Cervical cancer ranks fourth in women in terms of incidence and mortality. The RNA-binding protein YTH N6-methyladenosine RNA-binding protein F2 (YTHDF2) contributes to cancer progression by incompletely understood mechanisms. We show how YTHDF2 controls the fate of cervical cancer cells and whether YTHDF2 could be a valid target for the therapy of cervical cancer. Sphere formation and alkaline phosphatase staining assays were performed to evaluate tumor stemness of cervical cancer cells following YTHDF2 knockdown. Apoptosis was detected by flow cytometry and TUNEL assay. The compounds 4PBA and SP600125 were used to investigate the correlation between JNK, endoplasmic reticulum stress, tumor stemness, and apoptosis. Data from The Cancer Genome Atlas (TCGA) databases and Gene Expression Omnibus (GEO) revealed that GLI family zinc finger 2 (GLI2) might be the target of YTHDF2. The transcription inhibitor actinomycin D and dual-luciferase reporter gene assays were employed to investigate the association between the GLI2 mRNA and YTHDF2. Nude mouse xenografts were generated to assess the effects of YTHDF2 knockdown on cervical cancer growth in vivo. Knockdown of YTHDF2 up-regulated the expression of GLI2, leading to JNK phosphorylation and endoplasmic reticulum stress. These processes inhibited the proliferation of cervical cancer cells and their tumor cell stemness and promotion of apoptosis. In conclusion, the knockdown of YTHDF2 significantly affects the progression of cervical cancer cells, making it a potential target for treating cervical cancer.

Abstract The DC‐side stability of the grid‐tied converter under different control modes is fully investigated using electrical torque analysis. The small‐signal model of a single converter connected to an ideal DC bus under various control modes is formulated. Accordingly, the damping and synchronising coefficient contributed by the DC network and controllers of grid‐tied converter are separately accessed using the electrical torque analysis method and the stabilising conditions of the grid‐tied converter operating under different control modes are further derived. The system stability mainly corresponds with DC network dynamics under constant active power control mode. On the contrary, the grid‐tied converter under constant DC‐link voltage control mode has no stability problem. Generally, elevating the DC‐link capacitance or decreasing the droop gain can greatly improve the stability margin reserve of the VSC‐HVDC links. In addition, the control gains of the classical PQ controller are proven to have limited impacts on DC‐side system stability. Finally, the results of numerical simulation prove the validity of the proposed stability analysis method and the stable boundary for the grid‐tied converter with different control modes.

The High Friction Surface Treatment (HFST) paving technique is widely employed in engineering practices globally, serving to enhance the long-term anti-skid performance of pavements. However, there still exist significant gaps in research regarding the modified and optimized composition and corresponding content of the polymer binding materials used in HFST, as well as the performance of the pavements laid with modified and optimized blended aggregates during service. To address these issues, this research first modified and optimized the composition ratio of traditional epoxy resin materials. Subsequently, a toughened, high-flow epoxy bonding system suitable for the maintenance of concrete pavements was prepared. Additionally, the study explored the bonding performance of this system with both the cement base surface and aggregates. The results established that the optimal ratio range for the modified epoxy adhesive system is E51 epoxy resin to curing agent to toughening agent to diluent in the proportions of 100: 30–35: 20–30: 5. The experimental group used a high-flow epoxy resin adhesive ratio of E51 epoxy resin: curing agent: toughener: diluent = 100:30:20:5. For control group 1, the ratio of epoxy resin adhesive was E51 epoxy resin: curing agent = 100:30, and for control group 2, the ratio was E44 epoxy resin: curing agent = 1:1. Compared with control groups 1 and 2, the modified epoxy system with the cement base demonstrated a 149 % increase in average pull-off strength and an 18 % increase in average shear strength. Similarly, in comparison with control groups 1 and 2, the modified epoxy system with aggregates exhibited a 174 % increase in average pull-off strength and a 541 % increase in average shear strength. Based on the pull-off and shear test results, it was ultimately determined that the dosage of modified epoxy binder for HFST mixtures should be controlled at 1.0–1.2 kg/m2, and the dosage of aggregate should be controlled at 3.3–4.3 kg/m2. These findings provide valuable insights into optimizing HFST pavement compositions for improved performance and longevity.

AbstractContext Gastrodin has been used as antihypertension therapy in China; however, the mechanisms underlying the effects of gastrodin have yet to be fully elucidated.Objective To explore the therapeutic efficiency of gastrodin as an antihypertensive and determine the mechanisms underlying this effect.Materials and methods C57BL/6 mice were continuously administered angiotensin II (Ang II) (500 ng/kg/min) to induce hypertension. Mice were randomly divided into control, Ang II and Ang II + gastrodin groups. Mice received intragastric administration of gastrodin (5 mg/kg) or double distilled water once a day for 4 weeks. Blood pressure, pulse wave velocity (PWV), thickness of the abdominal aorta, pathological morphology and differential expression transcripts (DETs) were assessed. Abdominal aorta rings and primary isolated vascular smooth muscle cells were subjected to Ang II stimulation to induce hypertension as ex vivo and in vitro models, respectively. Vascular ring tension, release of Ca2+ and levels of proteins involved in the myosin light chain kinase (MLCK)/phospho-myosin light chain 2 (p-MLC2) pathway were determined.Results Gastrodin treatment attenuated increases in blood pressure, PWV and thickness of the abdominal aorta. Treatment with gastrodin resulted in 2785 DETs and the enrichment of vascular contraction and calcium signalling pathways. Gastrodin treatment attenuated Ang II-induced vasoconstriction, produced a norepinephrine-precontracted vasodilation effect (attenuated by verapamil), and reduced intracellular Ca2+ release. Furthermore, gastrodin suppressed activation of the MLCK/p-MLC2 pathway in vivo and in vitro.Conclusions Gastrodin treatment lowers blood pressure, suppresses Ang II-induced vascular contraction and MLCK/p-MLC2 pathway activation, thereby demonstrating the mechanisms underlying the therapeutic efficacy of gastrodin as an antihypertensive.

During the manufacturing of surgical forceps, the flashes of the blanks need to be removed. Manual production has problems such as high labor intensity, low efficiency, and high-risk factors. To solve this problem and realize fully automatic resection, a novel modular workstation was designed and a corresponding process method was proposed. The workstation adopts robots, non-standard automation equipment, and image recognition technology instead of manual loading and blanking, but the blank storage still needs to be performed manually. The critical components were selected according to the workstation design scheme and process method, and the control system design was completed. The reliability of the separation unit was studied through a test platform, and the failure problem caused by uneven force was solved using a blank locking device, which showed that the separation success rate was stabilized at 100%. The detection speed of the image recognition system can reach 100 ms/piece, and the product qualification rate can reach 95.7%. The advantages of the workstation in terms of output and productivity were further analyzed by comparing it to manual production, where the average daily output increased by 12.5% (4500 pieces). In addition, the results of long-term test experiments and power consumption comparison tests showed that the workstations are highly stable and consume little additional power.

Abstract Current polyamide lithium extraction nanofiltration membranes are susceptible to chlorine degradation and/or low permeance, two problems that are hard to reconcile. Here we simultaneously circumvented these problems by designing a quaternized-spiro piperazine monomer and translating its beneficial properties into large-area membranes (1 × 2 m2) via interfacial polymerization with trimesoyl chloride. The quaternary ammonium and spiral conformation of the monomer confer more positive charge and free volume to the membrane, leading to one of the highest permeance (~22 L m−2 h−1 bar−1) compared to the state-of-the-art Mg2+/Li+ nanofiltration membranes. Meanwhile, membrane structures are chlorine resistant as the amine–acyl bonding contains no sensitive N-H group. Thus the high performance of membrane is stable versus 400-h immersion in sodium hypochlorite, while control membranes degraded readily. Molecular simulations show that the high permeance and chlorine resistance, which were reproducible at the membrane module level, arise from the spiral conformation and secondary amine structures of the monomer.

Incorporating the advantages of line commutated converters (LCCs) and voltage-source converters (VSCs), hybrid high voltage DC (HVDC) links have bright prospects in bulk power transmission. For this new technique, however, there is a risk of oscillatory instability in the DC link, and the mechanism behind the instability is still unclear. This paper derives analytical DC-side stabilizing conditions for hybrid HVDC links by using dominant frequency model reduction. The small-signal model of the LCC-VSC link is first truncated by reserving only the state variables highly relevant to the dominant mode so that the expression of the dominant oscillation frequency can be obtained. Dynamics of other state variables are reintroduced and then simplified while leaving their properties nearby the dominant frequency intact. Based on the reduced model, an analytical stability criterion is obtained, which reveals that the DC-side stability of hybrid HVDC links will deteriorate with reduced DC voltage operation, a heavy load, a small DC-link capacitor, slow inner loop dynamics, a small proportional and a large integral gain of the DC voltage regulator. In addition, simplified sufficient stabilizing conditions of hybrid HVDC links are further derived for control parameter design. Case studies validate the accuracy of dominant frequency model reduction and the derived stabilizing conditions.

Natural polyphenols have drawbacks such as instability and low bioavailability, which can be overcome by encapsulated slow-release systems. Natural polymer hydrogels are ideal materials for slow-release systems because of their high biocompatibility. In this study, Longzhua mushroom polysaccharide hydrogel (LMPH) was used to encapsulate rambutan peel polyphenols (RPP) and delay their release time to improve their stability and bioavailability. The mechanical properties, rheology, stability, swelling properties, water-holding capacity, RPP loading, and slow-release behavior of LMPH were investigated. The results showed that LMPH has adequate mechanical and rheological properties, high thermal stability, excellent swelling and water-holding capacity, and good self-healing behavior. Increasing the polysaccharide content not only improved the hardness (0.17–1.13 N) and water-holding capacity of LMPH (90.84–99.32%) but also enhanced the encapsulation efficiency of RPP (93.13–99.94%). The dense network structure slowed down the release of RPP. In particular, LMPH5 released only 61.58% at 48 h. Thus, a stable encapsulated slow-release system was fabricated using a simple method based on the properties of LMPH. The developed material has great potential for the sustained release and delivery of biologically active substances.

Abstract Converter interfaced generation (CIG) may switch into different control modes under different fault scenarios, which poses a significant challenge to the fault calculation of modern power systems. This paper first establishes a reduced fault analysis model of CIG considering control mode switching. By neglecting the fast dynamic processes of the converter controller, CIG with each control mode can be expressed by equality and inequality constraints. Then, an alternative iteration algorithm is proposed to determine the control mode of each grid‐tied converter after fault, which mainly contains the estimation and discrimination steps. In the estimation step, the voltage of the point of common coupling (PCC) and output current of CIG are obtained by simultaneously solving the equality constraints of CIG and the reduced network equation. In the discrimination step, the inequality constraints of CIG are utilized to judge and revise the control mode of each converter. With alternative execution of the estimation and discrimination steps, the fault network can be solved. Simulation studies based on PSCAD/EMTP validate that the proposed method has high accuracy in fault calculation of the CIG embedded system under various fault locations and fault resistances.

Abstract Background Tomato yellow leaf curl virus (TYLCV) is a major monopartite virus in the family Geminiviridae and has caused severe yield losses in tomato and tobacco planting areas worldwide. Wall-associated kinases (WAKs) and WAK-like kinases (WAKLs) are a subfamily of the receptor-like kinase family implicated in cell wall signaling and transmitting extracellular signals to the cytoplasm, thereby regulating plant growth and development and resistance to abiotic and biotic stresses. Recently, many studies on WAK/WAKL family genes have been performed in various plants under different stresses; however, identification and functional survey of the WAK/WAKL gene family of Nicotiana benthamiana have not yet been performed, even though its genome has been sequenced for several years. Therefore, in this study, we aimed to identify the WAK/WAKL gene family in N. benthamiana and explore their possible functions in response to TYLCV infection. Results Thirty-eight putative WAK/WAKL genes were identified and named according to their locations in N. benthamiana. Phylogenetic analysis showed that NbWAK/WAKLs are clustered into five groups. The protein motifs and gene structure compositions of NbWAK/WAKLs appear to be highly conserved among the phylogenetic groups. Numerous cis-acting elements involved in phytohormone and/or stress responses were detected in the promoter regions of NbWAK/WAKLs. Moreover, gene expression analysis revealed that most of the NbWAK/WAKLs are expressed in at least one of the examined tissues, suggesting their possible roles in regulating the growth and development of plants. Virus-induced gene silencing and quantitative PCR analyses demonstrated that NbWAK/WAKLs are implicated in regulating the response of N. benthamiana to TYLCV, ten of which were dramatically upregulated in locally or systemically infected leaves of N. benthamiana following TYLCV infection. Conclusions Our study lays an essential base for the further exploration of the potential functions of NbWAK/WAKLs in plant growth and development and response to viral infections in N. benthamiana.

Abstract Here, the stability region of grid‐connected VSC with phase locked loop (PLL) is estimated. First, the conservative stability region is derived via the classical Lyapunov method. Then an energy function is constructed as the general stable condition for the VSC system. Based on the constructed energy function, two estimation methods of the system stability region by approximation of phase trajectory are proposed. The stability regions by the proposed methods are less conservative than that by the classical Lyapunov method. A stability evaluation function for the initial states of the VSC system is further proposed based on the estimated stability region. Time‐domain simulations verify the correctness of the proposed methods and the effectiveness of the stability evaluation function.

Roasting is an important step in the pretreatment of biomass upgrading. Roasting can improve the fuel quality of biomass, reduce the O/C and H/C ratios in the biomass, and provide the biomass with a fuel quality comparable to that of lignite. Therefore, studying the structure and component evolution laws during biomass roasting treatment is important for the rational and efficient utilization of biomass. When the roasting temperature is 200–300 °C, the cellulose and hemicellulose in the biomass undergo a depolymerization reaction, releasing many monocyclic aromatic hydrocarbons with high reactivity. The proportion of monocyclic aromatic hydrocarbons in biomass roasting products can be effectively regulated by controlling the reaction temperature, residence time, catalyst, baking atmosphere, and other factors in the biomass roasting process. This paper focuses on the dissociation law of organic components in the pretreatment process of biomass roasting.

Studies have shown that functional abnormalities in the locus coeruleus (LC) are strongly associated with depressive symptoms, but the pattern of LC functional connectivity in Alzheimer's disease patients with depressive symptoms (D-AD) remains unclear. The current study aimed to investigate the characteristics of LC functional connectivity (FC) in D-AD using resting-state functional magnetic resonance imaging (rsfMRI). We obtained rsfMRI data in 24 D-AD patients (aged 66–76 years), 14 non-depressive AD patients (nD-AD) (aged 69–79 years) and 20 normal controls (aged 67–74 years) using a 3 T scanner. We used the FC approach to investigate abnormalities in the LC brain network of D-AD patients. One-way ANCOVA and post-hoc two-sample t-tests were performed to compare the strength of functional connectivity from the LC among the three groups. Our results showed that, compared with normal controls, D-AD showed decreased left LC FC with the right caudate and left fusiform gyrus, whereas nD-AD showed decreased left LC FC with the right caudate, right middle frontal gyrus and left fusiform gyrus. Compared to nD-AD, D-AD showed increased left LC FC with right superior frontal gyrus and right precentral gyrus. These findings contribute to our understanding of the neural mechanisms of D-AD.

Abstract Background Depression is a common symptom of Alzheimer’s disease (AD), but the underlying neural mechanism is unknown. The aim of this study was to explore the topological properties of AD patients with depressive symptoms (D-AD) using graph theoretical analysis. Methods We obtained 3-Tesla rsfMRI data from 24 D-AD patients, 20 non-depressed AD patients (nD-AD), and 20 normal controls (NC). Resting state networks were identified using graph theory analysis. ANOVA with a two-sample t-test post hoc analysis in GRETNA was used to assess the topological measurements. Results Our results demonstrate that the three groups show characteristic properties of a small-world network. NCs showed significantly larger global and local efficiency than D-AD and nD-AD patients. Compared with nD-AD patients, D-AD patients showed decreased nodal centrality in the pallidum, putamen, and right superior temporal gyrus. They also showed increased nodal centrality in the right superior parietal gyrus, the medial orbital portion of the right superior frontal gyrus, and the orbital portion of the right superior frontal gyrus. Compared with nD-AD patients, NC showed decreased nodal betweenness in the right superior temporal gyrus, and increased nodal betweenness in medial orbital part of the right superior frontal gyrus. Conclusions These results indicate that D-AD is associated with alterations of topological structure. Our study provides new insights into the brain mechanisms underlying D-AD.

Abstract Several human host proteins play important roles in the lifecycle of severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). Many drugs targeting these host proteins have been investigated as potential therapeutics for coronavirus disease 2019 (COVID‐19). The tissue‐specific expressions of selected host proteins were summarized using proteomics data retrieved from the Human Protein Atlas, ProteomicsDB, Human Proteome Map databases, and a clinical COVID‐19 study. Protein expression features in different cell lines were summarized based on recent proteomics studies. The half‐maximal effective concentration or half‐maximal inhibitory concentration values were collected from in vitro studies. The pharmacokinetic data were mainly from studies in healthy subjects or non‐COVID‐19 patients. Considerable tissue‐specific expression patterns were observed for several host proteins. ACE2 expression in the lungs was significantly lower than in many other tissues (e.g., the kidneys and intestines); TMPRSS2 expression in the lungs was significantly lower than in other tissues (e.g., the prostate and intestines). The expression levels of endocytosis‐associated proteins CTSL, CLTC, NPC1, and PIKfyve in the lungs were comparable to or higher than most other tissues. TMPRSS2 expression was markedly different between cell lines, which could be associated with the cell‐dependent antiviral activities of several drugs. Drug delivery receptor ICAM1 and CTSB were expressed at a higher level in the lungs than in other tissues. In conclusion, the cell‐ and tissue‐specific proteomics data could help interpret the in vitro antiviral activities of host‐directed drugs in various cells and aid the transition of the in vitro findings to clinical research to develop safe and effective therapeutics for COVID‐19.

Abstract Background Chrysanthemum virus B (CVB), a key member of the genus Carlavirus, family Betaflexiviridae, causes severe viral diseases in chrysanthemum (Chrysanthemum morifolium) plants worldwide. However, information on the mechanisms underlying the response of chrysanthemum plants to CVB is scant. Methods Here, an integrated next-generation sequencing and comparative transcriptomic analysis of chrysanthemum leaves was conducted to explore the molecular response mechanisms of plants to a Chinese isolate of CVB (CVB-CN) at the molecular level. Results In total, 4934 significant differentially expressed genes (SDEGs) were identified to respond to CVB-CN, of which 4097 were upregulated and 837 were downregulated. Gene ontology and functional classification showed that the majority of upregulated SDEGs were categorized into gene cohorts involved in plant hormone signal transduction, phenylpropanoid and flavonoid biosynthesis, and ribosome metabolism. Enrichment analysis demonstrated that ethylene pathway-related genes were significantly upregulated following CVB-CN infection, indicating a strong promotion of ethylene biosynthesis and signaling. Furthermore, disruption of the ethylene pathway in Nicotiana benthamiana, a model plant, using virus-induced gene silencing technology rendered them more susceptible to cysteine-rich protein of CVB-CN induced hypersensitive response, suggesting a crucial role of this pathway in response to CVB-CN infection. Conclusion This study provides evidence that ethylene pathway has an essential role of plant in response to CVB and offers valuable insights into the defense mechanisms of chrysanthemum against Carlavirus.

In this work, an efficient synthesis of 3-hydroxy chromones was accomplished through oxidative cyclization mediated by lipase. This enzymatic process has salient features of environmental friendliness, mild reaction condition, satisfactory yield, green reusable biocatalyst, and short reaction time. The proposed method expands the application of lipase in organic synthesis.

Background: 5-Fluorouracil (5-FU) is one of the most commonly prescribed anticancer agents. However, intestinal mucositis is a well-known adverse event, which limits its therapeutic use. Refined Baohe formula (RBF) is derived from Baohe Pills with a potent gastrointestinal protective effect. However, the effects of RBF on 5-FU-induced intestinal mucositis remain unknown. Thus, this study investigated the effects and mechanism of RBF on 5-FU-induced intestinal mucositis in a CT-26 xenograft mice colorectal cancer (CRC) model. Methods: The composition of the RBF preparation was analyzed by high-pressure liquid chromatography. CT-26 cells bearing mice were intraperitoneally administered 150 mg/kg of 5-FU on day 1 to construct a 5-FU-induced intestinal injury model. Mice in the 5-FU + RBF group were intragastrically administrated with RBF (12.4 g/kg) for 4 days. Tumor volume and weight were determined, body weight and diarrhea scores were monitored, leukocytes were calculated, and hematoxylin-eosin staining, immunohistochemistry, and terminal deoxynucleotidyl transferase dUTP nick end labeling were performed to investigate the therapeutic efficiency and underlying mechanisms of RBF on 5-FU-induced intestinal mucositis mice. Results: RBF didn`t affect the effect of tumor suppression, while attenuated diarrhea associated with 5-FU-induced intestinal mucositis and significantly reduced leucocyte toxicity. Additionally, RBF significantly promoted cell proliferation and inhibited cell apoptosis, and up-regulated CDK4, c-Myc, and B-cell lymphoma-2 expression, while down-regulated Bax expression, as well as enhanced both p-AKT and AKT expression in intestinal crypts of 5-FU-treated mice. Conclusions: RBF attenuated 5-FU-induced intestinal mucositis by inhibiting apoptosis and promoting cell proliferation by modulating the AKT pathway and its downstream effectors in a CT-26 xenograft mouse CRC model.

The trade-off between environment and economy has been one of the hot topics discussed in academia, but there are many disputes among them. Based on the micro data of Chinese listed companies from 2016 to 2020, this paper uses the shock of the Environmental Protection Tax Law (EPTL) in 2018 as a quasi-natural experiment. Our Differences-in-Differences (DID) method is used to study the impact of the environmental governance pressure caused by the EPTL on firms’ total factor productivity (TFP). The conclusions suggest that: 1) The EPTL has significantly improved heavily polluting enterprises’ TFP, and this finding remains stable after a range of robustness checks. 2) The subgroup analysis shows that. The smaller the size of the administrative expenses paid by the enterprises, the larger the effect of the EPTL on firms’ TFP; the positive effect of the EPTL on the TFP of heavily polluting enterprises is not pronounced in the sample of small-scale enterprises; when the degree of financial slack is small, the effect of the EPTL on firms’ TFP is greater; the effect of the EPTL is greater when the enterprises are located in provincial capital and municipalities. 3) Meanwhile, we also find that promoting firm value and stimulating green innovation may be the potential mechanisms of the EPTL affecting firms’ TFP. This paper provides experience summary for the firms’ high-quality development under the background of government environmental governance.

With the increase in cadmium (Cd) release into the environment, it is necessary to find appropriate solutions to reduce soil Cd pollution. Microorganisms are a green and effective means for the remediation of Cd-contaminated soil. In this study, in a Cd-contaminated farmland, we screened and identified novel Cd-resistant strains, Paenarthrobactor nitroguajacolicus, Lysinibacillus fusiformis, Bacillus licheniformis, and Methyllobacium brachiatum, with minimum inhibitory concentrations of 100, 100, 50, and 50 mg/L, respectively, and added them each to pots containing Cd-contaminated rape plants to explore their remediation ability. The results showed that treatment with each of the four strains significantly increased the abundance of Nitrospirae, Firmicutes, Verrucomicrobia, and Patescibacterium in the rhizosphere soil of the plants. This led to changes in soil physical and chemical indices; pH; and available phosphorus, urease, and catalase activities, which were significantly negatively correlated with bioavailable Cd, reducing 28.74–58.82 % Cd enrichment to plants and 23.72–43.79 % Cd transport within plants, and reducing 5.52–10.68 % available cadmium in soil, effectively reducing the biotoxicity of Cd. Thus, this study suggests microbial remediation as a reliable option, forming a basis for the remediation of Cd-contaminated soil.

Protein, fat, shear force, color, and chewiness were the primary factors to evaluate the characteristics of cooked marinated meat products. Overall contents of protein and fat of cooked marinated meat products in black pigs (144.0 g/100 g) were higher than that of white pigs (131.3 g/100 g). The tenderness of cooked marinated meat products of black pigs was higher than white pigs. Total content and number of flavour components of cooked marinated meat products from black pigs were higher than those of white pigs. The flavour contents were 11 163.33 and 3 478.79 μg/g of cooked marinated meat products in black and white pigs, respectively. 2-Pentyl-furan, nonanal, octanal, hexanal, eucalyptol, hexanoic acid, 1-octen-3-ol, 2-octen-1-ol were the most prevalent odor-active compounds of cooked marinated meat products. The cooked marinated meat products from black pigs were associated with aldehydes, ketones, alcohol, esters, acid, pyrrole, and hydroxylamine. The cooked marinated meat products from white pigs were mainly associated with alcohol, ester, acid, phenol, furfural and alkane. Theoretical basis and guidance from these findings support the use of modern meat science and technology for enhancing the quality of traditional Chinese meat products in large-scale production. This would facilitate the global trade of these products. Meanwhile, this study, for the first time, provided some profound insights into the industrial production and flavour control of traditional cooked marinated meat products.

High temperatures are considered one of the most significant limitations to subtropical fishery production. Largemouth bass (Micropterus salmoides) is an economically important freshwater species grown in subtropical areas, which are extremely sensitive to heat stress (HS). However, comprehensive transcriptomic data for the livers of largemouth bass in response to HS are still lacking. In this study, a comparative transcriptomic analysis was performed to investigate the gene expression profiles of the livers of largemouth bass under HS treatment. As a result, 6114 significantly differentially expressed genes (DEGs), which included 2645 up-regulated and 3469 down-regulated genes, were identified in response to HS. Bioinformatics analyses demonstrated that the ‘ECM-receptor interaction’ pathway was one of the most dramatically changed pathways in response to HS, and eight DEGs assigned to this pathway were taken as hub genes. Furthermore, the expression of these eight hub genes was determined by quantitative reverse transcription PCR, and all of them showed a significant change at the transcriptional level, suggesting a crucial role of the ‘ECM-receptor interaction’ pathway in the response of largemouth bass to HS. These findings may improve our understanding of the molecular mechanisms underlying the response of largemouth bass to HS.

In order to reduce the use of fungicide and ensure food safety, it is necessary to develop fungicide with low toxicity and high efficiency to reduce residues. Azoxystrobin (AZOX), which is derived from mushrooms, is an excellent choice. However, conventional AZOX release is difficult to regulate. In this paper, a pH-responsive fungicide delivery system for the preparation of AZOX by impregnation method was reported. The Zinc metal–organic framework/Biomass charcoal (ZIF-8/BC) support was first prepared, and subsequently, the AZOX-ZIF-8/BC nano fungicide was prepared by adsorption of AZOX onto ZIF-8/BC by dipping. Gray mold, caused by Botrytis cinerea, is one of the most important crop diseases worldwide. AZOX-ZIF-8/BC could respond to oxalic acid produced by Botrytis cinerea to release loaded AZOX. When pH = 4.8, it was 48.42% faster than when pH = 8.2. The loading of AZOX on ZIF-8/BC was 19.83%. In vitro and pot experiments showed that AZOX-ZIF-8/BC had significant fungicidal activity, and 300 mg/L concentration of AZOX-ZIF-8-BC could be considered as a safe and effective control of Botrytis cinerea. The above results indicated that the prepared AZOX-ZIF-8/BC not only exhibited good drug efficacy but also demonstrated pH-responsive fungicide release.

Abstract CAR T-cell therapy is well tolerated and effective in patients with Hodgkin lymphoma (HL) and anaplastic large cell lymphoma (ALCL). However, even second- generation anti-CD30 CAR T-cells with CD28 (28z) costimulatory domains failed to achieve the desired rate of complete responses. In the present study, we developed second-generation (CD28z) and third-generation (CD28BBz) CAR T-cells targeting CD30 and investigated their efficacy in vitro and in vivo. Both of CD28z and CD28BBz anti-CD30 CAR T cells were similar regarding amplification, T cell subsets distribution, T cell activity, effector/memory and exhaustion. However, we found that the 28BBz anti-CD30 CAR T-cells persist long-term, specifically homing to the tumor and mediating powerful antitumor activity in tumor xenograft models. Subsequently, we also demonstrated that the third generation anti-CD30 CAR T-cells have miner side effects or potential risks of tumorigenesis. Thus, anti-CD30 CAR T-cells represent a safe and effective treatment for Hodgkin lymphoma.

Background: Neferine exhibits therapeutic effects on anti-hypertension. However, the effect of neferine on hypertensive vascular remodeling remains unexplored. Therefore, the current study was to investigate the effect of neferine on hypertensive vascular remodeling and its underlying mechanisms. Methods: Total 30 male spontaneously hypertensive rats (SHRs) were divided randomly into five groups, including SHR, Neferine-L (2.5 mg/kg/day), Neferine-M (5 mg/kg/day), Neferine-H (10 mg/kg/day), and Valsartan (10 mg/kg/day) groups (n = 6 for each group). Wistar Kyoto (WKY) rats were set as control group (n = 6). Noninvasive blood pressure system, ultrasound, hematoxylin and eosin staining, masson trichrome staining were used to detect the blood pressure, pulse wave velocity (PWV), pathological changes and collagen content in abdominal aortas of SHRs. RNA-sequencing and immunohistochemistry(IHC) analyses were used to identify and verify the differentially expressed transcripts and activation of associated signaling pathways in SHRs. Results: Various concentrations of neferine or valsartan treatment substantially reduced the elevation of blood pressure, PWV, and abdominal aortic thickening of SHRs. RNA-sequencing and KEGG analyses recognized 441 differentially expressed transcripts and several enriched pathways (including PI3K/AKT and TGF-β/Smad2/3 signaling pathways) after neferine treatment. Masson trichromatic staining and IHC analysis demonstrated that neferine treatment decreased the collagen content and down-regulated the protein expression of PCNA, collagen I & III, and fibronectin, as well as p-PI3K, p-AKT, TGF-β1 and p-Smad2/3 in abdominal aortic tissues of SHRs. Conclusion: Neferine treatment exhibits therapeutic effects on anti-hypertension and reduces vascular remodeling, as well as suppresses the abnormal activation of multiple signaling pathways, including PI3K/AKT and TGF-β1/Smad2/3 pathways.

Background: Gastrodin has been widely used clinically in China as an antihypertensive drug. However, its effect on hypertensive renal injury is yet to be elucidated. The current study aimed to investigate the effects of gastrodin on hypertensive renal injury and its underlying mechanisms by network pharmacology analysis and validation in vivo and in vitro.Methods: A total of 10 spontaneously hypertensive rats (SHRs) were randomly categorized into the following two groups: SHR and SHR + Gastrodin groups. Wistar Kyoto (WKY) rats were used as the control group (n = 5). The SHR + Gastrodin group was intragastrically administered gastrodin (3.5 mg/kg/day), and the rats in both WKY and SHR groups were intragastrically administered an equal amount of double-distilled water for 10 weeks. Hematoxylin-eosin, Masson’s trichrome, and Sirius red staining were used to detect the pathological changes and collagen content in the renal tissues. Network pharmacology analysis was performed to explore its potential targets and related pathways. In vitro, the CCK-8 assay was used to determine the cell viability. Immunohistochemistry and western-blotting analyses were employed to assess the protein expression associated with renal fibrosis and transforming growth factor-β1 (TGF-β1) pathway-related proteins in the renal tissues or in TGF-β1-stimulated rat kidney fibroblast cell lines (NRK-49F).Results: Gastrodin treatment attenuates renal injury and pathological alterations in SHRs, including glomerular sclerosis and atrophy, epithelial cell atrophy, and tubular dilation. Gastrodin also reduced the accumulation of collagen in the renal tissues of SHRs, which were confirmed by downregulation of α-SMA, collagen I, collagen III protein expression. Network pharmacology analysis identified TGFB1 and SMAD2 as two of lead candidate targets of gastrodin on against hypertensive renal injury. Consistently, gastrodin treatment downregulated the increase of the protein expression of TGF-β1, and ratios of both p-Smad2/Smad2 and p-Samd3/Smad3 in renal tissues of SHRs. In vitro, gastrodin (25–100 μM) treatment significantly reversed the upregulation of α-SMA, fibronectin, collagen I, as well as p-Smad2 and p-Smad3 protein expressions without affecting the cell viability of TGF-β1 stimulated NRK-49F cells.Conclusion: Gastrodin treatment significantly attenuates hypertensive renal injury and renal fibrosis and suppresses TGF-β1/Smad2/3 signaling in vivo and in vitro.

Lung cancer seriously threatens human health. To explore the molecular mechanism of 20(S)-Protopanaxadiol (PPD) on human non-small cell lung cancer cells, we investigated the transcriptional profile of PPD-treated NCI-H1299 cells. Cell proliferation, cell cycle, and apoptosis were detected using cell counting kit-8 and flow cytometry, respectively. Differentially expressed genes (DEGs) between PPD-treated and untreated cells were determined using RNA sequencing and bioinformatic analysis. Protein phosphorylation was detected using Western blotting. Data of mRNA expression profiles of lung cancer were from The Cancer Genome Atlas (TCGA) and analyzed using R software version 4.3.1. PPD showed an inhibitory effect on the proliferation of NCI-H1299 cells and induced apoptosis. There were 938 upregulated genes and 466 downregulated genes in PPD-treated cells, and DEGs were primarily enriched in the MAPK signaling pathway. The detection of phosphorylation revealed that the phosphorylation of ERK and p38 MAPK was significantly reduced in PPD-treated cells. Further comparison of PPD-regulated DEGs with clinical data of lung adenocarcinoma demonstrated that most downregulated genes in tumor tissues were upregulated in PPD-treated cells or vice versa. Two PPD-downregulated genes HSPA2 and EFNA2 were associated with patients’ overall survival. Therefore, PPD could inhibit NCI-H1299 cells by affecting gene expression and regulating ERK and p38 MAPK pathways.

With the rapid development of artificial intelligence and deep learning technologies, their applications in the field of agriculture, particularly in plant disease detection, have become increasingly extensive. This study focuses on the high-precision detection of tomato diseases, which is of paramount importance for agricultural economic benefits and food safety. To achieve this aim, a tomato disease image dataset was first constructed, and a NanoSegmenter model based on the Transformer structure was proposed. Additionally, lightweight technologies, such as the inverted bottleneck technique, quantization, and sparse attention mechanism, were introduced to optimize the model’s performance and computational efficiency. The experimental results demonstrated excellent performance of the model in tomato disease detection tasks, achieving a precision of 0.98, a recall of 0.97, and an mIoU of 0.95, while the computational efficiency reached an inference speed of 37 FPS. In summary, this study provides an effective solution for high-precision detection of tomato diseases and offers insights and references for future research.

Abstract This paper introduces a novel cascading frequency regulation scheme (CFRS) for VSC‐HVDC to provide strong support for the connected weak AC grid without phase‐locked loop and remote‐communication considering the cost‐effective property. The proposed CFRS utilises a DC voltage synchronisation control to enable the self‐synchronisation of VSC‐HVDC without phase‐locked loop, while the corresponding inertia response can be exerted via utilising the stored HVDC capacitor energy. Meanwhile, the DC voltage acts as the reflection of system frequency, enabling the coordination between two‐end VSC stations without communication. More importantly, to fully utilise the VSC‐HVDC potentials, the CFRS that sequentially activates HVDC capacitor and primary frequency control is implemented so that the energy loss and the detrimental control impacts on the sending‐end systems can be minimised while making sure the requirement of system frequency support. Analytical derivations have been done to evaluate the contribution of the proposed CFRS to system frequency regulation. Furthermore, the effectiveness of the proposed scheme has been well validated in PSCAD/EMTDC under several power system contingencies by fully comparing with existing control schemes. The proposed CFRS stands out by the reduced complexity of the control structure, the robustness for connecting the very weak AC grid and the fast frequency regulation ability with consideration of cost‐effective property.

Abstract Background Colorectal cancer (CRC) is one of the most highly malignant tumors and has a complicated pathogenesis. A preliminary study identified syntrophin beta 1 (SNTB1) as a potential oncogene in CRC. However, the clinical significance, biological function, and underlying mechanisms of SNTB1 in CRC remain largely unknown. Thus, the present study aimed to investigate the role of SNTB1 in CRC. Methods The expression profile of SNTB1 in CRC samples was evaluated by database analysis, cDNA array, tissue microarray, quantitative real-time PCR (qPCR), and immunohistochemistry. SNTB1 expression in human CRC cells was silenced using short hairpin RNAs (shRNA)/small interfering RNAs (siRNA) and its mRNA and protein levels were assessed by qPCR and/or western blotting. Cell viability, survival, cell cycle, and apoptosis were determined by the CCK-8 assay, colony formation, and flow cytometry assays, respectively. A xenograft nude mouse model of CRC was established to validate the roles of SNTB1 in vivo. Immunohistochemistry and TUNEL staining were used to determine the expression of SNTB1, PCNA, and cell apoptosis in tissue samples. Isobaric tag for relative and absolute quantification (iTRAQ) was used to analyze the differentially expressed proteins after knockdown of SNTB1 in CRC cells. Silence of protein kinase N2 (PKN2) using si-PNK2 was performed for rescue experiments. Results SNTB1 expression was increased in CRC tissues compared with adjacent noncancerous tissues and the increased SNTB1 expression was associated with shorter overall survival of CRC patients. Silencing of SNTB1 suppressed cell viability and survival, induced cell cycle arrest and apoptosis in vitro, and inhibited the growth of CRC cells in vivo. Further elucidation of the regulation of STNB1 on CRC growth by iTRAQ analysis identified 210 up-regulated and 55 down-regulated proteins in CRC cells after SNTB knockdown. A PPI network analysis identified PKN2 as a hub protein and was up-regulated in CRC cells after SNTB1 knockdown. Western-blot analysis further confirmed that SNTB1 knockdown significantly up-regulated PKN2 protein expression in CRC cells and decreased the phosphorylation of both ERK1/2 and AKT. Moreover, rescue experiments indicated that PKN2 knockdown significantly rescued SNTB1 knockdown-mediated decrease in cell viability, survival, and increase of cell cycle arrest at G0/G1 phase and apoptosis of CRC cells. Conclusions These findings indicate that SNTB1 is overexpressed in CRC. Elevated SNTB1 levels are correlated with shorter patient survival. Importantly, SNTB1 promotes tumor growth and progression of CRC, possibly by reducing the expression of PKN2 and activating the ERK and AKT signaling pathway. Our study highlights the potential of SNTB1 as a new prognostic factor and therapeutic target for CRC.

Rapid urban development, increasing urban population, and drastic land-use changes may cause deterioration of ecosystem health and pose a serious threat to sustainable development. In this study, the analytic hierarchy process-driving-pressure-state-response (AHP-DPSR) model was used to evaluate the ecosystem health of the Liaohe River Basin in Jilin Province, China. The DPSR model was used to select indicators, and 29 indicators were selected for the ecosystem health evaluation index system, including four aspects of driving, pressure, state, and response. At the same time, the weights of each index were determined using AHP, and the spatiotemporal change pattern of ecosystem health in the past 15 years was assessed. The results of the ecosystem health assessment in the study area were spatially spread using ArcGIS software. Finally, the influence of climate and human activities on ecosystem health status was explored. Spatial autocorrelation analysis of the ecosystem health structure was conducted to provide suggestions for the subsequent improvement of ecosystem health quality and sustainable development in the region. The results showed the following: (1) From 2005 to 2020, the health level of the ecosystem in the study area generally showed a trend of improvement. The percentages of ecosystem health in the study area, from high to low in 2020 were 26%, 24%, 31%, 12%, and 7%. (2) The regions with high spatial autocorrelation in 2020 were Gongzhuling City, Dongliao County, Xi'an District, and Longshan District. These areas can be prioritized for ecological management of the region. (3) The spatial and temporal variation in ecosystem health in the Liaohe River Basin of Jilin Province is significantly influenced by precipitation, temperature, and GDP in the region. The conclusions of this study can provide policy references and scientific guidance for the ecological environmental protection and sustainable development of the Liaohe River Basin in Jilin Province.

Currently commercialized CAR-T cell therapies targeting CD19 and BCMA show great efficacy to cure B cell malignancies. However, intravenous infusion of these CAR-T cells severely destroys both transformed and normal B cells in most tissues and organs, in particular lung, leading to a critical question that what the impact of normal B cell depletion on pulmonary diseases and lung cancer is. Herein, we find that B cell frequency is remarkably reduced in both smoking carcinogen-treated lung tissues and lung tumors, which is associated with advanced cancer progression and worse patient survival. B cell depletion by anti-CD20 antibody significantly accelerates the initiation and progression of lung tumors, which is mediated by repressed tumor infiltration of T cells and macrophage elimination of tumor cells. These findings unveil the overall antitumor activity of B cells in lung cancer, providing novel insights into both mechanisms underlying lung cancer pathogenesis and clinical prevention post CAR-T cell therapy.

This study aimed to investigate the relationship among physicochemical quality, water status and protein degradation of pork samples during superheated steam (SHS) cooking. Pork samples were cooked with SHS (120, 150 and 180 ℃ and traditional steam (TS) to 40, 60 and 80 ℃. The results showed that SHS cooking at 150 and 180 ℃ significantly reduced the values of lightness (L*), yellowness (b*), cooking loss and increased the value of redness (a*). Moreover, SHS cooked samples had lower shear force, hardness and chewiness value than TS cooked samples, indicating a better mouth feel quality. Low-field nuclear magnetic resonance (LF-NMR) analysis results showed that relaxation time T21, T22 and T23 increased with SHS temperature, T2 (TS-cooked) < T2 (SHS-cooked), SHS had higher P22 values but lower P23 values than TS. The secondary structure of pork protein cooked by TS tends to be loose than SHS, promoting more immobilized water into free water. Furthermore, SHS led to a low exposure of hydrogen bonds and hydrophobic bonds which reduced protein aggregation. The protein degradation and water status could explain the quality differences between SHS and TS cooked pork.

Chrysanthemum (Chrysanthemum morifolium) is an important ornamental and medicinal plant suffering from many viruses and viroids worldwide. In this study, a new carlavirus, tentatively named Chinese isolate of Carya illinoinensis carlavirus 1 (CiCV1-CN), was identified from chrysanthemum plants in Zhejiang Province, China. The genome sequence of CiCV1-CN was 8795 nucleotides (nt) in length, with a 68-nt 5′-untranslated region (UTR) and a 76-nt 3′-UTR, which contained six predicted open reading frames (ORFs) that encode six corresponding proteins of various sizes. Phylogenetic analyses based on full-length genome and coat protein sequences revealed that CiCV1-CN is in an evolutionary branch with chrysanthemum virus R (CVR) in the Carlavirus genus. Pairwise sequence identity analysis showed that, except for CiCV1, CiCV1-CN has the highest whole-genome sequence identity of 71.3% to CVR-X6. At the amino acid level, the highest identities of predicted proteins encoded by the ORF1, ORF2, ORF3, ORF4, ORF5, and ORF6 of CiCV1-CN were 77.1% in the CVR-X21 ORF1, 80.3% in the CVR-X13 ORF2, 74.8% in the CVR-X21 ORF3, 60.9% in the CVR-BJ ORF4, 90.2% in the CVR-X6 and CVR-TX ORF5s, and 79.4% in the CVR-X21 ORF6. Furthermore, we also found a transient expression of the cysteine-rich protein (CRP) encoded by the ORF6 of CiCV1-CN in Nicotiana benthamiana plants using a potato virus X-based vector, which can result in a downward leaf curl and hypersensitive cell death over the time course. These results demonstrated that CiCV1-CN is a pathogenic virus and C. morifolium is a natural host of CiCV1.

In this work, TiO2-MXene/poly (3,4-ethylenedioxythiophene): poly(styrenesulfonate) (PEDOT:PSS) composite was utilized as an electrode material for the sensitive electrochemical detection of baicalein. The in-situ growth of TiO2 nanoparticles on the surface of MXene nanosheets can effectively prevent their aggregation, thus presenting a significantly large specific surface area and abundant active sites. However, the partial oxidation of MXene after calcination could reduce its conductivity. To address this issue, herein, PEDOT:PSS films were introduced to disperse the TiO2-MXene materials. The uniform and dense films of PEDOT:PSS not only improved the conductivity and dispersion of TiO2-MXene but also enhanced its stability and electrocatalytic activity. With the advantages of a composite material, TiO2-MXene/PEDOT:PSS as an electrode material demonstrated excellent electrochemical sensing ability for baicalein determination, with a wide linear response ranging from 0.007 to 10.0 μM and a lower limit of detection of 2.33 nM. Furthermore, the prepared sensor displayed good repeatability, reproducibility, stability and selectivity, and presented satisfactory results for the determination of baicalein in human urine sample analysis.

Abstract Traditional permanent magnet synchronous generator (PMSG)‐based wind turbine (WT) normally utilises phase lock loop (PLL) for obtaining accurate phase angle of AC grid to make sure of maximum power point tracking (MPPT). However, a weak AC system with a low short circuit ratio (SCR) and high control bandwidth of PLL might have impaired effects on the stability of the system. To overcome these, a novel power synchronous control of PMSG‐based WT with grid forming ability of PMSG for integrating very weak AC system is first proposed in this study. Grid‐side converter (GSC) of PMSG emulates the inertia response of synchronous generator (SG) by artificially coupling the DC‐link voltage with the grid frequency based on the certain droop property, while the converter voltage magnitude is regulated according to DC‐link voltage deviation by controlling the modulation index of GSC. This novel scheme can realise the grid synchronisation without PLL by mimicking the natural characteristic of SG. Small‐signal analysis of the proposed scheme indicates the DC‐link capacitor energy can be exerted for system inertia support during system disturbances, while the system damping can be provided by spontaneously alternating converter voltage magnitude. More importantly, MPPT property of PMSG is not compromised while providing inertia and damping for the system, and the related inertia and damping coefficient of WT with the proposed control can be easily tuned for satisfying the system requirement. Non‐linear simulations of one PMSG connected with one large SG considering a sudden change of active power reference and variable wind speeds have been studied to verify the effectiveness of the proposed grid‐forming control.

Abstract Soybean is a major crop that provides oil and protein worldwide. Soybean products have been consumed for centuries in many different kinds of food. The regulatory network that controls the accumulation of fatty acids (FAs) and storage proteins (SSPs) in seeds is poorly understood. To gain new insights into the molecular mechanisms that contribute to high‐quality seeds, seed FA and SSP contents were analyzed for the effects of chromosome segment substitution in a wild soybean line (CSSL). High‐throughput transcriptomics and tandem mass tag (TMT)‐based quantitative proteomics were performed on the dry seeds of the CSSL and parent lines. In total, 665 differentially expressed genes (DEGs) and 83 differentially accumulated proteins (DAPs) were identified. Of these 27 DEGs and 23 DAPs were found to regulate the seed storage profile. These genes encode proteins involved in photosynthesis, protein processing, protein sorting, and storage protein accumulation. Data are presented showing that FA synthesis was decreased by the regulation of SSP‐accumulation‐related genes and proteins. Taken together, the results provide new insights into the regulation network involved in the accumulation of seed storage compounds involving both source and sink processes that may be used as markers in future soybean breeding programs.

It is well known that integer-order neural networks with diffusion have rich spatial and temporal dynamical behaviors, including Turing pattern and Hopf bifurcation. Recently, some studies indicate that fractional calculus can depict the memory and hereditary attributes of neural networks more accurately. In this paper, we mainly investigate the Turing pattern in a delayed reaction–diffusion neural network with Caputo-type fractional derivative. In particular, we find that this fractional neural network can form steadily spatial patterns even if its first-derivative counterpart cannot develop any steady pattern, which implies that temporal fractional derivative contributes to pattern formation. Numerical simulations show that both fractional derivative and time delay have influence on the shape of Turing patterns.

Yeasts are one of the predominant microbial groups in fermented meats. In this study, yeast communities of Chinese Dong fermented pork (Nanx Wudl) were investigated and the technological properties of 73 yeast isolates were evaluated. Through culture-dependent and high-throughput sequencing methods, the main yeast species identified included Pichia membranifaciens, Kazachstania bulderi, Millerozyma farinosa, Candida zeylanoides, Kazachstania exigua, Candida parapsilosis and Saccharomyces cerevisiae. Among these yeasts, P. membranifaciens, M. farinosa, K. exigua and K. bulderi were detected in fermented meats for the first time. A total of 73 yeast isolates was investigated for their lipolytic and proteolytic activities. All yeast species showed lipolytic activity, while proteolytic activity against meat protein was only detected in S. cerevisiae. Assay of aroma-producing potential was performed in a model simulating fermented sausage condition. Inoculation of yeast strains increased volatiles production, especially esters and alcohols. The highest ester production was observed in S. cerevisiae Y70 strain, followed by K. exigua Y12 and K. bulderi Y19. C. zeylanoides Y10 and S. cerevisiae Y70 were the highest producers of benzeneethanol and 3-methyl-1-butanol. S. cerevisiae Y70 with its highest production of branched alcohols and esters could be a promising candidate as aroma enhancer in the manufacture of fermented sausages.

The effect of a proteolytic starter culture isolated from Nanx Wudl, on microbiological, biochemical and organoleptic attributes of dry fermented sausages was investigated during processing. Based on preliminary screening, the combination of Staphylococcus xylosus SX16 and Lactobacillus plantarum CMRC6, showing excellent proteolytic activity in vitro, was selected as the multi-strain starter (starter LS). For comparison, the single-strain starter culture of L. plantarum CMRC6 (starter LB) and non-inoculated control were also tested. During fermentation, lactic acid bacteria and staphylococci dominated the microbiota and suppressed the Enterobacteriaceae growth in LS-inoculated sausages. The addition of LS starter accelerated acidification and proteolysis during ripening, improving the contents of total free amino acids and several essential free amino acids (Phe, Ile and Leu). Volatile compounds analysis revealed that LS-fermented sausage showed the highest abundance of 3-methyl-1-butanol, probably due to the inoculated S. xylosus. The inoculation of LS starter improved the sensory properties of sausages, especially the flavor attribute. Therefore, S. xylosus SX16 and L. plantarum CMRC6 are promising candidates for inclusion as multi-strain starters in the manufacture of gourmet fermented dry sausage.

The unique processing technology of dry-cured meat products leads to strong proteolysis, which produces numerous peptides. The purpose of this investigation was the systematic isolation, purification, and identification of potentially cardioprotective bioactive peptides from dry-cured pork coppa during processing. According to the results of anti-platelet-activating factor acetyl hydrolase activity and radical scavenging ability in vitro, the inhibitory effect of M1F2 in purified fractions on cardiovascular inflammation was higher than that of M2F2. The peptide of M1F2 was identified by nano-liquid chromatography–tandem mass spectrometry. A total of 30 peptides were identified. Based on bioinformatics methods, including in silico analysis and molecular docking, LTDKPFL, VEAPPAKVP, KVPVPAPK, IPVPKK, and PIKRSP were identified as the most promising potential platelet-activating factor acetyl hydrolase inhibitory peptides. Overall, bioactive peptides produced during dry-cured pork coppa processing demonstrate positive effects on human health.

Multiple new subtypes of breast cancer (BRCA) are identified in women each year, rendering BRCA the most common and rapidly expanding form of cancer in females globally. NUF2 has been identified as a prognostic factor in various human cancers, regulating cell apoptosis and proliferation. However, its role in BRCA prognosis has not been clarified. This study explored the role of NUF2 in breast cancer development and prognosis using informatic analysis combined with in vivo intracellular studies. Through the online website TIMER, we evaluated the transcription profile of NUF2 across a variety of different cancer types and found that NUF2 mRNA was highly expressed in BRCA patients. Its transcription level was found to be related to the subtype, pathological stage, and prognosis of BRCA. The R program analysis showed a correlation of NUF2 with cell proliferation and tumor stemness in the BRCA patient samples. Subsequently, the association between the NUF2 expression level and immune cell infiltration was analyzed using the XIANTAO and TIMER tools. The results revealed that NUF2 expression was correlated with the responses of multiple immune cells. Furthermore, we observed the effect of NUF2 expression on tumor stemness in BRCA cell lines in vivo. The experimental results illuminated that the overexpression of NUF2 statistically upregulated the proliferation and tumor stemness ability of the BRCA cell lines MCF-7 and Hs-578T. Meanwhile, the knockdown of NUF2 inhibited the abilities of both cell lines, a finding which was verified by analyzing the subcutaneous tumorigenic ability in nude mice. In summary, this study suggests that NUF2 may play a key role in the development and progression of BRCA by affecting tumor stemness. As a stemness indicator, it has the potential to be one of the markers for the diagnosis of BRCA.

Background: Qingda granules (QDG) exhibit antihypertension and multiple-target-organ protection. However, the therapeutic potential of QDG on hypertensive renal injury remains unknown. Therefore, the main objective of the current study is to explore the effects and underlying mechanisms of QDG treatment on renal injury in angiotensin (Ang) II-infused mice.Methods and results: Mice were infused with Ang II (500 ng/kg/min) or saline for 4 weeks with subcutaneously implanted osmotic pumps. After infusion, mice in the Ang II + QDG group were intragastrically administrated with QDG daily (1.145 g/kg/day), whereas the control group and Ang II group were intragastrically administrated with the same amount of double-distilled water. Blood pressure of the mice monitored using the CODA™ noninvasive blood pressure system revealed that QDG treatment significantly attenuated elevated blood pressure. Moreover, hematoxylin–eosin staining indicated that QDG treatment ameliorated Ang II-induced renal morphological changes, including glomerular sclerosis and atrophy, epithelial cell atrophy, and tubular dilatation. RNA-sequencing (RNA-seq) identified 662 differentially expressed transcripts (DETs) in renal tissues of Ang II-infused mice, which were reversed after QDG treatment. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis based on DETs in both comparisons of Ang II vs. Control and Ang II + QDG vs. Ang II identified multiple enriched pathways, including apoptosis and p53 pathways. Consistently, terminal deoxynucleotidyl transferase (TdT) dUTP nick-end labeling (TUNEL) staining and Annexin V staining revealed that QDG treatment significantly attenuated Ang II-induced cell apoptosis in renal tissues and cultured renal tubular epithelial cell lines (NRK-52E). Furthermore, western blot analysis indicated that Ang II infusion significantly upregulated the protein expression of p53, BCL2-associated X (BAX), cle-caspase-9, and cle-caspase-3, while downregulating the protein expression of BCL-2 in renal tissues, which were attenuated after QDG treatment.Conclusion: Collectively, QDG treatment significantly attenuated hypertensive renal injury, partially by attenuating renal apoptosis and suppressing p53 pathways, which might be the underlying mechanisms.

Abstract Windows are the least energy efficient part of the buildings, as building accounts for 40% of global energy consumption. Traditional smart windows can only regulate solar transmission, while all the solar energy on the window is wasted. Here, for the first time, the authors demonstrate an energy saving and energy generation integrated smart window (ESEG smart window) in a simple way by combining louver structure solar cell, thermotropic hydrogel, and indium tin oxides (ITO) glass. The ESEG smart window can achieve excellent optical properties with ≈90% luminous transmission and ≈54% solar modulation, which endows excellent energy saving performance. The outstanding photoelectric conversion efficiency (18.24%) of silicon solar cells with louver structure gives the smart window excellent energy generation ability, which is more than 100% higher than previously reported energy generation smart window. In addition, the solar cell can provide electricity to for ITO glass to turn the transmittance of hydrogel actively, as well as the effect of antifreezing. This work offers an insight into the design and preparation together with a disruptive strategy of easy fabrication, good uniformity, and scalability, which opens a new avenue to realize energy storage, energy saving, active control, and antifreezing integration in one device.

We assessed the role of the protein-coding gene chaperonin-containing TCP1 subunit 6A (CCT6A) in osteosarcoma, as this is currently unknown. Using data from the R2 online genomic analysis and visualization application, we found that CCT6A messenger ribonucleic acid (RNA) expression is increased in osteosarcoma tissue and cells. Transfection of CCT6A small interfering RNA into cultured osteosarcoma cells revealed that CCT6A knockdown attenuates cell growth, cell viability, cell survival, and induced apoptosis and cell cycle progression at the G0/G1 phases. Moreover, CCT6A knockdown downregulated phospho-protein kinase B (p-Akt), cyclinD1 and B-cell lymphoma-2, whereas upregulated Bcl-2-associated X-protein expression. Thus, CCT6A knockdown inhibits cell proliferation, induces cell apoptosis, and suppresses the Akt pathway.

This study presents a comprehensive analysis of the impact that a supplementary damping controller of the permanent magnet synchronous generator (PMSG)-based wind turbine (WT) has on low-frequency oscillation (LFO) damping. A reduced mathematical model of an interarea system is first established. Then, an auxiliary damping controller is designed using the PMSG active power control loop, enabling the WT to actively support the interarea LFO mitigation. Based on this, the damping torque analysis (DTA) method is applied to explore the contribution of the PMSG-based WT with an auxiliary damping controller to LFO damping enhancement, whose analytical expressions of the damping torque coefficients reveal the impacts of the control parameters and the operation conditions on the system damping characteristics. It is evident that the installation location of the wind power plant (WPP) plays a leading role in determining whether the damping provided by the PMSG controller is positive or negative. Also, the contribution of damping from the PMSG can be improved by tuning the droop coefficient of the PMSG controller properly. The results indicate that the proposed damping control is always helpful in improving the system damping when the wind power penetration increases. Accordingly, analytical conclusions can serve as guidelines for the control design. Case studies of a two-area test system integrated with a PMSG-based wind farm have been conducted to verify the theoretical analysis.

Due to the poor weldability of Inconel 738 alloy, defects (cracks, pores) always exist in the selective laser melting(SLM) process. In this paper, the molten pool evolution of the single track, including the morphology, porosity and crack formation, was systematically investigated by numerical simulation, and the effects of different preheating temperature and process parameters were verified by experimental analysis. The detailed temperature-dependent thermal physical properties were applied into the Volume of Fluid (VOF) method to simulate a powder bed computational domain which was established by Discrete Element Model (DEM).The simulation results show that the change of molten pool width by different laser power and preheating was not as significant as that of depth. The results also demonstrate that higher energy density and preheating temperature induced the generation of pores at the bottom of molten pool, which can be explained by the variation in cooling rates . Moreover, the change of temperature gradients in three-dimensional directions with the increase of preheating temperature was also investigated, which explained the reason for crack forming and extending through the center line. Finally, the evolution of molten pool at a higher preheating temperature 1000℃ was simulated to further underpin the positive effect of preheating on preventing cracks formation.

Objectives: To investigate the effects of swimming on cancer induced muscle wasting and explore its underlying mechanism in CT-26 bearing mice.Methods: BALB/c mice (n = 16) injected with CT-26 cells were divided into two groups, including Tumor group (n = 8) and Swimming group (n = 8). Another 8 un-injected mice were set as Control group. Mice in Swimming group were subjected to physical training for swimming twice per day for 30 min intervals and 6 days per week for a total of 4 weeks. The tumor volume was monitored every 3 days and tumor weight was measured at the end of experiment. The changes of muscle function, pathological and cell apoptosis of quadriceps muscles were further assessed, and its underlying mechanisms were further explored using multiple biological technologies.Results: Swimming obviously alleviated tumor volume and weight in CT-26 bearing mice. Moreover, swimming attenuated the decrease of muscle tension, autonomic activities, and increase of muscle atrophy, pathological ultrastructure, as well as cell apoptosis of quadriceps muscles in CT-26 bearing mice. Furthermore, swimming significantly down-regulated the protein expression of NF-κB, p-NF-κB, TNF-α, IL-1β, IL-6 and Bax, while up-regulated the expression of Bcl-2. Further differential expressed metabolites (DEMs) analysis identified a total of 76 (in anion mode) and 330 (in cationic mode) DEMs in quadriceps muscles of CT-26 bearing mice after swimming, including taurochenodeoxycholic acid, taurocholic acid, ascorbic acid and eicosapentaenoic acid.Conclusion: Swimming attenuates tumor growth and muscle wasting, and by suppressing the activation of NF-κB signaling pathway mediated inflammation, reducing the level of Bax medicated cell apoptosis, as well as modulating multiple metabolites might be the importantly underlying mechanisms.
