Abstract

Abstract Background The epithelial lining of the gut expresses intestinal fatty-acid binding proteins (I-FABPs), which increase in circulation and in plasma concentration during intestinal damage. From the perspective of obesity, the consumption of a diet rich in fat causes a disruption in the integrity of the gut barrier and an increase in its permeability. Hypothesis There is an association between the expression of I-FABP in the gut and various metabolic changes induced by a high-fat (HF) diet. Methods Wistar albino rats (n = 90) were divided into three groups (n = 30 per group), viz. One control and two HF diet groups (15 and 30%, respectively) were maintained for 6 weeks. Blood samples were thus collected to evaluate the lipid profile, blood glucose level and other biochemical tests. Tissue sampling was conducted to perform fat staining and immunohistochemistry. Results HF diet-fed rats developed adiposity, insulin resistance, leptin resistance, dyslipidemia, and increased expression of I-FABP in the small intestine compared to the control group. Increased I-FABP expression in the ileal region of the intestine is correlated significantly with higher fat contents in the diet, indicating that higher I-FABP expression occurs due to increased demand of enterocytes to transport lipids, leading to metabolic alterations. Conclusion In summary, there is an association between the expression of I-FABP and HF diet-induced metabolic alterations, indicating that I-FABP can be used as a biomarker for intestinal barrier dysfunction.

Abstract

Reproductive longevity is essential for fertility and influences healthy ageing in women1,2, but insights into its underlying biological mechanisms and treatments to preserve it are limited. Here we identify 290 genetic determinants of ovarian ageing, assessed using normal variation in age at natural menopause (ANM) in about 200,000 women of European ancestry. These common alleles were associated with clinical extremes of ANM; women in the top 1% of genetic susceptibility have an equivalent risk of premature ovarian insufficiency to those carrying monogenic FMR1 premutations3. The identified loci implicate a broad range of DNA damage response (DDR) processes and include loss-of-function variants in key DDR-associated genes. Integration with experimental models demonstrates that these DDR processes act across the life-course to shape the ovarian reserve and its rate of depletion. Furthermore, we demonstrate that experimental manipulation of DDR pathways highlighted by human genetics increases fertility and extends reproductive life in mice. Causal inference analyses using the identified genetic variants indicate that extending reproductive life in women improves bone health and reduces risk of type 2 diabetes, but increases the risk of hormone-sensitive cancers. These findings provide insight into the mechanisms that govern ovarian ageing, when they act, and how they might be targeted by therapeutic approaches to extend fertility and prevent disease. Hundreds of genetic loci associated with age at menopause, combined with experimental evidence in mice, highlight mechanisms of reproductive ageing across the lifespan.

Abstract

Posterior reversible encephalopathy syndrome (PRES) is a clinical and radiological entity initially described in 1996. PRES frequently develops in patients with preeclampsia and eclampsia. There is not much literature on risk factors causing PRES in pregnant patients with eclampsia. This study aimed to determine the incidence of PRES in eclampsia, its association with pregnancy, risk factors, and maternal and perinatal outcomes. Patients and methods: All patients who were admitted with eclampsia and developed PRES in an intensive care unit of a tertiary medical facility between 1997 and 2017 were included in the study. Patients’ demographics, pregnancy and gestational data, treatment mode, and outcomes were retrospectively obtained from their medical charts/files. Data were entered using SPSS program version 23. Chi-square test was used to compare the variables, and a p value of Results: A total of 151 patients were admitted during the study period, and 25 developed PRES. The diagnosis was common in patients older than 25 years. Eclampsia patients who developed PRES were without any pregnancy-associated comorbidities (p p p p p Conclusion: Of eclampsia patients, 16% developed PRES, which is on the lower side than the reviewed literature (10%–90%). Eclampsia on presentation, recurrent seizures, postpartum eclampsia, cesarean delivery, and labetalol use were associated with increased risk of PRES development.

Abstract

This experiment aimed to determine the crop evapotranspiration (ETc) and groundwater input in total water used by wheat (Triticum aestivum) crop and to use the CROPWAT model to predict the crop ETc. To estimate the on-field ETc and groundwater contribution, the combining lysimeter technique was used. The water levels below the soil surface were kept at 1.60, 2.20, and 2.80 m and each water table depth was replicated three times. A field experiment was conducted under silt loam (SL) and silty-clay loam (SCL) soil conditions. The climatic parameters and water balance components were recorded accordingly. The on-field ETc was compared with the predicted ETc by CROPWAT model. Under SL soil, the observed ETc was 442, 427 and 401 mm and under SCL soil conditions it was observed as 419, 402 and 389 mm at the water table depth of 1.60, 2.20, and 2.80 m, respectively. The ETc was reduced with an increase in water level below the surface. The groundwater contribution under SL and SCL soil was observed as 50%, 41%, and 30% and 43%, 34%, and 24%, at 1.60, 2.20 and 2.80 m water table depth, respectively. The predicted ETc by CROPWAT model for SL and SCL soil conditions was 428.8 and 410.7 mm, respectively. The projected ETc was equal to the average ETc observed under lysimeter experiment. Thus, the use of CROPWAT model is recommended to overcome waterlogging and salinity problems and to conserve water resources. Abstract

This study was conducted to estimate the evapotranspiration and to probe the seasonal groundwater contribution in total crop water use by the sesame crop under different water table depths. The method of combining lysimeter was executed to investigate the groundwater contribution in total water used by the crop. The water table depths were maintained at 1.60, 2.0 and 2.40 m, respectively. Each water table depth was replicated for three times. Climate conditions under which crop was grown were monitored and all the water balance components were recorded accordingly. The obtained evapotranspiration (ET) from lysimeter experiment was compared with the predicted ET by CROPWAT model. The experimentally observed ET were 457.5, 452 and 428 mm under the water table depths of 1.60, 2.0 and 2.40 m, respectively. The predicted ET using CROPWAT model was 434 mm, which was very close to the observed values. Under the lysimeter experiment, the groundwater contribution in total crop water use was observed as 37%. The predicted crop coefficient was ranged from 0.38 to 0.98, whereas the experimental crop coefficient was 1.0. The trend of soil moisture balance predicted by the model revealed the same as that experimentally observed. Thus, the use of CROPWAT model is recommended to redesign irrigation amounts and to prevent soil salinity and waterlogging problems. [ABSTRACT FROM AUTHOR]
