IntroductionMuscovy duck parvovirus (MDPV), Goose parvovirus (GPV), Duck circovirus, (DuCV) and Duck adenovirus 3 (DAdV-3) are important pathogens that cause high morbidity and mortality in ducks, causing huge economic loss for the duck industry.MethodsThe present study, a quadruplex one-step real time quantitative PCR method for the detection of MDPV, GPV, DuCV, and DAdV-3 was developed. ResultsThe results showed that assay had no cross-reactivity with other poultry pathogens [Duck plague virus (DPV), Duck tembusu virus (DTMUV), H6 avian influenza virus (H6 AIV), New duck reovirus (NDRV), Newcastle disease virus (NDV), H4 avian influenza virus (H4 AIV), Escherichia coli (E. coli), Muscovy duck reovirus (MDRV), Egg drop syndrome virus (EDSV), Pasteurella multocida (P. multocida)]. The sensitivity result showed that the limits of detection for MDPV, GPV, DuCV, and DAdV-3 were 10, 10, 1 and 10 copies/µl, respectively; The coefficients of variation intra- and inter-method was 1-2%; The range of linear (109 to 103 copies/µL) demonstrated the R2 values for MDPV, GPV, DuCV, and DAdV-3 as 0.9975, 0.998, 0.9964, and 0.996, respectively. The quadruplex real time quantitative PCR method efficiency was 90.30%, 101.10%, 90.72%, and 90.57% for MDPV, GPV, DuCV, and DAdV-3, respectively. 396 clinical specimens collected in some duck sausages from June 2022 to July 2023 were simultaneously detected using the established quadruplex real time quantitative PCR method and the reported assays. The detection rates for MDPV, GPV, DuCV, and DAdV-3 were 8.33% (33/396), 17.93% (71/396), 33.58% (133/396), and 29.04% (115/396), respectively. The agreement between these assays was greater than 99.56%.DiscussionThe developed quadruplex real-time quantitative PCR assay can accurately detect these four viruses infecting ducks, providing a rapid, sensitive, specific and accurate technique for clinical testing.

Rock mass may be exposed to high temperature and cooled by water in fire, geothermal resource exploitation and deep underground engineering, which affects the safety of these engineering. In this study, cracked straight through Brazilian disc (CSTBD) specimen was used to study the fracture properties of granite treated with different water-cooling time from 25 °C to 800 °C. A camera was used to record the fracture process of the specimen, the evolution of strain at the specimen surface was obtained by using digital image correlation technology, and the morphological characteristics of fracture surface were observed by using scanning electron microscopy. The results show that water-cooling time has some effects on fracture properties of granite, but its effect is less than the temperature. With the increase of water-cooling time, the mode I fracture toughness decreases first, then increases and finally decreases at the same temperature, and the fracture toughness ranges from 0.419 MPa m1/2 to 0.567 MPa m1/2 at 400 °C. The crack tip opening displacement (CTOD), crack propagation path and fracture surface are related to the energy released during the specimen failure. When the temperature is 25 °C or 200 °C, the energy released is large and the fracture surface is relatively flat with irregular sharp edges, the main cracks are straight lines. At 600 °C, the main crack is zigzag, the CTOD and the maximum principal strain in order can reach 0.143 mm and 0.731 %. This study can help to understand the effect of water-cooling time on engineering rock mass.

In tunnelling and mining applications, the rock-shotcrete interface is crucial for structural stability at the initial support stage, but its complex stress distribution remains poorly understood due to the rock surface depressions. This study employs a series of simulations and model tests to investigate the influence of depression shape and depth on the interface. The results indicate that increased depression depth negatively impacts nominal tensile strength (NTS) and crack initiation energy, although these effects are lessened with rectangular-toothed depressions. The tensile stress on non-depressed surfaces is unaffected by depression depth but is influenced by its shape. Due to the concentration of tensile stress, non-concave interfaces are most prone to failure. In underground rock early support, the construction method of rectangular depression chiselling is more conducive to improving the strength of the shotcrete-rock interface.

This study employs a two-sample bidirectional Mendelian randomization (MR) approach to systematically evaluate the causal relationship between gut microbiota and oral cavity cancer (OCC).ObjectiveTo address the challenge in establishing the causal relationship between gut microbiota and OCC, we applied a systematic MR analysis.MethodsUtilizing GWAS data from the MiBioGen consortium (18,340 individuals) and UK Biobank (n = 264,137), we selected instrumental variables and employed MR-Egger, weighted median, IVW, and weighted mode analyses. Heterogeneity and pleiotropy were assessed using Cochran’s Q test and MR-Egger intercept test.ResultsOur findings indicate, at the order level, Bacteroidales (OR = 0.9990, 95% CI = 0.9980–1.0000, P = 0.046), Burkholderiales (OR = 1.0009, 95% CI = 1.0001–1.0018, P = 0.033), and Victivallales (OR = 0.9979, 95% CI = 0.9962–0.9995, P = 0.037) exhibit causality on OCC in the Weighted median, IVW, and MR-Egger analyses, respectively. At the family level, Alcaligenaceae (OR = 1.0012, 95% CI = 1.0004–1.0019, P = 0.002) and Clostridiaceae1 (OR = 0.9970, 95% CI = 0.9948–0.9992, P = 0.027) show causality on OCC in IVW and MR-Egger analyses. At the genus level, Clostridiumsensustricto1 (IVW, OR = 0.9987, 95% CI = 0.9980–0.9995, P = 0.001; MR-Egger, OR = 0.9978, 95% CI = 0.9962–0.9995, P = 0.035), Desulfovibrio (IVW, OR = 1.0008, 95% CI = 1.0001–1.0015, P = 0.016), Eggerthella (IVW, OR = 0.9995, 95% CI = 0.9990–1.0000, P = 0.048), Eubacterium fissicatena group (IVW, OR = 1.0005, 95% CI = 1.0000–1.0009, P = 0.032), and Holdemanella (IVW, OR = 0.9994, 95% CI = 0.9989–0.9999, P = 0.018) are implicated in causing OCC in related analyses.ConclusionOur study identifies Burkholderiales order, Alcaligenaceae family, Desulfovibrio genus, and Eubacterium fissicatena group as causally increasing OCC risk. In contrast, Bacteroidales order, Victivallales order, Clostridiaceae1 family, Clostridiumsensustricto1 genus, Eggerthella genus, and Holdemanella genus are causally associated with a decreased OCC risk. However, further investigations are essential to delineate an optimal gut microbiota composition and unravel the underlying mechanisms of specific bacterial taxa in OCC pathophysiology.

Porcine reproductive and respiratory syndrome virus (PRRSV), a member of the Arteriviridae family, represents a persistent menace to the global pig industry, causing reproductive failure and respiratory disease in pigs. In this study, we delved into the role of histone deacetylases (HDAC2) during PRRSV infection. Our findings revealed that HDAC2 expression is downregulated upon PRRSV infection. Notably, suppressing HDAC2 activity through specific small interfering RNA led to an increase in virus production, whereas overexpressing HDAC2 effectively inhibited PRRSV replication by boosting the expression of IFN-regulated antiviral molecules. Furthermore, we identified the virus’s nonstructural protein 11 (nsp11) as a key player in reducing HDAC2 levels. Mutagenic analyses of PRRSV nsp11 revealed that its antagonistic effect on the antiviral activity of HDAC2 is dependent on its endonuclease activity. In summary, our research uncovered a novel immune evasion mechanism employed by PRRSV, providing crucial insights into the pathogenesis of this virus and guiding the development of innovative prevention strategies against PRRSV infection.

Cracked straight-through Brazilian disc (CSTBD) samples prepared using two rock materials were used for thermal treatment from room temperature to 700 °C. Uniaxial splitting experiments were performed using an automatic electro-hydraulic servo press to study the evolution laws of physical and fracture properties of different deep rock materials under high-temperature geological conditions. The fracture characteristics were measured using an industrial camera and digital image correlation technology to analyze the effect of high temperature on fracture properties and failure modes of the CSTBD samples after different thermal treatments. The micro-damage properties of green sandstone and granite materials were obtained using a scanning electron microscope (SEM). The following conclusions were drawn from the test results: (1) With the increasing temperature, the fracture characteristics of green sandstone and granite change from brittle fracture to plasticity fracture, the longitudinal wave velocity of granite decreases sharply at 600 °C, and the damage factor reaches 0.8748 at 700 °C. (2) The fracture toughness of green sandstone and granite decreases with increasing temperature; however, the decreasing range of granite is larger than that of green sandstone. (3) As the temperature increases, the fracture morphologies of green sandstone and granite materials become rougher, whereas thermal damage cracks of granite and intergranular fractures inside sandstone as well as pores of sandstone increase. (4) The crack tip opening displacement and peak strain corresponding to peak load increase with the temperature.

MXene based aerogels with efficient thermal insulation, excellent electrical conductivity, reversible compressibility, and high durability hold great potential for various applications, ranging from wearable electronics to multifunctional scaffolds. How to balance various performance to adapt to more application scenarios is an urgent problem to be solved. In the present work, honeycomb AuCu alloy nanoparticles decorated Ti3C2Tx MXene/RGO aerogels (MXene/RGO/AuCu NPs aerogels) were prepared by a simple reduction and directional freeze-drying method. The optimized MXene/RGO/AuCu NPs aerogels have low density (0.023 g/cm3), flame retardant, robust mechanical properties (longitudinal and transverse compressive modulus of 450.9 kPa and 79.9 kPa at 90% strain, respectively) and 80% compressive elasticity. The deformable porous structure enables it to respond to pressure, which has been proved to be applied to pulse and human respiration monitoring. The ordered pore structure and the LSPR effect of MXene and AuCu particles make aerogels have heat resistance, flame retardancy, good electrothermal (can rise to 245.2 °C in 6 s at 7 V drive voltage) and photothermal properties (109 °C in 10 s at 1sun sunlight intensity). The excellent thermal management capability and pressure sensing characteristics make it a prospect application in wearable flexible heaters and artificial skin.

Persistent infection of high-risk human papillomavirus (HPV) and the expression of E6 and E7 oncoproteins are the main causes of cervical cancer. Several prophylactic HPV vaccines are used in the clinic, but these vaccines have limited efficacy in patients already infected with HPV. Since HPV E7 is vital for tumor-specific immunity, developing a vaccine against HPV E7 is an attractive strategy for cervical cancer treatment. Here, we constructed an HPV16 E7 mutant that loses the ability to bind pRb while still eliciting a robust immune response. In order to build a therapeutic DNA vaccine, the E7 mutant was packaged in an adenovirus vector (Ad-E7) for efficient expression and enhanced immunogenicity of the vaccine. Our results showed that the Ad-E7 vaccine effectively inhibited tumor growth and increased the proportion of interferon-gamma (IFN-γ)-secreting CD8+ T cells in the spleen, and tumor-infiltrating lymphocytes in a mouse cervical cancer model was achieved by injecting with HPV16-E6/E7-expressing TC-1 cells subcutaneously. Combining the Ad-E7 vaccine with the PD-1/PD-L1 antibody blockade significantly improved the control of TC-1 tumors. Combination therapy elicited stronger cytotoxic T lymphocyte (CTL) responses, and IFN-γ secretion downregulated the proportion of Tregs and MDSCs significantly. The expressions of cancer-promoting factors, such as TNF-α, were also significantly down-regulated in the case of combination therapy. In addition, combination therapy inhibited the number of capillaries in tumor tissues and increased the thickness of the tumor capsule. Thus, Ad-E7 vaccination, in combination with an immune checkpoint blockade, may benefit patients with HPV16-associated cervical cancer.

Donkeys’ gut microbe is critical for their health and adaptation to the environment. Little research has been conducted on the donkey gut microbiome compared with other domestic animals. The Tibetan Plateau is an extreme environment. In this study, 6 Qinghai donkeys (QH) from the Tibetan Plateau and 6 Dezhou donkeys (DZ) were investigated, and the contents of 4 parts—stomach, small intestine, cecum, and rectum—were collected. 16S rRNA sequencing and metagenomic sequencing were used to analyze the composition and diversity of gut microbial communities in donkeys. N7-methylguanosine (m7G) modification on internal RNA positions plays a vital role in several biological processes. Recent research shows m7G modification is associated with multiple cancers. However, in hepatocellular carcinoma (HCC), its implications remain to be determined. In this place, we need to interrogate the mRNA patterns for 29 key regulators of m7G RNA modification and assess their prognostic value in HCC. Initial, the details from The Cancer Genome Atlas (TCGA) database concerning transcribed gene data and clinical information of HCC patients were inspected systematically. Second, according to the mRNA profiles of 29 m7G RNA methylation regulators, two clusters (named 1 and 2, respectively) were identified by consensus clustering. Furthermore, robust risk signature for seven m7G RNA modification regulators was constructed. Last, we used the Gene Expression Omnibus (GEO) dataset to validate the prognostic associations of the seven-gene risk signature. We figured out that 24/29 key regulators of m7G RNA modification varied remarkably in their grades of expression between the HCC and the adjacent tumor control tissues. Cluster one compared with cluster two had a substandard prognosis and was also positively correlated with T classification (T), pathological stage, and vital status (fustat) significantly. Consensus clustering results suggested the expression pattern of m7G RNA modification regulators was correlated with the malignancy of HCC strongly. In addition, cluster one was extensively enriched in metabolic-related pathways. Seven optimal genes (METTL1, WDR4, NSUN2, EIF4E, EIF4E2, NCBP1, and NCBP2) were selected to establish the risk model for HCC. Indicating by further analyses and validation, the prognostic model has fine anticipating command and this probability signature might be a self supporting presage factor for HCC. Finally, a new prognostic nomogram based on age, gender, pathological stage, histological grade, and prospects were established to forecast the prognosis of HCC patients accurately. In essence, we detected association of HCC severity and expression levels of m7G RNA modification regulators, and developed a risk score model for predicting prognosis of HCC patients’ progression.

The decoupling of cropland and livestock due to the industrialization of livestock production is a difficult problem for sustainable agricultural development in many global locations, including China. As population and urbanization increase, this decoupling is likely to become more serious. To date, the relationship between cropland and livestock has been mainly studied from a single perspective, and mostly at the regional and the local scales. Thus, the objective of our study is to systematically assess the coupling relationship between cropland and livestock from multiple aspects on a large scale. Here, we used a complex system covering cropland, livestock and environment subsystems to comprehensively analyze the spatio-temporal variation of the coupling coordination between cropland and livestock and its influencing factors in China over the past two decades. Elaborating on the data, we constructed a comprehensive system of evaluation indexes for cropland–livestock systems. We used a coupling coordination degree model to evaluate the coupling coordination relationship between cropland and livestock in 31 provinces of China during 2000–2020. The results show that the range of cropland–livestock and cropland–livestock–environment coupling coordination degree was 0.4–0.9. In most of the provinces, there was no risk of cropland and livestock decoupling; however, the coupling coordination degree needed to be increased. More attention should be paid to the coordinated development of cropland and livestock coupling in urbanized areas such as Beijing and Tianjin, where cropland and livestock decoupling was more likely to occur. Among the assessed 29 factors, 15 and 16 had an impact on the cropland–livestock and the cropland–livestock–environment coupling coordination degrees, respectively. Our study provides science-based evidence to support estimating the coupling relationship between cropland and livestock in the future.

BackgroundSince the introduction of the neurodevelopmental perspective of schizophrenia research on individuals at ultra-high risk for psychosis (UHR) has gained increasing interest, aiming at early detection and intervention. Results from fMRI studies investigating behavioral and brain functional changes in UHR during facial emotion recognition, an essential component of social cognition, showed heterogenous results, probably due clinical diversity across these investigations. This fMRI study investigated emotion recognition in a sub-group of the UHR spectrum, namely non-help-seeking, drug-naïve UHR with high cognitive functioning to reveal the neurofunctional underpinnings of their social functioning in comparison to healthy controls.MethodsTwo large cohorts of students from an elite University (n1 = 4,040, n2 = 4,364) were screened firstly with the Prodromal Questionnaires and by surpassing predefined cut-offs then interviewed with the semi-structured Interview for Psychosis-Risk Syndromes to verify their UHR status. Twenty-one identified non-help-seeking UHR and 23 non-UHR control subjects were scanned with functional magnetic resonance imaging while classifying emotions (i.e., neutral, happy, disgust and fear) in a facial emotion recognition task.ResultsBehaviorally, no group differences were found concerning accuracy, reaction times, sensitivity or specificity, except that non-help-seeking UHR showed higher specificity when recognizing neutral facial expressions. In comparison to healthy non-UHR controls, non-help-seeking UHR showed generally higher activation in the superior temporal and left Heschl's gyrus as well as in the somatosensory, insular and midcingulate cortex than the control subjects during the entire recognition task regardless of the emotion categories. In an exploratory analysis, in the non-help-seeking UHR group, functional activity in the left superior temporal gyrus was significantly correlated with deficits in the ability to experience emotions at uncorrected statistical thresholds.ConclusionsCompared to healthy controls, non-help-seeking UHR show no behavioral deficits during facial emotion recognition, but functional hyperactivities in brain regions associated with this cognitive process. Our study may inspire future early intervention and provide loci for treatment using neural stimulation.

The detection of dissolved gases in oil is an important method for the analysis of transformer fault diagnosis. In this article, the potential-doped structure of the Ag3 cluster on the HfSe2 monolayer and adsorption behavior of CO and C2H4 upon Ag3–HfSe2 were studied theoretically. Herein, the binding energy, adsorption energy, band structure, density of state (DOS), partial density of state (PDOS), Mulliken charge analysis, and frontier molecular orbital were investigated. The results showed that the adsorption effect on C2H4 is stronger than that on CO. The electrical sensitivity and anti-interference were studied based on the bandgap and adsorption energy of gases. In particular, there is an increase of 55.49% in the electrical sensitivity of C2H4 after the adsorption. Compared to the adsorption energy of different gases, it was found that only the adsorption of the C2H4 system is chemisorption, while that of the others is physisorption. It illustrates the great anti-interference in the detection of C2H4. Therefore, the study explored the potential of HfSe2-modified materials for sensing and detecting CO and C2H4 to estimate the working state of power transformers.

In this study, the adsorption of noxious gas molecules (NO, Cl2, and O3) on GaN and Au-decorated GaN was systematically scrutinized, and the adsorption energy, bond length, charge, density of state (DOS), partial density of state (PDOS), electron deformation density (EDD), and orbitals were analyzed by the density functional theory (DFT) method. It is found that the interaction between NO and pristine GaN is physical adsorption, while GaN chemically reacts with Cl2 and O3. These observations suggest that pristine GaN may be a candidate for the detection of Cl2 and O3. The highly activated Au-decorated GaN can enhance the adsorption performance toward NO and convert the physical adsorption for NO into chemical adsorption, explaining the fact that precious metal doping is essential for regulating the electronic properties of the substrate material. This further confirms the well-established role of Au-decorated GaN in NO gas-sensing applications. In addition, the adsorption performance of Au-decorated GaN for Cl2 and O3 molecules is highly improved, which provides guidance to scavenge toxic gases such as Cl2 and O3 by the Au-decorated GaN material.

As an effective anticancer drug, the clinical limitation of doxorubicin (Dox) is the time- and dose-dependent cardiotoxicity. Yes-associated protein 1 (YAP1) interacts with transcription factor TEA domain 1 (TEAD1) and plays an important role in cell proliferation and survival. However, the role of YAP1 in Dox-induced cardiomyopathy has not been reported. In this study, the expression of YAP1 was reduced in clinical human failing hearts with dilated cardiomyopathy and Dox-induced in vivo and in vitro cardiotoxic model. Ectopic expression of Yap1 significantly blocked Dox-induced cardiomyocytes apoptosis in TEAD1 dependent manner. Isorhapontigenin (Isor) is a new derivative of stilbene and responsible for a wide range of biological processes. Here, we found that Isor effectively relieved Dox-induced cardiomyocytes apoptosis in a dose-dependent manner in vitro. Administration with Isor (30 mg/kg/day, intraperitoneally, 3 weeks) significantly protected against Dox-induced cardiotoxicity in mice. Interestingly, Isor increased Dox-caused repression in YAP1 and the expression of its target genes in vivo and in vitro. Knockout or inhibition of Yap1 blocked the protective effects of Isor on Dox-induced cardiotoxicity. In conclusion, YAP1 may be a novel target for Dox-induced cardiotoxicity and Isor might be a new compound to fight against Dox-induced cardiotoxicity by increasing YAP1 expression.

We report on a comprehensive multiwavelength study of the pulsars in the globular cluster (GC) M5, including the discovery of M5G, a new compact noneclipsing “black widow” pulsar. Thanks to the analysis of 34 yr of radio data taken with the Five-hundred-meter Aperture Spherical radio Telescope and Arecibo telescopes, we obtained new phase-connected timing solutions for four pulsars and improved those of the other three. These have resulted in, among other things, (a) much improved proper motions for five pulsars, with transverse velocities (relative to the cluster) that are smaller than their respective escape velocities; (b) 3 σ and 1.5 σ detections of Shapiro delays in M5F and M5D, respectively; and (c) greatly improved measurement of the periastron advance in M5B, whose value of $\dot{\omega }=0\buildrel{\circ}\over{.} 01361(6)$ implies that M5B is still likely to be a heavy ( ${m}_{p}={1.981}_{-0.088}^{+0.038}\,{M}_{\odot }$ ) neutron star. The binary pulsars M5D, M5E, and M5F are confirmed to be in low-eccentricity binary systems, the low-mass companions of which are newly identified to be He white dwarfs using Hubble Space Telescope data. Four pulsars are also found to be associated with X-ray sources. Similarly to the eclipsing pulsar M5C, M5G shows little or no nonthermal X-ray emission, indicative of weak synchrotron radiation produced by intrabinary shocks. Recently, slowly cooling white dwarfs (WDs) are a new class of WD that have been identified in two globular clusters (namely M13 and NGC 6752), showing a horizontal branch (HB) morphology with an extended blue tail. The cooling rate of these WDs is reduced by stable thermonuclear hydrogen burning in their residual envelope, and they are thought to originate by stars that populate the blue tail of the HB and then skip the asymptotic giant branch phase. Consistently, no evidence of such kind of WDs has been found in M3, a similar cluster with no blue extension of the HB. To further explore this phenomenon, we took advantage of deep photometric data acquired with the Hubble Space Telescope in the near-ultraviolet and investigated the bright portion of the WD cooling sequence in M5, another Galactic globular cluster with HB morphology similar to M3. The normalized WD luminosity function derived in M5 was found to be impressively similar to that observed in M3, in agreement with the fact that the stellar mass distribution along the HB of these two systems is almost identical. The comparison with theoretical predictions is consistent with the fact that the cooling sequence in this cluster is populated by canonical (fast cooling) WDs. Thus, the results presented in this paper provide further support to the scenario proposing a direct causal connection between the slow cooling WD phenomenon and the horizontal branch morphology of the host stellar cluster.

We report on the discovery of the companion star to the millisecond pulsar PSR J1835−3259B in the Galactic globular cluster NGC 6652. Taking advantage of deep photometric archival observations acquired through the Hubble Space Telescope in near-UV and optical bands, we identified a bright and blue object at a position compatible with that of the radio pulsar. The companion is located along the helium-core white dwarf cooling sequence, and the comparison with binary evolution models provides a mass of 0.17 ± 0.02 M _⊙ , a surface temperature of 11,500 ± 1900 K, and a very young cooling age of only 200 ± 100 Myr. The mass and the age of the companion are compatible with a progenitor star of about 0.87 M _⊙ , which started transferring mass to the primary during its evolution along the subgiant branch and stopped during the early red giant branch phase. Combining together the pulsar mass function and the companion mass, we found that this system is observed at an almost edge-on orbit and hosts a neutron star with a mass of 1.44 ± 0.06 M _⊙ , thus suggesting a highly nonconservative mass accretion phase. The young age of the WD companion is consistent with the scenario of a powerful, relatively young MSP indicated by the earlier detection of gamma-rays from this system.

Subjects with ultra-high risk (UHR) states for psychosis show brain structural volume changes similar to first-episode psychosis and also elevated incidence of environmental risk factors like childhood trauma. It is unclear, however, whether early neurodevelopmental trajectories are altered in UHR. We screened a total of 12,779 first-year Chinese students to enroll 36 UHR subjects (based on clinical interviews) and 59 non-UHR healthy controls for a case-control study of markers of early neurodevelopment. Subjects underwent 3T MRI scanning and clinical characterization, including the childhood trauma questionnaire (CTQ). We then used the CAT12 toolbox to analyse structural brain scans for cortical surface complexity, a spherical harmonics-based marker of early neurodevelopmental changes. While we did not find statistically significant differences between the groups, a trend level finding for reduced cortical complexity (CC) in UHR vs. non-UHR subjects emerged in the left superior temporal cortex (and adjacent insular and transverse temporal cortices), and this trend level association was significantly moderated by childhood trauma (CTQ score). Our findings indicate that UHR subjects tend to show abnormal cortical surface morphometry, in line with recent research; more importantly, however, this association seems to be considerably modulated by early environmental impacts. Hence, our results provide an indication of environmental or gene × environment interactions on early neurodevelopment leading up to elevated psychosis risk.

Summary: Cryptorchidism, commonly known as undescended testis, affects 1%–9% of male newborns, posing infertility and testis tumor risks. Despite its prevalence, the detailed pathophysiology underlying male infertility within cryptorchidism remains unclear. Here, we profile and analyze 46,644 single-cell transcriptomes from individual testicular cells obtained from adult males diagnosed with cryptorchidism and healthy controls. Spermatogenesis compromise in cryptorchidism links primarily to spermatogonium self-renewal and differentiation dysfunctions. We illuminate the involvement of testicular somatic cells, including immune cells, thereby unveiling the activation and degranulation of mast cells in cryptorchidism. Mast cells are identified as contributors to interstitial fibrosis via transforming growth factor β1 (TGF-β1) and cathepsin G secretion. Furthermore, significantly increased levels of secretory proteins indicate mast cell activation and testicular fibrosis in the seminal plasma of individuals with cryptorchidism compared to controls. These insights serve as valuable translational references, enriching our comprehension of testicular pathogenesis and informing more precise diagnosis and targeted therapeutic strategies for cryptorchidism.
