The aim of this study was to evaluate the effects of alcoholic garlic extract on the expression of ABCA1 in macrophages."n"nMethods: Cell viability assay was used in order to detect the cytotoxic dose of alcoholic garlic extract on macrophages. Real-time PCR and Western blotting were performed to study the effects of alcoholic garlic extract on the expression of ABCA1. Macrophage cells were treated by different concentrations of alcoholic garlic extract for 48 h. The total RNA of the treated macrophages were extracted and analyzed by real-time PCR. ABCA1 protein expression was also analyzed using the Western blotting technique."n"nResults: Alcoholic garlic extract increased the ABCA1 mRNA (20-23%) and protein expression (18-37%) in THP-1 macrophage cells compared with the controls (untreated cells)."n"nConclusion: The results of this study are suggestive of the potential effects of alcoholic garlic extract in increasing ABCA1 expression in macrophages, the possibility of promoting reverse cholesterol efflux in macrophages and preventing atherosclerosis.

Objective(s): To quantify the relationship between serum PTH, vitamin D and bone mineral density to determine normal range of vitamin D in normal population. Methods: A total of 830 healthy individuals aged 30 to 76 years old (60.8% women, 39.2% men) living in Tehran were randomly selected, following exclusion of people suffering from endocrine, rheumatologic or chronic diseases, fractures, other conditions or using drugs that affecting bone metabolism. Vitamin D and Ca intakes were assessed with a food frequency questionnaire. Self-reported daily time of sun exposure was used to categorize exposure to sunshine. All participants underwent clinical examination and lumbar spine and proximal of femur BMD by using dual-energy X-ray absorptiometry technique. Plasma level of 25(0H) D, Ca, Phosphate (P), Alkaline phosphatase (ALK P), parathyroid hormone {PTH) measured during winter. The correlation between these results was investigated. Results: PTH correlated inversely with 25(0H) D .In multivariate analysis PTH correlated inversely with whole body BMD (mostly lumbar spine) but not with the hip BMD. No association was found between 25(0H) D levels and BMD at any site in univariate or multivariate analysis. Based on this relationship between PTH, vitamin D and BMD we obtained normal range of vitamin D. Cut -off point for sever vitamin D deficiency was lower than l 8nmol/l and range of moderate & mild were I 8-23 & 23-36 nmol/1, respectively. Conclusion: Normal range determination of vitamin D based on relationship between PTH, vitan..n D and BMD can be shown biological bone variation and is useful in prevention strategy.

Lipoprotein (a) [Lp (a) is formed by assembly of LDL-particles and the carbohydrate rich protein, apolipoprotein (a) [apo (a)]. Elevated plasma Lp (a) levels are an independent predictor of the development of premature coronary artery disease (CAD), but is not clear whether the apo (a) isoform plays on additional and independent role or not. To investigate the possible effect of apo (a) isoform on premature CAD (in patients < 55 years of age), we have analyzed apo (a) isoforms, Lp (a) level and their relation with many recognized CAD risk factors, in 60 male patients with angiographically defined CAD and in 60 male control with no angiographic evidence of CAD. Traditional knowledge (TK) based medicines have gained worldwide attention and presently the scientific community is focussing on proper pharmacological validation and identification of lead compounds for the treatment of various diseases. The North East region of India is the home of valuable traditional herbal remedies. Garcinia morella Desr. (Guttiferae) is one such medicinal plant used by traditional healers for the treatment of inflammatory disorders. The present study was aimed to evaluate the antioxidant and anticancer activity of methanol extracts of the leaf, bark and fruit of G. morella (GM) in different in vitro and in vivo experimental conditions. The results of this study showed that GM methanol extracts possessed in vitro antioxidant and anticancer properties, where the fruit extract (GF) showed maximum activity. The anticancer activity was further confirmed by the results of in vivo administration of GF (200 mg/kg) for ten days to Dalton's Lymphoma (DLA) induced mice. GF extract significantly increased the mean survival time (MST) of the animals, decreased the tumour volume and restored the haematological and biochemical parameters. The present study for the first time reported the anticancer property of GF on DLA. Further from the experiments conducted to elucidate the mechanism of action of GF on DLA, it can be concluded that GF exerts its anticancer effect through induction of caspases and DNA fragmentation that ultimately leads to apoptosis. However further experimentation is required to elucidate the active principle and validate these findings in various in vivo settings.

Abstract Background There are multiple studies in different countries regarding the prevalence of vitamin D deficiency. These studies showed high prevalence of vitamin D deficiency in Asian countries. This study tries to elucidate the prevalence of vitamin D deficiency and its influencing factors in population of Tehran. Methods 1210 subjects 20–64 years old were randomly selected. 25 (OH) D serum levels were measured. Duration of exposure to sunlight, the type of clothing and level of calcium intake and BMI were quantified based on a questionnaire. Results A high percentage of vitamin D deficiency was defined in the study population. Prevalence of severe, moderate and mild Vitamin D deficiency was 9.5%, 57.6% and 14.2% respectively. Vitamin D serum levels had no significant statistical relation with the duration of exposure to sunlight, kind of clothing and BMI. Calcium intake in the normal vitamin D group was significantly higher than the other groups (714.67 ± 330.8 mg/day vs 503.39 ± 303.1, 577.93 ± 304.9,595.84 ± 313.6). Vitamin D serum levels in young and middle aged females were significantly lower than the older group. Conclusions Vitamin D deficiency has a high prevalence in Tehran. In order to avoid complications of vitamin D deficiency, supplemental dietary intake seems essential.

Background and Objective: Respiratory failure is one of the most important respiratory problems in premature infants. Several studies have shown the efficacy of corticosteroids in gestational age less than 34 weeks. This study was done to determine the effect of prenatal betamethasone injection during 34 to 36 weeks of pregnancy on the prevention of newborn respiratory failure. Methods: This clinical trial study was conducted on 140 women with risk of preterm labor at 34 to 36 weeks of gestational age. Women in interventional group were received betamethasone 12 mg IM (2 doses, 12 hours apart). Women in control group were received the same volume of normal saline. Sex and birth weight, respiratory distress syndrome, requiring hospitalization in neonatal intensive care units and require respiratory support were recorded for each newborn. Results: No significant difference was seen in sex and weight of newborns between two groups. The precent of newborns with respiratory distress syndrome in the intervention and control groups was 12.5% and 22%, respectively. This difference was not significant. No significant difference was seen in the need to respiratory support, hospitalized in the NICU between intervention and control groups. Conclusion: Adminestration of betamethasone in 36-34 weeks of pregnancy has no effect on the prevention of respiratory failure in preterm infants.

A three dimensional simulation of cooling and freezing process of fish as a complex geometry is presented. The 3d geometry of the fish was produced by digitizing and the thermo-physical properties of the fish were considered as functions of temperature and water content of the fish. Heating loads and product temperatures during chilling, freezing and post cooling processes for immersion cooling, method in brine and for cold air blasting method were predicted. The influence of effective parameters such as initial temperature and fish water content are also investigated on processing time. Because of the slow freezing rate in immersion cooling technique, the results show that the initial temperature of fish highly affects freezing time. We suggest that a combination of immersion and cold air blasting methods will decrease the freezing time in comparison with immersion method when solely applied. Comparison of temperature distribution in present study shows a good agreement with other numerical and experimental results.

Microthrombi and circulating cell clusters are common microscopic findings in patients with coronavirus disease 2019 (COVID-19) at different stages in the disease course, implying that they may function as the primary drivers in disease progression. Inspired by a recent flow imaging cytometry study of the blood samples from patients with COVID-19, we perform computational simulations to investigate the dynamics of different types of circulating cell clusters, namely white blood cell (WBC) clusters, platelet clusters, and red blood cell clusters, over a range of shear flows and quantify their impact on the viscosity of the blood. Our simulation results indicate that the increased level of fibrinogen in patients with COVID-19 can promote the formation of red blood cell clusters at relatively low shear rates, thereby elevating the blood viscosity, a mechanism that also leads to an increase in viscosity in other blood diseases, such as sickle cell disease and type 2 diabetes mellitus. We further discover that the presence of WBC clusters could also aggravate the abnormalities of local blood rheology. In particular, the extent of elevation of the local blood viscosity is enlarged as the size of the WBC clusters grows. On the other hand, the impact of platelet clusters on the local rheology is found to be negligible, which is likely due to the smaller size of the platelets. The difference in the impact of WBC and platelet clusters on local hemorheology provides a compelling explanation for the clinical finding that the number of WBC clusters is significantly correlated with thrombotic events in COVID-19 whereas platelet clusters are not. Overall, our study demonstrates that our computational models based on dissipative particle dynamics can serve as a powerful tool to conduct quantitative investigation of the mechanism causing the pathological alterations of hemorheology and explore their connections to the clinical manifestations in COVID-19., Competing Interests: Declaration of interests The authors declare no competing interests., (Copyright © 2022 Biophysical Society. Single ventricle hearts have only one ventricle that can pump blood effectively and the treatment requires three stages of operations to reconfigure the heart and circulatory system. At the second stage, Glenn procedure is performed to connect superior vena cava (SVC) to the pulmonary arteries (PA). For the third and most complex operation, called Fontan, an extracardiac conduit is used to connect inferior vena cava (IVC) to the PL and thereafter no deoxygenated blood goes to the heart. Predicting Hemodynamic Performance of Fontan Operation using computational fluid dynamics (CFD) is hypothesized to improve outcomes and optimize this treatment planning in children with single-ventricle heart disease. An important reason for this surgical planning is to reduce the development of pulmonary arteriovenous malformations (PAVM) and the need to perform Fontan revisions. The purpose of this study was to develop amodel for Fontan surgical planning and use this model to compare blood circulation in two designed graft types of Fontan operation known as T-shape and Y-graft. The functionality of grafts was compared in terms of power loss (PL) and hepatic flow distribution (HFD), a known factor in PAVM development. To perform this study, ten single-ventricle children with Glenn physiology were included and a CFD model was developed to estimate the blood flow circulation to the left and right pulmonary arteries. The estimated blood flow by CFD was compared with that measured by cardiovascular magnetic resonance. Results showed that there was an excellent agreement between the net blood flow in the right and left pulmonary arteries computed by CFD and CMR (ICC= 0.98, P-value ≥0.21). After validating the accuracy of each CFD model, Fontan operations using T-shape and Y-graft conduits were performed in silico for each patient and the developed CFD model was used to predict the post-surgical PL and HFD. We found that the PL in the Y-graft was significantly lower than in the T-shape (P-value ≤0.001) and HFD was significantly better balanced in Y-graft compared to the T-shape (P-value=0.004)., Competing Interests: Conflict of interests: None. Hemorheology is known to be a major diagnostic tool for many blood-altering diseases. While hemorheological measures of blood, such as the general flow curve, shear-thinning behavior, and its yield stress, are much more studied in detail, thixotropic behavior and thermokinematic memory formation in blood are less understood. Here, we study the thermokinematic memory formation in blood, resulting in a clear sensitivity to the flow history, i.e., thixotropic behavior. We also measure the thixotropic timescale for blood flow using a well-defined flow protocol. Employing a series of in silico flow loops in which the blood is subject to a sweep down/up flow, we measure and discuss the dependence of the thixotropic timescale to the concentration of fibrinogen in the plasma as the main driver of structural evolution under flow.

The crucial role of the hemorheological characteristics of blood in a range of diagnoses, treatments and drug delivery mechanisms is widely accepted. Nonetheless, the literature on blood rheology remains inconclusive and sometimes even contradictory. This is in part due to natural variance of blood samples from one study to another, but also stems from fundamental differences in the consequences of the choice of rheometric flow employed. Here, and using a detailed and accurate computational scheme, we thoroughly study the role of flow type in measurement of blood viscosity. Performing these in silico measurements, we isolate the role of flow type and geometry at different hematocrit levels. We show that flow curves obtained in pressure-driven flows relevant to laminar circulatory flows deviate greatly from ones obtained in drag flow at the same hematocrit level. Our numerical platform also allows for the yield stress to be measured under quiescent conditions and without imposing any flow for different hematocrits. We discuss the scaling of the yield stress with the hematocrit level, and show that the differences in pressure vs. drag flows stem from the Red Blood Cell (RBC) orientation at different flow rates as well as the existence of a cell free layer close to the walls.

Hyperviscosity syndrome (HVS) is characterized by an increase of the blood viscosity by up to seven times the normal blood viscosity, resulting in disturbances to the circulation in the vasculature system. HVS is commonly associated with an increase of large plasma proteins and abnormalities in the properties of red blood cells, such as cell interactions, cell stiffness, and increased hematocrit. Here, we perform a systematic study of the effect of each biophysical factor on the viscosity of blood by employing the dissipative particle dynamic method. Our in silico platform enables manipulation of each parameter in isolation, providing a unique scheme to quantify and accurately investigate the role of each factor in increasing the blood viscosity. To study the effect of these four factors independently, each factor was elevated more than its values for a healthy blood while the other factors remained constant, and viscosity measurement was performed for different hematocrits and flow rates. Although all four factors were found to increase the overall blood viscosity, these increases were highly dependent on the hematocrit and the flow rates imposed. The effect of cell aggregation and cell concentration on blood viscosity were predominantly observed at low shear rates, in contrast to the more magnified role of cell rigidity and plasma viscosity at high shear rates. Additionally, cell-related factors increase the whole blood viscosity at high hematocrits compared with the relative role of plasma-related factors at lower hematocrits. This paper presents an overview of the different methods used for sensitivity (i.e., responsivity and noise equivalent power) determination of state-of-the-art field-effect transistor-based THz detectors/sensors. We point out that the reported result may depend very much on the method used to determine the effective area of the sensor, often leading to discrepancies of up to orders of magnitude. The challenges that arise when selecting a proper method for characterisation are demonstrated using the example of a 2×7 detector array. This array utilises field-effect transistors and monolithically integrated patch antennas at 620 GHz. The directivities of the individual antennas were simulated and determined from the measured angle dependence of the rectified voltage, as a function of tilting in the E- and H-planes. Furthermore, this study shows that the experimentally determined directivity and simulations imply that the part of radiation might still propagate in the substrate, resulting in modification of the sensor effective area. Our work summarises the methods for determining sensitivity which are paving the way towards the unified scientific metrology of FET-based THz sensors, which is important for both researchers competing for records, potential users, and system designers.

Normal haemostasis is an important physiological mechanism that prevents excessive bleeding during trauma, whereas the pathological thrombosis especially in diabetics leads to increased incidence of heart attacks and strokes as well as peripheral vascular events. In this work, we propose a new multiscale framework that integrates seamlessly four key components of blood clotting, namely transport of coagulation factors, coagulation kinetics, blood cell mechanics and platelet adhesive dynamics, to model the development of thrombi under physiological and pathological conditions. We implement this framework to simulate platelet adhesion due to the exposure of tissue factor in a three-dimensional microchannel. Our results show that our model can simulate thrombin-mediated platelet activation in the flowing blood, resulting in platelet adhesion to the injury site of the channel wall. Furthermore, we simulate platelet adhesion in diabetic blood, and our results show that both the pathological alterations in the biomechanics of blood cells and changes in the amount of coagulation factors contribute to the excessive platelet adhesion and aggregation in diabetic blood. Taken together, this new framework can be used to probe synergistic mechanisms of thrombus formation under physiological and pathological conditions, and open new directions in modelling complex biological problems that involve several multiscale processes.

Low-level laser has been indicated to have the capability to facilitate the differentiation of the osteoclastic and osteoblastic cells which are responsible for the bone remodeling process. The aim of this study was to evaluate the effects of InGaAlP laser with a wavelength of 660 nm on the rate of tooth movement and histological status. Thirty male Wistar rats of 7 weeks old were selected for this study. The rats were randomly divided into two groups of 15 each to form the experimental (laser-irradiated) and control (non-irradiated) groups. The control group received unilateral orthodontic appliance design (one quadrant), but the laser-irradiated group received split-mouth design, with orthodontic appliance on both sides and laser irradiation on one side only (group b) and on the contralateral side (group c). The orthodontic appliance consisted of a NiTi closed coil spring with a length of 5 mm which was ligated to maxillary molar and incisor. A total of 60 g of force was applied to the rat molar. The diode laser (660 nm) was irradiated with an output power of 25 mW in continuous mode for a total time of 5 min in the laser-irradiated group. After 14 days of orthodontic tooth movement, the amount of tooth movements was measured. In the laser-irradiated group, the amount of tooth movement was significantly greater than that of the non-irradiated group (2.3-fold), but there was no significant difference between the non-irradiated and indirectly irradiated groups. Histopathological studies revealed that the number of osteoclasts in the laser-irradiated group was significantly greater than that of the non-irradiated group (1.5-fold) while this number was almost the same in the non-irradiated and indirectly irradiated groups. The results suggested that low-level laser can accelerate the rate of bone remodeling. However, in order to utilize the low-level laser as an adjunct in orthodontic practice on patients, further research studies are needed for finding the appropriate dosage for the human tissues.

Objectives. To compare the analgesic efficacy of intrathecal clonidine and fentanyl added to bupivacaine after cesarean section. Methods. Ninety patients scheduled for cesarean section under spinal anesthesia were randomly allocated to one of the three following groups to receive bupivacaine 10 mg combined with 75 µg clonidine (group C), bupivacaine 10 mg combined with 0.5 mL fentanyl (group F), and bupivacaine 10 mg combined with 0.5 mL distilled water (group P), intrathecally. The time to first analgesic request, analgesic requirement in the first 24 hours after surgery, sensory and motor blockade onset time, duration of sensory and motor blockade, the incidence of hypotension, ephedrine requirements, bradycardia, and hypoxemia were recorded. Results. The duration of anesthesia in clonidine group (275.10 ± 96.09) was longer compared to the placebo (211.73 ± 74.80) and fentanyl (192.33 ± 30.36) groups. This difference between group C versus F (P = 0.006) and P groups (P < 0.001) was significant. Similarly, the mean time to first analgesic request was also longer in group C (519.44 ± 86.25) than in groups F (277.88 ± 94.25) and P (235.43 ± 22.35 min). This difference between group C versus F (P < 0.001) and P groups (P < 0.001) was significant. Conclusion. Intrathecal clonidine 75 µg with bupivacaine prolonged the time to first analgesic request compared to fentanyl; however, the total analgesic consumption within the first 24 h postoperative was similar in fentanyl and clonidine groups following cesarean section. Oral actinomycosis is not a common disease, but it can cause massive destruction. This article reports a case of implant failure associated with actinomycosis. A 55-year-old Caucasian male patient had tooth #20 extracted years ago and an implant placed 3 years ago. The #20 implant area developed an abscess about 1½ years after implant placement. Radiographic findings revealed a large radiolucency on the mesial aspect of the #20 implant. The implant was surgically removed and the lesion thoroughly debrided. The patient experienced severe pain when the apical soft tissue was curreted following implant removal. A periapical radiograph revealed that the lesion approached the mental foramen. A short course of antibiotics was prescribed. Histological observation found sulfur granules, which were found to be actinomycotic colonies. Peri-implant actinomycosis was diagnosed. No recurrence had occurred at the 1-year follow-up.

The purpose of this study was to investigate the effect of S-allylcysteine (SAC), the most abundant organosulfur compound in aged garlic extract, on the expression of ATP-binding cassette transporter A1 in human THP-1 macrophages. The human monocyte THP-1 cells were differentiated to macrophage cells in the presence of phorbol 12-myristate13-acetate (PMA). Macrophage cells were then treated with different concentrations (10, 20 and 40 mM) of SAC for 24 h. Total RNA of treated macrophages was extracted and analyzed with real-time RT-PCR. ABCA1 protein expression was also analyzed with western blotting. Results showed that SAC increased the ABCA1 mRNA (1.82-, 2.07- and 2.23-fold) and protein (1.37-, 1.55- and 2.08-fold) expression in macrophage THP-1 cells compared with control (untreated cells).
