Abstract Background Primary focal hyperhidrosis (PFH) may be attributed to the up-regulation of the cholinergic receptor nicotinic alpha 1 subunit (CHRNA1) in eccrine glands. Plasminogen activator inhibitor-1 (PAI1, encoded by SERPINE1) is reported to inhibit the expression of CHRNA1, while the role of PAI1 in hyperhidrosis is unknown. Methods Serpine1 KO mice, Serpine1-Tg mice, and wild type BALB/c mice were intraperitoneally injected with pilocarpine hydrochloride to induce PFH. Cisatracurium (CIS, antagonist of CHRNA1) or PAI-039 (small-molecule inhibitor of PAI1) was pre-administrated before the induction of hyperhidrosis. On the other hand, Chrna1-expressing AAV was constructed and administered to Serpine1-Tg mice with hydrochloride stimulation. Hydrochloride-related biomarkers, such as acetylcholine (ACH) in the serum, calcium voltage-gated channel subunit alpha1 C (CACNA1C), and aquaporin 5 (AQP5) in sweat glands of mice were assayed with ELISA, RT-PCR, and Western blot. Results The administration of PAI-039 or Pai1 knock-out increased Chrna1 expression, sweat secretion, and hydrochloride-related biomarkers (ACH, CACNA1C, and AQP5) expression. On the other hand, CIS administration diminished the strengthened hyperhidrosis phenotype induced by Pai1 knock-out with decreased sweat gland secretion. Conclusion PAI1 inhibits CHRNA1-mediated hydrochloride-induced hyperhidrosis, with decreased sweat gland secretion and diminished ACH, AQP5, and CACNA1C expression. These results indicate the potential to utilize PAI1 to alleviate PFH.

Machine vision systems have become key unmanned vehicle (UAV) sensing systems. However, under different weather conditions, the lighting direction and the selection of exposure parameters often lead to insufficient or missing object features in images, which could fail to perform various tasks. As a result, images need to be restored to secure information that is accessible when facing a light exposure difference environment. Many applications require real-time and high-quality images; therefore, efficiently restoring images is also important for subsequent tasks. This study adopts supervised learning to solve the problem of images under lighting discrepancies using a U-Net as our main architecture of the network and adding suitable modules to its encoder and decoder, such as inception-like blocks, dual attention units, selective kernel feature fusion, and denoising blocks. In addition to the ablation study, we also compared the quality of image light restoration with other network models using BAID and considered the overall trainable parameters of the model to construct a lightweight, high-exposure-difference image restoration model. Abstract Background Primary focal hyperhidrosis (PFH) is an autonomic neurological disease in which exocrine glands are oversecreted due to autonomic dysfunction of the sympathetic nervous system. Chrna1 promotes the pathogenesis of PFH. We aimed to check if downregulating of Chrna1 by cisatracurium could alleviate the symptoms of PFH. Methods The effect of cisatracurium in a hyperhidrosis mice model induced by pilocarpine hydrochloride was monitored for sweat gland secretion, and ultrastructural sweat secretory granules in sweat glands were analyzed. Meanwhile, markers of hyperhidrosis were checked, and release of Bdnf and Nrg1 from sympathetic ganglia axon was tested. Furthermore, the mechanism of cisatracurium function was evaluated in vitro using HEK293 expressing Chrna1. Finally, the effect of cisatracurium was determined in the hyperhidrosis mice model with overexpression or downregulation of Chrna1. Results In hyperhidrosis mice, pretreatment with cisatracurium effectively inhibited sweat secretion, along with fewer particle secretion in sweat glands. The molecular markers of hyperhidrosis (Aqp5 and Cacna1c) were inhibited by cisatracurium, acetylcholine (Ach) level in serum was found decreased. Neurotrophic factors (Bdnf and Nrg1) secreted by sympathetic axon activation were also inhibited. At last, it was confirmed that cisatracurium could not alter the gene or protein expression level of Chrna1, but could block the ion channel. Overexpression of Chrna1 abolished the effect of cisatracurium on hyperhidrosis, while cisatracurium could not function more in siChrna1‐treated mice. Conclusion Our results suggested that pretreatment of cisatracurium could alleviate hyperhidrosis in mice, probably through blocking the ion channel function of Chrna1.

Lung cancer is one of the most common malignant tumors. It is clinically divided into two types: small cell lung cancer and non-small cell lung cancer. Populus simonii distributed in East Asia region including China used as traditional medicine, which is an important medicinal plant for anti-lung cancer activity. The complete chloroplast genome sequence of P. simonii was characterized from Illumina pair-end sequencing. The chloroplast genome of P. simonii was 156,559 bp in length, containing a large single-copy region (LSC) of 84,825 bp, a small single-copy region (SSC) of 47,561 bp, and two inverted repeat (IR) regions of 16,494 bp. The overall GC content is 36.70%, while the corresponding values of the LSC, SSC, and IR regions are 34.5%, 30.0%, and 42.0%, respectively. The genome contains 131 complete genes, including 86 protein-coding genes (68 protein-coding gene species), 37 tRNA genes (29 tRNA species), and 8 rRNA genes (4 rRNA species). The neighbour-joining phylogenetic analysis showed that P. simonii and P. qamdoensis clustered together as sisters to other Populus species.

Abstract Background The present standard of surgical treatment for esophageal cancer is country dependent. The aim of the present study was to investigate the basic aspects of surgical procedures performed for esophageal cancer, and provide information about the present state of esophageal cancer surgery in China. Methods Data were obtained from a database administered by the Chinese Ministry for Health. A total of 542 participating hospitals were divided into seven geographic areas, and 10% of hospitals in each area were randomly chosen for inclusion. All patients with esophageal cancer, who underwent esophagectomy in these participating hospitals from January 1 to December 31, 2015, were included in the present study. The clinical characteristics, stage of tumor at diagnosis, operation summary and outcomes, and histological findings of patients were extracted and analyzed. Results The present study included 11,791 patients, and the average number of patients per hospital was 218. Squamous cell carcinoma was the most common pathological type, while the mid-esophagus was the most common location. Open procedures were performed in 63.8% of patients, while minimally invasive esophagectomy was performed in 36.2% of patients. Multiple approaches to transthoracic esophagectomy were utilized. Two-field lymphadenectomy was the most frequently performed (64.8%), followed by three-field lymphadenectomy (21.8%). Gastric tubes, thoracic duct ligation and postoperative enteral nutrition were implemented to minimize complications. Conclusion The standard operative procedure and detailed technique for esophageal carcinoma surgery is presently being debated in China. This survey provides some basic information about the present state of esophageal cancer surgery countrywide.

Background. The regulatory effect of integrin β6 (ITGB6) on sweat gland cells in primary palmar hyperhidrosis (PPH) remains unclear. Objectives. This study investigated the involvement of ITGB6 in the pathogenesis of PPH. Materials and methods. Sweat gland tissues were collected from PPH patients and healthy volunteers. The expression levels of ITGB6 in sweat gland tissues were detected with quantitative polymerase chain reaction (qPCR), western blot and immunohistochemical staining. Sweat gland cells were extracted from PPH patients, and identified with immunofluorescence staining of CEA and CK7. The expression of aquaporin 5 (AQP5) and Na-K-Cl cotransporter 1 (NKCC1) in primary sweat gland cells that overexpress ITGB6 were also detected. Through a series of bioinformatic methods, differentially expressed genes in sweat gland tissues were examined and validated via comparing PPH samples and controls. The key proteins and biological functions enriched in PPH were determined using Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses. Results. The ITGB6 was upregulated in sweat gland tissues of PPH patients compared to that of healthy volunteers. The CEA and CK7 were positively expressed in sweat gland cells extracted from PPH patients. The overexpression of ITGB6 upregulated AQP5 and NKCC1 protein expression in the sweat gland cells of PPH patients. A total of 562 differentially expressed mRNAs were identified using high-throughput sequencing (394 upregulated, 168 downregulated), which were mainly active in the chemokine and Wnt signaling pathways. After verification with qPCR and western blot, the overexpression of ITGB6 significantly upregulated CXCL3, CXCL5, CXCL10, and CXCL11, and downregulated Wnt2 mRNA and protein expression in sweat gland cells. Conclusions. The ITGB6 is upregulated in PPH patients. It may be involved in the pathogenesis of PPH by upregulating AQP5, NKCC1, CXCL3, CXCL5, CXCL10, and CXCL11, and downregulating Wnt2 expression in sweat glands. [ABSTRACT FROM AUTHOR]

During the production process of the components of precision machines, a stable quality is the key consideration of the vendors. During the production process of metal parts, a thermal expansion error of the spindle will occur due to long time operations, which causes machining errors during the production process. The chatter phenomenon that occurs during the production of components causes knife marks on the processing surface. Thus, chatter is also a major factor that affects the stability of quality. The occurrence of chatter phenomenon not only affects the quality of metal components, it also causes severe wear of the blade surface and shortens the life of the knife tool, and makes the cutting quality more difficult to control. Therefore, this paper uses the lathe machine to simulate the production process in the factory, to collect signals of the thermal expansion error of the spindle phenomenon and the coexistence of chatter, for timely compensation of the thermal expansion error and the development of the chatter suppression system, while spending extra efforts to implement real-time dynamic detection with the Lorenz chaotic system and applying spindle speed selection to control the chatter phenomenon, thus enhancing the processing quality and lengthening the longevity of the knife tool. As for the compensation of temperature, the system applies multi-regression analysis to detect the front and rear bearings of the spindle and room temperature and to compensate the error of the spindle in real time. The system takes real-time control of the chatter phenomenon and compensation of long-time thermal expansion error as its core architecture, while the secondary architecture combines the Internet of Things with Skymars to enable the machine to stabilize communication and to combine with heterogeneous machines such as robotic arm, milling machine, and grinding machine. It is a framework with horizontal development and establishes a database to record the critical value of the occurrence of chatter and implement two-time quality detection, achieving management of the production line, improvement of quality, and cloud information storage. In this study, the nonlinear dynamic analysis of the hybrid squeeze film damper mounted on a gear-rotor-bearing system is presented. The strong nonlinear oil film force, nonlinear rub-impact force between the rotor and stator, and the gear meshing force are considered in this study. The bifurcation diagrams are plotted using a dimensionless imbalance factor, damping coefficient, and speed ratio as control parameters in this study. The beginning of chaotic motion is identified by phase diagrams, power spectrum, Poincaré maps, bifurcation diagrams, maximum Lyapunov exponent, and the fractal dimension of the system. The numerical results provide useful insights into the design and development of a gear-rotor-bearing system for rotating machinery that operates at high speeds and highly nonlinear conditions. Hybrid squeeze film damper is used to suppress irregular dynamic behaviors and reduce greater amplitude, even if the rotational speed of the rotating machine is higher, which is also confirmed in this study. The rapid development of technology is causing the replacement of many traditional manufacture industries by automation. Robotic arms are now commonly used in many sectors. The requirements for robotic arms in different sectors are quite different; but, in general, robotic arms not only save cost and man power, they also improve safety. The aim of this study was an investigation of the integration of image identification with robotic arms. To do this, the Denavit–Hartenberg transformation matrix was used to analyze the mechanical kinematics of the joints of each robotic arm axis. This allowed the spatial relationships between the Cartesian 3 D coordinate system and the joint of each axis to be determined and communication between the robotic arm and image identification to be established. A robotic arm prototype platform with automatic image identification and calibration was constructed using a quick and robust method. Several of the variables that exist in real robotic arm applications have been solved in this study: primarily the accuracy error that occurs when repeated gripping of workpieces is done and a movement and placement track is set up. Deviations occur frequently and if image identification can be applied to offset repeated inaccuracy, the quality of finished products will be more consistent. The Universal Robots 5 six-axis robot and cameras, which use a filter, Sobel computation, and Hough transformation computer vision image processing technology, together comprise a system that can automatically identify the workpiece and carry out loading and unloading accurately.
