Abstract Parasitoid-host interactions form the foundation of biological control strategies against many agriculture and forest insect pests. The emerald ash borer (EAB), Agrilus planipennis (Coleoptera: Buprestidae), is a serious invasive pest of ash (Fraxinus spp.) trees in North America. Tetrastichus planipennisi (Hymenoptera: Eulophidae) is a gregarious, koinobiont endoparasitoid, attacking late (3rd to 4th) instars of EAB larvae, which feed in the live phloem of ash trunks or branches, making serpentine-like galleries filled with larval frass. In the present study, we tested the hypothesis that T. planipennisi regulates the host metabolism and feeding activity to optimize its offspring development and fitness. We first compared the respiration rate of parasitized and unparasitized host larvae at different times after parasitism, and then measured feeding activity of both parasitized and unparasitized host larvae inside their feeding galleries. Although parasitized host larvae increased metabolic rate and feeding activity in the first few days of parasitism, T. planipennisi parasitism induced an overall reduction of the metabolic rate and decrease in feeding activity of parasitized host larvae over their development period. In addition, there was a negative relationship between feeding activity of parasitized hosts and brood sizes of the parasitoid progeny—i.e., the more parasitoid progeny a host larva received, the less feeding activity the host had. These findings suggest that T. planipennisi has limited ability to optimize its offspring development and fitness through regulations of the host metabolism and feeding activity and its parasitism reduces feeding damage of parasitized EAB larvae to infested ash trees.

Abstract Titanium has been widely used in prosthetic valves, but they are associated with serious defects in titanium‐based prosthetic valves, such as thrombosis, calcification, and decay. Therefore, it is very important to biofunctionalize titanium‐based valves to reduce inflammation and accelerate endothelialization of stents and antithrombosis. The titanium dioxide nanotubes were prepared from pure titanium (Ti) by anodic oxidation method in this study. The effects of titanium dioxide nanotubes on the metabolism of macrophages and the inflammatory reaction as implants were studied in vitro. The polarization state of macrophages and the ability to accelerate endothelialization were analyzed. The results demonstrated that titanium nanotubes promote M2 polarization of macrophages by inhibiting glycolysis and activating the Adenosine monophosphate‐activated protein kinase (AMPK) signaling pathway. In general, biofunctionalization titanium with nanotube could inhibit macrophage glycolysis, reduce inflammatory factor release and promote M2 polarization by activating the AMPK signaling pathway. And endothelialization was accelerated in vitro. Our result demonstrated that titanium nanotube could act as a potential approach to biofunctionlize titanium‐based prosthetic valves for endothelialization.

Traditional work on bystander intervention in violent emergencies has found that the larger the group, the less the chance that any individual will intervene. Here, we tested the impact on helping behavior of the affiliation of the bystanders with respect to the participants. We recruited 40 male supporters of the U.K. Arsenal football club for a two-factor between groups study with 10 participants per group. Each participant spoke with a virtual human Arsenal supporter (V), the scenario displayed in a virtual reality system. During this conversation, another virtual character (P), not an Arsenal fan, verbally abused V for being an Arsenal fan leading eventually to physical pushing. There was a group of three virtual bystanders who were all either Arsenal supporters indicated by their shirts, or football fans wearing unbranded shirts. These bystanders either encouraged the participant to intervene or dissuaded him. We recorded the number of times that participants intervened to help V during the aggression. We found that participants were more likely to intervene when the bystanders were out-group with respect to the participant. By comparing levels of intervention with a “baseline” study (identical except for the presence of bystanders), we conclude that the presence of in-group bystanders decreases helping. We argue therefore that, other things being equal, diffusion of responsibility is more likely to be overcome when participant and victim share group membership, but bystanders do not. Our findings help to develop understanding of how diffusion of responsibility works by combining elements of both the bystander effect and the social identity approach to bystander behavior.

Abstract Background Arthropods comprise the largest and most diverse phylum on Earth and play vital roles in nearly every ecosystem. Their diversity stems in part from variations on a conserved body plan, resulting from and recorded in adaptive changes in the genome. Dissection of the genomic record of sequence change enables broad questions regarding genome evolution to be addressed, even across hyper-diverse taxa within arthropods. Results Using 76 whole genome sequences representing 21 orders spanning more than 500 million years of arthropod evolution, we document changes in gene and protein domain content and provide temporal and phylogenetic context for interpreting these innovations. We identify many novel gene families that arose early in the evolution of arthropods and during the diversification of insects into modern orders. We reveal unexpected variation in patterns of DNA methylation across arthropods and examples of gene family and protein domain evolution coincident with the appearance of notable phenotypic and physiological adaptations such as flight, metamorphosis, sociality, and chemoperception. Conclusions These analyses demonstrate how large-scale comparative genomics can provide broad new insights into the genotype to phenotype map and generate testable hypotheses about the evolution of animal diversity.

We analyzed Trim2A/A mice, generated by CRISPR-Cas9, which have a recessive, null mutation of Trim2. Trim2A/A mice develop ataxia that is associated with a severe loss of cerebellar Purkinje cells and a peripheral neuropathy. Myelinated axons in the CNS, including those in the deep cerebellar nuclei, have focal enlargements that contain mitochondria and neurofilaments. In the PNS, there is a loss of myelinated axons, particularly in the most distal nerves. The pathologically affected neuronal populations – primary sensory and motor neurons as well as cerebellar Purkinje cells – express TRIM2, suggesting that loss of TRIM2 in these neurons results in cell autonomous effects on their axons. In contrast, these pathological findings were not found in a second strain of Trim2 mutant mice (Trim2C/C), which has a partial deletion in the RING domain that is needed for ubiquitin ligase activity. Both the Trim2A and the Trim2C alleles encode mutant TRIM2 proteins with reduced ubiquitination activity. In sum, Trim2A/A mice are a genetically authentic animal model of a recessive axonal neuropathy of humans, apparently for a function that does not depend on the ubiquitin ligase activity.

Abstract Asian Longhorned Beetle (ALB) Anoplophora glabripennis is a serious invasive forest pest in several countries including the United States, Canada, and Europe. RNA interference (RNAi) technology is being developed as a novel method for pest management. Here, we identified the ALB core RNAi genes including those coding for Dicer, Argonaute, and double-stranded RNA-binding proteins (dsRBP) as well as for proteins involved in dsRNA transport and the systemic RNAi. We also compared expression of six potential reference genes that could be used to normalize gene expression and selected gapdh and rpl32 as the most reliable genes among different tissues and stages of ALB. Injection of double-stranded RNA (dsRNA) targeting gene coding for inhibitor of apoptosis (IAP) into larvae and adults resulted in a significant knockdown of this gene and caused the death of 90% of the larvae and 100% of adults. No mortality of both larvae and adults injected with dsRNA targeting gene coding for green fluorescence protein (GFP, as a negative control) was observed. These data suggest that functional RNAi machinery exists in ALB and a potential RNAi-based method could be developed for controlling this insect.

Abstract The ingestion of double-strand RNAs (dsRNA) targeting essential genes in an insect could cause mortality. Based on this principle, a new generation of insect control methods using RNA interference (RNAi) are being developed. In this work, we developed a bioassay for oral delivery of dsRNA to an invasive forest and urban tree pest, the emerald ash borer (EAB, Agrilus planipennis). EAB feeds and develops beneath the bark, killing trees rapidly. This behavior, coupled with the lack of a reliable artificial diet for rearing larvae and adults, make them difficult to study. We found that dsRNA is transported and processed to siRNAs by EAB larvae within 72 h after ingestion. Also, feeding neonate larvae with IAP (inhibitor of apoptosis) or COP (COPI coatomer, β subunit) dsRNA silenced their target genes and caused mortality. Both an increase in the concentration of dsRNA fed and sequential feeding of two different dsRNAs increased mortality. Here we provide evidence for successful RNAi in EAB, and demonstrate the development of a rapid and effective bioassay for oral delivery of dsRNA to screen additional genes.

Abstract We present a novel approach for automatically detecting and tracking facial landmarks across poses and expressions from in-the-wild monocular video data, e.g., YouTube videos and smartphone recordings. Our method does not require any calibration or manual adjustment for new individual input videos or actors. Firstly, we propose a method of robust 2D facial landmark detection across poses, by combining shape-face canonical-correlation analysis with a global supervised descent method. Since 2D regression-based methods are sensitive to unstable initialization, and the temporal and spatial coherence of videos is ignored, we utilize a coarse-todense 3D facial expression reconstruction method to refine the 2D landmarks. On one side, we employ an in-the-wild method to extract the coarse reconstruction result and its corresponding texture using the detected sparse facial landmarks, followed by robust pose, expression, and identity estimation. On the other side, to obtain dense reconstruction results, we give a face tracking flow method that corrects coarse reconstruction results and tracks weakly textured areas; this is used to iteratively update the coarse face model. Finally, a dense reconstruction result is estimated after it converges. Extensive experiments on a variety of video sequences recorded by ourselves or downloaded from YouTube show the results of facial landmark detection and tracking under various lighting conditions, for various head poses and facial expressions. The overall performance and a comparison with state-of-art methods demonstrate the robustness and effectiveness of our method.

Aprostocetus fukutai is a specialist egg parasitoid of the citrus longhorned beetle Anoplophora chinensis, a high-risk invasive pest of hardwood trees. The parasitoid overwinters as diapausing mature larvae within the host egg and emerges in early summer in synchrony with the egg-laying peak of A. chinensis. This study investigated the parasitoid’s diapause survival in parasitized host eggs that either remained in potted trees under semi-natural conditions in southern France or were removed from the wood and held at four different humidities (44, 75, 85–93 and 100% RH) at 11 °C or four different temperature regimes (2, 5, 10 and 12.5 °C) at 100% RH in the laboratory. The temperature regimes reflect overwintering temperatures across the parasitoid’s geographical distribution in its native range. Results show that the parasitoid resumed its development to the adult stage at normal rearing conditions (22 °C, 100% RH, 14L:10D) after 6- or 7-months cold chilling at both the semi-natural and laboratory conditions. It had a low survival rate (36.7%) on potted plants due to desiccation or tree wound defense response. No parasitoids survived at 44% RH, but survival rate increased with humidity, reaching the highest (93.7%) at 100% RH. Survival rate also increased from 21.0% at 2 °C to 82.8% at 12.5 °C. Post-diapause developmental time decreased with increased humidity or temperature. There was no difference in the lifetime fecundity of emerged females from 2 and 12.5 °C. These results suggest that 100% RH and 12.5 °C are the most suitable diapause conditions for laboratory rearing of this parasitoid.

Patient-derived xenograft (PDX) tumor models have emerged as a new approach to evaluate the effects of cancer drugs on patients’ personalized tumor grafts enabling to select the best treatment for the cancer patient and providing a new tool for oncology drug developers. Here, we report that human tumors engrafted in immunodeficient mice are susceptible to formation of B-and T-cell PDX tumors. We xenografted human primary and metastatic tumor samples into immunodeficient mice and found that a fraction of PDX tumors generated from patients’ samples of breast, colon, pancreatic, bladder and renal cancer were histologically similar to lymphocytic neoplasms. Moreover, we found that the first passage of breast and pancreatic cancer PDX tumors after initial transplantation of the tumor pieces from the same human tumor graft could grow as a lymphocytic tumor in one mouse and as an adenocarcinoma in another mouse. Whereas subcutaneous PDX tumors resembling human adenocarcinoma histology were slow growing and non-metastatic, we found that subcutaneous PDX lymphocytic tumors were fast growing and formed large metastatic lesions in mouse lymph nodes, liver, lungs, and spleen. PDX lymphocytic tumors were comprised of B-cells which were Epstein-Barr virus positive and expressed CD45 and CD20. Because B-cells are typically present in malignant solid tumors, formation of B-cell tumor may evolve in a wide range of PDX tumor models. Although PDX tumor models show great promise in the development of personalized therapy for cancer patients, our results suggest that confidence in any given PDX tumor model requires careful screening of lymphocytic markers.

In order to reduce the data size and simplify the process of creating characters’ 3D models, a new and interactive ordinary differential equation (ODE)-based C2 continuous surface creation algorithm is introduced in this paper. With this approach, the creation of a three-dimensional surface is transformed into generating two boundary curves plus four control curves and solving a vector-valued sixth order ordinary differential equation subjected to boundary constraints consisting of boundary curves, and first and second partial derivatives at the boundary curves. Unlike the existing patch modeling approaches which require tedious and time-consuming manual operations to stitch two separate patches together to achieve continuity between two stitched patches, the proposed technique maintains the C2 continuity between adjacent surface patches naturally, which avoids manual stitching operations. Besides, compared with polygon surface modeling, our ODE C2 surface creation method can significantly reduce and compress the data size, deform the surface easily by simply changing the first and second partial derivatives, and shape control parameters instead of manipulating loads of polygon points.

Since non-automatic rigging requires heavy human involvements, and various automatic rigging algorithms are less efficient in terms of computational efficiency, especially for current curve-based skin deformation methods, identifying the iso-parametric curves and creating the animation skeleton requires tedious and time-consuming manual work. Although several automatic rigging methods have been developed, but they do not aim at curve-based models. To tackle this issue, this paper proposes a new rigging algorithm for automatic generation of dynamic skin deformation to quickly identify iso-parametric curves and create an animation skeleton in a few milliseconds, which can be seamlessly used in curve-based skin deformation methods to make the rigging process fast enough for highly efficient computer animation applications.

Literature studies in North America (US and Canada), Europe, and Asia (particularly Russia, China, Japan, and the Korean peninsula) were reviewed to identify parasitoid guilds associated with Agrilus woodborers. There are at least 12 species of hymenopteran parasitoids attacking eggs of Agrilus beetles and 56 species (36 genera), attacking Agrilus larvae infesting various host plants in North America, Asia, and Europe. While most of the egg parasitoids (9 species) belong to the family Encyrtidae, a majority of the larval parasitoids are members of five families: Braconidae (24 species/11 genera), Eulophidae (8 species/4 genera), Ichneumonidae (10 species/9 genera), and Eupelmidae (6 species/5 genera). The highest rate of Agrilus egg parasitism (>50%) was exerted by encyrtid wasps (4 species) in North America, Asia, and Europe. In contrast, the highest rate of Agrilus larval parasitism (>50%) was caused by species in two genera of braconids: Atanycolus (North America) and Spathius (Asia), and one eulophid genus, Tetrastichus (Asia and Europe). Medical volume data are rapidly increasing, growing from gigabytes to petabytes, which presents significant challenges in organisation, storage, transmission, manipulation, and rendering. To address the challenges, we propose an end-to-end architecture for data compression, leveraging advanced deep learning technologies. This architecture consists of three key modules: downsampling, implicit neural representation (INR), and super-resolution (SR). We employ a trade-off point method to optimise each module's performance and achieve the best balance between high compression rates and reconstruction quality. Experimental results on multi-parametric MRI data demonstrate that our method achieves a high compression rate of up to 97.5% while maintaining superior reconstruction accuracy, with a Peak Signal-to-Noise Ratio (PSNR) of 40.05 dB and Structural Similarity Index (SSIM) of 0.96. This approach significantly reduces GPU memory requirements and processing time, making it a practical solution for handling large medical datasets. [ABSTRACT FROM AUTHOR]

Oobius agrili Zhang and Huang (Hymenoptera: Encyrtidae) is an important egg parasitoid of the emerald ash borer (EAB), Agrilus planipennis Fairmaire (Coleoptera: Buprestidae). Methods for laboratory-rearing O. agrili have been developed but its mass-production depends on the continuous production and storage of freshly laid EAB eggs as well as diapaused parasitoid progeny (inside parasitized EAB eggs). The purpose of this study was to determine optimal environmental conditions for long-term storage of host eggs as well as diapaused parasitoid progeny. Fresh host eggs and diapaused parasitoid progeny were stored at two low storage temperatures (1.7 and 12.8 °C) and three levels of relative humidity (low ∼31%, medium ∼74%, and high ∼99.9%) for various length of time (15–270 days) and then evaluated for host egg suitability and the reproductive fitness of stored parasitoid progeny. EAB eggs were stored for approximately 30 days without significant reduction of their viability and suitability to O. agrili parasitism at low storage temperatures under high and medium relative humidity. Neither storage temperature or humidity had any significant effects on adult parasitoid emergence for storage durations of up to 270 days. When storage durations were over 120 days, however, both adult parasitoid longevity and fecundity declined approximately 20–30% across all temperature and humidity treatments.
