- Abstract
Fish species show distinct differences in their muscular concentrations of imidazoles and free amino acids (FAA). This study was conducted to investigate whether metabolic response to mildly elevated water temperature (MEWT) relates to species-dependent muscular concentrations of imidazoles and FAA. Thirteen carp and 17 Nile tilapia, housed one per aquarium, were randomly assigned to either acclimation (25°C) or MEWT (30°C) for 14 days. Main muscular concentrations were histidine (HIS; P<0.001) in carp versus N-α-acetylhistidine (NAH; P<0.001) and taurine (TAU; P=0.001) in tilapia. Although the sum of imidazole (HIS+NAH) and TAU in muscle remained constant over species and temperatures (P>0.05), (NAH+HIS)/TAU ratio was markedly higher in carp versus tilapia, and decreased with MEWT only in carp (P<0.05). Many of the muscular FAA concentrations were higher in carp than in tilapia (P<0.05). Plasma acylcarnitine profile suggested a higher use of AA and fatty acids in carp metabolism (P<0.05). On the contrary, the concentration of 3-hydroxyisovalerylcarnitine, a sink of leucine catabolism, (P=0.009) pointed to avoidance of leucine use in tilapia metabolism. Despite a further increase of plasma longer-chain acylcarnitines in tilapia at MEWT (P=0.009), their corresponding beta-oxidation products (3-hydroxy-longer-chain acylcarnitines) remained constant. Together with higher plasma non-esterified fatty acids (NEFA) in carp (P=0.001), the latter shows that carp, being a fatter fish, more readily mobilises fat than tilapia at MEWT, which coincides with more intensive muscular mobilization of imidazoles. 6. Impact of parity on bone metabolism throughout the reproductive cycle in sows.

- Abstract
Bone metabolism fluctuates throughout the reproductive cycle of sows to enable foetal growth and milk production. Although increased bone mineralisation is conceivable in sows during reproduction, a study of mineralisation in function of parity has not been performed. This study evaluated the fluctuations of markers for bone metabolism in primiparous and multiparous sows throughout a reproductive cycle. The experiment included ten multiparous and five primiparous commercial hybrid sows from one herd. The sows were monitored for one reproductive cycle and fed according to commercial dietary standards. Blood samples were taken in the morning before feeding at fixed time intervals before (day -5) and during gestation (insemination (day 0), 21, 42, 63, 84), around parturition (day 108, 112, parturition (115), 118), and during lactation (day 122, 129, 143). Serum osteocalcin (OC) concentration increased in early and mid-gestation (P=0.002) and decreased at the end of gestation (P=0.001), whereas crosslaps (CTX) concentration decreased during early and mid-gestation (P=0.002) and increased towards the end of gestation (P=0.001). Towards the end of lactation serum levels of both markers increased (P=0.007 and 0.013, respectively). For hydroxyproline (HYP) no significant fluctuation in function of the reproductive cycle was detected. Matrix metalloproteinase 2 (MMP2) concentration increased towards parturition for both primiparous and multiparous sows (P=0.001), whereas during lactation no significant fluctuations in function of the reproductive cycle were found. A parity effect was found for OC and CTX (P<0.010), but not for the other markers. These results demonstrate that bone metabolism differed between primiparous and multiparous sows, although in both groups a similar fluctuation throughout the reproductive cycle was observed.

7. Locomotion Disorders and Skin and Claw Lesions in Gestating Sows Housed in Dynamic versus Static Groups.

- Abstract
Lameness and lesions to the skin and claws of sows in group housing are commonly occurring indicators of reduced welfare. Typically, these problems are more common in group housing than in individual housing systems. Group management type (dynamic versus static) and stage of gestation influence the behavior of the animals, which in turn influences the occurrence of these problems. The present study compared prevalence, incidence and mean scores of lameness and skin and claw lesions in static versus dynamic group housed sows at different stages of gestation during three consecutive reproductive cycles. A total of 10 Belgian sow herds were monitored; 5 in which dynamic groups and 5 in which static groups were utilized. All sows were visually assessed for lameness and skin lesions three times per cycle and the claws of the hind limbs were assessed once per cycle. Lameness and claw lesions were assessed using visual analogue scales. Static groups, in comparison with dynamic groups, demonstrated lower lameness scores (P<0.05) and decreased skin lesion prevalence (24.9 vs. 47.3%, P<0.05) at the end of gestation. There was no difference between treatment group regarding claw lesion prevalence with 75.5% of sows demonstrating claw lesions regardless of group management. Prevalences of lameness (22.4 vs. 8.9%, P<0.05) and skin lesions (46.6 vs. 4.4%, P<0.05) were highest during the group-housed phase compared to the individually housed phases. Although the prevalence of lameness and skin lesions did not differ three days after grouping versus at the end of the group-housing phase, their incidence peaked during the first three days after moving from the insemination stalls to the group. In conclusion, the first three days after grouping was the most risky period for lameness incidence, but there was no significant difference between static or dynamic group management., Competing Interests: The authors have declared that no competing interests exist.

Background: Inflammatory bowel disease (IBD) refers to a diverse group of chronic gastrointestinal diseases, and gut microbial dysbiosis has been proposed as a modulating factor in its pathogenesis. Several studies have investigated the gut microbial ecology of dogs with IBD but it is yet unclear if this microbial profile can alter the nutrient metabolism of the host. The aim of the present study was to characterize the faecal bacterial profile and functionality as well as to determine host metabolic changes in IBD dogs. Twenty-three dogs diagnosed with IBD and ten healthy control dogs were included. Dogs with IBD were given a clinical score using the canine chronic enteropathy clinical activity index (CCECAI). Faecal short-chain fatty acids (SCFA) and ammonia concentrations were measured and quantitative PCR was performed. The concentration of plasma amino acids, acylcarnitines, serum folate, cobalamin, and indoxyl sulfate was determined., Results: No significant differences in the abundance of a selection of bacterial groups and fermentation metabolites were observed between the IBD and control groups. However, significant negative correlations were found between CCECAI and the faecal proportion of Lactobacillus as well as between CCECAI and total SCFA concentration. Serum folate and plasma citrulline were decreased and plasma valine was increased in IBD compared to control dogs. Increased plasma free carnitine and total acylcarnitines were observed in IBD compared with control dogs, whereas short-chain acylcarnitines (butyrylcarnitine + isobutyrylcarnitine and, methylmalonylcarnitine) to free carnitine ratios decreased. Dogs with IBD had a higher 3-hydroxyisovalerylcarnitine + isovalerylcarnitine to leucine ratio compared to control dogs., Conclusions: Canine IBD induced a wide range of changes in metabolic profile, especially for the plasma concentrations of short-chain acylcarnitines and amino acids, which could have evolved from tissue damage and alteration in host metabolism. In addition, dogs with more severe IBD were characterised by a decrease in faecal proportion of Lactobacillus.

9. Assessment of feed resources, feeding practices and coping strategies to feed scarcity by smallholder urban dairy producers in Jimma town, Ethiopia.

- Abstract
Smallholder dairy production is increasingly becoming popular in Jimma town. However, feed shortage is a major constraint to dairy production. The objectives of this study was to assess feed resources, feeding practices and farmers' perceived causes of feed shortage and coping strategies to feed scarcity in smallholder dairy producers in Jimma town, Oromia Regional State, Ethiopia. A total of 54 randomly selected dairy farmers were interviewed using a pre-tested structured questionnaire and through direct observations. Twenty major feed types used by dairy farmers were identified and categorized into five classes: natural pasture grazing, green feeds, hay, concentrate (commercial mix and agro-industrial by-products) and non-conventional feed resources. Green feeds-fresh or succulent grasses and legumes (mean rank = 0.361), concentrate (0.256), hay (0.198), non-conventional feeds (0.115) and natural pasture grazing (0.070) were ranked as the main feed resources in that order of importance. Green feed (94.4 % of the respondents) was found to be the main basal diet of dairy cattle. Overall, wheat bran (85.2 % of the respondents), commercial concentrate (55.6 %), noug (Guizotia abyssinica) cake (20.4 %), cotton seed cake (7.4 %) and molasses (7.4 %) were the main concentrate supplements used (P > 0.05). Local brew waste (attela) (77.8 % of the respondents), bean and pea hulls (42.6 %) enset (Ensete ventricosum) leaf and pseudo-stem (37 %), sugarcane tops (33.3 %), banana leaf and stem/stover (16.7 %) and papaya stem (16.7 %) were the dominant non-conventional feed resources in the surveyed area (P > 0.05). About 79.6, 7.4, 1.9 and 11.1 % of the farmers used zero-, semi-zero-, and the combination of zero- and semi-zero- and free-grazing systems, respectively. Most farmers (90.7 %) offered concentrate supplements to milking cows. However, supplementation did not consider milk yield, physiological status and condition of cows. All the farmers (100 %) offered common salt to their cattle as mineral supplement. The majority (98.1 %) of the farmers experience feed shortage in the dry season. Land scarcity (55.6 % of the respondents) was reported as the most important cause of feed scarcity followed by a combination of land scarcity and poor feed availability (42.2 %). Increasing use of agro-industrial by-products and commercial concentrate mix (87 % of the respondents), increasing use of hay (74.1 %), increasing use of non-conventional feeds (50 %), purchasing green feeds (19.8 %) and reducing herd size (2.7 %) were the strategies adopted for coping with feed scarcity. From results of this study, it could be concluded that to ensure sustainable availability of dairy cattle feed in the surveyed area, technological, technical and institutional innovations would be vital.

10. Effect of rubber flooring on group-housed sows' gait and claw and skin lesions.

- Abstract
This study evaluated the influence of floor type on sow welfare in terms of lameness, claw lesions, and skin lesions. In a 2 × 3 factorial design, we have investigated the effect of rubber coverings on concrete floors and the effect of 3 levels of dietary zinc supplementation on locomotion and claw and skin lesions in group-housed sows. Six groups of 21 ± 4 hybrid sows were monitored during 3 successive reproductive cycles. The sows were group housed from d 28 after insemination (d 0) until 1 wk before expected farrowing date (d 108) in pens with either exposed concrete floors or concrete floors covered with rubber in part of the lying area and the fully slatted area. During each reproductive cycle, locomotion and skin lesions were assessed 4 times (d 28, 50, 108, and 140) and claw lesions were assessed twice (d 50 and 140). Results are given as least squares means ± SE. Locomotion and claw scores were given in millimeters, on analog scales of 150 and 160 mm, respectively. Here, we report on the effect of floor type, which did not interact with dietary zinc concentration ( > 0.10 for all variables). At move to group (d 28) and mid gestation (d 50), no differences between floor treatments were seen in locomotion ( > 0.10). At the end of gestation (d 108), sows housed on rubber flooring scored 9.9 ± 4.1 mm better on gait ( < 0.001). Regarding claw disorders, both parameters "heel overgrowth and erosion" (difference of 4.6 ± 1.8 mm; = 0.01) and "heel-sole crack" (difference of 3.1 ± 1.5 mm; = 0.04) scores were better for sows on rubber flooring at mid gestation (d 50). However, sows on rubber flooring scored worse for "vertical cracks in the wall horn" (difference of 3.4 ± 1.7 mm; = 0.04). At the end of lactation (d 140), both "white line" (difference of 2.9 ± 1 mm; = 0.02) and "claw length" (difference of 4.7 ± 1.4 mm; < 0.001) had better scores on rubber flooring. No differences for skin lesions were observed between floor treatments. The improved scores for gait toward the end of gestation and some types of claw disorders at mid gestation suggest that rubber flooring in group housing has a beneficial effect on the overall leg health of sows. The documented increase in vertical cracks in the wall horn at d 50 requires further investigation.

11. Does release of encapsulated nutrients have an important role in the efficacy of xylanase in broilers?

- Abstract
The non-starch polysaccharides (NSPs) in cell walls can act as a barrier for digestion of intracellular nutrients. This effect is called "cage effect." Part of the success of fibrolytic enzymes in broiler feed is assumed to be attributed to cage effect reduction. Further, changes in viscosity and potential prebiotic action should also be considered. The aim of this study was to gain insight into the relative importance of the cage effect in xylanase efficacy in broilers. Using a 2×2 factorial design, 24 pens with 30 Ross 308 male chicks were fed corn-soy based diets consisting of normal and freeze-thawed (5 d at -18°C) corn, both with and without xylanase. The freeze-thaw method was used to eliminate the cage effect, whereas a corn-based diet was used to exclude viscosity effects. Body weights (BW), feed intake (FI), and feed conversion ratio (FCR) were determined at d 13, 26, and 39. A balance study was executed at the end of the growing phase. These birds were euthanized at d 34 (non-fasted) to determine the viscosity of digesta, blood metabolites, intestinal morphology, and microbiota composition. During the finisher period, there was a significant interaction between enzyme supplementation and freeze-thawing for FCR, in which FCR was improved by freeze-thawed corn and tended to be improved by normal corn+enzyme compared with the control group. The improvement in performance (finisher period) of freeze-thawed corn and xylanase coincided with increased gut absorption of glucose (based on postprandial plasma concentrations) and increased number of Clostridiumcluster IV in the caecum, and agreed with the higher gut villus height. In addition, xylanase inclusion significantly increased the postprandial plasma glycine and triglycerides concentration, and led to elevated bacterial gene copies of butyryl CoA:acetate CoA-transferase, suggesting a prebiotic effect of xylanase addition through more than just the cage effect reduction. 12. Factors affecting mechanical nociceptive thresholds in healthy sows.

Objective: To describe anatomical and methodological factors influencing mechanical nociceptive thresholds (MNTs) and intra-site variability in healthy sows., Study Design: Prospective, randomized validation., Animals: Eight pregnant, healthy, mixed-parity sows (176-269 kg)., Methods: Repeated MNT measurements were taken: 1) with a hand-held probe and a limb-mounted actuator connected to a digital algometer; 2) at nine landmarks on the limbs and tail; and 3) at 1 and 3 minute intervals. Data were analysed using linear mixed regression models., Results: The MNTs (±SEM) of the limbs were lower with the probe (14.7 ± 1.2 N) than with the actuator (21.3 ± 1.2 N; p < 0.001), in the pelvic versus the thoracic limbs (16.7 ± 1.2 versus 19.2 ± 1.2 N; p < 0.001), and in the lateral versus the dorsal metatarsi and metacarpi (17.6 ± 1.2 versus 18.4 ± 1.2 N; p = 0.002). MNTs were higher in all subsequent measurements compared with the first (p < 0.001) and in the morning compared with the afternoon (p = 0.04). We found no evidence of MNT differences based on interval between consecutive measurements (1 versus 3 minutes). Variability was lower in the thoracic limbs [mean back-transformed log10 coefficient of variation (CV) ± SE = 25.5 ± 1.5% versus 30.6 ± 1.5% in the pelvic limbs; p < 0.001], with the actuator (22.7 ± 1.5% versus 33.4 ± 1.5% with the probe; p < 0.001), and on the left (CV = 26.9 ± 1.5% versus 29.3 ± 1.5% on the right; p = 0.01). Tail data (probe only) were analysed separately: mean MNT (± SE) was 11.7 (±1.8); MNT increased in days 3-6 of testing compared with day 1 (p < 0.001). 13. Selenium Digestibility and Bioactivity in Dogs: What the Can Can, the Kibble Can't.

There is a growing concern for the long-term health effects of selenium (Se) over- or underfeeding. The efficiency of utilization of dietary Se is subject to many factors. Our study in dogs evaluated the effect of diet type (canned versus kibble) and dietary protein concentration on Se digestibility and bioactivity. Canned and kibble diets are commonly used formats of dog food, widely ranging in protein concentration. Twenty-four Labrador retrievers were used and four canned and four kibble diets were selected with crude protein concentrations ranging from 10.1 to 27.5 g/MJ. Crude protein concentration had no influence on the digestibility of Se in either canned or kibble diets, but a lower Se digestibility was observed in canned compared to kibble diets. However, the biological activity of Se, as measured by whole blood glutathione peroxidase, was higher in dogs fed the canned diets than in dogs fed the kibble diets and decreased with increasing crude protein intake. These results indicate that selenium recommendations in dog foods need to take diet type into account.

The aim of the present study was to explore whether marginal dietary zinc (Zn) concentrations affect claw quality measurements in weaned pigs. Twenty-four weaned pigs were randomly assigned to two dietary treatment groups: (1) 42 mg Zn/kg diet from ingredients only (unsupplemented, marginal dietary Zn concentration below Zn requirements of 80 mg Zn/kg feed); and (2) 106 mg Zn/kg diet, where Zn was added as ZnO (common commercial dietary Zn concentration). Claw conformation characteristics were measured at the start (day 0, 4 weeks of age) and at the end (day 36) of the study, and the histological claw characteristics of horn wall and heel horn were examined on samples collected at 9 weeks of age. Non-supplemented pigs had narrower claw widths (P= 0.028) and lower toe heights (P= 0.010) at 9 weeks. The length of the dorsal border tended to be lower for the non-supplemented piglets (P= 0.092). Claw volume and claw horn size were lower (P= 0.003 and P < 0.001, respectively) for the non-supplemented pigs at 9 weeks of age. Horn growth and wear were lower for the non-supplemented pigs (P= 0.044 and P < 0.001, respectively), but net horn growth (horn growth minus wear) was not different (P= 0.406). No changes in the histological claw characteristics were observed. Differences in claw quality measurements were found between lateral and medial claw digits and between fore and hind claws. It was concluded that marginal dietary Zn concentration affected various claw quality measurements. Background: Inadequate dietary selenium (Se) intake in humans and animals can lead to long term health problems, such as cancer. In view of the owner's desire for healthy longevity of companion animals, the impact of dietary Se provision on long term health effects warrants investigation. Little is currently known regards biomarkers, and rate of change of such biomarkers in relation to dietary selenium intake in dogs. In this study, selected biomarkers were assessed for their suitability to detect changes in dietary Se in adult dogs within eight weeks., Results: Twenty-four dogs were fed a semi-purified diet with an adequate amount of Se (46.1 μg/MJ) over an 8 week period. They were then divided into two groups. The first group remained on the adequate Se diet, the second were offered a semi-purified diet with a low Se concentration (6.5 μg/MJ; 31% of the FEDIAF minimum) for 8 weeks. Weekly urine and blood was collected and hair growth measurements were performed. The urinary Se to creatinine ratio and serum Se concentration were significantly lower in dogs consuming the low Se diet from week 1 onwards, by 84% (adequate 25.3, low 4.1) and 7% (adequate 257 μg/L, low 238 μg/L) respectively. Serum and whole blood glutathione peroxidase were also significantly lower in dogs consuming the low Se diet from weeks 6 and 8 respectively. None of the other biomarkers (mRNA expression and serum copper, creatine kinase, triiodothyronine:thyroxine ratio and hair growth) responded significantly to the low Se diet over the 8 week period., Conclusions: This study demonstrated that urinary Se to creatinine ratio, serum Se and serum and whole blood glutathione peroxidase can be used as biomarkers of selenium status in dogs. Urinary Se to creatinine ratio and serum Se concentrations responded faster to decreased dietary Se than the other parameters. This makes these biomarkers candidates for early screening of long term effects of dietary Se provision on canine health.

It has been suggested that goats (typical browser) are better adapted to digest tannin-rich diets than sheep (typical grazer). To evaluate this, Bonga sheep and Kaffa goats were used in a 2 × 3 randomized crossover design with two species, three diets, and three periods (15-day adaptation + 7-day collection). The dietary treatments consisted of grass-based hay only (tannin-free diet = FT), a high-tannin diet (36% Albizia schimperiana (AS) + 9% Ficus elastica (FE) + 55% FT (HT)), and HT + polyethylene glycol 6000 (PEG). Animals were individually fed at 50 g dry matter (DM)/kg body weight (BW) and had free access to clean drinking water and mineralized salt licks. Nutrient intake, apparent nutrient digestibility, nutrient conversion ratios, and live weight changes were determined. Condensed tannin concentrations in AS and FE were 110 and 191 g/kg DM, respectively. Both sheep and goats ate 47% more of HT than FT, and dry matter intake further increased by 9% when PEG was added, with clear difference in effect size between goats and sheep (P < 0.001). The effects of the tannin-rich diet and PEG addition were similarly positive for DM digestibility between sheep and goats, but crude protein (CP) digestibility was higher in HT + PEG-fed goats than in sheep fed the same diet. However, PEG addition induced a larger improvement in growth performance and feed efficiency ratio in sheep than in goat (P < 0.001). The addition of PEG as a tannin binder improved digestion and performance in both species, but with the highest effect size in sheep.

Deoxynivalenol (DON) and fumonisins (FBs) are secondary metabolites produced by Fusarium fungi that frequently contaminate broiler feed. The aim of this study was to investigate the impact of DON and/or FBs on the intestinal barrier in broiler chickens, more specifically on the mucus layer and antioxidative response to oxidative stress. One-day-old broiler chicks were divided into four groups, each consisting of eight pens of seven birds each, and were fed for 15 days either a control diet, a DON-contaminated diet (4.6 mg DON/kg feed), a FBs-contaminated diet (25.4 mg FB1 + FB2/kg feed), or a DON+FBs-contaminated diet (4.3 mg DON and 22.9 mg FB1 + FB2/kg feed). DON and FBs affected the duodenal mucus layer by suppressing intestinal mucin (MUC) 2 gene expression and altering the mucin monosaccharide composition. Both mycotoxins decreased gene expression of the intestinal zinc transporter (ZnT)-1 and regulated intracellular methionine homeostasis, which are both important for preserving the cell's critical antioxidant activity. Feeding a DON- and/or FBs-contaminated diet, at concentrations close to the European Union maximum guidance levels (5 mg DON and 20 mg FB1 + FB2/kg feed) changes the intestinal mucus layer and several intestinal epithelial antioxidative mechanisms.

Microbiota in the gastro-intestinal tract are closely related to both the intestinal and overall health of the host. Experimental chickens have always been euthanized in order to identify and quantify the bacteria in cecal content. In this study, quantification and identification of the microbial populations in cecal drop, cecal content and fecal drop samples from chickens showed that cecal drop contains a bacterial community that is very similar (concerning bacterial diversity, richness and species composition) to cecal content, as opposed to the bacterial community found in fecal drop. Cecal drop analysis thus allows for longitudinal experiments on chickens' cecal bacteria. 19. Effect of locomotion score on sows' performances in a feed reward collection test.

Sows housed in groups have to move through their pen to fulfil their behavioural and physiological needs such as feeding and resting. In addition to causing pain and discomfort, lameness may restrict the ability of sows to fulfil such needs. The aim of our study was to investigate the extent to which the mobility of sows is affected by different degrees of lameness. Mobility was measured as the sow's willingness or capability to cover distances. Feed-restricted hybrid sows with different gait scores were subjected to a feed reward collection test in which they had to walk distances to obtain subsequent rewards. In all, 29 group-housed sows at similar gestation stage (day 96.6 ± 7 s.d.) were visually recorded for gait and classified as non-lame, mildly lame, moderately lame or severely lame. All sows received 2.6 kg of standard commercial gestation feed per day. The test arena consisted of two feeding locations separated from each other by a Y-shaped middle barrier. Feed rewards were presented at the two feeders in turn, using both light and sound cues to signal the availability of a new feed reward. Sows were individually trained during 5 non-consecutive days for 10 min/day with increasing barrier length (range: 0 to 3.5 m) each day. After training, sows were individually tested once per day on 3 non-consecutive days with the maximum barrier length such that they had to cover 9.3 m to walk from one feeder to the other. The outcome variable was the number of rewards collected in a 15-min time span. Non-lame and mildly lame sows obtained more rewards than moderately lame and severely lame sows (P<0.01). However, no significant difference was found between non-lame and mildly lame sows (P=0.69), nor between moderately lame and severely lame sows (P=1.00). This feed reward collection test indicates that both moderately lame and severely lame sows are limited in their combined ability and willingness to walk, but did not reveal an effect of mild lameness on mobility. These findings suggest that moderately and more severely lame sows, but not mildly lame sows, might suffer from reduced access to valuable resources in group housing systems.

Due to the high prevalence of obesity in some horses and ponies (especially in the leisure horse sector), effective and safe weight loss strategies are required. The present study evaluated the effect of two different energy restriction rates on physical, morphometric and welfare parameters in 18 obese (body condition score [BCS] 7-9/9) Shetland geldings. The trial was divided into three periods: (1) a 4 week adaptation period, during which the maintenance energy intakes to maintain a stable obese bodyweight were determined (100% MERob); (2) a 16.5-week weight loss period during which the ponies were randomly divided into three groups (n = 6/group) comprising a control group (CONTROL), moderate energy restricted (MOD), and severe energy restricted (SEV) groups that were respectively fed at 100%, 80% and 60% of their individual MERob; and (3) a 3 week follow up period in which the ponies were again fed at their outset individual 100% MERob. Between the start and end of the weight loss period, significant pairwise differences between the three treatment groups were seen for bodyweight, BCS, heart girth, belly girth, and relative ultrasound fat depth at the level of loin and ribs at several time points (P < 0.05). The higher energy restriction was associated with a faster decrease in BCS, tail head, and heart plus belly girth, but no gastric ulcers or stereotypic behaviours were seen., (Copyright © 2015 Elsevier Ltd. Fluctuations in Zn metabolism throughout gestation and lactation might affect Zn requirements. However, scientific data on Zn requirements for breeding sows are limited. The objective of the present study was to assess the Zn status of primiparous and multiparous sows using different Zn status biomarkers, to identify periods of critical Zn status throughout the reproductive cycle at different parities. Blood samples were taken after overnight fasting before feeding in the morning from five primiparous and ten multiparous sows at fixed time intervals during gestation (days - 5, 0 (insemination), 21, 42, 63 and 84), around parturition (days 108, 112, 115 (parturition) and 118) and during lactation (days 122, 129 and 143 (weaning)). At parturition, blood samples were collected from two randomly selected piglets per sow before colostrum intake. Plasma was analysed for Zn and Cu contents, whereas serum was analysed for alkaline phosphatase, metallothionein and albumin concentrations. Independently of parity, all biomarkers fluctuated differently during gestation and lactation (P< 0·050). This reflects their different roles in Zn metabolism, and suggests that the choice of a Zn status biomarker necessitates careful consideration. Low average plasma Zn concentration at the end of gestation and throughout lactation seem to be replenished towards weaning.

Se bioavailability in commercial pet foods has been shown to be highly variable. The aim of the present study was to identify dietary factors associated with in vitro accessibility of Se (Se Aiv) in pet foods. Se Aiv is defined as the percentage of Se from the diet that is potentially available for absorption after in vitro digestion. Sixty-two diets (dog, n 52; cat, n 10) were in vitro enzymatically digested: fifty-four of them were commercially available (kibble, n 20; pellet, n 8; canned, n 17; raw meat, n 6; steamed meat, n 3) and eight were unprocessed (kibble, n 4; canned, n 4) from the same batch as the corresponding processed diets. The present investigation examined if Se Aiv was affected by diet type, dietary protein, methionine, cysteine, lysine and Se content, DM, organic matter and crude protein (CP) digestibility. Se Aiv differed significantly among diet types (P< 0·001). Canned and steamed meat diets had a lower Se Aiv than pelleted and raw meat diets. Se Aiv correlated positively with CP digestibility in extruded diets (kibbles, n 19; r 0·540, P =0·017) and negatively in canned diets (n 16; r - 0·611, P =0·012). Moreover, the canning process (n 4) decreased Se Aiv (P =0·001), whereas extrusion (n 4) revealed no effect on Se Aiv (P =0·297). These differences in Se Aiv between diet types warrant quantification of diet type effects on in vivo Se bioavailability.

Global competition for high standard feed-food resources between man and livestock, such as industrial broilers, is a concerning problem. In addition, the low productivity of scavenger chickens in developing countries leaves much to be desired. Changing the ingredients, and therefore, the nutrient composition of feed intake by commercial fed as well as scavenger chickens seems like an obvious solution. In this study, the ability of four broiler chicken breeds to perform on a commercial versus a scavenger diet was tested. The four broiler breeds differed genetically in growth potential. A significant (P < 0.01) negative effect of the scavenger diet on the bodyweight of the fast growing breeds was found and this effect decreased with decreasing growth rate in the other breeds. These differences in bodyweight gain could not be explained by differences in nutrient digestibility but were caused by the lack of ability of the fast growing breeds to increase their feed intake sufficiently.

Small-scale urban dairy farms (n = 16) in and around Jimma, Ethiopia with cross-bred (Bos indicus × Bos taurus) cows were enrolled in a double-blinded intervention study to investigate the effect of a trace element supplementation programme on trace element status and milk concentrations as well as performance [body condition score (BCS), milk yield, leptin], milk composition, antioxidant status (ferric-reducing ability of plasma (FRAP), thiobarbituric acid-reactive substances (TBARS)], blood biochemistry, serum proteins and immune response (antibody titre upon rabies vaccination). The farms were allocated to a (1) placebo or (2) Cu, Zn, Se, Co and I supplementation treatment for 150 d. On days 0 and 120, four lactating cows per farm were sampled for milk and plasma, and on day 150 for serum, following primo-vaccination. Cu deficiency was present in 17% and marginal Se deficiency in 30% of initially sampled cows, while no Zn shortage was detected. Over 120 days, trace element supplementation caused a bigger increase in plasma Se and Cu concentrations, but also a larger decrease of plasma Fe concentrations. A larger increase in milk Se concentrations was observed in the supplemented group, whereas none of the other elements were affected. BCS decreased more over time in the supplemented group. None of the other parameters of performance and antioxidant status nor milk composition or blood biochemistry was affected by treatment. Antibody response to rabies vaccination did not differ between groups, whereas α1-globulins tended to be lower and β-globulins tended to be higher in the supplemented group. Understanding the symbiotic relationship between gut microbes and their animal host requires characterization of the core microbiota across populations and in time. Especially in captive populations of endangered wildlife species such as the cheetah (Acinonyx jubatus), this knowledge is a key element to enhance feeding strategies and reduce gastrointestinal disorders. In order to investigate the temporal stability of the intestinal microbiota in cheetahs under human care, we conducted a longitudinal study over a 3-year period with bimonthly faecal sampling of 5 cheetahs housed in two European zoos. For this purpose, an integrated 16S rRNA DGGE-clone library approach was used in combination with a series of real-time PCR assays. Our findings disclosed a stable faecal microbiota, beyond intestinal community variations that were detected between zoo sample sets or between animals. The core of this microbiota was dominated by members of Clostridium clusters I, XI and XIVa, with mean concentrations ranging from 7.5-9.2 log10 CFU/g faeces and with significant positive correlations between these clusters (P<0.05), and by Lactobacillaceae. Moving window analysis of DGGE profiles revealed 23.3-25.6% change between consecutive samples for four of the cheetahs. The fifth animal in the study suffered from intermediate episodes of vomiting and diarrhea during the monitoring period and exhibited remarkably more change (39.4%). This observation may reflect the temporary impact of perturbations such as the animal's compromised health, antibiotic administration or a combination thereof, which temporarily altered the relative proportions of Clostridium clusters I and XIVa. In conclusion, this first long-term monitoring study of the faecal microbiota in feline strict carnivores not only reveals a remarkable compositional stability of this ecosystem, but also shows a qualitative and quantitative similarity in a defined set of faecal bacterial lineages across the five animals under study that may typify the core phylogenetic microbiome of cheetahs.

26. Dietary factors associated with faecal consistency and other indicators of gastrointestinal health in the captive cheetah (Acinonyx jubatus).

Gastrointestinal diseases pose significant risks to captive cheetah survival and welfare. Multiple factors are thought to be associated with these diseases, but to date a comprehensive epidemiological survey of disease risk factors has not been conducted. A survey of diet and health parameters was completed for 184 captive cheetahs in 86 international facilities. Comparisons were made among dietary factors with respect to disease status and observed faecal consistency, incidence of vomiting and diarrhoea in the past 4 weeks. Extremely dry faeces were most common in cheetahs fed carcasses, but was still of low incidence (15%). Contrastingly, cheetahs fed commercially prepared diets had the highest prevalence of liquid faeces "always" or "often" (9%). Cheetahs fed raw meat diets had the highest prevalence of soft faeces with no shape (22%), as well as of firm and dry faeces (40%). No broad category of diet exerted any influence on the health parameters investigated. However, feeding of ribs at least once per week reduced the odds of diarrhoea (P = 0.020) and feeding of long bones (limbs) at least once per week was associated with a lower odds of vomiting (P = 0.008). Cheetahs fed muscle meat at least once per week had reduced odds of suffering from chronic gastritis (P = 0.005) or non-specific gastrointestinal disease (P < 0.001). The only factor identified as increasing the odds of chronic gastritis was feeding of horse "often" or "always" (P = 0.023). The findings of the current study build on existing empirical research to support a recommendation towards a greater inclusion of skeletal components. Current husbandry guidelines advocating the use of supplemented raw meat diets are likewise supported, but the use of horse meat, as well as commercially prepared diets for captive cheetahs, warrants caution until further research is conducted.

This study was executed to investigate the effect of dietary β-alanine (BA) on amino acid (AA) metabolism and voluntary feed intake in carp (Cyprinus carpio) at mildly elevated temperature to exert AA catabolism. Twenty-four fish in 12 aquaria were randomly assigned to either a control diet or the same diet with 500 mg BA/kg. A 14-day period at an ideal temperature (23 °C) was followed by 15 days at chronic mildly elevated temperature (27 °C). After the 15 days, all fish were euthanised for muscle analysis on histidine-containing dipeptides (HCD), whole blood on free AA and carnitine esters. The carnosine and anserine analysis indicated that all analyses were below the detection limit of 5 µmol/L, confirming that carp belongs to a species that does not store HCD. The increases in free AA concentrations due to BA supplementation failed to reach the level of significance. The effects of dietary BA on selected whole blood carnitine esters and their ratios were also not significant. The supplementation of BA tended to increase body weight gain (P = 0.081) and feed intake (P = 0.092). The lack of differences in the selected nutrient metabolites in combination with tendencies of improved growth performance warrants further investigation to unravel the mechanism of BA affecting feed intake. This first trial on the effect of BA supplementation on AA catabolism showed that its metabolic effect in carp at chronic mildly elevated temperature was very limited. Further studies need to evaluate which conditions are able to exert an effect of BA on AA metabolism.

Domestic cats are obligate carnivores and in this light hindgut fermentation has been considered unimportant in this species. However, a diverse microbiota has been found in the small and large intestines of domestic cats. Additionally, in vitro and in vivo studies support the hypothesis that microbial fermentation is significant in felines with potential benefits to the host. Results on microbiota composition and microbial counts in different regions of the feline gastrointestinal tract are compiled, including a description of modulating host and technical factors. Additionally, the effects of dietary fibre supplementation on the microbiota composition are described. In a second section, in vitro studies, using inocula from fresh feline faeces and focusing on the fermentation characteristics of diverse plant substrates, are described. In vivo studies have investigated the effects of dietary fibre on a broad range of physiological outcomes. Results of this research, together with studies on effects of plant fibre on colonic morphology and function, protein and carbohydrate metabolism, and the effects of plant fibre on disease conditions that require a decrease in dietary protein intake, are shown in a third section of the present review. Conclusively, for fructans and beet pulp, for example, diverse beneficial effects have been demonstrated in the domestic cat. Both dietary fibre sources are regularly used in the pet food industry. More research is warranted to reveal the potential benefits of other fibre sources that can be used on a large scale in feline diets for healthy and diseased cats.

- Abstract
The present study evaluated the effect of different levels of energy restriction on metabolic parameters in obese ponies. Relative weight changes, markers of lipid metabolism and oxidant/antioxidant balance were monitored. A total of eighteen obese (body condition score ≥ 7/9) Shetland ponies were studied over a 23·5-week trial, which was divided into three periods. The first period involved a 4-week adaptation period in which each animal was fed 100% of their maintenance energy requirements needed to maintain a stable obese body weight (MERob). This was followed by a 16·5-week weight-loss period in which ponies were assigned to receive either 100% (control group, CONTROL), 80% (slow weight-loss (SLOW) group) or 60% (rapid weight-loss (RAPID) group) of their MERob. During the 3-week end-phase period, all ponies were again fed 100% of their MERob. Relative weight loss was higher in the RAPID group (P< 0·001) compared with the SLOW group. No linear relationship was found as a doubling of the percentage of energy restriction was accompanied by a tripling of the percentage of weight loss. Relative weight gain afterwards in the end-phase period was higher in the RAPID group (P< 0·001) compared with the SLOW and CONTROL groups. During the weight-loss period, TAG and NEFA concentrations were highest in the RAPID group, as were α-tocopherol and ferric-reducing ability of plasma concentrations. After 8 weeks of weight loss, the concentrations of advanced oxidation protein products were higher in the RAPID group compared with the SLOW and CONTROL groups (P< 0·001). In conclusion, the level of energy restriction influences the extent of changes in oxidant/antioxidant balance. Practically, more severe energy restriction regimens may be associated with a greater regain of weight after the restriction period.

Twenty-four intact male Bonga lambs were studied in a 100-day experiment to evaluate the effect of feeding tannin rich leaves of Albizia gummifera (AG) on nutrient utilization, growth performance, and carcass composition. The dietary treatments consisted of hay alone (T1, control), AG at 30 % of control diet + T1 (T2), and T2 + polyethylene glycol 6000 (PEG) (T3, 40 mg PEG, 1 kg AG/head on a dry mass (DM) basis. The lambs were individually fed at 50 g DM/kg live weight. In the last 10 days of the experiment, all animals from each treatment were harnessed with feces collection bags. At the end of the experiment, lambs were slaughtered after overnight fasting for measurements of carcass characteristics. Crude protein, neutral detergent fiber, and condensed tannin contents of AG were 300, 586, and 108 g/kg DM, respectively. Lambs fed AG with PEG had higher (P < 0.001) feed intake, digestibility, carcass weight, and weight gain compared with other treatments. Although lambs fed diet T2 were able to utilize AG, the efficiency of its utilization was lower (P < 0.001) than the PEG-treated groups. The improvement in nutrient intake and digestibility, carcass weight, and growth performance of lambs supplemented with PEG emphasizes the negative effect of tannins on livestock performance. Leaves of AG enhance digestibility and performance in tropical hay-fed sheep, and the addition of PEG further improves this effect, likely due to the binding of tannins.

Both mycotoxin contamination of feed and Clostridium perfringens-induced necrotic enteritis have an increasing global economic impact on poultry production. Especially the Fusarium mycotoxin deoxynivalenol (DON) is a common feed contaminant. This study aimed at examining the predisposing effect of DON on the development of necrotic enteritis in broiler chickens. An experimental Clostridium perfringens infection study revealed that DON, at a contamination level of 3,000 to 4,000 µg/kg feed, increased the percentage of birds with subclinical necrotic enteritis from 20±2.6% to 47±3.0% (P<0.001). DON significantly reduced the transepithelial electrical resistance in duodenal segments (P<0.001) and decreased duodenal villus height (P = 0.014) indicating intestinal barrier disruption and intestinal epithelial damage, respectively. This may lead to an increased permeability of the intestinal epithelium and decreased absorption of dietary proteins. Protein analysis of duodenal content indeed showed that DON contamination resulted in a significant increase in total protein concentration (P = 0.023). Furthermore, DON had no effect on in vitro growth, alpha toxin production and netB toxin transcription of Clostridium perfringens. In conclusion, feed contamination with DON at concentrations below the European maximum guidance level of 5,000 µg/kg feed, is a predisposing factor for the development of necrotic enteritis in broilers. These results are associated with a negative effect of DON on the intestinal barrier function and increased intestinal protein availability, which may stimulate growth and toxin production of Clostridium perfringens.

Renal and gastrointestinal pathologies are widespread in the captive cheetah (Acinonyx jubatus) population but are often diagnosed at a late stage, because diagnostic tools are limited to the evaluation of clinical signs or general blood examination. Presently, no data are available on serum proteins and acute-phase proteins in cheetahs during health or disease, although they might be important to improve health monitoring. This study aimed to quantify serum proteins by capillary electrophoresis in 80 serum samples from captive cheetahs, categorized according to health status and disease type. Moreover, serum amyloid A concentrations were measured via a turbidimetric immunoassay validated in domestic cats, whereas haptoglobin and C-reactive protein were determined by non-species-specific functional tests. Cheetahs classified as healthy had serum protein and acute phase protein concentrations within reference ranges for healthy domestic cats. In contrast, unhealthy cheetahs had higher (P < 0.001) serum amyloid A, alpha2-globulin, and haptoglobin concentrations compared with the healthy subgroup. Moreover, serum amyloid A (P = 0.020), alpha2-globulin (P < 0.001) and haptoglobin (P = 0.001) concentrations in cheetahs suffering from chronic kidney disease were significantly greater compared to the reportedly healthy cheetahs. Our study indicates that serum proteins in the cheetah can be analyzed by routine capillary electrophoresis, whereas acute-phase proteins can be measured using available immunoassays or non-species-specific techniques, which are also likely to be applicable in other exotic felids. Moreover, results suggest that serum amyloid A and haptoglobin are important acute-phase proteins in the diseased cheetah and highlight the need to evaluate their role as early-onset markers for disease.

- Abstract
In strict carnivorous domestic cats, a metabolic competition arises between the need to use amino acids for gluconeogenesis and for protein synthesis both in health and disease. The present study investigated the amino acid-sparing potential of propionic acid in cats using dietary propionylated starch (HAMSP) supplementation. A total of thirty cats were fed a homemade diet, supplemented with either HAMSP, acetylated starch (HAMSA) or celite (Control) for three adaptation weeks. Propionylated starch was hypothesised to provide propionic acid as an alternative gluconeogenic substrate to amino acids, whereas acetic acid from HAMSA would not provide any gluconeogenic benefit. Post-adaptation, a 5-d total faecal collection was carried out to calculate apparent protein digestibility coefficients. Fresh faecal and blood samples were collected to analyse fermentation endproducts and metabolites. The apparent protein digestibility coefficients did not differ between supplements (P = 0·372) and were not affected by the protein intake level (P = 0·808). Faecal propionic acid concentrations were higher in HAMSP than in HAMSA (P = 0·018) and Control (P = 0·003) groups, whereas concentrations of ammonia (P = 0·007) were higher in HAMSA than in HAMSP cats. Tendencies for or higher propionylcarnitine concentrations were observed in HAMSP compared with HAMSA (P = 0·090) and Control (P = 0·037) groups, and for tiglyl- + 3-methylcrotonylcarnitine concentrations in HAMSP as compared with Control (P = 0·028) cats. Methylmalonylcarnitine concentrations did not differ between groups (P = 0·740), but were negatively correlated with the protein intake level (r -0·459, P = 0·016). These results suggest that HAMSP cats showed more saccharolytic fermentation patterns than those supplemented with HAMSA, as well as signs of sparing of valine in cats with a sufficient protein intake.

Research showed a positive association between back fat (BF) change the week before farrowing and colostrum yield (CY). This study tested the causality of this association, hence to optimize CY by altering the sows' peripartal feeding strategy. Sows were randomly divided into 2 treatment groups at d 108 of gestation. The first group (L, n = 28) received 1.5 kg feed·d(-1), the second group (H, n = 22) received 3 times 1.5 kg feed·d(-1) until farrowing. Daily feed intake and CY were measured. Colostrum was analyzed for nutrient composition, AA and fatty acids, IgG and IgA. Sow serum was obtained at d 108 of gestation and d 1 of lactation after overnight fasting and analyzed for NEFA, (iso)butyrylcarnitine (C4), creatinine, urea, 3-OH-butyrylcarnitine (3-OH-C4), IgG, and IgA. Based on BF at d 108, sows were divided into body condition (BC) groups: skinny (<17 mm, n = 15), moderate (17 to 23 mm, n = 21), fat (>23 mm, n = 14). We performed ANOVA with treatment and BC as fixed factors and Scheffé post-hoc test. The week before farrowing, the L group had the lowest daily feed intake (DFI; 1.5 kg), and within the H group, fat sows (3.8 kg) had a lower DFI than skinny sows (4.3 kg; p = 0.006). The H group tended to have a greater total CY (P = 0.074) and had a greater CY/kg liveborn piglet (P = 0.018) than the L group. Compared with sows in moderate BC, fat sows had a lower total CY (P = 0.044) and a lower CY/kg liveborn piglet (P = 0.005). The H group had a greater concentration of lactose (p = 0.009) and n-3 PUFA (p < 0.001) but a lower concentration of protein (p = 0.040) in colostrum than the L group. The concentration of IgG and IgA did not differ between treatment and BC groups. Serum parameters at d 108 were similar between the treatment groups and BC groups. At d 1, the H group mobilized less body fat (NEFA: p = 0.002) and protein (creatinine: p < 0.001, C4: p = 0.016) reserves but had a greater ratio urea:NEFA (p < 0.001) and less ketone bodies (3-OH-C4: p < 0.001) compared with the L group. This indicates a more balanced entry of metabolites in the citric acid cycle and thus a better support of the maternal peripartal metabolism in the H group. Serum parameters did not differ between BC groups. Both CY and composition can be influenced by the peripartal feeding strategy and BC. The highest CY and most beneficial colostrum composition were obtained when sows entered the farrowing unit in a moderate BC and were provided a high peripartal feeding strategy.

Copper deficiency is a commonly diagnosed problem in cattle around the globe. In Jimma, Ethiopia, 8 zebu (Bos indicus) and 8 zebu ×: Holstein Friesian cross (Bos taurus ×: Bos indicus) heifers were used in an 11-wk study to investigate breed type differences and effects of Cu deficiency on concentrations of trace elements in plasma and edible tissues as well as mRNA expression of Cu-related genes. Heifers were fed a grass diet (6.4 ± 0.2 [SEM] mg Cu/kg DM) supplemented with 1 mg Mo/kg DM in wk 1 to 4 and 2 mg Mo/kg DM in wk 5 to 11, with blood samples collected every 2 wk and tissue collection postmortem. Plasma, liver, kidney, and semitendinosus and cardiac muscle were analyzed for Zn, Cu, Fe, Se, Mo, Co, and Mn. Expression of mRNA Cu-related genes was measured in aorta (lysyl oxidase [LOX]), liver (Cu transporting β-polypeptide [Atp7b], Cu chaperone for superoxide dismutase [CCS], cytochrome c oxidase assembly homolog 17 [Cox17], Cu transporter 1 homolog [Ctr1], and superoxide dismutase 1 [Sod1]), and duodenum (diamine oxidase [DAO] and metallo-thionein-1A [Mt1a]) as well as the Se-related glutathione peroxidase 1 (Gpx1). Zebu cattle maintained initial plasma Cu concentrations just below the threshold value for deficiency, whereas crossbred cattle gradually became severely Cu deficient over time (P < 0.001). In contrast, plasma Zn and Co were greater in zebu cattle at the onset of the trial but became similar to crossbred cattle towards the end of the trial (P < 0.001). Liver Cu (P = 0.002) and Fe (P ≤ 0.001), kidney Se (P < 0.001), and kidney and cardiac muscle Co (P ≤ 0.001) concentrations were greater in zebu than in crossbred cattle. Increased hepatic mRNA expression of the Cu regulatory genes Atp7b, Ctr1 (P = 0.02), CCS (P = 0.03), and Cox17 (P = 0.009) and Cu-related Sod1 (P = 0.001) as well as the Se-related Gpx1 (P ≤ 0.001) were greater in zebu than in crossbred cattle. However, duodenal mRNA expression of DAO (P = 0.8) and Mt1a (P = 0.2) and aortic expression of LOX (P = 0.8) were not different. Both the differences in Cu status indices (plasma and liver concentrations) and hepatic mRNA expression of Cu regulatory genes point to the possibility of a more efficient use of dietary Cu in B. indicus as compared to B. taurus ×: B. indicus cattle resulting in greater sensitivity to Cu deficiency in B. taurus crossbred cattle.

The effect of supplementing leaves of four tannin-rich plant species with polyethylene glycol 6000 (PEG) on nutrient intake and digestibility as well as on weight gain, feed conversion ratio (FCR) and N retention of zebu bulls (Bos indicus) was studied. Leaves of Albizia gummifera, Grewia ferruginea, Prunus africana and Syzygium guineense, containing, respectively, 85, 55, 76 and 172 g condensed tannins (CT) per kg dry matter (DM), were combined with natural pasture hay in a ratio of 40:60 on DM basis. The four diets were fed both without and with addition of PEG, at a dose of 40 g per kg DM, to eight zebu bulls during trials of 25 days in an 8 × 8 randomized crossover design. Supplementation with PEG increased nutrient intake, digestibility, FCR, N retention and average daily gain (p < 0.01). A diet × PEG interaction was observed for nutrient intake as well as for crude protein, neutral detergent fibre and acid detergent fibre digestibility (p < 0.05), but the effect size of PEG addition could not be attributed to the CT content as such, and also digestibility without PEG was not related to CT content of the diets. Background: Oxidative stress and inflammation can be altered by dietary factors in various species. However, little data are available in true carnivorous species such as domestic cats. As numerous anti-inflammatory and anti-oxidative additives become available and might be of use in cats with chronic low-grade inflammatory diseases, the current study aimed to develop a model of diet-induced inflammation by use of two opposite diets. It was hypothesized that a high fat diet enhanced in n-6 PUFA and with lower concentrations of antioxidants would evoke inflammation and oxidative stress in domestic cats., Results: Sixteen healthy adult cats were allocated to two groups. One group received a moderate fat diet, containing pork lard and salmon oil (AA:(EPA + DHA) ratio 0.19) (MFn-3), while the other group was fed a high fat diet, containing pork lard and chicken fat (AA:(EPA + DHA) ratio 2.06) (HFn-6) for 12 weeks. Prior to and 2, 4, 6, 8, 10 and 12 weeks after starting the testing period, blood samples were collected. Erythrocytic fatty acid profile showed clear alterations in accordance to the dietary fatty acid profile. Serum thiobarbituric acid reactive substances was higher when fed MFn-3 compared to the HFn-6, suggesting augmented oxidative stress. This was associated with a reduced serum vitamin E status, as serum α-tocopherol concentrations were lower with MFn-3, even with higher dietary levels of vitamin E. Serum cytokine and serum amyloid A concentrations were not influenced by diet., Conclusion: These results point towards a resistance of cats to develop dietary fat-induced inflammation, but also suggest a high susceptibility to oxidative stress when fed a fish oil-supplemented diet even with moderate fat level and additional vitamin E.

Different species of chironomids larvae (Diptera: Chironomidae) so-called bloodworms are widely distributed in the sediments of all types of freshwater habitats and considered as an important food source for amphibians. In our study, three species of Chironomidae (Baeotendipes noctivagus, Benthalia dissidens, and Chironomus riparius) were identified in 23 samples of larvae from Belgium, Poland, Russia, and Ukraine provided by a distributor in Belgium. We evaluated the suitability of these samples as amphibian food based on four different aspects: the likelihood of amphibian pathogens spreading, risk of heavy metal accumulation in amphibians, nutritive value, and risk of spreading of zoonotic bacteria (Salmonella, Campylobacter, and ESBL producing Enterobacteriaceae). We found neither zoonotic bacteria nor the amphibian pathogens Ranavirus and Batrachochytrium dendrobatidis in these samples. Our data showed that among the five heavy metals tested (Hg, Cu, Cd, Pb, and Zn), the excess level of Pb in two samples and low content of Zn in four samples implicated potential risk of Pb accumulation and Zn inadequacy. Proximate nutritional analysis revealed that, chironomidae larvae are consistently high in protein but more variable in lipid content. Accordingly, variations in the lipid: protein ratio can affect the amount and pathway of energy supply to the amphibians. Our study indicated although environmentally-collected chironomids larvae may not be vectors of specific pathogens, they can be associated with nutritional imbalances and may also result in Pb bioaccumulation and Zn inadequacy in amphibians. Farrowing induction in sows is frequently applied and different protocols were described. Most studies induce farrowing between day 111 and 113 of gestation, but hardly any later during gestation. The aim of this study was to investigate the effects of different induction protocols when applied on d114 of gestation. We randomly assigned 118 sows to four treatment groups: single injection of prostaglandin (1×PG), prostaglandin combined with oxytocin 24 hours later (PG+OT), half the dosage of prostaglandin twice with six hours interval (2×1/2PG), and a control group of which farrowing wasn't induced. All injections were administered intramuscularly in the neck on day 114 of gestation. A significantly higher percentage of sows started farrowing between 22 and 32 hours after induction in the PG+OT-group (68 per cent) and the 2×1/2PG-group (52 per cent) compared to the control group (23 per cent). The 1×PG-group (46 per cent) tended to differ from the control group (P=0.087). More sows from the PG+OT-group were constipated at farrowing (P=0.042). Farrowing duration, birth interval, per cent stillborn piglets, dystocia and other variables were similar between the groups. In the present herd and with continuous farrowing supervision, applying PG+OT or 2×1/2PG on d114 of gestation was efficient to induce onset of farrowing in most of the sows 22-32 hours after induction without considerable negative effects.

Most gait-scoring scales for pigs have a limited number of categories, supposedly to improve repeatability. However, reducing the number of categories could lead to loss of information if the observers' discriminative capacities are underused. With a recently estimated within-herd prevalence of sow lameness of 8.8% to 16.9% in the European Union and the associated losses, the availability of reliable tools for the timely detection of initial cases warrants attention. This study investigated the intra- and inter-observer repeatability (intra-OR and inter-OR) of three gait-scoring scales for sows: a continuous 'tagged' visual analogue scale (tVAS, measured in mm), a 5-point and a 2-point ordinal scale (5P and 2P), all with the same descriptors. Veterinary medicine students (n=108) were trained to use the scales and then asked to score 90 videos (30 per scale) of sows with normal and abnormal gait. Thirty-six videos were shown once and 18 were randomly shown three times, of which one mirrored horizontally. The students' opinions on the scales were also collected. Intra- and inter-OR were higher with the tVAS than the 2P scale (inter-OR: 0.73 v. 0.60; P<0.05. Intra-OR: 0.80 v. 0.67; P<0.05). Intra-OR was higher with the 5P (0.81) than the 2P scale (0.67; P<0.05). For all three scales, repeatabilities were lower (P<0.05) for non-lame sows (gait score of ⩽45 mm on the tVAS) than for sows showing some signs of lameness (gait score>45 mm). Video order (first 45 v. last 45 clips), mirroring, users' opinions on the scales, and previous declared experience in handling pigs or scoring lameness in other species had no effect on repeatabilities. Correlations between the students' and experts' scores were high (tVAS=0.92; 5P=0.91; 2P=0.88) but the association for the 2P was not linear and the frequency distribution showed lower correlations for a group of students. This study confirms recent evidence that it is possible to design high-resolution gait-scoring scales that do not reduce observer repeatability. Visual gait-scoring scales with fewer than five categories are likely to entail loss of information on lameness in individual sows.

To overcome negative energy balance during the peripartal period of sows, an ad libitum feeding strategy (ADLIB) as alternative for commonly used restricted feeding (STANDARD, on average 3kg feed/day) was evaluated. Plasma metabolites and thyroid hormones, change of back fat thickness (BF), reproductive traits, and piglet performance were monitored. Voluntary feed intake of ADLIB sows declined at farrowing but was still more than twice the amount of what was offered to STANDARD sows. Consequently, ADLIB sows lost less BF than STANDARD sows (P=0.041). Additionally, BF change was affected by body condition. LEAN sows (BF<18mm on d 105 of gestation) lost less BF than MODERATE sows (18mm≤BF≤22mm) which lost less BF than FAT sows (BF>22mm) (P<0.001). Except for a decreased percentage of stillborn piglets for MODERATE sows (P=0.044), reproduction results were not affected. Piglet weaning weight of ADLIB-FAT and STANDARD-MODERATE sows was reduced in comparison with that of ADLIB-LEAN sows (P=0.005). Regardless of body condition, all metabolites and thyroid hormones measured showed a time dependent profile (P<0.001). On d 112 of gestation increased concentrations of creatinine (P=0.004), non-esterified fatty acids (P=0.039), and serum crosslaps (P=0.016) for STANDARD sows were observed. Triglycerides were increased for FAT sows (P<0.001), and decreased faster over time for ADLIB (P=0.013) and for FAT (P=0.012). Although ad libitum feeding during the peripartal period only resulted in less mobilization of muscle, fat, and bone reserves on d 112 of gestation, results of BF change and piglet weaning weight indicated that ad libitum feeding is beneficial for sow performance provided that BF is below 22mm., (Copyright © 2014 Elsevier B.V. Background: Imbalanced feeding regimes may initiate gastrointestinal and metabolic diseases in endangered felids kept in captivity such as cheetahs. Given the crucial role of the host's intestinal microbiota in feed fermentation and health maintenance, a better understanding of the cheetah's intestinal ecosystem is essential for improvement of current feeding strategies. We determined the phylogenetic diversity of the faecal microbiota of the only two cheetahs housed in an EAZA associated zoo in Flanders, Belgium, to gain first insights in the relative distribution, identity and potential role of the major community members., Results: Taxonomic analysis of 16S rRNA gene clone libraries (702 clones) revealed a microbiota dominated by Firmicutes (94.7%), followed by a minority of Actinobacteria (4.3%), Proteobacteria (0.4%) and Fusobacteria (0.6%). In the Firmicutes, the majority of the phylotypes within the Clostridiales were assigned to Clostridium clusters XIVa (43%), XI (38%) and I (13%). Members of the Bacteroidetes phylum and Bifidobacteriaceae, two groups that can positively contribute in maintaining intestinal homeostasis, were absent in the clone libraries and detected in only marginal to low levels in real-time PCR analyses., Conclusions: This marked underrepresentation is in contrast to data previously reported in domestic cats where Bacteroidetes and Bifidobacteriaceae are common residents of the faecal microbiota. Next to methodological differences, these findings may also reflect the apparent differences in dietary habits of both felid species. Thus, our results question the role of the domestic cat as the best available model for nutritional intervention studies in endangered exotic felids.

The amount of trace elements present in edible bovine tissues is of importance for both animal health and human nutrition. This study presents data on trace element concentrations in semitendinosus and cardiac muscles, livers and kidneys of 60 zebu (Bos indicus) bulls, sampled at Jimma, Ethiopia. From 28 of these bulls, blood samples were also obtained. Deficient levels of copper were found in plasma, livers, kidneys and semitendinosus muscles. Suboptimal selenium concentrations were found in plasma and semitendinosus muscles. Semitendinosus muscles contained high iron concentrations. Trace elements were mainly stored in the liver, except for iron and selenium. Cardiac muscles generally contained higher concentrations of trace elements than semitendinous muscles except for zinc. A strong association was found between liver and kidney concentrations of copper, iron, cobalt and molybdenum. Liver storage was well correlated with storage in semitendinosus muscle for selenium and with cardiac muscle for cobalt and selenium. Plasma concentrations of copper, selenium, cobalt were well related with their respective liver concentrations and for cobalt and selenium, also with cardiac muscle concentrations. The data suggest multiple trace element deficiencies in zebu cattle in South-West Ethiopia, with lowered tissue concentrations as a consequence. Based on the comparison of our data with other literature, trace element concentrations in selected edible tissues of Bos indicus seem quite similar to those in Bos taurus. However, tissue threshold values for deficiency in Bos taurus cattle need to be refined and their applicability for Bos indicus cattle needs to be evaluated.

Background: Insulin resistance in horses is an emerging field of interest as it is thought to be a contributing factor in the pathogenesis of many equine conditions., Objectives: The objectives of the present study were to determine the effects of long-term oral administration of L-carnitine on insulin sensitivity, glucose disposal, plasma leptin concentrations and acylcarnitine spectrum both in plasma and urine., Animals and Methods: Six 3-year-old healthy warmblood geldings were used. In a double blind 2 × 2 Latin square design at a dosage of 100 mg/kg body weight (BW)/day for 28 days the effects of oral supplementation of L-carnitine (as fumarate) were assessed. Glucose disposal and insulin sensitivity were measured by means of the euglycemic-hyperinsulinemic clamp technique. Radioimmunoassays were used to determine plasma leptin and insulin concentrations. Electrospray tandem mass spectrometry was used to assess acylcarnitines both in plasma and urine. Statistical analysis was performed using a linear mixed-effects model and P values <0.05 were considered significant., Results: Long-term L-carnitine administration did not affect insulin sensitivity. Plasma leptin and free carnitine concentrations in plasma and urine increased significantly (P = 0.047 and 0.000, respectively) following L-carnitine administration as well as short-chain acylcarnitines in plasma and urinary excretion of short- and medium-chain acylcarnitines., Conclusion and Clinical Relevance: Given the effects of oral administration of L-carnitine further clinical study is necessary in order to assess the potential beneficial effects in equine patients suffering from metabolic myopathies such as acquired multiple acyl-CoA dehydrogenase deficiency., Impact for Human Medicine: The current study supports the treatment rationale of short-chain acyl-CoA dehydrogenase deficiency in humans with L-carnitine at an oral dosage of 100 mg/kg BW/day.

The present study assessed the effect of silo emptying and feed transport by conveyor systems on particle size and nutrient content of the feed delivered to the pigs. Experiment 1 sampled feed from four feeders along the conveyor system of two barns. Samples were taken immediately after filling the feed silo (Begin) and when the silo was almost empty (End). In Experiment 2, three barns with drag-type conveyors, three with auger-type conveyors and two with spiral-type conveyors were sampled. Along the different conveyors, samples at 10, 20, 50 and 85 m distance from the feed silo were taken from the feeders. In each barn, sampling was repeated for two subsequent batches of feed delivered. In all samples, particle size profile was determined and nutrient content was analysed. In Experiment 2, mineral content was also determined. In Experiment 1, the size of the different particle fractions decreased from Begin to End. An interaction (p < 0.05) between sampling time and conveyor type was detected for the 10% smallest particles. In Experiment 2, an effect of sampling time on the 10% largest particles was detected (p < 0.05). The effect of sampling time on several nutrients was observed in Experiments 1 and 2, but the affected nutrients differed between both experiments. Results implied that it was mainly emptying of the silo that affected mash particle size profile and nutrient content. The potential impact of these changes on pig performance requires further investigation.

Background: In humans, adipose tissue (AT) originating from different depots shows varying gene expression profiles. In horses, the risk of certain metabolic disorders may also be influenced by the impact of specific AT depots. Macrophage infiltration in human and rat AT is considered to be a source of inflammatory changes. In horses, this relationship has not been extensively studied yet. Reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR), a useful method to evaluate differences in mRNA expression across different tissues, can be used to evaluate differences between equine AT depots. For a correct interpretation of the RT-qPCR results, expression data have to be normalized by the use of validated reference genes. The main objectives of this study were to compare mRNA expression of inflammation-related genes, as well as adipocyte morphology and number between different equine AT depots; and in addition, to investigate the presence of antigen presenting cells in equine AT and any potential relationship with adipokine mRNA expression., Results: In this study, the mRNA expression of inflammation-related genes (leptin, chemokine ligand 5, interleukin 1β, interleukin 6, interleukin 10, adiponectin, matrix metalloproteinase 2, and superoxide dismutase 2) and candidate reference gene stability was investigated in 8 different AT depots collected from the nuchal, abdominal (mesenteric, retroperitoneal, and peri-renal) and subcutaneous (tail head and loin) AT region. By using GeNorm analysis, HPRT1, RPL32, and GAPDH were found to be the most stable genes in equine AT. The mRNA expression of leptin, chemokine ligand 5, interleukin 10, interleukin 1β, adiponectin, and matrix metalloproteinase 2 significantly differed across AT depots (P < 0.05). No significant AT depot effect was found for interleukin 6 and superoxide dismutase 2 (P > 0.05). Adipocyte area and number of antigen presenting cells per adipocyte significantly differed between AT depots (P < 0.05)., Conclusions: Adipose tissue location was associated with differences in mRNA expression of inflammation-related genes. This depot-specific difference in mRNA expression suggests that the overall inflammatory status of horses could be partially determined by the relative proportion of the different AT depots.

Directing protein and energy sources towards lactation is crucial to optimise milk production in sows but how this influences colostrum yield (CY) remains unknown. The aim of this study was to identify associations between CY and the sow's use of nutrient resources. We included 37 sows in the study that were all housed, fed and managed similarly. Parity, back fat change (ΔBF), CY and performance parameters were measured. We obtained sow serum samples 3 to 4 days before farrowing and at D1 of lactation following overnight fasting. These were analysed for non-esterified fatty acids (NEFA), urea, creatinine, (iso)butyrylcarnitine (C4) and immunoglobulins G (IgG) and A (IgA). The colostrum samples collected 3, 6 and 24 h after the birth of the first piglet were analysed for their nutrient and immunoglobulins content. The technical parameters associated with CY were parity group (a; parities 1 to 3=value 0 v. parities 4 to 7=value 1) and ΔBF D85-D109 of gestation (mm) (b): CY (g)=4290-842a-113b. (R 2=0.41, P<0.001). The gestation length (P<0.001) and the ΔBF between D109 and D1 of lactation (P=0.050) were identified as possible underlying factors of the parity group. The metabolic parameters associated with CY were C4 at 3 to 4 days before farrowing (a), and 10logC4 (b) and 10logNEFA (c) at D1 of lactation: CY (g)=3582-1604a+1007b-922c (R 2=0.39, P=0.001). The colostrum composition was independent of CY. The negative association between CY and ΔBF D85-D109 of gestation could not be further explained based on our data. Sows that were catabolic 1 week prior to farrowing seemed unable to produce colostrum to their full potential. This was especially the case for sows with parities 4 to 7, although they had a similar feed intake, litter birth weight and colostrum composition compared with parities 1 to 3 sows. In conclusion, this study showed that parity and the use of body fat and protein reserves during late gestation were associated with CY, indicating that proper management of the sow's body condition during late gestation could optimise the intrinsic capacity of the sow's CY.

Lameness is a frequently occurring, painful condition of breeding sows that may result in hyperalgesia, i.e., an increased sensitivity to pain. In this study a mechanical nociception threshold (MT) test was used (1) to determine if hyperalgesia occurs in sows with naturally-occurring lameness; (2) to compare measurements obtained with a hand-held probe and a limb-mounted actuator connected to a digital algometer; and (3) to investigate the systematic left-to-right and cranial-to-caudal differences in MT. Twenty-eight pregnant sows were investigated, of which 14 were moderately lame and 14 were not lame. Over three testing sessions, repeated measurements were taken at 5 min intervals on the dorsal aspects of the metatarsi and metacarpi of all limbs. The MT was defined as the force in Newtons (N) that elicited an avoidance response, and this parameter was found to be lower in limbs affected by lameness than in normal limbs (P<0.05). Forelimbs had higher MTs than hindlimbs (P<0.001). The hand-held probe systematically yielded lower values than the actuator (P<0.001), and the MT differed between morning and afternoon testing sessions (P<0.001), as well as between days (P<0.001). The findings provide evidence that lame sows experience hyperalgesia. According to human research, the location of fat accumulation seems to play an important role in the induction of obesity-related inflammatory complications. To evaluate whether an inflammatory response to obesity depends on adipose tissue location, adipokine gene expression, presence of immune cells and adipocyte cell size of subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT) were compared between lean and obese cats. Additionally, the present study proposes the cat as a model for human obesity and highlights the importance of animal models for human research. A total of ten chronically obese and ten lean control cats were included in the present study. Body weight, body condition score and body composition were determined. T-lymphocyte, B-lymphocyte, macrophage concentrations and adipocyte cell size were measured in adipose tissue at different locations. Serum leptin concentration and the mRNA expression of leptin and adiponectin, monocyte chemoattractant protein-1, chemoligand-5, IL-8, TNF-alpha, interferon-gamma, IL-6 and IL-10 were measured in blood and adipose tissues (abdominal and inguinal SAT, and omental, bladder and renal VAT). Feline obesity was characterised by increased adipocyte cell size and altered adipokine gene expression, in favour of pro-inflammatory cytokines and chemokines. Consequently, concentration of T-lymphocytes was increased in the adipose tissue of obese cats. Alteration of adipose tissue was location dependent in both lean and obese cats. Moreover, the observed changes were more prominent in SAT compared with VAT.

Chickens have been raised as food for human consumption for over 4000 years. Over this time they have been continuously selected for specific desirable characteristics by active selection of parents to produce birds which fit perceived needs. Despite this long history of selective breeding and improvements in rearing techniques, the efficiency with which broiler meat is produced has shown a remarkable leap in recent decades. Persistent selection for rapid growth, high feed utilisation efficiency and large cut yield has resulted in modern meat-type poultry lines with superior genetic potential with regard to productivity. However, mortality and the incidence of metabolic diseases has increased in parallel with growth rate. One such disease is broiler ascites syndrome, which has been shown to be closely associated with the fast growth and high meat yield resulting from intense selection and with modern rearing techniques.
