In this study, the inhibitory effect of different doses of hydrogen peroxide nano-silver ion composite disinfectant (HPS) on DIV1 and bacteria was analyzed, and the antibacterial activity of three disinfectants, HPS, potassium monopersulfate (KMPS) and calcium hypochlorite (Ca(ClO)2), was compared. In addition, 16S rDNA amplification sequencing technology was used to analyze the effects of these three disinfectants on the intestinal microflora of Litopenaeus vannamei and the structure and composition of water microflora. The results showed that HPS did not positively affect the survival rate of prawns infected with DIV1, which needs to be verified in future studies. In the experimental design range, the higher the dosage of HPS, the more obvious the killing effect on the number of Vibrio and total bacteria in rearing water, and the two showed a negative correlation. 5,104 OTUs were obtained based on 16S rDNA high-throughput sequencing, of which 3,012 (59.01%) and 1,475 (28.90%) OUTs were annotated at the phylum and genus levels, respectively. Proteobacteria, Firmicutes, Actinobacteriota, Bacteroidetes, and Cyanobacteria dominated the water samples at the phylum level. At the genus level, the dominant bacterial genera in the intestinal bacterial community of shrimp were Photobacterium, Vibrio, and Ruegeria. The most dominant bacteria genera in water samples were Vibrio, Ruegeria, Pseudoalteromonas, and Nautella. In the water samples, the composition and structure of the Ca(ClO)2 microbial community were relatively simple, and the species richness and diversity of Ca(ClO)2 were significantly lower than those of HPS and KMPS disinfectant groups at 12 h and 24 h (p < 0.05). In terms of inhibiting the diversity and richness of the microbial community, Ca(ClO)2 had a significant effect (p < 0.05), but the intestinal microbial community diversity of shrimp treated with HPS was higher than that of the other two groups. It is worth noting that, compared with the three disinfectants, HPS has the strongest killing effect on Vibrio and total bacteria, and has a certain positive significance for maintaining the stable state of the microbial community. This study provides a scientific basis for applying HPS in aquaculture and broadens the application range of HPS.

To solve the problem of the large gap in the regulation resources of the power system due to the large number of wind power connected to the grid and to help achieve the strategic energy goal of “carbon neutrality and carbon peaking,” the demand-side adjustable energy-intensive load is taken as the research object in this paper, and a two-stage low-carbon economic scheduling method of the wind power system with multi-form energy-intensive load (MEL) is proposed. First, the operating characteristics of the system with MEL are analyzed. At the same time, a refined mathematical model that conforms to the actual operation characteristics is established. Then, the complementary low-carbon characteristics of the source-charge side and the difference in the response characteristics of the demand-side MEL are considered. Meanwhile, combining with the energy storage system, a day-ahead and intra-day two-stage low-carbon economic scheduling model of the wind power system with MEL is constructed. Finally, an example of the improved IEEE-39 bus system is analyzed to verify the effectiveness of the proposed scheduling method in the low-carbon economic scheduling of wind power systems and to provide a reference for power grid scheduling.

The study investigates the outcomes of adding sodium humate and sodium humate with probiotics (Lysinibacillus, Bacillus subtilis) to culture water on the growth performance, enzyme activity, and microbial environment of shrimp. The sodium humate and probiotics concentrations were 3 mg/L and 105 CFU/mL in the culture water. Litopenaeus vannamei (0.31 ± 0.03 g) at a density of 300 shrimps/m3 were cultured in nine buckets for a 43-days culture experiment. The results showed that the survival rate (SR) of L. vannamei in sodium humate group and sodium humate with probiotics group were (88.44%) and (86.07%), with the sodium humate group outcome being significantly better than the control group. The shrimps’ final body weight and specific growth rate (SGR) in the two experimental groups were significantly higher than the control group. The feed conversion rate (FCR) was significantly lower than the control group. The ammonia nitrogen concentration in sodium humate with probiotics group was significantly lower than the control group on the 15th day. The activities of intestinal amylase, lipase, trypsin and hepatopancreas superoxide dismutase (SOD), phenol oxidase (PO), and catalase (CAT) in sodium humate group were significantly higher than the control group. Notably, SOD, PO, and lipase activities in sodium humate with probiotics group were significantly higher than the control group. Illumina MiSeq high-throughput sequencing showed that the Chao and Ace indices of the culture water microflora in experimental groups were higher than the control group, and the sodium humate group was significantly higher than the control group. At the phylum level, Proteobacteria, Bacteroidetes, and Actinobacteria were the dominant bacterial communities in the intestine and culture water of L. vannamei, Proteobacteria was the most abundant phyla. At the genus level, the relative abundance of Pseudohongiella of water microflora in the control group was significantly lower than the experimental groups. Ruggiella (15.22-19.56%) was the most abundant genera of intestinal microflora. These results infer that the addition of sodium humate enhances the growth performance, digestive enzymes, and some immune enzyme activities, improving the richness of the water microbial community of L. vannamei.

Rolling bearings, as the main components of the large industrial rotating equipment, usually work under complex conditions and are prone to break down. It can provide a certain theoretical basis for identifying the sub-health state of the industrial equipment by the analysis from the incipient weak signals. Thus, a sub-health recognition offline algorithm based on Refined Composite Multiscale Dispersion Entropy (RCMDE) and Deep Belief Network-Extreme Learning Machine (DBN-ELM) optimized by Improved Firework Algorithm (IFWA) is proposed. First of all, in light of the drawbacks that it is easy to fall into local optima and cross the boundary for exploding fireworks in Firework Algorithm (FWA), Cauchy mutation and adaptive dynamic explosion radius factor coefficient is introduced into IFWA. Secondly, Maximum Correlation Kurtosis Deconvolution (MCKD) optimized by the improved parameters is used to process the incipient vibration signals with nonlinearity, nonstationary, and IFWA is used to adaptively adjust to the period $T$ and the filter length $L$ in MCKD(IFWA-MCKD). Then, each sequence of signals is further extracted the feature—RCMDE to rich sample diversity. Finally, combining the powerful unsupervised learning capability from DBN and the generalization capability from ELM, DBN-ELM can be established. What’s more, in order to avoid the interference of human on the parameters, IFWA is used to optimize the number of hidden nodes in DBN-ELM, and the IFWA-DBN-ELM is established. It shows that the algorithm has the higher sub-health recognition accuracy, better robustness and generalization, which has a better industrial application prospect.

Biofloc technology (BFT) is one of the most promising technologies in global aquaculture for the purpose of improving water quality, waste treatment, and disease prevention in intensive aquaculture systems. However, characterization of the microbial species and antibiotic resistance potentially present in biofloc-based aquaculture environments is needed. In this study, we used high-throughput sequencing technology to comprehensively compare the bacterial communities in mariculture ponds of Penaeus monodon (P. monodon), by testing of water, biofloc, and intestine of P. monodon. Operational taxonomic units (OTUs) cluster analysis showed that the nine samples tested divided into 45 phyla and 457 genera. Proteobacteria was the dominant bacteria in water, biofloc and prawn intestine. In biofloc and intestine, the Ruegeria (2.23–6.31%) genus represented the largest proportion of bacteria, with Marivita (14.01–20.94%) the largest group in water. Microbial functional annotation revealed that in all the samples, genes encoding metabolism were predominant. The antibiotic resistance gene annotation showed the highest absolute abundance of patB, adeF, OXA-243, and Brucella_suis_mprF from Proteobacteria. PatB (11.33–15.01%), adeF (15.79–18.16%), OXA-243 (35.65%), and Brucella_suis_mprF (10.03%) showed the highest absolute abundance of antibiotic resistance genes in water, biofloc, and intestines, respectively. These findings may greatly increase our understanding of the characteristics of the microbiota of shrimp biofloc-based aquaculture systems and the complex interactions among shrimp, ambient microflora, and environmental variables. It provides a reference basis for policy on breeding, environmental safety, and maintaining food safety in the production of P. monodon.

As a new type of shrimp lethal virus, decapod iridescent virus 1 (DIV1) has caused huge economic losses to shrimp farmers in China. Up to now, DIV1 has been detected in a variety of shrimps, but there is no report in Marsupenaeus japonicus. In the current study, we calculated the LC50 to evaluate the toxicity of DIV1 to M. japonicus and determined through nested PCR that M. japonicus can be the host of DIV1. Through enzyme activity study, it was found that DIV1 can inhibit the activities of superoxide dismutase, catalase, lysozyme, and phenoloxidase, which could be a way for DIV1 to achieve immune evasion. In a comprehensive study on the transcriptomic changes of M. japonicus in response to DIV1 infection, a total of 52,287 unigenes were de novo assembled, and 20,342 SSR markers associated with these unigenes were obtained. Through a comparative transcriptomic analysis, 6,900 differentially expressed genes were identified, including 3,882 upregulated genes and 3,018 downregulated genes. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that some GO terms related to virus invasion, replication, and host antiviral infection were promoted under DIV1 infection, such as carbohydrate binding, chitin binding, chitin metabolic process, and DNA replication initiation, and some KEGG pathways related to immune response were significantly influenced by DIV1 infection, including Toll and IMD signaling pathway, JAK-STAT signaling pathway, IL-17 signaling pathway, C-type lectin receptor signaling pathway, complement and coagulation cascades, antigen processing and presentation, necroptosis, apoptosis, NOD-like receptor signaling pathway, apoptosis—multiple species, and TNF signaling pathway. Further analysis showed that STAT, Dorsal, Relish, heat shock protein 70 (HSP70), C-type lectins, and caspase play an important role in DIV1 infection. This is the first detailed study of DIV1 infection in M. japonicus, which initially reveals the molecular mechanism of DIV1 infection in M. japonicus by using the transcriptome analysis of hemocytes combined with enzyme activity study.

Although crumb rubber modified asphalt (CRMA) has a better performance than matrix asphalt (MA) in road engineering, the application bottlenecks of this material lies in its high viscosity, poor workability and bad storage stability. Microwave technology is one of the solutions to the above problems. The objective of this study was to investigate the physical and chemical changes before and after aging of CRMA which was prepared by microwave activated crumb rubber, and the aging mechanism of microwave-activated crumb rubber modified asphalt (MACRMA). Through penetration, softening point, ductility, viscosity and DSR test, the high and low temperature performance and temperature sensitivity of MA, CRMA and MACRMA before and after aging were studied. These tests results revealed that microwave treatment can improve the anti-rutting ability and the anti-aging performance of rubber asphalt. According to gel permeation chromatography (GPC) test, the distribution changes of the internal molecular content of the three asphalts were determined during the aging process. Its test results showed that aging effect accelerated the conversion of small molecules to large molecules in asphalt, and the value of large molecular size (LMS) increases more after long-term aging. The microwave activation made the swelling reaction more intense, thereby delayed the conversion of LMS in the rubber asphalt. In addition, LMS has a high correlation with test indicators. The fourier transform infrared spectroscopy (FTIR) test was used to measure the functional group changes of each asphalt. The FTIR spectra indicated that microwave activation is mainly a process of physical change. The carbonyl and sulfoxide functional group indices of MACRMA were lower than those of MA and CRMA, which also confirmed that MACRMA has a better anti-aging ability.

White Spot Syndrome Virus (WSSV) can cause a large-scale death of cultured shrimp and significant damage to the shrimp farming industry. Marsupenaeus japonicus is one of the world's most important economically farmed shrimp. This study found that some M. japonicus survived the spontaneous outbreak of WSSV. Surprisingly, these virus-carrying shrimp showed no apparent illnesses or outbreaks of white spot disease in the subsequent cultivation, and their body size was substantially smaller than healthy shrimp, indicating a long-term fight between the host and the virus. To investigate this interesting phenomenon, we analyzed the transcriptomes of healthy shrimp and survived shrimp through the RNA-Seq platform, attempting to reveal the underlying molecular mechanism of the struggle between M. japonicus and WSSV. Transcriptional analysis showed that a total of 37,815 unigenes were assembled, with an average length of 1,193.34 bp and N50 of 2,049 bp. In the KEGG pathway, enrichment analysis of DEGs pathways related to immunity, biosynthesis, and growth metabolism was enriched, including pentose phosphate pathway, glycerophospholipid metabolism, fatty acid biosynthesis, Wnt signaling pathway, biosynthesis of amino acids, ascorbate, and aldarate metabolism. Our data showed a delicate balance between M. japonicus and WSSV infection: On the one hand, WSSV infection can cause host metabolism and biosynthesis disorders in the host, and the virus consumes a portion of the material and energy required for shrimp average growth and reproduction. If WSSV infection persisted for a long time, then the growth rate of M. japonicus decreased. On the other hand, the host can regulate immune defense to resist subsequent viral infection. This study reveals the underlying molecular mechanism of a long-term battle of M. japonicus against WSSV infection, providing novel insights for preventing WSSV persistent infection in M. japonicus and other farmed shrimp species.
