Abstract Diabetes mellitus, a significant global public health challenge, severely impacts human health worldwide. The organoid, an innovative in vitro three-dimensional (3D) culture model, closely mimics tissues or organs in vivo. Insulin-secreting islet organoid, derived from stem cells induced in vitro with 3D structures, has emerged as a potential alternative for islet transplantation and as a possible disease model that mirrors the human body’s in vivo environment, eliminating species difference. This technology has gained considerable attention for its potential in diabetes treatment. Despite advances, the process of stem cell differentiation into islet organoid and its cultivation demonstrates deficiencies, prompting ongoing efforts to develop more efficient differentiation protocols and 3D biomimetic materials. At present, the constructed islet organoid exhibit limitations in their composition, structure, and functionality when compared to natural islets. Consequently, further research is imperative to achieve a multi-tissue system composition and improved insulin secretion functionality in islet organoid, while addressing transplantation-related safety concerns, such as tumorigenicity, immune rejection, infection, and thrombosis. This review delves into the methodologies and strategies for constructing the islet organoid, its application in diabetes treatment, and the pivotal scientific challenges within organoid research, offering fresh perspectives for a deeper understanding of diabetes pathogenesis and the development of therapeutic interventions.

Abstract Chemoresistance remains a significant challenge for effective breast cancer treatment which leads to cancer recurrence. CRISPR-directed gene editing becomes a powerful tool to reduce chemoresistance by reprogramming the tumor microenvironment. Previous research has revealed that Chinese herbal extracts have significant potential to overcome tumor chemoresistance. However, the therapeutic efficacy is often limited due to their poor tumor targeting and in vivo durability. Here we have developed a tumor microenvironment responsive nanoplatform (H-MnO2(ISL + DOX)-PTPN2@HA, M(I + D)PH) for nano-herb and CRISPR codelivery to reduce chemoresistance. Synergistic tumor inhibitory effects were achieved by the treatment of isoliquiritigenin (ISL) with doxorubicin (DOX), which were enhanced by CRISPR-based gene editing to target protein tyrosine phosphatase non-receptor type 2 (PTPN2) to initiate long-term immunotherapy. Efficient PTPN2 depletion was observed after treatment with M(I + D)PH nanoparticles, which resulted in the recruitment of intratumoral infiltrating lymphocytes and an increase of proinflammatory cytokines in the tumor tissue. Overall, our nanoparticle platform provides a diverse technique for accomplishing synergistic chemotherapy and immunotherapy, which offers an effective treatment alternative for malignant neoplasms. BackgroundStroke is the leading cause of acquired disability and the second leading cause of death worldwide. Its rate of incidence, disability, mortality, and recurrence is high, and the patients experience various symptoms of discomfort, which not only affect their rehabilitation function but also reduce their ability to perform daily activities and their quality of life. Nowadays, with the improvement of China’s medical standards, patients are increasingly attentive to their quality of life and health status. However, diagnostic techniques and effective treatments for patients with stroke are still limited but urgently required. ObjectiveThis study aimed to evaluate the quality of life during hospitalization using a stroke patient-reported outcomes (PROs) scale and additionally to recognize potential factors and risk indicators that may impact recurrent events, facilitating early intervention measures. MethodsThis is a registry-based, retrospective observational cross-sectional study on patients with stroke. A convenient sampling method was used to select various indicators of patients. The Stroke-PRO scale was then used to assess patients’ conditions across physical, psychological, social, and therapeutic domains. Multiple linear regression analysis was applied to identify factors influencing stroke PROs, while correlation analysis was conducted to explore the relationship between these outcomes and blood lipid levels. This study investigated the potential of gamma-aminobutyric acid (GABA), a potent bioactive compound and inhibitory neurotransmitter, in mitigating chronic ammonia stress-induced liver toxicity in crucian carp (Carassius carassius). GABA is known for its role in alleviating stress responses and enhancing antioxidant defense. In this study, experimental fishes (weighing 20.51 ± 0.39 g) underwent 56 days of chronic ammonia stress (10 ± 0.053 mg/L), with or without supplemented with GABA (100 mg/kg) in the diet. Serum ammonia (152.14 ± 2.94, CK-95.82 ± 0.81 μmol/L), serum MDA content (3.41 ± 1.24, CK-1.25 ± 0.08 μmol/L), and liver MDA content (1.98 ± 1.72, CK-0.84 ± 0.02 μmol/g prot) were significantly increased in the ammonia exposure group compared with the CK group. GABA supplementation effectively alleviates the detrimental effects of chronic ammonia stress, specifically on growth parameters, serum ammonia levels, and malondialdehyde content. GABA supplementation elevated the expressions of antioxidant genes (zu/cu sod, cat), promoted the expressions of nrf2, and reduced the expression of keap1. Moreover, GABA significantly alleviated liver injury and reduced gene expression of three types of transmembrane proteins (perk, ire1, and atf6). GABA further regulated the expression of downstream apoptotic target genes (upregulation of bcl2; downregulation of bax, caspase3, and caspase9) through endoplasmic reticulum stress (ERS) pathways. This finding indicated the multifaceted benefits of GABA in combating chronic ammonia stress in crucian carp, providing valuable insights for the development of sustainable aquaculture practices.

Abstract Ratios of glycerol dialkyl glycerol tetraethers (GDGT), which are membrane lipids of bacteria and archaea, are at the base of several paleoenvironmental proxies. They are frequently applied to soils as well as lake‐ and marine sediments to generate records of past temperature and soil pH. To derive meaningful environmental information from these reconstructions, high analytical reproducibility is required. Based on submitted results by 39 laboratories from across the world, which employ a diverse range of analytical and quantification methods, we explored the reproducibility of brGDGT‐based proxies (MBT′5ME, IR, and #ringstetra) measured on four soil samples and four soil lipid extracts. Correct identification and integration of 5‐ and 6‐methyl brGDGTs is a prerequisite for the robust calculation of proxy values, but this can be challenging as indicated by the large inter‐interlaboratory variation. The exclusion of statistical outliers improves the reproducibility, where the remaining uncertainty translates into a temperature offset from median proxy values of 0.3–0.9°C and a pH offset of 0.05–0.3. There is no apparent systematic impact of the extraction method and sample preparation steps on the brGDGT ratios. Although reported GDGT concentrations are generally consistent within laboratories, they vary greatly between laboratories. This large variability in brGDGT quantification may relate to variations in ionization efficiency or specific mass spectrometer settings possibly impacting the response of brGDGTs masses relative to that of the internal standard used. While ratio values of GDGT are generally comparable, quantities can currently not be compared between laboratories.

With the ongoing advancements in porous asphalt pavement and the recycling of reclaimed asphalt pavement (RAP), there is a need to develop a new class of high-performance epoxy asphalt (EA) to enhance the performance of the porous structure and facilitate the efficient utilization of RAP. To achieve this objective, polyethylene glycol (PEG) with number-average molecular weights (Mn) of 200, 400, 800, and 1000 g/mol were individually incorporated into epoxy asphalt. The critical performance of different PEG-modified EAs in comparison with high-viscosity asphalt or commercial EA was investigated for PAM containing RAP. The results indicate that the addition of PEG leads to higher initial viscosity and extended construction time for EA with aged asphalt from RAP. The EA with higher Mn PEG causes a slight reduction in high-temperature performance for PAM containing RAP. However, it mitigates the deformability, resistance to water damage, long-term friction stability, and anti-strip performance deterioration induced by RAP. PEG-modified EA imparts excellent skid resistance and surface friction performance to RAP porous mixtures. Overall, PEG-modified EA porous structures containing RAP exhibit superior workability and toughness compared to commercial EA, and higher strength and road performance compared to high-viscosity asphalt. In terms of one-way ANOVA, Mn significantly influenced various key physical and functional parameters of EA mixtures with RAP. EA modified with PEG having Mn in the range of 800–1000 g/mol is recommended as a binder for porous structures containing RAP. The research is highly relevant towards the exploratory design and applications of thermosetting PEG-modified EA in the porous mixture and waste concrete.

[INLINE:1] Activated G-protein-coupled receptor 39 (GPR39) has been shown to attenuate inflammation by interacting with sirtuin 1 (SIRT1) and peroxisome proliferator-activated receptor-γ coactivator 1α (PGC-1α). However, whether GPR39 attenuates neuropathic pain remains unclear. In this study, we established a Sprague-Dawley rat model of spared nerve injury-induced neuropathic pain and found that GPR39 expression was significantly decreased in neurons and microglia in the spinal dorsal horn compared with sham-operated rats. Intrathecal injection of TC-G 1008, a specific agonist of GPR39, significantly alleviated mechanical allodynia in the rats with spared nerve injury, improved spinal cord mitochondrial biogenesis, and alleviated neuroinflammation. These changes were abolished by GPR39 small interfering RNA (siRNA), Ex-527 (SIRT1 inhibitor), and PGC-1α siRNA. Taken together, these findings show that GPR39 activation ameliorates mechanical allodynia by activating the SIRT1/PGC-1α pathway in rats with spared nerve injury.

Composition design of high-entropy carbides is a topic of great scientific interest for the hot-end parts in the aerospace field. A novel theoretical method through an inverse composition design route, i.e. initially ensuring the oxide scale with excellent anti-ablation stability, is proposed to improve the ablation resistance of the high-entropy carbide coatings. In this work, the (Hf0.36Zr0.24Ti0.1Sc0.1Y0.1La0.1)C1-δ (HEC) coatings were prepared by the inverse design concept and verified by the ablation resistance experiment. The linear ablation rate of the HEC coatings is −1.45 ​μm/s, only 4.78 % of the pristine HfC coatings after the oxyacetylene ablation at 4.18 ​MW/m2. The HEC possesses higher toughness with a higher Pugh's ratio of 1.55 in comparison with HfC (1.30). The in-situ formed dense (Hf0.36Zr0.24Ti0.1Sc0.1Y0.1La0.1)O2-δ oxide scale during ablation benefits to improve the anti-ablation performance attributed to its high structural adaptability with a lattice constant change not exceeding 0.19 % at 2000–2300 ​°C. The current investigation demonstrates the effectiveness of the inverse theoretical design, providing a novel optimization approach for ablation protection of high-entropy carbide coatings.

This study was to demonstrate that the alleviate effect of synbiotics (Bacillus subtilis and xylo-oligosaccharide) on NaHCO3-induced intestinal barrier dysfunction and autophagy of crucian carp (Carassius auratus). In a 2×2 factorial design, crucian carp were divided into 4 groups, fed with diets contained two levels of synbiotics (0 and 1× 108 CFU/g) and treated with two levels of NaHCO3 (0 and 50 mmol/L) for 56 days. Thermosetting resins have advantages such as high strength, corrosion resistance, and aging resistance, and have excellent prospects for practical application as asphalt modifiers. In order to promote the research of thermosetting resin modified asphalt, to provide direction for its further research, this paper reviews the research progress of thermosetting resin modified asphalt in recent years. The material composition, modification mechanism, and curing behavior of epoxy asphalt, thermosetting polyurethane modified asphalt, unsaturated polyester modified asphalt, and other thermosetting resin modified asphalts are overviewed. Different types of thermosetting resin modified asphalt have different performances, the performance advantages of different thermosetting resin modified asphalts are summarized. At the same time, the existing problems in thermosetting resin modified asphalt and further research directions are provided. Encouraging researchers to produce thermosetting resin modified asphalts using waste or bio-based materials, and to study the recycling technologies and life cycle assessment of thermosetting resin modified asphalt. This paper provides a reference for the study of thermosetting resin modified asphalt.

The ratio of leaf carotenoid to chlorophyll (Car/Chl) is an indicator of vegetation photosynthesis, development and responses to stress. However, the correlation between Car and Chl, and their overlapping absorption in the visible spectral domain pose a challenge for optical remote sensing of their ratio. This study aims to investigate combinations of vegetation indices (VIs) to minimize the influence of Car-Chl correlation, thus being more sensitive to the variability in the ratio across vegetation species and sites. VIs sensitive to Car and Chl variability were combined into four candidates of combinations, using a simulated dataset from the PROSPECT model. The VI combinations were then tested using six simulated datasets with different Car-Chl correlations, and evaluated against four independent datasets. The ratio of the carotenoid triangle ratio index (CTRI) with the red-edge chlorophyll index (CIred-edge) was found least influenced by the Car-Chl correlation and demonstrated a superior ability for estimating Car/Chl variability. Compared with published VIs and two machine learning algorithms, CTRI/CIred-edge also showed the optimal performance in the four field datasets. This new VI combination could be useful to provide insights in spatiotemporal variability in the leaf Car/Chl ratio, applicable for assessing vegetation physiology, phenology, and response to environmental stress. Abstract Background Ankyrin repeat domain 49 (ANKRD49) has been found to be highly expressed in multiple cancer including lung adenocarcinoma (LUAD) and lung squamous carcinoma (LUSC). However, the function of ANKRD49 in the pathogenesis of NSCLC still remains elusive. Previously, ANKRD49 has been demonstrated to promote the invasion and metastasis of A549 cells, a LUAD cell line, via activating the p38-ATF-2-MMP2/MMP9 pathways. Considering the heterogeneity of tumor cells, the function and mechanism of ANKRD49 in NSCLC need more NSCLC-originated cells to clarify. Methods Real-time qPCR was employed to test ANKRD49 expression levels in nine pairs of fresh NSCLC tissues and the corresponding adjacent normal tissues. The function of ANKRD49 was investigated using overexpression and RNA interference assays in lung adenocarcinoma cell line (NCI-H1299) and lung squamous carcinoma cell line (NCI-H1703) through gelatin zymography, cell counting kit-8, colony formation, wound healing, migration and invasion assays mmunoprecipitation was performed to in vitro. Immunoprecipitation was performed to test the interaction of c-Jun and ATF2. Chromatin immunoprecipitation was conducted to assess the transcriptional regulation of ATF2/c-Jun on MMP-2/9. Moreover, the tumorigenicity of ANKRD49 was evaluated in nude mice models and the involved signal molecular was also measured by immunohistochemical method. Results We found that the levels of ANKRD49 in cancerous tissues were higher than those in adjacent normal tissues. in vitro assay showed that ANKRD49 promoted the migration and invasion of NCI-H1299 and NCI-H1703 cells via enhancing the levels of MMP-2 and MMP-9. Furthermore, ANKRD49 elevated phosphorylation of JNK and then activated c-Jun and ATF2 which interact in nucleus to promote the binding of ATF2:c-Jun with the promoter MMP-2 or MMP-9. In vivo assay showed that ANKRD49 promoted lung metastasis of injected-NSCLC cells and the high metastatic rate was positively correlated with the high expression of ANKRD49, MMP-2, MMP-9, p-JNK, p-c-Jun and p-ATF2. Conclusion The present study indicated that ANKRD49 accelerated the invasion and metastasis of NSCLC cells via JNK-mediated transcription activation of c-Jun and ATF2 which regulated the expression of MMP-2/MMP-9. The molecular mechanisms of ANKRD49’s function is different from those found in A549 cells. The current study is a supplement and improvement to the previous research.

Hepatic stellate cells (HSCs) represent a significant component of hepatocellular carcinoma (HCC) microenvironments which play a critical role in tumor progression and drug resistance. Tumor-on-a-chip technology has provided a powerful in vitro platform to investigate the crosstalk between activated HSCs and HCC cells by mimicking physiological architecture with precise spatiotemporal control. Here we developed a tri-cell culture microfluidic chip to evaluate the impact of HSCs on HCC progression. On-chip analysis revealed activated HSCs contributed to endothelial invasion, HCC drug resistance and natural killer (NK) cell exhaustion. Cytokine array and RNA sequencing analysis were combined to indicate the iron-binding protein LIPOCALIN-2 (LCN-2) as a key factor in remodeling tumor microenvironments in the HCC-on-a-chip. LCN-2 targeted therapy demonstrated robust anti-tumor effects both in vitro 3D biomimetic chip and in vivo mouse model, including angiogenesis inhibition, sorafenib sensitivity promotion and NK-cell cytotoxicity enhancement. Taken together, the microfluidic platform exhibited obvious advantages in mimicking functional characteristics of tumor microenvironments and developing targeted therapies.

In order to study the characteristics of metabolites in Monascus cheese, Monascus cheese BC20 and Monascus cheese ZX-99 were used as experimental groups, and commercially available blue mold cheese and white mold cheese were used as control groups. The low molecular weight metabolites involved in the four cheeses were analyzed by non-targeted metabolomics. Metabolites that were only enriched in the experimental groups (VIP>1，P2) and had no significant difference between the experimental groups (VIP≤1，P≥0.05) were considered as the characteristics of Monascus cheese. The results showed that among the 2510 metabolites identified in positive and negative ion modes, 23 substances were preliminarily considered as characteristic metabolites of Monascus cheese. Eight of these substances, including cucurbitacin e, fusidic acid, quinapril, methyl dopa, orcinol, licoricesaponin h2, alfuzosin, and 25-desacetyl rifapentin had certain beneficial activities. Through comparison between the experimental groups and control groups, there were 10 identical differential metabolic pathways. The enrichment degree of the protein digestion and absorption pathway was relatively high, and the proportion of pathways related to amino acids was the largest. Abstract Background There is a lack of research regarding the relationship between creatinine to cystatin C to waist circumference ratio (CCR/WC ratios) and the development of type 2 diabetes mellitus (T2DM). We aimed to evaluate the association between CCR/WC ratios and incident T2DM in Chinese adults. Methods This prospective study was from the China Health and Retirement Longitudinal Study (2011, 2013, 2015, and 2018). The participants were divided into three groups by tertiaries of the CCR/WC ratios. Cox proportional‐hazards models were used to identify the relationship between CCR/WC and T2DM. Results Overall, 5938 participants were included for analysis, 766 of whom developed T2DM between 2011 and 2018. Risk of incident T2DM was decreased with tertiaries 2, 3 versus tertiary 1 of the CCR/WC index (adjusted hazard ratio [HR] 0.772 [95% confidence interval 0.647–0.921] and 0.724 [0.596–0.880], p for trend = .001 across tertiaries of the CCR/WC index). The results were consistent excluding participants with T2DM in the first 2 years. Conclusions This study demonstrated that CCR/WC was negatively correlated with the risk of T2DM in Chinese adults. Early detection is necessary to control the development of T2DM in Chinese with low CCR/WC levels.

Chronic pancreatitis (CP) is a progressive and irreversible fibroinflammatory disorder, accompanied by pancreatic exocrine insufficiency and dysregulated gut microbiota. Recently, accumulating evidence has supported a correlation between gut dysbiosis and CP development. However, whether gut microbiota dysbiosis contributes to CP pathogenesis remains unclear. Herein, an experimental CP was induced by repeated high-dose caerulein injections. The broad-spectrum antibiotics (ABX) and ABX targeting Gram-positive (G+) or Gram-negative bacteria (G–) were applied to explore the specific roles of these bacteria. Gut dysbiosis was observed in both mice and in CP patients, which was accompanied by a sharply reduced abundance for short-chain fatty acids (SCFAs)-producers, especially G+ bacteria. Broad-spectrum ABX exacerbated the severity of CP, as evidenced by aggravated pancreatic fibrosis and gut dysbiosis, especially the depletion of SCFAs-producing G+ bacteria. Additionally, depletion of SCFAs-producing G+ bacteria rather than G– bacteria intensified CP progression independent of TLR4, which was attenuated by supplementation with exogenous SCFAs. Finally, SCFAs modulated pancreatic fibrosis through inhibition of macrophage infiltration and M2 phenotype switching. The study supports a critical role for SCFAs-producing G+ bacteria in CP. Therefore, modulation of dietary-derived SCFAs or G+ SCFAs-producing bacteria may be considered a novel interventive approach for the management of CP.

The objective of this study was to compare the quantitative radiomics data between malignant mixed Müllerian tumors (MMMTs) and endometrial carcinoma (EC) and identify texture features associated with overall survival (OS). This study included 61 patients (36 with EC and 25 with MMMTs) and analyzed various radiomic features and gray-level co-occurrence matrix (GLCM) features. These variables and patient clinicopathologic characteristics were compared between EC and MMMTs using the Wilcoxon Rank sum and Fisher’s exact test. The area under the curve of the receiving operating characteristics (AUC ROC) was calculated for univariate analysis in predicting EC status. Logistic regression with elastic net regularization was performed for texture feature selection. This study showed that skewness (p = 0.045) and tumor volume (p = 0.007) significantly differed between EC and MMMTs. The range of cluster shade, the angular variance of cluster shade, and the range of the sum of squares variance were significant predictors of EC status (p ≤ 0.05). The regularized Cox regression analysis identified the “256 Angular Variance of Energy” texture feature as significantly associated with OS independently of the EC/MMMT grouping (p = 0.004). The volume and texture features of the tumor region may help distinguish between EC and MMMTs and predict patient outcomes.

Summary: Orexin is a neuropeptide released from hypothalamus regulating feeding, sleeping, arousal, and cardiovascular activity. Past research has demonstrated that orexin receptor 2 (OX2R) agonist infusion in the brain results in sympathoexcitatory responses. Here, we found that epicardial administration of OX2R agonism leads to opposite responses. We proved that OX2R is expressed mainly in DRG neurons and transported to sensory nerve endings innervating the heart. In a capsaicin-induced cardiac sympathetic afferent reflex (CSAR) model, we recorded the calcium influx in DRG neurons, measured heart rate variability, and examined the PVN c-Fos activity to prove that epicardial OX2R agonism administration could attenuate capsaicin-induced CSAR. We further showed that OX2R agonism could partially rescue acute myocardial infarction by reducing sympathetic overactivation. Our data indicate that epicardial application of OX2R agonist exerts a cardioprotective effect by attenuating CSAR. This OX2R-mediated heart-brain axis may provide therapeutic targets for acute cardiovascular diseases.

The three-dimensional (3D) plant chlorophyll distribution plays an essential role in plant physiological and ecological analysis. The hyperspectral LiDAR (HSL) technology, as a new active technology, has the advantage of directly and simultaneously obtaining plant hyperspectral and 3D spatial information. It provides an alternative opportunity for fast and nondestructive 3D plant chlorophyll estimation. Current estimation models are developed by statistical relationships between HSL spectra and plant chlorophyll content, which perform limited robustness and lack physical mechanisms. The leaf-canopy radiative transfer models (RTMs) could describe physical responses between spectra and chlorophyll content, and show great robustness from optical hyperspectral data, which have the potential to overcome the limitation of current estimation models. However, it is unclear whether leaf-canopy RTMs could be applied to HSL spectra for 3D chlorophyll mapping. Therefore, this study explored the potential of a leaf-canopy RTM with HSL spectra for 3D plant chlorophyll estimation. Firstly, an optimal leaf RTM was selected from a series of PROSEPCT models, and determined by estimating leaf-scale chlorophyll content across multi-species and multi-state leaves. Then, the 4SAIL model, a canopy RTM, was optimized to meet the active detection mechanism of the HSL technology. Next, coupling the best PROSPECT with optimized 4SAIL models was used as the leaf-canopy RTM. The 3D chlorophyll distribution was estimated by the leaf-canopy RTM with a lookup table (LUT) algorithm under six cost functions. Finally, the performance of 14 vegetation indice (VI) models was tested as a comparison. Results demonstrated that the PROSPECT-5 model was the best leaf RTM with the best R2 of 0.60. The combination of PROSPECT-5 and optimized 4SAIL model could successfully achieve 3D plant chlorophyll estimation. This leaf-canopy estimation RTM with the best cost function (Least square estimator) had the best accuracy with an R2 of 0.73. Its accuracy was the same as the median accuracy of the best VI model (MTCI model), and higher than that of other VI models. The estimated and actual 3D chlorophyll distributions were consistent. The application of the leaf-canopy RTM with HSL technology offers an alternative approach to plant physiological and ecological research.

Within this context, the dual-specificity tyrosine phosphorylation-regulated kinase 3 (DYRK3) has emerged as a potential key player with implications for prognosis and tumor progression. Methods. Transabdominal ultrasound is a promising imaging modality for pancreatic cystic lesions. This study aims to determine if transabdominal ultrasonography with CT fusion (TAUS-f) using volume navigation can be used to measure pancreatic cystic lesions (PCLs) compared to CT alone. We evaluated 33 patients prospectively with known PCLs. The readers evaluated each PCL’s size and imaging characteristics on TAUS-f and CT alone. These were compared to endoscopic ultrasonography reports. A total of 43 PCLs from 32 patients were evaluated. The detection rate by TAUS-f was 93%. Two of the three undetected PCLs were in the tail of the pancreas. Inter-reader variabilities for TAUS-f and CT were 0.005 cm and 0.03 cm, respectively. Subgroup analysis by size and location demonstrated that inter-modality variability between TAUS-f and CT was smallest for lesions < 1.5 cm with a size difference of −0.13 cm for each reader and smallest in the pancreatic head with a size difference of −0.16 cm and −0.17 cm for readers 1 and 2. We found that TAUS-f effectively evaluates PCLs compared to CT alone, thus suggesting that it should be considered part of the surveillance algorithm for a subset of patients.

Enzyme-driven micro/nanomotors consuming in situ chemical fuels have attracted lots of attention for biomedical applications. However, motor systems composed by organism-derived organics that maximize the therapeutic efficacy of enzymatic products remain challenging. Herein, swimming proteomotors based on biocompatible urease and human serum albumin are constructed for enhanced antitumor therapy via active motion and ammonia amplification. By decomposing urea into carbon dioxide and ammonia, the designed proteomotors are endowed with self-propulsive capability, which leads to improved internalization and enhanced penetration in vitro. As a glutamine synthetase inhibitor, the loaded l-methionine sulfoximine further prevents the conversion of toxic ammonia into non-toxic glutamine in both tumor and stromal cells, resulting in local ammonia amplification. After intravesical instillation, the proteomotors achieve longer bladder retention and thus significantly inhibit the growth of orthotopic bladder tumor in vivo without adverse effects. We envision that the as-developed swimming proteomotors with amplification of the product toxicity may be a potential platform for active cancer treatment.

We present a review of in situ observations regarding the interactions between tropical cyclones and the ocean in the western North Pacific for the year 2023. A total of at least 13 tropical cyclones occurred during this period. According to the Japan Meteorological Agency, Typhoon Mawar recorded the yearly minimum pressure at 900 hPar. On average, each tropical cyclone captured 7.4 surface drifters and 25.2 Argo floats when the search radius is 300 km. During Guchol, the maximum in situ Lagrangian current reached 1.23 m/s, with sustained wind speeds of the tropical cyclone up to 31.7 m/s and a relative position of 174 km. Additionally, several Argo floats were active during tropical cyclones, with maximum sea surface temperature cooling reaching 0.66 °C. This annual review provides a comprehensive summary of the current state of in situ observations regarding tropical cyclone–ocean interaction. These findings serve as valuable references for both scientific research and operational forecasting.

ABSTRACTPrecise classification of Light Detection and Ranging (LiDAR) point cloud is a fundamental process in various applications, such as land cover mapping, forestry management, and autonomous driving. Due to the lack of spectral information, the existing research on single wavelength LiDAR classification is limited. Spectral information from images could address this limitation, but data fusion suffers from varying illumination conditions and the registration problem. A novel multispectral LiDAR successfully obtains spatial and spectral information as a brand-new data type, namely, multispectral point cloud, thereby improving classification performance. However, spatial and spectral information of multispectral LiDAR has been processed separately in previous studies, thereby possibly limiting the classification performance of multispectral LiDAR. To explore the potential of this new data type, the current spatial–spectral classification framework for multispectral LiDAR that includes four steps: (1) neighborhood selection, (2) feature extraction and selection, (3) classification, and (4) label smoothing. Three novel highlights were proposed in this spatial – spectral classification framework. (1) We improved the popular eigen entropy-based neighborhood selection by spectral angle match to extract a more precise neighborhood. (2) We evaluated the importance of geometric and spectral features to compare their contributions and selected the most important features to reduce feature redundancy. (3) We conducted spatial label smoothing by a conditional random field, accounting for the spatial and spectral information of the neighborhood points. The proposed method demonstrated by a multispectral LiDAR with three channels: 466 nm (blue), 527 nm (green), and 628 nm (red). Experimental results demonstrate the effectiveness of the proposed spatial – spectral classification framework. Moreover, this research takes advantages of the complementation of spatial and spectral information, which could benefit more precise neighborhood selection, more effective features, and satisfactory refinement of classification result. Finally, this study could serve as an inspiration for future efficient spatial–spectral process for multispectral point cloud.

Abstract Background Both low‐carbohydrate (LC) and calorie-restricted (CR) diets have been shown to have metabolic benefits. However, the two regimens have yet to be thoroughly compared. We conducted a 12-week randomized trial to compare the effects of these diets separately and in combination on both weight loss and metabolic risk factors in overweight/obese individuals. Methods A total of 302 participants were randomized to LC diet (n = 76), CR diet (n = 75), LC + CR diet (n = 76), or normal control (NC) diet (n = 75) using a computer-based random number generator. The primary outcome was the change in body mass index (BMI). The secondary outcomes included body weight, waist circumference, waist-to-hip ratio, body fat, and metabolic risk factors. All participants attended health education sessions during the trial. Results A total of 298 participants were analyzed. BMI change over 12 weeks was − 0.6 (95% CI, − 0.8 to − 0.3) kg/m2 in NC, − 1.3 (95% CI, − 1.5 to − 1.1) kg/m2 in CR, − 2.3 (95% CI, − 2.6 to − 2.1) kg/m2 in LC, and − 2.9 (95% CI, − 3.2 to − 2.6) kg/m2 in LC + CR. Abstract Cholangiocarcinoma (CCA) is an aggressive solid tumour with a 5-year survival rate ranging from 7% to 20%. It is, therefore, urgent to identify novel biomarkers and therapeutic targets to improve the outcomes of patients with CCA. SPRY-domain containing protein 4 (SPRYD4) contains SPRY domains that modulate protein–protein interaction in various biological processes; however, its role in cancer development is insufficiently explored. This study is the first to identify that SPRYD4 is downregulated in CCA tissues using multiple public datasets and a CCA cohort. Furthermore, the low expression of SPRYD4 was significantly associated with unfavourable clinicopathological characteristics and poor prognosis in patients with CCA, indicating that SPRYD4 could be a prognosis indicator of CCA. In vitro experiments revealed that SPRYD4 overexpression inhibited CCA cells proliferation and migration, whereas the proliferative and migratory capacity of CCA cells was enhanced after SPRYD4 deletion. Moreover, flow cytometry showed that SPRYD4 overexpression triggered the S/G2 cell phase arrest and promoted apoptosis in CCA cells. Furthermore, the tumour-inhibitory effect of SPRYD4 was validated in vivo using xenograft mouse models. SPRYD4 also showed a close association with tumour-infiltrating lymphocytes and important immune checkpoints including PD1, PD-L1 and CTLA4 in CCA. In conclusion, this study elucidated the role of SPRYD4 during CCA development and highlighted SPRYD4 as a novel biomarker and tumour suppressor in CCA.

Leaf functional traits are key indicators of plant functions useful for inferring complex plant processes, including their responses to environmental changes. Vegetation indices (VIs) composed of a few reflectance wavelengths hold the advantages of being relatively simple and effective and have been widely used within remote sensing to estimate leaf traits. However, the difference between the reflectance from the upper and lower part of the leaf suggests that leaf reflectance mainly provides one-sided information, constraining its ability to estimate leaf functional traits. Leaf transmittance, on the other hand, gives information about the whole leaf and has more potential to be sensitive to changes in leaf biochemistry. As transmittance-based VI is rare, this study aims to propose new transmittance-based VIs for accurate estimations of leaf traits. Three forms, i.e. the normalized difference VI, the simple ratio VI, and the difference VI were employed, and wavelength selection for transmittance-based and reflectance-based VIs were conducted, respectively. The applicability of these VIs for estimating four leaf functional traits (leaf chlorophyll (Cab), leaf carotenoids (Car), equivalent water thickness (EWT), and leaf mass per area (LMA)) were evaluated. Cross-validation using three datasets of field observations and sensitivity analysis showed that the VIs constructed using transmittance were relatively less affected by interferences from other leaf parameters, improving the estimation accuracy of Car, EWT, and LMA compared to their optimal reflectance counterparts (RMSE reduced by 2% to 15%, and MAE reduced by 7% to 20% for the pooled dataset). Our study revealed that the normalized difference VI based on transmittance showed considerable sensitivity to Car, EWT, and LMA, whereas the difference VI based on reflectance was effective in indicating Cab. The proposed transmittance-based VIs will aid remote monitoring of leaf traits and thereby plant adaptations and acclimation to changes in environmental conditions.

Leaf mass per area (LMA) and equivalent water thickness (EWT) are key indicators providing information on plant growth status and agricultural management, and their retrieval is commonly done through radiative transfer models (RTMs) such as the PROSPECT model. However, the PROSPECT model is frequently hampered by the ill-posed problem as a consequence of measurement and model uncertainties. Here, we propose a wavelength selection method to improve the inversion of EWT and LMA by integrating PROSPECT with a machine learning algorithm (Gaussian process regression (GPR); PROSPECT-GPR for short). The GPR model conducted sorting of wavelengths and the PROSPECT-D was used to determine the optimal number of characteristic wavelengths. The results demonstrated that the estimation of EWT (R2 = 0.80; RMSE = 0.0021) and LMA (R2 = 0.71; RMSE = 0.0021) using the proposed wavelengths and PROSPECT inversion all exhibited superior accuracy in comparison with those from previous studies. The efficacy of PROSPECT-GPR in exploring the spectral linkage among vegetation traits was demonstrated by selecting wavelengths associated with leaf structure parameter N and EWT (1368 nm) that turn out to contribute to the estimation of LMA. The findings lay a strong foundation for understanding the spectral linkage among vegetation traits, and the proposed wavelength selection method provides valuable insights for selecting informative spectral wavelengths for RTMs inversion and designing future remote sensors.

Abstract Sepsis is a highly heterogeneous syndrome normally characterized by bacterial infection and dysregulated systemic inflammatory response that leads to multiple organ failure and death. Single anti‐inflammation or anti‐infection treatment exhibits limited survival benefit for severe cases. Here a biodegradable tobramycin‐loaded magnesium micromotor (Mg‐Tob motor) is successfully developed as a potential hydrogen generator and active antibiotic deliverer for synergistic therapy of sepsis. The peritoneal fluid of septic mouse provides an applicable space for Mg‐water reaction. Hydrogen generated sustainably and controllably from the motor interface propels the motion to achieve active drug delivery along with attenuating hyperinflammation. The developed Mg‐Tob motor demonstrates efficient protection from anti‐inflammatory and antibacterial activity both in vitro and in vivo. Importantly, it prevents multiple organ failure and significantly improves the survival rate up to 87.5% in a high‐grade sepsis model with no survival, whereas only about half of mice survive with the individual therapies. This micromotor displays the superior therapeutic effect of synergistic hydrogen‐chemical therapy against sepsis, thus holding great promise to be an innovative and translational drug delivery system to treat sepsis or other inflammation‐related diseases in the near future.

Early prediction of neoadjuvant systemic therapy (NAST) response for triple-negative breast cancer (TNBC) patients could help oncologists select individualized treatment and avoid toxic effects associated with ineffective therapy in patients unlikely to achieve pathologic complete response (pCR). The objective of this study is to evaluate the performance of radiomic features of the peritumoral and tumoral regions from dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) acquired at different time points of NAST for early treatment response prediction in TNBC. This study included 163 Stage I-III patients with TNBC undergoing NAST as part of a prospective clinical trial (NCT02276443). Peritumoral and tumoral regions of interest were segmented on DCE images at baseline (BL) and after two (C2) and four (C4) cycles of NAST. Ten first-order (FO) radiomic features and 300 gray-level-co-occurrence matrix (GLCM) features were calculated. Area under the receiver operating characteristic curve (AUC) and Wilcoxon rank sum test were used to determine the most predictive features. Multivariate logistic regression models were used for performance assessment. Pearson correlation was used to assess intrareader and interreader variability. Seventy-eight patients (48%) had pCR (52 training, 26 testing), and 85 (52%) had non-pCR (57 training, 28 testing). Forty-six radiomic features had AUC at least 0.70, and 13 multivariate models had AUC at least 0.75 for training and testing sets. The Pearson correlation showed significant correlation between readers. In conclusion, Radiomic features from DCE-MRI are useful for differentiating pCR and non-pCR. Similarly, predictive radiomic models based on these features can improve early noninvasive treatment response prediction in TNBC patients undergoing NAST.

Introduction:Cynanchum auriculatum (CA) is usually used to treat digestive disorders, such as anorexia, enteritis, dysentery, and indigestion. Functional dyspepsia (FD) is characterized by a group of symptoms associated with the gastroduodenal region. Recent pharmacological studies have demonstrated the efficacy of CA for treating FD. However, the pharmacokinetics (PK) and tissue distribution of CA in physiological and FD states is still unclear. The present study aimed to clarify the differences in PK parameters and tissue distribution of the four major active components of CA (baishouwu benzophenone, deacylmet-aplexigenin, qingyangshengenin, and syringic acid) under both physiological and FD states.Methods: For this, normal and FD rats were orally administered 10 mg/kg CA extract. Then, plasma and tissue (heart, liver, spleen, lung, kidney, brain, stomach, and small intestine) samples were obtained. The four active components of CA in rat plasma and tissues were quantified by developing and validating a fast and reliable ultra–high–performance liquid chromatography–mass spectrometry method.Results: The area under the plasma concentration–time curve from time zero to time t (AUC0-t) of baishouwu benzophenone was significantly lower in the FD group than in the normal group (p < 0.01). The FD group had significantly lower (p < 0.001) apparent volume of distribution and plasma clearance of qing-yangshengenin and significantly higher (p < 0.05) AUC0-t of deacylmetaplexigenin and qingyangshengenin. The four active components were rapidly distributed into various tissues, and the main target organs of CA activity were the stomach and small intestine. In addition, baishouwu benzophenone, deacylmetaplexigenin, and qingyangshengenin could cross the blood-brain barrier, indicating that the brain may be another target organ in the treatment of FD.Discussion: These results indicate that the pathological state of FD alters the PK behavior and tissue distribution characteristics of baishouwu benzophenone, deacylmetaplexigenin, qingyangshengenin, and syringic acid in the CA extract, providing an experimental basis for the role of CA in FD treatment.

Polymer-derived ultra-high-temperature ceramic (UHTC) nanocomposites have attracted growing attention due to the increasing demands for advanced thermal structure components in aerospace. Herein, hafnium carbide (HfC) whiskers are successfully fabricated in carbon fiber preforms via the polymer-derived ceramic (PDC) method. A novel carbon nanotube (CNT) template growth mechanism combined with the PDC method is proposed in this work, which is different from the conventional vapor–liquid–solid (VLS) mechanism that is commonly used for polymer-derived nanostructured ceramics. The CNTs are synthesized and proved to be the templates for fabricating the HfC whiskers, which are generated by the released low-molecular-weight gas such as CO, CO2, and CH4 during the pyrolysis of a Hf-containing precursor. The formed products are composed of inner single crystal HfC whiskers that are measured to be several tens of micrometers in length and 100–200 nm in diameter and outer HfC/HfO2 particles. Our work not only proposes a new strategy to prepare the HfC whiskers, but also puts forward a new thinking of the efficient utilization of a UHTC polymer precursor.

Abstract Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer. Neoadjuvant systemic therapy (NAST) followed by surgery are currently standard of care for TNBC with 50-60% of patients achieving pathologic complete response (pCR). We investigated ability of deep learning (DL) on dynamic contrast enhanced (DCE) MRI and diffusion weighted imaging acquired early during NAST to predict TNBC patients’ pCR status in the breast. During the development phase using the images of 130 TNBC patients, the DL model achieved areas under the receiver operating characteristic curves (AUCs) of 0.97 ± 0.04 and 0.82 ± 0.10 for the training and the validation, respectively. The model achieved an AUC of 0.86 ± 0.03 when evaluated in the independent testing group of 32 patients. In an additional prospective blinded testing group of 48 patients, the model achieved an AUC of 0.83 ± 0.02. These results demonstrated that DL based on multiparametric MRI can potentially differentiate TNBC patients with pCR or non-pCR in the breast early during NAST.

Nitric oxide (NO)/cyclic guanosine 3′,5′-monophosphate (cGMP) signaling has been shown to act as a mediator involved in pain transmission and processing. In this review, we summarize and discuss the mechanisms of the NO/cGMP signaling pathway involved in chronic pain, including neuropathic pain, bone cancer pain, inflammatory pain, and morphine tolerance. The main process in the NO/cGMP signaling pathway in cells involves NO activating soluble guanylate cyclase, which leads to subsequent production of cGMP. cGMP then activates cGMP-dependent protein kinase (PKG), resulting in the activation of multiple targets such as the opening of ATP-sensitive K+ channels. The activation of NO/cGMP signaling in the spinal cord evidently induces upregulation of downstream molecules, as well as reactive astrogliosis and microglial polarization which participate in the process of chronic pain. In dorsal root ganglion neurons, natriuretic peptide binds to particulate guanylyl cyclase, generating and further activating the cGMP/PKG pathway, and it also contributes to the development of chronic pain. Upregulation of multiple receptors is involved in activation of the NO/cGMP signaling pathway in various pain models. Notably the NO/cGMP signaling pathway induces expression of downstream effectors, exerting both algesic and analgesic effects in neuropathic pain and inflammatory pain. These findings suggest that activation of NO/cGMP signaling plays a constituent role in the development of chronic pain, and this signaling pathway with dual effects is an interesting and promising target for chronic pain therapy.

Antimony selenide (Sb2Se3) semiconducting material possesses a band gap of 1.05–1.2 eV and has been widely applied in single-junction solar cells. Based on its band gap, Sb2Se3 can also be used as the bottom cell absorber material in tandem solar cells. More importantly, Sb2Se3 solar cells exhibit excellent stability with nontoxic compositional elements. The band gap of organic–inorganic hybrid perovskite is tunable over a wide range. In this work, we demonstrate for the first time a perovskite/antimony selenide four-terminal tandem solar cell with a specially designed and fabricated transparent electrode for an optimized spectral response. By adjusting the thickness of the transparent electrode layer of the top cell, the wide-band-gap perovskite top solar cell achieves an efficiency of 17.88%, while the optimized antimony selenide bottom cell delivers a power conversion efficiency of 7.85% by introducing a double electron transport layer. Finally, the four-terminal tandem solar cell achieves an impressive efficiency exceeding 20%. This work provides a new tandem device structure and demonstrates that antimony selenide is a promising absorber material for bottom cell applications in tandem solar cells.

Corn silk (Zea mays L.) is the stigma of an annual gramineous plant named corn, which is distributed in many regions worldwide and has a long history of medicinal use. In recent years, with the sustainable development of traditional Chinese medicine, studies of corn silk based on modern technologies, such as GC–MS, LC–MS, and other analytical means, have offered more comprehensive analyses. Phytochemistry studies have shown that the main bioactive components in corn silk include flavonoids, polyphenols, phenolic acids, fatty acids, and terpenoids. Pharmacological studies have shown that corn silk extract has various pharmacological effects, such as reducing blood lipids, lowering blood pressure, regulating blood sugar levels, anti-inflammatory effects, and anti-oxidation effects. In this paper, the related research on corn silk from the past few years is summarized to provide a theoretical reference for the further development and utilization of corn silk.

Suspicious non-calcified mammographic findings have not been evaluated with modern mammographic technique, and the purpose of this work is to compare the likelihood of malignancy for those findings. To do this, 5018 consecutive mammographically guided biopsies performed during 2016–2019 at a large metropolitan, community-based hospital system were retrospectively reviewed. In total, 4396 were excluded for targeting calcifications, insufficient follow-up, or missing data. Thirty-seven of 126 masses (29.4%) were malignant, 44 of 194 asymmetries (22.7%) were malignant, and 77 of 302 architectural distortions (AD, 25.5%) were malignant. The combined likelihood of malignancy was 25.4%. Older age was associated with a higher likelihood of malignancy for each imaging finding type (all p ≤ 0.006), and a possible ultrasound correlation was associated with a higher likelihood of malignancy when all findings were considered together (p = 0.012). Two-view asymmetries were more frequently malignant than one-view asymmetries (p = 0.03). There were two false-negative biopsies (98.7% sensitivity and 100% specificity). In conclusion, the 25.4% likelihood of malignancy confirms the recommendation for biopsy of suspicious, ultrasound-occult, mammographic findings. Mammographically guided biopsies were highly sensitive and specific in this study. Older patient age and a possible ultrasound correlation should raise concern given the increased likelihood of malignancy in those scenarios.

Leaf area index (LAI) is an important biophysical parameter of vegetation and serves as a significant indicator for assessing forest ecosystems. Multi-source remote sensing data enables large-scale and dynamic surface observations, providing effective data for quantifying various indices in forest and evaluating ecosystem changes. However, employing single-source remote sensing spectral or LiDAR waveform data poses limitations for LAI inversion, making the integration of multi-source remote sensing data a trend. Currently, the fusion of active and passive remote sensing data for LAI inversion primarily relies on empirical models, which are mainly constructed based on field measurements and do not provide a good explanation of the fusion mechanism. In this study, we aimed to estimate LAI based on physical model using both spectral imagery and LiDAR waveform, exploring whether data fusion improved the accuracy of LAI inversion. Specifically, based on the physical model geometric-optical and radiative transfer (GORT), a fusion strategy was designed for LAI inversion. To ensure inversion accuracy, we enhanced the data processing by introducing a constraint-based EM waveform decomposition method. Considering the spatial heterogeneity of canopy/ground reflectivity ratio in regional forests, calculation strategy was proposed to improve this parameter in inversion model. The results showed that the constraint-based EM waveform decomposition method improved the decomposition accuracy with an average 12% reduction in RMSE, yielding more accurate waveform energy parameters. The proposed calculation strategy for the canopy/ground reflectivity ratio, considering dynamic variation of parameter, effectively enhanced previous research that relied on a fixed value, thereby improving the inversion accuracy that increasing on the correlation by 5% to 10% and on R2 by 62.5% to 132.1%. Based on the inversion strategy we proposed, data fusion could effectively be used for LAI inversion. The inversion accuracy achieved using both spectral and LiDAR data (correlation=0.81, R2 = 0.65, RMSE=1.01) surpassed that of using spectral data or LiDAR alone. This study provides a new inversion strategy for large-scale and high-precision LAI inversion, supporting the field of LAI research.

Liquid polymeric precursors have found wide applications as a matrix source for continuous fiber reinforced ceramic matrix composites (CMCs) because of their excellent fluidity and high ceramic yield. In the present study, a processable liquid single-source-precursor was synthesized using allylhydropolycarbosilane (AHPCS) and low-cost hafnium tetrachloride (HfCl4) as raw materials to form final SiC-HfC-C nanocomposites by well-known polymer-derived ceramic (PDC) approach. The precursor synthesis, thermal behavior, polymer-to-ceramic transformation and ceramic phase evolution were investigated by FT-IR, TGA, XRD and TEM. The TGA results show that the obtained single-source-precursor exhibits very high ceramic yields of over 80.0 wt% after heat treated at 1400 °C. After annealing at 1600 °C, the resultant SiC-HfC-C nanocomposites contain SiC@C and HfC@C nanoparticles with SiC and HfC as cores and amorphous carbon as shells. The present single-source-precursor is candidate material for CMC applications due to its liquidity and high ceramic yield.

The lateral hypothalamus' orexinergic system has been associated with anxiety-related behaviors, and electroacupuncture (EA) modifies orexin neurons to control the anti-anxiety process. However, in a rat model of post-traumatic stress disorder (PTSD), the important role of LH orexin neurons (OXNs) in the anxiolytic effects induced by EA has not been explored. In this study, rats underwent modified single prolonged stress (MSPS) for seven days before developing EA. The rats were then subjected to elevated plus maze (EPM) and open field (OFT) tests, and western blot and c-Fos/orexin double labeling investigations were carried out to determine the functional activation of LH orexinergic neurons. It is difficult to use a single edge operator in image processing to extract continuous and accurate contours of a porous bioelastomer due to the fuzzy boundary and complex background in ultrasound images. To solve this problem, this paper proposes a joint algorithm for bioelastomer contour detection and a texture feature extraction method for monitoring the degradation performance of bioelastomers. First, the mean-shift clustering method is utilized to obtain the clustering feature information of bioelastomers and native tissue from manually segmented images, and this information is used as the initial information in the image binarization algorithm for image partitioning. Second, Otsu's thresholding method and mathematical morphology are applied in the process of image binarization. Finally, the Canny edge detector is employed to extract the complete bioelastomers contour from the binary image. To verify the robustness of the proposed joint algorithm, the results using the proposed joint algorithm, where mean-shift clustering is replaced with k-means clustering are also obtained. The proposed joint algorithm based on mean-shift clustering outperforms the joint algorithm based on k-means clustering, as well as algorithms that directly apply the Canny, Sobel and Laplacian methods. Texture feature extraction is based on the computer-aided recognition of bioelastomers. The region of interest (ROI) is set in the scaffold region, and the first-order statistical features and second-order statistical features of the greyscale values of the ROI are extracted and analysed. The proposed joint algorithm can not only extract ideal bioelastomers contours from ultrasound images but also provide valuable feedback on the degradation behaviour of bioelastomers at implant sites.

Background: Recent research has shown that the adhesion G protein-coupled receptor F1 (Adgrf1; also known as GPR110; PGR19; KPG_012; hGPCR36) is an oncogene. The evidence is mainly based on high expression of Adgrf1 in numerous cancer types, and knockdown Adgrf1 can reduce the cell migration, invasion, and proliferation. Adgrf1 is, however, mostly expressed in the liver of healthy individuals. The function of Adgrf1 in liver has not been revealed. Interestingly, expression level of hepatic Adgrf1 is dramatically decreased in obese subjects. Here, the research examined whether Adgrf1 has a role in liver metabolism. Methods: We used recombinant adeno-associated virus-mediated gene delivery system, and antisense oligonucleotide was used to manipulate the hepatic Adgrf1 expression level in diet-induced obese mice to investigate the role of Adgrf1 in hepatic steatosis. The clinical relevance was examined using transcriptome profiling and archived biopsy specimens of liver tissues from non-alcoholic fatty liver disease (NAFLD) patients with different degree of fatty liver. Results: The expression of Adgrf1 in the liver was directly correlated to fat content in the livers of both obese mice and NAFLD patients. Stearoyl-coA desaturase 1 (Scd1), a crucial enzyme in hepatic de novo lipogenesis, was identified as a downstream target of Adgrf1 by RNA-sequencing analysis. Treatment with the liver-specific Scd1 inhibitor MK8245 and specific shRNAs against Scd1 in primary hepatocytes improved the hepatic steatosis of Adgrf1-overexpressing mice and lipid profile of hepatocytes, respectively. Conclusions: These results indicate Adgrf1 regulates hepatic lipid metabolism through controlling the expression of Scd1. Downregulation of Adgrf1 expression can potentially serve as a protective mechanism to stop the overaccumulation of fat in the liver in obese subjects. Overall, the above findings not only reveal a new mechanism regulating the progression of NAFLD, but also proposed a novel therapeutic approach to combat NAFLD by targeting Adgrf1. Sedimentary organic matter provides carbon substrates and energy sources for microorganisms, which drive benthic biogeochemical processes and in turn modify the quantity and quality of dissolved organic matter (DOM). However, the molecular composition and distribution of DOM and its interactions with microbes in deep-sea sediments remain poorly understood. Here, molecular composition of DOM and its relationship with microbes were analyzed in samples collected from two sediment cores (∼40 cm below the sea floor), at depths of 1157 and 2253 m from the South China Sea. Results show that niche differentiation was observed on a fine scale in different sediment layers, with Proteobacteria and Nitrososphaeria dominating the shallow sediments (0–6 cm) and Chloroflexi and Bathyarchaeia prevailing in deeper sediments (6–40 cm), indicating correspondence of microbial community composition with both geographical isolation and the availability of organic matter. An intimate link between the DOM composition and microbial community further indicates that, microbial mineralization of fresh organic matter in the shallow layer potentially resulted in the accumulation of recalcitrant DOM (RDOM), while relatively low abundance of RDOM was linked to anaerobic microbial utilization in deeper sediment layers. In addition, higher RDOM abundance in the overlying water, as compared to that in the surface sediment, suggests that sediment might be a source of deep-sea RDOM. These results emphasize the close relation between the distribution of sediment DOM and different microbial community, laying a foundation for understanding the complex dynamics of RDOM in deep-sea sediment and water column.

Based on core, well-logging and seismic data, the gravity flow characteristics and sedimentary model of the 1st member of the Tengger formation in the eastern steep slope belt of the Sai-4 tectonic belt in the Saihantala depression (Erlian Basin, China) have been systematically studied; further, the sublacustrine fan sedimentary model in the study area has been established. The main types of gravity flow identified in the study area are sandy debris flow deposits and turbidity flow deposition. Affected by paleo-geomorphology, provenance and other factors, sublacustrine fans in the eastern steep slope zone are formed by gravity flows from different periods. The sublacustrine fan is divided into three subfacies: inner-fan, mid-fan and outer-fan. The subfacies can be subdivided into six types of microfacies: main channel, levee of main channel, distributary channel, inter-channel, levee of distributary channel and terminals. The formation of the sublacustrine fan is caused by steep slopes and triggered by earthquakes. The sedimentary model of the sublacustrine fan and its sedimentary characteristics provide a foundation for exploring and developing oil and gas in the study area.

Highlights Multidiscipline application of triboelectric nanogenerators (TENGs) for intelligent Internet of Things (IoTs) are summarized from the aspects of agriculture, industry, city, emergency monitoring, and artificial intelligence. Perspectives on the challenges and future research directions of TENGs in IoTs have been proposed.

Abstract Copper-induced death, also termed cuproptosis, is a novel form of programmed cell death and is promising as a new strategy for cancer therapeutics. Elevated copper levels in tumor cells are positively associated with high PD-L1 expression. Nonetheless, the prognostic significance of cuproptosis-related immune checkpoint genes (CRICGs) in hepatocellular carcinoma remains to be further clarified. This study aimed to construct the prognostic CRICG signature to predict the immunotherapy response and outcomes of HCC patients. The co-expressed CRICGs were first screened through Pearson correlation analysis. Based on the least absolute shrinkage and selection operator-COX regression analyses, we identified a prognostic 5-CRICGs model, which closely correlates with poor outcomes, cancer development, and immune response to hepatocellular carcinoma. External validation was conducted using the GSE14520 dataset. Lastly, qRT-PCR was performed to determine the expression of the CRICGs in HCC. In summary, we developed and validated a novel prognostic CRICG model based on 5 CRICGs. This prognostic signature could effectively forecast the outcomes and immune response of HCC patients, which may serve as biomarkers for anticancer therapy.
