The development of isothermal nucleic acid amplification techniques has great significance for highly sensitive biosensing in modern biology and biomedicine. A facile and robust exponential rolling circle amplification (RCA) strategy is proposed based on primer-remodeling amplification jointly via a repair enzyme and polymerase, and uracil-DNA glycosylase (UDG) is selected as a model analyte. Two kinds of complexes, complex I and complex II, are preprepared by hybridizing a circular template (CT) with a uracil-containing hairpin probe and tetrahydrofuran abasic site mimic (AP site)-embedded fluorescence-quenched probe (AFP), respectively. The target UDG specifically binds to complex I, resulting in the generation of an AP site, followed by cleavage via endonuclease IV (Endo IV) and the successive trimming of unmatched 3' terminus via phi29 DNA polymerase, thus producing a useable primer-CT complex that actuates the primary RCA. Then, numerous complex II anneal with the first-generation RCA product (RP), generating a complex II-RP assembly containing AP sites within the DNA duplex. With the aid of Endo IV and phi29, AFP, as a pre-primer in complex II, is converted into a mature primer to initiate additional rounds of RCA. So, countless AFPs are cleaved, releasing remarkably strong fluorescent signals. An electrochemical biosensing platform for kanamycin analysis based on SDA and a DNA walker.

Circulating extracellular vesicles (EVs) are promising biomarkers for the early diagnosis and prognosis of cancer in a non-invasive manner. However, the rapid and accurate identification of EVs in complex biological samples is technically challenging, which is attributed to the requirement of extensive sample purification and unsatisfactory detection accuracy due to the disturbance of interfering proteins. Herein, a simultaneous binding of double-positive EV membrane protein-based recognition mode (DRM) is proposed. By the combination of DRM-mediated toehold activation and G-quadruplex DNAZyme-catalyzed etching of Au@Ag nanorods (Au@Ag NRs), we have developed an accurate, non-purified, low-cost, and visual strategy for EV identification. The synchronous binding of double-positive proteins on EV membranes is validated by confocal laser scanning microscopy analysis. This approach exhibits excellent specificity and sensitivity toward EVs ranging from 1.0 × 105 to 1.0 × 109 particles/mL with a detection limit of 6.31 × 104 particles/mL. Moreover, we have successfully realized non-purified EV quantification in complex biological media. In addition, target-initiated catalyzed hairpin assembly (CHA) is integrated with G-quadruplex DNAZyme-catalyzed color variation of Au@Ag NRs; thus, low-background EV detection can be achieved by the naked eye. Furthermore, our strategy is easy to adapt to high-throughput formats by using an automatic microplate reader, which could be expected to meet the requirements for high-throughput detection of clinical samples. With its capacities of rapidness, portability, affordability, high throughput, non-purification, and visual detection, this strategy could provide a practical tool for accurate identification of EVs and early diagnosis of cancer.

The design of pressure hull structure plays an essential role in the research and development of underwater operation equipment such as deep-sea submersibles. As a bionic structure, egg-shaped shell has apparent advantages in the application of an underwater pressure hull. Still, the egg-shaped shell is a positive Gauss surface, so it is difficult to control the manufacturing accuracy and quality. In this paper, the technology of solid particle flexible medium forming and the principle of high pressure bulging in tubes are proposed to manufacture high-precision egg-shaped shell parts. By optimizing the shape of the punch structure, optimizing the process parameters, improving the stress and strain distribution of the egg-shaped shell, analyzing the deformation mechanism in the bulging process, and mastering the deformation and quality control methods of the granular medium flexible bulging of non-equal curvature tubular structures.

Statistics show that food poisoning caused by Salmonella typhimurium (S. Typhimurium) often tops the list of bacterial food poisoning types in countries around the world. However, detecting traces of S. Typhimurium in real samples remains challenging. In recent years, primer exchange reaction (PER), a new isothermal amplification strategy, has rapidly attracted the attention of researchers in the field of biosensing. In this work, We developed a nanostructure called DNA arch bridge (DAB) and combined the DAB with cascade PER technology to construct a novel bidirectional PER (B-PER) for ultra-sensitive detection of pathogenic bacteria as a novel fluorescent biosensor. This strategy relies on the B-PER reaction mediated by binding of the target and adaptor, which occurs with the assistance of Klenow Fragment (KF) (3'-5'exo) polymerase and produces a good deal of G-quadruplex sequences that generate a fluorescent signal by embedding fluorescent dyes. Under the best conditions, the biosensor achieves ultrasensitive detection of S. Typhimurium, and the detection limit of the strategy is 9.3 cfu mL

DNA walkers, as intelligent artificial DNA nanomachines, have been widely used as efficient nucleic acid amplification tools that the detection sensitivity can be improved by incorporating DNA walkers into DNA biosensors. Nevertheless, since the premature release or flameout in a region of locally exhausted substrate, the walking efficiency of DNA walkers remains unsatisfactory. In this work, we design a smart tripedal DNA walker that is formed by target-initiated catalyzed hairpin assembly (CHA), which can move along the DNA duplex tracks on electrode driven by toehold-mediated DNA strand displacement (TMSD) for transduction and amplification of electrochemical signals. Emphatically, this flexible tripedal DNA walker is capable of walking freely along the tracks with unconstrained walking range. Moreover, the design of multi-legged walker can weaken the derailment of leg DNA and shorten the moving time on electrode, ensuring the processive walking with high efficiency. Additionally, the persistent walking of tripedal walker is driven by cascading TMSD, which eliminates the defects of high cost and instability of enzyme-assisted amplification technology. Therefore, the tripedal DNA walker-based electrochemical biosensor has enormous potential for the applications of OTA detection, and reveals a new avenue for food safety analysis and clinical diagnosis.

A novel fluorescence sensing strategy for ultrasensitive and highly specific detection of adenosine triphosphate (ATP) has been developed by the combination of the proximity ligation assay with bidirectional enzymatic repairing amplification (BERA). The strategy relies on proximity binding-triggered the release of palindromic tail that initiates bidirectional cyclic enzymatic repairing amplification reaction with the aid of polymerase and two DNA repairing enzymes, uracil-DNA glycosylase (UDG) and endonuclease IV (Endo IV). A fluorescence-quenched hairpin probe with a palindromic tail at the 3' end is skillfully designed that functions as not only the recognition element, primer, and polymerization template for BERA but also the indicator for fluorescence signal output. On the basis of the amplification strategy, this biosensor displays excellent sensitivity and selectivity for ATP detection with an outstanding detection limit of 0.81 pM. Through simultaneously enhancing the target response signal value and reducing nonspecific background, this work deducted the background effect, and showed high sensitivity and reproducibility. Moreover, our biosensor also shows promising potential in real sample analysis. Therefore, the proximity-enabled BERA strategy indeed creates a simple and valuable fluorescence sensing platform for ATP identification and related disease diagnosis and biomedical research.

Age-related jawbone loss directly impact the function of oral cavity resulted from tooth loss, implant failure, and jaw fracture. Numerous evidences show that age-related senescence of bone marrow stromal cells (BMSCs) play a critical role in bone loss, but little attention has been paid to jawbone. Here, we delineated the critical role of sirtuin family protein 6 (SIRT6) in senescence, autophagy, and osteogenesis of BMSCs from jawbones. Radiography analysis showed less jawbone quality in elderly than young people. We also showed that SIRT6 expression decreased in bone tissue and BMSCs from the elderly by immunochemical staining. BMSCs from the elderly exhibited decreased osteogenic differentiation and inclined senescence which these phenotypes could be simulated by SIRT6 knockdown. Furthermore, accompanied with the inhibition of SIRT6, the autophagy level and ostogenesis of BMSCs was also decreased. However, using rapamycin, an autophagy activator, could rescue these adverse effects of BMSCs caused by SIRT6 inhibition. Mechanistically, SIRT6 regulated the autophagy and osteogenesis of BMSCs by activating AKT-mTOR pathway, at least in part. Finally, a decreased jawbone quality was shown in SIRT6 haploinsufficiency mice by Wnt1 specific tissue knockdown (Wnt1-Cre;SIRT6fl/+) model. Taken together, our data revealed that SIRT6 adjusted senescence and osteogenesis of BMSCs via altering autophagy level, and associated with age-related bone loss. SIRT6 could be as a promising therapeutic target for age-related osteoporosis of jawbone.

In this study, a novel, rapid and ultrasensitive fluorescence strategy using the three-dimensional (3D) dynamic DNA walker (DW)-induced branched hybridization chain reaction (bHCR) has been proposed for the detection of ampicillin (AMP). The sensing system was composed of an Nt·Bbvcl-powered DNA walker blocked by an AMP aptamer, hairpin-shaped DNA track probe (TP) and four kinds of metastable hairpin probes as the substrates of bHCR, which triggered the formation of the split G-quadruplex as the signal molecule. Due to the reasonable design, the specific binding between AMP and its aptamer activated the DW, and the DW moved on the surface of the gold nanoparticles (AuNPs) with the help of Nt·Bbvcl to produce primer probes (PPs), which induced bHCR. The products of the bHCR gathered two split G-quadruplex sequences together to form one complete G-quadruplex. The formed G-quadruplex emitted a strong fluorescence signal in the presence of thioflavin-T (ThT) to achieve the purpose of detecting AMP. The sensitivity of this method was greatly improved by the use of the 3D DNA walker and bHCR. The split G-quadruplex enhanced the signal-to-noise ratio (SNR). Under the optimal experimental conditions, a good correlation was obtained between the fluorescence intensity of the sensing system and the concentration of AMP ranging from 5 pM to 500 nM with a limit of detection (LOD) of 3.68 pM. Simultaneously, the method has been applied to the detection of antibiotics in spiked milk samples with satisfactory results.

Textile fibre is very common in daily life, and its classification and identification play an important role in textile recycling, archaeology, public security, and other industries. However, traditional identification methods are time-consuming, laborious, and often destructive to the samples. In order to quickly, accurately, and nondestructively classify and recognize textile fibres, this study established a textile fibre classification and recognition method based on hyperspectral imaging (HSI) and a one-dimensional convolutional neural network (1D-CNN) model. Hyperspectral images of 25 kinds of commercial textile fibres were collected and denoised by pixel fusion. Four traditional machine learning classification models, k-nearest neighbors (KNN), support vector machine (SVM), random forest (RF), and partial least squares-discriminant analysis (PLS-DA), were used to identify the data. The results show that RF has the highest classification accuracy, reaching 91.4%. Then a back propagation neural network (BPNN) model and a one-dimensional convolutional neural network (1D-CNN) model were constructed and compared with the traditional machine learning methods. The results show that the 1D-CNN models have 97.9% and 98.6% accuracy on the training and test sets, respectively. The precision (Pr), sensitivity (Se), specificity (Sp), and F1 score (F1 score) of the models reached 98.7%, 98.6%, 99.9%, and 98.6%, respectively, which were significantly better than the four traditional machine learning models. It seems that 1D-CNN combined with the HSI technique may be a potential method in the detection and classification of textile fibres.

This work was mainly about the understanding of how urea and ammonium affect growth, glucose consumption and ethanol production of S. cerevisiae, in particular regarding the basic physiology of cell. The basic physiology of cell included intracellular pH, ATP, NADH and enzyme activity. Results showed that fermentation time was reduced by 19% when using urea compared with ammonium. The maximal ethanol production rate using urea was 1.14 g/L/h, increasing 30% comparing with the medium prepared with ammonium. Moreover, urea could decrease the synthesis of glycerol from glucose by 26% comparing with ammonium. The by-product of acetic acid yields decreased from 40 mmol/mol of glucose (with urea) to 24 mmol/mol of glucose (with ammonium). At the end of ethanol fermentation, cell number and pH were greater with urea than ammonium. Comparing with urea, ammonium decreased the intracellular pH by 14% (from 7.1 to 6.1). Urease converting urea into ammonia resulted in a more than 50% lower of ATP when comparing with ammonium. The values of NADH/DCW were 0.21 mg/g and 0.14 mg/g respectively with urea and ammonium, suggesting a 33% lower NADH. The enzyme activity of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was 0.0225 and 0.0275 U/mg protein respectively with urea and ammonium, which was consistent with the yields of glycerol.

We have developed a novel amplification strategy termed Endo IV-assisted feedback amplification (EFA) taking advantages of rolling circular amplification (RCA) and Endo IV-assisted signal amplification (ESA) biosensing platform for detecting of adenosine triphosphate (ATP). Two kinds of specially programmed DNA complexes were employed into EFA system, one composed of a split aptamer fragment and a circular template, and the other composed of AP probe and the same circular template. Hence, ATP as a target induced the self-assembly of spilt aptamer fragments and initiated RCA reaction generating a linear DNA, which consists of hybridization elements with Complex II and formation elements of G-quadruplex. More importantly, the addition of endonuclease IV can cut the Complex II into two parts, and one of which can be trimming by phi29 DNA polymerase initiating the new round of RCA reaction producing more RCA products. Thus significantly enhanced fluorescent signal can be measured for ATP as expected, and our proposed strategy exhibits improved performances toward ATP ultrasensitive detection with a limit of detection (LOD) as low as 0.09 nM. Additionally, our developed biosensor demonstrates high selectivity and the superiority of simplicity towards ATP. Above these significant aspects, our proximity binding-induced RCA reaction-based fluorescent assay and Endo IV-fueled feedback signal amplification strategy presents an optimal detection performance towards ATP for potential application in related research and clinical diagnosis.

In this paper, a novel image watermarking method is proposed which is based on discrete wave transformation (DWT), Hessenberg decomposition (HD), and singular value decomposition (SVD). First, in the embedding process, the host image is decomposed into a number of sub-bands through multi-level DWT, and the resulting coefficients of which are then used as the input for HD. The watermark is operated on the SVD at the same time. The watermark is finally embedded into the host image by the scaling factor. Fruit fly optimization algorithm, one of the natural-inspired optimization algorithms is devoted to find the scaling factor through the proposed objective evaluation function. The proposed method is compared to other research works under various spoof attacks, such as the filter, noise, JPEG compression, JPEG2000 compression, and sharpening attacks. The experimental results show that the proposed image watermarking method has a good trade-off between robustness and invisibility even for the watermarks with multiple sizes.

Herein, a split G-quadruplex DNAzyme as a signal reporter was integrated into an electrochemical sensing platform for the detection of antibiotics with specificity and sensitivity. To improve the signal-to-noise ratio, two G-rich oligonucleotide sequences (G1 and G2) were blocked into two different hairpin probes, preventing the two segments from assembling into a spilt G-quadruplex structure. Moreover, we designed a double-arch probe, consisting of an aptamer as the recognition element and two-step enzymatic signal amplification. Concretely, the first is the Nt.BbvCI-assisted nicking cyclic reaction activated by target-aptamer binding, and the second is exonuclease III-aided cyclic amplification for generating abundant G1 and G2. The modified capture probe on the electrode was used to combine G1 and G2 to form the spilt G-quadruplex/hemin when K+ and hemin were present. This complex plays the role of DNAzyme with superior horseradish peroxidase activity in catalyzing the decomposition of H2O2. Under optimal conditions, this biosensor showed an excellent performance for sensing kanamycin with a detection limit of 83 fM for kanamycin concentrations ranging from 100 fM to 1 nM. Hence, the proposed strategy has potential as an efficient and actual platform for small molecule analysis.

A facile signal-on electrochemical DNA biosensor has been developed for ultrasensitive detection of pathogenic bacteria using an Exo III-assisted autonomous multiple-cycle amplification strategy. The strategy relies on pathogens and aptamer binding-initiated release of a trigger, which combines with the 3′-protruding terminus of the hairpin probe 1, leading to the formation of double-stranded DNA with a blunt 3′ terminus which starts the Exo III-assisted multiple signal amplification reaction. In addition, hairpin probe 2 labeled with an electroactive reporter at the middle of the loop region is ingeniously designed to contain a short hairpin-embedded segment, which can fold into a hairpin structure via an Exo III-assisted cleavage reaction, thus bringing the redox molecule in proximity to the electrode surface for “signal-on” sensing. Under optimal conditions, this biosensor exhibits a very low detection limit as low as 8 cfu mL−1 and a wide linear range from 1.0 × 101 to 1.0 × 107 cfu mL−1 of target pathogenic bacteria. As far as we know, this is the first time that the Exo III-assisted autonomous multiple-cycle amplification strategy has been used for signal-on electrochemical sensing of pathogenic bacteria. In addition, the proposed sensor can also be used for highly sensitive detection of other targets by changing the aptamer sequence, such as nucleic acids, proteins and small molecules. Therefore, the proposed signal-on electrochemical sensing strategy might provide a simple and practical new platform for detection of pathogenic bacteria and related biological analysis, food safety inspection and environmental monitoring.

We have formulated a rapid and high-efficiency fluorescent biosensing platform based on a target-activated triple-helix molecular switch (THMS)-conformation-change-induced exponential rolling circular amplification (RCA) strategy for the ultrasensitive detection of miR-21. In this strategy, there are several aspects that are worthwhile. First, the functionalized THMS, comprising a typical triplex structure (T-A·T), specific recognition sequence for nicking endonuclease, complementary sequence for miR-21, and RCA product-annealing sequence, was concurrently used to perform signal transduction with one fluorophore and one quencher. As compared to the traditional double-helix molecular switches or molecular beacons, one of the biggest differentiating factors is that the properties of THMSs are independent of any specific binding sequence that they may contain. As far as we know, this is the first time that an ingeniously designed THMS not only contains the primer for exponential RCA, but also functions as the tracer for fluorescence assay. In the presence of miR-21, targets can induce conformation changes in THMS with the release of the trapped DNA segment (P), which, in turn, can activate the first run of the RCA process. Meanwhile, the RCA reaction is also initiated by the formation of a similar primer (SP) as the trapped DNA through a continuous "extension-nicking" reaction. Secondly, the resultant first-generation RCA product consists of numerous tandem repeated regions that can attach to countless THMSs, resulting in the release of the trapped DNA segment (P) for initiating the second run of the RCA reaction. Significantly, a large amount of THMSs were continuously consumed to yield a remarkably strong fluorescent signal. In addition, this biosensor was demonstrated to exhibit improved sensitivity owing to the high efficiency and rapid amplification kinetics of the exponential RCA and high selectivity toward miR-21 with a limit of detection as low as 1.1 aM. Hence, the target-mediated THMS-conformation-change-initiated exponential RCA strategy presents an optimal detection performance toward analytes for potential applications in related fundamental research and clinical diagnosis.

A simple and robust fluorescence sensing strategy has been developed for the detection of pathogenic bacteria by the combination of the dual functionality of phi29 DNA polymerase with isothermal circular strand displacement polymerization (ICSDP). The strategy relies on target-triggered formation of a mature primer that initiates the cyclic strand displacement polymerization reaction with the aid of dual functional phi29; thus, amplified detection of the target can be achieved. To our knowledge, this work is the first report where dual functional phi29-assisted ICSDP has been employed for fluorescence sensing of pathogenic bacteria. It is worth noting that a hairpin pre-primer is introduced that can be trimmed into a mature primer for initiating ICSDP via the 3' → 5' proofreading exonuclease activity of phi29, which contributes to the ultrahigh specificity of the strategy owing to the elimination of the unwished nonspecific extension. On the basis of the present amplification strategy, our biosensor exhibits excellent specificity and sensitivity toward S. typhimurium with an excellent detection limit as low as 1.5 cfu mL-1. In addition, the strategy offers the advantages of a simplified operation, shortened analysis time, and highly sensitive detection of pathogens with only a one-step reaction. Furthermore, by redesigning the corresponding binding molecules, the proposed strategy can be easily extended for the detection of a wide spectrum of analytes. Hence, the dual functional phi29-assisted ICSDP strategy indeed creates a robust and convenient fluorescence sensing platform for the identification of pathogenic bacteria and related food safety analysis.

Uracil-DNA glycosylase (UDG) is a common glycosylase that can expressly recognize and remove damaged uracil bases, and the ultrasensitive detection of which is significant to maintain genomic stability and early clinical diagnosis of disease. Herein, we proposed a sensitive colorimetric sensing platform to detect UDG. Combined with target-manipulated drawstring DNAzyme and Au@Ag nanorods (Au@Ag NRs) indicator, we achieved in naked-eyes observation and ultrasensitive detection of UDG. As an extremely important post-transcriptional regulator, miRNAs are involved in a variety of crucial biological processes, and the abnormal expressions of miRNAs are closely related to a variety of diseases. In this work, for the first time, we designed a nucleic acid lock nanostructure for specific detection of miRNA-21, which changes the self-structure to "active conformation" by binding the target, in order to generate triggers to initiate the subsequent reaction. Emphatically, this flexible nucleic acid lock is capable of self-cleaving without the assistance of external component, overcoming the disadvantages of the complex design and requiring protease assistance in traditional nanostructure. Moreover, the combination of DNAzyme and RCA technology not only greatly improves the efficiency of signal amplification but also enables primer generation to simultaneous cascade RCA amplification. Additionally, the electrochemical detection technology based on silver nanoclusters overcomes the shortcomings of traditional detection methods such as low sensitivity and complex operation. The detection limit achieved was 9.3 aM with a wide dynamic response ranging from 10 aM to 100 pM (at the DPV peak of - 0.5 V), which is comparable to most of the reported studies. Therefore, our work provided an ultra-sensitive way for the detection of miRNAs using nanostructures and revealed an effective means for disease theranostics and cancer diagnosis. In this work, for the first time, we designed a nucleic acid lock nanostructure based on its self-structural transformation for the specific detection of miRNA. And the combination of DNAzyme and cascade RCA reaction greatly improved the signal amplification efficiency.

The cassava-alcohol fermentation process employing cassava requires nitrogen source to maximize yields by a commercial strain of S. cerevisiae TG1348. In this study, a factorial experimental design was used to assess a suitable nitrogen source for growth and fermentative performance of S. cerevisiae in cassava-ethanol fermentation. The alcohol fermentation time was about 39 hours for urea and ammonium acetate, which was 48 hours for ammonium chloride and ammonium sulphate. The fermentation time was reduced by 19% when using urea and ammonium acetate as nitrogen source. Ammonium acetate leaded to the highest alcohol yield, which was 4% higher than for ammonium sulphate. In addition, byproduct formation differed obviously between the nitrogen sources. The glycerol yields were similar for urea, ammonium sulphate and ammonium chloride but were 24% lower for ammonium acetate. However, glycerol yield for ammonium carbonate was higher than for other nitrogen sources. Clearly, in batch cultures the ammonium acetate not only increased ethanol generation, but also decreased glycerol generation. In order to understand why ammonium acetate promotes alcohol fermentation, acetic acid was added to different nitrogen sources. The weight loss effect of ammonium sulphate adding acetic acid and ammonium acetate as nitrogen source was the same. The fermentation time was shortened by adding acetic acid. And pH was increased by addition of acetic acid when ammonium sulfate and urea were used as nitrogen sources. The results showed that the acetate root plays an important role in ammonium acetate. The results of this study could facilitate the development of new strategies to control fermentation performance.

DNA walkers, one of the artificial molecular machines which are constructed via smart synthetic DNA, have attracted rapidly growing attention from researchers in the biosensing field. In this work, we design an Exonuclease III (Exo III)-aided target-aptamer binding recycling (ETBR) activated bipedal DNA machine for highly sensitive electrochemical detection of antibiotics. To the best of our knowledge, this is the first time that a bipedal DNA machine has been applied in electrochemical sensing for antibiotics. On the one hand, the bipedal DNA walker exceeds the conventional single swing arm DNA walker in terms of walking efficiency and stability. On the other hand, the ETBR strategy, along with efficient strand displacement amplification via stepwise movement of a bipedal DNA walker significantly promotes the signal amplification efficiency. Under optimal conditions, this bipedal DNA machine possesses a detection limit of 7.1 fM within a linear detection range from 10 fM to 100 pM. Moreover, this electrochemical biosensor is expected to detect a wide variety of analytes using the corresponding target recognition probes. Thus, our proposed strategy offers a highly efficient, stable and practical platform for small molecule analysis.

An ultrasensitive fluorescence sensing strategy for kanamycin (KANA) determination using endonuclease IV (Endo IV)-powered DNA walker, and hybridization chain reaction (HCR) amplification was reported. The sensing system consists of Endo IV-powered 3D DNA walker using for the specific recognition of KANA and the formation of the initiators, two metastable hairpin probes as the substrates of HCR and a tetrahydrofuran abasic site (AP site)-embeded fluorescence-quenched probe for fluorescence signal output. On account of this skilled design of sensing system, the specific binding between KANA and its aptamer activates DNA walker, in which the swing arm can move autonomously along the 3D track via Endo IV-mediated hydrolysis of the anchorages, inducing the formation of initiators that initiates HCR and the following Endo IV-assisted cyclic cleavage of fluorescence reporter probes. The use of Endo IV offers the advantages of simplified and accessible design without the need of specific sequence in DNA substrates. Under the optimal experimental conditions, the fluorescence biosensor shows excellent sensitivity toward KANA detection with a detection limit as low as 1.01 pM (the excitation wavelength is 486 nm). The practical applicability of this strategy is demonstrated by detecting KANA in spiked milk samples with recovery in the range of 98 to 102%. Therefore, this reported strategy might create an accurate and robust fluorescence sensing platform for trace amounts of antibiotic residues determination and related safety analysis. BackgroundVenous malformations (VMs), most of which associated with activating mutations in the endothelial cells (ECs) tyrosine kinase receptor TIE2, are characterized by dilated and immature veins with scarce smooth muscle cells (SMCs) coverage. However, the underlying mechanism of interaction between ECs and SMCs responsible for VMs has not been fully understood.MethodsHere, we screened 5 patients with TIE2-L914F mutation who were diagnosed with VMs by SNP sequencing, and we compared the expression of platelet-derived growth factor beta (PDGFB) and α-SMA in TIE2 mutant veins and normal veins by immunohistochemistry. In vitro, we generated TIE2-L914F-expressing human umbilical vein endothelial cells (HUVECs) and performed BrdU, CCK-8, transwell and tube formation experiments on none-transfected and transfected ECs. Then we investigated the effects of rapamycin (RAPA) on cellular characteristics. Next we established a co-culture system and investigated the role of AKT/FOXO1/PDGFB in regulating cross-talking of mutant ECs and SMCs.ResultsVMs with TIE2-L914F mutation showed lower expression of PDGFB and α-SMA than normal veins. TIE2 mutant ECs revealed enhanced cell viability and motility, and decreased tube formation, whereas these phenotypes could be reversed by rapamycin. Mechanically, RAPA ameliorated the physiological function of mutant ECs by inhibiting AKT-mTOR pathway, but also facilitated the nuclear location of FOXO1 and the expression of PDGFB in mutant ECs, and then improved paracrine interactions between ECs and SMCs. Moreover, TIE2 mutant ECs strongly accelerated the transition of SMCs from contractile phenotype to synthetic phenotype, whereas RAPA could prevent the phenotype transition of SMCs.ConclusionsOur data demonstrate a previously unknown mechanistic linkage of AKT-mTOR/FOXO1 pathway between mutant ECs and SMCs in modulating venous dysmorphogenesis, and AKT/FOXO1 axis might be a potential therapeutic target for the recovery of TIE2-mutation causing VMs.Graphical abstract

A fluorescence sensing strategy was described for the determination of melamine with ultrasensitivity and high specificity. This strategy relies on the combination of proximity ligation assay (PLA)-modulated thymine–melamine–thymine (T–melamine–T) triplex structure along with exonuclease III (Exo III)-aided recycling amplification. The reaction system involves three DNA probes (poly A, tDNA and molecular beacon (MB)). Probe ploy A is an adenine enriched sequence containing an apurinic/apyrimidinic site (AP site). It can provide a hydrophobic vacancy for specially recognizing melamine to bind the nucleobase through pseudo-base pairing and participate in PLA modulated T-melamine-T triplex structure. Probe tDNA can form T-melamine-T with probe poly A via PLA only in the presence of melamine. Probe MB is modified with a fluorophore at its 5′-terminus and a quencher at an internal position, which can self-hybridize to form a hairpin structure, positioning the fluorophore in close proximity to the quencher. Therefore, the binding of MB did not yield any fluorescence signal. However, in the presence of melamine, a T-melamine-T triplex structure is formed through PLA. Probe MB can hybridize with t T-melamine-T triplex structure to yield a substrate for interaction with Exo III. As a result, the T-melamine-T triplex structure is released. Next, the released T-melamine-T triplex structure will hybridize with another MB probe to achieve the Exo III-aided recycle amplification. This is the first example of PLA-modulated T-melamine-T triplex structure coupled with Exo III-aided recycling amplification. Under optimal conditions, the assay has a detection range over 4 orders of magnitude and a limit of detection of 4.4 nM. Moreover, the assay is rapid, simple and inexpensive. Hence, the strategy may open new avenues for ultrasensitive and highly specific detection of melamine encounted in food analysis.

In the present work, a simple, rapid, isothermal, and ultrasensitive homogeneous electrochemical biosensing platform for target Hg2+ detection was developed on the basis of an exonuclease III (Exo III)-aided target recycling amplification strategy. In the assay, a label-free hairpin probe (HP1) was ingeniously designed, containing a protruding DNA fragment at the 3′-termini as the recognition unit for target Hg2+. Also, the DNA fragment in the loop region and 5′-termini (Helper) could be used when a secondary target analog is introduced, but it is caged in the stem region of HP1 when without such a target. The produced secondary target Helper opened the methylene blue (MB)-labeled hairpin probe (HP2) and triggered the Exo III cleavage process, accompanied with the secondary target recycling. This accordingly resulted in the autonomous reduction of the electroactive material MB on the electrode, inducing a distinct decrease in the electrochemical signal. The current developed homogeneous strategy provides a means for the ultrasensitive electrochemical detection of Hg2+ down to the 227 pM level, with high selectivity. It could be further used as a general autocatalytic and homogeneous strategy toward the detection of a wide spectrum of analytes and may be associated with more analytical techniques.

In this work, a rapid, one-step and ultrasensitive signal-on fluorescence sensing for the detection of adenosine triphosphate (ATP) based on target-activated cascaded digestion amplification with Exo III aid was developed. Our strategy integrates the target-activated cascaded digestion amplification with Exo III assistance into a signal-on fluorescence sensing platform. This advance greatly enhances the specificity and significantly improves the sensitivity for ATP detection. Specifically, an arch probe containing an anti-ATP aptamer (Apt) and a trigger probe (T) is used for recognizing the target. In the presence of ATP, the cascaded digestion amplification with the help of Exo III was activated, generating a remarkably strong signal-on fluorescence response. Our strategy features several virtues. First, the background signal is effectively eliminated owing to the specially designed hairpin probes (one is named HAP, the other is named MB short for molecular beacon) successfully avoiding the effect of Exo III. Second, the combination of target-activation with Exo III-aided cascade digestion amplification is integrated with fluorescence sensing, affording high specificity and ultrasensitive detection of ATP. Third, our method achieved the reliable and accurate detection of ATP in human serum. Furthermore, under optimized conditions, the results suggest that the proposed fluorescence sensing exhibits excellent specificity and ultrasensitivity toward ATP with the limit of detection as low as 0.63 pM and the broad analytical range from 1 pM to 1.0 × 105 pM. This target-activated cascaded digestion amplification with Exo III assistance combined with the fluorescence sensing strategy paves a luciferous and practical way for detecting ATP and other analytes with trace amounts in bio-analysis and clinical biomedicine.

Uracil-DNA glycosylase (UDG) is a crucial damage repair enzyme that initiates the cellular base excision repair pathway that maintains the integrity of the genome. Abnormal UDG activity may induce the malfunction of uracil excision repair that is directly related to a range of diseases including cancers, genotypic diseases, and human immunodeficiencies. In this work, a simple, robust and cost effective biosensing platform for the ultrasensitive detection of UDG activity is established based on the combination of base excision repair-initiated primer generation for rolling circular amplification (RCA) with Endo IV-assisted signal amplification. In the presence of target UDG, UDG can catalyze the removal of uracil on a hairpin probe (HP) leaving an apurinic/apyrimidinic (AP site) which can be cleaved by Endo IV to generate a primer for triggering the RCA reaction. Subsequently, numerous AP site-embedded signal probes, acting as fluorescence-quenched probes, combine with the RCA products to perform signal transduction and quadradic signal amplification through an Endo IV-catalyzed cleavage reaction, thus significantly enhancing the fluorescence signal, which can be used for UDG activity screening. Under optimum conditions, this biosensor exhibits improved sensitivity toward target UDG with a detection limit of as low as 9.3 × 10-5 U mL-1 and a wide detection range across 5 orders of magnitude. Additionally, our biosensor demonstrates high selectivity toward UDG for simple, rapid, and low-cost detection. Furthermore, by redesigning the modification of HP and using of suitable endonuclease enzymes, this RCA coupled with Endo IV-assisted signal amplification strategy might be applied for the detection of various other targets, such as thymine DNA glycosylase, 8-oxoguanine DNA glycosylase, DNA methyltransferase, and so on. Hence, the proposed strategy provides a useful and versatile biosensing platform for the ultrasensitive detection of UDG activity and related fundamental biomedicine research and clinical diagnosis.

A novel fluorescence biosensing strategy for ultrasensitive and specific detection of pathogenic bacteria based on target-triggered enzymatic repairing amplification (ERA) has been developed. This strategy relies on target–aptamer binding mediated ERA reaction, which is carried out cyclically with the help of polymerase and two DNA repairing enzymes, uracil-DNA glycosylase (UDG) and endonuclease IV (Endo IV) to produce amplified fluorescence signal. In our assay, the specially designed hairpin probe (HAP) is used as DNA template responsible for producing a great quantity of reporter oligonucleotides and secondary primers, which can initiate a new cycle of polymerization-repairing amplification. Moreover, by the combination of polymerase-catalyzed incorporation of lesion bases with UDG and Endo IV-assisted ERA, multiple cycle of amplification of the recognition event is achieved, enabling ultrasensitive detection of pathogenic bacteria. Under optimal conditions, this biosensor exhibits ultrasensitivity toward target pathogenic bacteria with detection limits of 9.86 cfu mL−1 and a detection range of 5 orders of magnitude. Additionally, the biosensor has the ability of combating nonspecific background. Furthermore, an archer probe containing the anti-target aptamer sequence and a primer sequence is designed, which translates the binding of target to aptamer into the presence of primer sequence, enabling the detection of various targets, such as protein, DNA, small molecular, and any substance possessing its aptamer. Hence, the proposed target-triggered ERA-based signal-on fluorescence sensing strategy indeed create a versatile and useful platform for detection of pathogenic bacteria, related food safety analysis and clinical diagnosis.

The authors describe a label-free electrochemical aptasensor for ultrasensitive and highly specific detection of the antibiotic based on cascade enzymatic recycling couple with DNAzyme amplification. The assay involves two sequential reactions: the first reaction is a λ exonuclease-assisted cyclic digestion reaction triggered by target-aptamer binding. The second reaction is a nicking endonuclease-aided cyclic nicking reaction, which produces a large amount of G-rich nucleic acid segments. These form a G-quadruplex/hemin complex in the presence of K(I) ions and hemin. Because the G-quadruplex/hemin complex acts as a horseradish peroxidase-mimicking DNAzyme with excellent redox activity, the electrochemical signal transduction is accomplished due to the electroreduction of H2O2. It appears that this work is the first example that cascade enzymatic recycling coupled to DNAzyme amplification is used for antibiotic detection. The aptasensor was applied to the quantitation of kanamycin and gave a response that is linear in the 1 pM to 10 nM kanamycin concentration range, with a detection limit as low as 0.5 pM. The working voltage (vs. Ag/AgCl) at which data can be acquired best is −0.35 V. The assay offers the advantages of remarkably improved sensitivity, use of affordable instrumentation, and simplified operation without the need for electrochemical labeling or addition of labile reagents. Thus, cascade enzymatic recycling coupled to DNAzyme amplification represents a versatile platform for highly sensitive and specific antibiotics detection.

Objective In this study, the objective is to construct graphene-oxide-coated titanate on titanium foils as drug vehicle to enhance cell proliferation and osteo-differentiation of rat bone mesenchymal stem cells (rBMSCs). Methods Graphene oxide (GO) sheets obtained using the modified Hummer’s method and characterized by AFM were coupled with bioactive titanate on Ti implants (GO-Ti) pretreated by alkali, followed by reduction (rGO-Ti). They were characterized by Raman spectroscopy, XPS, SEM, FTIR and contact angle. After dexamethasone (DEX) was loaded onto them (DEX-GO-Ti and DEX-rGO-Ti), cell proliferation of rBMSCs on them was evaluated by CCK-8 and F-actin staining, and differentiation through alkaline phosphatase activity, mRNA expression, and calcium nodules. Results The obtained GO sheets were monolayers from AFM. Raman spectra exhibited two prominent peaks at D and G bands, and the I(D)/I(G) ratios increased from 0.96 to 1.68 after reduction. XPS proved the existence of oxygenated functional groups for GO-Ti and the reduction of their intensity for rGO-Ti. From SEM, GO and rGO were evenly coated on nanostructures. DEX-GO-Ti absorbed most amount of DEX and released in a sustained manner. CCK-8 results showed that DEX-GO-Ti showed excellent performance on promoting cell proliferation. RMBSCs on DEX-GO-Ti presented greatly high expression of calcium, proteins and mRNA related to osteogenic differentiation. Significance GO coated titanate nanostructrues on surfaces of Ti foils by a simple self-assembly method, showed excellent vechiles for DEX. The construct promoted proliferation and accelerated osteogenic differentiation of rBMSCs, and would be prosperous for their further applications.
