Introduction: In this study, we aim to describe the radiological characteristics of degenerative cervical kyphosis (DCK) with cervical spondylotic myelopathy (CSM) and discuss the relationship between DCK and the pathogenesis of spinal cord dysfunction. Methods: In total, 90 patients with CSM hospitalized in our center from September 2017 to August 2022 were retrospectively examined in this study; they were then divided into the kyphosis group and the nonkyphosis group. The patients' demographics, clinical features, and radiological data were obtained, including gender, age, duration of illness, cervical Japanese Orthopaedic Association (JOA) score, cervical lordosis (CL), height of intervertebral space, degree of wedging vertebral body, degree of osteophyte formation, degree of disc herniation, degree of spinal cord compression, and anteroposterior diameter of the spinal cord. In the kyphosis group, kyphotic segments, apex of kyphosis, and segmental kyphosis angle were recorded. Radiological characteristics between the two groups were also compared. Correlation analysis was performed for different spinal cord compression types. Results: As per our findings, the patients in the kyphosis group showed more remarkable wedging of the vertebral body, more severe anterior compression of the spinal cord, and a higher degree of disc herniation, while the posterior compression of the spinal cord was relatively mild when compared with the nonkyphosis group. CL was related to the type of spinal cord compression, as cervical kyphosis is an independent risk factor for anterior spinal cord compression. Conclusions: DCK might play a vital role in the pathogenesis of spinal cord dysfunction. In patients with DCK, it was determined that the anterior column is less supported, and more severe anterior spinal cord compression is present. The anterior approach is supposed to be preferred for CSM patients with DCK.

To raise the performance of the current image saliency detection method and promote the extraction effect of salient regions in complex images, from the perspective of multi feature fusion, the image saliency detection method is studied. Corresponding saliency maps is obtained from color and texture channels respectively, and the final saliency map is obtained through linear fusion. On the basis of deep prior information, an improved deep convolutional neural network is used to extract relevant feature vectors such as image pixels, and the nearest neighbor classifier is trained to determine the ownership between pixels and image regions. Zero sample learning iterations are performed to improve the effectiveness of image saliency detection. On this basis, the combination of the above two methods is carried out to obtain the final image saliency detection method. Experimental design is conducted and Matlab software is selected, with a learning rate of 0.001 and 100 iterations. The results showed that compared to other algorithms, the improved algorithm had better detection performance. The detection error of this algorithm was smaller and the accuracy was higher. Its average absolute error value in the JUDD dataset was the smallest, 0.143, which was 0.34 less than the original algorithm. Its detection accuracy in the PASCAL dataset was the highest at 0.786. The missed detection rate and error detection rate of the improved algorithm were both higher than those of the original algorithm, with a missed detection rate of 4.7%, which was 2.5% lower than the original algorithm. Its error detection rate was 5.1%, while the original algorithm’s missed detection rate was 3.2%. The improved algorithm resulted in better image quality, with a maximum peak signal-to-noise ratio of 39.45-dB, which was 6.82-dB higher than the original algorithm. The maximum similarity index value was 0.892. Research methods can effectively detect the saliency of complex images.

Background Research has showcased a correlation between disruptions in gut microbiota and primary membranous nephropathy (pMN), giving rise to the concept of the ‘gut-kidney axis’. However, the precise relationship between gut microbiota and pMN remains elusive. Hence, this study endeavors to investigate whether a causal relationship exists between gut microbiota and pMN utilizing Mendelian randomization (MR) analysis.Methods The primary method employed for MR analysis is the inverse variance weighting method, supplemented by MR-Egger and the weighted median method, to infer causality. This approach was validated within the pMN cohort across two distinct populations.Results At the species level, the abundance of Bifidobacterium bifidum and Alistipes indistinctus was negatively correlated with the risk of pMN. Conversely, pMN was positively associated with Bacilli abundance at the class level, Lachnospiraceae abundance at the family level, and Dialister abundance at the genus level. Specifically, at the species level, pMN was positively correlated with the abundance of Ruminococcus lactaris, Dialister invisus, and Coprococcus_sp_ART55_1.Conclusion These findings lay the groundwork for future research exploring the interplay between pMN and the gut microbiota, with substantial implications for the prevention and treatment of pMN and its associated complications.

BackgroundPrevious research has indicated that Victimization Experiences (VE) may be linked to a heightened likelihood of developing psychological symptoms and Internet Addiction (IA) among adolescents. However, the precise mechanism through which VE contributes to IA in adolescents remains uncertain. This study aimed to investigate whether Social Anxiety (SA) serves as a mediation between VE and IA, utilizing the framework of General Strain Theory.MethodsA cross-sectional survey among 11 middle schools or high schools in Macao was conducted from October to December 2022. Respondents in the victimized group and non-victimized group were 1:1 paired using Propensity Score Matching (PSM) to control the potential confounding factors.ResultsA total of 1,089 questionnaires were valid for analysis and 311 pairs were generated through PSM. The protection pattern of typical marine ecosystems is an overall summary of the regional marine ecosystem protection, indicating the protection status of typical marine ecosystems. The coastal waters of China and ASEAN countries have a highly diverse range of ecosystem types, including mangroves, coral reefs, and seagrass beds. The need to protect these ecosystems is urgent, but establishing marine protected areas often involves conflicts with stakeholders highly dependent on these ecosystems. By constructing an analysis model of the protection pattern and driving mechanisms of typical marine ecosystems, with a focus on the China-ASEAN region, this study aimed to analyze the characteristics of typical marine ecosystems protection patterns in China-ASEAN, evaluate the current protection status of these ecosystems, discuss the driving factors, and derive a regression function to clarify the quantitative relationship between the protection pattern and driving factors of typical marine ecosystems in the China-ASEAN region. The results indicate that there are four main driving factors influencing the protection pattern of typical marine ecosystems in China-ASEAN countries. The most significant driving factor is the Natural Geographic and Resource Factor, followed by Urbanization, Industrial Structure, and Population Size and GDP Factor. Among these, the Natural Geographic and Resource Factor and the Industrial Structure Factor have a positive impact, while the others have a negative impact. In the future, it is crucial to fully consider the diversity and distribution of marine ecosystems to promote joint efforts among China-ASEAN countries in protecting marine ecosystems through international cooperation.

Predicting remaining useful life (RUL) is crucial for tool condition monitoring (TCM) systems. Inaccurate predictions can lead to premature tool replacements or excessive usage, resulting in resource wastage and potential equipment failures. This study introduces a novel tool RUL prediction method that integrates the enhanced northern goshawk optimization (MSANGO) algorithm with a bidirectional long short-term memory (BiLSTM) network. Initially, key statistical features are extracted from collected signal data using multivariate variational mode decomposition. This is followed by effective feature reduction, facilitated by the uniform information coefficient and Mann–Kendall trend tests. The RUL predictions are subsequently refined through a BiLSTM network, with the MSANGO algorithm optimizing the network parameters. Comparative evaluations with BiLSTM, BiGRU, and NGO-BiLSTM models, as well as tests on real-world datasets, demonstrate this method’s superior accuracy and generalizability in RUL prediction, enhancing the efficacy of tool management systems.

Grazing exclusion (GE) is considered an effective strategy for restoring the degradation of overgrazed grasslands on the global scale. Soil microbial diversity plays a crucial role in supporting multiple ecosystem functions (multifunctionality) in grassland ecosystems. However, the impact of grazing exclusion on soil microbial diversity remains uncertain. Here, we conducted a meta-analysis using a dataset comprising 246 paired observations from 46 peer-reviewed papers to estimate how GE affects microbial diversity and how these effects vary with climatic regions, grassland types, and GE duration ranging from 1 to 64 years. Meanwhile, we explored the relationship between microbial diversity and its functionality under grazing exclusion. Overall, grazing exclusion significantly increased microbial Shannon (1.9%) and microbial richness (4.9%) compared to grazing group. For microbial groups, GE significantly increased fungal richness (8.6%) and bacterial richness (5.3%), but decreased specific microbial richness (-11.9%). The responses of microbial Shannon to GE varied among climatic regions, grassland types, and GE duration. Specifically, GE increased microbial diversity in in arid, semi-arid, and dry sub-humid regions, but decreased it in humid regions. Moreover, GE significantly increased microbial Shannon in semidesert grasslands (5.9%) and alpine grasslands (3.0%), but not in temperate grasslands. The paulownia bagworm, Clania variegata Snell, is an economically important pest of agriculture and forests. The sex pheromone of this pest and its stereoisomers were synthesized, and two of the stereoisomers were prepared for the first time. Our strategy was efficient and mainly included the ring-opening reaction of (S)-2-methyloxirane, the coupling of chiral sulfonate, the oxidative cleavage of olefin, and Yamaguchi esterification. Moreover, the overall yields of our synthesis were 23–29%, with eight steps in the longest route.

Based on the water quality monitoring data in the spring, summer and autumn of 2018 in Xiamen Bay, this study used the fuzzy comprehensive evaluation method to comprehensively evaluate the water quality. The results showed that the Western sea area of Xiamen, Tong'an Bay and Jiulong River estuary were seriously polluted areas, which were class Ⅳ water bodies; the water quality of southern sea area, eastern sea area and Dadeng sea area was in good condition, mainly class Ⅰ water body; DIN and DIP were the main pollutants affecting the water quality of Xiamen Bay; using the area ratio method on the basis of the results obtained by the fuzzy comprehensive evaluation method, the results showed that in 2018, Xiamen Bay as a whole was in Class Ⅲ water body condition, and the water quality needed to be further improved.

Achieving the goal of water resource allocation improvement is significant for the ecological conservation of basins. Considering the water supply and demand contradiction and the competition among stakeholders within water-deficient regions, we developed a multi-objective optimization model and proposed the most effective method for allocating water resources based on the study of supply and demand. This study reveals the following findings: 1) the total demand for the Fen River Basin in 2030 will be 6.68 billion m3. Compared with previous years, the actual consumption has increased, but the growth rate has slowed down. 2) The basin’s total supply of water in 2030 will be 7.855 billion m3. With the tightening of the management practice, the consumption and supply in each county showed a decreasing trend year by year. 3) The results of optimal water allocation for 2030 indicate that the FRB should allocate 1.655 billion m3 for domestic water, 4.57 billion m3 for production consumption, and 57 million m3 for ecological use.

Background: Ferroptosis is an emerging type of regulated cell death and associated with antitumoral therapy, while some microRNAs have been shown to regulate the tumorigenesis and cancer progression. Meanwhile, polyphyllin I (PPI) has exhibited antitumoral effects by promoting cancer cell apoptosis and ferroptosis. However, it is unclear whether PPI induces cancer cell ferroptosis by regulating microRNAs.Methods: We used two gastric cancer cell lines (AGS and MKN-45) to set up a tumor model of the nude mice, which were then treated daily with PPI to measure the cancer growth in vitro and in vivo. Ferroptosis was measured using immunofluorescence staining and flow cytometric analysis according to levels of intracellular ROS, lipid ROS and ferrous ions. Moreover, NRF2 expression was measured by Western blotting. In some experiments, the mimics or inhibitors of miR-124-3p were used to further confirm its involvement in PPI-induced cancer cell ferroptosis.Results: Here we found that miR-124-3p mediated cancer ferroptosis and tumor repression induced by PPI since PPI increased miR-124-3p expression in gastric cancer cells and promoted their ferroptosis, whereas inhibition of miR-124-3p mostly abolished the effects of PPI on tumor growth, ferroptosis and NRF2 expression. Moreover, miR-124-3p mimics promoted cancer cell ferroptosis by downregulating NRF2 through directly targeting 3′-UTR region of NRF2, confirming a role for miR-124-3p in regulating PPI-induced ferroptosis.Conclusion: PPI exerts its antitumoral effects on the gastric cancer by promoting cell ferroptosis via regulating miR-124-3p. Our findings have clinical implications for cancer chemotherapy.

Acquiring phenotypic data from livestock constitutes a crucial yet cumbersome phase in the breeding process. Traditionally, obtaining livestock phenotypic data primarily involves manual, on-body measurement methods. This approach not only requires extensive labor but also induces stress on animals, which leads to potential economic losses. Presently, the integration of next-generation Artificial Intelligence (AI), visual processing, intelligent sensing, multimodal fusion processing, and robotic technology is increasingly prevalent in livestock farming. The advantages of these technologies lie in their rapidity and efficiency, coupled with their capability to acquire livestock data in a non-contact manner. Based on this, we provide a comprehensive summary and analysis of the primary advanced technologies employed in the non-contact acquisition of livestock phenotypic data. This review focuses on visual and AI-related techniques, including 3D reconstruction technology, body dimension acquisition techniques, and live animal weight estimation. We introduce the development of livestock 3D reconstruction technology and compare the methods of obtaining 3D point cloud data of livestock through RGB cameras, laser scanning, and 3D cameras. Subsequently, we explore body size calculation methods and compare the advantages and disadvantages of RGB image calculation methods and 3D point cloud body size calculation methods. Furthermore, we also compare and analyze weight estimation methods of linear regression and neural networks. Finally, we discuss the challenges and future trends of non-contact livestock phenotypic data acquisition. Through emerging technologies like next-generation AI and computer vision, the acquisition, analysis, and management of livestock phenotypic data are poised for rapid advancement.

Abstract Background: Long‐term treatment‐free remission (TFR) represents a new goal for chronic myeloid leukemia (CML). Optimizing dose of tyrosine kinase inhibitors (TKIs) in the CML treatment maybe a new challenge to maintain effective and improving patients’ quality of life. We hypothesized that administration of low‐dose TKIs does not compromise major molecular response (MMR) in patients with CML who have a deep molecular response (DMR). Methods: We did an open‐label, randomized trial at eight hospitals in China. Eligible CML‐CP patients (aged 18–70 years) had shown continuous response to TKI more than 5 years and maintained MR4.5 (BCR‐ABLIS ≤ 0.0032%) in recent 18 months. Patients were randomly assigned (1:1) to the TKI de‐escalation group or the discontinuation group. Randomization was done with permuted blocks (block size four) and implemented through an interactive web‐based randomization system. Recurrence was defined as the single sample with real time Quantitative PCR (RT‐qPCR) measurement greater than 0.1% (MMR). The primary endpoint was 12‐month MMR rate in patients who received de‐escalation or discontinuation of TKIs. This study was registered at ClinicalTrials.gov (NCT04143087). Results: Around 125 patients were enrolled between October 23, 2019 and October 31, 2020, 62 patients received dose de‐escalation of TKIs, while 63 patients in the discontinuation group. In the de‐escalation group, molecular recurrence‐free survival at 12 months was 88.32% (95% CI 79%–98%), whereas molecular recurrence‐free survival in the discontinuation group at 12 months was 59.98% (95% CI 47–73). No progressions occurred at the data cut‐off date. All 29 recurrence cases restart TKI treatment returned to MMR. Cytolytic NK cells as a proportion of lymphocyte cells were significantly increased from baseline after 6 months whether in the de‐escalation or TKIs cessation group (P = 0.048, 0.001, respectively); compared with the relapsing patients, Tregs proportion was decreased (P = 0.003), and higher proportion of NK cells were found in non‐relapsing patients whether in TKI de‐escalation or discontinuation group (P = 0.011, 0.007, respectively). We also found that the de‐escalation group showed better disease‐specific HRQOL in regards to its impact on emotional functioning, fatigue, pain, and financial difficulties. Conclusion: With 88.32% MMR in 12‐months follow‐up after de‐escalation TKIs’ treatment, dose‐halving could become a new treatment paradigm for CML patients who with DMR under continuing maintenance therapy with TKIs.

Abstract Magnetic tunnel junction (MTJ) with magnesium oxide (MgO) tunnel barrier is the core element of spin transfer torque-based magnetic random access memory. For the application in the space environment, the total ionizing dose radiation effects on MTJs need to be evaluated. In this work, the MTJs were exposed to X-ray radiation with different doses of up to 10 Mrad(Si). Measurements of current induced magnetization switching (CIMS) behavior of these MTJs were performed before and after radiation. The results show negligible changes in the tunneling magnetoresistance and current switching properties after 8 Mrad(Si) X-ray radiation. However, with a total dose of 9 Mrad(Si), a significant reduction in junction resistance of a fairly large number of MTJs was observed, which showed characteristics of MTJ breakdown. Moreover, in this study, all experimental MTJs became functionally disabled due to MgO breakdown under 10 Mrad(Si) X-ray radiation. The CoFeB/MgO/CoFeB interface microstructure was observed using X-ray photoelectron spectroscopy and high-resolution transmission electron microscopy (HRTEM). Interfacial structural results indicate that the MgO degradation and breakdown behavior caused by X-ray ionizing radiation can give rise to radiation-induced oxygen vacancies across the tunnel barrier oxide layer.

Objective: Heart failure (HF), a worldwide health condition, is the result of many cardiovascular diseases. The traditional Chinese medicine (TCM) Xiaoyu Jiangzhi capsule (XYC) has long been in use in China to treat hyperlipidemia and inhibit platelet aggregation. This study explores the effects of XYC on heart failure (HF) and its detailed mechanisms. Methods: Isoproterenol (ISO, 30 mg/kg) was injected intraperitoneally for 7 days to copy a HF model of 10–12 weeks old, 20–30 g male mice. We then compared the CON (control) group, ISO (HF model) group, MET (metoprolol) group, and XYC group. Cardiac systolic function and left wall thickness were evaluated by echocardiograph. Using western blot analysis, we detected the proteins of calmodulin dependent protein kinase II (CaMKII) and sarco/endoplasmic reticulum Ca2+-ATPase (Serca). Furthermore, tsA201 cells were cultured and the human CaV1.2 calcium channel current (hCaV1.2) were detected by patch clamp experiments. Results: XYC reduced HF, inhibiting the protein expression of CaMKII, but Serca did not change significantly. Moreover, XYC inhibited the peak amplitude of the hCaV1.2 current, depolarizing shifted the activation curve 27.6 mV, and shifted the inactivation curve toward a positive potential 17.6 mV. The fraction recovered from inaction was reduced in XYC group compared with that in CON group. Conclusion: XYC could inhibit ISO-induced HF by reducing the Ca2+/CaMKII signaling pathway in mice.

BackgroundFew studies have focused on the performance of Briganti 2012, Briganti 2017 and MSKCC nomograms in the Chinese population in assessing the risk of lymph node invasion(LNI) in prostate cancer(PCa) patients and identifying patients suitable for extended pelvic lymph node dissection(ePLND). We aimed to develop and validate a novel nomogram based on Chinese PCa patients treated with radical prostatectomy(RP) and ePLND for predicting LNI.MethodsWe retrospectively retrieved clinical data of 631 patients with localized PCa receiving RP and ePLND at a Chinese single tertiary referral center. All patients had detailed biopsy information from experienced uropathologist. Multivariate logistic-regression analyses were performed to identify independent factors associated with LNI. The discrimination accuracy and net-benefit of models were quantified using the area under curve(AUC) and Decision curve analysis(DCA).The nonparametric bootstrapping were used to internal validation.ResultsA total of 194(30.7%) patients had LNI. The median number of removed lymph nodes was 13(range, 11-18). In univariable analysis, preoperative prostate-specific antigen(PSA), clinical stage, biopsy Gleason grade group, maximum percentage of single core involvement with highest-grade PCa, percentage of positive cores, percentage of positive cores with highest-grade PCa and percentage of cores with clinically significant cancer on systematic biopsy differed significantly. The multivariable model that included preoperative PSA, clinical stage, biopsy Gleason grade group, maximum percentage of single core involvement with highest-grade PCa and percentage of cores with clinically significant cancer on systematic biopsy represented the basis for the novel nomogram. Based on a 12% cutoff, our results showed that 189(30%) patients could have avoided ePLND while only 9(4.8%) had LNI missing ePLND. Our proposed model achieved the highest AUC (proposed model vs Briganti 2012 vs Briganti 2017 vs MSKCC model: 0.83 vs 0.8 vs 0.8 vs 0.8, respectively) and highest net-benefit via DCA in the Chinese cohort compared with previous nomograms. In internal validation of proposed nomogram, all variables had a percent inclusion greater than 50%.ConclusionWe developed and validated a nomogram predicting the risk of LNI based on Chinese PCa patients, which demonstrated superior performance compared with previous nomograms.

ObjectiveThe viv-FOAM-SJTU solver is used to carry out the numerical simulation of the vortex induced vibrations of a riser under different top oscillation excitation amplitudes. MethodsThe flow field is computed by the strip theory and Reynolds-averaged Navier-Stokes (RANS) method, while the Bernoulli-Euler bending beam theory and finite element method are employed to model the structure of the riser. ResultsThe simulation results show that the cross-flow vibrations transform from multi-mode to single mode with the decrease in oscillation amplitude. At the same time, the inline vibrations maintain single mode, but the amplitude of the mode vibration decreases. ConclusionWith the variation in top oscillation excitation amplitudes, the washing out of the riser by shed vortices affects the characteristics of vortex induced vibrations.

ObjectivesThis study seeks to optimize the total resistance and seakeeping performance of ships sailing in waves by adding appendages and changing their size. MethodsFirst, numerical calculations are performed on a KCS ship with two anti-pitching fins and a stern flap, and both kinds of appendage separately, to study the effects of the appendages on the hull. Second, thirty KCS ships with deformed appendages are obtained by changing the width of the anti-pitch fins and the length of the stern flap. The in-house viscous flow solver naoe-FOAM-SJTU is used for the calculations. Third, a Kriging surrogate model is established and applied to a multi-objective genetic algorithm to obtain the Pareto front of the hull with optimal appendages. Three KCS ships with optimal appendages are selected from the Pareto front for further analysis. ResultsBy adding appendages to a KCS ship, the pitch magnitude and average total resistance in an encounter period are reduced by about 12.7% and 12.5% respectively. ConclusionsThe resistance and pitch magnitude of a KCS ship in head waves can be optimized by adding appendages such as anti-pitch fins and stern flaps, and changing their principal dimensions.

The aerodynamic properties of rotor systems operating within low Reynolds number flow field conditions are profoundly influenced by their geometric and flight parameters. Precise estimation of optimal airfoil parameters at different angles of attack is indispensable for enhancing these aerodynamic properties. This study presents a technique for optimizing the airfoil parameters of a Mars helicopter by employing machine learning methods in conjunction with computational fluid dynamics (CFD) simulations, thereby circumventing the need for expensive experiments and simulations. The effectiveness of diverse machine learning algorithms for prediction is evaluated, and the resultant models are utilized for airfoil optimization. Ultimately, the aerodynamic properties of the optimized airfoil are experimentally validated. The experimental findings exhibit agreement with the simulated predictions, indicating the successful optimization of the aerodynamic properties. This research offers valuable insights into the influence of airfoil parameters on the aerodynamic properties of the Mars helicopter, along with guidance for airfoil optimization.

In order to clarify the combined impact of tree species composition, site quality, and stand age on the growth and harvest of mixed forests, the prediction models of average DBH and stand volume for mixed forests were established, respectively. The interval period and tree species composition coefficient (TSCC) were considered as independent variables. These models were then optimized by using the particle swarm optimization algorithm for reparameterization and evaluating their applicability. It was found that after introducing the site quality grade and TSCC, the average stand height prediction model showed a better fitting result. The fit accuracy of the average DBH prediction model and the stand volume prediction model were both improved with the help of the TSCC, mainly because the tree species composition affects the growth rate of the average stand height and average DBH and the maximum growth rate of the stand volume. The degree of the impact can be sorted as Cunninghamia lanceolata > Pinus massoniana > hard broad-leaved tree species (group). Overall, the established growth and harvest prediction models for mixed forests with the interval period and TSCC as independent variables have high fit accuracy and applicability.

Background: Multifocal atherosclerosis has dramatically increased annual risk of adverse cardiovascular events than single artery affected. Triglyceride-rich lipoprotein (TRL) has been implicated in the early development of atherosclerosis.However, evidence on the effect of remnant cholesterol, a major atherogenic component of TRL, on multifocal atherosclerosis in Chinese health asymptomatic subjects is insufficient. This study aimed to investigate the association of remnant cholesterol with intra- and extra-cranial atherosclerosis in Chinese population. Methods: This study enrolled 3665 participants (median age 52.31 years) from the Asymptomatic Polyvascular Abnormalities Community study. Parameters of intra- and extra-cranial atherosclerosis includes intracranial artery stenosis (ICAS), carotid plaque, carotid artery stenosis (CAS), and carotid hypertrophy (intima-media thickness >0.9 mm). Previous studies suggested that S. epidermidis possessed low virulence, but recent studies confirmed that it can acquire high virulence from Staphylococcus aureus and with the increasing detection of methicillin-resistant S. epidermidis. It has become a major pathogen of graft-associated and hospital-acquired infections. In previous studies, we modified the antimicrobial peptide Cec4 (41 amino acids) and obtained the derived peptide C9 (16 amino acids) showing better antimicrobial activity against S. epidermidis with an MIC value of 8 μg/mL. The peptide has rapid bactericidal activity without detectable high-level resistance, showing certain inhibition and eradication ability on S. epidermidis biofilms. The damage of cell membrane structures by C9 was observed by scanning emission microscopy (SEM) and transmission electron microscopy (TEM). In addition, C9 altered the S. epidermidis cell membrane permeability, depolarization levels, fluidity, and reactive oxygen species (ROS) accumulation and possessed the ability to bind genomic DNA. Analysis of the transcriptional profiles of C9-treated cells revealed changes in genes involved in cell wall and ribosome biosynthesis, membrane protein transport, oxidative stress, and DNA transcription regulation. At the same time, the median lethal dose of C9 in mice was more than 128 mg/kg, and the intraperitoneal administration of 64 mg/kg was less toxic to the liver and kidneys of mice. Furthermore, C9 also showed a certain therapeutic effect on the mouse bacteremia model. In conclusion, C9 may be a candidate drug against S. epidermidis, which has the potential to be further developed as an antibacterial therapeutic agent. IMPORTANCE S. epidermidis is one of the most important pathogens of graft-related infection and hospital-acquired infection. The growing problem of antibiotic resistance, as well as the emergence of bacterial pathogenicity, highlights the need for antimicrobials with new modes of action. Antimicrobial peptides have been extensively studied over the past 30 years as ideal alternatives to antibiotics, and we report here that the derived peptide C9 is characterized by rapid bactericidal and antibiofilm activity, avoiding the development of resistance by acting on multiple nonspecific targets of the cell membrane or cell components. In addition, it has therapeutic potential against S. epidermidis infection in vivo. This study provides a rationale for the further development and application of C9 as an effective candidate antibiotic.

Objectives Previous studies suggested an inverse association between lipoprotein cholesterols and bleeding risk, while limited data were available about the predictive value of lipoproteins on intracerebral haemorrhage (ICH). ABSTRACT Antibiotic resistance in Acinetobacter baumannii is on the rise around the world, highlighting the urgent need for novel antimicrobial drugs. Antimicrobial peptides (AMPs) contribute to effective protection against infections by pathogens, making them the most promising options for next-generation antibiotics. Here, we report two designed, cationic, antimicrobial-derived peptides: Mt6, and its dextroisomer D-Mt6, belonging to the analogs of MAF-1, which is isolated from the instar larvae of houseflies. Both Mt6 and D-Mt6 have a broad-spectrum antimicrobial activity that is accompanied by strong antibacterial activities, especially against A. baumannii planktonic bacteria and biofilms. Additionally, the effect of D-Mt6 against A. baumannii is stable in a variety of physiological settings, including enzyme, salt ion, and hydrogen ion environments. Importantly, D-Mt6 cleans the bacteria on Caenorhabditis elegans without causing apparent toxicity and exhibits good activity in vivo. Both Mt6 and D-Mt6 demonstrated synergistic or additive capabilities with traditional antibiotics against A. baumannii, demonstrating their characteristics as potential complements to combination therapy. Scanning electron microscopy (SEM) and laser scanning confocal microscope (LSCM) experiments revealed that two analogs displayed rapid bactericidal activity by destroying cell membrane integrity. Furthermore, in lipopolysaccharide (LPS)-stimulated macrophage cells, these AMPs drastically decreased IL-1β and TNF-a gene expression and protein secretion, implying anti-inflammatory characteristics. This trait is likely due to its dual function of directly binding LPS and inhibiting the LPS-activated mitogen-activated protein kinase (MAPK) signaling pathways in macrophages. Our findings suggested that D-Mt6 could be further developed as a novel antimicrobial/anti-inflammatory agent and used in the treatment of A. baumannii infections. IMPORTANCE Around 700,000 people worldwide die each year from antibiotic-resistant pathogens. Acinetobacter baumannii in clinical specimens increases year by year, and it is developing a strong resistance to clinical drugs, which is resulting in A. baumannii becoming the main opportunistic pathogen. Antimicrobial peptides show great potential as new antibacterial drugs that can replace traditional antibiotics. In our study, Mt6 and D-Mt6, two new antimicrobial peptides, were designed based on a natural peptide that we first discovered in the hemlymphocytes of housefly larvae. Both Mt6 and D-Mt6 showed broad-spectrum antimicrobial activity, especially against A. baumannii, by damaging membrane integrity. Moreover, D-Mt6 showed better immunoregulatory activity against LPS induced inflammation through its LPS-neutralizing and suppression on MAPK signaling. This study suggested that D-Mt6 is a promising candidate drug as a derived peptide against A. baumannii.

Insects have the phenomenon of immune priming by which they can have enhanced protection against reinfection with the same pathogen, and this immune protection can be passed on to their offspring, which is defined as “trans-generational immune priming (TGIP).” But whether housefly possesses TGIP is still unclear. Therefore, we used the housefly as the insect model and Candida albicans as the pathogen to explore whether the housefly is capable of eliciting TGIP, and RNA sequencing (RNA-seq) was performed to explore the molecular mechanism of TGIP of the housefly. We found that the housefly possesses TGIP, and adults pre-exposed to heat-killed C. albicans could confer protection to itself and its offspring upon reinfection with a lethal dose of C. albicans. RNA-seq results showed that 30 and 154 genes were differentially expressed after adults were primed with heat-killed C. albicans (CA-A) and after offspring larvae were challenged with a lethal dose of C. albicans (CA-CA-G), respectively. Among the differentially expressed genes (DEGs), there were 23 immune genes, including 6 pattern recognition receptors (PRRs), 7 immune effectors, and 10 immunoregulatory molecules. More importantly, multiple DEGs were involved in the Toll signaling pathway and phagosome signaling pathway, suggesting that the Toll signaling pathway and phagocytosis might play important roles in the process of TGIP of housefly to C. albicans. Our results expanded on previous studies and provided parameters for exploring the mechanism of TGIP.

PurposeHirayama disease (HD) has been largely believed to affect only distal muscles. However, the proximal upper extremities have been affected in some cases, which can be confused with motor neuron diseases.MethodsBaseline data, deep tendon reflex, Hoffmann sign, cervical curvature, sagittal Cobb angle, atrophied spinal cord, high signal intensity, loss of attachment, and affected muscles and segments on electromyography (EMG) were retrospectively obtained and compared between patients with HD with proximal involvement and patients with simple distal HD in one center from September 2007 to April 2022.ResultsIn this study, fifteen patients with proximal HD and 30 patients with simple distal HD were included. The proximal group had a larger proportion of patients with decreased biceps reflex, decreased triceps reflex, brisk or hyperactive knee reflex, positive Hoffmann sign, and cervical kyphosis. The curvatures of the upper part of the cervical spine (C2-4) were lost to a greater degree in the proximal group. More affected segments were observed on magnetic resonance imaging (MRI) and electromyography in the proximal group.ConclusionThe injured segments were longer and the upper curvature of the cervical spine was poorer in patients with HD with proximal involvement. These findings indicated that proximal involvement may indicate more serious HD.

Fruit softening is an important characteristic of peach fruit ripening. The auxin receptor TIR1 (Transport Inhibitor Response 1) plays an important role in plant growth and fruit maturation. Still, little research has been conducted on the relation of TIR1 to the softening of peach fruits. In this study, the hardness of isolated peach fruits was reduced under exogenous NAA treatment at low concentrations. At the same time, the low concentration of NAA treatment reduced the transcription level of PpPG and Ppβ-GAL genes related to cell wall softening and PpACS1 genes related to ethylene synthesis. The transient overexpression of the PpTIR1 gene in peach fruit blocks caused significant down-regulation of the expression of early auxin-responsive genes, ethylene synthesis, and cell wall metabolic genes related to fruit firmness. Through yeast two-hybrid technology, bimolecular fluorescence complementary technology, and a firefly luciferase complementation imaging assay, we were able to unveil an interaction between PpTIR1 and PpIAA1/3/5/9/27 proteins. Furthermore, it was determined that the interaction depended on auxin and its type and concentration. These results show that the PpTIR1-Aux/IAA module has a possible regulatory effect on fruit ripening and softening.

Objective: MicroRNAs (miRNAs) in exosomes had been implicated differentially expressed in patient with moyamoya disease (MMD), but the miRNAs expression in circulating leukocytes remains unclear. This study was investigated on the differential expression of miRNAs in peripheral leukocytes between MMD patients and healthy adults, and among patients with subtypes of MMD.Materials and methods: A total of 30 patients with MMD and 10 healthy adults were enrolled in a stroke center from October 2017 to December 2018. The gene microarray was used to detect the differential expression profiles of miRNA in leukocytes between MMD patients and controls, and the differentially expressed miRNAs were verified by the method of real-time PCR. Objective: To accurately achieve distal contact force, a novel temperature-compensated sensor is developed and integrated into an atrial fibrillation (AF) ablation catheter. Methods: A dual elastomer-based dual FBGs structure is used to differentiate the strain on the two FBGs to achieve temperature compensation, and the design is optimized and validated by finite element simulation. Results: The designed sensor has a sensitivity of 90.5 pm/N, resolution of 0.01 N, and root–mean–square error (RMSE) of 0.02 N and 0.04 N for dynamic force loading and temperature compensation, respectively, and can stably measure distal contact forces with temperature disturbances. Conclusion: Due to the advantages, i.e., simple structure, easy assembly, low cost, and good robustness, the proposed sensor is suitable for industrial mass production.

The control algorithm could greatly help the suspension system improve the comprehensive performance of the vehicle. Existing control methods need to obtain the intermediate states, which are difficult to obtain directly or accurately when estimated by filters or observers. Thus, this paper proposed a new practical finite state LQR control method to deal with this problem. By combining with the output state of the finite sensor of the vehicle suspension system and weakening the unknown state as the goal, an optimization model is established with the design variables as the LQR weight coefficients. Then, the direct relationship between the current control input and the finite sensor output is obtained, and the finite state LQR control is realized. Taking the quarter-car suspension model as an example, the corresponding noise is added considering sensor accuracy, and the control performance of the four control methods is studied considering the uncertainties of suspension system parameters. In addition, the acceleration of sprung mass and the dynamic travel coefficient of suspension have been separately calculated by methods of finite state LQR control, LQR control, and PID control. The results show that there is not much difference between them under shock excitation or random excitation. However, the finite state LQR control method has the best comprehensive control performance in that its dynamic tire load coefficient is better than other methods; it could take into account the suspension work stroke coefficient, dynamic tire load coefficient, and sprung mass’ acceleration of the vehicle suspension system at the same time. In order to realize the optimal control effect with limited sensor arrangement, the finite state LQR control method only needs to obtain the current sensor output and the current control input, without estimating the unknown intermediate state. By this means, the proposed control method greatly simplifies the design of the control system and has great advantages on practical value.

Abstract A cyanate ester (CE) resin co‐modified with epoxy (EP) and bismaleimide (BMI) as the continuous phase, and phenolphthalein poly (ether sulfone) (PES‐C) as the dispersed phase were used for developing liquid oxygen (LOX) composite tanks. Gaseous oxygen (GOX) anti‐oxidation, LOX impact sensitivity, ductility, and cycle durability is a critical index for materials in LOX applications. Added components significantly improved material ductility at low temperature versus neat CE resin because of microstructure changes. However, improvement in thermal resistance was not achieved without tradeoffs; the addition of BMI improved thermal resistance but decreased durability. The failure mechanism of a tank containing GOX was related to reaction with oxygen. LOX impact sensitivity (IRS) is associated with anti‐oxidation and flaming retardance of the samples. Micro‐domains tended to separate from the continuous phase more during the LOX impact test versus the GOX test and was accompanied by increasing ductility and anti‐oxidation. Cycle durability was affected more by the difference in coefficient of thermal expansion between the continuous and PES‐C phases rather than immersion time. In summary, CE resin (CYEP6BM5) co‐modified with PES‐C, EP, and BMI showed good LOX compatibility including IRS value of 2%, 19 cycles durability, and flaming retardance of UL94V0 and may be applied for LOX composite tanks.

Hirayama disease (HD) is characterized by the juvenile onset of unilateral or asymmetric weakness and amyotrophy of the hand and ulnar forearm and is most common in males in Asia. A perception of compliance with previous standards of diagnosis and treatment appears to be challenged, so the review is to update on HD. First, based on existing theory, the factors related to HD includes, (1) cervical cord compression during cervical flexion, (2) immunological factors, and (3) other musculoskeletal dynamic factors. Then, we review the clinical manifestations: typically, (1) distal weakness and wasting in one or both upper extremities, (2) insidious onset and initial progression for 3–5 years, (3) coarse tremors in the fingers, (4) cold paralysis, and (5) absence of objective sensory loss; and atypically, (1) positive pyramidal signs, (2) atrophy of the muscles of the proximal upper extremity, (3) long progression, and (4) sensory deficits. Next, updated manifestations of imaging are reviewed, (1) asymmetric spinal cord flattening, and localized lower cervical spinal cord atrophy, (2) loss of attachment between the posterior dural sac and the subjacent lamina, (3) forward displacement of the posterior wall of the cervical dural sac, (4) intramedullary high signal intensity in the anterior horn cells on T2-weighted imaging, and (5) straight alignment or kyphosis of cervical spine. Thus, the main manifestations of eletrophysiological examinations in HD include segmental neurogenic damages of anterior horn cells or anterior roots of the spinal nerve located in the lower cervical spinal cord, without disorder of the sensory nerves. In addition, definite HD needs three-dimensional diagnostic framework above, while probable HD needs to exclude other diseases via “clinical manifestations” and “electrophysiological examinations”. Finally, the main purpose of treatment is to avoid neck flexion. Cervical collar is the first-line treatment for HD, while several surgical methods are available and have achieved satisfactory results. This review aimed to improve the awareness of HD in clinicians to enable early diagnosis and treatment, which will enable patients to achieve a better prognosis.

Heart failure (HF) affects millions of patients in the world. Shexiang Baoxin Pills (SXB) are extensively applied to treat coronary artery diseases and HF in Chinese hospitals. However, there are still no explanations for why SXB protects against HF. To assess the protective role, we created the HF model in rats by isoproterenol (ISO) subcutaneous injection, 85 milligrams per kilogram body weight for seven days. Four groups were implemented: CON (control), ISO (HF disease group), CAP (captopril, positive drug treatment), and SXB groups. Echocardiography was used to evaluate rats’ HF in vivo. The human CaV1.2 (hCaV1.2) channel currents were detected in tsA-201 cells by patch clamp technique. Five different concentrations of SXB (5, 10, 30, 50, and 100 mg/L) were chosen in this study. The results showed that SXB increased cardiac systolic function and inhibited rats’ cardiac hypertrophy and myocardial fibrosis induced by ISO. Abstract Acidic pH of lysosomes is closely related to autophagy; thus, well known of the precise lysosomes, pH changes will give more information on the autophagy process and status. So far, however, only pH changes in a relatively broad range could be indicated, the exact lysosomes pH detection has never arrived. In our study, we established an endo/lysosome pH indicator based on the self-decomposable SiO2 nanoparticle system with specific synthesis parameters. The central concentrated methylene blue (MB) in the central-hollow structural nanoparticles presented sensitive release as a function of pH values from pH 4.0–4.8, which is exactly the pH range of lysosomes. The linear correlation of the optical density (OD) values and the pH values has been built up, which has been used for the detection of lysosomes pH in 6 different cell lines. Moreover, by this system, we succeeded in precisely detecting the pH average changes of lysosomes before and after black mesoporous silicon (BPSi) NP endocytosis, clarifying the mechanism of the autophagy termination after BPSi endocytosis. So, the self-decomposable nanoparticle-based luminal pH indicator may provide a new methodology and strategy to know better of the lysosome pH, then indicate more details on the autophagy process or other important signaling about metabolisms.

Abstract Background It has been reported that there are more than ten antimicrobial peptides (AMPs) belonging to the cecropin family in Musca domestica; however, few of them have been identified, and the functions of the other molecules are poorly understood. Methods Sequences of the M. domestica cecropin family of genes were cloned from cDNA template, which was reverse-transcribed from total mRNA isolated from third-instar larvae of M. domestica that were challenged with pathogens. Sequence analysis was performed using DNAMAN comprehensive analysis software, and a molecular phylogenetic tree of the cecropin family was constructed using the Neighbour-Joining method in MEGA v.5.0 according to the mature peptide sequences. Antibacterial activity of the synthetic M. domestica cecropin protein was detected and the minimum inhibitory concentration (MIC) values were determined using broth microdilution techniques. Time-killing assays were performed on the Gram-negative bacteria, Acinetobacter baumannii, at the logarithmic or stabilizing stages of growth, and its morphological changes when treated with Cec4 were assessed by scanning electron microscopy (SEM) and detection of leakage of 260 nm absorbing material. Results Eleven cecropin family genes, namely Cec01, Cec02 and Cec1-9, show homology to the Cec form in a multigene family on the Scaffold18749 of M. domestica. In comparing the encoded cecropin protein sequences, most of them have the basic characteristics of the cecropin family, containing 19 conservative amino acid residues. To our knowledge, this is the first experimental demonstration that most genes in the Cec family are functional. Cec02, Cec1, Cec2, Cec5 and Cec7 have similar antibacterial spectra and antibacterial effects against Gram-negative bacteria, while Cec4 displays a more broad-spectrum of antimicrobial activity and has a very strong effect on A. baumannii. Cec4 eliminated A. baumannii in a rapid and concentration-dependent manner, with antibacterial effects within 24 h at 1× MIC and 2× MIC. Furthermore, SEM analysis and the leakage of 260 nm absorbing material detection indicated that Cec4 sterilized the bacteria through the disruption of cell membrane integrity. Conclusions Although there are more than ten cecropin genes related to M. domestica, some of them have no preferred antibacterial activity other than Cec4 against A. baumannii.

Background and Purpose: Peripheral artery disease (PAD) is a manifestation of systemic atherosclerosis with increased risk of severe cardiovascular and cerebrovascular events. The relationship between one-time measuring of low-density lipoprotein cholesterol (LDL-C) and PAD is inconsistent. Increasing evidence shows that the predictive value of non-high-density lipoprotein cholesterol (non-HDLC) on atherosclerosis disease is superior to LDL-C. We aimed to investigate the relationship between cumulative exposure to increased LDL-C and the risk of newly developed PAD and compare the predictive value of LDL-C with non-HDLC.Materials and Methods: In the Asymptomatic Polyvascular Abnormalities Community study, we enrolled 2,923 participants with LDL-C and non-HDLC measured every 2 years from 2006 to 2012. Cumulative exposure to increased LDL-C and non-HDLC, defined as LDL-C burden and non-HDLC burden, respectively, was calculated as the weighted sum of the difference between the measured value and the cutoff value. A new occurrence of PAD was identified through ankle brachial index measured in 2010 and 2012. Multivariate models were adopted to analyze the association of LDL-C burden and non-HDLC burden with the newly developed PAD. The receiver operating curve was drawn, and the area under the curve was calculated to compare the predictive performance of LDL-C burden with a single measure of LDL-C in 2006 and non-HDL-C burden adjusted with a model including traditional risk factors.Results: Of the 2,923 participants, 5.4% (158/2,923) were diagnosed as newly developed PAD. In the univariate analysis, the highest quartile of LDL-C burden was significantly associated with new occurrence of PAD [odds ratio (OR) 1.75, 95% confidence interval (CI) 1.13–2.73]. After adjustment for confounding factors, the same result was obtained (OR 1.59, 95%CI 1.01–2.49). The non-HDLC burden failed to show any statistical significance on the newly developed PAD (OR 1.31, 95% CI 0.84–2.04). Though LDL-C burden had a tendency to show better predictive performance than non-HDLC, it did not reach statistical significance (AUCLDL−C = 0.554 vs. AUCnon−HDLC = 0.544, P = 0.655).Conclusions: Cumulative exposure to increased LDL-C is an independent risk factor of newly developed PAD. The predictive value of non-HDLC burden was not revealed.

Groupers with an initial body weight of 9.10 ± 0.03 g were selected to investigate whether dietary addition of 0 (G0) and 1800 mg/kg glycerol monolaurate (GML, G1800) could alleviate the oxidative stress response and intestinal flora imbalance after 0, 6, 12, and 24 h of salinity change in grouper. Experimental results show that the dietary addition of GML significantly reduced the liver MDA content and increased the SOD activity of grouper. The gene expression of CAT and SOD increased and then decreased with time after adding 1800 mg/kg GML, and the highest values were significantly higher than those of the control group. Salinity change had a slight effect on the top four intestinal flora composition of grouper at 0, 12, and 24 h, with changes occurring only at 6 h when Cyanobacteria replaced Actinobacteria. The addition of dietary GML slowed down the intestinal flora disorder, inhibited the colonization of harmful bacterium Vibrio, and promoted the abundance of beneficial bacterium Bacillus. In conclusion, dietary GML significantly reduced the oxidative damage caused by sudden changes in salinity, improved the antioxidant capacity, and alleviated the intestinal flora imbalance in juvenile grouper.

Chlorpyrifos is an organophosphorus insecticide, which can be used to control a variety of chewing and piercing mouthparts pests in agricultural production. It can destroy the normal nerve impulse conduction by inhibiting the activity of acetylcholinesterase or cholinesterase in the nerves, causing a series of poisoning symptoms. In order to achieve the quantitative analysis of chlorpyrifos residues in agricultural products, an aptamer-controlled signal molecule release method was developed in this study. The signal molecule 4-ATP of surface-enhanced Raman spectroscopy (SERS) was loaded into aminated mesoporous silica nanoparticles (MSNs-NH2) prepared by the one pot method, and then coated with an aptamer of chlorpyrifos through electrostatic interaction. The specific binding of the aptamer and chlorpyrifos led to the release of 4-ATP, and the amount of 4-ATP released was positively correlated with the amount of chlorpyrifos. Finally, the standard curve of chlorpyrifos quantitative detection based on SERS was established. Meanwhile, Ag-carrying mesoporous silica (Ag@MSNs) was prepared as the reinforcement substrate for SERS detection. The results showed that there was a good linear correlation between the Raman intensity and the concentration of chlorpyrifos at 25–250 ng/mL, and the limit of detection (LOD) was 19.87 ng/mL. The recoveries of chlorpyrifos in the apple and tomato samples were 90.08–102.2%, with RSD < 3.32%. This method has high sensitivity, specificity, reproducibility and stability, and can be used for the quantitative detection of chlorpyrifos in the environment and agricultural products.

In ultra-precision positioning equipment, the positioning accuracy is affected by the friction characteristics, especially the pre-slip stage. At present, the research on friction mainly includes contact theory and the dynamic friction process. There is no time variable in contact theory models, so they only apply to the stage of static contact, while the establishment of a friction dynamics model depends on parameter identification and cannot reflect the influence of a rough morphology and load. Therefore, neither theory can elucidate the ultra-precise slip mechanism. In this paper, maximum static friction force, tangential stiffness, and tangential damping models of contact surfaces were deduced through fractal contact theory. By substituting the contact parameters into the modified LuGre model, a dynamic slip model of the mask was established. Finally, the above model was verified by a reticle dynamic slip measurement system. The experimental results showed that in the pre-slip stage of the reticle, with an increase in the normal external load, the slippage of the reticle decreased. The theoretical value calculated by the model was basically consistent with the experimental value, and the slippage of the reticle mainly originated from the tangential deformation and relative sliding of the contact surface. With the increase in the normal external load, the proportion of the tangential deformation in the sliding of the entire contact surface was larger. For the change in slippage during different motion stages, the theoretical value was close to the experimental value after eliminating system errors such as vibration and fiber bending, which proved the correctness of the model in this paper.

Acetamiprid (ACE) is widely used in various vegetables to control pests, resulting in residues and posing a threat to human health. For the rapid detection of ACE residues in vegetables, an indirect competitive chemiluminescence enzyme immunoassay (ic-CLEIA) was established. The optimized experimental parameters were as follows: the concentrations of coating antigen (ACE-BSA) and anti-ACE monoclonal antibody were 0.4 and 0.6 µg/mL, respectively; the pre-incubation time of anti-ACE monoclonal antibody and ACE (sample) solution was 30 min; the dilution ratio of goat anti-mouse-HRP antibody was 1:2500; and the reaction time of chemiluminescence was 20 min. The half-maximum inhibition concentration (IC50), the detection range (IC10–IC90), and the detection limit (LOD, IC10) of the ic-CLEIA were 10.24, 0.70–96.31, and 0.70 ng/mL, respectively. The cross-reactivity rates of four neonicotinoid structural analogues (nitenpyram, thiacloprid, thiamethoxam, and clothianidin) were all less than 10%, showing good specificity. The average recovery rates in Chinese cabbage and cucumber were 82.7–112.2%, with the coefficient of variation (CV) lower than 9.19%, which was highly correlated with the results of high-performance liquid chromatography (HPLC). The established ic-CLEIA has the advantages of simple pretreatment and detection process, good sensitivity and accuracy, and can meet the needs of rapid screening of ACE residues in vegetables.

Providing both personal and social benefits, vaccination may be motivated by collective responsibility (CR). Some previous studies have indicated the relationship between CR and vaccination but could not exclude confounding bias and had little knowledge about the boundary conditions. This study aimed to examine the association between CR and COVID-19 vaccine uptake and its boundary conditions in an extended version of the theory of planned behavior. A cross-sectional survey with 608 participants from six tourism satellite industries in Macao was conducted from 28 July 2021 to 20 August of 2021. Respondentss in CR-lower and CR-higher groups were 1:1 paired using propensity score matching (PSM) to control the potential confounding factors. Results showed participants in the CR-higher group reported significantly higher COVID-19 vaccine uptake than those in the CR-lower group (64.7% vs. 49.7%, p = 0.005). Multivariate logistic regression results indicated a positive association between CR and COVID-19 vaccine uptake (p = 0.012, OR = 2.070, 95% CI= 1.174 to 3.650) and its interaction effect with COVID-19 vaccine attitude (p = 0.019, OR = 0.922, 95% CI = 0.861 to 0.987). Spotlight analysis further illustrated that CR was more effective among individuals with a more negative COVID-19 vaccine attitude. These findings may help promote understanding of vaccine hesitancy, and hence optimize vaccination communication strategies during the COVID-19 pandemic.

Flexure leaf spring (FLS) with large deformation is the basic unit of compliant mechanisms with large stroke. The stiffness along the non-working directions of FLSs with large width-to-length ratio (w/L) is high. The motion stability of the compliant mechanism based on this type of FLS is high. When this type of FLS is loaded along the width direction, the shear deformation needs to be characterized. Nevertheless, currently available compliance modeling methods for FLS are established based on Euler–Bernoulli beam model and cannot be used to characterize shear models. Therefore, these methods are not applicable in this case. In this paper, a new six-DOF compliance model for FLSs with large w/L is established under large deformation. The shear deformation along the width direction model is characterized based on the Timoshenko beam theory. The new constraint model and differential equations are established to obtain a high-precision compliance model expression for this type of FLS. The effects of structural parameters on the compliance of the FLS are analyzed. Finally, the accuracy of the model is verified both experimentally and by finite element simulation. The relative error between theoretical result and experiment result is less than 5%.

Background: Macrophages regulate the inflammatory response and affect re-endothelialization. Inflammation and macrophages play important roles in promoting tissue repair, but p38α mitogen-activated protein kinase's role in re-endothelialization is unknown. Methods and results: Wire injuries of carotid arteries and Evans blue staining were performed in macrophage-specific p38α-knockout (p38αfl/flLysMCre+/-) mice and control mice (p38αfl/fl). Re-endothelialization of the carotid arteries at 3, 5 and 7 days was significantly promoted in p38αfl/flLysMCre+/- mice. In vitro experiments indicated that both the proliferation and migration of endothelial cells were enhanced in conditioned medium from peritoneal macrophages of p38αfl/flLysMCre+/- mice. Interleukin-6 (IL-6) level was decreased significantly in macrophages of p38αfl/flLysMCre+/- mice and an IL-6-neutralizing antibody promoted endothelial cell migration in vitro and re-endothelialization in p38αfl/fl mice in vivo. Phosphoproteomics revealed that the phosphorylation level of S544/T545/S549 sites in megakaryocytic leukemia 1 (MKL1) was decreased in p38αfl/flLysMCre+/- mice. The mutation of either S544/S549 or T545/S549 sites could reduce the expression of IL-6 and the inhibition of MKL1 reduced the expression of IL-6 in vitro and promoted re-endothelialization in vivo. Conclusion: p38α in macrophages aggravates injury of arteries by phosphorylating MKL1, and increasing IL-6 expression after vascular injury.

In order to obtain a new structure of beam flexure hinge with good performance, the flexure hinge based on the X-lattice structure is researched in this paper. The truss model in the finite element method is used to model the 6-DOF compliance of the flexure hinge based on the X-lattice structure. The influence of structural parameters on the compliance and compliance ratio of flexure hinges is analyzed based on this model, and the performance is compared with the traditional beam flexure hinge of the same size. In order to design a flexure hinge based on the X-lattice structure with good comprehensive performance, this paper proposes an intelligent structure optimization method based on a genetic algorithm. The feasibility of the optimization algorithm is verified by an example.

Background and Purpose: Intracranial atherosclerosis has gained increasing attention due to the high risk of recurrent clinical or subclinical ischemic events, while the relationship between low-density lipoprotein cholesterol (LDL-C) measured at a single time point and intracranial atherosclerotic stenosis (ICAS) is inconsistent. This study aims to assess the association between cumulative exposure to increased LDL-C and the prevalence of asymptomatic ICAS.Methods: The Asymptomatic Polyvascular Abnormalities Community study was investigated on the epidemiology of asymptomatic polyvascular abnormalities in Chinese adults. In this study, we included 4,523 participants with LDL-C measured at 3 examinations in 2006, 2008, and 2010. Cumulative exposure to increased LDL-C was calculated as following: LDL-C burden2006−2008 = [(LDL-C2006-1.8) + (LDL-C2008-1.8)]/2 * time2006−2008; LDL-C burden = LDL-C burden2006−2008 + LDL-C burden2008−2010. Transcranial doppler ultrasonography was performed in 2010 to detecting the ICAS.Results: Of the 4,347 patients, 13.3% (580/4,347) were diagnosed with ICAS. In univariate analysis, the association between LDL-C burden and ICAS prevalence was significant, the odds ratios (95% confidence interval) from the lowest to the highest quartile were 1 (reference), 1.30 (0.99–1.70), 1.32 (1.01–1.73), and 2.14 (1.66–2.75), respectively (P < 0.05). After adjustment for potential confounding factors, the same result was reached.Conclusions: Cumulative exposure to increased LDL-C is concentration-dependently associated with increased prevalence of asymptomatic ICAS, especially in those under the age of 65 y or free of hypertension, diabetes mellitus, and hyperlipidemia.

The drug resistance rate of Acinetobacter baumannii increases year on year, and the drugs available for the treatment of carbapenem-resistant A. baumannii (CRAB) infection are extremely limited. A. baumannii, which forms biofilms, protects itself by secreting substrates such as exopolysaccharides, allowing it to survive under adverse conditions and increasing drug resistance. Antimicrobial peptides are small molecular peptides with broad-spectrum antibacterial activity and immunomodulatory function. Previous studies have shown that the antimicrobial peptide Cec4 has a strong effect on A. baumannii, but the antibacterial and biofilm inhibition of this antimicrobial peptide on clinical carbapenem resistance A. baumannii is not thoroughly understood. In this study, it was indicated that most of the 200 strains of CRAB were susceptible to Cec4 with a MIC of 4 μg/ml. Cec4 has a strong inhibitory and eradication effect on the CRAB biofilm; the minimum biofilm inhibition concentration (MBIC) was 64–128 μg/ml, and the minimum biofilm eradication concentration (MBEC) was 256–512 μg/ml. It was observed that Cec4 disrupted the structure of the biofilm using scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM). A comparative transcriptome analysis of the effects of the antimicrobial peptide Cec4 on CRAB biofilm, identified 185 differentially expressed genes, including membrane proteins, bacterial resistance genes, and pilus-related genes. The results show that multiple metabolic pathways, two-component regulation systems, quorum sensing, and antibiotic synthesis-related pathways in A. baumannii biofilms were affected after Cec4 treatment. In conclusion, Cec4 may represent a new choice for the prevention and treatment of clinical infections, and may also provide a theoretical basis for the development of antimicrobial peptide drugs.

The aerial manipulator is a complex system with high coupling and instability. The motion of the robotic arm will affect the self-stabilizing accuracy of the unmanned aerial vehicles (UAVs). To enhance the stability of the aerial manipulator, a composite controller combining conventional proportion integration differentiation (PID) control, fuzzy theory, and neural network algorithm is proposed. By blurring the attitude error signal of UAV as the input of the neural network, the anti-interference ability and stability of UAV is improved. At the same time, a neural network model identifier based on Maxout activation function is built to realize accurate recognition of the controlled model. The simulation results show that, compared with the conventional PID controller, the composite controller combined with fuzzy neural network can improve the anti-interference ability and stability of UAV greatly.

Aims: To explore the effect and mechanism of gastrodin (GAS) on human umbilical vein endothelial cells (HUVECs) apoptosis induced by oxidative stress and its function in wound healing.Main methods: HUVECs were incubated with tert-butyl hydroperoxide (TBHP) to induce endothelial cell dysfunction and GAS was used as a protector. Cell viability was detected by Counting Kit-8 (CCK-8). HUVECs apoptosis was evaluated by TUNEL assay and western blotting for cleaved caspase3 (C-caspase3) and other apoptosis-related proteins. Transwell migration assay, tube formation assay, and cell-matrix adhesion assay were performed to evaluated cell function of HUVECs. Transfection with nuclear factor-erythroid 2-related factor 2 (Nrf2) small interfering ribonucleic acid and western blotting for Nrf2, HO-1, and apoptosis-related proteins were performed to prove that Nrf2/HO-1 pathway is involved in the protective effects of GAS. The skin wound model of rat was used to assess the protective effects of GAS in vivo.Key Findings: The results show that treating HUVECs with GAS attenuated TBHP-induced apoptosis and cellular dysfunction, including cellular tube formation, migration, and adhesion. Mechanistically, we found that GAS protects HUVECs from TBHP-induced cellular apoptosis by activating the nuclear factor (erythroid-derived 2)-like 2 (Nrf2)/heme oxygenase 1 (HO-1) pathway. An in vivo study illustrated that the oral administration of GAS enhances vascularization in regenerated tissue and facilitates wound healing.Significance: The findings of this study demonstrated that GAS may serve as a potential agent that accelerates wound healing.

The steering arm has recently been frequently broken in a kind of mining truck with Macpherson suspension. To accelerate replacing the broken parts and minimize the economic cost, a fast calculation method for improving the steering arm is proposed in this paper. In this method, the forces on the steering arm are calculated by quasi-static analysis under a low vehicle velocity. Dynamic characteristics of the tire and road are partly included by considering the ranges of the rolling resistance coefficient and friction coefficient from the empirical values, which determines the torque on the steering arm under extreme conditions. The rigid–flexible coupling model for the left steering mechanism in ANSYS Workbench is established and solved to obtain the distribution stress on the steering arm under extreme conditions. Then, the reliability of the simulation results based on this fast calculation method is verified by the experiment. After determining an improvement scheme considering the economic and time cost, the satisfactory strength is obtained. The results illustrate that the strength of the improved steering arm has nearly doubled. Finally, the effectiveness of the improved steering arm is demonstrated by the users’ feedback after it is manufactured, installed, and used.
