Abstract Background Cyclic-dependent kinase (CDK) 4/6 kinases, as the critical drivers of the cell cycle, are involved in the tumor progression of various malignancies. Pharmacologic inhibitors of CDK4/6 have shown significant clinical prospects in treating hormone receptor-positive and human epidermal growth factor receptor-negative (HR + /HER2-) breast cancer (BC) patients. However, acquired resistance to CDK4/6 inhibitors (CDK4/6i), as a common issue, has developed rapidly. It is of great significance that the identification of novel therapeutic targets facilitates overcoming the CDK4/6i resistance. PARP1, an amplified gene for CDK4/6i-resistant patients, was found to be significantly upregulated during the construction of CDK4/6i-resistant strains. Whether PARP1 drives CDK4/6i resistance in breast cancer is worth further study. Method PARP1 and p-YB-1 protein levels in breast cancer cells and tissues were quantified using Western blot (WB) analysis, immunohistochemical staining (IHC) and immunofluorescence (IF) assays. Bioinformatics analyses of Gene Expression Profiling Interactive Analysis (GEPIA), Genomics of Drug Sensitivity in Cancer (GDSC) and Cancer Cell Line Encyclopedia (CCLE) datasets were applied to explore the relationship between YB-1/PARP1 protein levels and CDK4/6i IC50. Cell Counting Kit-8 (CCK-8) and crystal violet staining assays were performed to evaluate cell proliferation rates and drug killing effects. Flow cytometry assays were conducted to assess apoptosis rates and the G1/S ratio in the cell cycle. An EdU proliferation assay was used to detect the DNA replication ratio after treatment with PARP1 and YB-1 inhibitors. A ChIP assay was performed to assess the interaction of the transcription factor YB-1 and associated DNA regions. A double fluorescein reporter gene assay was designed to assess the influence of WT/S102A/S102E YB-1 on the promoter region of PARP1. Subcutaneous implantation models were applied for in vivo tumor growth evaluations. Results Here, we reported that PARP1 was amplified in breast cancer cells and CDK4/6i-resistant patients, and knockdown or inhibition of PARP1 reversed drug resistance in cell experiments and animal models. In addition, upregulation of transcription factor YB-1 also occurred in CDK4/6i-resistant breast cancer, and YB-1 inhibition can regulate PARP1 expression. p-YB-1 and PARP1 were upregulated when treated with CDK4/6i based on the WB and IF results, and elevated PARP1 and p-YB-1 were almost simultaneously observed during the construction of MCF7AR-resistant strains. Inhibition of YB-1 or PAPR1 can cause decreased DNA replication, G1/S cycle arrest, and increased apoptosis. We initially confirmed that YB-1 can bind to the promoter region of PARP1 through a ChIP assay. Furthermore, we found that YB-1 phosphorylated at S102 was crucial for PARP1 transcription according to the double fluorescein reporter gene assay. The combination therapy of YB-1 inhibitors and CDK4/6i exerted a synergistic antitumor effect in vitro and in vivo. The clinical data suggested that HR + /HER2- patients with low expression of p-YB-1/PARP1 may be sensitive to CDK4/6i in breast cancer. Conclusion These findings indicated that a ‘‘YB-1/PARP1’’ loop conferred resistance to CDK4/6 inhibitors. Furthermore, interrupting the loop can enhance tumor killing in the xenograft tumor model, which provides a promising strategy against drug resistance in breast cancer.

Tulou dwellings in Southeast China have captivated global interest due to their distinctive appearance, sustainable construction materials and technologies, and their defensive and collective housing functions. Despite several being recognized as World Cultural Heritage sites, the vast majority of tulou buildings are undergoing irreversible decline and destruction, necessitating a comprehensive and systematic study. Taking 83 tulou buildings in Raoping County, Chaozhou City, Guangdong Province as the research object, this study reconstructs the historical scenes and systematically reveals the emergence, popularity, and consolidation process of tulou dwellings as integrated defensive and residential buildings for ordinary people by conducting a comprehensive analysis of historical documents and local chronicles. Based on an extensive field investigation, the study systematically analyzes the geospatial distribution and the spatial characteristics of Raoping tulou and its residential unit. The results demonstrate the adaptability and flexibility of tulou dwellings, showcasing their developmental process and revealing the inclusive nature of these traditional residences, as well as the initiative of those who reside within them. The research findings contribute to a more dynamic, comprehensive, and authentic understanding of tulou and Chinese traditional residences, providing valuable references for the preservation and sustainable development of tulou architectural heritage.

Abstract Background The effectiveness of the quadratus lumborum block (QLB) for postoperative pain management depends on the injection pathway used. There is limited research on the block area produced by intramuscular injection of local anesthesia in the quadratus lumborum muscle. This study aimed to determine the cutaneous sensory blockade area produced by an intramuscular quadratus lumborum block (QLBi) at the L2 level. Methods Twenty patients aged 18–60 years with ASA grade I-II and a BMI of 18–30 kg/m2 who were scheduled for open inguinal hernia repair with mesh underwent ultrasound-guided QLBi injection of 20 ml of 0.5% ropivacaine. The cutaneous sensory blockade area was measured by applying a cold stimulus 1 h after the block and then measured every hour after surgery until the sensation returned to normal. The duration of a blockade is defined as the time it takes for all affected areas to fully regain normal sensation following a blockade. Pain scores (numeric rating scale, NRS) were recorded at 2, 4, 8, 12, and 24 h after surgery. Adverse reactions to QLBi were recorded 24 h after surgery. Results All 20 patients had reduced or lost cold sensation areas. The greatest extent of cold sensation reduction occurred at T7 (10%), and the least amount of cold sensation reduction occurred at L3 (10%). The block level covered T8 (20%), T9 (30%), T10 (45%), T11 (90%), T12 (95%), L1 (100%), and L2 (15%). Eighteen patients experienced areas of sensory loss, with the highest range at T11 and the lowest at L2. The duration of the blockade was 8.9 ± 3.8 h, with a maximum of 24 h and a minimum of 5 h. One patient experienced quadriceps weakness after surgery. Conclusion Quadratus lumborum block of intramuscular pathway can produce effective cutaneous sensory blockade, which can be used for postoperative analgesia of indirect inguinal hernia operation, and may also be beneficial to analgesia of other lower abdominal operations. However, the best method needs further confirmation to determine specific anesthesia methods for various operations. Chinese clinical trial registry June 2, 2018; ChiCTR1800016457.

Insects sense intraspecific or interspecific information about the chemical substances in the habitat through the sensitive olfactory system to carry out foraging, mating, oviposition, and other activities. The antennae serve as the primary olfactory organs in insects. The olfactory process involves the participation of many proteins, such as odorant-binding proteins (OBPs) and odorant receptors (ORs), but ORs play a central role in olfactory specificity and sensitivity. The beet webworm, Loxostege sticticalis, is an omnivorous agricultural pest that endangers crops and poses a significant risk to the agricultural and animal husbandry production in northern China. In this study, Illumina sequencing was conducted on the antennal transcriptome of male L. sticticalis trapped by three different sex pheromone blends. A total of 10,320 DEGs were identified, from which 46 candidate olfactory genes were selected for further analysis. These candidate olfactory genes comprise 13 odorant receptors, 6 ionotropic receptors (IRs), 3 gustatory receptors (GRs), 12 odorant-binding proteins, and 13 chemosensory proteins (CSPs). In summary, we analyzed the antennal transcriptome of male L. sticticalis trapped by three different sex pheromone blends and identified several candidate olfactory genes. This discovery offers a foundation for further molecular-level investigations into the olfactory system of L. sticticalis.

With advancements in drone driving technology, drones can now collaborate with trucks to execute tasks. However, existing drone–truck collaborative systems are limited to single-task objectives and lack efficiency in large-scale multi-task scenarios. Enhancing the efficiency of drone–truck cooperative systems necessitates the coordination of drone and truck paths to execute multiple tasks simultaneously. Addressing time conflicts in such scenarios remains a significant challenge. This study proposes an innovative drone–truck collaborative system enabling the concurrent execution of delivery and monitoring tasks within specified time windows. To minimize travel costs, a two-stage greedy genetic algorithm (TGGA) is introduced. The methodology initially separates tasks, processes them in batches, and subsequently recombines them to determine the final route. The simulation results indicate that TGGA outperforms existing heuristic algorithms.

Abstract With the coming of the post‐antibiotic era, there is an increasingly urgent need for safe and efficient antibacterial vaccines. Bacterial outer membrane vesicles (OMVs) have received increased attention recently as a potential subunit vaccine. OMVs are non‐replicative and contain the principle immunogenic bacterial antigen, which circumvents the safety concerns of live‐attenuated vaccines. Here, we developed a novel nano‐vaccine by coating OMVs onto PEGylated nano‐Rehmannia glutinosa polysaccharide (pRL) in a structure consisting of concentric circles, resulting in a more stable vaccine with improved immunogenicity. The immunological function of the pRL‐OMV formulation was evaluated in vivo and in vitro, and the underlying mechanism was studied though transcriptomic analysis. The pRL‐OMV formulation significantly increased dendritic cell (DC) proliferation and cytokine secretion. Efficient phagocytosis of the formulation by DCs was accompanied by DC maturation. Further, the formulation demonstrated superior lymph node targeting, contributing to a potent mixed cellular response and bacterial‐specific antibody response against Bordetella bronchiseptica infection. Specifically, transcriptomic analysis revealed that the immune protection function correlated with T‐cell receptor signalling and Th1/Th2/Th17 differentiation, among other markers of enhanced immunological activity. These findings have implications for the future application of OMV‐coated nano‐carriers in antimicrobial immunotherapy.

Methane is a common intestinal gas that has been linked to a variety of gastrointestinal disorders. In this study, we prepared a LIG-based electrochemical sensor modified with PEDOT:PSS/Au-Pt nanocomposites for high-sensitivity methane detection. LIG’s high porosity, flexibility, and excellent electrical conductivity are beneficial for electrochemical detection. The PEDOT:PSS film is electrodeposited on the electrode surface to amplify the signal further. The synergistic effect of bimetallic nanocomposites can improve the electrochemical catalytic activity and increase the specific surface area and the advantages of active sites. The experimental results show that the LIG sensor has a wide linear detection range (2–500 ppm), a low detection limit (about 0.36 ppm), high sensitivity, and good repeatability. The sensor is easy to manufacture, is low-cost, operates at room temperature, has high sensitivity, has a low detection limit, and is expected to be used for methane breath detection in the future.

Compared with discrete emotion space, image emotion analysis based on dimensional emotion space can more accurately represent fine-grained emotion. Meanwhile, this high-precision representation of emotion requires dimensional emotion prediction methods to sense and capture emotional information in images as accurately and richly as possible. However, the existing methods mainly focus on emotion recognition by extracting the emotional regions where salient objects are located while ignoring the joint influence of objects and background on emotion. Furthermore, in the existing literature, when fusing multi-level features, no consideration has been given to the varying contributions of features from different levels to emotional analysis, which makes it difficult to distinguish valuable and useless features and cannot improve the utilization of effective features. This paper proposes an image emotion prediction network named ARMNet. In ARMNet, a unified affective region extraction method that integrates eye fixation detection and attention detection is proposed to enhance the combined influence of objects and backgrounds. Additionally, the multi-level features are fused with the consideration of their different contributions through an improved channel attention mechanism. In comparison to the existing methods, experiments conducted on the CGnA10766 dataset demonstrate that the performance of valence and arousal, as measured by Mean Squared Error (MSE), Mean Absolute Error (MAE), and Coefficient of Determination (R²), has improved by 4.74%, 3.53%, 3.62%, 1.93%, 6.29%, and 7.23%, respectively. Furthermore, the interpretability of the network is enhanced through the visualization of attention weights corresponding to emotional regions within the images.

Abstract Iron participates in various crucial metabolic processes as an essential cofactor of many enzymes, which are vital to the survival of plants and their pathogens. However, excessive iron is toxic to the cells of plants and pathogens. Iron plays a complex role in the interactions between plants and pathogens. Plants and pathogens have evolved sophisticated mechanisms to modulate iron status at a moderate level for maintaining fitness. Iron competition extensively exists on both sides of plants and pathogens during infection. Plants employ iron withholding, local iron accumulation, or iron deficiency to trigger resistance against pathogens. Pathogens counteract host-derived iron stress or interfere with plant iron homeostasis to ensure virulence during infection. This review focuses on the recent progress in understanding the roles of iron in plant-pathogen interactions and proposes prospects for future studies.

In recent years, 5G has been commercialized around the world in a large scale.However, considering the cost and geographical restrictions, traditional 5G networks still have trouble in providing seamless global communication.To reach the global area coverage, satellite and 5G technology integration has become a new hot topic in the communication industry.In 3GPP, the first release of 5G NTN technical specifications based on transparent payload architecture has been completed.Firstly, the standardization status of 5G NTN network architecture was introduced.Secondly, the shortcomings or issues existing in the current network architecture of 5G NTN were analyzed.Finally, three 5G NTN network architecture enhancement and optimization schemes were proposed, which including IAB-based network architecture, NTN adaptive network architecture and NTN multi-connection network architecture for intersatellite link construction and data shunt.The proposed architectures aim to providing valuable suggestions and references for the subsequent research, standardization and network deployment of 5G NTN.

To detect damage in mechanical structures, acoustic emission (AE) inspection is considered as a powerful tool. Generally, the classical acoustic emission detection method uses a sparse sensor array to identify damage and its location. It often depends on a pre-defined wave velocity and it is difficult to yield a high localization accuracy for complicated structures using this method. In this paper, the passive guided wave phased array method, a dense sensor array method, is studied, aiming to obtain better AE localization accuracy in aluminum thin plates. Specifically, the proposed method uses a cross-shaped phased array enhanced with four additional far-end sensors for AE source localization. The proposed two-step method first calculates the real-time velocity and the polar angle of the AE source using the phased array algorithm, and then solves the location of the AE source with the additional far-end sensor. Both numerical and physical experiments on an aluminum flat panel are carried out to validate the proposed method. It is found that using the cross-shaped guided wave phased array method with enhanced far-end sensors can localize the coordinates of the AE source accurately without knowing the wave velocity in advance. The proposed method is also extended to a stiffened thin-walled structure with high localization accuracy, which validates its AE source localization ability for complicated structures. Finally, the influences of cross-shaped phased array element number and the time window length on the proposed method are discussed in detail.

Epidermal wax is strategically situated at the interface between plants and air; therefore, it plays a key role in plants’ interactions with their surroundings. It is also unstable and susceptible to light intensity. Hosta plants are shade-loving herbs with admirable flowers and leaves. Hosta ‘Halcyon’ and Hosta ensata F. Maek. are two species of Hosta with a glaucous and a glossy appearance, respectively. Light intensity can affect the composition of epicuticular wax on the leaf surface, which influences the leaf color phenotype and ornamental value. In this paper, the crystal micromorphology, content, and components of epicuticular wax on the leaves of two species of Hosta under different light conditions (10%-, 30%-, 50%-, 70%-, and 100%-intensity sunlight, relative light intensity (RLI)) have been studied using pot experiments. The results indicate that the epicuticular wax crystals of H. ‘Halcyon’ and H. ensata are tubular and platelet-like, respectively. The wax crystals of H. ‘Halcyon’ melted and formed a thick crust under 100% RLI, and those of H. ensata melted and formed a thick crust under 70% and 100% RLI conditions. The primary ingredients of the epicuticular wax of the two species of Hosta contained primary alcohols, alkanes, fatty acids, and esters; β-diketones were only detected in H. ‘Halcyon’. The quantity of epicuticular wax of H. ‘Halcyon’ reduced at first and then increased with an RLI increase, achieving its lowest value at 50% RLI, but that of H. ensata declined little by little. The amounts of C28 primary alcohols, C31 alkanes, and C18 fatty acids were significantly higher than those of other carbon atoms in the two genotypes of Hosta. The C31β-diketones content decreased with the increase in light intensity, which caused the white frost phenotype to gradually weaken in H. ‘Halcyon’.

Platycodon grandiflorus set ornamental, edible, and medicinal plant with broad prospects for further application development. However, there are no reports on the YABBY transcription factor in P. grandiflorus. Identification and analysis of the YABBY gene family of P. grandiflorus using bioinformatics means. Six YABBY genes were identified and divided into five subgroups. Transcriptome data and qRT-PCR were used to analyze the expression patterns of YABBY. YABBY genes exhibited organ-specific patterns in expression in P grandiflorus. Upon salt stress and drought induction, P. grandiflorus presented different morphological and physiological changes with some dynamic changes. Under salt treatment, the YABBY gene family was down-regulated; PgYABBY5 was up-regulated in leaves at 24 h. In drought treatment, PgYABBY1, PgYABBY2, and PgYABBY3 were down-regulated to varying degrees, but PgYABBY3 was significantly up-regulated in the roots. PgYABBY5 was up-regulated gradually after being down-regulated. PgYABBY5 was significantly up-regulated in stem and leaf at 48 h. PgYABBY6 was down-regulated at first and then significantly up-regulated. The dynamic changes of salt stress and drought stress can be regarded as the responses of plants to resist damage. During the whole process of salt and drought stress treatment, the protein content of each tissue part of P grandiflorus changed continuously. At the same time, we found that the promoter region of the PgYABBY gene contains stress-resistant elements, and the regulatory role of YABBY transcription factor in the anti-stress mechanism of P grandiflorus remains to be studied. PgYABBY1, PgYABBY2, and PgYABBY5 may be involved in the regulation of saponins in P. grandiflorus. PgYABBY5 may be involved in the drought resistance mechanism in P. grandiflorus stems and leaves. This study may provide a theoretical basis for studying the regulation of terpenoids by the YABBY transcription factor and its resistance to abiotic stress.

Biomedical sensing technology is developing at a tremendous pace and is expected to become an effective clinical tool for the diagnosis and monitoring of human health. The development of sensing devices has successfully transformed the specific sensor prototype designed in the laboratory into a commercially feasible clinical disease detection device. Recently, sensing devices have been accelerated and extended to various fields beyond disease detection, including the measurement of gastrointestinal physiological parameters such as pH, VOC detection, small-molecule gas sensing, and noninvasive screening of oral and lung diseases such as oral cancer, gastric cancer, and other major diseases. In this review, the applications of sensors and electronic nose devices in the diagnosis and monitoring of oral, pulmonary, and gastrointestinal diseases are reviewed, as well as the design and application of sensor materials in disease markers and in situ detection. This article also introduces the practical application of sensing devices in human disease detection, critically analyzes their detection mechanisms and clinical utility, and discusses their future development in medicine. We believe that this review will help readers, especially practitioners in the medical field, provide ideas for the development of sensing devices.

This study aimed to determine the prevalence of bacterial diseases in pig farms in various regions of Zhejiang Province and surrounding areas. A total of 526 samples were collected from 85 pig farms in Zhejiang Province and surrounding areas. In this study, samples were analyzed using bacterial isolation and purification, Gram staining, PCR amplification, and antimicrobial susceptibility testing. A total of 36 Pasteurella multocida (Pm) isolates were detected, with an isolation rate of 6.84%; 37 Bordetella bronchiseptica (Bb) isolates were detected, with an isolation rate of 7.03%; 60 Glasserella parasuis (G. parasuis) isolates were detected, with an isolation rate of 11.41%; 170 Escherichia coli (E. coli) isolates were detected, with an isolation rate of 32.32%; 67 Streptococcus suis (SS) isolates were detected, with an isolation rate of 12.74%; 44 Actinobacillus pleuropneumoniae (APP) isolates were detected, with an isolation rate of 8.37%; and 7 Salmonella enteritis (SE) isolates were detected, with an isolation rate of 1.33%. Antimicrobial drug susceptibility testing against 21 types of antibiotics was carried out on the isolated strains, and the results showed that 228 strains had varying degrees of resistance to 21 antibiotics, including Pm, Bb, E. coli, and APP, with the highest resistance to lincomycin, at 100%. Pm and APP were the most sensitive to cephalothin, with resistance rates of 0. In terms of strains, Pm had the highest overall sensitivity to 21 antibiotics, and E. coli had the highest resistance. In short, bacterial diseases in Zhejiang and the surrounding areas were harmful, and the drug resistance situation was severe. This study provides scientific guidance for the clinical treatment of bacterial diseases.

BackgroundActive-empathic listening (AEL) is the active and emotional involvement of a listener that can take place in at least three key stages of the listening process. Bodie has developed and validated a self-reported, 11-item, three-factor active-empathic listening scale (AELS) in English with good reliability (Cronbach’s alpha = 0.86) to assess AEL abilities. Nevertheless, a Chinese version of the AELS had not been established and validated yet.ObjectiveThe objective of the present study was to examine the reliability and validity of the Chinese version of the AELS.MethodsAfter translating the scale into the Chinese version, 834 college students completed the test. After 4 weeks, 206 participants were tested again on the Chinese AELS to examine retest reliability. The critical ratio method and the item-total correlations were used for the item analysis. Exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) were performed to examine the construct validity. The internal consistency of the scale was analyzed with Cronbach’s alpha and McDonald’s Omega. Interclass correlation coefficient (ICC) was used to examine the scale’s retest reliability. The Interpersonal Reactivity Index (IRI) was used to examine the convergent validity. Pearson correlation analysis was conducted.ResultsEach item of the Chinese AELS had a good discrimination, and the item-total correlation of each item ranged from 0.51 to 0.73. EFA extracted three factors with characteristic root values greater than 1, which could explain 70.72% of the total variance. CFA indicated an adequate fit of the three-factor model (χ2/df = 2.250, root mean square error of approximation [RMSEA] = 0.055, the comparative fit index [CFI] = 0.971, the Tucker-Lewis index [TLI] = 0.959, and the goodness of fit index [GFI] = 0.959). The internal consistency reliability was acceptable (sensing: α = 0.79/ω = 0.78, processing: α = 0.83/ω = 0.83, responding: α = 0.79/ω = 0.79, and AELS: α = 0.87/ω = 0.87). Retest reliability of the scale at 4-week intervals by an ICC was 0.563. The Chinese AELS was significantly correlated with each dimension of IRI.ConclusionThe reliability and validity of the Chinese AELS met the basic psychometrics requirements. Therefore, the scale can be potentially used to assess the active empathic listening abilities of people in China.

Acer ukurunduense refers to a deciduous tree distributed in Northeast Asia and is a widely used landscaping tree species. Although several studies have been conducted on the species’ ecological and economic significance, limited information is available on its phylo-genomics. Our study newly constitutes the complete chloroplast genome of A. ukurunduense into a 156,645-bp circular DNA, which displayed a typical quadripartite structure. In addition, 133 genes were identified, containing 88 protein-coding genes, 37 tRNA genes, and eight rRNA genes. In total, 107 simple sequence repeats and 49 repetitive sequences were observed. Thirty-two codons indicated that biased usages were estimated across 20 protein-coding genes (CDS) in A. ukurunduense. Four hotspot regions (trnK-UUU/rps16, ndhF/rpl32, rpl32/trnL-UAG, and ycf1) were detected among the five analyzed Acer species. Those hotspot regions may be useful molecular markers and contribute to future population genetics studies. The phylogenetic analysis demonstrated that A. ukurunduense is most closely associated with the species of Sect. Palmata. A. ukurunduense and A. pubipetiolatum var. pingpienense diverged in 22.11 Mya. We selected one of the hypervariable regions (trnK-UUU/rps16) to develop a new molecular marker and designed primers and confirmed that the molecular markers could accurately discriminate five Acer species through Sanger sequencing. By sequencing the cp genome of A. ukurunduense and comparing it with the relative species of Acer, we can effectively address the phylogenetic problems of Acer at the species level and provide insights into future research on population genetics and genetic diversity.

Information from multiple sensory modalities interacts. Using functional magnetic resonance imaging (fMRI), we aimed to identify the neural structures correlated with how cooccurring sound modulates the visual motor response execution. The reaction time (RT) to audiovisual stimuli was significantly faster than the RT to visual stimuli. Signal detection analyses showed no significant difference in the perceptual sensitivity (d’) between audiovisual and visual stimuli, while the response criteria (β or c) of the audiovisual stimuli was decreased compared to the visual stimuli. The functional connectivity between the left inferior parietal lobule (IPL) and bilateral superior temporal gyrus (STG) was enhanced in Go processing compared with No-go processing of audiovisual stimuli. Furthermore, the left precentral gyrus (PreCG) showed enhanced functional connectivity with the bilateral STG and other areas of the ventral stream in Go processing compared with No-go processing of audiovisual stimuli. These results revealed that the neuronal network correlated with modulations of the motor response execution after the presentation of both visual stimuli along with cooccurring sound in a multisensory Go/Nogo task, including the left IPL, left PreCG, bilateral STG and some areas of the ventral stream. The role of the interaction between the IPL and STG in transforming audiovisual information into motor behavior is discussed. The current study provides a new perspective for exploring potential brain mechanisms underlying how humans execute appropriate behaviors on the basis of multisensory information.

Evidence of active tuberculosis (ATB) in patients with rheumatic diseases are research priorities but limited data from China have been reported. Research targeting patients not taking anti-TNF biologics are especially insufficient. We aimed to investigate the prevalence and risk factors of ATB in this at-risk population. We conducted a tertiary hospital-based, multi-center, cross-sectional study by using stratified multi-stage cluster sampling strategy to screen ATB in patients with rheumatic diseases. We estimated the prevalence of ATB in patients with rheumatic diseases and identified risk factors among those who were not taking anti-TNF biologic. A total of 13,550 eligible patients were enrolled, and the result showed the standardized prevalence of ATB according to the composition ratio of various types of rheumatic disease was 882/100000 (95% confidence interval (CI): 706-1057). Multivariable logistic regression analysis in patients not taking anti-TNF biologics showed that the independent risk factors of ATB were having systemic lupus erythematosus (SLE) (OR=2.722, 95% CI: 1.437-5.159, p=0.002), having Behcet’s disease (BD) (OR= 5.261, 95% CI: 2.071-13.365, p

The role of NADPH oxidases (NOXs) in pathogenesis and development in the Curvularia leaf spot agent Curvularia lunata remains poorly understood. In this study, we identified C. lunata ClNOX2, which localized to the plasma membrane and was responsible for reactive oxygen species (ROS) generation. Scavenging the ROS production inhibited the conidial germination and appressorial formation. The ClNOX2 and ClBRN1 deletion mutants were defective in 1,8-dihydroxynaphthalene (DHN) melanin accumulation, appressorial formation, and cellulase synthesis and exhibited lower virulence. However, disruption of the ClNOX2 and ClBRN1 genes facilitated hyphal growth, enhanced stress adaptation to cell-wall-disrupting agents, and promoted developmental processes such as conidiation, conidial germination, and pseudothecium and ascus formation. Interestingly, loss of ClM1, the cell wall integrity (CWI) mitogen-activated protein kinase gene in C. lunata, led to morphology and pathogenicity phenotypes similar to ClNOX2 and ClBRN1 deletion mutants such as abnormal conidia, fewer appressoria, less melanin, increased hyphal growth, and enhanced tolerance to Congo red (CR). These results indicated that the ClNOX2 gene plays an important role in C. lunata development and virulence via regulating intracellular DHN melanin biosynthesis. Quantitative reverse-transcription PCR revealed that the ClNOX2-related ROS signaling pathway and ClM1-mediated CWI signaling pathway are cross-linked in regulating DHN melanin biosynthesis. Therapeutic hypothermia (TH) is potentially an important therapy for central nervous system (CNS) trauma. However, its clinical application remains controversial, hampered by two major factors: (1) Many of the CNS injury sites, such as the optic nerve (ON), are deeply buried, preventing access for local TH. The alternative is to apply TH systemically, which significantly limits the applicable temperature range. (2) Even with possible access for ‘local refrigeration’, cold-induced cellular damage offsets the benefit of TH. Here we present a clinically translatable model of traumatic optic neuropathy (TON) by applying clinical trans-nasal endoscopic surgery to goats and non-human primates. This model faithfully recapitulates clinical features of TON such as the injury site (pre-chiasmatic ON), the spatiotemporal pattern of neural degeneration, and the accessibility of local treatments with large operating space. We also developed a computer program to simplify the endoscopic procedure and expand this model to other large animal species. Moreover, applying a cold-protective treatment, inspired by our previous hibernation research, enables us to deliver deep hypothermia (4 °C) locally to mitigate inflammation and metabolic stress (indicated by the transcriptomic changes after injury) without cold-induced cellular damage, and confers prominent neuroprotection both structurally and functionally. Intriguingly, neither treatment alone was effective, demonstrating that in situ deep hypothermia combined with cold protection constitutes a breakthrough for TH as a therapy for TON and other CNS traumas.

An implantation cyst after orbital wall reconstruction may present as a late complication, which can cause local pain, proptosis, diplopia, and vision impairment. Previous surgical strategies prefer transconjunctival or subtarsal approach for cyst drainage, a similar approach to orbital wall reconstruction. These strategies may have risk of secondary infection. Herein, we propose an endoscopic transnasal surgical approach, through which the removal of implant and cyst drainage can be performed conveniently. The residual, medial single-layer cyst wall is generally strong enough to support the orbital soft tissues without the need for a new implant. We believe this surgical approach can simplify the procedure, reduce the complications, and prevent cyst recurrence.

In this study, caffeic acid (CA) and chlorogenic acid (CGA) were incorporated onto chitosan (CS) using free radical grafting initiated by a hydrogen peroxide/ascorbic acid (H2O2/Vc) redox system. The structural properties of the CA (CA-g-CS) and CGA (CGA-g-CS) derivatives were characterized by UV–Vis absorption, Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), nuclear magnetic resonance (NMR), and thermal stability analysis. Then, the antioxidant and antibacterial properties were evaluated, and the effect of CGA-g-CS on the postharvest quality of Saimaiti apricot was studied. It proved that phenolic acids were successfully grafted onto the CS. The grafting ratios of CA-g-CS and CGA-g-CS were 126.21 mg CAE/g and 148.94 mg CGAE/g. The antioxidation and antibacterial activities of CGA-g-CS were better than those of CA-g-CS. The MICs of CGA-g-CS against E. coli, S. aureus, and B. subtilis were 2, 1, and 2 mg/mL. The inhibitory zones of 20 mg/mL CGA-g-CS against the three bacteria were 19.16 ± 0.35, 16.33 ± 0.91, and 16.24 ± 0.05 mm. The inhibitory effects of 0.5% CGA-g-CS on the firmness, weight loss, SSC, TA, relative conductivity, and respiration rate of the apricot were superior. Our results suggest that CGA-g-CS can be potentially used as an edible coating material to preserve apricots.

The purpose of this study was to evaluate the effects of dietary coated lysozyme on growth performance, serum biochemical indexes, antioxidant activity, digestive enzyme activity, intestinal permeability, and the cecal microbiota in weaned piglets. In total, 144 weaned Large White × Landrace piglets were divided into six treatment groups, with 3 replicates and 8 piglets per replicate: CN, a basal diet; CL-L, CL-M, and CL-H, basal diet supplemented with 100, 150, 500 mg/kg coated lysozyme; UL, basal diet supplemented with 150 mg/kg lysozyme; and Abs, basal diet supplemented with 150 mg/kg guitaromycin for 6 weeks. Compared with the CN and UL diets, dietary CL-H inclusion increased the average daily gain (ADG) and decreased the feed/gain (F/G) ratio of piglets (p < 0.05). The addition of 500 mg/kg coated lysozyme to the diet significantly increased the total protein (TP) and globulin (Glob) plasma levels of weaned piglets (p < 0.05). Supplementation with 500 mg/kg coated lysozyme significantly increased the serum IgM concentration and increased lipase activity in the duodenum (p < 0.05). The addition of coated lysozyme and lysozyme significantly decreased the malondialdehyde (MDA) content, while the superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and total antioxidant capacity (T-AOC) levels all increased (p < 0.05). High-throughput sequencing results showed that CL-H treatment effectively improved the intestinal microbiome. The relative abundance of Terrisporobacter in the CL-H and CL-M groups was significantly lower than that in the other groups (p < 0.05). LEfSe analysis results showed that the relative abundance of Coprococcus_3 was higher in the CL-M treatment group. The marker species added to the CL-H treatment group was Anaerofilum. In summary, as a potential substitute for feed antibiotics, lysozyme is directly used as a dietary additive, which is inefficient. Therefore, we used palm oil as the main coating material to coat lysozyme. Lysozyme after coating can more effectively improve the growth performance of piglets by improving the intestinal flora, improving the activity of digestive enzymes, reducing the damage to intestinal permeability and oxidative stress in piglets caused by weaning stress, and improving the immunity of piglets.

Non-orthogonal multiple access (NOMA) cooperative communication technology can combine the advantages of NOMA and cooperative communication, providing high spectrum efficiency and increasing user coverage for next-generation wireless systems. However, the research on NOMA cooperative communication technology is still in a preliminary stage and has mainly concentrated on the scenario of fewer users. This paper focuses on a user-centered NOMA collaboration system in an ultra-dense network, and it constructs a resource allocation optimization problem to meet the demands of each user. Then, this paper decomposes the optimization problem into two subproblems; one is the grouping match among multiple relays and users, and the other is jointly allocating power and subcarrier resources. Accordingly, a dynamic packet matching algorithm based on Gale–Shapley and an iterative algorithm based on the difference of convex functions programing are proposed. Compared with existing schemes, the proposed algorithms can improve system throughput while ensuring the quality of service of users.

Evidence have indicated that RR-type MYB-related transcription factors (TFs) are functionally diverse in regulating floral development, fruit development, leaf senescence, ABA response, and drought and salt responses. Several RR-type MYB-related TFs in Arabidopsis, Antirrhinum and rice are identified and characterized. However, the complete RR-type MYB-related family in tomato has not been studied to date. Here, a genome-wide identification of tomato RR-type MYB-related TFs (SlMYBR) was performed by bioinformatics analysis, and their expression patterns were analyzed. A total of thirteen SlMYBR genes, which were mainly distributed in the head or tail of the chromosome, were identified from tomato and were divided into three groups. Group II was all MYBR genes from eudicots without genes from monocots. For Group I and Group III, the phylogenetic tree was in accord with the evolutionary relationship of these species. SlMYBR proteins were unstable proteins and located in the nucleus. The promoters of SlMYBR contained multiple important cis-acting elements related to abiotic stress or hormone responses. SlMYBR genes had various temporal and spatial expression patterns. Experiments of spraying exogenous hormone demonstrated that the expression of most genes containing hormone response elements was changed, indicating that the expression patterns were associated with the amount of cis-acting elements. The comprehensive investigation of tomato SlMYBR genes in the present study helps to clearly understand the evolution of RR-type MYB-related TFs and provides a useful reference for the further functional study of SlMYBR genes in tomato.

Increased expression of mineral dust-induced gene (mdig, also named as mina53, MINA, or NO52) has been observed in a number of human cancers. The mechanism of how mdig contribute to the pathogenesis of cancer remains to be fully elucidated. In this report, we demonstrated that overexpression of mdig decreased the nuclear staining signal by 4',6-diamidino-2-phenylindole (DAPI), along with a considerable enhancement in cell proliferation. Silencing mdig by shRNA resulted in a statistically significant decrease of cell proliferation. Intriguingly, mdig overexpression reduced the capacity of the cells in migration and invasion in vitro, whereas silencing mdig by shRNA/siRNA enhanced migration and invasion. Clinically, we found that increased expression of mdig in cancer tissues correlates with poorer overall survival of the lung cancer patients, esp., for those without lymph node metastasis. Taken together, our results suggest that mdig plays opposite roles on cell growth and motility, which possibly indicates the paradoxical effect of mdig at the different stages of carcinogenesis.

We present here a case of cough, expectoration and fever for six days. After investigation by the doctor and epidemiologist, it was confirmed that this patient had a history of contact with a novel influenza virus. All the results of the real time PCR on novel Swine Influenza H1 were positive. Blood-gas assay PO250.6 mmHg, hypoxemia and computed tomography (CT) of lungs indicated patchy dense shadow scattered in both lungs in which the inflatable bronchial shadow was observed. A visible change in leakage around the shadow was seen. This was a serious case of infection from a novel influenza virus and the patient received systemic treatment: oseltamivir 75mg bid po, methylprednisolone 40mg qd iv, biapenem 0.6 q12h iv, and moxifloxacin 0.4qd po. On discharge from hospital, Swine influenza H1 was negative. Lungs showed apparent absorption of the inflammation. Respiratory failure had been corrected. Patients infected with a novel influenza virus should be given low-dose hormone and an antiviral drug as soon as possible after the infection is confirmed.
