Abstract Background Compassion fatigue in nursing interns contributes to career indecision and worsens the nursing shortage. While work environment and psychological factors are well-studied, the ethical dimension remains unexplored. Understanding these mechanisms, particularly the role of moral courage, is essential for designing interventions to combat compassion fatigue and address the workforce crisis. This study investigates the influence of moral courage on compassion fatigue among Chinese nursing interns, focusing on the mediating roles of moral sensitivity and professional identity. Methods A quantitative, cross-sectional study was conducted in accordance with the STROBE guidelines. We used the convenience sampling method to recruit 467 nursing interns from four public junior colleges in Hunan Province, China in February, 2024. Data were collected using Compassion Fatigue Short Scale, Moral Courage Scale, Revised Moral Sensitivity Questionnaire, and Professional Identity Scale. Data analyses were conducted using SPSS 22.0 and Amos 21.0. Results The modified model exhibited a good fit (χ2/df = 3.437, AGFI = 0.928, IFI = 0.984, TLI = 0.976, CFI = 0.984, NFI = 0.977, RMSEA = 0.072). Moral sensitivity positively influenced both moral courage and professional identity, while professional identity negatively impacted compassion fatigue. Importantly, the effect of moral courage on compassion fatigue was entirely mediated by moral sensitivity and professional identity (β = -0.114, P = 0.001). Conclusion This study suggests that moral courage in nursing interns mitigates compassion fatigue through the combined mediating effects of moral sensitivity and professional identity. Ethics education programs fostering moral courage, moral sensitivity, and professional values in nursing students could be crucial in alleviating compassion fatigue.

Abstract Wheat grain starch content displays large variations within different pearling fractions, which affecting the processing quality of corresponding flour, while the underlying mechanism on starch gradient formation is unclear. Here, we show that wheat caryopses acquire sugar through the transfer of cells (TCs), inner endosperm (IE), outer endosperm (OE), and finally aleurone (AL) via micro positron emission tomography-computed tomography (PET-CT). To obtain integrated information on spatial transcript distributions, developing caryopses are laser microdissected into AL, OE, IE, and TC. Most genes encoding carbohydrate transporters are upregulated or specifically expressed, and sugar metabolites are more highly enriched in the TC group than in the AL group, in line with the PET-CT results. Genes encoding enzymes in sucrose metabolism, such as sucrose synthase, beta-fructofuranosidase, glucose-1-phosphate adenylyltransferase show significantly lower expression in AL than in OE and IE, indicating that substrate supply is crucial for the formation of starch gradients. Furthermore, the low expressions of gene encoding starch synthase contribute to low starch content in AL. Our results imply that transcriptional regulation represents an important means of impacting starch distribution in wheat grains and suggests breeding targets for enhancing specially pearled wheat with higher quality.

Abstract This paper proposes a preventive maintenance decision model for electric vehicle charging stations based on mutation operators and lifecycle optimization to address the impact of potential faults on maintenance effectiveness. By introducing the particle swarm optimization algorithm with mutation operator, a comprehensive analysis of opportunity service age factor and safety failure probability factor was conducted to establish an indicator system for the operation status of charging piles, and a potential fault identification model was constructed. By optimizing the life cycle, the balance problem between optimal maintenance life and optimal opportunity maintenance life has been solved, thus completing preventive maintenance decisions. The experimental results show that the accuracy of this method in preventive maintenance decision‐making for electric vehicle charging piles can reach 98%, with an average preventive maintenance decision‐making time of 1.6 s for load piles. At the same time, the risk probability value and load loss value are effectively controlled. This study has good application prospects in improving the preventive maintenance effect of electric vehicle charging piles.

The objective of this study was to optimize the extraction process of total sterols from wild Pyracantha fortuneana leaves (WPFL), and explore their antioxidant and whitening activities. The yield of total sterols (WPFLTPS) was used as indicator, and the single factor experiment was used to investigate the effects of extraction solvent, solid-liquid ratio, ultrasonic temperature and ultrasonic time on the yield of WPFLTPS. Based on this, the orthogonal experimental design was used to optimize the extraction conditions. The DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] and nitrite methods were used to evaluate the antioxidant activity of WPFLTPS in vitro. The whitening activity was evaluated by the inhibitory effect on tyrosinase. The results showed that for optimal extraction process of WPFLTPS, the extraction solvent was absolute ethanol, ultrasonic temperature was 50 ℃, solid-liquid ratio was 1:16 g/mL, and ultrasonic time was 25 minutes. Under these conditions, the yield of WPFLTPS was (11.00±0.03) mg/g. In the range of 0.5~5 mg/mL, WPFLTPS had strong scavenging effects on DPPH radicals, ABTS+ radicals and nitrite ions, as well as a strong inhibitory effect on tyrosinase, demonstrating a good dose-response relationship. This extraction process was stable and reliable for extracting WPFLTPS. The WPFLTPS can be used as a new, natural antioxidant and tyrosinase inhibitor in functional foods.

Non-Saccharomyces yeasts have great potential in improving wine quality, showing personality characteristics, and highlighting the terroir of wine. In this study, we evaluated the impact of simultaneous inoculation with the non-Saccharomyces yeasts Torulaspora delbrueckii or (and) Hanseniaspora uvarum in combination with Saccharomyces cerevisiae (EC1118 or VL3) on the aromatic compounds and sensory quality of Sauvignon blanc wines. The growth of yeast groups in the alcoholic fermentation process was tracked using fluorescence in situ hybridization. The presence of non-Saccharomyces yeast notably impacted the distribution of S. cerevisiae and was related to the species of yeast. The co-fermentation of H. uvarum and S. cerevisiae improved the content of total esters, especially acetate esters. Simultaneous inoculation of T. delbrueckii or (and) H. uvarum significantly increased the content of total terpenes, especially linalool. Similar results were found for some higher alcohols and organic acids. Sensory evaluation showed that the wines mixed fermentation with H. uvarum had significantly tropical fruit aroma characteristics. Citrus and mineral notes, typical aroma characteristics of Sauvignon blanc wine, were enhanced by mixed fermentation strategies with T. delbrueckii or (and) H. uvarum and different S. cerevisiae. Hence, co-fermentation by T. delbrueckii or H. uvarum combined with S. cerevisiae could significantly improve the sensory quality of Sauvignon blanc wine.

AbstractContext Therapeutic effects of Qiangjing tablets (QJT) on sperm vitality and asthenozoospermia (AZS) have been confirmed. However, the mechanism of action remains unclear.Objective This study investigates the effects of QJT on AZS and the underlying mechanism of action.Materials and methods Sixty Sprague–Dawley rats were randomly divided into six groups: Control, ORN (ornidazole; 200 mg/kg), ORN + QJT-low (0.17 g/mL), ORN + QJT-middle (0.33 g/mL), ORN + QJT-high (0.67 g/mL), and ORN + QJT + Radicicol (0.67 g/mL QJT and 20 mg/kg radicicol) groups. Pathological evaluation and analysis of mitophagy were conducted by H&E staining and transmission electron microscopy, respectively. Reactive oxygen species were detected by flow cytometry. Protein expression was determined by Western blotting.Results QJT significantly improved ORN-treated sperm motility and kinematic parameters, as well as the pathological symptoms of testicular and epididymal tissues. Stylocarotid artery syndrome (SAS) is a rare variant of Eagle’s syndrome that may lead to transient ischemic attack or stroke. The underlying pathophysiological mechanism involves compression of the internal carotid artery by an elongated styloid process (ESP), potentially resulting in vascular occlusion or dissection. An ESP exceeding 2.5 cm is deemed elongated, with a length of 3.0 cm considered clinically significant. Although the prevalence of ESP ranges from 4.0% to 7.3%, symptomatic cases are rare; symptoms are present in only approximately 4.0% of individuals with an ESP. Unlike the typical symptoms of Eagle’s syndrome, SAS may not cause pharyngeal discomfort, the sensation of a foreign body in the throat, dysphagia, or facial pain. This absence of characteristic symptoms as well as the development of central nervous system symptoms often leads patients to seek care from neurologists instead of otolaryngologists, increasing the likelihood of misdiagnosis or underdiagnosis. We herein report a unique case of ischemic stroke caused by SAS and present a literature review on cases of SAS-associated ischemic stroke published in the past decade. The reporting of this study conforms to the CARE guidelines.

Abstract Background Despite significant progress in sanitation status and public health awareness, intestinal infectious diseases (IID) have caused a serious disease burden in China. Little was known about the spatio-temporal pattern of IID at the county level in Zhejiang. Therefore, a spatio-temporal modelling study to identify high-risk regions of IID incidence and potential risk factors was conducted. Methods Reported cases of notifiable IID from 2008 to 2021 were obtained from the China Information System for Disease Control and Prevention. Moran’s I index and the local indicators of spatial association (LISA) were calculated using Geoda software to identify the spatial autocorrelation and high-risk areas of IID incidence. Bayesian hierarchical model was used to explore socioeconomic and climate factors affecting IID incidence inequities from spatial and temporal perspectives. Results From 2008 to 2021, a total of 101 cholera, 55,298 bacterial dysentery, 131 amoebic dysentery, 5297 typhoid, 2102 paratyphoid, 27,947 HEV, 1,695,925 hand, foot and mouth disease (HFMD), and 1,505,797 other infectious diarrhea (OID) cases were reported in Zhejiang Province. The hot spots for bacterial dysentery, OID, and HEV incidence were found mainly in Hangzhou, while high-high cluster regions for incidence of enteric fever and HFMD were mainly located in Ningbo. The Bayesian model showed that Areas with a high proportion of males had a lower risk of BD and enteric fever. People under the age of 18 may have a higher risk of IID. High urbanization rate was a protective factor against HFMD (RR = 0.91, 95% CI: 0.88, 0.94), but was a risk factor for HEV (RR = 1.06, 95% CI: 1.01–1.10). BD risk (RR = 1.14, 95% CI: 1.10–1.18) and enteric fever risk (RR = 1.18, 95% CI:1.10–1.27) seemed higher in areas with high GDP per capita. The greater the population density, the higher the risk of BD (RR = 1.29, 95% CI: 1.23–1.36), enteric fever (RR = 1.12, 95% CI: 1.00–1.25), and HEV (RR = 1.15, 95% CI: 1.09–1.21). Among climate variables, higher temperature was associated with a higher risk of BD (RR = 1.32, 95% CI: 1.23–1.41), enteric fever (RR = 1.41, 95% CI: 1.33–1.50), and HFMD (RR = 1.22, 95% CI: 1.08–1.38), and with lower risk of HEV (RR = 0.83, 95% CI: 0.78–0.89). Precipitation was positively correlated with enteric fever (RR = 1.04, 95% CI: 1.00–1.08), HFMD (RR = 1.03, 95% CI: 1.00–1.06), and HEV (RR = 1.05, 95% CI: 1.03–1.08). Higher HFMD risk was also associated with increasing relative humidity (RR = 1.20, 95% CI: 1.16–1.24) and lower wind velocity (RR = 0.88, 95% CI: 0.84–0.92). Conclusions There was significant spatial clustering of IID incidence in Zhejiang Province from 2008 to 2021. Spatio-temporal patterns of IID risk could be largely explained by socioeconomic and meteorological factors. Preventive measures and enhanced monitoring should be taken in some high-risk counties in Hangzhou city and Ningbo city.

In order to understand the diversity of endophytic bacteria community in different tissues of Pyracantha fortuneana, the 16S rRNA V5-V7 variable region of endophytic bacteria in P. fortuneana was sequenced by high-throughput sequencing technology, and the diversity of endophytic bacteria community in different tissue parts of P. fortuneana was analyzed. The results were as follows: (1) A total of 1 818 endophytic bacteria OTU were obtained, in which 754 endophytic bacteria were from root, 308 from stem and 756 from leaf, and with a total of 152 OTU. (2) Species classification showed that endophytic bacteria in different P. fortuneana tissues had abundant community diversities, and the endophytic bacteria in root belonged to 557 species, 373 genera, 216 families, 137 orders, 53 classes, 23 phyla, Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium and Streptomyces were the dominant genera, and their relative abundance was 10.57% and 8.00%, respectively. The endophytic bacteria in stem belonged to 270 species, 204 genera, 126 families, 76 orders, 32 classes, 21 phyla, Massilia and unclassified_f_Comamonadaceae were the dominant genera, and their relative abundance was 31.10% and 12.82%, respectively. The endophytic bacteria in leaf belonged to 581 species, 380 genera, 210 families, 130 orders, 52 classes, 21 phyla, Geobacillus and Pseudomonas were the dominant genera, and their relative abundance was 12.31% and 9.84%, respectively. (3) PICRUSt function prediction analysis showed that the species richness of root endophytic bacteria involved in te regulation of various metabolic pathways was the highest. These results provide a reference for endophytic bacteria functions and exploiting new beneficial microbial resources.

Abstract Background The detection of peritoneal metastasis (PM) is limited by current imaging tools. In this prospective study, we aimed to evaluate the sensitivity and specificity of peritoneal cell-free DNA (cfDNA) for diagnosis of PM. Methods Colorectal cancer (CRC) patients with/without PM were enrolled. The cfDNA experimental personnel and statists were blinded to the diagnosis of PM. Ultradeep sequencing covering large genomic regions (35000X, Next-generation sequencing) of cfDNA in peritoneal lavage fluid (FLD) and matched tumor tissues was performed. Results A total of 64 cases were recruited prospectively and 51 were enrolled into final analysis. In training cohort, 100% (17/17) PM patients obtained positive FLD cfDNA, comparing to 5/23 (21.7%) in patients without PM. Peritoneal cfDNA had a high sensitivity of 100% and specificity of 77.3% for diagnosis of PM (AUC: 0.95). In validation group of 11, 5/6 (83%) patients with PM obtained positive FLD cfDNA, comparing to 0/5 in non-PM (P = 0.031) with a sensitivity of 83.3% and specificity of 100%. Positive FLD cfDNA was associated with poor recurrence-free survival (P = 0.013) and was preceding radiographic evidence of recurrence. Conclusions Peritoneal cfDNA is a promising sensitive biomarker for earlier detection of PM in CRC than current radiological tools. It can potentially guide selection for targeted therapies and serve as a surrogate instead of laparoscopic explore in the future. Background Imbalances in bile acid (BA) synthesis and metabolism are involved in the onset of diabetes and depression in humans and rodents. However, the role of BAs and the farnesoid X receptor (FXR)/fibroblast growth factor (FGF) 15 signaling pathway in the development of diabetes and depression is still largely unknown. Therefore, we investigated the potential molecular mechanisms of BAs that may be associated with glucolipid metabolism disorders in diabetic mice subjected to chronic stress. Methods The type 2 diabetes mellitus (T2DM) mouse model was induced by feeding mice a high-fat diet and administering an intraperitoneal injection of streptozotocin (STZ). The chronic unpredictable mild stress (CUMS) procedure was performed by introducing a series of mild stressors. Forty mice were randomly divided into the regular chow feeding group and the high-fat diet feeding group. After two weeks of feeding, the mice were randomly divided into four groups: the Control group, CUMS group, T2DM group, and T2DM+CUMS group. The T2DM group and T2DM+CUMS group received an intraperitoneal injection of STZ to induce the T2DM model. The CUMS and T2DM+CUMS groups were exposed to CUMS to induce depressive-like phenotypes. Blood and tissue samples were obtained for pertinent analysis and detection. Results Compared with the T2DM mice, T2DM+CUMS mice had higher blood glucose and lipid levels, insulin resistance, inflammation of the liver and pancreas, impaired liver function, and increased total bile acids. These changes were accompanied by attenuated FXR signaling. Chronic stress was found to attenuate FXR expression and its downstream target, FGF15, in the ileum when compared with the T2DM group. Conclusion FXR may play a role in the diabetic disorder of glucolipid metabolism when aggravated by chronic stress. FXR and its downstream target, FGF15, may be therapeutic targets for treating comorbid T2DM and depression.

Abstract Background Approximately 10–15% of 46,XY disorders of sex development (DSDs) have an SRY mutation residing in the high mobility group (HMG) domain. Here, we present a case of 46,XY DSD caused by a novel missense mutation in the HMG region of SRY rapidly progressing to germ cell tumors (GCTs). Case presentation An adolescent female (15 years old) exhibiting primary amenorrhea was later diagnosed as a 46,XY female with bilateral gonadal dysplasia on the basis of peripheral lymphocyte karyotype 46,XY and a novel missense mutation in SRY (c.281 T > G, p.L94R). The novel missense mutation (c.281 T > G, p.L94R) and its adjacent region were conserved. Protein structure analysis showed that the mutant site was located in the middle of the HMG domain, and the mutant protein had a diminished ability to bind to DNA. Imaging examination revealed an adolescent female with a naive uterus. Laparoscopy and initial pathological examination revealed left gonadal dysplasia and right gonadal dysplasia with gonadoblastoma (GB). Right gonadectomy by laparoscopy was performed upon consent from the patient’s parents. Less than 1 year postoperatively, the left gonadal gland deteriorated as observed by the findings of a mass in the left adnexal region by pelvic MRI and serum AFP > 1000 ng/ml by serological tests, and then total hysterectomy and adnexal and left gonadectomy by laparoscopy were performed. The GCT stage was classified as stage Ic according to FIGO. At this time, pathologic examination showed that the left gonad had progressed to yolk sac tumor and dysgerminoma. The patient underwent chemotherapy post-operatively but developed type III myelosuppression and tumor recurrence several months later. Conclusions The patient initially presented with right gonadoblastoma but chose only right gonadectomy by laparoscopy to preserve the female sex characteristics, which resulted in rapid deterioration of the left gonad and poor treatment outcomes. This case demonstrates the importance of early genetic diagnosis and treatment of 46,XY female DSD.

Two new sesquiterpenoid derivatives, elgonenes M (1) and N (2), and a new shikimic acid metabolite, methyl 5-O-acetyl-5-epi-shikimate (3), were isolated from the mangrove sediment-derived fungus Roussoella sp. SCSIO 41427 together with fourteen known compounds (4–17). The planar structures were elucidated through nuclear magnetic resonance (NMR) and mass spectroscopic (MS) analyses. The relative configurations of 1–3 were ascertained by NOESY experiments, while their absolute configurations were determined by electronic circular dichroism (ECD) calculation. Elgonene M (1) exhibited inhibition of interleukin-1β (IL-1β) mRNA, a pro-inflammatory cytokine, at a concentration of 5 μM, with an inhibitory ratio of 31.14%. On the other hand, elgonene N (2) demonstrated inhibition at a concentration of 20 μM, with inhibitory ratios of 27.57%.

BackgroundPrevious studies have shown that abnormal increases in autoimmune antibodies in pregnant women may increase the risk of maternal thrombosis. However, at our hospital, two pregnant women presented with umbilical artery thrombosis and positive maternal autoantibodies were detected in both, which led us to consider whether maternal autoantibodies also played a role in umbilical artery thrombosis.Case presentationCase 1: Fetal ultrasound of a 34-year-old pregnant woman at 30+4 weeks gestation showed two umbilical arteries, with an inner diameter of approximately 0.15 cm for the smaller was artery. However, only a single umbilical artery blood flow signal was detected. Due to fetal distress, which was noted on abnormal cardiotocography and Doppler ultrasound, an emergency cesarean section was performed at 31+1 weeks gestation. The Apgar score of the newborn was 3-8-8. Umbilical cord examination detected thrombosis in the two umbilical arteries. Moreover, blood test results during pregnancy showed nRNP/Sm antibody (+) and SS antibody (+++). Case 2: The first systematic ultrasound of a 33-year-old twin pregnancy at 24+3 weeks gestation was normal, but routine fetal ultrasound at 27+1 weeks gestation showed only one umbilical artery between fetus A and the placenta. Blood test results showed that the patient was anti-nRNP/Sm antibody (+) in the rheumatoid immune activity test at 27+3 weeks gestation. An emergency cesarean section was performed at 34+6 weeks gestation because of the single umbilical artery and abnormal maternal coagulation. Both umbilical cords of fetus A and B blood test results showed anti-nRNP/Sm antibody (++). The pathological examination of the umbilical cord and placenta showed the presence of old thrombosis in one of the umbilical arteries of fetus A.ConclusionsAbnormal maternal autoantibodies may be a risk factor for umbilical artery thrombosis. For these pregnant women, conducting more detailed ultrasound monitoring might get early detection of UAT formation and avoid the occurrence of adverse pregnancy outcomes.

Aiming at the limitations of current fixed charging facilities and considering the charging needs of users in multiple scenarios, a mobile charging complex with real-time monitoring of rest, consumption and charging safety has been developed to meet the needs of users in multiple scenarios. Firstly, according to the development status of mobile charging, the technical route of mobile charging complex is proposed and the functions and requirements of the complex are clarified. Secondly, the mobile charging complex device that can be used in multiple scenarios such as high-speed service areas, residential areas and so on. A mobile charging integrated cabin, a charging cabin and a rest cabin are respectively designed. And the functional composition and commercial promotion value of each cabin are researched, which has been applied in Quzhou service area of G60 Shanghai–Kunming expressway in Zhejiang, China. Finally, the development of mobile charging complex is prospected from the perspectives of ergonomics, demand response, intelligent parking and bionic charging arm. Through the practical application, it is verified that the technical solution can effectively reduce the operation and maintenance cost of charging facilities under the premise of safety and reliability. It realizes peak shaving and valley filling, which is conducive to the healthy and stable development of charging facilities.

Abstract Background The screening biomarkers for early detection of colorectal cancer (CRC) is lacking. The aim is to identify epigenetic silenced genes and clarify its roles and underlying mechanism in CRC. We conducted integrative analyses of epigenome-wide Human Methylation 450 K arrays and transcriptome to screen out candidate epigenetic driver genes with transcription silencing. Methylated silencing HAND2 were identified and verified in large CRC cohort. The mechanism of HAND2 expression by promoter inhibition were clarified both in vitro and vivo assays. Cell biofunctional roles of HAND2 methylation was investigated in CRC cells. HAND2 reconstitution were constructed by lentivirus plasmid and tumor xenograft model of HAND2 were built subcutaneously. Genomic mRNA analysis by RNA-sequencing and subsequent GSEA analysis were performed to identify potential target of HAND2 and qPCR/WB was conducted to identify the results. Results We firstly reported high frequency of HAND2 methylation in promoter in CRC and hypermethylation was negatively correlated with expression silencing and leaded to poor survival in several CRC cohort patients. 5-Aza treatment to demethylated HAND2 could revert its expression in CRC cells. Functionally, HAND2 reconstitution can inhibit cell proliferation, invasion and migration in vitro. In tumor xenograft, HAND2 reconstruction significantly repressed tumor growth when compared to control vector. Thousands of aberrant expressed genes were observed in the heatmap of RNA-sequencing data. HAND2 reconstitution could bind to ERK and reduce its phosphorylation by CoIP assay. These above results showed HAND2 reconstitution perturbed the activation of MAPK/ERK signaling by reduction of ERK phosphorylation. Conclusions HAND2 is one tumor suppressor by targeting ERK signaling and one potential epigenetic driver gene in CRC. Video Abstract

Purpose: Mammary Paget’s disease (PD) in young women has seldom been reported. The aim of this study was to improve the knowledge of the clinicopathological characteristics in young patients with PD to provide a basis for the precise treatment of young patients. Methods: The medical records and pathological slides of 8 young patients (younger than 40 years old) with PD were reviewed. The data of 20 patients over 40 years old within the same period were used as controls. Results: The average age was 32.00 ± 3.96 years for the young patient group, with the youngest aged 27 years. The first symptom, physical examination, Paget cell morphology, and immunohistochemical marks were the same in different age groups. But young patients have varied tumor distribution patterns, fewer interstitial inflammatory cells, and advanced pathological local lymphatic metastasis than older patients in the same period. Conclusions: PD in young women has unique histopathological features. These manifestations seem to provide personalized treatment for PD treatment in young patients. More research is needed to clarify the significance of this research.

Low-temperature stress during stem elongation is a major factor limiting wheat yield. While calcium (Ca2+) is known to enhance stress tolerance, it’s potential as an alternative to cold priming and the underlying mechanisms in wheat remains unclear. The current study assessed the effects of exogenous Ca2+ and calcium inhibitors on wheat growth and related physiology mechanisms under low-temperature stress. The results revealed that exogenous Ca2+ increased photosynthesis and antioxidant capacity, lowered cell membrane damage, and ultimately enhanced tolerance to low-temperature stress during the stem elongation stage, compared with the non-exogenous Ca2+ treatment. Moreover, exogenous Ca2+ induced endogenous Ca2+ content and triggered the upregulation of Ca2+ signaling and cold-responsive related genes. This study highlights the significance of exogenous Ca2+ in enhancing stress tolerance and contributing to wheat yield improvement under low-temperature stress.

Grain filling is essential for wheat yield formation, but is very susceptible to environmental stresses, such as high temperatures, especially in the context of global climate change. Grain RGB images include rich color, shape, and texture information, which can explicitly reveal the dynamics of grain filling. However, it is still challenging to further quantitatively predict the days after anthesis (DAA) from grain RGB images to monitor grain development. Results: The WheatGrain dataset revealed dynamic changes in color, shape, and texture traits during grain development. To predict the DAA from RGB images of wheat grains, we tested the performance of traditional machine learning, deep learning, and few-shot learning on this dataset. The results showed that Random Forest (RF) had the best accuracy of the traditional machine learning algorithms, but it was far less accurate than all deep learning algorithms. The precision and recall of the deep learning classification model using Vision Transformer (ViT) were the highest, 99.03% and 99.00%, respectively. In addition, few-shot learning could realize fine-grained image recognition for wheat grains, and it had a higher accuracy and recall rate in the case of 5-shot, which were 96.86% and 96.67%, respectively. Materials and Methods: In this work, we proposed a complete wheat grain dataset, WheatGrain, which covers thousands of wheat grain images from 6 DAA to 39 DAA, which can characterize the complete dynamics of grain development. At the same time, we built different algorithms to predict the DAA, including traditional machine learning, deep learning, and few-shot learning, in this dataset, and evaluated the performance of all models. Conclusions: To obtain wheat grain filling dynamics promptly, this study proposed an RGB dataset for the whole growth period of grain development. In addition, detailed comparisons were conducted between traditional machine learning, deep learning, and few-shot learning, which provided the possibility of recognizing the DAA of the grain timely. These results revealed that the ViT could improve the performance of deep learning in predicting the DAA, while few-shot learning could reduce the need for a number of datasets. This work provides a new approach to monitoring wheat grain filling dynamics, and it is beneficial for disaster prevention and improvement of wheat production.

Stomata play a critical role in balancing photosynthesis and transpiration, which are essential processes for plant growth, especially in response to abiotic stress. Drought priming has been shown to improve drought tolerance. Lots of studies have been done with the response of stomatal behavior to drought stress. However, how the stomatal dynamic movement in intact wheat plants response to drought priming process is not known. Here, a portable microscope was used to take microphotographs in order to in-stiu determination of stomatal behavior. Non-invasive micro-test technology was used for measurements of guard cell K+, H+ and Ca2+ fluxes. Surprisingly, the results found that primed plants close stomatal much faster under drought stress, and reopening the stomatal much quicker under recovery, in relation to non-primed plants. Compared with non-primed plants, primed plants showed higher accumulation of ABA and Ca2+ influx rate in guard cells under drought stress. Furthermore, genes encoding anion channels were higher expressed and K+ outward channels activated, leading to enhanced K+ efflux, resulting in faster stomatal closure in primed plants than non-primed plants. During recovery, both guard cell ABA and Ca2+ influx of primed plants were found to be significantly reducing K+ efflux and accelerating stomatal reopening. Collectively, a portable non-invasive stomatal observation of wheat found that priming promoted faster stomatal closure under drought stress and faster reopening during post-drought recovery in relation to non-primed plants, thereby enhancing overall drought tolerance.

Introduction. The interaction between diabetes, obesity, and bone metabolism was drawing increasing public attention. However, the osteometabolic changes in diabetes mellitus type 2 (T2DM) patients with abdominal obesity have not been fully revealed. This study is aimed at investigating the association between abdominal obesity indices and bone turnover markers among T2DM participants. Methods. 4351 subjects were involved in the METAL study. Abdominal obesity indices included neck, waist, and hip circumference, visceral adiposity index (VAI), lipid accumulation product (LAP), waist-to-hip ratio (WHR), and Chinese visceral adiposity index (CVAI). They were applied to elucidate the nexus between β-C-terminal telopeptide (β-CTX), osteocalcin (OC), and intact N-terminal propeptide of type I collagen (P1NP). Results. Abdominal obesity indices were strongly negatively associated with β-CTX and OC. Among males, five indices were negatively correlated with β-CTX (BMI, WC, LAP, WHR, and CVAI) and OC (BMI, NC, WC, WHR, and CVAI). There were no significant associations with P1NP. Among females, all eight indices were negatively associated with β-CTX. Seven indices were negatively related to OC (BMI, NC, WC, HC, LAP, WHR, and CVAI). The VAI was negatively correlated with P1NP. Conclusions. The present study demonstrated that in T2DM, abdominal obesity had an obviously negative correlation with bone metabolism. Abdominal obesity indices were significantly negatively associated with skeletal destruction (β-CTX) and formation (OC). In routine clinical practice, these easily obtained indices could be used as a preliminary screening method and relevant factors for osteodysfunction incidence risk at no additional cost and may be of particular value for postmenopausal women in T2DM populations.

Chemical treatment is one of the encouraging approaches to refine the drought tolerance of wheat due to the possibility to be applied by both seed soaking and seedling foliar application at various growth stages of the plant. The effects of various chemical molecules treatment on wheat physiology and its productivity under drought were evaluated in this paper. As the climate changes continuously, it adversely treats crop production, particularly wheat cultivation. Chemical treatment is identified to stabilize the drought effect on wheat by improving drought-responsive defense hormones and enzyme activity by boosting stress protein expression and antioxidant enzymes for counteracting the formation of reactive oxygen species. The use of multiple chemicals with pre-and post-emergence treatment has refined drought tolerance to uphold the normal physiology of wheat under changing climates. Furthermore, exploring the potential priming chemicals and evaluating the molecules with varying growth stages, a combination of chemicals, and methods of treatment should be emphasized for an effective treatment approach.

Lindera chienii Cheng 1934 is an important medicine plant. The first complete plastid genome sequence of L. chienii was assembled and analyzed in this study. The plastid genome is 152,744 bp in length with a GC content of 39.15%, contains a large single-copy region of 93,767 bp and a small single-copy region of 18,843 bp, which were separated by a pair of inverted repeat regions of 20,067 bp. A total of 128 genes were detected in the plastid genome, including eight ribosomal RNA genes, 36 transfer RNA genes, and 81 protein-coding genes. The phylogenomic analysis based on plastid genomes supports the close relationships among Lindera chienii, L. megaphylla and Litsea acutivena.

Abstract Background Synchronous peritoneal metastasis of colorectal cancer usually predicts a bleak prognosis. Hyperthermic intraperitoneal chemotherapy (HIPEC) and cytoreductive surgery (CRS) have brought a glimmer of hope to the treatment of peritoneal cancer. Few cases treated with lobaplatin have been reported in the literature and the regimen is controversial. In this case, the comprehensive treatment scheme of lobaplatin-based HIPEC plus CRS and rechallenge using cetuximab plus systemic chemotherapy is effective, especially for the patients with left colon cancer (wild-type RAS). Case presentation A 49 year-old man with signet ring cell carcinoma of sigmoid colon with extensive abdominal metastasis (wild-type RAS) was hospitalized with prolonged abdominal pain, distention and abdominal mass. After receiving HIPEC with lobaplatin and XELOX regimen combined with cetuximab for eight cycles, the patient had been treated with the FOLFIRI regimen and cetuximab for 24 cycles, which discontinued due to myelosuppression. Because the disease recurred unfortunately 4 months later, the FOLFIRI + cetuximab regimen was initiated again and stopped after two cycles. Intestinal obstruction occurred 1 month later, so open total colectomy, CRS + HIPEC and ileorectal anastomosis were performed. Capecitabine adjuvant chemotherapy was administered, followed by the maintenance therapy with FOLFIRI + cetuximab regimen. After that, the patient has been in relatively stable condition. By August 2021, the overall survival is more than 45 months, which displays significant curative effect. Conclusion For peritoneal metastasis from left colon cancer, the management with CRS + lobaplatin HIPEC and rechallenge of systemic chemotherapy plus targeted medicine based on gene detection can dramatically improve prognosis and extend the overall survival.

Large-scale artificial neural networks have many redundant structures, making the network fall into the issue of local optimization and extended training time. Moreover, existing neural network topology optimization algorithms have the disadvantage of many calculations and complex network structure modeling. We propose a Dynamic Node-based neural network Structure optimization algorithm (DNS) to handle these issues. DNS consists of two steps: the generation step and the pruning step. In the generation step, the network generates hidden layers layer by layer until accuracy reaches the threshold. Then, the network uses a pruning algorithm based on Hebb’s rule or Pearson’s correlation for adaptation in the pruning step. In addition, we combine genetic algorithm to optimize DNS (GA-DNS). Experimental results show that compared with traditional neural network topology optimization algorithms, GA-DNS can generate neural networks with higher construction efficiency, lower structure complexity, and higher classification accuracy.

Antenna-in-Package has been widely applied for millimeter-wave systems that integrate the MMIC chip and antenna into one package. For organic substrate Antenna-in-Package solutions, probe feeding and aperture-coupled feeding patches are generally used as radiation units. In this work, a patch antenna is proposed using coupled resonation; a wideband, high-gain and compact 3-D antenna package integration is realized. Equivalent circuit model is analysed for the coupled structure. $\boldsymbol {\lambda }\mathrm {/\mathbf {4}}$ coupled line is adopted for impedance matching, the whole antenna is designed on the same thick radiation patch substrate, with less organic substrate lamination layers. For millimeter-wave Antenna-in-Package structures, vias are generally used for vertical interconnections, which function well in the lower millimeter-wave frequency range. For operation frequency higher than 100 GHz, design difficulties may be encountered for via interconnection, with performance degeneration on impedance matching and transmission loss. In our work a H-shaped slot is integrated instead of vias on an organic substrate, realizing a high frequency and wideband mm-wave transition. An interconnection structure is also adopted to convert a microstrip line to differential microstrip line in the feeding network. Antenna prototype with $2\times 4$ radiation array is designed for verification purpose at 20 GHz frequency range with simulated bandwidth of 23%. The prototype is fabricated with separate PCB boards assembled with screws; measured antenna gain is 12.7 dBi - 13.5 dBi. The proposed structure provides a wideband and low-loss solution for potential higher frequency mm-wave Antenna-in-Package applications.

The low-frequency disturbances transmitted by flexible cables are difficult to be attenuated for a novel disturbance-free payload spacecraft, which decreases the payload’s pointing accuracy and stability. In this research, a new spacecraft configuration with a high-precision inertial reference unit composed of capacitive sensors and a spherical test mass is proposed. The disturbance attenuation and pointing control system is subdivided into four interconnected control loops. The payload can be isolated from disturbances in the all-frequency band by the active vibration isolation control loop and the drag-free control loops, and its high-precision pointing requirement can be satisfied with the attitude pointing control loop and the attitude tracking control loop. An integrated control strategy is proposed, and the control system is decoupled into 12 single-input single-output control loops by pre-compensating, which lays the foundation for feedback design. Through the amplitude-frequency response analysis, the control bandwidth is designed according to the Proportional-Integral-Differentive control algorithm. The numerical simulations show that the disturbance attenuation performance is better than −20 dB in the all-frequency band, and the pointing accuracy and the pointing stability are better than 10−6 deg and 10−7 deg/s, respectively. The new spacecraft configuration and the disturbance attenuation and pointing control system provide a general technical solution for payloads with high-precision and high-stability requirements.

Background COVID-19 vaccination has been accepted widely. However, there are only a few reports on patients’ willingness to be vaccinated. This study investigated the willingness of Chinese outpatients to accept COVID-19 vaccination as well as influencing factors. Methods A cross-sectional survey was conducted in Jiangsu province, China in December 2020. Self-administered questionnaires, which were distributed to 625 outpatients among secondary hospitals, addressed demographic characteristics, sociological characteristics, and subjective reasons. There were 522 (83.5%) participants who gave completely valid responses. Logistic regression analysis was performed to explore the risk factors for willingness of COVID-19 vaccination. Results 71.5% of participants were willing to receive the COVID-19 vaccine. “worried about contracting COVID-19” (49.6%) and “vaccines have just been introduced and need time to consider” (38.9%) were the main reasons for being willing and unwilling to receive COVID-19 vaccine, respectively. The logistic regression analysis showed that “vaccines are an effective way to prevent diseases” (OR = 5.07, 95%CI: 3.32–7.75), “the price you are willing to pay for non-free vaccines (yuan) (101–500 vs ≤100)″ (OR = 1.87, 95%CI: 1.16–3.02), “per capital monthly income(yuan) (>6000 vs ≤3000)”(OR = 2.13, 95%CI: 1.03–4.41), and “self- assessed health status (Good vs Bad)″ (OR = 1.71, 95%CI: 1.01–2.90) were the main risk factors for outpatients to be willing to receive the COVID-19 vaccine. Conclusions The willingness of Chinese outpatients to receive COVID-19 vaccine was not high. The government should do more to increase publicity of knowledge about COVID-19 vaccine thus increasing willingness to vaccinate, and provide free vaccine to eliminate the cost impact.

ObjectiveHuman adenovirus (HAdV) coinfection with other respiratory viruses is common, but adenovirus infection combined with human coronavirus-229E (HCoV-229E) is very rare.Study design and settingClinical manifestations, laboratory examinations, and disease severity were compared between three groups: one coinfected with HAdV-Ad7 and HCoV-229E, one infected only with adenovirus (mono-adenovirus), and one infected only with HCoV-229E (mono-HCoV-229E).ResultsFrom July to August 2019, there were 24 hospitalized children: two were coinfected with HAdV-Ad7 and HCoV-229E, and 21 were infected with a single adenovirus infection. Finally, one 14-year-old boy presented with a high fever, but tested negative for HAdV-Ad7 and HCoV-229E. Additionally, three adult asymptotic cases with HCoV-229E were screened. No significant difference in age was found in the coinfection and mono-adenovirus groups (11 vs. 8 years, p = 0.332). Both groups had the same incubation period (2.5 vs. 3 days, p = 0.8302), fever duration (2.5 vs. 2.9 days, p = 0.5062), and length of hospital stay (7 vs. 6.76 days, p = 0.640). No obvious differences were found in viral loads between the coinfection and mono-adenovirus groups (25.4 vs. 23.7, p = 0.570), or in the coinfection and mono-HCoV-229E groups (32.9 vs. 30.06, p = 0.067). All cases recovered and were discharged from the hospital.ConclusionHAdV-Ad7 and HCoV-229E coinfection in healthy children may not increase the clinical severity or prolong the clinical course. The specific interaction mechanism between the viruses requires further study.

Background: Basement membranes (BMs) are associated with cell polarity, differentiation, migration, and survival. Previous studies have shown that BMs play a key role in the progression of cancer, and thus could serve as potential targets for inhibiting the development of cancer. However, the association between basement membrane-related genes (BMRGs) and clear cell renal cell carcinoma (ccRCC) remains unclear. To address that gap, we constructed a novel risk signature utilizing BMRGs to explore the relationship between ccRCC and BMs.Methods: We gathered transcriptome and clinical data from The Cancer Genome Atlas (TCGA) and randomly separated the data into training and test sets to look for new potential biomarkers and create a predictive signature of BMRGs for ccRCC. We applied univariate, least absolute shrinkage and selection operator (LASSO) and multivariate Cox regression analyses to establish the model. The risk signature was further verified and evaluated through principal component analysis (PCA), the Kaplan-Meier technique, and time-dependent receiver operating characteristics (ROC). A nomogram was constructed to predict the overall survival (OS). The possible biological pathways were investigated through functional enrichment analysis. In this study, we also determined tumor mutation burden (TMB) and performed immunological analysis and immunotherapeutic drug analysis between the high- and low-risk groups.Results: We identified 33 differentially expressed genes and constructed a risk model of eight BMRGs, including COL4A4, FREM1, CSPG4, COL4A5, ITGB6, ADAMTS14, MMP17, and THBS4. The PCA analysis showed that the signature could distinguish the high- and low-risk groups well. The K-M and ROC analysis demonstrated that the model could predict the prognosis well from the areas under the curves (AUCs), which was 0.731. Moreover, the nomogram showed good predictability. Univariate and multivariate Cox regression analysis validated that the model results supported the hypothesis that BMRGs were independent risk factors for ccRCC. Furthermore, immune cell infiltration, immunological checkpoints, TMB, and the half-inhibitory concentration varied considerably between high- and low-risk groups.Conclusion: Employing eight BMRGs to construct a risk model as a prognostic indicator of ccRCC could provide us with a potential progression trajectory as well as predictions of therapeutic response.

To discover bioactive natural products from mangrove sediment-derived microbes, a chemical investigation of the two Beibu Gulf-derived fungi strains, Talaromyces sp. SCSIO 41050 and Penicillium sp. SCSIO 41411, led to the isolation of 23 natural products. Five of them were identified as new ones, including two polyketide derivatives with unusual acid anhydride moieties named cordyanhydride A ethyl ester (1) and maleicanhydridane (4), and three hydroxyphenylacetic acid derivatives named stachylines H–J (10–12). Their structures were determined by detailed nuclear magnetic resonance (NMR) and mass spectroscopic (MS) analyses, while the absolute configurations were established by theoretical electronic circular dichroism (ECD) calculation. A variety of bioactive screens revealed three polyketide derivatives (1–3) with obvious antifungal activities, and 4 displayed moderate cytotoxicity against cell lines A549 and WPMY-1. Compounds 1 and 6 at 10 μM exhibited obvious inhibition against phosphodiesterase 4 (PDE4) with inhibitory ratios of 49.7% and 39.6%, respectively, while 5, 10, and 11 showed the potential of inhibiting acetylcholinesterase (AChE) by an enzyme activity test, as well as in silico docking analysis.

Wheat is a staple crop; its production must achieve both high yield and good quality due to worldwide demands for food security and better quality of life. It has been found that the grain qualities vary greatly within the different layers of wheat kernels. In this paper, the spatial distributions of protein and its components, starch, dietary fiber, and microelements are summarized in detail. The underlying mechanisms regarding the formation of protein and starch, as well as spatial distribution, are discussed from the views of substrate supply and the protein and starch synthesis capacity. The regulating effects of cultivation practices on gradients in composition are identified. Finally, breakthrough solutions for exploring the underlying mechanisms of the spatial gradients of functional components are presented. This paper will provide research perspectives for producing wheat that is both high in yield and of good quality.

Most cancer‐related deaths result from the progressive growth of metastases. Patients with peritoneal metastatic (PM) colorectal cancer have reduced overall survival. Currently, it is still unclear why colorectal cancer (CRC) cells home to and proliferate inside the peritoneal cavity, and there is no effective consolidation therapy for improved survival. Using a proteomic approach, we found that key enzymes of fatty acid oxidation (FAO) were decreased in patients with PM colorectal cancer. Furthermore, we confirmed that carnitine palmitoyltransferase IA (CPT1A), a rate‐limiting enzyme of FAO, was expressed at significantly low levels in patients with PM colorectal cancer, as determined by RT‐qPCR, IHC, and GEO dataset analysis. However, lipidomics revealed no difference in FFA levels between PM and non‐PM primary tumors. Here, we showed that cancer‐associated fibroblasts (CAFs) promote the proliferation, migration, and invasion of colon cancer cells via upregulating CPT1A to actively oxidize FAs and conduct minimal glycolysis. In addition, coculture‐induced glycolysis increased in cancer cells while fatty acid catabolism decreased with lower adiponectin levels. Importantly, inhibition of glycolysis significantly reduced the survival of CRC cells after incubation with conditioned medium from CAFsCPT1A‐OE in vitro and impaired the survival and growth of organoids derived from CRC‐PM. Finally, we found that directly blocking FAO in CAFsCPT1A‐OE with etomoxir inhibits migration and invasion in vitro and decreases tumor growth and intraperitoneal dissemination in vivo, revealing a role for CAF CPT1A in promoting tumor growth and invasion. In conclusion, our results suggest the possibility of testing FAO inhibition as a novel approach and clinical strategy against CAF‐induced colorectal cancer with peritoneal dissemination/metastases.

Abstract The terrestrial environment of a watershed is a source of potential carbon (C), nitrogen (N), and phosphorus (P) exports, and the hydrological regime provides the mechanism to turn the potential exports into reality when water is available. However, the extent to which the terrestrial environment alters the strength and nature of streamflow in transporting stream water nutrient ratios remains largely unknown. This study combined monthly stream discharge data with synchronously sampled stream water C:N:P ratios in 14 catchment streams in the Xitiao River Basin (XRB) in Zhejiang Province, China. The transport effect of streamflow on C:N:P ratios varied depending on the nutrient element, flow condition, and terrestrial environment. In the lower reaches of the XRB, there were negative relationships between C:N ratios, C:P ratios and watershed discharge, and positive relationships between N:P ratios and watershed discharge in both high and low flow conditions. In the middle and upper reaches of the XRB, the C:N-discharge relationship changed from negative to positive when the streamflow conditions altered from low to high flow. The C:P- and N:P-discharge relationships were negative regardless of high or low flows, but the regression coefficient significantly decreased with increasing streamflow. The C:N-discharge correlation over the course of the year shifted from negative to positive, as urban areas expanded within the catchment. The C:P-discharge relationship altered from negative to positive with more cropland and wetland but from positive to negative with a greater forest percentage and mean percentage slope. Our results indicate that changes in the terrestrial environment (e.g., the proportion of a particular land cover within a watershed) generally produced a threshold flow above which the coupling relationships between element fluxes from the terrestrial to riverine ecosystem changed sharply.

Abstract Purpose to describe the methodology of transurethral seminal vesiculoscopy and the anatomy of the area of the verumontanum, and to determine the safety of this procedure, especially in terms of postoperative complications. Methods This retrospective observational study enrolled 144 patients with intractable hematospermia from May 2011 and August 2019. A 4.5/6.5-Fr vesiculoscope was inserted into the seminal vesicle to deal with the positive findings. The solution of quinolones was used to rinse each seminal vesicle. Results In this study, Transurethral seminal vesiculoscopy was successfully performed in 139 patients (96.53%). Hematospermia was alleviated or disappeared in 116 (80.56%) patients by less than half a year after surgery. Common intraoperative manifestations were hemorrhage, stones, utricle polyps and cysts. The surgical approach in our study were categorized into four types, including 24 (16.7%), 73 (50.7%), 42 (29.2%), and 5 (3.5%) cases in Type A (natural opening of the ejaculatory duct), B (trans-duct fenestration), C (trans-utricle fenestration), and D (not founded), respectively. Sexual function change was recorded in 12 patients of 111 patients, all by the method of trans-utricle fenestration, including 8 (7.21%), 3 (2.70%), and 1 (0.90%) patients in shorter intravaginal ejaculatory latency time, worse erection hardness and loss of orgasm, respectively. Conclusion Transurethral seminal vesiculoscopy is an effective and safe procedure for the management of hematospermia. The anatomy of the distal seminal tract should be understood more deeply and Wu’method (uncover-curtain method) needs to be promoted to verify its universality and safety. Besides, the complications of the function dysfunction should be discussed in the future in multi-center clinical trials.

Litsea honghoensis Liou is an endangered tree endemic to south China. In this study, the first complete plastid genome of L. honghoensis was presented, which had a length of 152,605 base pairs (bp) with a GC content of 39.20%. The genome consisted of a large single-copy (LSC) region of 93,560 bp, a small single-copy (SSC) region of 18,905 bp, and two inverted repeat regions (IRa and IRb) of 20,070 bp. There were 125 genes in the plastid genome, including 81 protein-coding genes, 36 transfer RNA (tRNA) genes, and eight ribosomal RNA (rRNA) genes. Phylogenomic analysis based on 52 complete plastomes of Laureae in the family Lauraceae supports the close relationships among L. honghoensis, Lindera communis, Lindera nacusua, Lindera angustifolia, and Lindera glauca.

During the period of service, the infrastructure is subjected to various forms of impact. To investigate the impact responses of a reinforced concrete (RC) beam, a numerical model of the RC beam under impact was developed by the finite element package LS-DYNA in this study. The numerical model was verified by using the drop hammer test on the RC beam. Using the numerical model, the midspan displacement of the RC beam is analyzed under the interaction of impact mass and impact velocity. The results show that the response surface of midspan displacement can be fitted as a binary power function of impact mass and impact velocity. The midspan displacement under various impact conditions with equal impulse or equal impact energy is different. Within the scope of a low-speed impact, the midspan displacement decreases with an increase in the impact mass under the equal impulse, while it increases with an increase in the impact mass under the equal impact energy. In addition, the impact failure of the RC beam is judged by the deformation criterion. The threshold value of the ductility coefficient is recommended to be set as 15 in the impact-resistant design for RC beams in civil engineering structures within the scope of a low-speed impact.

A new trithiodiketopiperazine derivative, adametizine C (1), and five new alkane derivatives (7–11), were isolated from the mangrove sediment–derived fungus Penicillium ludwigii SCSIO 41408, together with five known dithiodiketopiperazine derivatives (2–6). Their structures were elucidated on the basis of spectroscopic analysis, and the absolute configuration of 1 was determined by X-ray crystallographic analysis. In a variety of bioactivity screening, 1–5 exhibited some selective antifungal or antibacterial activities. Compounds 1–3 showed cytotoxicity against prostate cancer cell line 22Rv1 with half maximal inhibitory concentration (IC50) values of 13.0–13.9 μM; moreover, 3 showed obvious activity against another prostate cancer PC-3 cells with an IC50 value of 5.1 μM. Further experiments revealed that 3 could significantly reduce PC-3 cells colony formation and induce apoptosis in a dose-dependent manner. Several compounds also exhibited obvious inhibitory activities of lipopolysaccharide–induced nuclear factor-κB with IC50 values range from 8.2 to 21.5 μM, and 1, 5, and 9 were further evaluated for their effects on receptor activator of NF-κB ligand (RANKL)-induced osteoclastogenesis. Adametizine C (1), with the strongest inhibitory activity against RANKL-induced osteoclast differentiation in bone marrow macrophage cells with 10 μM, was suggested to be the promising lead compound for the treatment of osteoclast-related diseases.

Starch plays an important role in food industry. In this study, three wheat cultivars with different protein contents were used to investigate the different ratios of starch addition on starch pasting properties, starch thermal performance, dough rheology, biscuit quality, and their relationships. Results showed that with the increase in starch content, gluten, protein and glutenin macropolymer (GMP), lactic acid solvent retention capacity (SRC), sucrose SRC, and onset temperature (To) decreased, while most pasting parameters and gelatinization enthalpy (ΔH) increased. Viscosity parameters were significantly negatively correlated with dough stability time, farinograph quality number (FQN), and sucrose SRC. Biscuit quality was improved by starch addition, indicated by lower thickness and hardness, higher diameter, spread ratio, and sensory score. Viscosity parameters were positively correlated to diameter, spread ratio, and sensory score of biscuit, while negatively correlated to hardness and thickness of biscuit. Image analysis showed that the crumbs of biscuit were improved as shown by bigger pores in the bottom side. The results provide useful information for the clarification of the role of starch in determining biscuit quality and the inter-relationships of flour, dough, and biscuit.

Drought stress is a limiting factor for wheat production and food security. Drought priming has been shown to increase drought tolerance in wheat. However, the underlying mechanisms are unknown. In the present study, the genes encoding the biosynthesis and metabolism of abscisic acid (ABA) and jasmonic acid (JA), as well as genes involved in the ABA and JA signaling pathways were up-regulated by drought priming. Endogenous concentrations of JA and ABA increased following drought priming. The interplay between JA and ABA in plant responses to drought priming was further investigated using inhibitors of ABA and JA biosynthesis. Application of fluridone (FLU) or nordihydroguaiaretic acid (NDGA) to primed plants resulted in lower chlorophyll-fluorescence parameters and activities of superoxide dismutase and glutathione reductase, and higher cell membrane damage, compared to primed plants (PD) under drought stress. NDGA + ABA, but not FLU + JA, restored priming-induced tolerance, as indicated by a finding of no significant difference from PD under drought stress. Under drought priming, NDGA induced the suppression of ABA accumulation, while FLU did not affect JA accumulation. These results were consistent with the expression of genes involved in the biosynthesis of ABA and JA. They suggest that ABA and JA are required for priming-induced drought tolerance in wheat, with JA acting upstream of ABA.

The rapid on-site nucleic acid detection method is urgently required in many fields. In this study, we report a portable and highly integrated device for DNA detection that combines ultrafast DNA adsorption and rapid DNA amplification. The device, known as silicon film mediated recombinase polymerase amplification (RPA) for nucleic acid detection (SMART), can detect target DNA in less than 25 min from plants, animals, and microbes. Utilizing SMART, transgenic maize was rapidly detected with high selectivity and sensitivity. The sensitivity threshold of the SMART for transgenic maize genomic DNA was 50 copies. The detection results of genuine samples containing plants, animals, and microbes by SMART were consistent with the conventional polymerase chain reaction (PCR) method, demonstrating the high robustness of SMART. Additionally, SMART does not require expensive equipment and is fast, affordable, and user-friendly, making it suited for the broad-scale on-site detection of nucleic acids.

Two new chlorinated metabolites, 8-chlorine-5-hydroxy-2,3-dimethyl-7-methoxychromone (1) and 3,4-dichloro-1H-pyrrole-2,5-dione (3), and eight known compounds (2 and 4–9) were isolated from the mangrove sediments-derived fungus Mollisia sp. SCSIO41409. Their structures were elucidated by physicochemical properties and extensive spectroscopic analysis. The absolute configuration of stemphone C (4) was established for the first time by the X-ray crystallographic analysis. Compounds 3 and 4 showed different intensity of antimicrobial activities against several pathogenic fungi and bacteria, and antiproliferative activities against two human prostate cancer cell lines (IC50 values 2.77 to 9.60 μM). Further, stemphone C (4) showed a reducing PC-3 cell colony formation, inducing apoptosis and blocking the cell cycle at S-phase in a dose-dependent manner; thus, it could be considered as a potential antiproliferative agent and a promising anti-prostate cancer lead compound.

Early arrival waveform inversion (EWI) is an essential approach to obtaining velocity structures in near-surface. Due to suffering from a cycle‐skipping issue, it is difficult to reach the global minima for conventional EWI with the misfit function of least-squares norm (L2‐norm). Following the optimal transportation theory, we developed an EWI solution with a new objective function based on quadratic‐Wasserstein‐metric (W2-norm) to maintain the geometric characteristics of the distribution and improve the stability and convexity of the inverse problem. First, we gave the continuous form of the adjoint source and the Frechet gradient of the Wasserstein metric for seismic early arrival, which leads to an easy and efficient way to implement in the adjoint-state method. Then, we conducted two synthetic experiments on the target model containing some velocity anomalies and hidden layers to test its effectiveness in mapping accurate and high-resolution near-surface velocity structure. The results show that the W2-normed EWI can mitigate cycle-skipping issues compared with the L2-normed EWI. In addition, it can deal with hidden layers and is robust in terms of noise. The application to a real dataset indicates that this new solution can recover more details in the shallow structure, especially in the aspect of dealing with hidden layers.

Wheat yield and grain protein content (GPC) are two main optimization targets for breeding and cultivation. Remote sensing provides nondestructive and early predictions of yield and GPC, respectively. However, whether it is possible to simultaneously predict yield and GPC in one model and the accuracy and influencing factors are still unclear. In this study, we made a systematic comparison of different deep learning models in terms of data fusion, time-series feature extraction, and multitask learning. The results showed that time-series data fusion significantly improved yield and GPC prediction accuracy with R2 values of 0.817 and 0.809. Multitask learning achieved simultaneous prediction of yield and GPC with comparable accuracy to the single-task model. We further proposed a two-to-two model that combines data fusion (two kinds of data sources for input) and multitask learning (two outputs) and compared different feature extraction layers, including RNN (recurrent neural network), LSTM (long short-term memory), CNN (convolutional neural network), and attention module. The two-to-two model with the attention module achieved the best prediction accuracy for yield (R2=0.833) and GPC (R2=0.846). The temporal distribution of feature importance was visualized based on the attention feature values. Although the temporal patterns of structural traits and spectral traits were inconsistent, the overall importance of both structural traits and spectral traits at the postanthesis stage was more important than that at the preanthesis stage. This study provides new insights into the simultaneous prediction of yield and GPC using deep learning from time-series proximal sensing, which may contribute to the accurate and efficient predictions of agricultural production.

Background Coronavirus disease (COVID-19) pandemic posed a critical threat to public health in the past year and has not been fully controlled so far. The nature of front-line young hospital pharmacists’ occupation puts them at an increased risk of contracting any contagious disease, including COVID-19. Recent survey indicated that hospital pharmacists in China are depressive, hostile amid the pandemic. Aim The present investigation aims to understand the job stress among young hospital pharmacists during the outbreak of COVID-19 and to provide basic information for pharmacy managers to help young fellows to cope with job stress. Method This study is adopting pharmacist job stress questionnaire as the key instrument of data collection through WJX App in mobile phone. Demographic information, career prospects and stress management proposals were obtained synchronously. Quantitative data were processed with SPSS. Significant differences were examined using analysis of variance and Chi-square analysis. Result About 60% of 289 questionnaire respondents complained of job stress (178 respondents). According to the narrative description of the data, young pharmacists’ gender, education background, hospital grade, and specific work post had no significant effect on job stress difference. However, young pharmacists in different age-groups and professional titles showed different job stress. Pharmacists at the age of 31–35 complained more stress than the others. Pharmacists with high professional title (deputy chief pharmacist) complained more stress than the others. About 65% of 289 respondents had long-term plan for their practice, although 61% of young pharmacists felt troubled or worried with their future. As for stress management proposal, almost all young pharmacists hoped to improve their professional identity via raising their wages. Conclusion More than half of young pharmacists suffer from job stress amidst the COVID-19 pandemic in China, and various intervention measures should be taken to relieve the stress and finally improve their social identity.

Abstract Background Microorganisms in rivers and lakes are essential for nutrient recycling in aquatic ecosystems. Understanding the ecological processes shaping microbial communities is of crucial importance for aquatic microbial ecology and biogeography. However, the diversity of microorganisms and the forces that control this diversity are poorly understood. This is particularly true within the framework of the river-lake continuum in arid regions. Results Using a whole catchment-sampling effort, we explored biogeographical patterns and mechanisms of microbial community (bacteria and archaea) assembly within the catchment of the largest inland once freshwater lake (Lake Bosten) in China. Water samples from headstream tributaries, the mainstream of the River Kaidu to downstream Lake Bosten were characterized using amplicon sequencing of 16S rRNA genes. Higher α-diversity was found in mainstream of River Kaidu and in the tributaries compared with Lake Bosten. And the microbial community composition was also significantly different between the lake and its connected river habitats. Canonical correspondence analysis demonstrated that salinity and total suspended solids were the most important environmental factors shaping the community variations. Overall, pure environmental and pure spatial factors explained 13.7 and 5.6% of the community variation, respectively, while 32.0% of the variation was explained by combined environmental and spatial variables. These observations suggested that spatially structured environmental variations mainly shaped the microbial biogeography in this region. Both deterministic and stochastic processes influenced the microbial community assembly in river and lake habitats, and the stochastic pattern was particularly pronounced for microbiome in river habitat. Co-occurrence network analysis revealed more abundant and complicated correlations among frequently occurred taxa in lake habitat compared with the river habitat, implying that ecological multispecies interactions (e.g., competition) shaped lake microbial community structures. Conclusions Our findings demonstrate an ecological succession along the river-lake continuum of microbial communities across the largest inland once freshwater lake basin in China, and highlight the effects of spatially structured environmental factors on regional microbial β-diversity and species interactions on local community assembly.

Abstract Background The prevalence of both placenta previa and cesarean are on the rise. Multiple adverse outcomes are critically increased when placenta previa is subsequent to prior cesarean. The purpose of the present study is to develop a pre-surgical method for predicting adverse outcomes in pregnancy complicated with both placenta previa and prior cesarean. Methods Clinical data was obtained from the medical history system at the First Affiliated Hospital of Sun Yat-sen University from February 2003 to December 2016. All cases with a final diagnosis of “placenta previa/low lying placenta (ICD:O44.001-105)” and “scarred uterus complicated with pregnancy (ICD: O34.200-202)” were collected and reviewed. Hysterectomy was taken as the primary outcome; and blood loss was taken as the secondary outcome. Results Of 219 pregnant women in the final analysis, 25 received a hysterectomy following delivery, and 48 had blood loss exceeding 1000 ml. Pre-surgical risk factors for hysterectomy are ultrasonic signs of vascular lacunae, central placenta previa, and loss of normal hypoechoic retroplacental zone. A pre-surgical predictive equation referred to as “Hysterectomy Index in Placenta Previa with Prior cesarean (HIPs)” was generated and each risk factor was weighted to create an 8-point scale. This index yielded an area under the curve of 0.972 for the prediction of hysterectomy. Conclusions Application of the HIPs score may provide an effective pre-surgical prediction of cesarean hysterectomy in pregnant women complicated with both placenta previa and prior cesarean.

Sonneratia apetala fruits are edible and medicinal. Our previous report demonstrated that the S. apetala fruits extract had the hepatoprotective activity. In the study, an acidic polysaccharide named SAP-2 was isolated and purified from S. apetala fruits. The preliminary structure of SAP-2 was characterized, and its hepatoprotective effect and the mechanism of action in Nrf2 signaling pathway were also evaluated in acetaminophen (acetyl-para-aminophenol, APAP)-induced liver damaged mice. Results manifested that SAP-2 could alleviate histological changes, inhibit oxidative stress and enhance antioxidant system via activating of Nrf2 signaling pathway on APAP-induced hepatic injury. These results indicated that S. apetala fruits could be developed into a new source of potential natural antioxidant and functional food for hepatoprotection.
