Abstract Transportation systems primarily depend on vehicular flow on roads. Developed countries have shifted towards automated signal control, which manages and updates signal synchronisation automatically. In contrast, traffic in underdeveloped countries is mainly governed by manual traffic light systems. These existing manual systems lead to numerous issues, wasting substantial resources such as time, energy, and fuel, as they cannot make real‐time decisions. In this work, we propose an algorithm to determine traffic signal durations based on real‐time vehicle density, obtained from live closed circuit television camera feeds adjacent to traffic signals. The algorithm automates the traffic light system, making decisions based on vehicle density and employing Faster R‐CNN for vehicle detection. Additionally, we have created a local dataset from live streams of Punjab Safe City cameras in collaboration with the local police authority. The proposed algorithm achieves a class accuracy of 96.6% and a vehicle detection accuracy of 95.7%. Across both day and night modes, our proposed method maintains an average precision, recall, F1 score, and vehicle detection accuracy of 0.94, 0.98, 0.96 and 0.95, respectively. Our proposed work surpasses all evaluation metrics compared to state‐of‐the‐art methodologies.

Abstract Background Spatial molecular profiling depends on accurate cell segmentation. Identification and quantitation of individual cells in dense tissues, e.g. highly inflamed tissue caused by viral infection or immune reaction, remains a challenge. Methods We first assess the performance of 18 deep learning-based cell segmentation models, either pre-trained or trained by us using two public image sets, on a set of immunofluorescence images stained with immune cell surface markers in skin tissue obtained during human herpes simplex virus (HSV) infection. We then further train eight of these models using up to 10,000+ training instances from the current image set. Finally, we seek to improve performance by tuning parameters of the most successful method from the previous step. Results The best model before fine-tuning achieves a mean Average Precision (mAP) of 0.516. Prediction performance improves substantially after training. The best model is the cyto model from Cellpose. After training, it achieves an mAP of 0.694; with further parameter tuning, the mAP reaches 0.711. Conclusion Selecting the best model among the existing approaches and further training the model with images of interest produce the most gain in prediction performance. The performance of the resulting model compares favorably to human performance. The imperfection of the final model performance can be attributed to the moderate signal-to-noise ratio in the imageset.

Abstract This paper focuses on radio resource management (RRM) in multi-user dual-function radar communication (DFRC) systems using orthogonal frequency division multiplexing (OFDM) waveforms. We propose two RRM schemes, one from the perspective of sum rate maximization and the other from the perspective of user fairness maximization. These optimization problems are non-convex due to the presence of mixed integer terms, making them difficult to solve. To address these challenges, we have employed a decomposition approach to transform these two complex problems into separate, more readily solvable ones. In addressing the sum rate maximization problem, we initially introduce a heuristic greedy algorithm to obtain a resource allocation scheme that satisfies radar performance requirements. Subsequently, we utilize a cyclic iterative method along with KKT conditions to solve the sum rate maximization problem for communication users. Concerning the fairness maximization problem for communication users, we similarly employ a heuristic greedy algorithm to obtain a resource allocation scheme that meets radar performance constraints. Then utilize the Lagrangian dual method to solve the multi-user fairness maximization problem for communication users. Our experimental results confirm the effectiveness of the proposed algorithms.

BackgroundHigh-intensity chemotherapy can cause life-threatening complications in pediatric patients. Therefore, this study investigated safety and efficacy of long-acting pegylated recombinant human granulocyte colony-stimulating factor (PEG-rhG-CSF; Jinyouli®) in children undergoing high-intensity chemotherapy.MethodsTreatment-naive patients received post-chemotherapy PEG-rhG-CSF as primary prophylaxis for two cycles. The primary endpoints were drug-related adverse events (AEs) and bone pain scores. Secondary endpoints included grade 3–4 neutropenia, duration of neutropenia recovery, absolute neutrophil count changes, febrile neutropenia (FN), reduced chemotherapy intensity, antibiotic usage, and AE severity. The cost-effectiveness of PEG-rhG-CSF was compared with that of rhG-CSF (Ruibai®).ResultsHere, 307 and 288 patients underwent one and two PEG-rhG-CSF cycles, respectively. Ninety-one patients experienced drug-related AEs, primarily bone pain (12.7%). Moreover, Grade 3–4 neutropenia and FN were observed. Median FN durations were 3.0 days in both cycles. No drug-related delays were observed during chemotherapy. One patient experienced grade 4 neutropenia-induced reduction in chemotherapy intensity during cycle 2. In total, 138 patients received antibiotics. PEG-rhG-CSF exhibited superior cost-effectiveness compared to rhG-CSF.ConclusionOur findings indicate that PEG-rhG-CSF is safe, efficient, and cost-effective in pediatric patients undergoing high-intensity chemotherapy, providing preliminary evidence warranting further randomized controlled trials.

Summary: Background: The combined vincristine, pegylated liposomal doxorubicin (PLD), and cyclophosphamide (VPC) regimen has never been studied in pediatric patients. Methods: This open-label, single-center, single-arm phase I study utilizing a “3 + 3” design enrolled children with relapsed/refractory (R/R) solid tumors. Three dose levels of PLD (Duomeisu®) were studied (30, 40, or 50 mg/m2) in combination with cyclophosphamide (1500 mg/m2), mesna (1500 mg/m2), and vincristine (1.5 mg/m2, maximum 2 mg) once every 3 weeks. The primary endpoints included safety, the maximum tolerated dose (MTD) of PLD (Duomeisu®), and the recommended phase 2 dose (RP2D) of PLD (Duomeisu®) for further phase 2 investigation. The secondary endpoints were objective response rate (ORR) and disease control rate (DCR). This study is registered with ClinicalTrials.gov, NCT04213612. Findings: Between January 7, 2020, and November 18, 2021, 34 patients were eligible and evaluable for toxicity, while 26 patients were evaluable for response. The MTD of PLD (Duomeisu®) was 30 mg/m2. The most common adverse event (AE) was grade 3 or 4 neutropenia (61.8%). The most common grade 1 or 2 non-hematologic AE and cardiotoxicity effects were vomiting (35.3%) and abnormal electrocardiogram T waves (20.6%), respectively. ORR and DCR to VPC regimen after two cycles were 50.0% and 92.3%, respectively. Targeted gene panel sequencing revealed the activation of TP53 mutation may be an adverse prognostic factor. Interpretation: The VPC regimen showed a promising safety profile and had preliminary efficacy in children with R/R solid tumors. The RP2D for PLD (Duomeisu®) combined with cyclophosphamide and vincristine is 30 mg/m2 once every 3 weeks. Abstract Polyimides externally deployed in spacecraft or satellites extensively have various aerospace hazards, including atomic oxygen (AO) erosion, irradiation degradation, and electrostatic charge/discharge (ESC/ESD). To cope with these challenges, we fabricate a ZnO/CuNi‐polyimide composite film with augmented permanence. Using spectroscopy and microscopy techniques, we have shown that the combination of chelation and cross‐linking in the interfacial architecture leads to enhanced interfacial compatibility and mechanical robustness. Besides, due to the positive AO diffusion barrier ability of the wurtzite ZnO, our composite film shows remarkable AO resistance and a very small Ey value of 6.88 × 10−26 cm3/atom, which is merely 2.29% of that of pristine polyimide. Moreover, the well‐defined nanocrystalline state with minimal lattice swelling (0.3%–0.7%) of the Fe+‐irradiated ZnO/CuNi‐polyimide at a damaging dose of 353.4 dpa demonstrates its excellent irradiation resistance. Finally, the ZnO/CuNi‐polyimide also shows sufficient electrostatic dissipation capacity to cope with the ESC/ESD events. Our fabrication approach for composite films based on multi‐technology integration shows potential for aerospace applications and deployment.

Abstract Nowadays, silver nanoparticles (AgNPs) have attracted the attention of many researchers due to their special physical, chemical, and biological properties. There is strong evidence that biogenic AgNPs can act as potent anticancer agents through the production of reactive oxygen species (ROS) and initiate the mitochondrial pathway of apoptosis. That is why we decided to use Nepeta bracteata Benth flower extract for the first time to bio-synthesize AgNPs and study their cytotoxic and apoptotic effects on SK-BR-3 cells. AgNPs were biosynthesized at 70 °C after mixing silver nitrate and flower extract with a specific ratio and concentration, then were characterized using various analytical techniques, such as FESEM, FTIR, EDS, and zeta potential. Studies have shown that AgNPs have an irregular and circular shape, with about 99% by weight of silver, carbon, and oxygen. On the other hand, the appropriate size (below 57 nm) and surface charge (− 11.52 mV) make them stable in biological fluids. The better cytotoxic effect of AgNPs compared to flower extract on SK-BR-3 cells was investigated using the MTT method. The positive effect of AgNPs on inhibiting the growth of SK-BR-3 breast cancer cells was again confirmed by the sulforhodamine B staining method, so that AgNPs were able to decrease the density of cancer cells in a concentration-dependent manner. In addition, the flow cytometry test proved that biosynthesized AgNPs using Nepeta bracteata Benth flower extract can induce apoptosis in SK-BR-3 cancer cells. Real-time PCR then proved that the ratio of Bak1/Bclx, as well as caspase-3 expression, was increased due to active ROS-producing biomolecules present in the plant extract, and therefore, AgNPs can activate the mitochondria-dependent apoptosis pathway in breast cancer cells. Finally, their negligible oxidative stress on erythrocytes was confirmed by the lipid peroxidation method and showed that biosynthesized AgNPs can be used for breast cancer treatment without showing adverse effects on erythrocytes.

Abstract MYCN amplification is an independent poor prognostic factor in patients with high-risk neuroblastoma (NB). Further exploring the molecular regulatory mechanisms in MYCN-amplified NB will help to develop novel therapy targets. In this study, methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) was identified as the differentially expressed gene (DEG) highly expressed in MYCN-amplified NB, and it showed a positive correlation with MYCN and was associated with a poor prognosis of NB patients. Knockdown of MTHFD1 inhibited proliferation and migration, and induced apoptosis of NB cells in vitro. Mouse model experiments validated the tumorigenic effect of MTHFD1 in NB in vivo. In terms of the mechanism, ChIP-qPCR and dual-luciferase reporter assays demonstrated that MTHFD1 was directly activated by MYCN at the transcriptional level. As an important enzyme in the folic acid metabolism pathway, MTHFD1 maintained the NADPH redox homeostasis in MYCN-amplified NB. Knockdown of MTHFD1 reduced cellular NADPH/NADP+ and GSH/GSSG ratios, increased cellular reactive oxygen species (ROS) and triggered the apoptosis of NB cells. Moreover, genetic knockdown of MTHFD1 or application of the anti-folic acid metabolism drug methotrexate (MTX) potentiated the anti-tumor effect of JQ1 both in vitro and in vivo. Taken together, MTHFD1 as an oncogene is a potential therapeutic target for MYCN-amplified NB. The combination of MTX with JQ1 is of important clinical translational significance for the treatment of patients with MYCN-amplified NB.

Abstract Radiative cooling is a zero-energy technology that enables subambient cooling by emitting heat into outer space (~3 K) through the atmospheric transparent windows. However, existing designs typically focus only on the main atmospheric transparent window (8–13 μm) and ignore another window (16–25 μm), under-exploiting their cooling potential. Here, we show a dual-selective radiative cooling design based on a scalable thermal emitter, which exhibits selective emission in both atmospheric transparent windows and reflection in the remaining mid-infrared and solar wavebands. As a result, the dual-selective thermal emitter exhibits an ultrahigh subambient cooling capacity (~9 °C) under strong sunlight, surpassing existing typical thermal emitters (≥3 °C cooler) and commercial counterparts (as building materials). Furthermore, the dual-selective sample also exhibits high weather resistance and color compatibility, indicating a high practicality. This work provides a scalable and practical radiative cooling design for sustainable thermal management.

Hypergraph neural networks have gained substantial popularity in capturing complex correlations between data items in multimodal datasets. In this study, we propose a novel approach called the self-supervised hypergraph learning (SHL) framework that focuses on extracting hypergraph features to improve multimodal representation. Our method utilizes a dual embedding strategy and leverages SHL to improve the accuracy and robustness of the model. To achieve this, we employ a hypergraph learning framework to extract global context effectively by capturing rich inter-modal dependencies. Additionally, we introduce a novel self-supervised learning (SSL) component that utilizes the interaction graph data, thereby strengthening the robustness of the model. By jointly optimizing hypergraph feature extraction and SSL, SHL significantly improves the performance of multimodal representation tasks. To validate the effectiveness of our approach, we construct two comprehensive multimodal micro-video recommendation datasets using publicly available data (TikTok and MovieLens-10M). Prior to dataset creation, we meticulously handle invalid entries and outliers and complete missing mode information using external auxiliary sources, such as YouTube. These datasets are made publicly available to the research community for evaluation purposes. Experimental results on the above recommendation datasets demonstrate that the proposed SHL approach outperforms state-of-the-art baselines, highlighting its superior performance in multimodal representation tasks.

Abstract Objective To early recognise and improve the prognosis of children systemic lupus erythematosus (cSLE)-associated pancreatitis by summarising and analysing clinical features and prognosis data from 12 cases. Methods Retrospective analysis of clinical data from 12 cases of cSLE-associated pancreatitis diagnosed and treated from January 2016 to December 2021 at hospitals such as Children’s Hospital of Capital Institute of Paediatrics. Results The median SLEDAI-2K score for disease activity was 18.00 (range 12.25–21.00) in the case group and 10.00 (range 7.00–18.00) in the control group, with a statistically significant difference (P < 0.05) between the two groups. The case group had a higher proportion of abdominal pain, vomiting, abdominal distension, pleural effusion, Raynaud’s phenomenon (RP), splenic infarction, and concurrent macrophage activation syndrome (MAS) than the control group, with a statistically significant difference (P < 0.05). Serum ferritin (SF), alanine transaminase (ALT), aspartate transaminase (AST), lactate dehydrogenase (LDH), amylase, and increased 24-h urine protein levels were statistically different between the two groups (P < 0.05); platelet counts (PLT) reduction was also statistically different (P < 0.05). The case group had a higher proportion of methylprednisolone pulse therapy, cyclophosphamide pulse therapy during remission induction, and therapeutic plasma exchange than the control group, with a statistically significant difference (P < 0.05) between the two. Conclusion CSLE-associated pancreatitis has a high fatality rate. The presence of RP, splenic infarction, pleural effusion, and MAS warrants attention from clinicians regarding the possibility of pancreatitis. Once pancreatitis is detected, the primary disease needs active treatment for better prognosis.

Abstract Background Lupus mesenteric vasculitis (LMV) as initial presentation is rare, especially in childhood-onset systemic lupus erythematosus (cSLE). It is a critical complication of lupus. At present, the research on cSLE with LMV as the initial presentation is few. The aim of this study was to analyze the clinical characteristics and prognosis of cSLE with LMV in the Chinese population, compared with non-LMV cSLE. Methods A retrospective case-controlled study was conducted on 55 cSLE patients between July 2018 and July 2021. The clinical data, laboratory findings, imaging, treatment, and follow-up data were collected and compared between the two groups of cSLE with LMV and non-LMV. Non-LMV cSLE patients were matched according to the age and sex of LMV patients. Results A total of 11 cSLE patients with LMV as the LMV group and 44 cSLE patients without LMV as the non-LMV group were included. The average age of onset was 12.55 ± 1.57 years old, the male-to-female ratio was 2:9, and high disease activity was observed in the LMV group. Abdominal pain was most common in LMV. Compared with the non-LMV, the percentage of abdominal pain, vomiting, abdominal distension, and diarrhea was higher, and gastrointestinal tract, serous cavity, kidney, and lung damage were higher in the LMV group (P

Abstract Recent text generation methods frequently learn node representations from graph‐based data via global or local aggregation, such as knowledge graphs. Since all nodes are connected directly, node global representation encoding enables direct communication between two distant nodes while disregarding graph topology. Node local representation encoding, which captures the graph structure, considers the connections between nearby nodes but misses out onlong‐range relations. A quantum‐like approach to learning better‐contextualised node embeddings is proposed using a fusion model that combines both encoding strategies. Our methods significantly improve on two graph‐to‐text datasets compared to state‐of‐the‐art models in various experiments.

Abstract General movements (GMs) have been widely used for the early clinical evaluation of infant brain development, allowing immediate evaluation of potential development disorders and timely rehabilitation. The infants’ general movements can be captured digitally, but the lack of quantitative assessment and well‐trained clinical pediatricians presents an obstacle for many years to achieve wider deployment, especially in low‐resource settings. There is a high potential to explore wearable sensors for movement analysis due to outstanding privacy, low cost, and easy‐to‐use features. This work presents a sparse sensor network with soft wireless IMU devices (SWDs) for automatic early evaluation of general movements in infants. The sparse network consisting of only five sensor nodes (SWDs) with robust mechanical properties and excellent biocompatibility continuously and stably captures full‐body motion data. The proof‐of‐the‐concept clinical testing with 23 infants showcases outstanding performance in recognizing neonatal activities, confirming the reliability of the system. Taken together with a tiny machine learning algorithm, the system can automatically identify risky infants based on the GMs, with an accuracy of up to 100% (99.9%). The wearable sparse sensor network with an artificial intelligence‐based algorithm facilitates intelligent evaluation of infant brain development and early diagnosis of development disorders.

Abstract The aim of this phase I study is to evaluate, for the first time, the safety and efficacy of sintilimab in pediatric patients diagnosed with advanced or recurrent malignancies. During the dose escalation phase, patients received a single intravenous infusion of sintilimab at varying doses of 1, 3, and 10 mg/kg. The primary endpoints included the identification of dose-limiting toxicities (DLTs) as well as the evaluation of safety and tolerance. Secondary endpoints focused on assessing objective response rate (ORR), progression-free survival (PFS), and overall survival (OS). A total of 29 patients were enrolled, including 10 individuals diagnosed with Hodgkin lymphoma (HL) and 19 patients with various other tumor categories. Notably, diverse pathological types such as thymoma, choroid plexus carcinoma, and NK/T-cell lymphoma were also included in the study cohort. By the safety data cutoff, most adverse events were grade 1 or 2, with grade 3 or higher treatment-related adverse events (TRAE) occurring in 10% of patients. Among the 27 evaluated subjects, four achieved confirmed complete response (CR) while seven patients exhibited confirmed partial response (PR). Additionally, seven patients maintained disease (SD) during the study period. Notably, sintilimab demonstrated remarkable tolerability without DLTs and exhibited promising anti-tumor effects in pediatric HL. Whole-exome sequencing (WES) was conducted in 15 patients to assess the mutational landscape and copy number variation (CNV) status. The completion of this phase I study establishes the foundation for potential combination regimens involving sintilimab in childhood cancer treatment. Objective The dry-wet cycles can promote the deterioration of the mechanical properties and stability of schist slope rock masses and induce geological disasters such as landslides and collapses. Methods Taking mica quartz schist, which is widely distributed in Shiyan, Hubei Province, as the research object, uniaxial compression tests and electron microscope scanning were carried out on mica quartz schist samples under different dry-wet cycles to reveal the degradation law and mechanism of its compressive property. Results The results show that under the action of dry-wet cycles, the degradation effect of the compressive strength and elastic modulus of mica quartz schist were obvious. The degradation rate was fast and the degradation range was large in the early stage, but the degradation rate was slow and the degradation range was low in the later stage. The macroscopic failure modes were gradually controlled by the schistosity planes, which gradually evolved from styles of "X" and "Y" failure to along the schistosity planes failure. The microstructure was characterized by continuous spalling and granulation of flake mineral particles, and the microcracks as well as pores were gradually developed, expanded and connected. The mica quartz schist produces secondary minerals, ions and rock cuttings and leaves the rock continuously under dry-wet cycles, which promotes the preferential expansion and cracking of schistosity, microcracks and other structural planes. which then degrades the overall mechanical properties of the mica quartz schist. Conclusion The research results can provide a theoretical basis for the stability analysis and treatment design of schist slopes.

Abstract Ice-templating technology holds great potential to construct industrial porous materials from nanometers to the macroscopic scale for tailoring thermal, electronic, or acoustic transport. Herein, we describe a general ice-templating technology through freezing the material on a rotating cryogenic drum surface, crushing it, and then re-casting the nanofiber slurry. Through decoupling the ice nucleation and growth processes, we achieved the columnar-equiaxed crystal transition in the freezing procedure. The highly random stacking and integrating of equiaxed ice crystals can organize nanofibers into thousands of repeating microscale units with a tortuous channel topology. Owing to the spatially well-defined isotropic structure, the obtained Al2O3·SiO2 nanofiber aerogels exhibit ultralow thermal conductivity, superelasticity, good damage tolerance, and fatigue resistance. These features, together with their natural stability up to 1200 °C, make them highly robust for thermal insulation under extreme thermomechanical environments. Cascading thermal runaway propagation in a high-capacity lithium-ion battery module consisting of LiNi0.8Co0.1Mn0.1O2 cathode, with ultrahigh thermal shock power of 215 kW, can be completely prevented by a thin nanofiber aerogel layer. These findings not only establish a general production route for nanomaterial assemblies that is conventionally challenging, but also demonstrate a high-energy-density battery module configuration with a high safety standard that is critical for practical applications.

From established latency, human herpes virus type 2 (HSV-2) frequently reactivates into the genital tract, resulting in symptomatic ulcers or subclinical shedding. Tissue-resident memory (TRM) CD8+ T cells that accumulate and persist in the genital skin at the local site of recrudescence are the “first responders” to viral reactivation, performing immunosurveillance and containment and aborting the ability of the virus to induce clinical lesions. This review describes the unique spatiotemporal characteristics, transcriptional signatures, and noncatalytic effector functions of TRM CD8+ T cells in the tissue context of human HSV-2 infection. We highlight recent insights into the intricate overlaps between intrinsic resistance, innate defense, and adaptive immunity in the tissue microenvironment and discuss how rapid virus–host dynamics at the skin and mucosal level influence clinical outcomes of genital herpes diseases.

Abstract Future communication networks are widely considered to be able to provide both high‐speed communication service and reliable sensing service. Integrated sensing and communication (ISAC) is considered to be the key technology to realize this vision. Appropriate resource allocation strategy is essential for ensuring the quality of both communication and sensing services in terms of ISAC. Existing ISAC‐based resource allocation schemes mainly focus on the coexistence of sensing and communication. However, in multi‐user ISAC networks, the services required by users are diverse, including sensing‐only, communication‐only and dual‐function cases. To this end, a resource allocation strategy is proposed based on user quality of service (QoS). Specifically, the sum rate of cellular network is maximized via optimizing spectrum resource selection and transmit power, while satisfying the sensing QoS. The optimal solution to the entire problem is achieved by first demonstrating that the communication rate in this paper is a monotonically increasing function of sensing power, which allows to obtain the optimal power allocation scheme. Subsequently, a matching method is employed to obtain the optimal spectrum sharing scheme. Numerical results validate the effectiveness of the proposed algorithm and also unveil a flexible trade‐off in ISAC systems over the benchmark scheme.

BackgroundCough is a common symptom during and after COVID-19 infection; however, few studies have described the cough profiles of COVID-19. ObjectiveThe aim of this study was to investigate the prevalence, severity, and associated risk factors of severe and persistent cough in individuals with COVID-19 during the latest wave of the Omicron variant in China. MethodsIn this nationwide cross-sectional study, we collected information of the characteristics of cough from individuals with infection of the SARS-CoV-2 Omicron variant using an online questionnaire sent between December 31, 2022, and January 11, 2023. ResultsThere were 11,718 (n=7978, 68.1% female) nonhospitalized responders, with a median age of 37 (IQR 30-47) years who responded at a median of 16 (IQR 12-20) days from infection onset to the time of the survey. Cough was the most common symptom, occurring in 91.7% of participants, followed by fever, fatigue, and nasal congestion (68.8%-87.4%). The median cough visual analog scale (VAS) score was 70 (IQR 50-80) mm. Being female (odds ratio [OR] 1.31, 95% CI 1.20-1.43), having a COVID-19 vaccination history (OR 1.71, 95% CI 1.37-2.12), current smoking (OR 0.48, 95% CI 0.41-0.58), chronic cough (OR 2.04, 95% CI 1.69-2.45), coronary heart disease (OR 1.71, 95% CI 1.17-2.52), asthma (OR 1.22, 95% CI 1.02-1.46), and gastroesophageal reflux disease (GERD) (OR 1.21, 95% CI 1.01-1.45) were independent factors for severe cough (VAS>70, 37.4%). Among all respondents, 35.0% indicated having a productive cough, which was associated with risk factors of being female (OR 1.44, 95% CI 1.31-1.57), having asthma (OR 1.84, 95% CI 1.52-2.22), chronic cough (OR 1.44, 95% CI 1.19-1.74), and GERD (OR 1.22, 95% CI 1.01-1.47). Persistent cough (>3 weeks) occurred in 13.0% of individuals, which was associated with the risk factors of having diabetes (OR 2.24, 95% CI 1.30-3.85), asthma (OR 1.70, 95% CI 1.11-2.62), and chronic cough (OR 1.97, 95% CI 1.32-2.94). ConclusionsCough is the most common symptom in nonhospitalized individuals with Omicron SARS-CoV-2 variant infection. Being female, having asthma, chronic cough, GERD, coronary heart disease, diabetes, and a COVID-19 vaccination history emerged as independent factors associated with severe cough, productive cough, and persistent cough.

Abstract Objective This study was designed to analyze the structural characteristics of the intestinal flora of elderly Uygur patients with sarcopenia, thereby providing new ideas for clinical treatment. Methods Firstly, fecal samples were collected from 40 elderly Uygur patients with sarcopenia (Sarcopenia group) and 40 healthy people (Control group). Next, significant differences in the intestinal flora between the two groups were analyzed based on 16S rDNA high‐throughput sequencing. The linear discriminant analysis effect size (LEfSe) was used to estimate the magnitude of the effect of each component (species) abundance on the differential effect. Additionally, an analysis was also performed on the relationship between the intestinal flora and the cytokines in the peripheral blood of patients with sarcopenia. Results The results of β diversity showed that there were differences in the structure of the intestinal flora between the two groups. Besides, the phylum level of intestinal flora between the two groups was not significantly different. However, the difference was significant in the intestinal flora at the order, family, and genus levels between the two groups. Among them, Lachnoclostridium, Photobacterium, Anaerobic Bacillus, Hydrogenophilus, and Eubacterium were enriched in the Sarcopenia group; Prevotella 9, Firmicutes FCS020 group, Streptobacillus, Aggregatibacter, Corynebacterium, Clostridium Difficile, and Haloanaerobium were enriched in the Control group. The LEfSe outcomes further showed that Lachnoclostridium was highly enriched in the Sarcopenia group; Prevotella 9 and Firmicutes FCS020 group were significantly enriched in the Control group. Furthermore, the relative abundance of Lachnoclostridium and Streptobacillus were significantly different in patients with high and low IL‐6 levels. Conclusion In conclusion, Lachnoclostridium is significantly enriched in the intestines of elderly Uygur patients with sarcopenia; the increase in Lachnoclostridium abundance and the decrease in Streptobacillus abundance are associated with high levels of IL‐6. Therefore, abnormal intestinal flora is related to inflammatory reflexes in patients with sarcopenia.

Objectives: This review aimed to harmoniously summarize and compare outlier rates for various cardiac troponin (cTn) assays, including high-sensitivity-cTn (hs-cTn) assays and contemporary cTn (generation of assays prior to hs-cTn ones) assays, from the published studies. Methods: The PRISMA guidelines were utilized to perform this systematic review. Five databases, including PubMed, Scopus, Embase, Cochrane Library, and Web of Science, were searched using specific keywords up to June 30th, 2023. Studies reporting specifically calculated outlier rates for cTn assays when conducting in-vitro diagnosis in human samples were included. Selected studies were then further assessed using the GRADE tool. Results: Thirteen studies were included. The data from the studies were summarized statistically in this review. The results showed substantial evidence of improved analytical robustness or reduced respective mean rates of outliers, critical outliers, and analytical outliers for hs-cTn assays (0.14 %, 0.18 %, and 0.18 %) compared to contemporary cTn assays (0.63 %, 0.71 %, and 0.50 %). Conclusion: The findings offer promisingly provide a comprehensive reference for laboratory scientists and clinical staff in choosing the most suitable cTn assay for patient care regrading outlier rates. Besides, this review reveals the advancements of hs-cTn assays with lower outlier rates than contemporary cTn assays. The emerging challenges for continuously improving analytical robustness of cTn assays are also elaborated.

Two-dimensional (2D) materials have attracted considerable research interest, leading to significant advances in energy applications in recent years, such as lithium batteries, catalysis, electronics, and thermoelectrics, owing to their rich controllable properties and excellent performances. Recently, pressure has been successfully employed as an effective method for property modulation of 2D materials, through tuning electronic orbitals and bonding patterns. In this review, we summarize recent progresses in the pressure-driven property modulations and elucidate the underlying mechanism of the pressure modulation of 2D materials. Further, we identify the remaining challenges and opportunities in this new, vibrant area of research for energy conversion and utilization. Among the different property modulation strategies, the in situ application of high pressure is systematically identified as a promising knob for 2D materials. This review is expected to inspire further research on the fundamental understanding and practical applications of high-pressure modulation in 2D materials.

Abstract Background Pegylated recombinant human granulocyte colony‐stimulating factor (PEG‐rhG‐CSF) can be used in pediatric patients. This study assessed the safety and efficacy of PEG‐rhG‐CSF as a primary prophylactic drug against neutropenia after chemotherapy in pediatric patients with solid tumors or non‐Hodgkin lymphoma (NHL). Patients and Methods This phase II study (between October 2020 and March 2022) enrolled pediatric patients with solid tumors or NHL treated with high‐intensity chemotherapy and with grade ≥3 myelosuppression for at least 14 days during chemotherapy. Prophylactic PEG‐rhG‐CSF was given at 100 μg/kg body weight (maximum total dosage of 6 mg) once 24–48 h following chemotherapy for two cycles. The primary endpoint was the incidence of PEG‐rhG‐CSF‐related adverse events (AEs). The key secondary endpoints were the rates of grade 3/4 neutropenia and febrile neutropenia (FN). Results This study included 160 pediatric patients with a median age of 6.22 (0.29, 18.00) years. Fifty‐eight patients (36.25%) were diagnosed with sarcoma. AEs potentially related to PEG‐rhG‐CSF included bone pain (n = 32), fatigue (n = 21), pain at the injection site (n = 21), and myalgia (n = 20). The rates of grade 3/4 neutropenia and FN during treatment were 57.28% and 29.45%, respectively. Conclusion PEG‐rhG‐CSF is well tolerated and effective in pediatric patients with solid tumors or NHL. Abstract Atmospheric chemistry transport models have been extensively applied in aerosol forecasts over recent decades, whereas they are facing challenges from uncertainties in emission rates, meteorological data, and over-simplified chemical parameterizations. Here, we developed a spatial-temporal deep learning framework, named PPN (Pollution-Predicting Net for PM2.5), to accurately and efficiently predict regional PM2.5 concentrations. It has an encoder-decoder architecture and combines the preceding PM2.5 observations and numerical weather prediction. Besides, the model proposes a weighted loss function to promote the forecasting performance in extreme events. We applied the proposed model to forecast 3-day PM2.5 concentrations over the Beijing-Tianjin-Hebei region in China on a three-hour-by-three-hour basis. Overall, the model showed good performance with R 2 and RMSE values of 0.7 and 17.7 μg m−3, respectively. It could capture the high PM2.5 concentration in the south and relatively low concentration in the north and exhibit better performance within the next 24 h. The use of the weighted loss function decreased the level of “high values underestimation, low values overestimation”, while incorporating the preceding PM2.5 observations into the encoder phase improved the predictive accuracy within 24 h. We also compared the model result with that from a state-of-the-art numerical model (WRF-Chem with pollutant data assimilation). The temporal R 2 and RMSE from the WRF-Chem were 0.30−0.77 and 19−45 μg m−3 while those from the PPN model were 0.42−0.84 and 15−42 μg m−3. The proposed model shows powerful capacity in aerosol forecasts and provides an efficient and accurate tool for early warning and management of regional pollution events.

AbstractThis study prepared and evaluated polymeric polybutylcyanoacrylate (PBCA) nanoparticles (NPs) that can be used as a new agent for contrast-enhanced intravascular ultrasound (IVUS) imaging with drug delivery capacity. The nanoformulation was successfully developed using suspension polymerisation and characterised in terms of size, size distribution, zeta potential, morphology, stability, toxicity effects, imaging effects and drug release study. The results showed that the nanoparticles were round and hollow, with a particle diameter of 215.8 ± 25.3 nm and a zeta potential of −22.2 ± 4.1 mV. In vitro experiments, the nanoparticles were safe, non-toxic, and stable in nature with the capacity to carry and release drug (ant-miR-126). Moreover, the nanoparticles can match the high-frequency probe of commercially IVUS as a contrast agent to improve the resolution of imaging (the mean echo intensity ratio in the vascular wall increased significantly from 10.89 ± 1.10 at baseline, to 24.51 ± 1.91 during injection and 43.70 ± 0.88 after injection, respectively p

Interfacial solar steam generation (ISSG) is a novel and potential solution to global freshwater crisis. Here, based on a facile sol-gel fabrication process, we demonstrate a highly scalable Janus aramid nanofiber aerogel (JANA) as a high-efficiency ISSG device. JANA performs near-perfect broadband optical absorption, rapid photothermal conversion and effective water transportation. Owning to these features, efficient desalination of salty water and purification of municipal sewage are successfully demonstrated using JANA. In addition, benefiting from the mechanical property and chemical stability of constituent aramid nanofibers, JANA not only possesses outstanding flexibility and fire-resistance properties, but its solar steaming efficiency is also free from the influences of elastic deformations and fire treatments. We envision JANA provides a promising platform for mass-production of high-efficiency ISSG devices with supplementary capabilities of convenient transportation and long-term storage, which could further promote the realistic applications of ISSG technology.

Abstract Background Compared with other pediatric tumors, little advances were achieved in studies on the stratified treatment in localized Ewing sarcoma. Most pediatric oncology groups treated Ewing sarcoma according to whether there was an existing metastasis, without involving more prognostic factors. In this study, patients with localized Ewing sarcoma were divided into resectable and unresectable groups at diagnosis and received chemotherapy with different intensity, for the purpose of achieving good efficacy, avoiding overtreatment and reducing unnecessary toxicity. Methods A total of 143 patients with a median age of 10 years old diagnosed with localized Ewing sarcoma in this retrospective study were divided into two cohorts (Cohort 1, n = 42; Cohort 2, n = 101) and patients in Cohort 2 received chemotherapy with different intensity (Regimen 1, n = 52; Regimen 2, n = 49). Outcomes were analyzed using the Kaplan–Meier method to estimate event‐free survival (EFS) and overall survival (OS), and the curves were compared using the log‐rank test. Results The 5‐year EFS and 5‐year OS for all the patients were 69.0% and 77.5%. The 5‐year EFS for Cohort 1 and Cohort 2 were 76.0% and 66.1% (p = 0.31), and the 5‐year OS were 83.0% and 75.1% (p = 0.30), respectively. In Cohort 2, the 5‐year EFS rate of patients treated with Regimen 2 was significantly higher than that of patients treated with Regimen 1 (74.5% vs. 58.3%, p = 0.03). Conclusions According to whether a grossly complete resection was received at the time of diagnosis, localized Ewing sarcoma patients in this study were stratified into two groups and received different intensities of chemotherapy, which achieved good efficacy and avoided overtreatment and reduced unnecessary toxicity.

Abstract Backgroud To summarize the clinical characteristics and identify the risk factors for pediatric Takayasu arteritis (TAK) with coronary artery lesions (CALs). Methods Clinical data of pediatric TAK patients in our center were retrospectively assessed. Independent risk factors for CALs were identified using multivariate logistic regression analysis. Survival analysis was used to compare differences in survival rates between the groups. Results Among the 66 pediatric TAK cases, the incidence of accompanying CALs was 39.4%. In the CAL group, 19 (73.1%) cases started within 36 months. None of the patients had symptoms of angina or ischemia on electrocardiogram (ECG), the CALs were detected using coronary ultrasound. The CALs most commonly were the left main and right coronary arteries. The lesions were mostly small or middle coronary artery aneurysms; some children may have giant coronary aneurysmal dilations, thrombosis and heart failure. Extreme multi-label text classification (XMTC) is an emerging and essential task in natural language processing. Its objective is to retrieve the most relevant labels for a text from a large set of labels while balancing time and accuracy. Although large-scale pre-trained models have brought new perspectives to this task, more attention should be given to valuable fine-tuned methods and the significant semantic gap between texts and labels. In this paper, we propose a novel guide network (GUDN) with a label reinforcement strategy based on label semantics to help fine-tune pre-trained models for classification. Experimental results demonstrate that GUDN outperforms state-of-the-art methods on Eurlex-4k and achieves competitive results on other popular datasets. In addition, we find that meaningless tokens can harm the Transformer-based model’s classification accuracy in another experiment. We conclude that GUDN is effective in the presence of solid semantics. Our source code is available at https://t.hk.uy/aFSH.

Abstract The application of totally implantable access ports (TIAPs) reduces treatment-related discomfort; however, the existence of catheter may cause side effects, with the most common one being the occurrence of TIAPs-associated thrombosis. The risk factors for TIAPs-associated thrombosis in pediatric oncology patients have not been fully described. A total of 587 pediatric oncology patients undergoing TIAPs implantation at a single center over a 5-year period were retrospectively analyzed in the present study. We investigated the risk factors for thrombosis, emphasizing the internal jugular vein distance, by measuring the vertical distance from the highest point of the catheter to the upper border of the left and right extremitas sternalis claviculae on chest X-ray images. Among 587 patients, 143 (24.4%) had thrombosis. Platelet count, C-reactive protein, and the vertical distance from the highest point of the catheter to the upper border of the left and right extremitas sternalis claviculae were demonstrated to be the main risk factors for the development of TIAPs-associated thrombosis. TIAPs-associated thrombosis, especially asymptomatic events, is common in pediatric cancer patients. The vertical distance from the highest point of the catheter to the upper border of the left and right extremitas sternalis claviculae was a risk factor for TIAPs-associated thrombosis, which deserved additional attention.

Abstract Video captioning aims at automatically generating a natural language caption to describe the content of a video. However, most of the existing methods in the video captioning task ignore the relationship between objects in the video and the correlation between multimodal features, and they also ignore the effect of caption length on the task. This study proposes a novel video captioning framework (ORMF) based on the object relation graph and multimodal feature fusion. ORMF uses the similarity and Spatio‐temporal relationship of objects in video to construct object relation features graph and introduce graph convolution network (GCN) to encode the object relation. At the same time, ORMF also constructs a multimodal features fusion network to learn the relationship between different modal features. The multimodal feature fusion network is used to fuse the features of different modals. Furthermore, the proposed model calculates the length loss of the caption, making the caption get richer information. The experimental results on two public datasets (Microsoft video captioning corpus [MSVD] and Microsoft research‐video to text [MSR‐VTT]) demonstrate the effectiveness of our method.

Abstract Sodium ion‐batteries (SIBs) are considered as a class of promising alternatives to lithium‐ion batteries (LIBs) to overcome their drawbacks of limited sources and safety problems. However, the lack of high‐performance electrode materials hinders the wide‐range commercialization of SIBs. Comparing to inorganic counterparts, organic electrode materials, which are benefitted from flexibly designable structures, low cost, environmental friendliness, and high theoretical gravimetric capacities, should be a prior choice. Here, a covalent organic polymer (COP) based material (denoted as CityU‐9) is designed and synthesized by integrating multiple redox motifs (benzoquinone and thioether), improved conductivity (sulfur induction), and intrinsic insolubility (rigid skeleton). The half‐cell SIBs exhibit ultrahigh specific capacity of 1009 mAh g−1 and nearly no capacity drop after 650 cycles. The first all‐COP symmetric full‐cell shows high specific capacity of 90 mAh g−1 and excellent rate capability. This work can extend the selection of redox‐active moieties and provide a rational design strategy of high‐performance novel organic electrode materials.

Abstract Background Circular RNAs (circRNAs) have important regulatory functions in cancer, but the role of circRNAs in the tumor microenvironment (TME) remains unclear. Moreover, we also explore the effects of si-circRNAs loaded in nanoparticles as therapeutic agent for anti-tumor in vivo. Methods We conducted bioinformatics analysis, qRT-PCR, EdU assays, Transwell assays, co-culture system and multiple orthotopic xenograft models to investigate the expression and function of circRNAs. Additionally, PLGA-based nanoparticles loaded with si-circRNAs were used to evaluate the potential of nanotherapeutic strategy in anti-tumor response. Results We identified oncogene SERPINE2 derived circRNA, named as cSERPINE2, which was notably elevated in breast cancer and was closely related to poor clinical outcome. Functionally, tumor exosomal cSERPINE2 was shuttled to tumor associated macrophages (TAMs) and enhanced the secretion of Interleukin-6 (IL-6), leading to increased proliferation and invasion of breast cancer cells. Furthermore, IL-6 in turn increased the EIF4A3 and CCL2 levels within tumor cells in a positive feedback mechanism, further enhancing tumor cSERPINE2 biogenesis and promoting the recruitment of TAMs. More importantly, we developed a PLGA-based nanoparticle loaded with si-cSERPINE2, which effectively attenuated breast cancer progression in vivo. Conclusions Our study illustrates a novel mechanism that tumor exosomal cSERPINE2 mediates a positive feedback loop between tumor cells and TAMs to promote cancer progression, which may serve as a promising nanotherapeutic strategy for the treatment of breast cancer.

The detection of the spine is crucial in automating the measurement of the Cobb Angle. While various segmentation models have been employed for vertebrae segmentation in X-ray images, there is a need to enhance segmentation performance. This paper proposes a comprehensive automatic measurement method for the Cobb angle. The RetinaNet model is employed to detect the region of interest corresponding to the spine, while the W-Net model is developed for accurate vertebrae segmentation. To address the issue of adjacent vertebrae adhesion in the segmented image, a post-processing technique is applied. Experimental results demonstrate that the W-Net model achieves superior performance, with a mean Intersection over Union (MIoU) of 0.9073 ± 0.0021, Dice Coefficient of 0.9446 ± 0.0139, and Precision of 0.9390 ± 0.0190. The post-processing step reduces adhesion at one end by approximately 83.4% and adhesion at both ends by approximately 83.6%. The reliability of the proposed method is evaluated through intra-group correlation coefficients (ICC) of 0.902 and 0.915, respectively, between two observers, both exceeding 0.9. The mean absolute deviation (MAD) is 3.08° and 2.91°, respectively. Therefore, the proposed method achieves automatic detection of the Cobb angle without the need for manual cropping or additional human intervention, while maintaining good reliability.

To improve the accuracy of feature extraction and description of various scales in traditional Oriented FAST and Rotated BRIEF (ORB) feature matching algorithm, this paper proposes an ORB feature matching algorithm based on multi-scale feature description fusion and feature point mapping error correction. Firstly, when establishing the feature scale pyramid, the method of using the same patch-size for description on each feature layer is adopted instead of using different patch-sizes on a unified feature layer in the original algorithm, which enhances the robustness of the descriptor and improves the matching accuracy. Secondly, FAST-SCORE maps are established on different scales, and the coordinates of high-level feature points mapped to the bottom layer are corrected to further improve the positioning accuracy of feature points. The algorithm is verified in remote sensing images, autonomous driving, and industrial automation fields. Experimental results show that when resisting theoretical interference, the average matching accuracy of the proposed algorithm is 67.9%, which is about 2.0 times that of the ORB algorithm, and the average stability is 14.0, which is about 1.5 times that of the ORB algorithm. After correcting the feature point mapping, the matching accuracy can be further improved by 19.2%, indicating that the improved algorithm has excellent robustness when resisting interference. In the experiments on the KITTI and custom datasets, the matching accuracy of the proposed algorithm reached 88.70% and 96.88%, respectively, which is an improvement of 10.15% and 1.2% compared to the ORB algorithm. At the same time, the matching time was reduced by 17.34% and 24.30%, ensuring the accuracy and real-time performance of the algorithm in practical scenarios.

In developing countries, funding is a significant obstacle to receiving higher education. Brilliant but needy students cannot complete their studies since their parents are unemployed and their countries’ economies are poor. As a result, the students’ talents are not harnessed to their full potential. In order to help students obtain higher education and harness their full potential, governments provide student loans to students in higher education. The government provides loans to students through the ministry of education. The students pay back the loan with interest when they start working. Governments have been the sole funders of student loans. The emergence of COVID-19 and the Russia-Ukraine war have resulted in a global economic crisis. Because of the global economic crisis, the government’s spending has increased. In order to help reduce the burden of government and thereby reduce spending, we intend to revolutionize the student loan program through blockchain and crowdsourcing. This work presents a blockchain-based crowdsourcing decentralized loan platform where investors will be brought on board to provide funds for students in higher education. The platform will allow students to apply for loans from investors through registered financial institutions. The students will pay back the loans with interest when they enter the workforce. The proposed platform will allow students to fund their education, investors will get interest on the money they invest, and governments can channel the money they put into student loan programs into other avenues. We perform a thorough security analysis and back the efficiency of our work with numerical results.

The high demand for rapid wound healing has spurred the development of multifunctional and smart bioadhesives with strong bioadhesion, antibacterial effect, real-time sensing, wireless communication, and on-demand treatment capabilities. Bioadhesives with bio-inspired structures and chemicals have shown unprecedented adhesion strengths, as well as tunable optical, electrical, and bio-dissolvable properties. Accelerated wound healing has been achieved via directly released antibacterial and growth factors, material or drug-induced host immune responses, and delivery of curative cells. Most recently, the integration of biosensing and treatment modules with wireless units in a closed-loop system yielded smart bioadhesives, allowing real-time sensing of the physiological conditions (e.g., pH, temperature, uric acid, glucose, and cytokine) with iterative feedback for drastically enhanced, stage-specific wound healing by triggering drug delivery and treatment to avoid infection or prolonged inflammation. Despite rapid advances in the burgeoning field, challenges still exist in the design and fabrication of integrated systems, particularly for chronic wounds, presenting significant opportunities for the future development of next-generation smart materials and systems.

Key Clinical Message Hyponatremia is common among children undergoing treatment for hematologic malignancies and may be attributed to multiple underlying causes. In cases of hyponatremia due to mixed physiology, the osmolal gap, can identify pseudohyponatremia that may be masked by other causes.

Photocatalytic degradation has been extensively investigated toward the removal emerging contaminants (ECs) from water. In this study, a series of Ag-Bi3O4Cl plasmon photocatalysts were synthesized through the photo-deposition of metallic Ag on the Bi3O4Cl surface. The effects of plasmon modification on the catalytic performance of bismuth oxychlorides were analyzed. Ag addition did not alter the morphology of Bi3O4Cl. With the increasing Ag content, the number of oxygen defects on the catalyst surface first increased and then decreased. Moreover, the surface plasmon resonance effect of Ag suppressed the recombination of electron–hole pairs, promoting the migration and separation of photocarriers and improving the light absorption efficiency. However, the addition of excessive Ag reduced the number of active sites on the Bi3O4Cl surface, hindering the catalytic degradation of pollutants. The optimal Ag-Bi3O4Cl photocatalyst (Ag ratio: 0.025; solution pH: 9; dosage: 0.8 g/L) achieved 93.8 and 94.9% removal of ciprofloxacin and tetrabromobisphenol A, respectively. The physicochemical and photoelectric properties of Ag-Bi3O4Cl were determined through various characterization techniques. This study demonstrates that introducing metallic Ag alters the electron transfer path of the catalyst, reduces the recombination rate of electron–hole pairs, and effectively improves the catalytic efficiency of Bi3O4Cl. Furthermore, the pathways of ciprofloxacin degradation products and their biotoxicity were revealed.

BackgroundProgrammed death receptor 1 (PD-1) inhibition has shown durable response and mild adverse events (AEs) in adult malignancies. However, data on the clinical activity of PD-1 inhibition in pediatric patients are lacking. We comprehensively assessed the efficacy and safety of PD-1 inhibitor-based regimens for pediatric malignancies.MethodsWe conducted a real-world, multi-institutional, retrospective analysis of pediatric malignancies treated with PD-1 inhibitor-based regimens. The primary endpoints were objective response rate (ORR) and progression-free survival (PFS). The secondary endpoints included disease control rate (DCR), duration of response (DOR), and AEs. The Kaplan–Meier method was used to calculate PFS and DOR. The National Cancer Institute Common Toxicity Criteria for AEs (version 5.0) were used to grade toxicity.ResultsA total of 93 and 109 patients were evaluated for efficacy and safety, respectively. For all efficacy-evaluable patients, PD-1 inhibitor monotherapy, combined chemotherapy, combined histone deacetylase inhibitor, and combined vascular endothelial growth factor receptor tyrosine kinase inhibitor cohorts, the ORR and DCR were 53.76%/81.72%, 56.67%/83.33%, 54.00%/80.00%, 100.00%/100.00%, and 12.50%/75.00%, respectively; the median PFS and DOR were 17.6/31.2 months, not achieved/not achieved, 14.9/31.2 months, 17.6/14.9 months, and 3.7/1.8 months, respectively; the incidence rate of AEs were 83.49%, 55.26%, 100.00%, 80.00%, and 100.00%, respectively. One patient in the PD-1 inhibitor-combined chemotherapy cohort discontinued treatment due to diabetic ketoacidosis.ConclusionsThis largest retrospective analysis demonstrate that PD-1 inhibitor-based regimens are potentially effective and tolerable in pediatric malignancies. Our findings provide references for future clinical trials and practice of PD-1 inhibitors in pediatric cancer patients.

ABSTRACTT/NK cell-based immunotherapy has achieved remarkable success in adult cancers but has limited efficacy in pediatric malignancies including high-risk neuroblastoma (NB). Immune defects of NB tumor microenvironment are poorly understood compared with adults. Here, we described the unique characteristics of NB immune contexture and determined the phenotype signatures of PD-L1-expressing CD8+ T and NK cells in NB tumors by systemically analyzing the spatial distribution of T and NK cells and the distinct expression of programmed death 1 (PD-1) and its ligand (PD-L1) in patients with NB. We found that PD-L1-expressing CD8+ T and NK cells in NB tumors were highly activated and functionally competent and associated with better clinical outcomes. Intratumoral NK cells were a favorable prognostic biomarker independent of CD8+ T cells, PD-1/PD-L1 expression, tumor stage, MYCN amplification, and risk classification. NK cells combined with anti-PD-1/PD-L1 antibodies showed potent antitumor activity against both MYCN-amplified and non-amplified NBs in vitro and in vivo, and PD-L1-expressing NK cells associated with improved antitumor efficacy. Collectively, we raise novel insights into the role of PD-L1 expression on CD8+ T-cell and NK-cell activation. We highlight the great potential of intratumoral NK cells in better defining risk stratification, and predicting survival and response to anti-PD-1/PD-L1 therapy in NB. These findings explain why single anti-PD-1/PD-L1 therapy may not be successful in NB, suggesting its combination with NK cell-adoptive cellular therapy as a promising strategy for relapsing/refractory NB. This study provides a potential prospect that patients with PD-L1-expressing NK cells may respond to anti-PD-1/PD-L1 therapy.

Objective To investigate the biomechanics of transforaminal lumbar interbody fusion (TLIF) with interspinous process device (IPD) or pedicle screw fixation under both static and vibration conditions by the finite element (FE) method. Method A validated FE model of the L1‐5 lumbar spine was used in this study. This FE model derived from computed tomography images of a healthy female adult volunteer of appropriate age. Then the model was modified to simulate L3‐4 TLIF. Four conditions were compared: (i) intact; (ii) TLIF combined with bilateral pedicle screw fixation (BPSF); (iii) TLIF combined with U‐shaped IPD Coflex‐F (CF); and (iv) TLIF combined with unilateral pedicle screw fixation (UPSF). The intact and surgical FE models were analyzed under static and vibration loading conditions respectively. For static loading conditions, four motion modes (flexion, extension, lateral bending, and axial rotation) were simulated. For vibration loading conditions, the dynamic responses of lumbar spine under sinusoidal vertical load were simulated. Result Under static loading conditions, compared with intact case, BPSF decreased range of motion (ROM) by 92%, 95%, 89% and 92% in flexion, extension, lateral bending and axial rotation, respectively. While CF decreased ROM by 87%, 90%, 69% and 80%, and UPSF decreased ROM by 84%, 89%, 66% and 82%, respectively. Compared with CF, UPSF increased the endplate stress by 5%–8% in flexion, 7%–10% in extension, 2%–4% in lateral bending, and decreased the endplate stress by 16%–19% in axial rotation. Compared with CF, UPSF increased the cage stress by 9% in flexion, 10% in extension, and decreased the cage stress by 3% in lateral bending, and 13% in axial rotation. BPSF decreased the stress responses of endplates and cage compared with CF and UPSF. Compared BPSF, CF decreased the facet joint force (FJF) by 6%–13%, and UPSF decreased the FJF by 4%–12%. During vibration loading conditions, compared with BPSF, CF reduced maximum values of the FJF by 16%–32%, and vibration amplitudes by 22%–35%, while UPSF reduced maximum values by 20%–40%, and vibration amplitudes by 31%–45%. Conclusion Compared with other surgical models, BPSF increased the stability of lumbar spine, and also showed advantages in cage stress and endplate stress. CF showed advantages in IDP and FJF especially during vertical vibration, which may lead to lower risk of adjacent segment degeneration. CF may be an effective alternative to pedicle screw fixation in TLIF procedures.

Abstract The 2020 COVID-19 pandemic has greatly accelerated the adoption of online learning and teaching in many colleges and universities. Video, as a key integral part of online education, largely influences student learning experiences. Though many guidelines on designing educational videos have been reported, the quantitative data showing the impacts of video length on students’ academic performance in a credit-bearing course is limited, particularly for an online-flipped college engineering course. The forced pandemic lockdown enables a suitable environment to address this research gap. In this paper, we present the first step to examine the impact of short videos on students’ academic performance in such circumstances. Our results indicate that short videos can greatly improve student engagement by 24.7% in terms of video viewing time, and the final exam score by 9.0%, both compared to the long-video group. The quantitative Likert questionnaire also indicates students’ preference for short videos over long videos. We believe this study has important implications for course design for future online-flipped engineering courses.

Abstract Biallelic pathogenic variants in NDUFS8, a nuclear gene encoding a subunit of mitochondrial complex I, result in a mitochondrial disorder characterized by varying clinical presentations and severity. Here, we expand the neuroimaging and clinical spectrum of NDUFS8‐related disorder. We present three cases from two unrelated families (a girl and two brothers) homozygous for a recurrent pathogenic NDUFS8 variant [c.460G>A, p.(Gly154Ser)], located in the [4Fe‐4S] domain of the protein. One of the patients developed auto‐antibody positive diabetic ketoacidosis. Brain MRIs performed in two of the three patients demonstrated diffuse cerebral and cerebellar white matter involvement including corticospinal tracts, but notably had sparing of deep gray matter structures. Our report expands the neuroimaging phenotype of NDUFS8‐related disorder to include progressive leukodystrophy with increasing brainstem and cerebellar involvement, with relative sparing of the basal ganglia. In addition, we describe autoimmune diabetes in association with NDUFS8‐related disorder, though the exact mechanism of this association is unclear. This paper provides a comprehensive review of case presentation and progressive neuroimaging findings of three patients from two unrelated families that have an identical pathogenic NDUFS8 variant, which expands the clinical spectrum of NDUFS8‐associated neurological disease.

Abstract Background There is insufficient evidence on the clinical effectiveness and safety of infliximab (IFX) treatment of Takayasu arteritis (TA) in infants. Methods We evaluated the therapeutic effectiveness and safety of IFX in a retrospective case series of 10 infantile TA patients. Observations included assessment of clinical symptoms, laboratory testing, and vascular imaging. Results Fever was the presenting symptom for 8 of 10 infants with TA. During acute episodes, leucocyte and inflammatory indices were significantly increased. Vascular imaging showed the most commonly involved arteries to be carotid arteries, abdominal aortas, and coronary arteries (9 cases, 90%). Two weeks after initiating IFX treatment, leukocyte and platelet counts decreased and hemoglobin levels increased. Abstract With the intensive therapeutic strategies, diffuse large B-cell lymphoma (DLBCL) is still a fatal disease due to its progressive characteristics. Indoleamine 2,3-dioxygenase 1 (IDO1) is a key regulator that catalyzes the commitment step of the kynurenine pathway in the immune system, its aberrant activation may contribute to malignant cell escape eradication. However, the role of IDO1 in DLBCL progression remains elusive. Our study showed IDO1 expression was upregulated in DLBCL and was associated with a poor prognosis and low overall survival. Inhibition of IDO1 suppressed DLBCL cell proliferation in vitro and impeded xenograft tumorigenesis in vivo. RNA-seq analyses revealed MDM2 was downregulated while TP53 was upregulated in IDO1 inhibition OCI-Ly10 cells. Mechanistically, IDO1 inhibition decreased the expression of MDM2, a major negative regulator of p53, and restored p53 expression in OCI-Ly3 and OCI-Ly10 cells, resulting in cell cycle arrest and apoptosis. IDO1 inhibition induced cell apoptosis coupled with PUMA and BAX upregulation, as well as BCL2 and BCL-XL downregulation. In addition, p21, a p53 transcriptional target, was upregulated in cell cycle arrest. Taken together, this study revealed IDO1 is essential for the proliferation of DLBCL cells and may be a potential therapeutic target for the treatment of DLBCL.

Abstract In this study, we examine the cross‐view gait recognition problem. Many existing methods establish global feature representation based on the whole human body shape. However, they ignore some important details of different parts of the human body. In the latest literature, positioning partial regions to learn fine‐grained features has been verified to be effective in human identification. But they only consider coarse fine‐grained features and ignore the relationship between neighboring regions. Taken the above insights together, we propose a novel model called GaitGP, which learns both important details through fine‐grained features and the relationship between neighboring regions through global features. Our GaitGP model mainly consists of the following two aspects. First, we propose a Channel‐Attention Feature Extractor (CAFE) to extract the global features, which aggregates the channel‐level attention to enhance the spatial information in a novel convolutional component. Second, we present the Global and Partial Feature Combiner (GPFC) to learn different fine‐grained features, and combine them with the global features extracted by the CAFE to obtain the relevant information between neighboring regions. Experimental results on the CASIA gait recognition dataset B (CASIA‐B), The OU‐ISIR gait database, multi‐view large population dataset, and The OU‐ISIR gait database gait datasets show that our method is superior to the state‐of‐the‐art cross‐view gait recognition methods.
