Henan Province is a typical key area of Chinese main agricultural and food planting. However, a key eco-restoration of degraded farmlands issue needs be resolved by biodiversity technology. Because sustainable agro-ecosystem quickly growth must inject new technology, just as nitrogen-fixing plants can replace the using of chemical nitrogen fertilizers, insect repellent plants can replace the use of chemical insecticides, (Robinia pseudoacacia L.) wood populations surrounding the damaged agricultural ecosystem can block wind impacting on the death of wheat, “plants of soil and water conservation” can avoid the loss of water and nutrients around farmland in low mountain and hilly areas in typical key area of Henan Province of Chinese. This work firstly has been resolved eco-restoration of damaged agro-ecosystem by biodiversity technology.

This study aimed to evaluate the composition of the gastric microbiota in the gastric mucosa and gastric fluid of patients with chronic antral gastritis. Specifically, we sought to determine whether Helicobacter pylori (Hp) infection changes the bacterial community in the gastric mucosa or alters the microbiota in the gastric fluid. The bacterial community at another site in the stomach was also investigated. DNA was extracted from 160 samples collected from 40 patients with chronic antral gastritis (20 Hp-positive and 20 Hp-negative cases). Three tissue samples of the gastric mucosa (gastric angle, body, and antral mucosa) and one tube of gastric fluid were collected from every patient. A 16S rRNA amplification library was created, and high-throughput sequencing was performed. A profile of the community composition was obtained using bioinformatics methods, including cluster, taxonomy, and diversity analyses. Analysis of the gastric bacterial community revealed that the community compositions of the gastric mucosa and gastric fluid of patients without Hp are similar to but show differences from those of Hp-positive patients. The microbiota in Hp-positive patients exhibited reduced microbial diversity, and the gastric fluid of these patients contained a small proportion of Hp. The richness of Leptotrichia in mucosal samples was greater than that in gastric fluid samples from Hp-negative patients with chronic antral gastritis. Hp changes the growth of other microbiota in the mucosa and affects the microbiota in the gastric fluid of patients with chronic antral gastritis. In addition to Hp, the presence of other bacteria might be related to the development of chronic antral gastritis.

To investigate the gene mutation occurved in AML patients with 29 kinds of fusion genes and 51 kinds of tumor gene.Next-generation sequencing (NGS) was used to detected the 49 kinds of targeted gene. FLT3 internal tandem duplication (FLT3-ITD), CALR, NPM1 and CEBPA mutation were detected by DNA-based PCR and Sanger sequencing. Twenty-nine kinds of fusion genes were dected by multiplex nested RT-PCR.The total gene mutation rate was 91% (109/121) in all the 121 patients. On average, 2.1 mutated genes per patient were identified, among these 121 patients, coexistence of ≥ 3 mutations was frequent (34.7%). The most commonly mutated genes were NRAS (23.96%, n=29), followed by NPM1 (14.04%, n=17), CEBPA double mutations (14.04%, n=17), KRAS (11.57%, n=14)，FLT3-ITD (10.74%, n=13), CSF3R (10.74%, n=13), TET2 (9.92%, n=12) and IDH1 (9.1%, n=11). Overall, fusion genes were detected in 47 (37.3%) patients, including AML/ETO (n=12), CBFβ/MYH11 (n=11), PML/RARa (n=12), MLL rearranagement realated mutation MLL-X (n=10). TLS/ERG (n=1) and DEK/CAN (n=1) in an order of decreasing frequency. Patients with normal karyotype (NK)- AML exhibited more mutations in CEBPA, NPM1, TET2, RUNX1 and IDH1, comparing with abnormal karyotype patients. KRAS mutation in abnormal kayotype patients was significantly higher than that in normal kayotype patients (P=0.014). TP53 mutations were predominantly associated with complex cytogenetics (P=0.199). KRAS mutations were more frequent in core binding factor (CBF) acute myeloid leukemia (AML) and 11q23/MLL rearrangement leukemia, compared with NK-AML (P=0.006 and 0.003, respectively). KIT mutations predominated in CBF-AML (P=0.006). JAK2V617F mutations were detected in two patients and co-occurred with AML-ETO fusions.At least one mutation is observed in more than 90% patients. On average, more than 2 mutated genes per patient are identified. Some gene mutations are associated with gene rearrangement.121例急性髓系白血病患者基因突变谱的分析.研究急性髓系白血病（AML）患者29种融合基因及51种肿瘤基因突变的发生情况.采用第二代DNA测序技术检测49种靶基因；采用DNA-PCR检测FLT3-ITD、NPM1基因12号外显子、CALR基因9号外显子及CEBPA的TAD、BZIP 2个功能区的突变发生情况；采用多重PCR法检测29种白血病融合基因.121例患者中，基因突变总发生率为90.1%（109/121），每例患者平均发生2.1个基因突变，34.7%的患者同时携带≥3个基因突变。突变检出率最高的基因为NRAS（23.96%，n=29），其他基因依次为：NPM1（14.04%，n=17）、CEBPA双突变（14.04%，n=17）、KRAS（11.57%，n=14）、FLT3-ITD（10.74%，n=13）、CSF3R（10.74%，n=13）、TET2（9.92%，n=12）和IDH1（9.1%，n=11）。共47例患者检出融合基因，包括AML/ETO 12例，CBFβ/MYH11 11例，PML/RARa 12例，涉及MLL重排的MLL-X融合基因10例，TLS/ERG 1例及DEK/CAN 1例。 CEBPA、NPM1、TET2、RUNX1及IDH1突变在正常核型中的检出率均高于异常核型，差异有统计学意义；KRAS突变在异常核型中的发生率高于正常核型，差异显著（P=0.014）。复杂核型患者的TP53突变检出率明显高于正常核型及其他异常核型（P=0.199）。KRAS突变在核心结合因子白血病及伴有11q23/MLL重排的M5患者中的阳性率均高于正常核型（P=0.006，P=0.003）。KIT基因突变主要见于核心结合因子白血病，两者有相关性（P=0.006）。共检测到2例JAK2V617F突变，均与t (8; 21)/AML1/ETO伴随出现.大于90%的AML患者至少携带1个基因突变，每例患者平均发生2个以上白血病常见致病基因突变，部分基因突变与基因重排有一定相关性.

Background/Aims: This study is aimed at identification of miR-195-5p/MMP14 expression in cervical cancer (CC) and their roles on cell proliferation and invasion profile of CC cells through TNF signaling pathway in CC. Methods: Microarray analysis, gene set enrichment analysis (GSEA) and DAVID were used to analyze differentially expressed miRNAs, mRNAs and signaling pathways. MiR-195-5p and MMP14 expression levels in CC cell were determined by qRT-PCR. Western blot was employed to measure MMP14 and TNF signaling pathway-relating protein level. Luciferase reporter system was used to confirm the targeting relationship between MMP14 and miR-195-5p. Cell proliferation and invasion was respectively deeded by CCK8, transwell. In vivo experiment was carried out to study the impact of MMP14 and miR-195-5p on CC development in mice. Results: The microarray analysis and the results of qRT-PCR determined that miR-195-5p was under-expressed and MMP14 was over-expressed in CC cells. GSEA and DAVID analysis showed that TNF signaling pathway was regulated by miR-195-5p/MMP14 and activated in cervical carcinoma cells. The miR-195-5p and MMP14 have a negative regulation relation. In vivo experiment found that down-regulated MMP14 and up-regulated miR-195-5p suppressed the tumor development. Conclusion: Our results suggest that MMP14 is a direct target of miR-195-5p, and down-regulated MMP14 and up-regulated miR-195-5p suppressed proliferation and invasion of CC cells by inhibiting TNF signaling pathway.

Polychlorinated biphenyls (PCBs) are a class of persistent organic environmental pollutants with a total of 209 homologs. The homolog 2,3',4,4',5-pentachlorobiphenyl (PCB118) is one of the most important dioxin-like PCBs and is highly toxic. PCB118 can accumulate in human tissues, serum and breast milk, which leads to direct exposure of the fetus during development. In the present study, pregnant mice were exposed to 0, 20 and 100 μg/kg/day of PCB118 during the stage of fetal primordial germ cell migration. Compared with the control group, we found morphological alterations of the seminiferous tubules and a higher sperm deformity rate in the male offspring in the treatment groups. Furthermore, the methylation patterns in the treatment groups of the imprinted genes H19 and Gtl2 in the sperm were altered in the male offspring. We also characterized the disturbance of the expression levels of DNA methyltransferase 1 (Dnmt1), Dnmt3a, Dnmt3b, Dnmt3l, and Uhrf1. The results indicated that intrauterine exposure to low doses of PCB118 could significantly damage the reproductive health of the male offspring. Therefore, attention should be paid to the adverse effects of PCB118 exposure during pregnancy on the reproductive system of male offspring.

Polychlorinated biphenyls (PCBs) are persistent organic pollutants, and the widespread use of PCBs has had adverse effects on human and animal health. This study experiment explored the effects of 2,3′,4,4′,5-pentachlorobiphenyl (PCB118) on the mammalian reproductive system. PCB118 was administered to pregnant mice from 7.5 to 12.5 days of gestation; F1 mice were obtained and the reproductive system of F1 male mice was examined. PCB118 damaged the reproductive system in male F1 mice, as evidenced by negative effects on the testicular organ coefficient (testes weight/bodyweight), a decrease in the diameter of seminiferous tubules and a significant reduction in the anogenital distance in 35-day-old F1 mice. In addition, methylation levels of genomic DNA were reduced, with reductions in the expression of the DNA methyltransferases DNMT1, DNMT3A and DNMT3B, as well as that of the epigenetic regulatory factor ubiquitin like with PHD and ring finger domains 1 (Uhrf1). Together, the results of this study provide compelling evidence that exposure of pregnant mice to PCB118 during primordial germ cell migration in the fetus affects the reproductive system of the offspring and decreases global methylation levels in the testis.

Objective: To analyze the effect of azithromycin, montelukast combined with pulmicort respulas therapy on the degree of inflammation and lung function in children with mycoplasma pneumonia. Methods: A total of 318 cases of children with mycoplasma pneumonia treated in our hospital from February 2013 to February 2016 were randomly divided into observation group and control group (n=159). Control group received azithromycin and montelukast therapy, observation group received azithromycin, montelukast combined with pulmicort respulas therapy, and then the degree of inflammation, lung function, etc. were compared between two groups. Results: V-T, t-PTEF/t-E, TEF25/PTEF, FEV1, FVC, FEV1/FVC and MVV values of observation group after treatment were higher than those of control groupand differences in MTIF/MTEF values were not statistically significant between groups; serum CCSP value was higher than that of control group, and IL-17, MCP-4, MDC and CysLTs values were lower than those of control group. Conclusion: Azithromycin, montelukast combined with pulmicort respulas therapy can reduce the systemic inflammatory state and optimize lung function in children with mycoplasma pneumonia, and it has positive clinical significance.

Objective: To study the effect of montelukast combined with procaterol treatment on airway remodeling and inflammatory response in children with mycoplasma pneumonia. Methods: A total of 60 mycoplasma pneumonia children with acute asthma attack treated in our hospital were selected and randomly divided into two groups, observation group received conventional anti-infection, cough-relieving and sputum-reducing combined with montelukast and procaterol spasmolysis and asthma-relieving treatment, control group received conventional anti-infection, cough-relieving and sputum-reducing combined with procaterol spasmolysis and asthma-relieving treatment, and then the degree of airway remodeling and inflammatory response was compared between two groups. Results: Serum MCP-4, MDC and CysLTs levels of both groups 1 week after treatment were significantly lower than those before treatment and serum MCP-4, MDC and CysLTs levels of observation group after treatment were lower than those of control group; 1 week after treatment, LD and the proportion of Treg in induced sputum of observation group were significantly higher than those of control group, WT, WA%, PEF25, PEF50 and PEF75 as well as FN1 and Col-1 levels and the proportion of Th17 in induced sputum were significantly lower than those of control group, and FVC, PEF and FEV1 were not different from those of control group. Conclusions: Montelukast combined with procaterol treatment can significantly improve the airway remodeling and inflammatory response in mycoplasma pneumonia children with acute asthma attack.

Rationale Primary peritoneal epithelioid mesothelioma of clear cell type is an extremely rare entity composed of clear cytoplasm. It is challenging to diagnose because of the morphological resemblance to clear cell tumor. Patients concerns A 69-year-old male patient had swollen lymph nodes in the right inguinal region for 7 months and was constipated for 1 month. Diagnosis The patient was diagnosed as peritoneal epithelioid mesothelioma of clear cell type based on computed tomography scan, pathology, immunohistochemistry, special staining and whole-exome sequencing. This patient harbored VHL gene alteration in exon 1 and homologous recombination defect (with a score of 45). This finding indicated that this patient might be sensitive to platinum-based therapy and Poly ADP-ribose Polymerase (PARP) inhibitor. This patient carried no microsatellite instability, a low level of tumor mutation burden, and a high extent of intratumoral heterogeneity. Eighteen neoantigens were detected. Interventions The patient received surgery-based multidisciplinary treatment by integrating cytoreductive surgery (CRS) with hyperthermic intraperitoneal chemotherapy (HIPEC). HIPEC was administered with docetaxel 120 mg plus cisplatin 120 mg, at 43°C, for 60 minutes. After operation, the patient received intravenous (IV) chemotherapy with docetaxel 60 mg, pemetrexed 750 mg and cisplatin 100 mg, and then intraperitoneal (IP) chemotherapy with docetaxel 40 mg. The patient received interventional therapy of hepatic artery embolization for 5 times. Outcomes Regular follow-up was performed until Oct 14, 2020. The patient died 31.6 months later owing to incomplete intestinal obstruction. Lessons Primary peritoneal epithelioid mesothelioma of clear cell type needs to be differentiated from a variety of clear cell tumors. This disease is characterized by specific genetic alteration. Whole-exome sequencing contributes to guide individualized therapy. CRS-HIPEC helps achieve long-term overall survival.

Transfer of aac(6')-aph(2″) transposons mediating high-level gentamicin resistance (HLGR) in Enterococcus faecalis is a serious problem in the clinic. However, factors affecting the transfer of aac(6')-aph(2″) have not yet been elucidated. The current study aimed to examine the genetic and molecular basis of HLGR in E. faecalis strains isolated in Beijing (China) and to clarify the relationship between transfer efficiency of aac(6')-aph(2″) transposons and the transposon structure/location. A total of five transposon structures were identified by PCR mapping of the corresponding transposon regions, including a Tn5281-like non-truncated transposon and four truncated transposons. A plasmid location study of aac(6')-aph(2″) by Southern blot following S1-PFGE and filter mating conjugation experiments demonstrated that plasmid location rates correlated with conjugation-positive rates. Chromosome walking to identify the sequence upstream of a representative type III truncated transposon found a truncated aph(2″)-Ia region, and further PCR analysis of this region among strains from different groups revealed similar a positive rate trend as the transposon plasmid location rate and conjugation-positive rate. In conclusion, aac(6')-aph(2″) transposons were of different structures in E. faecalis strains from Beijing, with two new transposon structures that have not been reported elsewhere. Presence of the truncated aph(2″)-Ia region upstream of some truncated transposons suggests recombination between aminoglycoside-modifying enzyme genes. Possible links exist among plasmid location, conjugation and the presence of truncated aph(2″)-Ia upstream of the transposon.

Objective To investigate the possible protective effects of sophocarpine on mucosal injury and epithelial barrier disruption on dextran sulfate sodium (DSS)-induced acute colitis. Methods Male BALB/c mice were randomly divided into three groups. The mice in normal group were given normal water, and those in model and sophocarpine-treated groups were given 2.5% DSS for 6 d to induce acute colitis. Sophocarpine (30 mg/kg) was ip administered once daily during the study period. Severity of colitis was evaluated by disease activity index (DAI), histological injury and inflammatory cytokine production including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and monocyte chemoattractant protein-1 (MCP-1). The colonic barrier disruption was assessed by testing the expression of junctional adhesion molecule-1 (JAM-1), E-cadherin (E-CAD), and desmocollin-2 (DSC-2) in colon mucosa. Expression of HNF4α in colon mucosa was detected by immunohistochemistry staining and real-time RT-PCR, respectively. Results Compared with normal group, DAI, colonic shortening, and histopathological injury in model group were elevated (P Conclusion Sophocarpine can enhance the expression of HNF4α, promote the expression of colonic intrecellular junctions, thus, maintain the integrity of the colonic barrier and inhibit the colitis process. We suggest that sophocarpine could enhance the production of cellular junction proteins to protect the intestinal barrier fuction, at least partly, in HNF4α-dependent pathway.

Wogonin, a natural flavonoid, is one of the bioactive compounds of the medicinal herb Eucommia ulmoides OLIV. widely used in southeastern Asia for treating hypertension. However, the molecular mechanisms for the therapeutic benefits remain largely unclear. The present study investigated the vasodilatory effect of wogonin and its possible mechanisms. The flavonoid (0.1-100 µM) caused concentration-dependent relaxations in endothelium-intact aortic rings precontracted with norepinephrine (NE, 1 µM) or potassium chloride (KCl, 60 mM). Preincubation with wogonin (10, 100 µM) for 20 min significantly inhibited the contractile responses to NE (0.1, 1, 10 µM) or KCl (7.5, 15, 30, 60 mM). Relaxant responses to wogonin were not inhibited by N(G)-nitro-L-arginine methylester (100 µM) or endothelial denudation. In a Ca(2+)-free Krebs' solution, wogonin not only blocked Ca(2+) influx-dependent vasoconstriction by either NE (1 µM) or KCl (100 mM), but also inhibited NE (1 µM)-induced tonic contraction, which is dependent on intracellular Ca(2+) release. Wogonin also suppressed the elevation of [Ca(2+)]i induced by KCl (60 mM) after exhausting the calcium store in sarcoplasmic and endoplasmic reticula with thapsigargin (1 µM) or by ATP (100 µM) in primary vascular smooth muscle cells. These findings suggest that wogonin-induced responses are mainly due to the inhibition of both intracellular Ca(2+) release and extracellular Ca(2+) influx.

Background/Aims: Pioglitazone, a peroxisome proliferator-activated receptor γ activator, is clinically used to treat insulin resistance. However, the underlying mechanism of pioglitazone's action remains unclear. We investigated whether, and how, pioglitazone modulates serum level of retinol binding protein 4 (RBP4), an adipocytokine associated with obesity and insulin resistance. Methods: Insulin sensitivity was determined by oral glucose tolerance test, and RBP4 expression was detected by RT-PCR and Western blotting. Results: Pioglitazone treatment significantly decreased serum RBP4 levels in obese rats, which was correlated with reduced body weight and increased insulin sensitivity. Moreover, pioglitazone greatly decreased RBP4 mRNA and protein levels in adipose tissue but not in the liver. Consistently, pioglitazone treatment significantly reduced RBP4 protein expression in 3T3-L1 adipocytes but not in HepG2 cells. Conclusion: These results demonstrate that pioglitazone inhibits the level of serum RPB4 by suppressing RBP4 expression in adipose tissue of obese rats, suggesting that inhibiting RBP4 expression in adipocytes may provide a mechanism by which pioglitazone improves insulin sensitivity in insulin-resistant subjects.

In order to detection wind turbine equipment and accurately assess the the equipments condition, this paper combines cloud theory, and propose a new comprehensive evaluation method about wind turbine operating status. Use AHP to establish wind turbine evaluation system, and taking advantage of variable weight mode correct quantitative indicators right weight. Adopt Cloud model function represent the indicator data, and establish improved evaluation model, finally, an example verify the effectiveness of the method.

Based on gas chromatography mass spectrometry techniques, the biomarker characteristics of the Hongqi Formation source rocks in the Tuquan basin are studied, information and petroleum geological significance on organic matter origin, sedimentary environment and organic matter maturity are revealed. Biomarker assemblage characteristics of the Hongqi Formation source rocks are described as follows: Long-chain tricyclic terpane and gammacerane is abundant, C27 regular steranes is predominant. The low content of gammacerane in organic matter shows that the source rocks were deposited in the low salinity environment. The mean value of steranes/hopanes is 0.40, showing more contribution from algae than bacteria. High content of C27 regular steranes indicate that the parent material input is mainly the low aquatic oSubscript textrganism. Biomarker parameters including Ts/(Ts+Tm) ratio, C29 steranes 20S/(20S+20R) ratio, C29 sterane αββ/(αββ+ααα) ratio indicate that Hongqi Formation source rock has reached or surpassed the main phase of oil generation.

As the lack of carbon source is a key problem of in-situ denitrification in groundwater, choosing appropriate solid carbon source materials is very important. In this study, wheatstraw and sawdust were selected as potential carbon sources to evaluate the performance for in-situ biological denitrification in groundwater by three laboratory-scale column experiments operated at a flow rate of 2.0 cm3/min for 78 d. The results showed that sawdust was a suitable carbon source with less release of nitrogen compounds and relatively stable release of organic carbon, compared with wheatstraw, and was applicable for further use as a filling material in in-situ groundwater bioremediation.

Use software Pro/E to build a blade model based on 1.5MW wind turbine, analyze stress characteristics of different regions including spar cap, webs and trailing edge of the dangerous section of blade at related loads by using different strength criteria to make finite element analysis to check the strength, through comparing the F.I.(fatigue index) and stress bringing about the idea that using different strength criteria to analyze different material of different regions can ensure the security and reliability of the designed blade at large extent.

Large collapse of underground house of a hydroelectric power station happened while excavation of the top arch. The volume of collapse was up to 5000m3. With consideration of the field geological conditions and practical construction, by strength reduction finite element method (FEM) and discrete element method (DEM), the process of collapse is simulated, which is compared with the field monitoring data. The mechanism of instability failure is analyzed. Some conclusions are obtained. Firstly, the development of fractured weak zone β80, faults, joints and fissures above the top arch are the basic factors to induce this collapse. Secondly, abrupt blasting without supporting is the direct factor. So collapse of underground house can be avoided by reinforcement and supporting on the geological weak zone in time.

For the cast-steel joint of large span roof system, the main difficulties of prototype stress test encountered are the large scale of the structure system, large loading system and large supporting system, etc. The hydraulic jack of high tonnage is adopted to apply the load. The frame structure of channel steel is used to fix the cast-steel joint and the loading system. The reverse loading system is elaborated to simulate the axial tensile force. So prototype stress test on the cast-steel joint of large span roof system is accomplished successfully. The test results show that, under the designing load condition, cast-steel joint is in a linear elastic state, the structure is globally safe; the maximum surface stress of cast-steel joint is adjacent to the crossing point of steel pipes; the stress value of test is well coincident with one of finite element analysis. This test device system is especially suitable for the stress state test of large components, and can also provide important reference to the similar projects.

Base on finite element method of composite, take 5MW horizontal axis wind turbine blades as example, skin uses a mixture of fiberglass and carbon fiber as ply, spar caps and web adopt carbon fiber ply entirely to build the finite element model of the blade. The total weigh of the blade is 20.2 ton. Use Bladed software calculated the limit load of each cross-section, analyzed the stress distribution of each section and the modal characteristics of the blade, these provide a theoretical reference for the application of carbon fiber using on MW class wind turbine blade.

Several studies showed that the up-regulation of glial glutamate transporter-1 (GLT-1) participates in the acquisition of brain ischemic tolerance induced by cerebral ischemic preconditioning or ceftriaxone pretreatment in rats. To explore whether GLT-1 plays a role in the acquisition of brain ischemic tolerance induced by intermittent hypobaric hypoxia (IH) preconditioning (mimicking 5,000 m high-altitude, 6 h per day, once daily for 28 days), immunohistochemistry and western blot were used to observe the changes in the expression of GLT-1 protein in hippocampal CA1 subfield during the induction of brain ischemic tolerance by IH preconditioning, and the effect of dihydrokainate (DHK), an inhibitor of GLT-1, on the acquisition of brain ischemic tolerance in rats. The basal expression of GLT-1 protein in hippocampal CA1 subfield was significantly up-regulated by IH preconditioning, and at the same time astrocytes were activated by IH preconditioning, which appeared normal soma and aplenty slender processes. The GLT-1 expression was decreased at 7 days after 8-min global brain ischemia. When the rats were pretreated with the IH preconditioning before the global brain ischemia, the down-regulation of GLT-1 protein was prevented clearly. Neuropathological evaluation by thionin staining showed that 200 nmol DHK blocked the protective role of IH preconditioning against delayed neuronal death induced normally by 8-min global brain ischemia. Taken together, the up-regulation of GLT-1 protein participates in the acquisition of brain ischemic tolerance induced by IH preconditioning in rats.

In this paper, a method of carbon nanotube coated with the dendrimer encapsulated platinum nanoparticles（Pt-DENs/CNTs） was introduced, and the properties of enzyme electrode modified by the Pt-DENs/CNTs were investigated. The formation of the self-assembled (GOx/Pt-DENs)n/CNTs construction was expiored by high resolution transmission electron microscopy (HRTEM). And the results indicated that the uniform growth of the layer-by-layer nanostructures onto carboxyl-functionalized CNTs. The electrochemical properties of the enzyme anode modified by (GOx/Pt-DENs)n/CNTs, which act as biofuel cell anode, was studied by electrochemica measurements. Owing to its nanostructure, the (GOx/Pt-DENs)n/CNTs heterostructures have a large specific area, a high surface active center, good conductivity and macro tunnel effect, therefore the electrode modified by (GOx/Pt-DENs)n/CNTs has series of attractive electrochemical characteristics so as to enhance biofuel cell’s function.

To study the preventive effect of sophocarpine (Soc) on dextran sulfate sodium (DSS)-induced colitis in mice, in order to analyze the influence of Soc on toll like receptor 4 (TLR4)/mitogen-activated protein kinases (MAPKs) and janus tyrosine kinase 2 signal transducer and activator of transcription 3 (JAK2/STAT3) signal pathways in mice intestinal tissues. The mice was given 2.5% DSS for 6 days to induce the acute colitis model. The Soc-treated group was intraperitoneally injected with sophocarpine 30 mg · kg(-1) · d(-1) since the day before the experiment to the end. The disease activity index (DAI) was assessed everyday, and the colonic morphology and histological damage were observed with HE staining. The mRNA expressions of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-6 (IL-6) were detected by real-time RT-PCR. The changes in key protein kinase p38 mitogen-activated protein kinase (p38MAPK), c-Jun NH2-terminal protein kinase1/2 (JNK1/2), extracellular signal-regulated kinase1/2 (ERK1/2), JAK2, STAT3 in TLR4/MAPKs and JAK2/STAT3 signaling pathways were detected by western blot. The result showed that the model group showed statistical significance in body weight, DAI, colon length and histopathological changes compared with the normal group (P0.05); however, the Soc-treated group showed significant improvements in the above indexes compared with the model group (P0.05). TNF-α, IL-1β and IL-6 in the model group was significantly higher than that in the normal group (P0.05), but lowered in the Soc-treated group to varying degrees (P0.05). In the normal group, the expressions of TLR4 and the phosphorylation of P38, JNK1/2, JAK2, STAT3 were at low levels; in the model group, the phosphorylation of P38, JNK1/2, JAK2, STAT3 increased; the Soc-treated group showed a decrease in TLR4 expression compared with the model group, with notable declines in the phosphorylation of TLR4, P38, JNK1/2, JAK2, STAT3. These findings indicate that Soc can inhibit TLR4/MAPKs, K2/STAT3 signaling pathway activation, reduce the expression of proinflammatory cytokines TNF-α, IL-1β and IL-6 and relieve inflammatory reactions, so as to effectively prevent experimental colitis.

Menopause is characterized by a decrease in life quality due to the appearance of uncomfortable symptoms. Nowadays, Understanding menopause-associated pathophysiology and developing new strategies to improve the treatment of menopausal-associated symptoms is an important issue. Our study was to evaluate the synergistic effects of Danshen (salvia miltiorrhiza bunge) and the phytoestrogenic effects of 3 modified Qing E formulas, to explore a better formula for menopausal disorders. 100 rats were randomized into 5 groups: Sham (Sham operation group), OVX (model group of ovariectomized rat), BDL (group with low concentration of Qing E Formula), BDH (group with high concentration of Qing E Formula) and BDD (group with high concentration of Qing E Formula Plus Danshen), receiving vehicle and extract of different modified Qing E formula respectively. The food intake, body weight, uterus weight, blood levels of triglycerides (TG), total cholesterol (TC) and cholesterol fractions were assessed. The mammary glands and uterus were morphologically analyzed. The bone density of tibias were measured by peripheral quantitative computed tomography (pQCT). Additionally, luciferase induction assays were performed in Hela cells with the mixtures derived from Qing E formula plus Danshen (BDD). Qing E formula plus Danshen significantly increased the uterus wet weight, enhanced the thickness of uterine wall, endometrial epithelium and glandular epithelium, improved trabecular bone and total density evidently, reduced the levels of low density lipoprotein cholesterol (LDL-C) and TG, possessed notable estrogen receptor beta (ERβ) and estrogen receptor alpha (ERα) agonist activity. Qing E formula plus Danshen exerted more evident estrogen-like effects, thus it has a potential therapeutic use to treat menopausal disorders.

Plateau zokor (Myospalax baileyi) is one of the blind subterranean mole rats that spend their life solely underground in sealed burrows. It is one of the special species of the Qinghai-Tibet plateau. In their burrows, oxygen is low and carbon dioxide is high and their contents fluctuate with the change of seasons, soil types, rain and depth of burrows. However, plateau zokors show successful adaptation to that extreme environment. In this study, their adapting mechanisms to the hypoxic hypercapnic environment were analyzed through the comparison of their blood-gas properties with that of pikas (Ochotona curzniae) and Sprague-Dawley rats. The results indicated that plateau zokors had higher red blood corpuscle counts (8.11+/-0.59 (10(12)/L)) and hemoglobin concentrations (147+/-9.85 g/L), but hematocrit (45.9+/-3.29%) and mean corpuscular volume (56.67+/-2.57 fL) were lower than the other rodents. Their arterial blood and venous blood pH were 7.46+/-0.07 and 7.27+/-0.07. Oxygen pressure in arterial blood of plateau zokors was about 1.5 times higher than that of pikas and rats, and it was 0.36 and 0.26 times in their venous blood. Partial pressure for carbon dioxide in arterial and venous blood of plateau zokors was 1.5-fold and 2.0-fold higher, respectively, than in rats and pikas. Oxygen saturation of plateau zokors was 5.7 and 9.3 times lower in venous blood than that of pikas and rats, respectively. As result, the difference of oxygen saturation in arterial blood to venous blood was 2- and 4.5-fold higher in plateau zokors as that of pikas and rats, respectively. In conclusion, plateau zokors had a high tolerance to pH changes in tissues, together with strong capabilities to obtain oxygen from their hypoxic-hypercapnic environment.

To evaluate the expression of proteasome β5 subunit in human atherosclerotic plaque.The specimens of carotid endarterectomy were collected from 16 carotid stenosis patients. For the case group, they were divided into 2 groups: plaque (group A) and areas adjacent to plaque (group B). And carotid endarterium was obtained from 6 patients undergoing aortic replacement surgery. Relatively normal arterial intima (subclavian arteryinnominate artery) was selected as the control group (group C). Content of proteasome β5 subunit was evaluated by hematoxylineosin (HE) staining, immunohistochemistry and Western blot.(1) In patients with diabetes, hyperlipidemia and probable smoking, proteasome β5 subunit was enhanced in all groups; (2) Proteasome β5 subunit was demonstrated in both cytoplasm and nucleus of vascular smooth muscle cells, but mainly confined to cytoplasm; (3)The positive expression rate of β5 was 9/16 in group A, 11/16 in group B and 6/6 in group C. There was significant divergence in the expression of β5 in different sample sub-groups: group A (atherosclerotic plaque)group B (areas adjacent to plaque)group C (control group).The expression of β5 decrease markedly in human atherosclerotic plaques.

This algorithm can simplify the computation significantly and improve the real time character of fault location, meanwhile, it suits the fault appears at the terminal of feeder line. The effectiveness of proposed method is verified by sample test.

The main generalizations come from an elementary inequality and by means of the modification of Medveď’s de-singular approach.

To explore the adapting metabolic mechanisms of the plateau zokors to the hypoxic-hypercapnic environment.The activities of lactate dehydrogenase in serum and tissues, and the content of lactate in serum of plateau zokors in spring, summer and autumn were determined by using method of enzyme analysis. The spectrums of lactate dehydrogenase isoenzymes in serum and tissues of plateau zokors in spring, summer and autumn were analyzed by using method of the discontinuous systemic poly-acrylamide perpendicular plank gel electrophoresis.The activities of lactate dehydrogenase in serum had obvious seasonally difference that were higher in spring and lower in autumn, and the content of lactate in serum showed same changing pattern. The spectrums of lactate dehydrogenase isoenzymes in serum showed five bands that were LDH1, LDH2, LDH3, LDH4 and LDH5 from positive pole to negative pole respectively, it showed clearly two bands in serum of summer that were LDH4 and LDH5 and one band in serum of autumn that was LDH5. The activities of LDH in tissues of skeleton muscle, cardiac muscle and brain were higher compared with the other tissues, it decreased markedly from spring to summer to autumn. In tissues of liver, kidney and lungs, activities of LDH were lower. Activities of LDH in livers, were significantly higher in spring compared that in summer and autumn, which had no obvious difference between summer and autumn. Activities of LDH in kidneys and lungs, showed no obviously difference between spring and summer, which decreased markedly in autumn. The spectrums of lactate dehydrogenase isoenzymes in tissues of cardiac muscle, liver, lungs, kidney, brain and skeleton muscle showed five bands, the spectrums were obvious different in different tissues, and the content of LDH isoenzymes showed seasonal changes in different tissues.Glycolysis levels in plateau zokors had obvious seasonally change which increased in spring and decreased in autumn significantly. It related to the activity of plateau zokors in different seasons and seasonal fluctuation of oxygen and carbon dioxide in burrows of plateau zokors.

With the better capabilities of anti-jamming, low-probability of interception and networking, frequency hopping communication is one of the most widely used technologies at present. In order to study and validate the anti-jamming capability of FH communication system, it is urgently needed to establish a high efficiency simulation platform. Based on the characteristics of FH communication system, this paper designs a kind of more high-efficient and practical simulation platform. The bit error rate (BER) performance is simulated under the two conditions of Gaussian noise and Gaussian noise plus partial-band jamming.

To investigate the detection limit of multicolor combinational probe coding real-time PCR (MCPC-PCR) in detection of Salmonella and Staphylococcus aureus suspended in the food samples, and to apply MCPC-PCR to detect the samples of food poisoning.Series concentration of bacterium suspension (10(1) - 10(9) CFU/ml) was prepared by using 22 simulated samples including fresh meat and cakes and then MCPC-PCR was applied to detect Salmonella and Staphylococcus aureus in 22 samples. Enrichment broth of 101 frozen samples and 5 early patients' anal swabs in food poisoning cases were detected after the DNA samples were extracted.The limits of MCPC-PCR assay in detecting Salmonella and Staphylococcus aureus were about 10(2) copies/test; 101 frozen enrichment broth of samples in food poisoning cases were detected by MCPC-PCR assay, of 23 positive samples, 18 were confirmed by bacteriology techniques; 96 samples detected by MCPC-PCR and bacteriology techniques had the same results, and the coincidence rate was 95.05%. Anal swabs, collected from 5 of early patients in a food poisoning case gave a clue to be Vibrio parahaemolyticus by MCPC-PCR assay and then were perfectly consistent with bacteriology assay.As a method of high sensitivity and good specificity, MCPC-PCR assay can quickly and conveniently detect multiple pathogens existing in food samples, therefore we recommend it to be used in rapidly screening or simultaneous detection of food-borne diseases.

Objective Proteasome is a mutiple-unit protease working on non-lysosomal protein degradation in cells. Recent studies have illustrated its potential roles in the whole process of atherosclerosis including endothelial dysfunction, formation of foam cells, smooth muscle cell proliferation, and vascular remodeling. This research aims to evaluate the expression of the proteasome β5 subunit in atherosclerotic plaques from patients subjected to aortic replacement surgery. Methods We assigned the atherosclerotic plaque tissue, from carotid endarterectomy for carotid stenosis patients, as the case group (16 cases). According to different degrees of arterial stenosis, we divided the samples into the edge portion (stenosis>50%, A group) and the core portion (stenosis≤50%, B group). Carotid endarteriums were obtained from four patients undergoing Aortic replacement surgery, selected relatively normal arterial intima (Subclavian artery and Innominate artery) as the control group; Collected clinical data of patients, such as age, sex, smoking history, hypertension, diabetes, hyperlipidemia, and the biochemical indicators. A portion of the sample was fixed in 10% formalin, dehydrationed in gradient alcohol, Transparented in xylene, embedded in paraffin, sections were prepared for HE staining and immunohistochemical analysis. Another part of the tissue was frozen in liquid nitrogen, for extracting Total protein, and Western Blot analysis. Data are expressed as percentage or mean±SE for continuous variables and by percentage for qualitative variables. The proteasome β5 subunit activity variables were compared by use of the χ2 test. Statistical significance was assumed for p Results (1) The decrease in the level of β5 had something to do with the risks of diabetes, Hyperlipidemia and possibility of smoking in the patients we observed; (2) β5 was expressed in all samples, most of which accumulated in cytoplasm with few exceptions in nucleus; (3) There was significant divergence in the expression of β5 in different sub-groups of samples: A group (arterial stenosis>50%) Conclusion The expression of β5 was decreased in the atherosclerotic plaques of patients, implying it might play an important role in the progress of atherosclerosis.
