Abstract Background Radical resection plus lymph node dissection is a common treatment for patients with T1-3N0M0 non-small cell lung cancer (NSCLC). Few models predicted the survival outcomes of these patients. This study aimed to developed a nomogram for predicting their overall survival (OS). Materials and methods This study involved 3002 patients with T1-3N0M0 NSCLC after curative resection between January 1999 and October 2013. 1525 Patients from Sun Yat-sen University Cancer Center were randomly allocated to training cohort and internal validation cohort in a ratio of 7:3. 1477 patients from ten institutions were recruited as external validation cohort. A nomogram was constructed based on the training cohort and validated by internal and external validation cohort to predict the OS of these patients. The accuracy and practicability were tested by Harrell's C-indexes, calibration plots and decision curve analyses (DCA). Results Age, sex, histological classification, pathological T stage, and HI standard were independent factors for OS and were included in our nomogram. The C-index of the nomogram for OS estimates were 0.671 (95% CI, 0.637–0.705),0.632 (95% CI, 0.581–0.683), and 0.645 (95% CI, 0.617–0.673) in the training cohorts, internal validation cohorts, and external validation cohort, respectively. The calibration plots and DCA for predictions of OS were in excellent agreement. An online version of the nomogram was built for convenient clinical practice. Conclusions Our nomogram can predict the OS of patients with T1-3N0M0 NSCLC after curative resection. The online version of our nomogram offer opportunities for fast personalized risk stratification and prognosis prediction in clinical practice.

Abstract Background There is an unmet need to identify novel predictive biomarkers that enable more accurate identification of individuals who can benefit from immune checkpoint inhibitor (ICI) therapy. The US FDA recently approved tumor mutational burden (TMB) score of ≥ 10 mut/Mb as a threshold for pembrolizumab treatment of solid tumors. Our study aimed to test the hypothesis that specific gene mutation signature may predict the efficacy of ICI therapy more precisely than high TMB (≥ 10). Methods We selected 20 candidate genes that may predict for the efficacy of ICI therapy by the analysis of data from a published cohort of 350 advanced non-small cell lung cancer (NSCLC) patients. Then, we compared the influences of various gene mutation signatures on the efficacy of ICI treatment. They were also compared with PD-L1 and TMB. The Kaplan-Meier method was utilized to evaluate the prognosis univariates, while selected univariates were adopted to develop a systematic nomogram. Results A high mutation signature, where three or more of the 20 selected genes were mutated, was associated with the significant benefits of ICI therapy. Specifically, patients with high mutation signature were confirmed to have better prognosis for ICI treatment, compared with those with wild type (the median PFS: 7.17 vs. 2.90 months, p = 0.0004, HR = 0.47 (95% [CI]:0.32–0.68); the median OS: unreached vs. 9 months, p = 1.8E-8, HR = 0.17 (95% [CI]:0.11–0.25)). Vehicle detection based on unmanned aerial vehicle (UAV) aerial images plays a significant role in areas such as traffic monitoring and management, disaster relief, and more, garnering extensive attention from researchers in recent years. However, datasets acquired from UAV platforms inevitably suffer from issues such as imbalanced class distribution, severe background interference, numerous small objects, and significant target scale variance, presenting substantial challenges to practical vehicle detection applications based on this platform. Addressing these challenges, this paper proposes an object detection model grounded in a background suppression pyramid network and multi-scale task adaptive decoupled head. Firstly, the model implements a long-tail feature resampling algorithm (LFRA) to solve the problem of imbalanced class distribution in the dataset. Next, a background suppression pyramid network (BSPN) is integrated into the Neck segment of the model. This network not only reduces the interference of redundant background information but also skillfully extracts features of small target vehicles, enhancing the ability of the model to detect small objects. Lastly, a multi-scale task adaptive decoupled head (MTAD) with varied receptive fields is introduced, enhancing detection accuracy by leveraging multi-scale features and adaptively generating relevant features for classification and detection. Experimental results indicate that the proposed model achieves state-of-the-art performance on lightweight object detection networks. Compared to the baseline model PP-YOLOE-s, our model improves the AP50:95 on the VisDrone-Vehicle dataset by 1.9%.

The objective of this study was to investigate the possible association between the single nucleotide polymorphism (SNP), rs35569394, of the vascular endothelial growth factor gene (VEGF) and the risk of esophageal cancer (EC) in the Han Chinese population. A total of 290 EC subjects and 322 ethnically matched unrelated healthy controls free from the esophageal disease were studied. Genomic DNA was isolated from peripheral blood by salting out. Genotyping of VEGF rs35569394 polymorphism was carried out via polymerase chain reaction followed by agarose gel electrophoresis. The results showed that the distribution of genotypes was significantly different across the gender groups (p=0.032) and clinical stages (p=0.034). VEGF rs35569394 was associated with EC risk (p= 0.012, OR=1.34). A gender analysis break-down showed that rs35569394-D allele frequency was significantly higher in females than in the controls (p=0.0004, OR=1.81). Moreover, significant associations were also found in females under the dominant model (II versus ID+DD: χ2=8.18, p=0.003, OR=2.12) and the recessive model (II+ID versus DD: χ2=8.25, p=0.004, OR=2.39). Additionally, we found that the genotype, rs35569394-DD, was associated with a complete response + partial response to chemotherapy when compared with rs35569394-II (χ2=4.67, p=0.030, OR=0.47). In conclusion, our case-control study showed that the VEGF rs35569394 was significantly associated with the clinical stages and the increased risk of EC in Han Chinese females. In addition, the genotype rs35569394-DD showed a better response to chemotherapy.

Subglacial lake exploration is of great interest to the science community. RECoverable Autonomous Sonde (RECAS) provides an exploration tool to measure and sample subglacial lake environments while the subglacial lake remains isolated from the glacier surface and atmosphere. This paper presents an electronic control system design of 200 m prototype of RECAS. The proposed electronic control system consists of a surface system, a downhole control system, and a power transfer and communication system. The downhole control system is the core element of RECAS, and is responsible for sonde status monitoring, sonde motion control, subglacial water sampling and in situ analysis. A custom RS485 temperature sensor was developed to cater for the limited size and depth requirements of the system. We adopted a humidity-based measurement to monitor for a housing leak. This condition is because standard leak detection monitoring of water conductivity may be inapplicable to pure ice in Antarctica. A water sampler control board was designed to control the samplers and monitor the on/off state. A high-definition camera system with built-in storage and self-heating ability was designed to perform the video recording in the subglacial lake. The proposed electronic control system is proven effective after a series of tests.

Drilling and sampling are the most direct and effective methods available to study Antarctic subglacial lakes. Based on the Philberth probe, a Recoverable Autonomous Sonde (RECAS) allows for in situ lake water measurement and sampling, through the addition of an upper thermal tip and a cable recoiling mechanism. RECAS-200, a prototype of RECAS, has a drilling depth of 200 m, a surface supply voltage of 800 VAC and a downhole power of ~9.6 kW during drilling. In this study, a heating control system for RECAS-200 was designed. The system avoids the need for high-power step-down converters, by separating heating power from control power, thereby reducing the overall weight of the probe and avoiding the need to increase cable diameter. We also introduce a self-developed, small, solid-state, 800 VAC power regulator and a fuzzy PID temperature control algorithm. Their purpose was to manage the power adjustment of each heating element and to provide closed-loop temperature control of certain heating elements which can easily burn out due to overheating. Test results indicated that the proposed RECAS-200 heating control system met all our design specifications and could be easily assembled into the RECAS-200 probe.

Monitoring the tension in cables is significant in some ice drill and deepwater applications. Take our RECoverable Autonomous Sonde (RECAS) for example. It is able to melt a hole to the bottom of ice sheet and is able to move upwards. A winch is installed inside RECAS to release and recover the cable, whose tension needs to be monitored in real time in order to control the behavior of the winch. The high pressure of deep water and limited installation space pose great challenges in sensor development. In this paper, two editions of newly designed deepwater tension sensors are proposed. The first edition is based on a fresh hydraulic load module that operates in high pressure environment and the second edition tension, which aims to improve the accuracy, applies a newly designed watertight load module. Detailed force transmission and characteristic analysis of the sensors are carried out. The sensors have got through a series of experiments, including calibration experiments, pressure experiments and field experiments. The resultant accuracy of the second edition sensor, which has a better performance, is over 2% under the measuring range of 1000 kg and the dimension of the final sensor is as compact as 150 mm × 137 mm × 86 mm.

BackgroundLung cancer is the third most frequently diagnosed cancer in the world, with lung adenocarcinoma (LUAD) as the most common pathological type. But studies on the predictive effect of a single gene on LUAD are limited. We aimed to discover new predictive markers for LUAD.MethodsDifferentially high-expressed genes at each stage were obtained from the TCGA and GTEx databases. The functions of these genes were investigated through GO enrichment and KEGG pathway analyses. Then, the key genes were selected by applying whole gene overall survival time. The expression of the key gene was studied in LUAD, and survival analysis was performed using Kaplan-Meier mapper, followed by univariate and multifactorial COX analysis. Finally, the gene expression and its prognostic significance in the pan-cancer were examined.ResultsA total of 10,106 DEGs were obtained from the two datasets. The top 266 differentially upregulated genes intersected with the top 1,497 overall survival-related genes, and 87 key genes were identified. High-expressed HMMR was associated with a poor prognosis of LUAD. Univariate and multifactorial Cox analysis showed that HMMR was an independent prognostic factor for LUAD patients. A high HMMR expression was strongly associated with the overall survival (OS) and disease-specific survival (DSS) in 11 cancer types and with poorer OS, DSS, and PFI in 10 cancer types.ConclusionHMMR may be an independent prognostic indicator and an important biomarker in diagnosing and predicting the survival of LUAD patients. Also, HMMR may be a key predictor of a variety of cancers.

Epithelial-mesenchymal transition (EMT) process, which is regulated by genes of inducible factors and transcription factor family of signaling pathways, transforms epithelial cells into mesenchymal cells and is involved in tumor invasion and progression and increases tumor tolerance to clinical interventions. This study constructed a multigene marker for lung predicting the prognosis of lung adenocarcinoma (LUAD) patients by bioinformatic analysis based on EMT-related genes. Gene sets associated with EMT were downloaded from the EMT-gene database, and RNA-seq of LUAD and clinical information of patients were downloaded from the TCGA database. Differentially expressed genes were screened by difference analysis. Survival analysis was performed to identify genes associated with LUAD prognosis, and overlapping genes were taken for all the three. Prognosis-related genes were further determined by combining LASSO regression analysis for establishing a prediction signature, and the risk score equation for the prognostic model was established using multifactorial COX regression analysis to construct a survival prognostic model. The model accuracy was evaluated using subject working characteristic curves. According to the median value of risk score, samples were divided into a high-risk group and low-risk group to observe the correlation with the clinicopathological characteristics of patients. Combined with the results of one-way COX regression analysis, HGF, PTX3, and S100P were considered as independent predictors of LUAD prognosis. In lung cancer tissues, HGF and PTX3 expression was downregulated and S100P expression was upregulated. Kaplan-Meier, COX regression analysis showed that HGF, PTX3, and S100P were prognostic independent predictors of LUAD, and high expressions of all the three were all significantly associated with immune cell infiltration. The present study provided potential prognostic predictive biological markers for LUAD patients, and confirmed EMT as a key mechanism in LUAD progression.

One of the main reasons for the lack of seismic capacity of existing historical masonry buildings in China is that the strength of the lime-clay mortar used in these buildings is insufficient. In this paper, a kind of modified oyster shell ash mortar, which is similar to the ancient mortar used in historical buildings in terms of material composition, is proposed to partially replace the original mortar, so as to strengthening the brick masonry wall. In order to evaluate the strengthening effect of this method, shear tests were carried out on specimens strengthened with modified oyster shell ash mortar. The test results show that the proposed strengthening method has significant effect on improving the shear strength of brick masonry wall with low strength mortar. Based on the test results, the calculation method of shear strength for brick masonry strengthened with partially replaced modified oyster shell mortar is proposed. The calculated results are in good agreement with the experimental ones.

Masonry bricks were widely used in construction of the walls in most of Chinese historical buildings. The low strength of lime–clay mortar used in existing historical brick masonry walls has usually led to poor performance such as cracking and collapse during earthquakes. As the composition of modified oyster shell ash mortar (MOSA mortar) with higher strength is similar to that of lime–clay mortar, it can be used to partially replace original lime–clay mortar for historical brick masonry buildings in order to improve their seismic performance. Previous research has proven that this strengthening method for brick masonry is effective in improving shear strength. In this paper, we present further experimental research regarding the compressive behaviors of brick masonry strengthened by replacing mortar with a MOSA mortar. The test results showed that the compressive strength of brick masonry specimens strengthened by the proposed method meets the design requirements. The formula for calculating compressive strength for brick masonry strengthened by replacing mortar was obtained by fitting the test results. The calculated values were consistent with the tested ones. In addition, the stress–strain relationship of tested specimens under axial compression was simulated using the parabolic model.

It is the key to the concrete repair and strengthening construction that the base concrete has a certain quality and strength. Based on the strengthening mechanism of sodium silicate, the sodium silicate was modified to develop a kind of compound strengthening liquid. Through the contrast test results of concrete impregnation. The strength of C5 concrete before and after strengthening is increased by 42.5%, while the strength of C10 concrete is increased by 21.4%, and the strength of C15 concrete is increased by 10%.

The quality of the the existing concrete surface in its repairing and strengthening is the key factor affecting the together working performance. The effective method to solve this problem is to use permeable crystal waterproof material to strengthen the existing concrete surface. In view of the existing old concrete which lacks free Ca2+ in the interior, the method of compensating calcium ion strengthening is proposed based on the action mechanism of permeable crystalline waterproof material. On the basis of DPS, calcium ion compensating agent Ca (OH)2, Na2SiO3 (sodium silicate) and Na2CO3 (potassium carbonate) solutes are added to prepare composite reinforcement solution for impregnation strengthening of concrete. Scanning electron microscopy (SEM) was used to observe the microstructure of the concrete before and after strengthening. It showed that after the entry of silicate ions, C-S-H colloid was formed by reaction with Ca (OH)2, and C-S-H gel blocked some pores, which made the inner microstructure more compact. The results of compressive strength before and after concrete strengthening show that the strengthening effect of calcium ion compensation method for low-strength concrete is better than that of permeable crystallization material strengthening method. The strength of C5 and C15 grade concrete is increased by 36.1% and 6% respectively, and the surface strength of 13.7 MPa concrete is increased by 16.7%.

Bathymetric mapping with Autonomous Underwater Vehicles (AUVs) receives increased attentions in recent years. AUVs offer a lower operational cost and smaller carbon footprint with reduced ship usage, and they can provide higher resolution data when surveying the seabed at a closer distance if compared to ships. However, advancements are still needed to improve the data quality of AUV-based surveys. Unlike mobile robots with deterministic mapping performance, multibeam sonars used in AUV-based bathymetric mapping often yields inconsistent swath width due to the varied seabed elevation and surficial properties. As a result, mapping voids may exist between planned lawnmower transects. Although this could be solved by planning closer lawnmower paths, mission time increases proportionally. Therefore, an onboard path planner is demanded to assure the defined survey objective, i.e., coverage rate. Here in this paper, we present a new data-driven coverage path planning (CPP) method, in which the vehicle automatically updates the waypoints intermittently based on an objective function constructed using the information about the exploration preference, sonar performance, and coverage efficiency. The goal of the proposed method is to plan a cost-effective path on-the-fly to obtain high quality mapping result meeting the requirements in coverage rate and uncertainty. The proposed CPP method has been evaluated in a simulated environment with a 6DOF REMUS AUV model and a realistic seafloor topography. A series of trials has been conducted to investigate the performance affected by the parameters in the objective function. We also compared the proposed method with traditional lawnmower and spiral paths. The results show that the weight assignment in the objective function is critical as they affect the overall survey performance. With proper weight settings, the AUV yields better survey performance, coverage rate and coverage efficiency, compared to traditional approaches. Moreover, the proposed method can be easily adjusted or modified to achieve different coverage goals, such as rapid data gathering of the entire region, survey of irregular workspace, or maintaining real time path planning.

Oxidative stress (OS) is intimately associated with tumorigenesis and has been considered a potential therapeutic strategy. However, the OS-associated therapeutic target for esophageal squamous cell carcinoma (ESCC) remains unconfirmed. In our study, gene expression data of ESCC and clinical information from public databases were downloaded. Through LASSO-Cox regression analysis, a risk score (RS) signature map of prognosis was constructed and performed external verification with the GSE53625 cohort. The ESTIMATE, xCell, CIBERSORT, TIMER, and ImmuCellAI algorithms were employed to analyze infiltrating immune cells and generate an immune microenvironment (IM). Afterward, functional enrichment analysis clarified the underlying mechanism of the model. Nomogram was utilized for forecasting the survival rate of individual ESCC cases. As a result, we successfully constructed an OS-related genes (OSRGs) model and found that the survival rate of high-risk groups was lower than that of low-risk groups. The AUC of the ROC verified the strong prediction performance of the signal in these two cohorts further. According to independent prognostic analysis, the RS was identified as an independent risk factor for ESCC. The nomogram and follow-up data revealed that the RS possesses favorable predictive value for the prognosis of ESCC patients. qRT-PCR detection demonstrated increased expression of MPC1, COX6C, CYB5R3, CASP7, and CYCS in esophageal cancer patients. In conclusion, we have constructed an OSRGs model for ESCC to predict patients' prognosis, offering a novel insight into the potential application of the OSRGs model in ESCC. [ABSTRACT FROM AUTHOR]

Compared with traditional metal contact wet mateable connectors, a noninsert wet mateable connector (NWMC) costs much less and is simpler and safer to plug underwater and can operate steadily for a long duration. This article proposes a newly designed NWMC that can transfer power and data simultaneously in deepsea conditions. Design and analysis methodology of a small volume and stable output inductive coupling power transfer (ICPT) system that combines calculation, simulation, and measurement methods are proposed. The operating parameters are monitored in real time through the communication path, and the heat is dissipated through the end cap. According to the experimental results, this system is able to maintain an output voltage of up to 48 V, transfer up to 130 W power, and simultaneously obtain 73 Mbps full-duplex noncontact communication. Conceiving tall building structure systems is a major step in the plan and design of tall buildings. Through summarizing the basic components and the basic lateral elements in various tall building structures, four conceiving methods for tall building structure systems are presented: (1) intercombination or alone forming by the basic lateral elements, (2) intercombination of different structures, (3) adding basic components in structure systems and (4) replacing basic components in structure systems. With these methods, designers could be able to establish the needed structure system to meet architectural functions, spatial arrangement, facade and other conditions, and these conceiving methods are helpful to architects or engineers to innovate the structure system in their special project.
