Winter wheat grain protein content (GPC) is an important criterion for assessing grain quality. A timely and simple GPC model is urgently required for GPC prediction ahead of maturity. The GPC model included regressional models of dry matter and N accumulation and translocation for anthesis and post-anthesis stages, and incorporated both soil nitrogen (N) supply and meterological factors based on historical as well as current season data, final GPC were calculated as the ratio of N accumulation to dry matter in grain at maturity. This study conducted six field experiments during the 2003–2006 and 2008–2011 growing seasons to establish and validate the model. A three-way factorial arrangement of N fertilization, sowing date, and cultivar was conducted using a split-plot design. Critical growth parameters were determined by field measurements, and historical seasonal meteorological data covering the growing period were collected.

Two new oleanane triterpenoids, sysamarins F (1) and G (2), along with one known analogue (3), were isolated from the leaves of Syzygium samarangense. Their structures were elucidated by means of extensive spectroscopic techniques, including interpretation of 1D and 2D NMR spectra and comparison with the values reported in the literature.

Plants of the genus Camellia are widely cultivated throughout the world as ornamentals because of their bright and large flowers. The widely cultivated varieties are mainly derived from the mutant lines and hybrid progenies of Camellia japonica Linn. and Camellia reticulata Lindl. While their geographical distributions and environmental adaptabilities are significantly different, no systematic comparison has been conducted between these two species. To investigate differences in how these plants have adapted to their environments, we measured photosynthesis and 20 leaf functional traits of C. japonica and C. reticulata grown under the same conditions. Compared with C. japonica, C. reticulata showed higher values for light saturation point, light-saturated photosynthetic rate, leaf dry mass per unit area and stomatal area, but lower values for apparent quantum efficiency, leaf size, stomatal density and leaf nitrogen content per unit mass. Stomatal area was positively correlated with light-saturated photosynthetic rate and light saturation point, but negatively correlated with stomatal density. The differences between C. reticulata and C. japonica were mainly reflected in their adaptations to light intensity and leaf morphological traits. C. reticulata is better adapted to high light intensity than C. japonica. This difference is related to the two species' differing life forms. Thus, leaf morphological traits have played an important role in the light adaptation of C. reticulata and C. japonica, and might be first noticed and selected during the breeding process. These findings will contribute to the cultivation of camellia plants.

2,2',4,4'-tetrabrominated diphenyl ether (BDE-47) is one of the most widely distributed PBDEs. BDE-47 is also the most abundant in organisms and the most toxic to humans and animals. Herein, we have studied the pathway of BDE-47 degradation and gene involvement in Acinetobacter pittii GB-2. This degradation is dominated by hydroxylation, resulting in hydroxylated products 6-OH-BDE-47, 5-OH-BDE-47 and 2'-OH-BDE-28, and bromophenol products 2,4-DBP and 4-BP. Transcriptome sequencing results showed 359 differentially expressed genes (DEGs) induced by BDE-47, of which 159 were up-regulated and 200 were down-regulated. The up-regulated ones were mainly related to substance transport, degradation and cell stress. From these results, we suggest that 1,2-dioxygenase, phenol hydroxylase and monooxygenase are involved in BDE-47 degradation. The function of AntA gene was identified by constructing a prokaryotic expression vector. Our study contributes to understanding how the metabolism of strain GB-2 changes under BDE-47 stress conditions, and sheds light on the mechanism of BDE-47 degradation.

The plant growth, development, and secondary metabolism are regulated by R2 R3-MYB transcription factors. This study identified the R2 R3-MYB genes in the genome of Andrographis paniculata and analyzed the chromosomal localization, gene structure, and conserved domains, phylogenetic relationship, and promoter cis-acting elements of these R2 R3-MYB genes. Moreover, the gene expression profiles of R2 R3-MYB genes under abiotic stress and hormone treatments were generated by RNA-seq and validated by qRT-PCR. The results showed that A. paniculata contained 73 R2 R3-MYB genes on 21 chromosomes. These members belonged to 34 subfamilies, 19 of which could be classified into the known subfamilies in Arabidopsis thaliana. The 73 R2 R3-MYB members included 36 acidic proteins and 37 basic proteins, with the lengths of 148-887 aa. The domains, motifs, and gene structures of R2 R3-MYBs in A. paniculata were conserved. The promoter regions of these genes contains a variety of cis-acting elements related to the responses to environmental factors and plant hormones including light, ABA, MeJA, and drought. Based on the similarity of functions of R2 R3-MYBs in the same subfamily and the transcription profiles, ApMYB13/21/35/67/73(S22) may regulate drought stress through ABA pathway; ApMYB20(S11) and ApMYB55(S2) may play a role in the response of A. paniculata to high temperature and UV-C stress; ApMYB5(S7) and ApMYB33(S20) may affect the accumulation of andrographolide by regulating the expression of key enzymes in the MEP pathway. This study provides theoretical reference for further research on the functions of R2 R3-MYB genes in A. paniculata and breeding of A. paniculata varieties with high andrographolide content.

Rhododendron delavayi Franch. is a famous ornamental plant. However, seasonal drought caused by a monsoon climate seriously limits its growth and flowering performance in urban gardens. Our previous study has found that brassinosteroids (BRs) can improve the tolerance of R. delavayi to drought stress. Here, we employed a data-independent acquisition (DIA) approach to compare the protein expression profiles under drought treatment (D) and pre-treatment with BR before drought treatment (BR). With an increase in drought stress, the net photosynthetic rate, stomatal conductance, and transpiration rate in the BR treatment showed more stable changes that were significantly higher than those in the D treatment. However, the contents of malondialdehyde, soluble sugar, soluble protein, and the activity of superoxide dismutase (SOD), peroxidase, and catalase showed opposite trends. The pre-treatment with BR alleviated the negative effect of drought stress on the photosynthetic performance of R. delavayi. A total of 3453 differentially expressed proteins (DEPs) were identified, and 683 DEPs were significantly expressed in the D and BR treatments. The DEPs uniquely expressed in the BR treatment participated in the pathways of “ribosome”, “ether lipid metabolism”, “photosynthesis”, and “oxidative phosphorylation”. The improvement effect of the BR treatment on the drought tolerance of R. delavayi was mainly attributed to improved photosynthesis by alleviating stomatal closure and oxidative stress, maintaining the integrity and stability of the ribosomal complex to mediate protein synthesis and the balance between energy metabolism and carbon metabolism. Our study presents a comprehensive understanding of the key proteins and metabolic pathways related to the response of R. delavayi to drought and will contribute to the breeding of drought-tolerant rhododendrons.

Two new phloroglucinol derivatives were isolated from the twigs and leaves of Syzygium brachyantherum. Their structures were elucidated based on extensive spectroscopic data. They exhibited weak inhibitory activities against α-glucosidase and PTP1B.

A new monoterpenoid glycoside, together with seven known aliphatic acid derivatives, was isolated from the twigs and leaves of Syzygium fluviatile. Their structures were elucidated by means of NMR and HR-ESI-MS data and comparison with the reported values. This is the first example of a monoterpenoid glycoside from Syzygium plants.

Upon a sudden transition from high to low light, the rate of CO2 assimilation (AN) in some plants first decreases to a low level before gradually becoming stable. However, the underlying mechanisms remain controversial. The activity of chloroplast ATP synthase (gH+) is usually depressed under high light when compared with low light. Therefore, we hypothesize that upon a sudden transfer from high to low light, the relatively low gH+ restricts ATP synthesis and thus causes a reduction in AN. To test this hypothesis, we measured gas exchange, chlorophyll fluorescence, P700 redox state, and electrochromic shift signals in Bletilla striata (Orchidaceae). After the transition from saturating to lower irradiance, AN and ETRII decreased first to a low level and then gradually increased to a stable value. Within the first seconds after transfer from high to low light, gH+ was maintained at low levels. During further exposure to low light, gH+ gradually increased to a stable value. Interestingly, a tight positive relationship was found between gH+ and ETRII. These results suggested that upon a sudden transition from high to low light, AN was restricted by gH+ at the step of ATP synthesis. Taken together, we propose that the decline in AN upon sudden transfer from high to low light is linked to the slow kinetics of chloroplast ATP synthase.

Leaves under high light may suffer from risks caused by excessive light energy and dehydration. However, it remains unclear how leaf water-related traits affect the photosynthetic acclimation of plants to light intensity. We investigated the photosynthesis and leaf water-related traits of two hybrids (Rhododendron ‘XXL’ and ‘Furnivall’s daughter’) and one native species (Rhododendron delavayi) under different growth irradiances to determine the importance of leaf water-related traits on the photosynthetic acclimation of rhododendrons to irradiance. Compared to plants under low light, the values for light saturation point (LSP), stomatal density (SD), leaf dry mass per unit area, and vein density (Dv) in the three rhododendrons were higher under high light, but the values for chlorophyll content and leaf size were lower. There were significant differences in light-saturated photosynthetic rate (Pmax), stomatal conductance (gs), transpiration rate (Tr), and non-photochemical quenching (NPQ) in R. delavayi and Rhododendron ‘XXL’ between plants grown under high and low irradiance levels, but not in ‘Furnivall’s daughter’. The positive correlations of Dv and SD with Pmax, Tr, and LSP indicate that the increase in water transport capacity can improve water supply to meet transpirational demand and regulate photosynthetic performance under high irradiance. Meanwhile, the decrease in chlorophyll content and the increase in NPQ prevent the photodamage of photosystem II by decreasing light absorption and enhancing heat dissipation, respectively, when the plants are exposed to high light. These results suggest that adjusting leaf water-related traits and reducing the damage on photosystem II caused by excess light play important roles in the photosynthetic acclimation of rhododendrons to growth irradiance.

This paper is concerned with small quadratic perturbations to one parameter family of generic reversible quadratic vector fields with a simple center. The first objective is to show that this system exhibits two small amplitude limit cycles emerging from a Hopf bifurcation. The second one we prove that the system has no limit cycle around the weak focus of order two. The results may be viewed as a contribution to proving the conjecture on cyclicity proposed by Iliev (1998).

Rhododendron delavayi is an alpine evergreen ornamental plant, but water shortage limits its growth and development in urban gardens. However, the adaptive mechanism of alpine evergreen rhododendrons to drought remains unclear. Here, a water control experiment was conducted to study the physiological and transcriptomic response of R. delavayi to drought. The drought treatment for 9 days decreased photosynthetic rate, induced accumulation of reactive oxygen species (ROS), and damaged chloroplast ultrastructure of R. delavayi. However, the photosynthetic rate quickly recovered to the level before treatment when the plants were re-watered. De novo assembly of RNA-Seq data generated 86,855 unigenes with an average length of 1870 bp. A total of 22,728 differentially expressed genes (DEGs) were identified between the control and drought plants. The expression of most DEGs related to photosynthesis were down-regulated during drought stress, and were up-regulated when the plants were re-watered, including the DEGs encoding subunits of light-harvesting chlorophyll-protein complex, photosystem II and photosystem I reaction center pigment-protein complexes, and photosynthetic electron transport. The expressions of many DEGs related to signal transduction, flavonoid biosynthesis and antioxidant activity were also significantly affected by drought stress. The results indicated that the response of R. delavayi to drought involved multiple physiological processes and metabolic pathways. Photosynthetic adjustment, ROS-scavenging system, abscisic acid and brassinosteroid signal transduction pathway may play important roles to improve drought tolerance of R. delavayi. Our findings provided valuable information for understanding the mechanisms of drought tolerance employed by Rhododendron species.

Members of the genus Paphiopedilum are world-famous for their large, colourful flowers, unique floral morphology and long floral lifespan. Most Paphiopedilum species bloom in spring or autumn. The control of flowering time is of great significance to the commercial production of floral crops, because it affects the sales and prices of flowers. However, the mechanism that regulates when Paphiopedilum species bloom is unclear. In the present study, floral bud initiation and development of P. micranthum (spring-flowering species with one flower per stalk), P. dianthum (autumn-flowering species with multiple flowers per stalk) and P. henryanum (autumn-flowering species with one flower per stalk) were investigated by morphological and anatomical methods. We divided Paphiopedilum floral bud differentiation into six phases: the initiation of differentiation, inflorescence primordium differentiation, flower primordium differentiation, sepal primordium differentiation, petal primordium differentiation and column primordium differentiation. We found that the timing of floral bud differentiation for the three species was synchronized when experiencing the same environment, while the period from initiation to flowering largely differed. In addition, initiation of floral bud differentiation in P. dianthum was earlier at a warmer environment. The difference in flowering time of three species was mainly caused by the duration of floral bud development, rather than the initiation time. Background The genus Rhododendron L. has been widely cultivated for hundreds of years around the world. Members of this genus are known for great ornamental and medicinal value. Owing to advances in sequencing technology, genomes and transcriptomes of members of the Rhododendron genus have been sequenced and published by various laboratories. With increasing amounts of omics data available, a centralized platform is necessary for effective storage, analysis, and integration of these large-scale datasets to ensure consistency, independence, and maintainability. Results Here, we report our development of the Rhododendron Plant Genome Database (RPGD; http://bioinfor.kib.ac.cn/RPGD/), which represents the first comprehensive database of Rhododendron genomics information. It includes large amounts of omics data, including genome sequence assemblies for R. delavayi, R. williamsianum, and R. simsii, gene expression profiles derived from public RNA-Seq data, functional annotations, gene families, transcription factor identification, gene homology, simple sequence repeats, and chloroplast genome. Additionally, many useful tools, including BLAST, JBrowse, Orthologous Groups, Genome Synteny Browser, Flanking Sequence Finder, Expression Heatmap, and Batch Download were integrated into the platform. Conclusions RPGD is designed to be a comprehensive and helpful platform for all Rhododendron researchers. Believe that RPGD will be an indispensable hub for Rhododendron studies.

In the present study, we studied the role of chloroplastic ATP synthase in photosynthetic regulation during leaf maturation. We measured gas exchange, chlorophyll fluorescence, P700 redox state, and the electrochromic shift signal in mature and immature leaves. Under high light, the immature leaves displayed high levels of non-photochemical quenching (NPQ) and P700 oxidation ratio, and higher values for proton motive force (pmf) and proton gradient (ΔpH) across the thylakoid membranes but lower values for the activity of chloroplastic ATP synthase (gH+) than the mature leaves. Furthermore, gH+ was significantly and positively correlated with CO2 assimilation rate and linear electron flow (LEF), but negatively correlated with pmf and ΔpH. ΔpH was significantly correlated with LEF and the P700 oxidation ratio. These results indicated that gH+ was regulated to match photosynthetic capacity during leaf maturation, and the formation of pmf and ΔpH was predominantly regulated by the alterations in gH+. In the immature leaves, the high steady-state ΔpH increased lumen acidification, which, in turn, stimulated photoprotection for the photosynthetic apparatus via NPQ induction and photosynthetic control. Our results highlighted the importance of chloroplastic ATP synthase in optimizing the trade-off between CO2 assimilation and photoprotection during leaf maturation.

The proton motive force (pmf) across the thylakoid membranes plays a key role for photosynthesis in fluctuating light. However, the mechanisms underlying the regulation of pmf in fluctuating light are not well known. In this study, we aimed to identify the roles of chloroplastic ATP synthase and cyclic electron flow (CEF) around photosystem I (PSI) in the regulation of the pmf in fluctuating light. To do this, we measured chlorophyll fluorescence, P700 parameters, and the electrochromic shift signal in the fluctuating light alternating between 918 (high light) and 89 (low light) μmol photons m-2 s-1 every 5 min. We found that the activity of chloroplastic ATP synthase (gH+), pmf, CEF activity, non-photochemical quenching (NPQ), and the P700 redox state changed rapidly in fluctuating light. During transition from low to high light, the decreased gH+ and the stimulation of CEF both contributed to the rapid formation of pmf, activating NPQ and optimizing the redox state of P700 in PSI. During the low-light phases, gH+ rapidly increased and the pmf declined sharply, leading to the relaxation of NPQ and down-regulation of photosynthetic control. These findings indicate that in fluctuating light the gH+ and CEF are finely regulated to modulate the pmf formation, avoiding the over-accumulation of reactive intermediates and maximizing energy use efficiency.

Background To primarily evaluate the effects and safety of a selective angiotensin II type 1 (AT1) receptor blocker (ARB) allisartan isoproxil combined with amlodipine or indapamide in the treatment of patients with essential hypertension who failed allisartan monotherapy. Methods Patients aged 18–75 years with mild-to-moderate essential hypertension [office systolic blood pressure (SBP) 140 to Results A total of 2,212 patients were enrolled, among them 2,126 patients were included in the efficacy analysis, with an average age of 55.1 ± 10.2 years. A total of 1,463 cases (68.8%) were effective after 4 weeks allisartan treatment, and the mean SBP and DBP were significantly decreased by 14.7 ± 12.2 and 8.0 ± 8.4 mm Hg compared with the baseline levels (all P < 0.001). In nonachievers, allisartan combined with indapamide for 8 weeks significantly lowered the sitting BP (SBP/DBP) by 14.0 ± 12.2/8.3 ± 9.2 mm Hg, respectively, compared with 4 weeks monotherapy with allisartan with a BP targeting rate of 57.7% (169/293). In the allisartan + amlodipine group, the SBP/DBP were significantly decreased by (14.4 ± 12.1/8.2 ± 8.2) mm Hg, respectively, with a BP targeting rate of 62.8% (181/288). There was no statistical significance in BP reduction, targeting rate, or adverse reactions between the 2 combined therapies. Conclusions Allisartan isoproxil combined with indapamide or amlodipine can further improve the BP targeting rate when allisartan monotherapy failed in essential hypertension. The 2 combined therapies have similar efficacy and safety.

Lhasa, the capital of Tibet, is located on the Tibetan Plateau. Accelerated economic development and flourishing tourism resulting from the opening of the Qinghai-Tibet Railway (QTR) have increased solid waste generation and contamination in recent years. Using data from Lhasa Statistical Yearbooks and previous studies, this study estimates the future population of permanent residents and tourists using the least squares method to extrapolate the population from 2015-2025, and evaluates the effects of the QTR on municipal solid waste (MSW) generation in Lhasa and estimates future MSW generation. There were approximately 1.35 million tourists in 2008 when the QTR had been operating for 2 years and MSW generation was approximately 470 tons per day. The amount of MSW generated increased dramatically with time after opening the QTR. This study estimates that MSW generation will reach 962 tons per day in 2025. Due to the existence of the QTR, increasing numbers of people are traveling to Lhasa, and tourism has driven the development of the local economy. During the studies, the proportion of MSW produced by tourists increased from 2.99% to 20.06%, and it is estimated that it will increase to 33.49% in 2025. If the current trend continues, Lhasa will face significant challenges from garbage disposal. This study analyzes the current situation of urban garbage treatment in Lhasa, and it suggests several options for improvement to MSW generation, transportation equipment, disposal, and resource recycling.

Photosystem I (PSI) is the primary target of photoinhibition under fluctuating light (FL). In angiosperms, cyclic electron flow (CEF) around PSI is thought to be the main player protecting PSI under FL. The activation of CEF is linked to the thylakoid stacking, which is in turn affected by light intensity. However, it is unknown how pre-illumination affects the CEF activation and PSI redox state under FL. To address this question, we conducted a spectroscopic analysis under FL in Bletilla striata and Morus alba after pre-illumination at moderate light (ML, 611 μmol photons m−2 s−1) or high light (HL, 1455 μmol photons m−2 s−1). Our results indicated that both species displayed a transient over-reduction of PSI after a transition from low to high light, but the extent of PSI over-reduction under FL was largely alleviated by pre-illumination at HL when compared with pre-illumination under ML. Furthermore, pre-illumination at HL accelerated the activation rate of CEF but did not affect the activation of non-photochemical quenching and linear electron flow from photosystem II under FL. Therefore, such increased CEF activity by pre-illumination under HL alleviated PSI over-reduction under FL by facilitating the electron sink downstream of PSI. Taking together, pre-illumination at HL protects PSI under FL through acceptor-side regulation.

Patrinia scabiosaefolia Fisch. (PSF), a well-known traditional Chinese medicine, has been demonstrated to show therapeutic effects on inflammatory bowel disease. In this study, a rapid and sensitive method using ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF-MS) was developed for identification of the major constituents in PSF. The separation analysis was performed on Waters Acquity UPLC system, and the accurate mass of molecules and their fragment ions were determined by Q-TOF-MS. Thirty-one constituents, including triterpenoids, iridoids, flavonoids, and organic acids were detected and tentatively deduced on the basis of their element compositions, tandem mass spectrometry (MS/MS) data, and relevant literatures. Twelve constituents were discovered for the first time in PSF. The results demonstrated that hederagenin-type and oleanolic acid-type saponins were the main constituents of PSF. Nine resorcinol derivatives including two new ones, 5-[(8Z,11Z,14Z)-nonadeca−8,11,14-trienyl] resorcinol (1) and 5-[(8Z,11Z,14E)-heptadeca-8,11,14-trienyl] resorcinol (2), were isolated from the leaves of Syzygium samarangense. The new structures were elucidated by means of extensive spectroscopic techniques including interpretation of 1D and 2D NMR spectra. Among them, compounds 3, 4, 6 and 7 exhibited significant α-glucosidase inhibitory activities with IC50 of 3.16, 3.16, 2.34 and 0.99 µM, respectively. This finding provides evidence that resorcinol derivatives with long aliphatic chain function as new promising antidiabetic alternatives.

How to characterize short protein sequences to make an effective connection to their functions is an unsolved problem. Here we propose to map the physicochemical properties of each amino acid onto unit spheres so that each protein sequence can be represented quantitatively. We demonstrate the usefulness of this representation by applying it to the prediction of cell penetrating peptides. We show that its combination with traditional composition features yields the best performance across different datasets, among several methods compared. For the convenience of users, a web server has been established for automatic calculations of the proposed features at http://biophy.dzu.edu.cn/SNumD/.

The water-water cycle (WWC) is thought to dissipate excess excitation energy and balance the ATP/NADPH energy budget under some conditions. However, the importance of the WWC in photosynthetic regulation remains controversy. We observed that three Camellia cultivars exhibited high rates of photosynthetic electron flow under high light when photosynthesis was restricted. We thus tested the hypothesis that the WWC is a major electron sink in the three Camellia cultivars when CO2 assimilation is restricted. Light response curves indicated that the WWC was strongly increased with photorespiration and was positively correlated with extra ATP supplied from other flexible mechanisms excluding linear electron flow, implying that the WWC is an important alternative electron sink to balance ATP/NADPH energy demand for sustaining photorespiration in Camellia cultivars. Interestingly, when photosynthesis was depressed by the decreases in stomatal and mesophyll conductance, the rates of photosynthetic electron flow through photosystem II declined slightly and the rates of WWC was enhanced. Furthermore, the increased electron flow of WWC was positively correlated with the ratio of Rubisco oxygenation to carboxylation, supporting the involvement of alternative electron flow in balancing the ATP/NADPH energy budget. We propose that the WWC is a crucial electron sink to regulate ATP/NADPH energy budget and dissipate excess energy excitation in Camellia species when CO2 assimilation is restricted.
