Abstract Solution-processed quantum dot light-emitting diodes (QLEDs) hold great potential as competitive candidates for display and lighting applications. However, the serious energy disorder between the quantum dots (QDs) and hole transport layer (HTL) makes it challenging to achieve high-performance devices at lower voltage ranges. Here, we introduce “giant” fully alloy CdZnSe/ZnSeS core/shell QDs (size ~ 19 nm) as the emitting layer to build high-efficient and stable QLEDs. The synthesized CdZnSe-based QDs reveal a decreased ground-state band splitting, shallow valence band maximum, and improved quasi-Fermi level splitting, which effectively flatten the energy landscape between the QD layer and hole transport layer. The higher electron concentration and accelerated hole injection significantly promote the carrier radiative recombination dynamics. Consequently, CdZnSe-based device exhibits a high power conversion efficiency (PCE) of 27.3% and an ultra-low efficiency roll-off, with a high external quantum efficiency (EQE) exceeding 25% over a wide range of low driving voltages (1.8-3.0 V) and low heat generation. The record-high luminance levels of 1,400 and 8,600 cd m-2 are achieved at bandgap voltages of 100% and 120%, respectively. Meanwhile, These LEDs show an unprecedented operation lifetime T95 (time for the luminance to decrease to 95%) of 72,968 h at 1,000 cd m-2. Our work points to a novel path to flatten energy landscape at the QD-related interface for solution-processed photoelectronic devices.

Abstract CDK4/6i, the first-line drug for treating ERα-positive breast cancer, significantly improves clinical outcomes. However, CDK4/6i resistance often develops and remains a major hurdle, and the underlying mechanisms remain challenging to fully investigate. Here, we used Genome-wide CRISPR/Cas9 library screening combined with single-cell sequencing to screen for molecules mediating CDK4/6i resistance and identified METTL14 as a determinant of CDK4/6i sensitivity. Clinical samples and datasets were analyzed and in vitro and in vivo experiments were performed to confirm the critical function of METTL14 in CDK4/6i resistance. Mechanistically, METTL14 can induce an increase in E2F1 expression in breast cancer cells via an m6A IGF2BP2-dependent mechanism and thus promote CDK4/6i resistance. Furthermore, through a small molecule screen, a novel METTL14 inhibitor named WKYMVM, which can restore sensitivity to CDK4/6i in CDK4/6i-resistant breast cancer cells, was identified. Treatment with folate-conjugated liposomes targeting breast cancer cells that contained both a CDK4/6i and WKYMVM revealed the synergistic effect of METTL14 inhibition with CDK4/6i therapy in a CDK4/6i-resistant PDX model. Together, our findings reveal the mechanism of CDK4/6i resistance and provide a strategy for overcoming CDK4/6i resistance via METTL14 inhibition. Graphical Abstract

In order to explore the effects of vacuum heat treatment and gluten on the gel properties of myofibrillar protein (MP), the effects of vacuum heat treatment and gluten addition on tilapia MP gel were studied by testing texture, water retention, intermolecular forces and other indicators. Under the condition of water bath, the addition of 0.7% gluten significantly improved the hardness and gel strength of MP. The microstructure results showed that the gluten made the MP gel network tend to be flat and the holes decreased, which was conducive to the formation of gel. Compared with the water bath heating, the cohesion, elasticity and gel strength of MP gel were significantly improved after 1 h of one-stage vacuum heat treatment. After 2 h of vacuum heat treatment, the cohesion and gel strength of MP gel were further improved. In addition, vacuum heat treatment and gluten addition improved the moisture state of MP gel, the content of T22 fixed water decreased, and the migration of T22 fixed water and T23 free water was restricted, resulting in the stability of the moisture in the gel. The results of intermolecular forces showed that the interaction between ionic bond and hydrophobic interaction was enhanced, which promoted the gelation of MP. In summary, vacuum processing promotes the structure extension of MP and the formation of a more compact three-dimensional gel network.

Abstract Porphyromonas gingivalis (P. gingivalis), the main pathogen responsible for periodontitis, is linked to systemic disorders via the oral-gut axis. Short-chain fatty acids (SCFAs) are vital for gut health, but their role in P. gingivalis-induced gut disorders remains unclear. This study utilized metabolomics and 16 S rRNA sequencing to explore gut microbiota and SCFAs levels in P. gingivalis-induced periodontitis mouse models. Significant changes were observed in gut, including a reduction in SCFAs-producing bacteria, such as Lactobacillus, Ligilactobacillus, Allobucalum, and a notable decrease in Firmicutes and Actinobacteriota. The intestinal permeability tests and histological analyses revealed that periodontitis led to epithelial inflammation, reduced mucin secretion, and compromised gut barrier integrity. In vitro experiments with Caco-2 cells co-cultured with P. gingivalis showed that the bacterium disrupted cellular junctions by impairing autophagy, specifically through the ATG5-LC3 pathway, leading to decreased expression of tight junction proteins and reduced SCFA absorption. Remarkably, rapamycin treatment both in vitro and in vivo restored gut barrier function by enhancing autophagy, increasing tight junction protein expression, and promoting SCFAs absorption via MCT1 and SMCT1, alongside GPR43/GPR109a pathway activation. These findings reveal autophagy’s novel role in regulating SCFAs metabolism in P. gingivalis-induced gut dysbiosis, offering insights for preventing and treating periodontitis-related systemic diseases.

Extensive research has been performed on the tensile properties of carbon fibre-reinforced polymer (CFRP) material used in both standalone and rehabilitation applications. Limited knowledge and design guidelines exist on the flexural behaviour of CFRP cable strands (herein strand) and reinforcing bars. This knowledge gap hinders the widespread use of strands and CFRP bars in prestressed structures and restricts their market share compared to reinforcing steel members. To address this gap, this study investigates the flexural behaviour of CFRP strands and reinforcing bars using a combination of experimental and numerical methods. A standard three-point bending test is employed to measure the flexural strength and ultimate curvature radius of strands and bars. The experimental study included strands of diameters 10.2 mm and 15.2 mm, and CFRP bars of diameters 9 mm, 12 mm, and 14 mm. Finite element analysis has been conducted to further investigate the flexural performance and failure mechanisms of the specimens. The test results indicate that the ultimate bending radius of the strands is approximately one-half of that of the bar, while their ultimate bending strength surpasses the bars by at least 10 %. Optimal lay lengths for the 10.2 mm and 15.2 mm cable strands are proposed based on the simulation results.

Acute kidney injury (AKI) is characterized by a rapid loss of renal metabolic function and a high mortality rate. Although significant progress has been made in developing targeted drugs for AKI treatment, issues such as inadequate antioxidant effects and poor renal enrichment efficiency remain. Nanomotors can enhance drug delivery efficiency in AKI treatments through self-propulsion in the microenvironment or via external stimuli. We reviewed recent progress in the targeted treatment of AKI with nanomotors, focusing on their contribution to targeted drug delivery at different stages and combined treatments. Current limitations and future development directions are also discussed.

Abstract Background The sustainable supply of medicinal plants is important, and cultivating and domesticating them has been suggested as an optimal strategy. However, this can lead to a loss of genetic diversity. Tripterygium wilfordii Hook. f. is a medicinal plant commonly used in traditional Chinese medicine, but its wild populations are dwindling due to excessive harvesting. To protect the species and meet the increasing demand, it is urgent to cultivate it on a large scale. However, distinguishing between T. wilfordii and T. hypoglaucum, two similar species with different medicinal properties, is challenging. Therefore, it is crucial to understand the genetic diversity and population structure of these species for their sustainable utilization. Results In this study, we investigated the genetic diversity and population structure of the two traditional medicinal semiwoody vines plant species, Tripterygium wilfordii and T. hypoglaucum, including wild and cultivated populations using chloroplast DNA (cpDNA) sequences and microsatellite loci. Our results indicated that the two species maintain a high level of genetic divergence, indicating possible genetic bases for the different contents of bioactive compounds of the two species. T. wilfordii showed lower genetic diversity and less subdivided population structures of both markers than T. hypoglaucum. The potential factors in shaping these interesting differences might be differentiated pollen-to-seed migration rates, interbreeding, and history of population divergence. Analyses of cpDNA and microsatellite loci supported that the two species are genetically distinct entities. In addition, a significant reduction of genetic diversity was observed for cultivated populations of the two species, which mainly resulted from the small initial population size and propagated vegetative practice during their cultivation. Conclusion Our findings indicate significant genetic divergence between T. wilfordii and T. hypoglaucum. The genetic diversity and population structure analyses provide important insights into the sustainable cultivation and utilization of these medicinal plants. Accurate identification and conservation efforts are necessary for both species to ensure the safety and effectiveness of crude drug use. Our study also highlighted the importance of combined analyses of different DNA markers in addressing population genetics of medicinal plants because of the contrasts of inheritance and rates of gene flow. Large-scale cultivation programs should consider preserving genetic diversity to enhance the long-term sustainability of T. wilfordii and T. hypoglaucum. Our study proposed that some populations showed higher genetic diversity and distinctness, which can be considered with priority for conservation and as the sources for future breeding and genetic improvement.

Precision pesticide application mainly relies on canopy volume, resulting in varied application effectiveness across different density areas of orchard trees. This study examined pesticide application effectiveness based on the spray wind, canopy volume, and leaf area within the canopy, providing variable bases for precise regulation of spray wind and pesticide dosage. The study addresses the knowledge gap by utilizing laser detection and ranging (LiDAR) to measure the thickness and leaf area of orchard tree canopies. The spray experiments were conducted on canopies of different regions, using an air-assisted sprayer with varying fan speeds of 1381 r/min, 1502 r/min, and 1676 r/min. The deposition effects were analyzed using water-sensitive papers. The inlet air speed within the canopy did not increase proportionally when the spray fan speed increased, and it showed a significant variation in locations with sparse foliage. Furthermore, droplets exhibited abnormal median volume diameters of the canopy regions with lower wind loss rates and smaller leaf areas. The influences were in the order of canopy thickness, leaf area, and inlet air speed on the cumulative deposition of droplets on both sides of the water-sensitive papers, as well as the ratio of deposition between the two sides, from big to small, are inlet air speed, leaf area, and canopy thickness. The study provides a scientific foundation for air control in precision pesticide application in apple orchards and contributes to the rapid development of precision spraying technologies.

Feed efficiency (FE) is a crucial trait in pig production that influences both economic viability and environmental sustainability. The jejunum, an essential organ for nutrient absorption, plays a significant role in determining FE by affecting how pigs process and utilize feed. To explore the genetic and regulatory mechanisms behind FE, we conducted an integrative multi-omics study using RNA sequencing (RNA-seq) and ATAC sequencing (ATAC-seq) on pigs with high and low FE. By comparing gene expression and chromatin accessibility profiles in the jejunum, we identified key differentially expressed genes (DEGs) and differentially accessible regions (DARs) associated with lipid metabolism and immune function, both of which are critical pathways for efficient growth. Notably, we identified transcription factors such as GATA4 and EHF and genes like SCARB1 and GRXCR1 that may play regulatory roles in FE. Our findings provide novel insights into the molecular mechanisms governing FE in pigs, offering potential targets for genetic selection and nutritional interventions to enhance feed efficiency and sustainability in pig production.

Ticks are important vectors for many pathogens affecting humans and animals. This study was aimed at investigating the species composition, genetic diversity, and potential transmission risks of tick-borne pathogens in the Guangxi Zhuang Autonomous Region, China. A total of 980 ticks were collected from 14 prefecture-level cities in Guangxi between March and July 2021. Ticks were identified morphologically and genetically, and the population diversity of Rhipicephalus microplus was analyzed according to 16S rDNA, COXI , and ITS-2 gene sequences. Population genetics was assessed through F ST , AMOVA, and phylogenetic analyses. PCR was used to detect five common tick-borne pathogens: Rickettsia , Babesia , Theileria , Borrelia burgdorferi , and Anaplasma phagocytophilum . Rhipicephalus microplus was the dominant tick species, and was observed in 92.04% of all samples. Genetic diversity analysis revealed higher haplotype diversity for the 16S rDNA and COXI genes than the ITS-2 gene; these findings suggested a faster evolutionary rate for the first two markers. Population genetic differentiation was observed between Rhipicephalus microplus populations in Guilin, Hechi, and other regions, probably because of geographic isolation. Pathogen analysis revealed a 13.27% overall infection rate, and identified three Rickettsia , two Babesia , and three Theileria species. Co-infection with Theileria and Rickettsia was also detected. Notably, neither Borrelia nor Anaplasma was found in the tick samples. Phylogenetic analyses confirmed the distinctiveness of the Rickettsia and Piroplasma groups. This study provides a comprehensive assessment of tick species diversity and tick-borne pathogens in Guangxi, highlighting the potential health risks posed by Rhipicephalus microplus . The findings emphasize the need for expanded tick surveillance, enhanced pathogen detection methods, and integrated management strategies to mitigate the spread of tick-borne diseases. Future research should include the collection of free questing ticks and metagenomic approaches to provide a more comprehensive understanding of pathogen risk in the region.

Developing eco-friendly polymeric plasticizers with excellent migration resistance is one of the research hotspots in the polyvinyl chloride (PVC) industry. A low-molecular-weight biobased polyester rubber (LMW-BPR) was synthesized from five biobased polyester monomers in a 100-L reactor and evaluated as a potential eco-friendly polymeric plasticizer for PVC. The obtained LMW-BPR is an amorphous polyester material with a low glass transition temperature of −48°C and a molecular weight of 22 kg/mol, which is lower than that of existing polyester rubber but higher than those of most polyester plasticizer commodities. Plasticized PVC composites with a total plasticizer content of 50 phr were prepared by using the mixture of LMW-BPR and di-isononyl cyclohexane-1,2-dicarboxylate (DINCH, an eco-friendly monomeric plasticizer commodity) as the plasticizer. The migration resistance test showed that the migration loss of plasticizer in plasticized PVC composite decreased significantly with the increase of LMW-BPR content. When the content of LMW-BPR reaches 30 phr, the plasticized PVC composites are almost nonmigratory. In addition, compared with PVC composite plasticized by pure DINCH, co-plasticized PVC composites containing LMW-BPR exhibit higher tensile strength and thermal stability, and their flexibility, low-temperature resistance and biocompatibility are also maintained at a similar level to the former. Overall, LMW-BPR is an effective eco-friendly polymeric plasticizer for PVC and also sustainable and scalable, thus it is worthy of wide application.

BackgroundEmerging evidence have suggested that dietary habits have potential implication on the development of Alzheimer’s disease (AD). However, elucidating the causal relationship between specific dietary factors and AD risk remains a challenge. Therefore, our study endeavors to investigate the causal association between dietary habits and the risk of AD.Materials and methodsWe analyzed data on 231 dietary habits sourced from the UK Biobank and MRC-IEU, and AD data obtained from the FinnGen database. Employing a framework based on the classic two-sample Mendelian randomization (MR) study, we utilized the inverse-variance weighted (IVW) method as the primary analysis. Additionally, we conducted Steiger filtering and other methods to mitigate horizontal pleiotropy. The robustness of our overall findings was confirmed through multiple sensitivity analysis methods, and forward MR and reverse MR to address potential reverse causality bias.ResultsOur study evaluated the causal effect between 231 dietary habits involving over 500,000 participants of European ancestry, and 10,520 AD cases. Only oily fish intake demonstrated a significant protective causal relationship with AD following FDR correction (raw p-value = 1.28e-4, FDR p-value = 0.011, OR = 0.60, 95%CI: 0.47–0.78). Additionally, six dietary habits potentially influenced AD risk, with protective causal effects observed for average monthly intake of other alcoholic drinks (raw p-value = 0.024, FDR p-value = 0.574, OR = 0.57, 95%CI: 0.35–0.93) and tea intake (raw p-value = 0.047, FDR p-value = 0.581, OR = 0.78, 95%CI: 0.603–1.00). Conversely, detrimental causal effects were observed for the average weekly champagne plus white wine intake (raw p-value = 0.006, FDR p-value = 0.243, OR = 2.96, 95%CI: 1.37–6.38), Danish pastry intake (raw p-value = 0.036, FDR p-value = 0.574, OR = 13.33, 95%CI: 1.19–149.69), and doughnut intake (raw p-value = 0.039, FDR p-value = 0.574, OR = 7.41, 95%CI: 1.11–49.57). Abstract Background Herb-drug interactions may result in increased adverse drug reactions or diminished drug efficacy, especially for drugs with a narrow therapeutic index such as warfarin. The current study investigates the effects of sodium ferulate for injection (SFI) on anticoagulation of warfarin from aspects of pharmacodynamics and pharmacokinetics in rats and predicts the risk of the combination use. Methods Rats were randomly divided into different groups and administered single- or multiple-dose of warfarin (0.2 mg/kg) with or without SFI of low dose (8.93 mg/kg) or high dose (26.79 mg/kg). Prothrombin time (PT) and activated partial thromboplastin time (APTT) were detected by a blood coagulation analyzer, and international normalized ratio (INR) values were calculated. UPLC-MS/MS was conducted to measure concentrations of warfarin enantiomers and pharmacokinetic parameters were calculated by DAS2.0 software. Results The single-dose study demonstrated that SFI alone had no effect on coagulation indices, but significantly decreased PT and INR values of warfarin when the two drugs were co-administered (P 0.05). C max and AUC of R/S-warfarin decreased but CL increased significantly in presence of SFI (P 0.05). Conclusions The present study implied that SFI could accelerate warfarin metabolism and weaken its anticoagulation intensity in rats.

Background/Objectives: Periodontitis is a common oral disease marked by gingival inflammation and alveolar bone loss. This study evaluated the efficacy of chamomile tincture and lidocaine hydrochloride (CLH) gel in mitigating periodontal inflammation and bone loss and uncovered the molecular mechanisms involved, both in vitro and in vivo. Methods: A periodontitis model was induced in Sprague Dawley rats by ligating the mandibular first molars. Sixty rats were divided into four groups: control (C), periodontitis (PD), periodontitis + CLH gel once daily (G1), and periodontitis + CLH gel thrice daily (G3). Clinical, micro-computed tomography (micro-CT), biological, and histological evaluations were performed, focusing on osteoclastogenesis, osteogenesis, and inflammatory cytokine production. The effect of CLH gel on inflammatory responses in RAW264.7 cells was also assessed through co-culture assays under Porphyromonas gingivalis (P. gingivalis) infection, with RNA-sequencing, qPCR, and Western blot analyses to explore underlying mechanisms. Results: CLH gel significantly reduced gingival and systemic inflammation and mitigated bone loss by enhancing the bone volume to tissue volume ratio and trabecular thickness via the RANKL/OPG axis in rats. The G3 group showed marked reductions in osteoclasts and increases in osterix-positive cells compared to other groups. In vitro, CLH gel reduced the inflammatory phenotype of macrophages in the periodontitis microenvironment by modulating Type II interferon (IFN-γ) networks. Conclusions: CLH gel reduced inflammation and bone loss in rat periodontitis, promoting osteogenesis and inhibiting osteoclastogenesis. It also suppressed macrophage inflammation via Type II interferon networks under P. gingivalis stimulation. These findings suggest that CLH gel has potential as an adjunctive therapy for periodontitis.

Abstract Valencene is a natural sesquiterpene with desirable bioactivity and aroma, making it a valuable ingredient in the food and cosmetics industries. Traditionally, valencene was extracted from the citrus fruits, and its applications were restricted by the low concentrations in natural sources and high costs for extraction. Photosynthetic biomanufacturing represents a promising route for efficient and stable production of valencene, while cyanobacteria have been considered one of the most promising platforms regarding biotechnological routes for the direct conversion of CO2. In this work, we engineered Synechocystis sp. PCC 6803 to synthesize valencene. By introducing a heterologous valencene synthase and modifying the native MEP pathway, we obtained an efficient cyanobacterial cell factory that produced 154 mg/L valencene during a semi‐continual cultivation, with an average productivity of 4.3 mg/L/day, and the cell factory exhibited robust growth and production in non‐sterilized conditions. We also achieved the production of other sesquiterpenes including bisabolene, amorpha‐4,11‐diene, farnesene, and nerolidol by engineered cyanobacteria with enhanced MEP pathway flux, showing promising potentials as a universal chassis.

In Rosaceae, the replacement of the traditional four-subfamily division (Amygdaloideae or Prunoideae, Maloideae, Rosoideae, and Spiraeoideae) by the three-subfamily division (Dryadoideae, Rosoideae, and Amygdaloideae), the circumscription, systematic position, and phylogeny of genera in Maleae need to be reconsidered. The study aimed to circumscribe Maleae, pinpoint its systematic position, and evaluate the status of all generally accepted genera in the tribe using complete chloroplast genome data. Results indicated that Maleae consisted of pome-bearing genera that belonged to Maloideae as well as four genera (Gillenia, Kageneckia, Lindleya, and Vauquelinia) that were formerly considered to be outside Maloideae. The tribe could be subdivided into four subtribes: Gilleniinae (Gillenia), Lindleyinae (Kageneckia and Lindleya), Vaugueliniinae (Vauquelinia), and Malinae (all other genera; the core Maleae). Among the 36 recognized genera, Aria, Docyniopsis, Chamaemespilus, and Mespilus were not considered distinct and more research is needed to determine the taxonomic status of Rhaphiolepis from Eriobotrya. Within the core Maleae, five groups were revealed, whereas Sorbus L. was split as its members belonged to different groups.

Abstract Glucose is the most abundant monosaccharide, serving as an essential energy source for cells in all domains of life and as an important feedstock for the biorefinery industry. The plant-biomass-sugar route dominates the current glucose supply, while the direct conversion of carbon dioxide into glucose through photosynthesis is not well studied. Here, we show that the potential of Synechococcus elongatus PCC 7942 for photosynthetic glucose production can be unlocked by preventing native glucokinase activity. Knocking out two glucokinase genes causes intracellular accumulation of glucose and promotes the formation of a spontaneous mutation in the genome, which eventually leads to glucose secretion. Without heterologous catalysis or transportation genes, glucokinase deficiency and spontaneous genomic mutation lead to a glucose secretion of 1.5 g/L, which is further increased to 5 g/L through metabolic and cultivation engineering. These findings underline the cyanobacterial metabolism plasticities and demonstrate their applications for supporting the direct photosynthetic production of glucose.

Abstract Background l-Methionine is the only bulk amino acid that has not been industrially produced by the fermentation method. Due to highly complex and strictly regulated biosynthesis, the development of microbial strains for high-level l-methionine production has remained challenging in recent years. Results By strengthening the l-methionine terminal synthetic module via site-directed mutation of l-homoserine O-succinyltransferase (MetA) and overexpression of metA fbr, metC, and yjeH, l-methionine production was increased to 1.93 g/L in shake flask fermentation. Deletion of the pykA and pykF genes further improved l-methionine production to 2.51 g/L in shake flask fermentation. Computer simulation and auxotrophic experiments verified that during the synthesis of l-methionine, equimolar amounts of l-isoleucine were accumulated via the elimination reaction of cystathionine γ-synthetase MetB due to the insufficient supply of l-cysteine. To increase the supply of l-cysteine, the l-cysteine synthetic module was strengthened by overexpression of cysE fbr, serA fbr, and cysDN, which further increased the production of l-methionine by 52.9% and significantly reduced the accumulation of the byproduct l-isoleucine by 29.1%. After optimizing the addition of ammonium thiosulfate, the final metabolically engineered strain MET17 produced 21.28 g/L l-methionine in 64 h with glucose as the carbon source in a 5 L fermenter, representing the highest l-methionine titer reported to date. Conclusions In this study, a high-efficiency strain for l-methionine production was derived from wild-type Escherichia coli W3110 by rational metabolic engineering strategies, providing an efficient platform for the industrial production of l-methionine.

Tomatoes are prone to mechanical damage due to improper gripping forces during automated harvest and postharvest processes. To reduce this damage, a dynamic viscoelastic model based on long short-term memory (LSTM) is proposed to fit the dynamic compression stress relaxation characteristics of the individual fruit. Furthermore, the classical stress relaxation models involved, the triple-element Maxwell and Caputo fractional derivative models, are compared with the LSTM model to validate its performance. Meanwhile, the LSTM and classical stress relaxation models are used to predict the stress relaxation characteristics of tomato fruit with different fruit sizes and compression positions. The results for the whole test dataset show that the LSTM model achieves a RMSE of 2.829×10−5 Mpa and a MAPE of 0.228%. It significantly outperforms the Caputo fractional derivative model by demonstrating a substantial enhancement with a 37% decrease in RMSE and a 36% reduction in MAPE. Further analysis of individual tomato fruit reveals the LSTM model’s performance, with the minimum RMSE recorded at the septum position being 3.438×10−5 Mpa, 31% higher than the maximum RMSE at the locule position. Similarly, the lowest MAPE at the septum stands at 0.375%, outperforming the highest MAPE at the locule position by a significant margin of 90%. Moreover, the LSTM model consistently reports the smallest discrepancies between the predicted and observed values compared to classical stress relaxation models. This accuracy suggests that the LSTM model could effectively supplant classical stress relaxation models for predicting stress relaxation changes in individual tomato fruit.

The teacher’s pets are a common occurrence in the field of education. To investigate the preferences teachers exhibit toward certain children, the study focused on kindergarten teachers and employed a mixed research methodology. Initially, qualitative interviews were conducted with 15 kindergarten teachers to identify specific criteria influencing teacher preferences. Subsequently, A comprehensive model of teacher’s pets was developed through a questionnaire survey involving 463 participants. This model encapsulated 32 distinct indicators, categorized into 7 types: children with good appearance (GA), exceptional abilities (OA), commendable conduct (GC), proactive and enthusiastic demeanor (PE), compliant and carefree nature (OC), children from vulnerable groups (VC), and those influenced by their parents (PI). The resulting model demonstrated a sound structure. Not only did it validate existing findings, but it also expanded upon the identified types of teacher’s pets. An analysis based on game theory revealed the weighted combinations, highlighting the top three types of teacher’s pets: children influenced by parental factors (24.3%), proactive and enthusiastic individuals (15.7%), and obedient, carefree children (14.8%), respectively. Conversely, the representation of vulnerable-concerned children (11.1%) was the lowest among the identified types.

Fritillaria ussuriensis Maxim. (F.M.) has been widely used in both food and medication for more than 2000 years. In order to achieve its comprehensive utilization and investigate the structural characterization and biology activity, response surface methodology (RSM) was used to optimize the ultrasound-assisted extraction conditions of F.M. polysaccharides. The optimal extraction conditions were ultrasonic power of 174.2 W, ratio of liquid to material of 40.7 mL/g and ultrasonic time of 82.0 min. In addition, a neutral polysaccharide F-1 was obtained, and its structure characterization, antioxidant and immunological activity were evaluated. The structural properties of the polysaccharide were characterized by UV, IR, GC-MS, NMR and AFM. Monosaccharide composition of F-1 (MW 18.11 kDa) was rhamnose, arabinose, glucosamine hydrochloride, galactose, and glucose which under the ratio of 0.9: 3.8: 0.2: 2.9: 92.2. The fractions of F-1 were mainly linked by → 6)-α-D-Glcp-(1 → with branch chain α-D-Glcp-(1 → 4)-α-D-Glcp-(1 → and 4,6)-α-D-Glcp-(1 → residues. Moreover, F-1 has a significant scavenging activity, which can clear hydroxyl radicals, superoxide anion, DPPH and ABTS. In addition, the immunological activity showed that F-1 had an effect on macrophage phagocytic activity. And it can increase the release of inflammatory factors including TNF-α, IL-1β and IL-6. F-1 is a novel polysaccharide with significant activity in antioxidant and immunological activity, which has great potential for antioxidant and immunizer in food, pharmaceutical and cosmetic industries. The study can provide a methodological basis for polysaccharide research and theoretical basis for the industrialized production and practical application.

Background: The emergence of dengue fever has prompted significant public health responses, highlighting the need for a comprehensive understanding of One Health in addressing vector-borne diseases. China's experience in dengue control and prevention programs offers valuable insights into the successful integration of multidisciplinary strategies. Aims: The review aims to: (1) systematically analyze lessons from China's dengue control and prevention programs, focusing on the integration of these efforts with the One Health approach; (2) underscore the reasons of optimizing the dengue control and prevention program; (3) highlight the alignment of China's dengue control strategies with the One Health framework; (4) contribute to global efforts in combating dengue, providing scientific evidence and strategic recommendations for other regions facing similar challenges. Results: Through a comprehensive literature review and expert interviews, this study found China's approach to dengue control and prevention implemented through a hierarchical system led by the government, with collaborative efforts across multiple departments. This multi-sectoral collaboration mechanism enables the technical interventions well executed by health and disease control institutions, optimizing the integration of multiple cost-effeteness approaches, such as case management, early detection and outbreak response, reducing local transmission, and minimizing severe cases and fatalities. It was found that community participation and public health education have played a vital role in raising awareness, promoting personal protective measures, and enhancing the overall effectiveness of control efforts. The implementation of these integrated interventions has resulted in reduced dengue cases and improved capacity of outbreak response. China's dengue control strategies under the One Health framework, with focus on interdisciplinary collaboration, incorporated environmental and ecological interventions, which reduced mosquito breeding sites and improved sanitation. The findings of the review underscore the need for continuous improvement in early warning systems, scientific research, and the adoption of the One Health approach to address emerging challenges posed by climate change and the cross-border spread of infectious diseases. Conclusion: China's dengue control and prevention programs provide a compelling case study for the effective application of the One Health approach. By systematically analyzing the integration of multidisciplinary strategies, this review reveals valuable lessons on optimizing public health responses to vector-borne diseases. The alignment of these strategies with One Health principles not only enhances the effectiveness of dengue control efforts in China but also offers a framework that can be adapted by other regions facing similar challenges. Ultimately, the insights gained from this analysis contribute to the global fight against dengue, emphasizing the need for collaborative and holistic approaches in public health initiatives.

Abstract Background Leonurus japonicus, a significant medicinal plant known for its therapeutic effects on gynecological and cardiovascular diseases, has genetic diversity that forms the basis for germplasm preservation and utilization in medicine. Despite its economic value, limited research has focused on its genetic diversity and divergence. Results The avg. nucleotide diversity of 59 accessions from China were 0.00029 and hotspot regions in petN-psbM and rpl32-trnL (UAG) spacers, which can be used for genotype discrimination. These accessions divided into four clades with significant divergence. The four subclades, which split at approximately 7.36 Ma, were likely influenced by the Hengduan Mountains uplift and global temperature drop. The initial divergence gave rise to Clade D, with a crown age estimated at 4.27 Ma, followed by Clade C, with a crown age estimated at 3.39 Ma. The four clades were not showed a clear spatial distribution. Suitable climatic conditions for the species were identified, including warmest quarter precipitation 433.20 mm ~ 1,524.07 mm, driest month precipitation > 12.06 mm, and coldest month min temp > -4.34 °C. The high suitability distribution showed contraction in LIG to LGM, followed by expansion from LGM to present. The Hengduan Mountains acted as a glacial refuge for the species during climate changes. Conclusions Our findings reflected a clear phylogenetic relationships and divergence within species L. japonicus and the identified hotspot regions could facilitate the genotype discrimination. The divergence time estimation and suitable area simulation revealed evolution dynamics of this species and may propose conservation suggestions and exploitation approaches in the future.

Natural polysaccharides have numerous biological activities, a stable structure, a large molecular weight, and a high level of safety, making them a popular choice for use as emulsifiers in the food business. However, polysaccharides' strong hydrophilicity and insolubility caused them to perform poorly during emulsification in high-temperature and high-salt environments, restricting their widespread use. To improve their ability to form emulsions, polysaccharides can have their molecular weight, structure, hydrophobicity, and other functional qualities altered. This paper reviews the effects of physical, chemical, and biological modification methods on the molecular structure and emulsification properties of polysaccharides, as well as the status and progress of research on the use of modified polysaccharides in emulsions, and also analyzes the issues with the current modification methods and forecasts the future development trend, with the goal of providing a theoretical foundation for further research on improving the emulsification property and broadening their application fields.

Cyanobacteria mutants with improved tolerance to combined high light and high temperature (HLHT) are rarely reported. Here, the authors use a hypermutation system for adaptive laboratory evolution and identify a mutant with improved HLHT tolerance by enhancing expression of shikimate kinase.

Rhizoma atractylodis, including Atractylodes lancea and Atractylodes chinensis, is widely utilized worldwide as a Chinese meteria medica (CMM) due to its rich content of various sesquiterpenoids and other medicinal components. Benefiting from the rapid and ongoing advancements in analytical technologies, significant strides have been achieved in advancing the analysis of Rhizoma atractylodis towards a more systematic, comprehensive, personalized, and intelligent approach. In this review, we highlight the progress made in the biogeography, biosynthesis, quality formation, and control of Rhizoma atractylodis over the past decade. Special attention is given to the incorporation of advanced genetics tools, modern omics technology, and chemometric methods that facilitate the accurate utilization of Rhizoma atractylodis. The elucidation of biosynthetic pathways, the formation of quality, and the control of active components continue to pose challenges in the study of Rhizoma atractylodis. Additionally, this review presents novel directions and opportunities aimed at enhancing ongoing research efforts.

Abstract Background Dicranostigma leptopodum (Maxim.) Fedde is a perennial herb with bright yellow flowers, well known as "Hongmao Cao" for its medicinal properties, and is an excellent early spring flower used in urban greening. However, its molecular genomic information remains largely unknown. Here, we sequenced and analyzed the chloroplast genome of D. leptopodum to discover its genome structure, organization, and phylogenomic position within the subfamily Papaveroideae. Results The chloroplast genome size of D. leptopodum was 162,942 bp, and D. leptopodum exhibited a characteristic circular quadripartite structure, with a large single-copy (LSC) region (87,565 bp), a small single-copy (SSC) region (18,759 bp) and a pair of inverted repeat (IR) regions (28,309 bp). The D. leptopodum chloroplast genome encoded 113 genes, including 79 protein-coding genes, 30 tRNA genes, and four rRNA genes. The dynamics of the genome structures, genes, IR contraction and expansion, long repeats, and single sequence repeats exhibited similarities, with slight differences observed among the eight Papaveroideae species. In addition, seven interspace regions and three coding genes displayed highly variable divergence, signifying their potential to serve as molecular markers for phylogenetic and species identification studies. Molecular evolution analyses indicated that most of the genes were undergoing purifying selection. Phylogenetic analyses revealed that D. leptopodum formed a clade with the tribe Chelidonieae. Conclusions Our study provides detailed information on the D. leptopodum chloroplast genome, expanding the available genomic resources that may be used for future evolution and genetic diversity studies.

IntroductionAtractylodes lancea is widely distributed in East Asia, ranging from Amur to south-central China. The rhizome of A. lancea is commonly used in traditional Chinese medicine, however, the quality of products varies across different regions with different geochemical characteristics.MethodThis study aimed to identify the chemotypes of A. lancea from different areas and screen for chemical markers by quantifying volatile organic compounds (VOCs) using a targeted metabolomics approach based on GC–MS/MS.ResultsThe A. lancea distributed in Hubei, Anhui, Shaanxi, and a region west of Henan province was classified as the Hubei Chemotype (HBA). HBA is characterized by high content of β-eudesmol and hinesol with lower levels of atractylodin and atractylon. In contrast, the Maoshan Chemotype (MA) from Jiangsu, Shandong, Shanxi, Hebei, Inner Mongolia, and other northern regions, exhibited high levels of atractylodin and atractylon. A total of 15 categories of VOCs metabolites were detected and identified, revealing significant differences in the profiles of terpenoid, heterocyclic compound, ester, and ketone among different areas. Multivariate statistics indicated that 6 compounds and 455 metabolites could serve as candidate markers for differentiating A. lancea obtained from the southern, northern, and Maoshan areas.DiscussionThis comprehensive analysis provides a chemical fingerprint of selected A. lancea. Our results highlight the potential of metabolite profiling combined with chemometrics for authenticating the geographical origin of A. lancea.

Considering the vulnerability of satellite positioning signals to obstruction and interference in orchard environments, this paper investigates a navigation and positioning method based on real-time kinematic global navigation satellite system (RTK-GNSS), inertial navigation system (INS), and light detection and ranging (LiDAR). This method aims to enhance the research and application of autonomous operational equipment in orchards. Firstly, we design and integrate robot vehicles; secondly, we unify the positioning information of GNSS/INS and laser odometer through coordinate system transformation; next, we propose a dynamic switching strategy, whereby the system switches to LiDAR positioning when the GNSS signal is unavailable; and finally, we combine the kinematic model of the robot vehicles with PID and propose a path-tracking control system. The results of the orchard navigation experiment indicate that the maximum lateral deviation of the robotic vehicle during the path-tracking process was 0.35 m, with an average lateral error of 0.1 m. The positioning experiment under satellite signal obstruction shows that compared to the GNSS/INS integrated with adaptive Kalman filtering, the navigation system proposed in this article reduced the average positioning error by 1.6 m.

In order to realize the automatic picking of shiitake mushrooms and reduce the risk of damage to shiitake mushrooms in the picking process, this paper designed a shiitake mushroom picking grasper. First, this paper carries out mechanical tests of compression and stress relaxation on sections of shiitake mushroom stipes, and establishes the component stress relaxation equations of shiitake mushroom stipes. The compression mechanical characteristics of the entire mushroom stipe are then analyzed using finite element analysis, with a mean square error of less than 5% compared to actual results. Second, based on the actual picking experience, this paper proposes an “L”-shaped three-finger picking grasper, and analyzes the mechanical relationship between the grasper’s gripping force, twisting separation torque, and servo output torque. Furthermore, according to the mechanical constitutive model of mushroom stipes, the optimal twisting separation torque and corresponding servo motor output torque for the grasper are determined. The picking grasper designed in this paper was tested for picking mushrooms of different growth periods, and the test results show that the picking grasper designed in this paper is able to grasp and separate the mushrooms quickly and without damage.

Coptis huanjiangensis, a new species of Ranunculaceae distributed in the valleys of Jiuwanshan National Natural Reserve in Huanjiang county (Guangxi, China), is described and illustrated for the first time based on morphological and plastome sequences data. It differs from C. chinensis, C. deltoidei and C. omeiensis mainly by having notably longer petiole, scape, bigger leaf blade with lobes obviously remote and robust rhizomes without stolons. Phylogenetic analyses support that C. huanjiangensis is sister to C. omeiensis and C. deltoidei.

It is challenging to restore Tibetan Plateau vegetation once it has been damaged.Highway construction, necessary for economic development, have significant negative impacts on the composition, structure, and functioning of the Qinghai-Tibet Plateau alpine grassland ecosystem. However, there is uncertainty regarding the extent of ecosystem damage in this region, such as whether quarries dug during highway construction may be restored and how recovery varies with increasing altitude. We conducted a transect survey of 39 quarry-restored grasslands reseeded with Elymus nutans and adjacent paired natural grasslands on the Gongyu and S308 highways on the Qinghai-Tibet Plateau (2800–5100 m above sea level). By comparing the community height, coverage, and species composition between restored and natural grasslands, we evaluated the degree of recovery and altitudinal gradient pattern of the grassland community and explained the limiting factors that led to the differences in recovery. After two-year restoration, quarry coverage and species composition were not restored to the degree of natural grassland. As altitude increased, the coverage recovery degree of quarry recovery community continued to decline, and the dissimilarity of community composition between the restored and natural grasslands was rising. We showed a breaking point at ca. 4100 m where recovery degree dropped dramatically, indicating a threshold for highway construction. The soil nutrients in quarry decreased significantly with increasing altitude. Structural equation modelling indicated that increasing altitude decreased the coverage of quarry by reducing temperature and soil nutrients. Due to the limitations of single species sowing, low temperature and nutrient availability, the effect of recovery decreased with increasing altitude. Therefore, we propose that during highway construction in the permafrost of the Qinghai-Tibet Plateau, quarries should avoid operation at high altitudes. When the altitude exceeds a certain height (e.g., 4100 m in this study), we do not recommend excavating quarries to destroy alpine grasslands.

The Chinese yam, an important orphan crop with both high nutrient and health promoting value, is mainly produced in the Yellow-Huai-Hai plain near the river basins in China. The protected designation of origin (PDO)-labeled Chinese yam differs greatly from others in market acceptance and price, which has led to fakes and the need for reliable authentication methods. Hence, stable isotope ratios of δ13C, δ15N, δ2D, and δ18O and 44 multielemental contents were used to explore the authentication of geographical origins and the effect of environmental factors. Twenty-two elements and δ15N were selected as the key variables to authenticate Chinese yams from three river basins as well as to authenticate them among traditional PDOs and others in the Yellow River basin. Moreover, six environmental factors, including the moisture index, maximum temperature, photosynthetically active radiation, soil organic carbon, total nitrogen and pH, were found to be highly related to these variances.
