With the rapid development of clinical diagnosis and treatment, many traditional and conventional in vitro diagnosis technologies are unable to meet the demands of clinical medicine development. In this situation, nanomaterials are rapidly developing and widely used in the field of in vitro diagnosis. Nanomaterials have distinct size-dependent physical or chemical properties, and their optical, magnetic, electrical, thermal, and biological properties can be modulated at the nanoscale by changing their size, shape, chemical composition, and surface functional groups, particularly because they have a larger specific surface area than macromaterials. They provide an amount of space to modify different molecules on their surface, allowing them to detect small substances, nucleic acids, proteins, and microorganisms. Combining nanomaterials with in vitro diagnosis is expected to result in lower detection limits, higher sensitivity, and stronger selectivity. In this review, we will discuss the classfication and properties of some common nanomaterials, as well as their applications in protein, nucleic acids, and other aspect detection and analysis for in vitro diagnosis, especially on aging-related nanodiagnostics. Finally, it is summarized with guidelines for in vitro diagnosis.

Accurate detection of tea diseases is essential for optimizing tea yield and quality, improving production, and minimizing economic losses. In this paper, we introduce TeaDiseaseNet, a novel disease detection method designed to address the challenges in tea disease detection, such as variability in disease scales and dense, obscuring disease patterns. TeaDiseaseNet utilizes a multi-scale self-attention mechanism to enhance disease detection performance. Specifically, it incorporates a CNN-based module for extracting features at multiple scales, effectively capturing localized information such as texture and edges. This approach enables a comprehensive representation of tea images. Additionally, a self-attention module captures global dependencies among pixels, facilitating effective interaction between global information and local features. Furthermore, we integrate a channel attention mechanism, which selectively weighs and combines the multi-scale features, eliminating redundant information and enabling precise localization and recognition of tea disease information across diverse scales and complex backgrounds. Extensive comparative experiments and ablation studies validate the effectiveness of the proposed method, demonstrating superior detection results in scenarios characterized by complex backgrounds and varying disease scales. The presented method provides valuable insights for intelligent tea disease diagnosis, with significant potential for improving tea disease management and production.

Abstract Background Pathogenic PAK1 variants were described to be causative of neurodevelopmental disorder with macrocephaly, seizures, and speech delay. Herein, we present a de novo PAK1 variant combine with a de novo terminal 1q microdeletion in a Chinese pediatric patient, aiming to provide more insights into the underlying genotype–phenotype relationship. Methods Enrolled in this study was a 6-year-old girl with clinical features of global developmental delay, severe intellectual disability, speech delay, and seizures from Quanzhou region of China. Karyotype and chromosomal microarray analysis (CMA) were performed to detect chromosome abnormalities in this family. Whole exome sequencing (WES) was performed to investigate additional genetic variants in this family. Results No chromosomal abnormalities were elicited from the entire family by karyotype analysis. Further familial CMA results revealed that the patient had a de novo 2.7-Mb microdeletion (arr[GRCh37] 1q44(246,454,321_249,224,684) × 1]) in 1q44 region, which contains 14 OMIM genes, but did not overlap the reported smallest region of overlap (SRO) responsible for the clinical features in 1q43q44 deletion syndrome. In addition, WES result demonstrated a de novo NM_002576: c.251C > G (p.T84R) variant in PAK1 gene in the patient, which was interpreted as a likely pathogenic variant. Conclusion In this study, we identify a novel PAK1 variant associated with a terminal 1q microdeletion in a patient with neurodevelopmental disorder. In addition, we believe that the main clinical features may ascribe to the pathogenic variant in PAK1 gene in the patient.

To achieve a micro-destructive and rapid measurement of the wood density of standing trees, this study investigated the possibility of the unified modeling of multiple tree species, the reliability of the micro drilling resistance method for measuring wood density, the relationship between drilling needle resistance and wood density, and whether moisture content has a significant impact on the model. First, 231 tree cores and drill resistance data were sampled from Pinus massoniana, Cunninghamia lanceolate, and Cryptomeria fortunei. The basic density and moisture content of each core were measured, and the average value of each resistance data record was calculated. Second, the average drill resistance, the natural logarithm of average drill resistance, and absolute moisture content were used as independent variables, while the basic wood density was used as the dependent variable. Third, the total model of the three tree species and sub-model for each tree species were established through a stepwise regression method. Finally, the accuracy of each model was compared and analyzed with that of using the average basic density of each tree species as an estimated density. The estimated accuracy of the total model, sub model, and average wood density modeling data were 90.070%, 93.865%, and 92.195%, respectively. The results revealed that the estimation accuracy of the sub-model was 1.670 percentage points higher than that of the average wood density modeling data, while the estimation accuracy of the total model was 2.125 percentage points lower than that of the average wood density modeling data. Additionally, except for Cryptomeria fortunei, the natural logarithm of drill resistance significantly influenced the wood density model at a significance level of 0.05. Moreover, moisture content significantly affected the total model and sub-models of Pinus massoniana at a significance level of 0.05. The results indicated the feasibility of using the micro-drilling resistance method to measure the wood density of standing trees. Moreover, the relationship between wood density and drill resistance did not follow a linear pattern, and moisture content slightly influenced the drill needle resistance. Furthermore, the establishment of a mathematical model for each tree species was deemed essential. This study provides valuable guidance for measuring the wood density of standing trees through the micro-drilling resistance method.

To improve the estimation accuracy of wood density and study the linear correlation between drill feed resistance and wood density, a new micro drill instrument prototype that can simultaneously measure the rotation resistance and feed resistance of the drill needle was designed. The test tree species included hard and soft broad-leaved trees and coniferous trees, and the absolute dry density of each wood sample was measured. The drill resistance data were tested by our newly proposed prototype and a Resistograph 650-SC, and four linear models were established to define the relation between drill resistance and the absolute dry density of the wood. The results showed that (1) the statistical indicators of each model for our proposed prototype were better than the corresponding indicators of the Resistograph 650-SC for three of the four species tested; (2) the coefficient of determination of the linear regression model between the feed resistance of our proposed prototype and the absolute dry density of wood was 0.946; and (3) the statistical indicators of the model that included rotation resistance and feed resistance were better than those of the model that only including rotation resistance. Although the proposed prototype produced a competitive level of accuracy and explicitly demonstrated that including feed resistance improved wood density measurement accuracy, the prototype should be considered a first iteration as further hardware design changes and in-forest performance assessments across wider diverse set of test species are required before a conclusive evaluation can be rendered.

A finite element simulation process based on methods of mutual calling between MATLAB and ANSYS software packages are proposed, which has the ability to consider the influence of tension insulator string weight on the sag of a span in a tension section. The convergence criterion of the process can be controlled artificially and is defines to converge when the maximum sag of two calculation steps is very small. A simplified theoretical analysis method is also developed for comparison and verification. These proposed methods are then applied to a tension section and a three-span actual line. Results show that when using a finite element simulation to obtain the curve of a wire, if the number of elements in the span is 10, the mid-span sag in the span is consistent with the theoretical result. And if the number of elements in the span is greater than 50, the sag curve of the conductor is almost unchanged. The two methods and the difference between them are very small, indicating the efficiency of the proposed process.

Corroded transmission towers, whose load-bearing capacities are lowered, are suffering from wind-induced damage. By simulating the member corrosion through section thinning, the structural dynamic finite element model of an angle-steel transmission tower with a corrosion depth of 0 mm to 1.4 mm was established using ANSYS/LS-DYNA software. The incremental dynamic analysis method was used to study the collapse mode of the tower under different degrees of corrosion, as well as the effects of corrosion on structural self-vibration characteristics, wind-induced member stress, and tower-top displacement. The results show that with the corrosion deepening, the first three vibration frequencies decrease, and the torsional mode becomes the first mode. When the corrosion depth is smaller, the overall structure failure occurs under strong wind. The failure starts at the most unfavorable oblique rod between the upper and middle cross arm, then expands to the rods under the lower cross arm. When the corrosion depth is larger, the overall structure failure occurs restrictedly between the upper and middle cross arm. Internal axial force and bending moment redistribution happen when corrosion deepens. Under the critical failure state, the oblique rods are always at a high stress level, while the stresses of the main rods decrease. Corrosion has a stronger impact on component strength and stability than on structural stiffness.

Abstract Aims There is little knowledge about partial trisomy 4q and the genotype–phenotype correlation. In this study, we presented the detail of two Chinese families with partial distal 4q duplication in an attempt to clarify the correlation between the genotype and the phenotype. Methods Two pedigrees with distal 4q duplication were enrolled in this study. Karyotype analysis and single-nucleotide polymorphism (SNP) array detection were performed for prenatal diagnosis. Fluorescence in situ hybridization analysis. (FISH) was conducted to verify the copy number variants. Results Two families with partial trisomy 4q were identified. The fetus in pedigree 1 exhibited multiple ultrasound anomalies including intrauterine growth restriction and an atrioventricular septal defect who had a duplication of 4q28.3-qter associate with 6p25.2-p25.3 deletion, which resulted from balanced translocation carried by his father t(4;6)(q28.3;p25.2). The fetus in pedigree 2 had a distal 4q28.3-qter duplication combined with monosomy of Xp21.3-p22.3, and the karyotype was described as 46,X,der(X)t(X;4)(p21.3;q28.3)mat, which originally inherited from the pregnant woman who exhibited a mild clinical phenotype limited to short stature. Conclusions In our study, we for the first time identified the partial trisomy 4q associate with 6p or Xp deletion. In addition, our finding further strengthens that mild clinical phenotype in 4q duplication case may be due to the spreading of X inactivation to the autosomal in derivation of chromosome X.

Background: Preterm human milk has advantages over preterm formula (PF), but it may compromise some functions after pasteurization. Objective: To explore the effects of preterm donor milk (DM) on growth, feeding tolerance, and severe morbidity in very-low-birth-weight infants. Method: This was a single-center, prospective cohort study that included 304 preterm infants weighing 0.05). Feeding intolerance in the DM group was significantly lower than that in PF group (P < 0.05), and parenteral nutrition time and hospitalization time were also shorter than that in the PF group (P < 0.05). Moreover, the incidence of necrotizing enterocolitis and sepsis was also significantly lower in the DM group (P < 0.05). Conclusion: The study indicated that preterm DM does not affect the growth of very-low-birth-weight infants. Further, it significantly reduces feeding intolerance, helps achieve full enteral feeding early, and has protective effects against necrotizing enterocolitis and sepsis. Thus, compared with formula, preterm DM can lower the rate of infection in preterm infants and is worthy of promotion.

Abstract Background Reprogrammed glucose metabolism, also known as the Warburg effect, which is essential for tumor progression, is regarded as a hallmark of cancer. MAP17, a small 17-kDa non-glycosylated membrane protein, is frequently dysregulated in human cancers. However, its role in hepatocellular carcinoma (HCC) remains largely unknown. Methods Immunohistochemistry was used to analyze the expression pattern of MAP17 in HCC. Loss-of-function and gain-of-function studies were performed to investigate the oncogenic roles of MAP17 in vitro and in vivo. RNA sequencing, co-immunoprecipitation, immunofluorescence and western blotting were used to study the molecular mechanism of MAP17 affecting the tumor growth and glycolytic phenotype of HCC. Results An integrative analysis showed that MAP17, a small 17-kDa non-glycosylated membrane protein, is significantly related to the glycolytic phenotype of hepatocellular carcinoma (HCC). Firstly, we found that MAP17 expression is hypoxia-dependent and predicts a poor prognosis in HCC. Genetic silencing of MAP17 reduced the rate of glucose uptake, lactate release, extracellular acidification rate, and expression of glycolytic genes. Ectopic expression of wild type MAP17 but not its PDZ binding domain mutant MAP17-PDZm increased tumor glycolysis. Further research showed that MAP17 knockdown markedly retarded in vivo tumor growth in HCC. Importantly, attenuation of tumor glycolysis by galactose largely hijacked the growth-promoting role of MAP17 in HCC cells. RNA sequencing analysis revealed that MAP17 knockdown leads to transcriptional changes in the ROS metabolic process, cell surface receptor signaling, cell communication, mitotic cell cycle progression, and regulation of cell differentiation. Mechanistically, MAP17 exerted an increased tumoral phenotype associated with an increase in reactive oxygen species (ROS), which activates downstream effectors AKT and HIF1α to enhance the Warburg effect. In HCC clinical samples, there is a close correlation between MAP17 expression and HIF1α or phosphorated level of AKT. Conclusions Our results show that MAP17 is a novel glycolytic regulator, and targeting MAP17/ROS pathway may be an alternative approach for the prevention and treatment of HCC.

Cognitive neuroscience research has found that individuals from different cultures have different neural responses and emotional perceptions. Differences in executives’ perception of external pay gaps in different cultures can affect their work attitudes and behavior. In this study, we explore the direct relationship between executive compensation fairness and executive innovation motivation. We also investigate the moderating effects of Confucian culture and western culture between executive compensation fairness and executive innovation motivation. Data were collected from the Chinese listed firms from 2011 to 2019 and test the relationship using the fixed-effect panel regression models. The results demonstrate that executive compensation fairness positively influences the executive innovation motivation. This effect is more pronounced in Confucian culture regions, while Western culture weakens this effect. The findings of this study confirmed that executive compensation fairness, provide a cross-cultural comparison for compensation research, validate the findings of cultural neuroscience, and provide useful insights into the research of common prosperity. To improve the corporate compensation structure, it is necessary to consider the relative pay equity with firms in the same region and the influence of corporate culture.

To achieve a comprehensive understanding of a three-dimensional (3D) wind field and the speed-up phenomenon in a downburst wind flow over a 3D hilly terrain, a succession of laboratory tests utilizing 12 hill models with cosine-squared cross-section was conducted using a physical downburst simulator with a jet diameter of 0.6 m. By placing the models in the strong horizontal wind region and the strong vertical wind region, the corresponding wind profiles for both the horizontal and vertical velocities were measured. It was found that the wind flowed predominantly over the crest of the hill in the case of low hills, whereas wind flow around the hill body became increasingly pronounced as the hill height increased. In addition, the speed-up region, where the horizontal wind velocity exceeds the impinging jet velocity, was identified, and found to move from the crest to the two sides of the hill as the hill height increased. Accordingly, the most significant topographic multipliers of all locations on the hill might appear at the crest, the hill foot, or elsewhere, depending largely on the hill height. Among all cases, the maximum topographic multiplier was 1.12, and occurred at the ridge, while the ratio of hill height to jet height was 5/12. Additionally, empirical equations are presented to facilitate the determination of wind loads induced by a downburst flow over an isolated hill.

Non-alcoholic fatty liver disease (NAFLD) is a common disease, which is characterized by the accumulation of triglycerides in the hepatocytes without excess alcohol intake. Circadian rhythms can participate in lipid, glucose, and cholesterol metabolism and are closely related to metabolism seen in this disease. Circadian clock genes can modulate liver lipid metabolism. Desynchrony of circadian rhythms and the influences imparted by external environmental stimuli can increase morbidity. By contrast, synchronizing circadian rhythms can help to alleviate the metabolic disturbance seen in NAFLD. In this review, we have discussed the current research connections that exist between the circadian clock and the metabolism of NAFLD, and we have specifically focused on the key circadian clock genes, Bmal1, Clock, Rev-Erbs, Rors, Pers, Crys, Nocturnin, and DECs.
