The application of warm mix asphalt (WMA) technology in polymer-modified asphalt becomes increasingly widespread, but it lacks thorough investigation on the interaction mechanism between warm mix additives and polymers. Accordingly, two new warm mix additives, an organic viscosity-reducing modifier (A) and a surfactant-based modifier (C), were selected to prepare the warm mix SBS-modified asphalt. The basic properties, viscosity-temperature characteristics, ageing resistance and rheological properties of modified asphalt were tested. At last, the modification mechanism was elucidated through a series of microscopic tests. The results indicated that A could more effectively enhance the construction workability of SBS-modified asphalt, while C had little effect on its basic properties. Both new modifiers, especially A, reduced the high-temperature property of asphalt, with partial recovery after short-term ageing. The microstructure test results indicated that the two modifiers were physically modified with SBS-modified asphalt. Both additives improved the dispersion uniformity of SBS particles in asphalt, and the microscopic morphology parameters obtained from quantitative analysis reflect the macroscopic properties of asphalt. Moreover, the light components and rubber powder of A improved the flexibility of asphalt, and C enhanced its low-temperature performance, which demonstrated that these two modifiers were more suitable for asphalt pavements in cold regions. Sandstone has poor mechanical properties. To facilitate the application of sandstone into asphalt mixtures, sandstone was treated by immersion in sodium silicate solution, and the dynamic modulus after reinforcement was used as a criterion. The results showed that the mechanical properties of the sandstone aggregate treated with sodium silicate were improved, and the dynamic modulus was increased by 18.2%, which will help to reduce rutting. The dynamic modulus and phase angle can be effectively predicted over a wide frequency range using the sigma function and the Kramers–Kronig relationship. Sandstone asphalt mixtures basically conform to linear viscoelasticity, but the phase angle changes are more complicated at high temperatures and do not vary monotonically with frequency. By calculating the rutting coefficient, fatigue coefficient, and DSRFn parameters for performance prediction, it was found that an increase in dynamic modulus resulted in a significant increase in the rutting coefficient but a decrease in the cracking resistance. Abstract Background An accurate diagnosis for high-suspicion nodules based on the 2015 American Thyroid Association (ATA) guidelines would reduce unnecessary invasive examinations. Elastography is a useful tool for discriminating benign and malignant thyroid nodules. The aim of this study is to investigate the diagnostic efficiency of elastography for high-suspicion thyroid nodules based on the 2015 ATA guidelines in the Chinese population. Methods Thyroid nodules with high-suspicion characteristics based on the 2015 ATA guidelines were subjected to conventional ultrasound (US) and ultrasound strain elastography (USE) examinations at 12 hospitals from 4 geographic regions across China. Cytology/histology of thyroid nodules was used as a reference method. Receiver operating characteristic (ROC) curves were plotted to evaluate the diagnostic performance of the elasticity score (ES) and strain ratio (SR). Logistic regression analysis was used to determine the predictors of malignancy. Results Overall, a total of 1445 thyroid nodules (834 malignant, 611 benign) from 12 centers were included in the final analysis. The areas under the curve of the ES and SR were 0.828 and 0.732, respectively. The sensitivity, specificity, accuracy, positive predictive value (PPV) and negative predictive value (NPV) of the ES were 92.4, 60.7, 79.0, 76.3 and 85.5%, respectively, and those of the SR were 81.1, 50.1, 68.9, 65.9 and 67.9%, respectively. The combination of the Thyroid Imaging Reporting and Data System (TI-RADS) and ES led to a significant increase in the sensitivity and NPV (97.1 and 91.9%, respectively) compared with the TI-RADS alone. Logistic regression analysis showed that microcalcifications (OR = 5.290), taller than wide (OR = 12.710), irregular margins (OR = 10.117), extrathyroidal extension (ETE; OR = 6.412), the ES (OR = 3.741) and the SR (OR = 1.083) were independent predictors of malignant thyroid nodules. Aims CD4+ T cells are activated during inflammatory dilated cardiomyopathy (iDCM) development to induce immunogenic responses that damage the myocardium. Low-intensity pulsed ultrasound (LIPUS), a novel physiotherapy for cardiovascular diseases, has recently been shown to modulate inflammatory responses. However, its efficacy in iDCM remains unknown. Here, we investigated whether LIPUS could improve the severity of iDCM by orchestrating immune responses and explored its therapeutic mechanisms. Methods and results In iDCM mice, LIPUS treatment reduced cardiac remodelling and dysfunction. Additionally, CD4+ T-cell inflammatory responses were suppressed. LIPUS increased Treg cells while decreasing Th17 cells. LIPUS mechanically stimulates endothelial cells, resulting in increased secretion of extracellular vesicles (EVs), which are taken up by CD4+ T cells and alter their differentiation and metabolic patterns. Moreover, EVs selectively loaded with microRNA (miR)-99a are responsible for the therapeutic effects of LIPUS. The hnRNPA2B1 translocation from the nucleus to the cytoplasm and binding to caveolin-1 and miR-99a confirmed the upstream mechanism of miR-99a transport. This complex is loaded into EVs and taken up by CD4+ T cells, which further suppress mTOR and TRIB2 expression to modulate cellular differentiation. Conclusion Our findings revealed that LIPUS uses an EVs-dependent molecular mechanism to protect against iDCM progression. Therefore, LIPUS is a promising new treatment option for iDCM. The two-step thermal stress calculation method (TTSCM) is commonly used to predict the cracking temperature of asphalt mixture. The aim of this study is to improve TTSCM's mathematical model so as to enhance its prediction accuracy. First, this study evaluated the errors of predicted cracking temperatures of original TTSCM for AC-16 and AC-25 asphalt mixtures by thermal stress-restrained specimen test (TSRST). Then, an improved method called the extended creep compliance test (ECCT) was developed to modify the TTSCM. The test results show that the cracking predictions of the original TTSCM are not always accurate. Particularly for AC-16 asphalt mixture, the predicted cracking temperature is 2.9 °C (−10.6%) higher than the measured value by the TSRST. The ECCT method has been proven to be an effective way to enhance the prediction accuracy of the TTSCM. The predicted cracking temperatures modified by the ECCT method for both asphalt mixtures are relatively accurate, having an error within ±2%. The ECCT method changed the calculated thermal stress values at different temperatures of the TTSCM; however, they still conformed to a basic changing trend with respect to the initial temperature and cooling rate. To evaluate the inter-observer variability and the intra-observer repeatability of pulmonary transit time (PTT) measurement using contrast-enhanced ultrasound (CEUS) in healthy rabbits, and assess the effects of dilution concentration of ultrasound contrast agents (UCAs) on PTT. Thirteen healthy rabbits were selected, and five concentrations UCAs of 1:200, 1:100, 1:50, 1:10, and 1:1 were injected into the right ear vein. Five digital loops were obtained from the apical 4-chamber view. Four sonographers obtained PTT by plotting the TIC of right atrium (RA) and left atrium (LA) at two time points (T1 and T2). The frame counts of the first appearance of UCAs in RA and LA had excellent inter-observer agreement, with intra-class correlations (ICC) of 0.996, 0.988, respectively. The agreement of PTT among four observers was all good at five different concentrations, with an ICC of 0.758–0.873. The reproducibility of PTT obtained by four observers at T1 and T2 was performed well, with ICC of 0.888–0.961. The median inter-observer variability across 13 rabbits was 6.5% and the median variability within 14 days for 4 observers was 1.9%, 1.7%, 2.2%, 1.9%, respectively; The PTT of 13 healthy rabbits is 1.01 ± 0.18 second. The difference of PTT between five concentrations is statistically significant. The PTT obtained by a concentration of 1:200 and 1:100 were higher than that of 1:1, while there were no significantly differences in PTT of a concentration of 1:1, 1:10, and 1:50. PTT measured by CEUS in rabbits is feasible, with excellent inter-observer and intra-observer reliability and reproducibility, and dilution concentration of UCAs influences PTT results. To know the mixing flowability in theory and quantitatively evaluate the mixing workability of the asphalt mixture, a variable speed mixing (VSM) test device was developed in our laboratory. Different asphalt mixtures are tested by recording the mixing power under different mixing speeds (20∼50 r·min-1) and different mixing temperatures (130∼180 °C) to analyze the mixing flowability. Afterwards, the mixing workability index was defined from rheological parameters by establishing the mixing flow model. The experimental results indicate that the mixing flowability of asphalt mixtures obeys the linear Bingham's visco-plastic model, which conducts two mixing rheological parameters including the mixing plastic limit and the mixing viscosity by the intercept and slope of the flow straight respectively. When the mixing temperature increases, the mixing viscosity decreases and the workability index increases, but the mixing plastic limit is not affected. With the increase of the nominal maximum particle size of asphalt mixtures, the mixing plastic limit increases and the workability index decreases, but the mixing viscosity is not affected. The asphalt becomes more viscous as the asphalt content increase, which leads to worse workability. The composition of aggregate determines the mixing plastic limit, and the viscosity of asphalt mortar determines the mixing viscosity. [ABSTRACT FROM AUTHOR]

In the process of intelligent compaction of roadbeds, the water content of the roadbed is one of the important influencing factors of compaction quality. In order to analyze the effect of water content on the compaction quality of roadbeds, this paper is developed by secondary development of Abaqus finite element numerical simulation software. At the same time, the artificial viscous boundary was set to eliminate the influence of boundary conditions on the results in the finite element modeling process, so that the numerical simulation can be refined to model. On this basis, the dynamic response analysis of the roadbed compaction process is performed on the finite element numerical simulation results. This paper established the correlation between compaction degree and intelligent compaction index CMV (Compaction Meter Value) and then analyzed the effect of water content on the compaction quality for the roadbed. The results of this paper show that the amplitude of the vertical acceleration is almost independent of the moisture content, and the vertical displacement mainly occurs in the static compaction stage. The vertical displacement changes sharply in the first 0.5 s when the vibrating wheel is in contact with the roadbed. The main stage of roadbed compaction quality increase is before the end of the first compaction. At the end of the first compaction, the roadbed compaction degree increased rapidly from 80% to 91.68%, 95.34% and 97.41%, respectively. With the increase in water content, the CMV gradually increased. At the end of the second compaction, CMV increased slightly compared with that at the end of the first compaction and stabilized at the end of the second compaction. The water content of the roadbed should be considered to be set slightly higher than the optimal water content of the roadbed by about 1% during the construction of the roadbed within the assumptions of this paper. To evaluate the water accumulation status at superelevation transitions of the freeway and establish a correlation with wet friction, laboratory tests and theoretical calculation approaches were carried out during water-film formation and sheet flow stages. The influencing factors on water depth and the water depth dependency of friction were analyzed using the self-developed multi-directional adjustment tester. The results indicate that the ponding is severe at flat road sections, adjusting alignment and improving drainage capacity are the key remedies. There is a good correlation between decay characteristics of dynamic friction and the water depth. The wet grip can be explained by the tribology principle and lubrication regimes, which is decomposed into the distinct contributions of tire, pavement, and water. Besides, the water depth threshold was determined. Finally, a prediction model of water depth at superelevation transitions was established based on the hydromechanics analysis. Aims Nicotine, a major component of tobacco, is an important factor contributing to atherosclerosis. However, the molecular mechanisms underlying the link between nicotine and atherosclerosis are unclear. As extracellular vesicles (EVs) are involved in intercellular communication in atherosclerosis, we investigated whether their influence on arterial pathophysiology under nicotine stimulation. Methods and results EVs from the serum of smokers (smoker-EVs) were significantly increased and exacerbated endothelial inflammation, as well as apoptosis according to functional studies. Meanwhile, inhibition of EVs blunted the nicotine-induced atherosclerosis progression, and injection of nicotine-induced EVs promoted atherosclerosis progression in ApoE–/– mice. Furthermore, quantitative reverse transcription-polymerase chain reaction analysis revealed a remarkable increase in miR-155 levels in smoker-EVs, which was correlated with carotid plaque formation in patients measured by ultrasound imaging. Moreover, CD14 levels were significantly increased in EVs from smokers (representing EVs derived from monocytes), indicating that monocytes are an important source of smoker-EVs. DNA methylation and the transcription factor HIF1α may contribute to increased miR-155 levels in monocytes, as assessed with bisulfite conversion sequencing and chromatin immunoprecipitation. Mechanistically, EVs encapsulated miR-155 induced endothelial cell dysfunction by directedly targeting BCL2, MCL1, TIMP3, BCL6, and activating NF-κB pathway, as verified in a series of molecular and biological experiments. Injecting EVs from nicotine-stimulated monocytes promoted plaque formation and triggered vascular endothelial injury in ApoE–/– mice, whereas inhibition of miR-155 weakened this effect. Conclusion Our findings revealed an EV-dependent mechanism of nicotine-aggravated atherosclerosis. Accordingly, we propose an EV-based intervention strategy for atherosclerosis management. Background/Aims: Intracellular calcium concentration ([Ca2+]i) homeostasis, an initial factor of cardiac hypertrophy, is regulated by the calcium-sensing receptor (CaSR) and is associated with the formation of autolysosomes. The aim of this study was to investigate the role of Calhex231, a CaSR inhibitor, on the hypertrophic response via autophagy modulation. Methods: Cardiac hypertrophy was induced by transverse aortic constriction (TAC) in 40 male Wistar rats, while 10 rats underwent a sham operation and served as controls. Cardiac function was monitored by transthoracic echocardiography, and the hypertrophy index was calculated. Cardiac tissue was stained with hematoxylin and eosin (H&E) or Masson's trichrome reagent and examined by transmission electron microscopy. An angiotensin II (Ang II)-induced cardiomyocyte hypertrophy model was established and used to test the involvement of active molecules. Intracellular calcium concentration ([Ca2+]i) was determined by the introduction of Fluo-4/AM dye followed by confocal microscopy. The expression of various active proteins was analyzed by western blot. Results: The rats with TAC-induced hypertrophy had an increased heart size, ratio of heart weight to body weight, myocardial fibrosis, and CaSR and autophagy levels, which were suppressed by Calhex231. Experimental results using Ang II-induced hypertrophic cardiomyocytes confirmed that Calhex231 suppressed CaSR expression and downregulated autophagy by inhibiting the Ca2+/calmodulin-dependent-protein kinase-kinase-ß (CaMKKß)- AMP-activated protein kinase (AMPK)-mammalian target of rapamycin (mTOR) pathway to ameliorate cardiomyocyte hypertrophy. Conclusions: Calhex231 ameliorates myocardial hypertrophy induced by pressure-overload or Ang II via inhibiting CaSR expression and autophagy.
