factor (VEGF) - receptor system, which maintains CP function, and 2) to determine the level of both congeners in blood plasma after their oral administration. Studies of both congeners were performed on ovariectomized ewes treated per os with low doses (0.1 mg/kg, three times a week for two weeks) of PCB153 (n=4) or PCB104 (n=4) and vehicle (control, n=4). The effects of PCB153 and PCB104 treatment on mRNA expression of two isoforms of VEGF (VEGF120 and VEGF164) and their receptors Flt-1 and KDR were determined using real-time PCR. Plasma concentration of PCBs was measured using high resolution chromatography/tandem mass spectrometry (HRGC/MS-MS). We observed that neither PCB153 nor PCB104 significantly altered the mRNA of the VEGF-receptor system in the CP. In PCB treated animals plasma concentration of PCB153 (1.425 ± 0.16 ng/g of dry mass, DM) was about 150 times higher than PCB104 (0.009 ± 0.007ng/g DM). In control animals the PCB153 level was 0.14 ± 0.031 ng/g DM, while the PCB104 level was below detection level. This indicates that increase in plasma PCB153 concentration to levels similar to those reported in humans and of PCB104 concentration to levels 100 times higher than those found in human plasma did not affect the VEGF-receptor system in the CP in adult ewes. The significantly lower increase of PCB104 than PCB153 concentration in blood after oral administration suggests different absorption of both congeners from the digestive tract.

In sheep, phytoestrogens are known to act at various levels on the hypothalamo–pituitary–gonadal axis, as well as the hypothalamo–pituitary–thyroid axis. The levels of genistein and daidzein and their metabolites, p-ethylphenol and equol, in the cerebrospinal fluid (CSF) were studied in ewes fed red clover silage. Moreover, to test the hypothesis that phytoestrogens may affect the access of thyroid hormones in the brain, the concentrations of both total (TT4) and free thyroxine (fT4) in the blood and CSF, were recorded. In the CSF, daidzein, equol and p-ethylphenol were present, but genistein absent. The total daidzein concentration increased from an undetectable level at 2 h, to 0.11 ± 0.05 μM at 6 h after feeding, representing 13.1% of the daidzein measured in the blood. Gradual increases in equol, from an undetectable level at 2 h, to 0.24 ± 0.13 μM at 6 h after feeding (5.1% of that in blood), and in p-ethylphenol, from 0.03 ± 0.02 μM at 2 h to 1.7 ± 0.74 μM at 6 h after feeding (0.1–5.8% of that in blood) were recorded. Sexually mature cycling gilts (n = 5) with two recorded oestrous cycles were anaesthetized on Day 10 of the oestrous cycle. On both sides of the uterus, catheters were inserted into branches of the uterine artery proximal to the ovary. The tip of each catheter was placed close to, but above anastomoses with ovarian artery branches. This allowed collection of uterine arterial blood mixed with ovarian arterial blood supplying the uterus and oviduct. A third catheter was placed in the jugular vein. Catheters were exteriorized subcutaneously to the backs of the animals for blood sample collection. Simultaneous blood samples were collected as follows: (a) in anaesthetized animals, every 5 min for 1 h immediately after catheters were inserted during surgery, and every 10 min for 2 h, when the surgery was completed; (b) from conscious animals on Day 2 after surgery, every 10 min for 2 h in the morning and every 10 min for 2 h in the afternoon. Blood plasma samples were assayed for progesterone and androstenedione by RIA method. Mean values (±SEM) for progesterone measured in samples collected from branches of the uterine arteries and from the jugular vein in anaesthetized animals were 34 ± 1.25 ng ml−1 and 26 ± 1.31 ng ml−1 of plasma, respectively. In conscious animals the measurements showed 33 ± 0.90 ng ml−1 and 24 ± 0.81 ng ml−1 of plasma, respectively. Plasma concentration of androstenedione in branches of the uterine arteries and from the jugular vein in anaesthetized and conscious animals were 107 ± 5.26 pg ml−1 and 72 ± 5.50 pg ml−1, and 78 ± 4.54 pg ml−1 and 57 ± 3.77 pg ml−1, respectively. It was concluded that counter-current transfer of steroid hormones in ovarian pedicle vasculature on Days 10–11 of the oestrous cycle significantly elevates (P < 0.0001) the local concentration of steroid hormones in arterial blood supplying the uterus and oviduct (35% for progesterone and 46% for androstenedione).

The effects of photoperiod on the cerebrospinal fluid (CSF) concentration of six ortho -substituted polychlorinated biphenyls (PCBs: PCB28, PCB52, PCB101, PCB138, PCB153, and PCB180), the effects of an orally administered low dose of PCB153 (0.3 mg/kg, three times a week for three weeks) on PCBs and thyroid hormones (THs) concentrations in the CSF and plasma, and the release of luteinizing hormone (LH) were determined in ovariectomized, estradiol-implanted ewes (2.5 years old) maintained indoors under artificial long day (LD, 16L: 8D) and short day (SD, 8L: 16D) conditions. Concentrations of two PCBs (PCB28 and PCB153) in the plasma and four PCBs in the CSF (PCB101, PCB138, PCB153, and PCB180) were significantly higher during LD than SD. Following PCB153 treatment, its concentration in the plasma was higher in SD (1.2±0.3 ng/ml) than LD (0.2±0.05 ng/ml), but similar in the CSF (10.2±3.7 pg/ml vs. 13±0.7 pg/ml) under both photoperiods. During SD, the concentration of PCB153 in the CSF was higher in treated animals than controls, while no differences were noted under LD. These findings indicate that in ewes, exposure of the brain to more highly chlorinated, ortho -substituted PCBs may be modulated by photoperiod. PCB153 treatment had no effect on plasma THs, but reduced total triiodothyronine concentration during LD and free thyroxine during SD in the CSF. Under both photoperiods, PCB153 reduced basal plasma LH and reinforced the inhibition of pulsatile LH release during LD. As PCB153 reduced LH and THs (which are involved in the seasonal control of reproduction in ewes), it may have a braking effect on seasonal transitions between active and inactive phases of reproduction.

Thyroid hormones (THs) are obligatory for transition from breeding season to anestrus in sheep. In this process, THs act during a very limited time of the year and primarily within the brain. In ewes chronically equipped for sampling cerebrospinal fluid (CSF) from the third ventricle, we have characterized the concentrations of total and free thyroxine (T4), triiodothyronine (T3), and total reverse T3 (rT3) in the CSF during breeding season, anestrus and during a critical period required for transition to anestrus (December-March). The total T4, T3, rT3 and free T3 average concentrations (+/- SEM) in CSF were 1.5 +/- 0.07 ng/ml, 14.5 +/- 1.2 pg/ml, 43 +/- 7.4 pg/ml, and 0.6 +/- 0.05 pg/ml, respectively, and all were significantly lower (p < 0.001) than in blood plasma except free T4 (12.6 +/- 1.1 pg/ml), which was similar to that in plasma. There was a seasonal trend (p < 0.05) in the concentration of total T3 (highest in December) and free T4 (highest in November) in the CSF that does not follow that in blood plasma. During the period of transition to anestrus the CSF total T3/TT4 molar ratio and free T3/T4 ratio were significantly lower (p < 0.05 and p < 0.01, respectively) than in blood plasma, while the total rT3/T4 ratio was significantly higher (p < 0.01) at the end of this period (March). Additionally, the CSF total rT3 concentrations were also significantly correlated with the CSF total T4 levels (r = 0.57; p < 0.05). In conclusion, the CSF in sheep may serve as a considerable source of thyroid hormones for neuroendocrine events. The lack of significant changes in THs concentrations in the CSF during the period of transition to anestrus indicate that neither seasonal changes of THs circulating in the blood plasma nor THs circulating in the CSF actively drive the transition to anestrus.

Sheep from temperate latitudes exhibit seasonal variations in many physiological functions such as reproduction, food intake, body weight, and pelage growth. Majority of seasonal changes are controlled by the annual photoperiodic cycle and melatonin secretion. For reproduction, the resulting key event is a modulation of the negative feedback of steroids on gonadotropin secretion. However, this seasonal effect could also depend on variable uptake of steroids by the brain. Seasonal regulation of food intake also involves numerous peripheral hormones, among which the protein hormone leptin informs the brain on the metabolic status of the animal. It has been shown previously that access of progesterone, estradiol and leptin to the cerebrospinal fluid (CSF) increases under long days. This physiological modulation of the passage of hormones to the brain could depend on regulation of the permeability of the blood-CSF barrier. This study therefore compared the tight junction proteins in the choroid plexus of ewes exposed to short days or long days. Levels of occludin, zonula occludens proteins (ZO) ZO-1 and ZO-2, afadin and cadherin were significantly higher during short days, but no statistical difference was observed for junctional adhesion molecule 1 (JAM-1), ZO-3 or claudins 1 and 5. These results are consistent with an increase in the blood-CSF barrier permeability during long days through a regulation of tight junctions and show that the permeability could depend upon physiological conditions such as photoperiodic status.

This study was conducted in order to scrutinise the countercurrent exchange of luteinising hormone releasing hormone (LH-RH), β-endorphin and progesterone in the carotid rete and cavernous sinus of sheep and gilts. The ewes (n=30) and gilts (n=20) were anaesthetised and had catheters inserted into their jugular veins and carotid common arteries from which heparinised blood was then collected. After exsanguination, the head with the neck was immediately removed and about 30 min later supplied with oxygenated and heated autologous blood mixed with saline (1:1), through one of the carotid arteries at a stable flow rate. Blood samples were collected every minute from the carotid artery contralateral to the arterial blood supply (blood from the carotid rete) and from both jugular veins. Arterial blood pressure was continuously measured and regulated. Radiolabelled hormones 3H-progesterone (3H-P4), 125I-luteinising hormone releasing hormone (125I-LH-RH), 125I-β-endorphin (125I-En), or 51Cr-labelled red blood cells (control) were infused for 5 min into the cavernous sinuses through catheterised angularis oculi veins. Infusions were carried out on gilts and sheep which were in different phases of the oestrous cycle and on sheep during anoestrus and after oestrogen treatment. As a control, 3H-P4 was infused into the facial vein instead of into the cavernous sinus. In all experiments with 3H-P4 and 125I-En infusions, labelled hormones were found in arterial blood collected from the carotid rete. No radioactivity was present in arterial blood in the experiment with seasonally anoestrous sheep. However, 125I-LH-RH was found in the arterial blood of anoestrous oestrogen-treated sheep and of sheep in the early luteal phase. No radioactivity was ascertained in arterial blood after 3H-P4 infusion into the facial vein (control) or 51Cr-labelled red blood cells into the cavernous sinus (control). These results demonstrate that two neuropeptides (LH-RH and β-endorphin) and a steroid hormone (progesterone) can be transferred from venous to arterial blood at the cavernous sinus and carotid rete in the base of the brain in sheep and pigs. This indicates that there is a powerful exchange system for resupplying hormones to the brain and that this system is dependent on the phase of the oestrous cycle.

The authors measured the effects of exogenous melatonin treatment on the concentrations of total (T) and free (f) fractions of thyroxine (T4) and triiodothyronine (T3) in cerebrospinal fluid (CSF) and blood plasma as well as the expression of their binding/transporter protein, transthyretin (TTR), in the choroid plexus of ewes from May to August. Melatonin implantation in May and July mainly prevented the decrease in plasma for fT3 and TT3 exhibited in untreated group, and induced a limited decrease in TT4 in June. By contrast, melatonin implantation prevented the decrease in CSF fT3 observed in the untreated group. No effect of melatonin was found on the expression of TTR mRNA in the choroid plexus There were a correlations between blood fT4 and CSF TT4 concentrations in both control and melatonin treated group (r(2)-0.4; P < 0.01 vs. r(2)-0.14; P < 0.05), as well as between blood fT3 and CSF TT3 concentrations but only in the melatonin-treated group (r(2)-0.26; P < 0.02). We conclude that T3, the active form of the hormone within the brain, is regulated by melatonin independently of the peripheral changes within the blood. The lack of correlation between plasma fT3 and CSF TT3 in the control group suggests that an increase in local T3 conversion could contribute as an additional source of T3 in the CSF during the period of increasing day length. These data seem to confirm a local nature for recently discovered connections between the pineal melatonin signal and thyroid-dependent seasonal biology in mammals.

The cerebrospinal fluid (CSF) is a major part of the extracellular fluid of the central nervous system. The function of the CSF and the tissue that secretes it, the choroid plexus (CP) has traditionally been thought as providing the brain with essential nutrients, removing products of neuronal activity of the central nervous system, and providing mechanical support for the brain's fragile cellular network. More recent studies suggest, however, that the CP and CSF system play a much more active role in the function of the central nervous system being a target, source and pathway for neuroendocrine signaling within the brain.

The present study examines the direct effect of luteinising hormone (LH) on the reactivity of the porcine uterine artery to norepinephrine (NE). Three-mm-long arterial segments collected during the luteal phase of the oestrous cycle were mounted in an organ bath for isometric tension recording. After 30 min of equilibration in optimal passive tone, one part of the vessels was treated with 10 ng/ml of LH in PBS (experimental), while a second part of the arterial segments was treated with 10 ng/ml of bovine serum albumin (BSA) in PBS (control). After 30 min of equilibration, NE was given to each organ bath in a cumulative concentration manner, ranging between 1 × 10 −8 mol/l to 3 × 10 −4 mol/l. NE caused a dose-dependent contraction of all experimental and control arteries. The addition of LH caused a rightward shift of the dose-response curve to NE. The corresponding EC 50 values were 2.17 (± 0.39) μmol/l in PBS-pretreated vessels and 3.35 (± 0.41) μmol/l in LH-pretreated vessels (P < 0.05). The results of the present study demonstrate that LH attenuates the vascular response to NE in third-order branches of the uterine artery. Therefore, it can be suggested that besides the known effect of LH-hCG on the formation of vasoactive eicosanoids, an additional mechanism is involved in the direct action of LH on blood flow in the uterine arteries in pigs.

The objective of the study was to determine whether luteinizing hormone (LH) and prolactin (PRL) can access the brain by way of transfer from the venous blood of the cavernous sinus to the arterial blood supplying the brain and hypophysis. Studies were performed on heads of 22 mature sheep isolated during different phases of the estrous cycle and perfused with autologous blood. We were not able to demonstrate any transfer of LH and PRL in the investigated periods. This suggests that molecular weight of hormone may be a main factor determining the permeation and transfer of hormones in the perihypophyseal vascular complex.

In female sheep, estradiol-dependent dopaminergic inhibition exerted by the A15 nucleus during long days (LD) results in a blockade of reproductive activity. This effect could involve the GnRH cell bodies or their terminals in the median eminence (ME). However, a vast majority of terminals of the A15 nucleus are located in neurohypophysis and only a few in the ME. Previously we demonstrated that tritiated dopamine (DA) was transferred from the venous blood of the cavernous sinus to the arterial blood supplying the brain. In the present paper, we tested the hypothesis that the transferred dopamine could reach further the brain and ME. Using isolated sheep heads harvested on short days vs. long days, we examined radioactivity in brain tissues following infusion of tritiated dopamine into the cavernous sinus. The experiment was performed in ovariectomized ewes treated with estradiol (E2) or vehicle. The mean level of radioactivity in brain was affected by season (p0.001) and E2 (p0.05) and was the highest during LD in E2-treated animals. In the next experiment on isolated sheep head we measured dopamine and its metabolites levels in blood and pituitary after infusion of non-radiolabeled dopamine. We observed an increase (p0.01) in dopamine concentration in arterial blood but not in the brain. The pituitary was the only structure examined in which a tendency (p=0.06) towards increased dopamine concentration following dopamine infusion was observed. Thus, even if part of DA released from terminals within the posterior and intermediate lobes of the pituitary reaches the vessels of the ME through local vascular pathways, it is unlikely that it could affect the LHRH terminals located in ME. In addition, our results suggest that brain capillaries in the isolated head are able to maintain a functional blood brain barrier.

In the present study we examined whether local transfer of intranasally administrated tritiated progesterone (3H-P4) would increase its concentration in blood supplying the brain and hypophysis in comparison with other organs. Additionally, the effect of estrous cycle on the P4 transfer was evaluated on isolated gilts' heads. In the first experiment 3H-P4 was instilled into the nasal cavities of anaesthetized, immature pigs (n=10). Simultaneous blood samples were collected for radioactivity measurement every minute from the same occluded carotid artery through two catheters; one catheter was pointed towards the head, the other one towards the heart. In eight animals the ratio calculated between the 'head' and 'heart' samples was significantly (p0.05) higher than 1 and reached a mean (+/- SEM) level of 3.23 +/- 0.81. In two animals a much higher ratio was observed. A head/heart ratio1 indicates an existence of local transfer of 3H-P4 from venous blood to the carotid blood. In the second experiment, heads of 26 mature, cycling gilts were perfused through the right carotid artery with autologous blood. The outflow from the left carotid artery was collected as 1 min samples. 3H-P4 was infused into the angularis oculi veins. Transfer of 3H-P4 from the venous blood into the arterial blood reached the mean (+/- SEM) level of 4.11 +/- 1.08 pg/ml on days 2-4, 3.2 +/- 0.70 on days 17-21 and 0.94 +/- 0.22 pg/ml on days 15-16 of the estrous cycle. No 3H-P4 transfer was observed on days 9-11. These findings demonstrate that nasally administered progesterone can reach the brain in locally higher concentration through the vascular pathway. Moreover, the between-vessel transfer of P4 is significantly affected by the stage of the estrous cycle.

It is generally accepted that pheromones act by stimulating of the dendritic receptors of the olfactory neurones massed in the olfactory epithelium. This study was designed to ascertain whether it is possible for the boar pheromone androstenol (5alpha-androst-16-en-3-ol) to be transported from the nasal cavity of anaesthetized gilts to the brain and hypophysis via local transfer from the blood in the perihypophyseal vascular complex. The experiment was performed on days 18-21 of the porcine oestrous cycle (crossbred gilts, n = 6). Tritiated androstenol (3H-A; total amount 10(8) d.p.m. (758 ng)) was applied for 1 min onto the respiratory part of the nasal mucosa, 4-6 cm from the opening of the nares. Arterial blood samples from the aorta and from the carotid rete were collected every 2 min during the 60 min period following administration of the steroid. Total radioactive venous effluent from the head was removed and an adequate volume of homologous blood was transfused into the heart through the carotid external vein. At the end of the experiment gilts were killed and tissue samples of the hypophysis and some brain structures were collected to measure radioactivity. In addition, corresponding control tissues were collected from three untreated gilts and from three heads of gilts 60 min after 3H-A was applied post mortem into the nasal cavity. The concentration of 3H-A was significantly higher (P0.0001) in the arterial blood of the carotid rete than that of aorta. The mean rate of 3H-A counter current transfer from venous to arterial blood in the perihypophyseal vascular complex, expressed as the ratio of the 3H-A concentration in arterial blood of the carotid rete to the 3H-A concentration in blood sampled simultaneously from the aorta, was 1.96 +/- 0.1. The concentration of 3H-A in plasma from the venous effluent from the head ranged from 1.3 to 1.8 pg x ml(-1). During the 60 min period of the experiment, 0.68% of the total applied dose of 3H-A was resorbed from the nasal cavity into the venous blood. Moreover, we found that 3H-A was present in the olfactory bulb (P0.01), amygdala, septum, hypothalamus, adenohypophysis, neurohypophysis (P0.05) and perihypophyseal vascular complex (P0.01). These results demonstrate that, in anaesthetized gilts, the boar pheromone androstenol may be resorbed from the nasal mucosa, transferred in the perihypophyseal vascular complex into arterial blood supplying the brain and hypophysis, and then arrested in the hypophysis and certain brain structures. We suggest that in addition to the standard neural pathway for signalling pheromones, another pathway exists whereby androstenol, as a priming pheromone, may be resorbed from the nasal cavity into the bloodstream and then pass locally from the perihypophyseal vascular complex into the arterial blood supplying the brain and hypophysis, thus avoiding the first passage metabolism in the liver.

Intraspecific communication by chemical signals (pheromones) plays important behavioral and physiological roles in coordinating reproduction in mammals. Females of a variety of domestic species respond to pheromones from males by undergoing an earlier onset of puberty (Brooks and Cole, 1970; Kirkwood et al., 1983; Kalbrom, 1982; Pearce and Paterson, 1992), alterations in ovarian cycles (Oldham et al., 1979; Booth and Baldwin, 1983; Booth and Signoret, 1992) and estrous behavior (Signoret, 1970; Domes et al., 1997). The precocious attainment of puberty in gilts was studied after contact with the boar (Kirkwood et al., 1983; Kalbrom, 1982) or following the treatment with the male pheromone androstenone, commercially available as an aerosol spray called Suidor (Glei et al., 1989). According to Pearce and Paterson (1992), physical contact with the boar is essential for the maximal pubertal acceleration as it allows the direct transfer of the priming pheromone from the boar to the snout of the recipient gilt; among adult females, nosing and sniffing of the genitalia corresponded to 47% of interactions between the boar and anestrous gilts, while the remaining 43% of interactions involved head-to-head contacts (Signoret, 1970).

This study was performed to answer the question of whether counter current retrograde transfer of -endorphin in the perihypophyseal cavernous sinus-carotid rete vascular complex depends on the reproductive activity of sheep and if this transfer depends on membrane Na + K + ATP-ase blocking by ouabain. Sheep were anaesthetised and the jugular vein and the carotid artery were cannulated on both sides. Multielectrolitical liquids (Solfin, Polfa Kutno, Poland): 500 ml of Solfin with heparin (25,000 IU), or Solfin with heparin and ouabain (Sigma, St. Louis, USA) in concentrations of 10 -5 or 10 -4 mol 1 -1 were infused into the brain through the carotid artery. Heparinized blood was collected through the carotid artery. After exsanguination, the head with the neck was removed. The isolated head was supplied with oxygenated, heated, autolog-ous blood diluted with Solfin (4:1) without or with ouabain in concentration of 10 -5 or 10 -4 mol 1 -1 . Blood pressure and temperature were measured throughout the duration of the experiment. During the experiment 125 I-β-endorphin (7.9 X 107 dpm) dissolved in 10 ml of Solfin was infused for 5 min (5 ml) into each cavernous sinus through the angularis oculi veins. Blood samples for radioactivity measurements were collected each min from the carotid rete (through the opposite carotid artery to the artery supplying the brain with arterial blood) and from both jugular veins. In all the experiments significant 125 I-β-endorphin radioactivity was found in arterial blood supplying the brain and hypophysis in the early luteal phase in sheep. No radioactivity was found (with the exception of one animal) in sheep during seasonal anoestrus. A blockage of Na + K + ATP-ase by ouabain administered during exsanguination and during head perfusion with dose of 10 -4 mol 1 - 1 ' reduced β-endor-phin counter current transfer to arterial blood, but this effect was not evident with the dose of 10 -5 mol 1 -1 . Increased blood pressure was observed in all the experiments with either dose of ouabain.

It has been reported that apoptosis plays an essential role in controlling the physiological cell kinetics in the human and rodent endometrium but this type of death has never been studied in the porcine endometrium. The aim of this study was to investigate the apoptotic cell death in the porcine endometrium during the middle (Days 9–11) and late (Day 13) luteal phase, during the luteolysis (Day 15) and early follicular phase (Days 17–19) of the oestrous cycle. Apoptotic cells were identified by in situ DNA 3′-end labelling method. it was revealed that the greatest number of apoptotic cells in the luminal and glandular epithelium was found on Days 17–19 and on Day 15 of the oestrous cycle, respectively. in the stroma, the greatest number of these cells was found on Days 9–11. Our data have shown that in the porcine endometrium, both epithelial and stromal cells undergo apoptosis and that the number of apoptotic cells varies depending on the phase of the oestrous cycle.
