BackgroundAn increasing number of patients with chronic heart failure (CHF) are demanding more convenient and efficient modern health care systems, especially in remote areas away from central cities. Telehealth is receiving increasing attention, which may be useful to patients with CHF. ObjectiveThis study aimed to evaluate the feasibility of a hospital-community-family (HCF)–based telehealth program, which was designed to implement remote hierarchical management in patients with CHF. MethodsThis was a single-arm prospective study in which 70 patients with CHF participated in the HCF-based telehealth program for remote intervention for at least 4 months. The participants were recruited from the clinic and educated on the use of smart health tracking devices and mobile apps to collect and manually upload comprehensive data elements related to the risk of CHF self-care management. They were also instructed on how to use the remote platform and mobile app to send text messages, check notifications, and open video channels. The general practitioners viewed the index of each participant on the mobile app and provided primary care periodically, and cardiologists in the regional central hospital offered remote guidance, if necessary. The assessed outcomes included accomplishments of the program, usability and satisfaction, engagement with the intervention, and changes of heart failure–related health behaviors. ResultsAs of February 2018, a total of 66 individuals, aged 40-79 years, completed the 4-month study. Throughout the study period, 294 electronic medical records were formed on the remote monitoring service platform. In addition, a total of 89 remote consultations and 196 remote ward rounds were conducted. Participants indicated that they were generally satisfied with the intervention for its ease of use and usefulness. More than 91% (21/23) of physicians believed the program was effective, and 87% (20/23) of physicians stated that their professional knowledge could always be refreshed and enhanced through a library hosted on the platform and remote consultation. More than 60% (40/66) of participants showed good adherence to the care plan in the study period, and 79% (52/66) of patients maintained a consistent pattern of reporting and viewing their data over the course of the 4-month follow-up period. Abstract Alzheimer’s disease (AD) is a complex neurodegenerative condition. 5α-epoxyalantolactone (5α-EAL), a eudesmane-type sesquiterpene isolated from the herb of Inula macrophylla, has various pharmacological effects. This work supposed to investigate the improved impact of 5α-EAL on cognitive impairment. 5α-EAL inhibited the generation of nitric oxide (NO) in BV-2 cells stimulated with lipopolysaccharide (LPS) with an EC50 of 6.2 μM. 5α-EAL significantly reduced the production of prostaglandin E2 (PGE2) and tumor necrosis factor-α (TNF-α), while also inhibiting the production of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) proteins. The ability of 5α-EAL to penetrate the blood–brain barrier (BBB) was confirmed via a parallel artificial membrane permeation assay. Scopolamine (SCOP)-induced AD mice model was employed to assess the improved impacts of 5α-EAL on cognitive impairment in vivo. After the mice were pretreated with 5α-EAL (10 and 30 mg/kg per day, i.p.) for 21 days, the behavioral experiments indicated that the administration of the 5α-EAL could alleviate the cognitive and memory impairments. 5α-EAL significantly reduced the AChE activity in the brain of SCOP-induced AD mice. In summary, these findings highlight the beneficial effects of the natural product 5α-EAL as a potential bioactive compound for attenuating cognitive deficits in AD due to its pharmacological profile. Abstract Intervertebral disc degeneration (IDD) is a highly prevalent musculoskeletal disorder affecting millions of adults worldwide, but a poor understanding of its pathogenesis has limited the effectiveness of therapy. In the current study, we integrated untargeted LC/MS metabolomics and magnetic resonance spectroscopy data to investigate metabolic profile alterations during IDD. Combined with validation via a large-cohort analysis, we found excessive lipid droplet accumulation in the nucleus pulposus cells of advanced-stage IDD samples. We also found abnormal palmitic acid (PA) accumulation in IDD nucleus pulposus cells, and PA exposure resulted in lipid droplet accumulation and cell senescence in an endoplasmic reticulum stress-dependent manner. Complementary transcriptome and proteome profiles enabled us to identify solute carrier transporter (SLC) 43A3 involvement in the regulation of the intracellular PA level. SLC43A3 was expressed at low levels and negatively correlated with intracellular lipid content in IDD nucleus pulposus cells. Overexpression of SLC43A3 significantly alleviated PA-induced endoplasmic reticulum stress, lipid droplet accumulation and cell senescence by inhibiting PA uptake. This work provides novel integration analysis-based insight into the metabolic profile alterations in IDD and further reveals new therapeutic targets for IDD treatment.

Abstract Background The global issue of ecological resource scarcity, worsened by climate change, necessitates effective methods to promote resource conservation. One commonly used approach is presenting ecological resource scarcity information. However, the effectiveness of this method remains uncertain, particularly in an unpredictable world. This research aims to examine the role of perceived environmental unpredictability in moderating the impact of ecological resource scarcity information on pro-environmental behavior (PEB). Methods We conducted three studies to test our hypothesis on moderation. Study 1 (N = 256) measured perceived general environmental unpredictability, perceived resource scarcity and daily PEB frequencies in a cross-sectional survey. Study 2 (N = 107) took it a step further by manipulating resource scarcity. Importantly, to increase ecological validity, Study 3 (N = 135) manipulated the information on both ecological resource scarcity and nature-related environmental unpredictability, and measured real water and paper consumption using a newly developed washing-hands paradigm. Results In Study 1, we discovered that perceived resource scarcity positively predicted PEB, but only when individuals perceive the environment as less unpredictable (interaction effect: 95% CI = [-0.09, -0.01], ΔR 2 = 0.018). Furthermore, by manipulating scarcity information, Study 2 revealed that only for individuals with lower levels of environmental unpredictability presenting ecological resource scarcity information could decrease forest resource consumption intention (interaction effect: 95%CI = [-0.025, -0.031], ΔR 2 = .04). Moreover, Study 3 found that the negative effect of water resource scarcity information on actual water and (interaction effect: 95%CI = [3.037, 22.097], η p 2 = .050) paper saving behaviors (interaction effect: 95%CI = [0.021, 0.275], η p 2 = .040), as well as hypothetical forest resource consumption (interaction effect: 95%CI = [-0.053, 0.849], η p 2 = .023) emerged only for people who receiving weaker environmental unpredictability information. Conclusion Across three studies, we provide evidence to support the moderation hypothesis that environmental unpredictability weakens the positive effect of ecological resource scarcity information on PEB, offering important theoretical and practical implications on the optimal use of resource scarcity to enhance PEB.

Abstract Mutations in isocitrate dehydrogenases (IDH) are oncogenic events due to the generation of oncogenic metabolite 2-hydroxyglutarate. However, the role of wild-type IDH in cancer development remains elusive. Here we show that wild-type IDH2 is highly expressed in triple negative breast cancer (TNBC) cells and promotes their proliferation in vitro and tumor growth in vivo. Genetic silencing or pharmacological inhibition of wt-IDH2 causes a significant increase in α-ketoglutarate (α-KG), indicating a suppression of reductive tricarboxylic acid (TCA) cycle. The aberrant accumulation of α-KG due to IDH2 abrogation inhibits mitochondrial ATP synthesis and promotes HIF-1α degradation, leading to suppression of glycolysis. Such metabolic double-hit results in ATP depletion and suppression of tumor growth, and renders TNBC cells more sensitive to doxorubicin treatment. Our study reveals a metabolic property of TNBC cells with active utilization of glutamine via reductive TCA metabolism, and suggests that wild-type IDH2 plays an important role in this metabolic process and could be a potential therapeutic target for TNBC.

Abstract Background Research on the fatty acid metabolism related gene SLC27A2 is currently mainly focused on solid tumors, and its mechanism of action in hematological tumors has not been reported. Method This study aims to explore the pathological and immune mechanisms of the fatty acid metabolism related gene SLC27A2 in hematological tumors and verify its functional role in hematological tumors through cell experiments to improve treatment decisions and clinical outcomes of hematological tumors. Result This study identified the fatty acid metabolism related gene SLC27A2 as a common differentially expressed gene between DLBCL and AML. Immune microenvironment analysis showed that SLC27A2 was significantly positively correlated with T cell CD4 + , T cell CD8 + , endothelial cells, macrophages, and NK cells in DLBCL. In AML, there is a significant negative correlation between SLC27A2 and B cells, T cell CD8 + , and macrophages. SLC27A2 participates in the immune process of hematological tumors through T cell CD8 + and macrophages. The GESA results indicate that high expression of SLC27A2 is mainly involved in the fatty acid pathway, immune pathway, and cell cycle pathway of DLBCL. The low expression of SLC27A2 is mainly involved in the immune pathway of AML. Therefore, SLC27A2 is mainly involved in the pathological mechanisms of hematological tumors through immune pathways, and cell experiments have also confirmed that SLC27A2 is involved in the regulation of DLBCL cells. Conclusion In summary, our research results comprehensively report for the first time the mechanism of action of SLC27A2 in the immune microenvironment of DLBCL and AML, and for the first time verify the cycle and apoptotic effects of the fatty acid related gene SLC27A2 in DLBCL cells through cell experiments. Research can help improve the treatment of AML and DLBCL patients.

Abstract Background Insufficient interfragmentary compression force (IFCF) frequently leads to unstable fixation of osteoporotic lateral tibial plateau fractures (OLTPFs). A combined cancellous lag screw (CCLS) enhances IFCF; however, its effect on OLTPF fixation stability remains unclear. Therefore, we investigated the effect of CCLS on OLTPF stability using locking plate fixation (LPF). Materials and methods Twelve synthetic osteoporotic tibial bones were used to simulate OLTPFs, which were fixed using LPF, LPF-AO cancellous lag screws (LPF-AOCLS), and LPF-CCLS. Subsequently, 10,000 cyclic loadings from 30 to 400 N were performed. The initial axial stiffness (IAS), maximal axial micromotion of the lateral fragment (MAM-LF) measured every 1000 cycles, and failure load after 10,000 cycles were tested. The same three fixations for OLTPF were simulated using finite element analysis (FEA). IFCFs of 0, 225, and 300 N were applied to the LPF, LPF-AOCLS, and LPF-CCLS, respectively, with a 1000-N axial compressive force. The MAM-LF, peak von Mises stress (VMS), peak equivalent elastic strain of the lateral fragment (EES-LF), and nodes of EES-LF > 2% (considered bone destruction) were calculated. Results Biomechanical tests revealed the LPF-AOCLS and LPF-CCLS groups to be superior to the LPF group in terms of the IAS, MAM-LF, and failure load (all p 2% in the LPF were higher than those in the LPF-AOCLS and LPF-CCLS. Conclusion IFCF was shown to enhance the stability of OLTPFs using LPF. Considering overscrewing, CCLS is preferably recommended, although there were no significant differences between CCLS and AOCLS.

Abstract Gastric cancer (GC) presents a formidable global health challenge, and conventional therapies face efficacy limitations. Ubiquitin‐specific protease 7 (USP7) plays pivotal roles in GC development, immune response, and chemo‐resistance, making it a promising target. Various USP7 inhibitors have shown selectivity and efficacy in preclinical studies. However, the mechanistic role of USP7 has not been fully elucidated, and currently, no USP7 inhibitors have been approved for clinical use. In this study, DHPO is identified as a potent USP7 inhibitor for GC treatment through in silico screening. DHPO demonstrates significant anti‐tumor activity in vitro, inhibiting cell viability and clonogenic ability, and preventing tumor migration and invasion. In vivo studies using orthotopic gastric tumor mouse models validate DHPO's efficacy in suppressing tumor growth and metastasis without significant toxicity. Mechanistically, DHPO inhibition triggers ferroptosis, evidenced by mitochondrial alterations, lipid Reactive Oxygen Species (ROS), Malondialdehyde (MDA) accumulation, and iron overload. Further investigations unveil USP7's regulation of Stearoyl‐CoA Desaturase (SCD) through deubiquitination, linking USP7 inhibition to SCD degradation and ferroptosis induction. Overall, this study identifies USP7 as a key player in ferroptosis of GC, elucidates DHPO's inhibitory mechanisms, and highlights its potential for GC treatment by inducing ferroptosis through SCD regulation.

The understanding on the growth mechanism of complex gold nanostructures both experimentally and theoretically can guide their design and fabrication toward various applications. In this work, we report a cysteine-directed overgrowth of penta-twinned nanorod seeds into jagged gold bipyramids with discontinuous stepped {hhk} facets. By monitoring the growth process, we find that {hhk} facets with large k/h values (∼7) are formed first at two ends of the nanorods, followed by the protrusion of the middle section exposing {hhk} facets with smaller indices (k/h ∼ 2–3). Molecular dynamics simulations indicate that the strong adsorption of cysteine molecules on {110} facets is likely responsible for the formation of stepped {hhk} facets, and the stronger adsorption of cysteine molecules on {hhk} facets with smaller k/h compared to that on {hhk} facets with larger k/h is a possible cause of the discontinuity of {hhk} facets at the middle of gold bipyramids. The obtained jagged gold bipyramids display large field enhancement under illumination due to their sharp nanostructures, demonstrating their application potentials in surface-enhanced spectroscopy and catalysis. Abstract The cGAS-STING signaling pathway has emerged as a critical mediator of innate immune responses, playing a crucial role in improving antitumor immunity through immune effector responses. Targeting the cGAS-STING pathway holds promise for overcoming immunosuppressive tumor microenvironments (TME) and promoting effective tumor elimination. However, systemic administration of current STING agonists faces challenges related to low bioavailability and potential adverse effects, thus limiting their clinical applicability. Recently, nanotechnology-based strategies have been developed to modulate TMEs for robust immunotherapeutic responses. The encapsulation and delivery of STING agonists within nanoparticles (STING-NPs) present an attractive avenue for antitumor immunotherapy. This review explores a range of nanoparticles designed to encapsulate STING agonists, highlighting their benefits, including favorable biocompatibility, improved tumor penetration, and efficient intracellular delivery of STING agonists. The review also summarizes the immunomodulatory impacts of STING-NPs on the TME, including enhanced secretion of pro-inflammatory cytokines and chemokines, dendritic cell activation, cytotoxic T cell priming, macrophage re-education, and vasculature normalization. Furthermore, the review offers insights into co-delivered nanoplatforms involving STING agonists alongside antitumor agents such as chemotherapeutic compounds, immune checkpoint inhibitors, antigen peptides, and other immune adjuvants. These platforms demonstrate remarkable versatility in inducing immunogenic responses within the TME, ultimately amplifying the potential for antitumor immunotherapy.

Abstract The pharmacological effects of natural product therapy have received sigificant attention, among which terpenoids such as sesquiterpene lactones stand out due to their biological activity and pharmacological potential as anti-tumor drugs. Inula sesquiterpene lactones are a kind of sesquiterpene lactones extracted from Inula species. They have many pharmacological activities such as anti-inflammation, anti-asthma, anti-tumor, neuroprotective and anti-allergic. In recent years, more and more studies have proved that they are important candidate drugs for the treatment of a variety of cancers because of its good anti-tumor activity. In this paper, the structure, structure–activity relationship, antitumor activities, mechanisms and targets of Inula sesquiterpene lactones reported in recent years were reviewed in order to provide clues for the development of novel anticancer drugs. The day-ahead dispatching of power system depends on the predicted renewable energy output and load power, which brings great challenges to the real-time power balance. This paper focuses on voltage sensitive loads (VSLs) that are widely distributed in space, thus exploring the real-time dispatching potential for VSLs. Firstly, based on the P-V relationship of VSLs clusters, a ZIP model is adopted to establish VSLs’ power controllable model. Meanwhile, considering the potential voltage instability risk during the process of voltage regulation, a linear relationship between voltage safety margin (VSM) index and node power variation was derived. Then, a two-stage linear dispatching model for controllable VSLs clusters was established to meet the needs of online application. In detail, the first stage mainly focuses on evaluating the scheduling potential of VSLs clusters and the second stage determines the optimal real-time response strategy for VSLs clusters. Finally, the effectiveness and feasibility of the proposed method are verified through an improved IEEE-30 node system.

Aspergillus fungi are renowned for producing a diverse range of natural products with promising biological activities. These include lovastatin, itaconic acid, terrin, and geodin, known for their cholesterol-regulating, anti-inflammatory, antitumor, and antibiotic properties. In our current study, we isolated three dimeric nitrophenyl trans-epoxyamides (1–3), along with fifteen known compounds (4–18), from the culture of Aspergillus terreus MCCC M28183, a deep-sea-derived fungus. The structures of compounds 1–3 were elucidated using a combination of NMR, MS, NMR calculation, and ECD calculation. Compound 1 exhibited moderate inhibitory activity against human gastric cancer cells MKN28, while compound 7 showed similar activity against MGC803 cells, with both showing IC50 values below 10 μM. Furthermore, compound 16 exhibited moderate potency against Vibrio parahaemolyticus ATCC 17802, with a minimum inhibitory concentration (MIC) value of 7.8 μg/mL. This promising research suggests potential avenues for developing new pharmaceuticals, particularly in targeting specific cancer cell lines and combating bacterial infections, leveraging the unique properties of these Aspergillus-derived compounds.

Background: The functional movement screen (FMS) has been used to identify deficiencies in neuromuscular capabilities and balance among athletes. However, its effectiveness in detecting movement anomalies within the population afflicted by knee osteoarthritis (KOA), particularly through the application of a family-oriented objective assessment technique, remains unexplored. The objective of this study is to investigate the sensitivity of the FMS and daily activities in identifying kinematic abnormalities in KOA people employing a markerless motion capture system.Methods: A total of 45 persons, presenting various Kellgren–Lawrence grades of KOA, along with 15 healthy controls, completed five tasks of the FMS (deep squat, hurdle step, and in-line lunge) and daily activities (walking and sit-to-stand), which were recorded using the markerless motion capture system. The kinematic waveforms and discrete parameters were subjected to comparative analysis.Results: Notably, the FMS exhibited greater sensitivity compared to daily activities, with knee flexion, trunk sagittal, and trunk frontal angles during in-line lunge emerging as the most responsive indicators.Conclusion: The knee flexion, trunk sagittal, and trunk frontal angles during in-line lunge assessed via the markerless motion capture technique hold promise as potential indicators for the objective assessment of KOA.

Amorphous structures can be induced by cold deformation in the NiTi shape-memory alloys, which transform into nanocrystalline structures after annealing. In this study, the effect of cold-drawn-induced amorphous phase on the microstructure and pseudoelasticity properties of nanocrystalline NiTi alloys was investigated. Ni-49.2 at.%Ti alloy samples were subjected to different cold-drawn deformations. Transmission electron microscopy was used to investigate the microstructure of the samples. Three types of microstructures: twin-related lamellae, nanocrystalline, and amorphous, were observed in the cold-drawn samples. The amorphous volume fractions of the samples were determined using differential scanning calorimetry. The results suggested that the amorphous volume fraction increased with increasing cold-drawn deformation. The pseudoelasticity properties of the samples with different cold-drawn deformations were investigated using tensile tests. Notably, 59.05% cold-drawn deformation, which induced approximately 39.1% volume fraction of the amorphous phase, was the turning point in the performance of the NiTi alloy. The stress hysteresis and upper plateau stress increased with increasing cold-drawn deformation when the cold-drawn deformation was lesser than 59.04%. Furthermore, the difference in stress hysteresis and upper plateau stress was not significant when the cold-drawn deformation was greater than 59.04% because a complete nanocrystalline microstructure could be obtained after annealing when the volume fraction of the amorphous phase reached 39.1% in the NiTi alloy.

Abstract The evaluation of the capability of network-based systems of systems has replaced the simple method that considers return on investment, becoming a new paradigm for planning national defence capabilities. However, the dual uncertainty of the key system attributes of scenes and weapons has brought great challenges for decision-making. Based on this, we developed a multiobjective optimization model with multiple stages and scenarios under uncertainty to determine plans. In this study, we consider planning risk and planning cost as the two objectives. To solve this problem, we propose a hybrid solution for a network-based optimization method integrated with fuzzy set theory. The network-based optimization method combines the NSGA-II-DE and complex network theory. We use the characteristics of the network to evaluate the capabilities of the WSoS, and the NSGA-II-DE is used to generate a development plan and finally output a set of Pareto optimal solutions. We use fuzzy sets to determine the fuzzy membership of each plan on the Pareto front and determine a satisfactory solution. Finally, we conduct simulation experiments to verify the rationality of the methods proposed in this article. The results can provide a set of efficient solutions for military planners, helping generate a variety of planning solutions and trade-offs according to their preferences. Graphical Abstract

Natural vegetation protects, maintains, and improves the environment through its ecological functions and is, thus, an important component of Earth’s ecosystems. The distribution of natural vegetation and its corresponding ecological roles vary with the topographic gradient. Understanding this role is essential for effective ecosystem management and conservation efforts. This study analyzes vegetation composition across altitude gradients and the spatiotemporal evolution of water conservation, soil conservation, and carbon storage in the southern hill and mountain belt of China. We then explored the drivers of the ecological functions of vegetation at different altitude gradients. The results showed that water conservation increased by 108.56%, soil conservation increased by 97.04%, and carbon storage increased only slightly. The ecological functions of vegetation varied across altitude gradients, with the 500–800 m gradient exhibiting markedly higher ecological functions than the other gradients. The effect of precipitation on soil conservation increases with altitude. In addition, at higher altitudes, evergreen coniferous forests had a greater effect on carbon storage. Based on the results, we propose vegetation management measures for different altitudes. This study provides a reference for decision-makers to develop and adjust ecological restoration programs in mountainous areas for the improvement of the local ecological environment.

Ecological engineering (EE) has a profound impact on land-use dynamics, leading to alterations in ecosystem services (ESs). However, an appropriate EE implementation intensity that can balance the tradeoffs associated with altered ESs well has always been a concern for researchers and policymakers. In this study, we set the transition probability of farmland, bare land, and desertification land to forest and natural shrub, with 2010–2020 as the natural implementation scenario, as 10% for the low-intensity implementation scenario (LIS), 30% for the medium-intensity scenario, and 50% for the high-intensity scenario. The patch-generating land-use simulation (PLUS) model was used to project land-use patterns and the Integrated Valuation of Ecosystem Service and Tradeoffs (InVEST) model was used to simulate changes in the quality of ESs under four EE implementation intensities in 2030. We then performed a quantitative tradeoff analysis on the dominant ESs under four scenarios and used the production possibility frontier (PPF) curve to identify the optimal EE implementation intensity scenario. Our results indicated that an increase in EE implementation intensity would lead to an increase in soil retention, water purification, habitat quality, and carbon storage, but also to a decrease in water yield, aggravating the tradeoffs between water yield and other ESs. In all EE implementation intensity scenarios, the LIS had the lowest tradeoff intensity index and balanced ESs well, and thus was the optimal EE implementation scenario in Qinghai province. Our results provide knowledge to help decision makers select the appropriate EE intensity to maintain sustainable development. The integrated methodology can also be applied in other conservation regions to carry out practical land management.

The aim of our study is to understand the patterns of variation in water use efficiency (WUE) in coniferous and broad-leaved mixed forest ecosystems across multiple scales and to identify its main controlling factors. We employ the eddy covariance method to gather data from 2017, 2018, and 2020, which we use to calculate the gross primary productivity and evapotranspiration of these forests in East China and to determine WUE at the ecosystem level. The mean daily variation in WUE ranges from 4.84 to 7.88 gC kg−1 H2O, with a mean value of 6.12 gC kg−1 H2O. We use ridge regression analysis to ascertain the independent effect of environmental factors on WUE variation. We find that WUE responds differently to environmental factors at different time scales. In mixed conifer ecosystems, temperature and relative humidity emerge as the most significant environmental factors influencing WUE variability. Especially at the seasonal scale, temperature and relative humidity can explain more than 51% of the WUE variation. Our results underscore the varied effects of environmental factors on WUE variation across different time scales and aid in predicting the response of WUE to climate change in coniferous and broad-leaved mixed forest ecosystems.

Understanding the effects of various forest management systems, including logging practices, on soil properties is essential for implementing sustainable management strategies. In Malaysia, two types of forest management systems were commonly used: Malayan Uniform System (MUS) and Selective Management System (SMS) practices. However, their effects on soil quality remained elusive, especially after decades of recovery. To address this need, we selected three plots for the MUS and SMS in Johor, Malaysia, to assess soil properties in logged-over forest plots. All the plots were natural forest reserves. Soil properties analyzed include soil acidity, electrical conductivity, cation exchange capacity, selected nutrient contents, and soil compaction. Generally, the results of the study indicate that forests logged using the SMS exhibit superior soil quality compared to those logged using the MUS according to several key soil properties. Specifically, significantly higher cation exchange capacity, potassium content, calcium content, and magnesium content with lower soil compaction was observed in the SMS when compared to MUS plots.
