Abstract Increased production of reactive oxygen species (ROS) leads to oxidative stress, with its damaging effect extending to the mitochondria and plasma membrane. Further, prolonged inflammation can result in chronic disease development. The marine microdiatom Odontella aurita is recognized for its potential in food and pharmaceutical development. Moreover, it contains antioxidant and anti-inflammatory properties. However, studies regarding the efficacy of their varying extract forms and their underlying mechanisms remain scarce. Therefore, this study aims to explore the antioxidant and anti-inflammatory effects of Odontella aurita extracts obtained using various extraction methods (hot water, 70% ethanol, and chloroform:methanol (CM)). Among the three Odontella aurita extracts, the CM extract demonstrated superior efficacy in protecting RAW 264.7 cells from H2O2-induced cytotoxicity. It significantly lowered the levels of ROS and enhanced the expression of superoxide dismutase and glutathione peroxidase. Furthermore, the CM extract outperformed other extracts in inhibiting LPS-induced nitric oxide production, reducing mRNA levels in nitric oxide synthase, cyclooxygenase, and the proinflammatory cytokines interleukin IL-1β, IL-6, TNFα. Additionally, CM extract effectively suppressed the activation of NF-κB/IκBα and JAK2-STAT3 in LPS-induced RAW 264.7 cells. HPLC–UV analysis revealed a remarkable 33-fold higher fucoxanthin content in CM compared to the ethanol extract. GC–MS analysis identified elevated levels of cholest-5-en-3-ol, phytol, eicosapentaenoic acid methyl ester, methyl palmitate, palmitoleic acid methyl ester, and neophytadiene in the CM extract. These findings suggest that Odontella aurita CM extract is a promising antioxidant candidate for preventing or treating inflammatory diseases, consequently emphasizing its potential for further development. Abstract Background Current approaches to profile the single-cell transcriptomics of human pancreatic endocrine cells almost exclusively rely on freshly isolated islets. However, human islets are limited in availability. Furthermore, the extensive processing steps during islet isolation and subsequent single cell dissolution might alter gene expressions. In this work, we report the development of a single-nucleus RNA sequencing (snRNA-seq) approach with targeted islet cell enrichment for endocrine-population focused transcriptomic profiling using frozen archival pancreatic tissues without islet isolation. Results We cross-compared five nuclei isolation protocols and selected the citric acid method as the best strategy to isolate nuclei with high RNA integrity and low cytoplasmic contamination from frozen archival human pancreata. We innovated fluorescence-activated nuclei sorting based on the positive signal of NKX2-2 antibody to enrich nuclei of the endocrine population from the entire nuclei pool of the pancreas. Our sample preparation procedure generated high-quality single-nucleus gene-expression libraries while preserving the endocrine population diversity. In comparison with single-cell RNA sequencing (scRNA-seq) library generated with live cells from freshly isolated human islets, the snRNA-seq library displayed comparable endocrine cellular composition and cell type signature gene expression. However, between these two types of libraries, differential enrichments of transcripts belonging to different functional classes could be observed. Conclusions Our work fills a technological gap and helps to unleash frozen archival pancreatic tissues for molecular profiling targeting the endocrine population. This study opens doors to retrospective mappings of endocrine cell dynamics in pancreatic tissues of complex histopathology. We expect that our protocol is applicable to enrich nuclei for transcriptomics studies from various populations in different types of frozen archival tissues.

Phosphor‐sensitized fluorescent (PSF) organic light‐emitting diodes (OLEDs) emerge as an attractive solution for realizing high‐performance displays with high color purity, meeting the Broadcast Television 2020 (BT.2020) requirements, by fully harnessing the benefits of both phosphorescent and fluorescent emitters. Herein, the synergistic effect of a Pt sensitizer and a boron (B)‐based multi‐resonance (MR)‐fluorescent emitter are introduced to enhance the efficiency of blue‐emitting PSF–OLEDs. Notably, it is confirmed that the interaction between the empty pz orbital in the boron atom of the MR emitter and the unpaired electrons in the dz2 orbital of the Pt emitter plays a crucial role. The combination results in an enhancement of the horizontal emitting dipole orientation (EDO) to 81%, a marked improvement compared to the individual components of Pt phosphor (69%) or MR emitter (78%). The synergistic approach leads to the realization of high‐performance blue‐emitting PSF‐OLEDs with a maximum external quantum efficiency of 25%, a peak intensity at 465 nm in the electroluminescence spectrum, and a full width at half maximum of 29 nm, achieved by enhancing the outcoupling. In these findings, insight is provided into design strategies for developing PSF systems based on Pt sensitizer and B‐terminal emitter to achieve efficient energy transfer as well as improved EDO.

Yuzu (Citrus junos Sieb.) is a peel-edible fruit with a pleasant aroma, but its bitter taste can impact consumer appeal. In this study, an efficient enzymatic method reduced bitterness in green yuzu powder (GYP). Cellulase KN and naringinase from Aspergillus oryzae NYO-2 significantly decreased naringin and neohesperidin content by over 87 %, while increasing total dietary fiber and soluble dietary fiber by up to 10 % and 51 %, respectively. Insoluble dietary fiber decreased by up to 22 %. Cellulose, hemicellulose, lignin, and pectin contents in enzyme-treated YP decreased by 1.15–2.00-fold, respectively. Enzyme-treated GYP exhibited improved physicochemical properties, including enhanced solubility, oil-holding capacity, and water swelling capacities. 3T3-L1 cells treated with cellulase-treated GYP and naringinase-treated GYP showed lower lipid accumulation and higher lipolysis capability than GYP, along with decreased fatty acid synthase contents. These findings suggest that enzyme-treated GYP holds potential as a functional ingredient in the food industry.

Abstract Heterogeneous integration of dissimilar crystalline materials has recently attracted considerable attention due to its potential for high-performance multifunctional electronic and photonic devices. The conventional method for fabricating heterostructures is by heteroepitaxy, in which epitaxy is performed on crystallographically different materials. However, epitaxial limitations in monolithic growth of dissimilar materials prevent implementation of high quality heterostructures, such as complex-oxides on conventional semiconductor platforms (Si, III-V and III-N). In this work, we demonstrate gallium nitride (GaN) high-electron-mobility transistors with crystalline complex-oxide material enabled by heterogeneous integration through epitaxial lift-off and direct stacking. We successfully integrate high-κ complex-oxide SrTiO3 in freestanding membrane form with GaN heterostructure via a simple transfer process as the gate oxide. The fabricated device shows steep subthreshold swing close to the Boltzmann limit, along with negligible hysteresis and low dynamic on-resistance, indicating very low defect density between the SrTiO3 gate oxide and GaN heterostructure. Our results show that heterogeneous integration through direct material stacking is a promising route towards fabricating functional heterostructures not possible by conventional epitaxy.

Abstract Melosira nummuloides is a marine diatom with potential use as food, fuel, and a dietary supplement. However, the efficacy of its extraction and drying techniques have not been explored. Here, M. nummuloides powders were prepared by two drying methods—hot-air drying (HAD) and freeze-drying (FD)—and extracted with hot water, ethanol, methanol, and chloroform:methanol (CM) at a ratio of 2:1 v/v. The antioxidant and anti-inflammatory activity of each extract was investigated. The CM extract had the greatest 2,2-diphenyl-1-picrylhydrazyl and 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity among the solvent extracts, and a slight difference in antioxidant activity was observed across the various drying methods. Compared to other extracts, both the FD-CM and HAD-CM extracts showed stronger anti-inflammatory effects by inhibiting nitric oxide production in lipopolysaccharide-stimulated RAW 264.7 cells. Furthermore, the FD-CM extract contained a wide range of lipophilic compounds. Notably, myristic acid (29.08 ± 0.45 mg/g dry weight powder extract (DW)), oleic acid (25.20 ± 0.92 mg/g DW), palmitoleic acid (10.77 ± 0.41 mg/g DW), eicosapentaenoic acid (12.53 ± 1.00 mg/g DW), neophytadiene (8.42 ± 0.51 mg/g DW), and α-linolenic acid (1.27 ± 0.005 mg/g DW) were among the prominent compounds identified. It is plausible to suggest that the abundance of these lipophilic compounds contributes to the remarkable antioxidative and anti-inflammatory potential exhibited by the FD-CM extract. Our results provide insights into the preferential drying methods and extraction solvents for producing M. nummuloides-based products with enhanced antioxidant and anti-inflammatory activity.

Abstract This study aimed to assess the impact of a prolonged carbapenem use-focused antimicrobial stewardship program (ASP) on antimicrobial consumption and clinical outcomes and to analyze factors affecting adherence to interventions. Patients prescribed carbapenems for ≥ 2 weeks received intervention. Interrupted time-series analysis was performed to compare antimicrobial consumption before and after intervention. Factors associated with non-adherence to intervention were investigated. Of 273 patients who were eligible for intervention, discontinuation or de-escalation was recommended in 256 (94.1%) and intervention was accepted in 136 (53.1%) patients. Before intervention, carbapenem consumption significantly increased to 1.14 days of therapy (DOT)/1000 patient days (PD)/month (P = 0.018).
