Abstract Background Low back pain is one of the most common symptoms of osteoporosis. The pain can seriously affect patients’ mood and quality of life; it can also further aggravate bone loss, causing a serious social burden. Minodronate is an oral bisphosphonate that needs to be administered daily. It significantly reduces levels of bone turnover markers (BTMs) and rapidly improves symptoms of low back pain in patients with osteoporosis. Osteoporosis requires long-term treatment, and daily dosing reduces patient compliance. Minodronate has a better safety profile than other bisphosphonates. The objective of the trial is to explore the efficacy and safety of minodronate in the treatment of low back pain in postmenopausal osteoporosis patients. Methods This is a single-centre, randomized, open-label controlled trial with a 24-week duration. Seventy-two eligible patients will be randomly divided into 4 groups. Acute hepatopancreatic necrosis (AHPND) is a bacterial disease caused by Vibrio bacteria that severely affects the Pacific white shrimp farming industry. Since AHPND was first identified in China and Vietnam in 2010, it was subsequently identified in Malaysia (2011), Thailand (2012), Mexico (2013), the Philippines (2014), South Korea (2016), Bangladesh (2017), the United States (US) (2017), and Japan (2020). AHPND causes significant economic losses of more than 7 billion US＄ annually to the global shrimp farming industry. It has severely restricted the development of the global shrimp farming industry. Many studies have shown that the pathogens of AHPND are Vibrio spp., including V. parahaemolyticus, V. owensii, V. campbellii, V. harveyi, V. punensis, and V. anguillarum. The virulence of these pathogenic strains was derived from a 70 kb virulent plasmid (pVA1-type plasmid). The pVA1-type plasmid carries pirAB genes encoding the binary toxin pirAB homologous to the insecticidal protein of Photobacterium phosphoreum. The pVA1-type plasmid has been confirmed to carry a novel trb type Ⅳ secretion system (T4SS). Trb-T4SS can mediate the conjugative transfer of the pVA1-type plasmid at high cell density for interspecific horizontal transfer.The quorum sensing (QS) system is also known as the density-sensing system. In the surrounding environment, the QS system allows bacteria to regulate the expression of multiple genes by sensing changes in the concentration of autoinducer signaling molecules. At high cell density, high concentrations of autoinducer signaling molecules bind to receptor proteins to inhibit phosphorylation cascades and high cell density master regulator OpaR is normally expressed. OpaR is involved in the regulation of various biological processes, such as biofilm-forming ability, motility, and the expression of the type Ⅲ and type Ⅳ secretion systems. Reportedly, high concentrations of signaling molecules in the QS system regulate the expression of key genes of the T4SS to increase the conjugative transfer efficiency of drug-resistant plasmids. However, the regulation of the T4SS by opaR in AHPND is not yet reported.In this study, OpaR, the high cell density master regulator of the QS system in V. parahaemolyticus, was selected to explore the relationship between the QS system and T4SS expression, as well as the conjugative transfer of the pVA1-type plasmid. The V. parahaemolyticus 20130629002S01::cat (Vp2S01::cat) strain, a pathogenic strain of AHPND, was used as the starting strain. The opaR gene was replaced with the erythromycin resistance gene (ermB) by homologous recombination and electroporation. The opaR mutant strain (Vp2S01::catΔopaR) was successfully constructed. The effects of OpaR on Vp2S01::cat were explored by performing growth curve and motility assays.Vp2S01::cat and Vp2S01::catΔopaR were cultured continuously for approximately 24 h in a shaking bed at 28 ℃ and 180 r/min. OD600 was measured every 2 h to compare the difference in growth. The results showed that there were no significant differences in the growth curves for Vp2S01::cat and Vp2S01::catΔopaR. Biofilm-forming ability was detected using a crystal violet staining assay. The results showed that the biofilm-forming ability of Vp2S01::catΔopaR was significantly reduced. Growth zone diameter was recorded in 0.3% (swimming) LB agar plates at 28 ℃ for 8 h to analyze the difference in swimming ability. We found that the swimming ability of Vp2S01::catΔopaR increased significantly by 2.67 times compared with that of Vp2S01::cat. Growth zone diameter was recorded in 1.5% (swarming) LB agar plates at 28 ℃ for 12 h to analyze the difference in swarming ability. The results showed that there was no significant difference in swarming ability between Vp2S01::cat and Vp2S01::catΔopaR. However, Vp2S01::catΔopaR had more missing colonies. Using VcLMB29 as the receptor strain, the conjugative transfer efficiency of Vp2S01::cat and Vp2S01::catΔopaR was compared at different time points. We found that the conjugative transfer efficiency of Vp2S01::catΔopaR increased after 12 and 24 h. The conjugative transfer efficiency after 24 h was increased by a factor of 265.43. RNA of Vp2S01::cat and Vp2S01::catΔopaR was extracted after 24 h of conjugative transfer and reverse-transcribed into cDNA for real-time quantitative fluorescence PCR (RT-qPCR). GyrB was used as the reference gene. AphA-qRT, opaR-qRT, and T4SS-qRT primers were used for RT-qPCR. The relative expression levels of genes were calculated using the 2–ΔΔCt method. The results showed that the relative expression levels of T4SS in the Vp2S01::catΔopaR experimental group were significantly increased by 1.13–3.21 times and that the relative expression levels of the key conjugative transfer genes traF, trbE, and traG were significantly increased by 1.96, 1.92, and 3.21 times, respectively.In conclusion, the opaR gene does not affect the growth and swarming motility but does affect the biofilm-forming and swimming ability of Vp2S01::cat. The high cell density master regulator OpaR may affect the conjugative transfer efficiency of the virulent plasmid by regulating the expression levels of T4SS. This study provides fundamental data for analyzing the mechanism by which the QS system regulates the expression of T4SS and the conjugative transfer of the pVA1-type plasmid in AHPND pathogenic bacteria, offering technical support for the control of the horizontal transfer of the virulent plasmid in AHPND-causing bacteria.

Macrobrachium rosenbergii is one of the most popular species in aquaculture. However, the M. rosenbergii farming industry has been facing an ongoing iron prawn syndrome (IPS) crisis since 2010, resulting in substantial economic losses to the farming industry. Infectious precocity virus (IPV) is a novel virus of Flaviviridae found in recent years that can cause sexual precocity and associated slow growth in healthy M. rosenbergii. It is believed to have a specific correlation with IPS. There are very few cell lines of crustaceans that can be used for studying the response of cells to pathogens. Even though the primary culture technology of blood and muscle cells in M. rosenbergii has gradually matured, there have been few studies on the primary culture of neural cells. Quantitative results of different tissues of IPV-positive M. rosenbergii have indicated that nerve-rich tissues, such as eyestalk, brain, and thoracic ganglion tissues, have a higher viral load, which explains why IPV has neurotropic tissue characteristics. To provide an in vitro cell platform for studying the virus-host interactions of IPV, a simple, stable, and feasible primary culture method was established for the nervous tissue primary cells of M. rosenbergii.In this study, healthy prawns with a body length of about 10–12 cm, healthy appendages, and vitality were selected for the experiments. The body surface of M. rosenbergii was first disinfected with 75% alcohol. On the clean bench, the nervous tissues, including the brain, X organ-sinus gland complex from the eyestalk, thoracic ganglion, and abdominal ganglion tissues, were isolated and washed in PBS buffer containing 100 U/mL penicillin and streptomycin, 100 U/mL amphotericin, and 80 U/mL gentamicin, 2–3 times. The tissues were then placed in 5 mL of 0.5% papain solution. After digestion at 25 ℃ for 5 min, 1× L-15 medium containing 15% FBS, 140 mmol/L D-glucose, and antibiotics (100 U/mL penicillin and streptomycin, 100 U/mL amphotericin, and 80 U/mL gentamicin) were added to terminate digestion. Next, the cells were seeded onto a 24-well plate and allowed to settle in darkness at 25 ℃ for 45 min. After adhesion, cells were transferred to a cell incubator and cultured in the dark at 28 ℃. The nervous tissues' primary cells at different time points were observed under an inverted microscope, and the morphological changes were recorded through imaging. The compound eye tissue of IPV-positive M. rosenbergii was placed in SM buffer, and the IPV mixture was obtained after multiple rounds of crushing and centrifugation. The brain tissue primary cells with large quantities and better culture effects were selected for the IPV challenge after cultured in vitro for 5 days. The virus crude extract was filtered through a 0.22 μm filter membrane and mixed according to the volume ratio of virus crude extract to serum-free medium = 1∶9. For the experimental group, 2 mL of the mixed solution was added to each well, while the control group received 2 mL of serum-free medium per well. Samples were taken at 0, 6, 12, 24, 48, and 96 h after virus infection. RT-qPCR was used to detect the IPV load.The results showed that the cultured primary cells grew well in a 1× L-15 medium containing glutamine and serum. According to the morphology of the cells, number of axons, and other characteristics, brain tissue primary cells, primary cells from the eyestalk X organ-sinus gland complex, and thoracic ganglion tissue primary cells were divided into four types: neurosecretory cells, pseudounipolar neural cell-like, bipolar neural cell-like, and multipolar neural cell-like, respectively. In comparison, only round neural cells and bipolar-like neural cells were found in the primary cell culture of the abdominal ganglion tissue under in vitro culture. The brain tissue primary cells and X organ-sinus gland complex cells survived for 15 days, while the thoracic ganglion and abdominal ganglion tissue primary cells survived for 9 days after being cultured in vitro. After IPV infected the brain tissue primary cells, the viral load of IPV was 15.30 copies/μg RNA at 6 h post-infection (hpi) and 35.59 copies/μg RNA at 12 hpi. IPV was not detected in IPV-infected brain tissue primary cells at 24 hpi and 48 hpi. Then, IPV viral load was detected at 96 hpi, reaching 104 copies/μg RNA.In conclusion, this study successfully established a simple and convenient primary culture technology for cells from M. rosenbergii neural tissues, providing preliminary data and a platform for neuroendocrine and virus-host interactions research. An in vitro infection model of IPV was also initially established in this study. Before establishing the shrimp cell line, the cells could be used to study the mechanism of viral infection, replication, and transcription, providing primary data and a platform for studying the norovirus and neuroendocrine factors of M. rosenbergii. They could also provide critical experimental materials for further research into infection mechanisms and the development of virus and disease prevention technologies.

Abstract Since the beginning of the twenty-first century, in alignment with globalization and the megatrend within the international socio-cultural sphere, China has initiated a large-scale “intangible cultural heritage (ICH) protection movement” through its proactive leadership and management of cultural heritage administration. As a significant component of Chinese cultural politics, this movement has profoundly influenced the cultural lives of Chinese society and its citizens and is poised to continue doing so. Some Chinese anthropologists have also participated in and significantly contributed to this movement in various ways. This paper comprehensively and meticulously examines these fundamental ways. Building upon this analysis, this paper further provides an overview and assessment of the research outcomes of anthropological research on Chinese cultural heritage, both domestically and internationally. The author posits that the paramount contribution of anthropologists, both domestically and internationally, to Chinese cultural heritages and the related protection endeavors lies in their academic research outcomes being predominantly based on their respective fieldwork practices. These outcomes, shaped and realized through on-the-ground fieldwork, hold exceptional value for cultural heritage administration and national cultural policies, as they effectively provide cultural criticism and dissenting perspectives.

Abstract Background According to the 5th edition of the Japanese Guidelines for the Treatment of Gastric Cancer, proximal gastrectomy is recommended for patients with early upper gastric cancer who can retain the distal half of the residual stomach after R0 resection. However, a large number of recent clinical studies suggest that surgical indications for proximal gastrectomy in the guidelines may be too narrow. Therefore, this meta-analysis included patients with early and advanced gastric cancer and compared short- and long-term postoperative outcomes between the two groups. At the same time, we only had high-quality clinical studies such as propensity score-matched studies and randomized controlled trials, which made our research more authentic and credible. Methods Data were retrieved from PubMed, EMBASE, Medline, and Cochrane Library up to June 2023, and included treatment outcomes after proximal gastrectomy with double-tract reconstruction and total gastrectomy with Roux-en-Y reconstruction. The primary results were Early-phase complications(Anastomotic leakage, Anastomotic bleeding, Abdominal abscess, Abdominal infection, Pulmonary infection, Incision infection, Intestinal obstruction, Dumping syndrome, Pancreatic fistula), Late-phase complications(Intestinal obstruction, Anastomosis stricture, Dumping syndrome, Reoperation, Internal hernia, Incidence of endoscopic gastroesophageal reflux), Serious complications (≥ Grade III C-D score), Quality of life[Gastroesophageal reflux symptom evaluation (Visick score)(≥ III), Los Angeles classification(C or D)], Nutritional status(Hemoglobin, Receipt of vitamin B12 supplementation), Oncologic Outcomes(The 5-year overall survival rates). Results After screening, 11 studies were finally included, including 1154 patients. Results from the combined literature showed that total gastrectomy had a significant advantage over proximal gastrectomy with double-tract reconstruction in mean operating time (MD = 4.92, 95% CI: 0.22∼9.61 P = 0.04). Abstract To enhance the capability of identifying unknown emitters in open spaces, an open‐multiscale attention kernel (MSAK)‐convolutional neural network‐long short‐term memory (CNN‐LSTM) structure is proposed. To this end, first, a MSAK module and CNN‐LSTM structure are introduced, and then, the depth and complexity of the feature extraction network are improved to enhance its representation capability. To classify unknown emitters accurately, the MSAK‐CNN‐LSTM model is improved to obtain an open‐MSAK‐CNN‐LSTM model with open‐set recognition capability. Additionally, the two preprocessing procedures are summarised, and their strengths and weaknesses are compared. Experimental results show that the proposed open‐MSAK‐CNN‐LSTM model achieves satisfactory accuracy in identifying unknown emitters in open space. In addition, it has significant advantages in low signal‐to‐noise ratio (SNR) scenarios.

Abstract Background Abnormal heart rate recovery (HRR), representing cardiac autonomic dysfunction, is an important predictor of cardiovascular disease. Prolonged sedentary time (ST) is associated with a slower HRR. However, it is not clear how much moderate-to-vigorous physical activity (MVPA) is required to mitigate the adverse effects of sedentary behavior on HRR in young and middle-aged adults. This study aimed to examine the joint association of ST and MVPA with abnormal HRR in this population. Methods A cross-sectional analysis was conducted on 1253 participants (aged 20–50 years, 67.8% male) from an observational study assessing cardiopulmonary fitness in Fujian Province, China. HRR measured via cardiopulmonary exercise tests on a treadmill was calculated as the difference between heart rate at peak exercise and 2 min after exercise. Abstract Objectives The purpose of this study was to investigate fatigue, mental workload, and burnout among health care workers (HCWs) and explore the possible underlying factors. Materials and methods An online cross-sectional survey design was used to collect data from HCWs in Chongqing, China. The online survey included the Fatigue Severity Scale, NASA Task Load Index, and Chinese version of the Maslach Burnout Inventory-General Survey to assess fatigue, mental workload, and burnout, respectively, and was conducted from February 1 to March 1, 2023. Results In this study, the incidence of fatigue and burnout among HCWs was 76.40% and 89.14%, respectively, and the incidence of moderate to intolerable mental workloads was 90.26%. Work–family conflict, current symptoms, number of days of COVID-19 positivity, mental workload, burnout and reduced personal accomplishment were significantly associated with fatigue. Mental workload was affected by fatigue and reduced personal accomplishment. Furthermore, burnout was influenced by marital status and fatigue. Moreover, there was a correlation among mental workload, fatigue, and burnout. Conclusions Fatigue, mental workload and burnout had a high incidence and were influenced by multiple factors during COVID-19 public emergencies in China.

Abstract Ferroptosis, an iron-dependent regulated cell death mechanism, holds significant promise as a therapeutic strategy in oncology. In the current study, we explored the regulatory effects of epigallocatechin gallate (EGCG), a prominent polyphenol in green tea, on ferroptosis and its potential therapeutic implications for non-small cell lung cancer (NSCLC). Treatment of NSCLC cell lines with varying concentrations of EGCG resulted in a notable suppression of cell proliferation, as evidenced by a reduction in Ki67 immunofluorescence staining. Western blot analyses demonstrated that EGCG treatment led to a decrease in the expression of glutathione peroxidase 4 (GPX4) and solute carrier family 7 member 11 (SLC7A11) while increasing the levels of acyl-CoA synthetase long-chain family member 4 (ACSL4). These molecular changes were accompanied by an increase in intracellular iron, malondialdehyde (MDA), and reactive oxygen species (ROS), alongside ultrastructural alterations characteristic of ferroptosis. Through small RNA sequencing and RT-qPCR, transfer RNA-derived small RNA 13502 (tsRNA-13502) was identified as a significant target of EGCG action, with its expression being upregulated in NSCLC tissues compared to adjacent non-tumorous tissues. EGCG was found to modulate the ferroptosis pathway by downregulating tsRNA-13502 and altering the expression of key ferroptosis regulators (GPX4/SLC7A11 and ACSL4), thereby promoting the accumulation of iron, MDA, and ROS, and ultimately inducing ferroptosis in NSCLC cells. This study elucidates EGCG’s multifaceted mechanisms of action, underscoring the modulation of ferroptosis as a viable therapeutic approach for enhancing NSCLC treatment outcomes.

Abstract H2O dissociation plays a crucial role in solar-driven catalytic CO2 methanation, demanding high temperature even for solar-to-chemical conversion efficiencies

Objective To comprehensively compare the quality and filtration efficiency of medicinal maltose between two domestic manufacturers(B, C) and one foreign manufacturer(A), compare their product quality as an excipient for intravenous immunoglobulin(IVIG), so as to determine the feasibility and substitutability of maltose as an excipient for IVIG from different manufacturers. Methods Quality inspection and comparison of maltose from different manufacturers, small-scale filtration tests, and product quality comparision of IVIG(pH4) were conducted. Results The comparis on results showed that there was no significant difference in the quality of medicinal maltose between manufacturer B and A, and there was no significant difference in the quality of IVIG (pH4) produced. The quality of manufacturer C did not meet the requirements. Conclusion There is no significant difference in the quality of medicinal maltose produced by manufacturer B and A, which can be used for the production of IVIG (pH4).

Abstract Rheumatoid arthritis (RA) is an autoimmune disease. Early studies hold an opinion that gut microbiota is environmentally acquired and associated with RA susceptibility. However, accumulating evidence demonstrates that genetics also shape the gut microbiota. It is known that some strains of inbred laboratory mice are highly susceptible to collagen-induced arthritis (CIA), while the others are resistant to CIA. Here, we show that transplantation of fecal microbiota of CIA-resistant C57BL/6J mice to CIA-susceptible DBA/1J mice confer CIA resistance in DBA/1J mice. C57BL/6J mice and healthy human individuals have enriched B. fragilis than DBA/1J mice and RA patients. Transplantation of B. fragilis prevents CIA in DBA/1J mice. We identify that B. fragilis mainly produces propionate and C57BL/6J mice and healthy human individuals have higher level of propionate. Fibroblast-like synoviocytes (FLSs) in RA are activated to undergo tumor-like transformation. Propionate disrupts HDAC3-FOXK1 interaction to increase acetylation of FOXK1, resulting in reduced FOXK1 stability, blocked interferon signaling and deactivation of RA-FLSs. We treat CIA mice with propionate and show that propionate attenuates CIA. Moreover, a combination of propionate with anti-TNF etanercept synergistically relieves CIA. These results suggest that B. fragilis or propionate could be an alternative or complementary approach to the current therapies.

Abstract The accuracy of target passive localization is influenced by the placement of signal receiving stations; therefore, many studies have been performed to optimize station placement. However, most of the present placement methods focus on the localization error of one target, and if the exact position of the target cannot be determined, but only the range of the target activity is known, how to study the localization station placement in a region is a problem that needs to be solved. This paper proposes a grey wolf optimization algorithm based on the regional target error model to solve the optimal station placement problem. Firstly, a regional target localization error model is established using the measured TDOA, and the overall error matrix within a region is derived. Then, by taking the trace of the error matrix as a criterion, the objective function is established to find the optimal location of the receiving station by grey wolf optimizer. The optimization parameters are also improved to increase the global search ability of the algorithm. Finally, the feasibility and reliability of the overall error model and the grey wolf algorithm proposed are verified by experiments from multiple perspectives. The station placement method proposed in this paper can effectively solve the localization problem of targets that are only known to be in a general activity region in advance, which is more realistic.

Abstract Background Survival prediction using high-dimensional molecular data is a hot topic in the field of genomics and precision medicine, especially for cancer studies. Considering that carcinogenesis has a pathway-based pathogenesis, developing models using such group structures is a closer mimic of disease progression and prognosis. Many approaches can be used to integrate group information; however, most of them are single-model methods, which may account for unstable prediction. Methods We introduced a novel survival stacking method that modeled using group structure information to improve the robustness of cancer survival prediction in the context of high-dimensional omics data. With a super learner, survival stacking combines the prediction from multiple sub-models that are independently trained using the features in pre-grouped biological pathways. In addition to a non-negative linear combination of sub-models, we extended the super learner to non-negative Bayesian hierarchical generalized linear model and artificial neural network. We compared the proposed modeling strategy with the widely used survival penalized method Lasso Cox and several group penalized methods, e.g., group Lasso Cox, via simulation study and real-world data application. Results The proposed survival stacking method showed superior and robust performance in terms of discrimination compared with single-model methods in case of high-noise simulated data and real-world data. The non-negative Bayesian stacking method can identify important biological signal pathways and genes that are associated with the prognosis of cancer. Conclusions This study proposed a novel survival stacking strategy incorporating biological group information into the cancer prognosis models. Additionally, this study extended the super learner to non-negative Bayesian model and ANN, enriching the combination of sub-models. The proposed Bayesian stacking strategy exhibited favorable properties in the prediction and interpretation of complex survival data, which may aid in discovering cancer targets.

Introduction: Rice flowering is a major agronomic trait, determining yield and ecological adaptability in particular regions. ABA plays an essential role in rice flowering, but the underlying molecular mechanism remains largely elusive. Objectives: In this study, we demonstrated a “SAPK8-ABF1-Ehd1/Ehd2” pathway, through which exogenous ABA represses rice flowering in a photoperiod-independent manner. Methods: We generated abf1 and sapk8 mutants using the CRISPR-Cas9 method. Using yeast two-hybrid, Pull down, BiFC and kinase assays, SAPK8 interacted and phosphorylated ABF1. ABF1 directly bound to the promoters of Ehd1 and Ehd2 using ChIP-qPCR, EMSA, and LUC transient transcriptional activity assay, and suppressed the transcription of these genes. Results: Under both long day and short day conditions, simultaneous knock-out of ABF1 and its homolog bZIP40 accelerated flowering, while SAPK8 and ABF1 over-expression lines exhibited delayed flowering and hypersensitivity to ABA-mediated flowering repression. After perceiving the ABA signal, SAPK8 physically binds to and phosphorylates ABF1 to enhance its binding to the promoters of master positive flowering regulators Ehd1 and Ehd2. Upon interacting with FIE2, ABF1 recruited PRC2 complex to deposit H3K27me3 suppressive histone modification on Ehd1 and Ehd2 to suppress these genes transcription, thereby leading to later flowering. Conclusion: Our work highlighted the biological functions of SAPK8 and ABF1 in ABA signaling, flowering control and the involvement of a PRC2-mediated epigenetic repression mechanism in the transcription regulation governed by ABF1 on ABA-mediated rice flowering repression.

Abstract Human bocavirus (HBoV) 1 is considered an important pathogen that mainly affects infants aged 6–24 months, but preventing viral transmission in resource‐limited regions through rapid and affordable on‐site diagnosis of individuals with early infection of HBoV1 remains somewhat challenging. Herein, we present a novel faster, lower cost, reliable method for the detection of HBoV1, which integrates a recombinase polymerase amplification (RPA) assay with the CRISPR/Cas12a system, designated the RPA‐Cas12a‐fluorescence assay. The RPA‐Cas12a‐fluorescence system can specifically detect target gene levels as low as 0.5 copies of HBoV1 plasmid DNA per microliter within 40 min at 37°C without the need for sophisticated instruments. The method also demonstrates excellent specificity without cross‐reactivity to non‐target pathogens. Furthermore, the method was appraised using 28 clinical samples, and displayed high accuracy with positive and negative predictive agreement of 90.9% and 100%, respectively. Therefore, our proposed rapid and sensitive HBoV1 detection method, the RPA‐Cas12a‐fluorescence assay, shows promising potential for early on‐site diagnosis of HBoV1 infection in the fields of public health and health care. The established RPA‐Cas12a‐fluorescence assay is rapid and reliable method for human bocavirus 1 detection. The RPA‐Cas12a‐fluorescence assay can be completed within 40 min with robust specificity and sensitivity of 0.5 copies/μl.

Hydrogen, as an important clean energy source that can be used as an alternative to fossil energy, can reduce global greenhouse gas emissions. However, the large-scale hydrogen industry requires large reservoirs to store hydrogen, and the use of underground reservoirs to store large amounts of hydrogen has become a new topic. In this paper, 11 coal samples with different metamorphic degrees of Carboniferous-Permian in Ordos Basin, Qinshui Basin and Ningwu Basin of China were investigated. Based on the specific surface area and total pore volume, the amount of hydrogen adsorbed by coal rock was evaluated by hydrogen isothermal adsorption experiment and Langmuir model. The mechanism of hydrogen adsorption by coal rock was discussed in combination with coal quality, organic geochemistry, mineral and pore structure characteristics. The results show that the Langmuir model is suitable for the calculation and evaluation of the potential of simulating the hydrogen adsorption of coal rock. Coal rock with different metamorphic degrees has high hydrogen adsorption capacity, and the higher the metamorphic degree, the higher the organic matter abundance and the more developed micropores, the higher the hydrogen adsorption capacity. Hydrogen in coal rock is mainly physically adsorbed on the surface of coal rock by van der Waals force. With the increase of coal rock metamorphic degree, the more micropores formed by organic matter, the more pore volume and total specific surface area provided by organic matter, which increases the amount of hydrogen adsorption, and the higher pore connectivity can improve the efficiency of hydrogen entering pores and being adsorbed. The amount of hydrogen adsorbed by coal rock will increase with the increase of pressure, but due to the reasons such as intermolecular repulsion, the adsorption rate will decrease, and the hydrogen storage will reach saturation. The development of deep coal seams in China provides a good foundation for hydrogen storage in coal seams. However, due to chemical reactions, competitive adsorption and other problems, hydrogen will be lost and lost in coal-rock reservoirs. The use of high pressure for hydrogen storage or acquisition of hydrogen will cause irreversible damage to the structure of coal-rock reservoirs, which has adverse effects on the use of coal-rock reservoirs for one or more storage of hydrogen. Deep coal seams with high metamorphic degree have certain potential as hydrogen storage horizons but further research is still needed to overcome some key problems such as hydrogen loss and reservoir damage.

Abstract Exposure to PM2.5, a harmful type of air pollution, has been associated with compromised male reproductive health; however, it remains unclear whether such exposure can elicit transgenerational effects on male fertility. Here, we aim to examine the effect of paternal exposure to real-world PM2.5 on the reproductive health of male offspring. We have observed that paternal exposure to real-world PM2.5 can lead to transgenerational primary hypogonadism in a sex-selective manner, and we have also confirmed this phenotype by using an external model. Mechanically, we have identified small RNAs (sRNAs) that play a critical role in mediating these transgenerational effects. Specifically, miR6240 and piR016061, which are present in F0 PM sperm, regulate intergenerational transmission by targeting Lhcgr and Nsd1, respectively. We have also uncovered that piR033435 and piR006695 indirectly regulate F1 PM sperm methylation by binding to the 3′-untranslated region of Tet1 mRNA. The reduced expression of Tet1 resulted in hypermethylation of several testosterone synthesis genes, including Lhcgr and Gnas, impaired Leydig cell function and ultimately led to transgenerational primary hypogonadism. Our findings provide insights into the mechanisms underlying the transgenerational effects of paternal PM2.5 exposure on reproductive health, highlighting the crucial role played by sRNAs in mediating these effects. The findings underscore the significance of paternal pre-conception interventions in alleviating the adverse effects of environmental pollutants on reproductive health.

Abstract Background The initial phase II stuty (NCT03215693) demonstrated that ensartinib has shown clinical activity in patients with advanced crizotinib‐refractory, anaplastic lymphoma kinase (ALK)‐positive non‐small cell lung cancer (NSCLC). Herein, we reported the updated data on overall survival (OS) and molecular profiling from the initial phase II study. Methods In this study, 180 patients received 225 mg of ensartinib orally once daily until disease progression, death or withdrawal. OS was estimated by Kaplan‒Meier methods with two‐sided 95% confidence intervals (CIs). Next‐generation sequencing was employed to explore prognostic biomarkers based on plasma samples collected at baseline and after initiating ensartinib. Circulating tumor DNA (ctDNA) was detected to dynamically monitor the genomic alternations during treatment and indicate the existence of molecular residual disease, facilitating improvement of clinical management. Results At the data cut‐off date (August 31, 2022), with a median follow‐up time of 53.2 months, 97 of 180 (53.9%) patients had died. The median OS was 42.8 months (95% CI: 29.3‐53.2 months). A total of 333 plasma samples from 168 patients were included for ctDNA analysis. An inferior OS correlated significantly with baseline ALK or tumor protein 53 (TP53) mutation. In addition, patients with concurrent TP53 mutations had shorter OS than those without concurrent TP53 mutations. High ctDNA levels evaluated by variant allele frequency (VAF) and haploid genome equivalents per milliliter of plasma (hGE/mL) at baseline were associated with poor OS. Additionally, patients with ctDNA clearance at 6 weeks and slow ascent growth had dramatically longer OS than those with ctDNA residual and fast ascent growth, respectively. Furthermore, patients who had a lower tumor burden, as evaluated by the diameter of target lesions, had a longer OS. Multivariate Cox regression analysis further uncovered the independent prognostic values of bone metastases, higher hGE, and elevated ALK mutation abundance at 6 weeks. Conclusion Ensartinib led to a favorable OS in patients with advanced, crizotinib‐resistant, and ALK‐positive NSCLC. Quantification of ctDNA levels also provided valuable prognostic information for risk stratification.

Abstract Objective Anterior cervical discectomy and fusion (ACDF) is the standard procedure for the treatment of cervical spinal stenosis (CSS), but complications such as adjacent segment degeneration can seriously affect the long-term efficacy. Currently, posterior endoscopic surgery has been increasingly used in the clinical treatment of CSS. The aim of this study was to compare the clinical outcomes of single-segment CSS patients who underwent full endoscopic laminotomy decompression or ACDF. Methods 138 CSS patients who met the inclusion criteria from June 2018 to August 2020 were retrospectively analyzed and divided into endoscopic and ACDF groups. The propensity score matching (PSM) method was used to adjust the imbalanced confounding variables between the groups. Then, perioperative data were recorded and clinical outcomes were compared, including functional scores and imaging data. Functional scores included Visual Analog Scale of Arms (A-VAS) and Neck pain (N-VAS), Japanese Orthopedic Association score (JOA), Neck Disability Index (NDI), and imaging data included Disc Height Index (DHI), Cervical range of motion (ROM), and Ratio of grey scale (RVG). Results After PSM, 84 patients were included in the study and followed for 24–30 months. The endoscopic group was significantly superior to the ACDF group in terms of operative time, intraoperative blood loss, incision length, and hospital stay (P 0.05). Conclusion Full endoscopic laminotomy decompression is demonstrated to be an efficacious alternative technique to traditional ACDF for the treatment of single-segment CSS, with the advantages of less trauma, faster recovery, and less impact on cervical spine kinematics and adjacent segmental degeneration.

Abstract This study aimed to assess the impact of light perception presence or absence on visual function recovery in patients with traumatic optic neuropathy (TON). A retrospective analysis was conducted on the clinical data of 206 TON patients. Based on the presence or absence of light perception after injury, patients were categorized into a light perception group and a non-light perception group. A comparison was made between the two groups regarding visual acuity recovery before and after treatment. The non-light perception group comprised 63 patients, with a treatment effectiveness rate of 39.68%. The light perception group consisted of 143 patients, with a treatment effectiveness rate of 74.83%. The difference between the two groups was statistically significant (χ2 = 23.464, P

Abstract The autonomic nervous system plays a crucial role in regulating bone metabolism, with sympathetic activation stimulating bone resorption and inhibiting bone formation. We found that fractures lead to increased sympathetic tone, enhanced osteoclast resorption, decreased osteoblast formation, and thus hastened systemic bone loss in ovariectomized (OVX) mice. However, the combined administration of parathyroid hormone (PTH) and the β-receptor blocker propranolol dramatically promoted systemic bone formation and osteoporotic fracture healing in OVX mice. The effect of this treatment is superior to that of treatment with PTH or propranolol alone. In vitro, the sympathetic neurotransmitter norepinephrine (NE) suppressed PTH-induced osteoblast differentiation and mineralization, which was rescued by propranolol. Moreover, NE decreased the PTH-induced expression of Runx2 but enhanced the expression of Rankl and the effect of PTH-stimulated osteoblasts on osteoclastic differentiation, whereas these effects were reversed by propranolol. Furthermore, PTH increased the expression of the circadian clock gene Bmal1, which was inhibited by NE-βAR signaling. Bmal1 knockdown blocked the rescue effect of propranolol on the NE-induced decrease in PTH-stimulated osteoblast differentiation. Taken together, these results suggest that propranolol enhances the anabolic effect of PTH in preventing systemic bone loss following osteoporotic fracture by blocking the negative effects of sympathetic signaling on PTH anabolism.

Abstract Background Cancer‐related pain is one of the common priority symptoms in advanced lung cancer patients at the end‐of‐life (EOL). Alleviating pain is undoubtedly a critical component of palliative care in lung cancer. Our study was initiated to examined trends in opioid prescription‐level outcomes as potential indicators of undertreated pain in China. Methods This study used data on 1330 patients diagnosed with lung cancer of urban city medical insurance in China who died between 2014 and 2017. Opioid prescription‐level outcomes were determined by annual trends of the proportion of patients filling an opioid prescription, the total dose of opioids filled by decedents, and morphine milligram equivalents per day (MMED) at the EOL (defined as the 60 days before death). We further analyzed monthly changes in the number of opioid prescriptions filled, MMED, and mean daily dose of opioids per prescription (MDDP) of the last 60 days of life by year at death and age, respectively. Results A total of 959 patients with exact dates of death were included, with 432 cases (45.06%; 95% CI: 44.36%–45.77%) receiving at least one opioid prescription at the EOL. The declining trends were shown in the proportion of patients filling any opioid prescription, the total dose of opioids filled by decedents and MMED, with an annual decrease of 0.341% (p = 0.01), 104.23 mg (p = 0.011) and 2.84 mg (p = 0.014), respectively. Within the 31–60 days to the 0–30 days of life, the MMED declined 6.08 mg (95% CI: −7.14 to −5.03; p = 0.000351), while the number of opioid prescriptions rose 0.66 (95% CI: 0.160–1.16; p = 0.025). Like the MMED, the MDDP fell 4.11 mg (95% CI: −5.86 to −2.37; p = 0.005) within the last month before death compared to the previous month. Conclusion Terminal lung cancer populations in urban China have experienced reduced access to opioids at the EOL. The clinicians did not prescribe a satisfactory dose of opioids per prescription, while the patients suffered increasing pain in the last 30 days of life. Sufficient opioid analgesic administration should be advocated for lung cancer patients during the EOL period.

Abstract Background The relationship between the triglyceride glucose (TyG) index and osteoarthritis (OA) remains unclear. The objective of this study was to examine potential associations between an elevated TyG index and an increased risk of OA prevalence. Methods 3,921 participants with OA from the National Health and Nutrition Examination Survey (2015–2020) were included in this study. Participants were categorized into quartiles based on TyG index, which was determined using the formula: Ln [triglyceride (mg/dL) fasting blood glucose (mg/dL)/2]. Weighted multivariable regression, subgroup analyses, and threshold effect analyses were performed to calculate the independent association between TyG index and OA. Results A total of 25,514 people were enrolled, with a mean TyG index of 8.48 ± 0.65. The results of multivariable logistic regression analysis after full adjustment showed a significant association between higher TyG index values and an increased risk of OA. Specifically, each incremental unit increase in the TyG index was associated with a 634% higher risk of OA [OR = 7.34; 95% CI: 2.25, 23.93; p = 0.0010]. Based on interaction tests, age, gender, BMI, and smoking status did not significantly affect the relationship between the TyG index and OA, while diabetes showed a stronger positive correlation between the TyG index and OA. Conclusion An increased risk of OA was associated with a higher TyG index. TyG could be a valuable predictor of OA and offer novel perspectives on the assessment and treatment of OA.

In this work, the (10 wt%, 30 wt%, 50 wt%) TiC reinforced AlFeCrCo medium entropy alloy (MEA) lightweight composite coatings with the extremely low porosity were successfully fabricated on Mg alloy substrate by resistance seam processing. The results showed that the microstructure of composite coating was consisting of a BCC-based MEA matrix and TiC particles, and a good metallurgical bonding with a semi-coherent relationship was formed between the coating and Mg alloy. Furthermore, the lightweight composite coatings (5.6–6.5 g cm−3) showed improved corrosion resistance over Mg alloy substrate. Specifically, the composite coatings exhibited optimized wear performance in the dry, deionized water and 3.5 wt% NaCl solution conditions with the increasing TiC content, surpassing related high/medium entropy alloy coatings, which is attributed to the formation of a coherent interface between the MEA matrix and the TiC particles. The first-principles calculations were performed to elucidate the nature for the higher bonding strength of TiC/MEA interface. In these wear conditions, the main wear mechanisms of composite coatings were discussed in terms of adhesive wear, oxidative wear and/or corrosive wear, in connection with their microstructure features and electrochemical behaviors. Based on the elevated anti-corrosion ability, this work has provided a strategy to fabricate advanced coatings on Mg alloys endowing with lower density, interfacial metallurgical bonding and optimized wear resistance, substantially contributing to the development of high-performance composite coatings.

BackgroundDuck enteritis virus (DEV) belongs to Alphaherpesvirinae; little is known about the DEV UL7 gene and its encoded protein. This study examined the molecular characteristics of DEV pUL7 in vitro and determined whether DEV pUL7 co-localizes with pUL51.ResultsThe results showed that UL7 can be regarded as a late gene. Moreover, immunofluorescence assay revealed that pUL7 was located around the perinuclear cytoplasmic region and co-localized with pUL51 in the cytoplasm and nucleus after transfection into duck embryo fibroblast cells (DEFs).ConclusionIn conclusion, we identified the molecular characteristics of the DEV UL7 gene, which is a late gene, and the co-localization of its encoded protein with pUL51 in transfected DEFs, enriching our understanding of pUL7 and future research directions.

Mesenchymal stem cells (MSCs) have emerged as promising candidates for idiopathic pulmonary fibrosis (IPF) therapy. Increasing the MSC survival rate and deepening the understanding of the behavior of transplanted MSCs are of great significance for improving the efficacy of MSC-based IPF treatment. Therefore, dual-functional Au-based nanoparticles (Au@PEG@PEI@TAT NPs, AuPPT) were fabricated by sequential modification of cationic polymer polyetherimide (PEI), polyethylene glycol (PEG), and transactivator of transcription (TAT) penetration peptide on AuNPs, to co-deliver retinoic acid (RA) and microRNA (miRNA) for simultaneously enhancing MSC survive and real-time imaging tracking of MSCs during IPF treatment. AuPPT NPs, with good drug loading and cellular uptake abilities, could efficiently deliver miRNA and RA to protect MSCs from reactive oxygen species and reduce their expression of apoptosis executive protein Caspase 3, thus prolonging the survival time of MSC after transplantation. In the meantime, the intracellular accumulation of AuPPT NPs enhanced the computed tomography imaging contrast of transplanted MSCs, allowing them to be visually tracked in vivo. This study establishes an Au-based dual-functional platform for drug delivery and cell imaging tracking, which provides a new strategy for MSC-related IPF therapy.

In recent years, there has been notable progress in the development of perovskite solar cells (PSCs), marked by significant advancements in efficiency, stability, and scalability. Among different device architectures and technical routes, mesoporous perovskite solar cells (MPSCs) based on TiO2/ZrO2/carbon scaffold and screen-printing fabrication process have shown unique advantages for mass production and commercialization due to the low material cost and scalable fabrication process. Through efforts on material optimization, interface engineering, and device design, the power conversion efficiency of MPSCs has steadily increased from the initial 6.64 ​% in 2013 to current 22.22 ​%. Attributing to the screen-printing-based fabrication process, printable mesoscopic PSCs have enabled large-area devices, from mini-modules to modules, and solar panels. In this review, we discuss the development and latest advances of MPSCs, and provide outlooks for further improving the performance of this promising photovoltaic technology.

Abstract Metal–organic frameworks (MOFs) are a class of promising nanomaterials for atmospheric water harvesting (AWH), especially in arid remote areas. However, several challenges are still faced for practical applications because of the dissatisfied water adsorption/desorption properties in terms of the capability, kinetics, and stability. Herein, we report the facile synthesis of a nano‐sized octahedral nitrogen‐modified MOF‐801 that exhibits superior solar‐powered AWH performance using a custom‐made device, with a state‐of‐the‐art water harvesting ability up to 4.64LH2OkgMOFs−1 from air upon 12‐h test under a relative humidity (RH) of 30% and simulated sunlight irradiation. The nitrogen‐modified MOF‐801 with rapid sorption–desorption kinetics, uptakes 0.29gH2OgMOFs‐1 of water at 30% RH within 30 min and releases 90% of the captured water within 10 min under 1‐sun illumination. The success relies on N‐doping‐induced mixed‐linkers in the form of 2,3‐diaminobutanedioic acid and fumaric acid in the unique pore structures of the MOFs for rapid and high‐capacity water capture. The N‐doped MOF‐801 with water uptake capacity, fast adsorption kinetics, and cycle stability sheds light on the practical use of MOFs for effective solar‐powered water harvesting from droughty air.

ObjectiveIn the past, the localization of seizure onset zone (SOZ) primarily relied on traditional EEG signal analysis methods. However, due to their limited spatial and temporal resolution, accurately pinpointing neural activity was challenging, thereby restricting their clinical applicability. Compared with traditional EEG signals, SEEG signals have superior spatial and temporal resolution, and can more accurately record neural activity near epileptic foci, making them better suited for studying SOZ. In addition, the traditional EEG signal analysis methods still have limitations, mainly focusing on the analysis of local signal features, while ignoring the complexity and interconnection of the overall brain network. How to more accurately locate SOZ is still not well resolved. The purpose of this study is to develop an effective positioning method for more accurate positioning.MethodTo overcome these limitations, this study proposed a model integrating brain functional network analysis with nonlinear dynamics. We utilized weighted phase lag index (WPLI) to construct brain functional network, epilepic network connectivity strength (ENCS) as the feature, and introduced persistence entropy (PE) for feature fusion, subsequently employing support vector machine (SVM) classification.ResultsThe proposed method was verified on the HUP-iEEG dataset, our solution identified the SOZ with 0.9440 accuracy, 0.9848 precision, 0.8974 recall rate, 0.9340 F1 score and 0.9697 area under the ROC curve across patients, which outperforms the existing approaches. It exhibits a 2.30 percentage point enhancement in localisation accuracy along with a 2.97 percentage points in AUC compared to others.ConclusionOur method consider the interactions between nodes in brain network connections, as well as the inherent nonlinear and non-stationary properties of neural signals, to be more robust.

It is a challenge to study the nucleation of cavitation bubbles, which critically depends on nanoscale morphological features. Our recent advances in synthesizing colloidal negative-curvature nanoparticles (NGC-NPs) offer a rare opportunity, in comparison to the conventional studies of bulk substrates, where it is difficult to obtain consistent and well-defined surface features. In order to quantitatively assess their effects, we exploit the radical-induced color change of [Fe(SCN)6]3−, which turned out to be a more convenient method than the bending of AgNWs and the fluorescence-based methods. We show that the NGC-NPs outperform positive-curvature nanoparticles (PSC-NPs) and homogeneous nucleation, in terms of promoting cavitation. The NGC-NPs provide a higher percentage of gas–solid interface, and thus reduces the activation barrier during the critical stage of bubble nucleation. This leads a higher probability of cavitation and transforms more energy from ultrasonication to radical formation and shockwaves.

Key Clinical Message One Kirsten Ras (KRAS) G12C mutated non‐small cell lung cancer (NSCLC) patient had improved poor performance status and obtained mixed response with the first‐line KRAS‐targeted treatment of sotorasib. After disease progression, partial response was achieved with chemotherapy plus immunotherapy. KRAS G12C mutated immunoenvironment in NSCLC may favor the immunotherapy. Abstract KRAS is one of the most commonly mutated genes, which used to be untargetable. The phase II CodeBreak 100 trial revealed 6.8‐month median progress‐free survival (PFS) and 12.5‐month overall survival (OS) in previously treated KRAS G12C‐mutant NSCLC patients treated with KRAS inhibitor, sotorasib. The specimens of the brain, lymph node (LN), and blood from the patient were analyzed by next‐generation sequencing. Hematoxylin and eosin staining and immunohistochemistry were performed for pathological characterization. Computed tomography (CT) and magnetic resonance imaging (MRI) scan were used for treatment response evaluation. The patient was diagnosed in a bad Eastern Cooperative Oncology Group performance status (ECOG‐PS) with metastatic KRAS G12C‐mutated lung adenocarcinoma who had achieved mixed response to sotorasib as the first‐line treatment. Although 5‐month PFS of the treatment with sotorasib was not surprising, the patient achieved significantly improved ECOG‐PS score from 4 to 1. Subsequently, partial response (PR) was achieved with the treatment of pemetrexed plus pembrolizumab. This case highlights superior efficacy of first‐line treatment with sotorasib for the advance untreated KRAS G12C‐mutant patients. The high efficacy of the treatment with chemotherapy plus immunotherapy revealed that immunoenvironment of KRAS G12C‐mutated patient may favor the immunotherapy.

A two-dimensional symmetric fluid model is established to study the influence of radio frequency (RF) wave driving frequency on the capacitively coupled plasma discharge. The relationship between the driving frequency and electron density is obtained by solving the electron energy balance equation. The calculation results show that the average electron density first increases rapidly with the increase in driving frequency and then gradually tends to saturation at a threshold frequency. A fluid simulation is also carried out, which provides similar results. Physical studies on this phenomenon are conducted, revealing that the essence of this phenomenon is due to the inability of electrons to quickly respond to potential changes within the boundary sheath when the driving frequency of RF exceeds the plasma frequency. In addition, it is also found that increasing gas pressure can enhance the electron density and the type of gas can also affect the electron density.

LED (Light Emitting Diode) lighting has the characteristics of efficiency, environmental protection, energy conservation, and so on. It is more and more widely used in the lighting system of urban rail vehicles. The research on the LED lighting system of rail vehicles has important practical significance. As the core of the LED lighting system of rail vehicles, the LED control system of urban rail vehicles is the key to ensure the stable operation of the entire LED lighting system. The lighting environment of urban rail vehicles needs LED lighting system with good power stability, long life of drive chip, high feedback accuracy, fast processing speed, intelligent algorithm efficiency and other characteristics to ensure continuous operation, and also should have strong anti-interference ability and excellent static performance. However, conventional lighting systems not only have low life, but also have poor stability. Therefore, this paper studied the uniqueness and functional module analysis of LED driving power supply by analyzing the causes and disadvantages of LED lighting, and finally optimized the design of LED lighting control system by using the adaptive PID (Proportional Integral Differential) algorithm. Through comparison, the brightness acquisition quality of LED lighting control system was 21.2 % higher than that of traditional lighting control system, and the lighting control effect was 18 % higher than that of traditional lighting control system. In short, the selection of LED lighting driving power affects the lighting effect of rail vehicles.

The successful application of new technologies such as remotely piloted aircraft systems, distributed electric propulsion systems, and automatic control systems on electric vertical take-off and landing(eVTOL) aircraft has prompted Urban Air Mobility (UAM) to be mentioned frequently. UAM is a newly raised transport mode of using eVTOL aircraft to transport people and cargo in urban areas, which is thought to share some of the traffic on the ground. One of the prerequisites for UAM to operate on a regular basis is that its demand can support the operating costs, so forecasting UAM demand is necessary. We conduct UAM demand forecasting based on the four-step method, focusing on improving the third-step modal split, and propose a demand forecasting model based on the logit model. The model combines a nested logit (NL) model with a multinomial logit (MNL) model to solve the problem of non-existent UAM sharing rates. We use Chengdu, China as an example, and focus on forecasting the UAM traffic demand in 2030 with the help of the four-step method. The results show that UAM is suitable for shared operation during the early stages. With a fully shared operation, the UAM share rate increases by 0.73% for every kilometer increase in distance. Moreover, UAM is more competitive than other modes for delivery distances exceeding 15 ​km. Finally, using the distributions of the share rate and traffic flow pattern from the simulation, we propose the routes that can be prioritized for UAM operations in Chengdu.

High-performance P-type cuprous oxide (Cu2O) film was prepared at room temperature by high power impulse magnetron sputtering. Optical emission spectra revealed that the ratio of Cu radicals/ions in the plasma significantly decreased with increasing process pressure due to the reduction of sputtering yield as the Cu target surface was oxidized by the increased oxygen radicals/ions. In addition, the increase of self-sputtering yield for Cu cations with increasing process pressure reduced the arrival ratio of Cu species at the substrate surface. As a result, the film crystal phase transformed from Cu2O to Cu4O3 and CuO with increasing process pressure. X-ray photoelectron spectra and Hall effect test results revealed that the oxygen vacancy defects in the films were passivated by the increased oxygen species at higher process pressure, leading to the inhibition of electron background and the emergence of net hole concentration. A mobility of 37.3 cm2/V·s was achieved, which is very high for room-temperature-deposited Cu2O film and comparable to high-temperature (300–600 °C) deposited/post-annealed Cu2O film. Finally, Cu2O thin film transistors (TFTs) exhibited reasonable switching characteristics with a low off-current of 0.3 nA without post-annealing treatment, showing an advantage in low-temperature preparation.

Abstract Cancer cell growth, metastasis, and drug resistance pose significant challenges in the management of lung adenocarcinoma (LUAD). However, there is a deficiency in optimal predictive models capable of accurately forecasting patient prognoses and guiding the selection of targeted treatments. Programmed cell death (PCD) pathways play a pivotal role in the development and progression of various cancers, offering potential as prognostic indicators and drug sensitivity markers for LUAD patients. The development and validation of predictive models were conducted by integrating 13 PCD patterns with comprehensive analysis of bulk RNA, single-cell RNA transcriptomics, and pertinent clinicopathological details derived from TCGA-LUAD and six GEO datasets. Utilizing the machine learning algorithms, we identified ten critical differentially expressed genes associated with PCD in LUAD, namely CHEK2, KRT18, RRM2, GAPDH, MMP1, CHRNA5, TMPRSS4, ITGB4, CD79A, and CTLA4. Subsequently, we conducted a programmed cell death index (PCDI) based on these genes across the aforementioned cohorts and integrated this index with relevant clinical features to develop several prognostic nomograms. Furthermore, we observed a significant correlation between the PCDI and immune features in LUAD, including immune cell infiltration and the expression of immune checkpoint molecules. Additionally, we found that patients with a high PCDI score may exhibit resistance to immunotherapy and standard adjuvant chemotherapy regimens; however, they may benefit from other FDA-supported drugs such as docetaxel and dasatinib. In conclusion, the PCDI holds potential as a prognostic signature and can facilitate personalized treatment for LUAD patients.

Abstract Background Problem-based learning (PBL) is a student-centred approach that triggers learning by presenting problems cenarios early in the learning process.Flipped classrooms have been used in various disciplines using various models.Pre-class e- learning in aflipped classrooms can enrich knowledge acquisition in PBL teaching. This study was conducted to explore nursing students’experience of flipped classroom combined with problem-based learning in a paediatric nursing course. Method This descriptive qualitative study was conducted between January and June 2022.Semi-structured interviews were conducted with nursing students who were participated in flipped classrooms combined with PBL teaching in a paediatric nursing course at Soochow university in China. Nursing students were selected using a purposive sampling method until no new data were generated (n = 16). Results We identified ten sub-themes and four higher-order themes based on these sub-themes: (1)stimulating interest in learning and enhancing autonomous learning,(2)improving independent thinking and problem-solving skills,(3)cultivating team work spirit, and (4) gaining knowledge and improving skills.The findings of our research contribute to show the effectiveness of the flipped classroom combined with PBL in a paediatric nursing course. Conclusion The flipped classroom combined with PBL in a paediatric nursing course can enhances communication and cooperation abilities among nursing students, promoting common progress and the comprehensive development of nursing students.

Abstract Our objective is to develop a prognostic model focused on cuproptosis, aimed at predicting overall survival (OS) outcomes among Acute myeloid leukemia (AML) patients. The model utilized machine learning algorithms incorporating stacking. The GSE37642 dataset was used as the training data, and the GSE12417 and TCGA-LAML cohorts were used as the validation data. Stacking was used to merge the three prediction models, subsequently using a random survival forests algorithm to refit the final model using the stacking linear predictor and clinical factors. The prediction model, featuring stacking linear predictor and clinical factors, achieved AUC values of 0.840, 0.876 and 0.892 at 1, 2 and 3 years within the GSE37642 dataset. In external validation dataset, the corresponding AUCs were 0.741, 0.754 and 0.783. The predictive performance of the model in the external dataset surpasses that of the model simply incorporates all predictors. Additionally, the final model exhibited good calibration accuracy. In conclusion, our findings indicate that the novel prediction model refines the prognostic prediction for AML patients, while the stacking strategy displays potential for model integration.

Abstract Objective To construct and validate a model based on the dual-energy computed tomography (DECT) quantitative parameters and radiological features to predict Ki-67 expression levels in pancreatic ductal adenocarcinoma (PDAC). Materials and methods Data from 143 PDAC patients were analysed. The variables of clinic, radiology and DECT were evaluated. In the arterial phase and portal venous phase (PVP), the normalized iodine concentration (NIC), normalized effective atomic number and slope of the spectral attenuation curves were measured. The extracellular volume fraction (ECVf) was measured in the equilibrium phase. Univariate analysis was used to screen independent risk factors to predict Ki-67 expression. The Radiology, DECT and DECT–Radiology models were constructed, and their diagnostic effectiveness and clinical applicability were obtained through area under the curve (AUC) and decision curve analysis, respectively. The nomogram was established based on the optimal model, and its goodness-of-fit was assessed by a calibration curve. Results Computed tomography reported regional lymph node status, NIC of PVP, and ECVf were independent predictors for Ki-67 expression prediction. The AUCs of the Radiology, DECT, and DECT–Radiology models were 0.705, 0.884, and 0.905, respectively, in the training cohort, and 0.669, 0.835, and 0.865, respectively, in the validation cohort. The DECT–Radiology nomogram was established based on the DECT–Radiology model, which showed the highest net benefit and satisfactory consistency. Conclusions The DECT–Radiology model shows favourable predictive efficacy for Ki-67 expression, which may be of value for clinical decision-making in PDAC patients. Critical relevance statement The DECT–Radiology model could contribute to the preoperative and non-invasive assessment of Ki-67 expression of PDAC, which may help clinicians to screen out PDAC patients with high Ki-67 expression. Abstract Soft tissue sarcoma is a broad family of mesenchymal malignancies exhibiting remarkable histological diversity. We portray the proteomic landscape of 272 soft tissue sarcomas representing 12 major subtypes. Hierarchical classification finds the similarity of proteomic features between angiosarcoma and epithelial sarcoma, and elevated expression of SHC1 in AS and ES is correlated with poor prognosis. Moreover, proteomic clustering classifies patients of soft tissue sarcoma into 3 proteomic clusters with diverse driven pathways and clinical outcomes. In the proteomic cluster featured with the high cell proliferation rate, APEX1 and NPM1 are found to promote cell proliferation and drive the progression of cancer cells. The classification based on immune signatures defines three immune subtypes with distinctive tumor microenvironments. Further analysis illustrates the potential association between immune evasion markers (PD-L1 and CD80) and tumor metastasis in soft tissue sarcoma. Overall, this analysis uncovers sarcoma-type-specific changes in proteins, providing insights about relationships of soft tissue sarcoma.

Abstract The carbon market plays a critical role in promoting the transition toward renewable energy sources and reducing greenhouse gas emissions in the electricity generation and transmission. Extant research has overlooked the dynamic bilateral causality that exists between electricity and carbon markets. Moreover, these studies have frequently treated the macroeconomic effect as exogenous. To bridge this research gap, this paper presents a holistic modeling framework that comprehensively captures the intertwined nature of electricity and carbon markets and their concomitant interactions with the overarching economy. The suggested modeling framework is an integration of three principal modules, namely, a carbon market, an electricity market, and economic system. This synergistic blend provides an exhaustive understanding of the entire market operation cycle. It offers detailed clearance rules, and most importantly, it adopts a macroeconomic systematic modeling approach for evaluating the impact emanating from the interconnected electricity and carbon markets. To illustrate the practicality and effectiveness of the proposed approach, a case study anchored on empirical data sourced from the electricity and carbon markets in China is conducted. The empirical findings underscore the fact that incorporating a green certificate market into the modeling framework can precipitate a reduction in greenhouse gas emissions. Additionally, the results indicate that expanding the scale of the green certificate market from 1.9% in 2021 to 33% by 2023 will increase the generation of green electricity by 10%.

Lymphatic metastasis is the main metastatic route for colorectal cancer, which increases the risk of cancer recurrence and distant metastasis. The properties of the lymph node metastatic colorectal cancer (LNM-CRC) cells are poorly understood, and effective therapies are still lacking. Here, we found that hypoxia-induced fibroblast activation protein alpha (FAPα) expression in LNM-CRC cells. Gain- or loss-function experiments demonstrated that FAPα enhanced tumor cell migration, invasion, epithelial–mesenchymal transition, stemness, and lymphangiogenesis via activation of the STAT3 pathway. In addition, FAPα in tumor cells induced extracellular matrix remodeling and established an immunosuppressive environment via recruiting regulatory T cells, to promote colorectal cancer lymph node metastasis (CRCLNM). Z-GP-DAVLBH, a FAPα-activated prodrug, inhibited CRCLNM by targeting FAPα-positive LNM-CRC cells. Our study highlights the role of FAPα in tumor cells in CRCLNM and provides a potential therapeutic target and promising strategy for CRCLNM.

Abstract Early diagnosis and pharmacological treatment of central nervous system (CNS) diseases has been a long-standing challenge for clinical research due to the presence of the blood–brain barrier. Specific proteins and RNAs in brain-derived extracellular vesicles (EVs) usually reflect the corresponding state of brain disease, and therefore, EVs can be used as diagnostic biomarkers for CNS diseases. In addition, EVs can be engineered and fused to target cells for delivery of cargo, demonstrating the great potential of EVs as a nanocarrier platform. We review the progress of EVs as markers and drug carriers in the diagnosis and treatment of neurological diseases. The main areas include visual imaging, biomarker diagnosis and drug loading therapy for different types of CNS diseases. It is hoped that increased knowledge of EVs will facilitate their clinical translation in CNS diseases.

The task of named entity recognition has been widely used in medical text analysis, but there is still the problem of poor transfer ability in practical applications. This work proposes a novel named entity recognition method based on a proposed knowledge distillation framework and Efficient GlobalPointer for Chinese biomedical and clinical data. Specifically, our study leverages the Efficienct GlobalPointer to address the issue of entity nesting and introduces a context shield window to mitigate interference from redundant information. Furthermore, the model’s generalization ability is enhanced through a novel knowledge distillation framework. The proposed knowledge distillation framework solves the problem of independent feature learning process in feature distillation by using linkage mechanism. The recognition accuracy is improved by the proposed knowledge distillation method while keeping the model complexity low, so that our method can meet the inference speed requirement in real applications while ensuring a certain recognition accuracy. Our method achieves excellent experimental results on three publicly available Chinese datasets, where the comprehensive evaluation metric F1 exceeds the best results achieved by existing methods.

Searchable encryption is widely used in electronic health record systems because it enables users to search ciphertext data without decryption. However, the existing traditional searchable encryption schemes lack fine-grained access policies with wildcards in electronic health record systems. And they also do not consider the problem of hiding policies, as well as the problem of incomplete search results caused by cloud servers. In order to solve all the above problems, this paper proposes a blockchain-aided attribute-based searchable scheme with the properties of inner product predicate. In the proposed scheme, the attribute encryption mechanism implements fine-grained access policies with wildcards, which improves the data owner’s ability to control access authorization precisely. Introducing the inner product predicate not only achieves fully hidden access policies but also prevents the leakage of sensitive medical data. The immutability of the blockchain ensures the integrity of multi-keyword search results, guaranteeing reliable data sharing. Finally, the security proof and the performance evaluation are conducted to confirm the security and effectiveness of the proposed scheme.

Font style recognition plays a vital role in the field of computer vision, particularly in document and pattern analysis, and image processing. In the industry context, this recognition of font styles holds immense importance for professionals such as graphic designers, front-end developers, and UI-UX developers. In recent times, font style recognition using Computer Vision has made significant progress, especially in English. Very few works have been done for other languages as well. However, the existing models are computationally costly, time-consuming, and not diversified. In this work, we propose a state-of-the-art model to recognize Bangla fonts from images using a quantized Convolutional Autoencoder (Q-CAE) approach. The compressed model takes around 58 KB of memory only which makes it suitable for not only high-end but also low-end computational edge devices. We have also created a synthetic data set consisting of 10 distinct Bangla font styles and a total of 60,000 images for conducting this study as no dedicated dataset is available publicly. Experimental outcomes demonstrate that the proposed method can perform better than existing methods, gaining an overall accuracy of 99.95% without quantization and 99.85% after quantization.
