Highly efficient precision gene editing is ultimately a desirable technology, but implementation remains challenging, especially in plants. Here, we tested a novel CRISPR-Cas-based tool for precision gene replacement in plants employing a microhomology-mediated end joining (MMEJ) mechanism. The MMEJ-mediated precise gene replacement employing 20-bp microhomology MMEJ donors (~100 bp lengths) and a cNHEJ inhibitor, NU7441, produced high targeted editing efficiencies, up to 1.60 ± 0.14, 4.47 ± 1.98, and 8.98 ± 4.73 % in tomato, lettuce, and cabbage, respectively. Our data also revealed the critical impacts of the microhomology length and NU7441 concentration on MMEJ-based precision gene editing in plants.

This study empirically investigated the effects of monetary policy shocks on farm prices and exchange rates by using vector auto-regression models with sign restrictions on impulse responses. The main empirical results are as follows. First, contractionary monetary policy shocks have significant negative effects on real farm prices, which suggests that farm prices respond more to monetary policy shocks than to the general price level. Second, the effects of monetary policy shocks on farm prices are stronger than the effects of monetary policy shocks on exchange rates. Third, farm price dynamics under monetary policy shocks show “delayed overshooting” as exchange rate dynamics do.

Summary Gene editing and/or allele introgression with absolute precision and control appear to be the ultimate goals of genetic engineering. Precision genome editing in plants has been developed through various approaches, including oligonucleotide‐directed mutagenesis (ODM), base editing, prime editing and especially homologous recombination (HR)‐based gene targeting. With the advent of CRISPR/Cas for the targeted generation of DNA breaks (single‐stranded breaks (SSBs) or double‐stranded breaks (DSBs)), a substantial advancement in HR‐mediated precise editing frequencies has been achieved. Nonetheless, further research needs to be performed for commercially viable applications of precise genome editing; hence, an alternative innovative method for genome editing may be required. Within this scope, we summarize recent progress regarding precision genome editing mediated by microhomology‐mediated end joining (MMEJ) and discuss their potential applications in crop improvement.

CRISPR/Cas9-based multiplexed editing of SlHyPRP1 resulted in precise deletions of its functional motif(s), thereby resulting in salt stress-tolerant events in cultivated tomato. Crop genetic improvement to address environmental stresses for sustainable food production has been in high demand, especially given the current situation of global climate changes and reduction of the global food production rate/population rate. Recently, the emerging clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein (Cas)-based targeted mutagenesis has provided a revolutionary approach to crop improvement. The major application of CRISPR/Cas in plant genome editing has been the generation of indel mutations via error-prone nonhomologous end joining (NHEJ) repair of DNA DSBs. In this study, we examined the power of the CRISPR/Cas9-based novel approach in the precise manipulation of protein domains of tomato hybrid proline-rich protein 1 (HyPRP1), which is a negative regulator of salt stress responses. We revealed that the precise elimination of SlHyPRP1 negative-response domain(s) led to high salinity tolerance at the germination and vegetative stages in our experimental conditions. CRISPR/Cas9-based domain editing may be an efficient tool to engineer multidomain proteins of important food crops to cope with global climate changes for sustainable agriculture and future food security.

OBJECTIVES: Metabolic syndrome (MetS) is closely associated with dietary intake; however, few studies have investigated whether the consumption of fruits and vegetables and multivitamin use affect MetS in the Korean population. This study aimed to examine these effects in Korean adults.METHODS: This was a cross-sectional study of 89,548 participants aged between 40 years and 69 years selected from the baseline data of the Health Examinees study conducted in Korea. Fresh vegetable and fruit consumption was assessed using a validated 106-item food frequency questionnaire. MetS and its components were defined using the National Cholesterol Education Program, Adult Treatment Panel III criteria. Multivariate logistic regression analyses were conducted to identify associations of fresh vegetable, fruit, and fresh vegetable+fruit consumption and multivitamin use with the prevalence of MetS.RESULTS: Female in the highest quartile of fresh vegetable, fruit, and fresh vegetable + fruit consumption exhibited a lower prevalence of MetS than those in the lowest quartile. An inverse association with the prevalence of MetS was observed among male with only fresh vegetable consumption. The interaction between the 3 categories and multivitamin intake on the prevalence of MetS was not significant (all pinteraction>0.05), regardless of sex.CONCLUSIONS: Multivitamin use and consumption of fresh vegetables and fruits had no significant synergistic effects. Although fresh vegetable and fruit consumption showed an inverse association with the prevalence of MetS, this relationship was not altered by multivitamin use.

Background Tuberculosis (TB) is a highly infectious disease. Negative perceptions and insufficient knowledge have made its eradication difficult. Recently, mobile health care interventions, such as an anti-TB chatbot developed by the research team, have emerged in support of TB eradication programs. However, before the anti-TB chatbot is deployed, it is important to understand the factors that predict its acceptance by the population. Objective This study aims to explore the acceptance of an anti-TB chatbot that provides information about the disease and its treatment to people vulnerable to TB in South Korea. Thus, we are investigating the factors that predict technology acceptance through qualitative research based on the interviews of patients with TB and homeless facility personnel. We are then verifying the extended Technology Acceptance Model (TAM) and predicting the factors associated with the acceptance of the chatbot. Methods In study 1, we conducted interviews with potential chatbot users to extract the factors that predict user acceptance and constructed a conceptual framework based on the TAM. In total, 16 interviews with patients with TB and one focus group interview with 10 experts on TB were conducted. In study 2, we conducted surveys of potential chatbot users to validate the extended TAM. Survey participants were recruited among late-stage patients in TB facilities and members of web-based communities sharing TB information. A total of 123 responses were collected. 척수 손상 장애인에게 있어 전동 휠체어는 이동과 독립성 증가를 가져오며 이로 인해 삶의 질 향상을 가져온다. 하지만 휠체어의 사용자 인터페이스는 연구 주제로 다뤄지지 않는 실정이다. 본 연구에서는 척수 손상 장애인 5명을 대상으로 인터뷰와 관찰을 진행하고 그 결과를 바탕으로 전동 휠체어 사용자 인터페이스는 무엇이고, 사용자 인터페이스가 척수 손상 장애인의 전동 휠체어 사용에 어떻게 영향을 주는지 살펴보는 것을 연구 목적으로 한다. 전동 휠체어 사용자 인터페이스는 세 가지로 구분될 수 있다. 이동을 위한 사용자 인터페이스는 바퀴, 조절 스위치(조이스틱, 컨트롤러), 조명 장치가 있으며, 지지를 위한 사용자 인터페이스는 머리, 몸통, 팔, 다리, 발 받침대와 고정 벨트, 좌석의 바닥면과 등받이, 가변 기능 컨트롤러, 견인고리가 있다. 활동 보조를 위한 사용자 인터페이스는 짐 보관함과 충전 잭이 있다. 추후 척수 손상 레벨과 같은 장애 정도에 따라 전동 휠체어 사용자 인터페이스가 어떻게 달라지는지에 대해 연구 대상자를 추가하여 연구를 진행하면 사용자에 따라 어떻게 사용자 인터페이스를 디자인해야 하는지 의미 있는 연구 결과를 도출할 수 있을 것이라 사료된다.

Callose is a plant cell wall polymer in the form of β-1,3-glucan, which regulates symplasmic channel size at plasmodesmata (PD). It plays a crucial role in a variety of processes in plants through the regulation of intercelluar symplasmic continuity. However, how to maintain callose homeostasis at PD in the molecular levels is poorly understood. To further elucidate the mechanism of PD callose homeostasis, we screened and identified an Arabidopsis mutant plant that exhibited excessive callose deposition at PD. Based on the Next-generation sequencing (NGS)-based mapping, other mutant allele analysis, and complementation assay, the mutated gene was shown to be α1-COP, which encodes a member of the COPI coatomer complex comprised of α, β, β′, γ, δ, ε, and ζ subunits. Since there is no report on the link between COPI and callose/PD, it was extremely curious to know the roles of α1-COP or COPI in PD regulation through callose deposition. Here, we report that loss-of-function of α1-COP directly elevates the callose accumulation at PD by affecting subcellular protein localization of callose degradation enzyme PdBG2. This process is linked to ERH1, an inositol phosphoryl ceramide synthase (IPCS), and glucosylceramide synthase (GCS) functions through physical interactions with the α1-COP protein. In addition, the loss-of-function of α1-COP also alters the subcellular localization of ERH1 and GCS proteins, results in a reduction of GlcCers and GlcHCers molecules, which are the key SL species for lipid raft formation. According to our findings, we propose that α1-COP protein, together with the SL modifiers controlling lipid raft compositions, regulates the function of GPI-anchored PD proteins and hence the callose turnover at PD and symplastic movement of biomolecules. Our findings provide the first key clue to link the COPI-mediated intracellular trafficking pathway to the callose-mediated intercellular signaling pathway through PD.One-sentence summaryPlant-specific coatomer protein functions as a negative regulator of callose accumulation by regulating the translocation of sphingolipid enzymes.

Precision genome editing is highly desired for crop improvement. The recently emerged CRISPR/Cas technology offers great potential applications in precision plant genome engineering. A prime editing (PE) approach combining a reverse transcriptase (RT) with a Cas9 nickase and a “priming” extended guide RNA has shown a high frequency for precise genome modification in mammalian cells and several plant species. Nevertheless, the applications of the PE approach in dicot plants are still limited and inefficient. We designed and tested prime editors for precision editing of a synthetic sequence in a transient assay and for desirable alleles of 10 loci in tomato by stable transformation. Our data obtained by targeted deep sequencing also revealed only low PE efficiencies in both the tobacco and tomato systems. Further assessment of the activities of the PE components uncovered potential reasons for the inefficiency of the PE complexes. Modifying the pegRNA sequences by shortening or introducing mismatches to the primer binding sequences (PBS) in order to reduce their melting temperatures (Tm) did not enhance the PE efficiency at the SlBMP21, SlALC and SlALS1 loci. Our data show challenges of PE approach in tomato, indicating a further improvement of the PE system for the successful applications such as use of improved expression systems. Our work provides an important clue for the successful application of the PE approach in crop improvement.

Behavioral intention was positively predicted by attitude toward the chatbot and facilitating conditions (PP=.03, respectively). The research model explained 55.4% of the variance in the use of anti-TB chatbots. The moderating effect of TB history was found in the relationship between attitude toward the chatbot and behavioral intention (P=.01) and between facilitating conditions and behavioral intention (P=.02). CONCLUSIONS This study can be used to inform future design of anti-TB chatbots and highlight the importance of services and the environment that empower people to use the technology.

본 연구는 제안된 휠체어 사용자 조사 방법론을 통해 척수 손상 장애인인 전동 휠체어 사용자의 섀도잉(인터뷰와 관찰) 결과이다. 전동 휠체어의 기본 컨트롤러, 개조 컨트롤러, 특수 컨트롤러 사용자가 휠체어 및 관련 보조기기, 보조인, 환경과 어떻게 상호작용하는지를 설명한다. 주제 분석 방법을 사용하여 섀도잉 결과를 분석하였으며 분석 결과, 세 가지 상호작용에서의 이슈 사항과 디자인 개선점을 도출하였다. 사용자-제품 상호작용에서는 사용자 몸과 같이 움직일 수 있는 전동 휠체어, 다른 보조기기의 컨트롤러 공유, 다양한 컨트롤러의 형태와 종류 고안, 짐 보관에 대한 고려가 필요하며, 사용자-보조인 상호작용에서는 휠체어 사용 전 과정, 특히 외출 준비에서의 보조인과의 의사소통을 고려해야 하며, 사용자-환경 상호작용에서는 휠체어 사용자가 불안감을 느끼거나 위험한 상황에 놓이게 되는 것을 예방하기 위한 해결책 강구와 같은 디자인 함의가 도출되었다. 이러한 섀도잉 관찰결과는 중증 척수 손상 장애인이 전동 휠체어와 어떻게 상호작용하는지에 대해 논의하였고, 논의 결과가 전동 휠체어 디자인 가이드로 활용될 수 있기를 기대한다.

기존 장애인의 휠체어 사용에 대한 연구 방법들은 대부분 통제 연구로 진행되어 왔다. 본 연구의 목적은 디자이너 또는 디자인 연구자가 기존 방법만을 통해서는 사용자의 실제 제품 사용에 대한 고려를 풍부하게 할 수 없다는 한계점을 보완하기 위한 사용자 조사 방법을 모색하고자 국내 척수 손상 장애인의 휠체어 디자인을 위한 사용자 조사 방법을 제안하는 데에 있다. 국내의 휠체어 디자인 및 사용성 관련 선행 연구, 국내의 장애인 사용자 조사 관련 선행 연구를 바탕으로 휠체어 사용자 조사 방법(인터뷰와 관찰)을 계획하였으며 척수 손상 장애인 7인을 대상으로 휠체어 사용자 조사를 진행하였고 연구 결과를 바탕으로 사용자 조사 방법에 대한 고찰을 진행하였다. 본 연구에서 제안하는 사용자 조사 방법은 사전 인터뷰, 관찰, 사후 인터뷰로 구성되어 있다. 사전 인터뷰에서는 사용자의 특성으로 기본 정보(장애 유형, 진단명, 일상생활 잔존기능, 합병증, 직업, 주 활동 보조인), 실내/실외 활동 보조기기, 실내/실외 활동으로 살펴보았다. 이는 휠체어 사용에 영향을 주는 요인을 분석하기 위함이다. 관찰은 휠체어를 사용하여 외출 준비, 이동, 작업, 마무리 4가지 시나리오에 따라 사용자와 휠체어, 보조기기, 사용자와 보조인, 사용자와 환경이라는 3가지 인터랙션의 사용 현황과 니즈를 살펴본다. 사후 인터뷰에서는 관찰 시 나타났던 사용 상 특이점에 대해 조사하였고, 휠체어 사용 만족도 평가를 진행하는 것으로 구성된다. 본 연구에서 제안한 사용자 조사 방법은 실제 휠체어 사용 맥락의 복잡성을 반영할 수 있도록 ‘사용자와 휠체어, 보조기기, 보조인, 환경’ 간의 다양한 인터랙션에 중점을 두고 관찰 할 수 있는 장점이 있다.

Background Spontaneous double-stranded DNA breaks (DSBs) frequently occur within the genome of all living organisms and must be well repaired for survival. Recently, more important roles of the DSB repair pathways that were previously thought to be minor pathways, such as single-strand annealing (SSA), have been shown. Nevertheless, the biochemical mechanisms and applications of the SSA pathway in genome editing have not been updated. Purpose and scope Understanding the molecular mechanism of SSA is important to design potential applications in gene editing. This review provides insights into the recent progress of SSA studies and establishes a model for their potential applications in precision genome editing. Summary and conclusion The SSA mechanism involved in DNA DSB repair appears to be activated by a complex signaling cascade starting with broken end sensing and 5'-3' resection to reveal homologous repeats on the 3' ssDNA overhangs that flank the DSB. Annealing the repeats would help to amend the discontinuous ends and restore the intact genome, resulting in the missing one repeat and the intervening sequence between the repeats. We proposed a model for CRISPR-Cas-based precision insertion or replacement of DNA fragments to take advantage of the characteristics. The proposed model can add a tool to extend the choice for precision gene editing. Genome editing via the homology-directed repair (HDR) pathway in somatic plant cells is very inefficient compared to error-prone repair by nonhomologous end joining (NHEJ). Here, we increased HDR-based genome editing efficiency approximately 3-fold compared to a Cas9-based single-replicon system via the use of de novo multi-replicon systems equipped with CRISPR/LbCpf1 in tomato and obtained replicon-free but stable HDR alleles. The efficiency of CRISPR/LbCpf1-based HDR was significantly modulated by physical culture conditions such as temperature and light. Ten days of incubation at 31°C under a light/dark cycle after Agrobacterium-mediated transformation resulted in the best performance among the tested conditions. Furthermore, we developed our single-replicon system into a multi-replicon system that effectively increased HDR efficiency. Although this approach is still challenging, we showed the feasibility of HDR-based genome editing of a salt-tolerant SlHKT1;2 allele without genomic integration of antibiotic markers or any phenotypic selection. Self-pollinated offspring plants carrying the HKT1;2 HDR allele showed stable inheritance and germination tolerance in the presence of 100 mM NaCl. Our work may pave the way for transgene-free editing of alleles of interest in asexually as well as sexually reproducing plants.

In this paper, we empirically investigate the effects of monetary policy shocks on exchange rates in Asian countries. To do so, we use VAR models which impose sign restrictions on impulse responses to identify monetary policy shocks. We find that contractionary monetary policy shocks lead to significant exchange rate appreciation in Malaysia, the People’s Republic of China, and the Republic of Korea. However, in India, Indonesia, the Philippines and Thailand, we find either a significant depreciation or no significant effect. These results suggest that an interest rate increase (or decrease) may not necessarily shield Asian countries from exchange rate depreciation (or appreciation) pressure following a U.S. interest rate increase (or decrease).

This paper analyses the conduct and the effects of macroprudential policy in 11 Asian economies, some of which used macroprudential instruments frequently already before the Great Financial Crisis. Data are quarterly for the period 2000–2014. The frequency of macroprudential measures has generally increased after the crisis, and policies are generally tightened during the upswing of a bank credit cycle. Estimates from panel vector autoregression models show that contractionary macroprudential policy shocks have negative effects on credit and output that are qualitatively similar to those of monetary policy. The results suggest that policy conflicts when credit growth is strong and the real economy is weak may be resolved through complementary use of macroprudential and monetary policy instruments.

We report the synthesis and characterization of a series of fused molecular triads, consisting of tetrathiafulvalene-pyrrole derivatives annulated with pyrrole, benzene, thiophene, furan, pyrazine and thiadiazole. Unique and readily tunable optical and electrochemical features are displayed by these new systems.

Influence of precursor intermediates (PI) on the synthesis of high efficient green emitting β-Ca2SiO4:Eu2+ (β-C2S:Eu) phosphors were demonstrated by a novel liquid-phase-precursor (LPP) method utilizing (a) CaCl2–EuCl3, (b) Ca(NO3)2–EuCl3, (c) CaCl2–Eu(NO3)3, and (d) Ca(NO3)2–Eu(NO3)3 combinations as precursors. The crystallinity and morphology of the PI analyzed using X-ray diffraction and high-resolution transmission electron microscopy measurements showed both the amorphous and crystalline type precursor intermediates (API and CPI) depending upon the metal-source materials. The final morphology and luminescence properties of β-C2S:Eu phosphors were found to be strongly influenced by the crystallinity (amorphous and crystalline) and morphology of the PIs. The APIs have led to regular-shaped homogeneous phosphor particles with higher luminescence when compared with the CPIs.

Synthesis of oxynitride solid solutions CaAl4-xSixO7-xNx:Eu2+ (x = 0 - 4) was attempted by the solid state reaction (SSR) methods using Si3N4 and AlN as nitrogen sources. The Ca3Al8Si4O17N4 (x = 4/3) sample with the high phase purity was obtained when AlN was used as a nitrogen source whereas the sample synthesized using Si3N4 as another nitrogen source contained a Ca2Al2SiO7 impurity. Thus, it was revealed that AlN was a preferable nitrogen source for the synthesis of Ca3Al8Si4O17N4 by the SSR method. The solid solutions around x = 4/3 activated with Eu2+ exhibited bluish-green luminescence with emission maxima at 480 nm by the excitation at 250 - 450 nm. Thus, the CaAl4-xSixO7-xNx: Eu2+ solid solutions especially for Ca3Al8Si4O17N4:Eu2+ (x = 4/3) were developed as novel Eu2+

We have conducted two kinds of the so-called charge-compensated aliovalent element substitutions to control the photoluminescence properties of NaAlSiO 4:Eu 2+ with a special focus on the enhancement of the excitation intensity at 400 nm. The aliovalent element substitutions include cation–cation and cation–anion co-substitutions according to the general formulas Na 1−x M x Al 1+x Si 1−x O 4:Eu 2+ and Na 1−x M x AlSiO 4−x N x :Eu 2+ (M=Mg 2+, Ca 2+, and Sr 2+), respectively. The increase in the relative excitation intensity at 400 nm has been achieved in both types of the co-substitutions. Thus, the present research has demonstrated the effectiveness of the charge-compensated element substitution.

We explored heritabilities of the State and Trait Anxiety Inventory (STAI) and the Alcohol Use Disorders Identification Test (AUDIT), and associations including genetic and environmental correlations between the phenotypes among Korean twins and their families. We analyzed the data of 1,748 participants (835 men, 913 women, 656 individuals of monozygotic twins, 173 individuals of same-sexed dizygotic twins, 919 non-twin family members, age 30–79 years) from the Healthy Twin study. Heritabilities and bivariate analyses were assessed using the SOLAR package software. In the methods of generalized estimation equations, women in the 4th quartile of state and trait scores were 17% and 15%, respectively more likely to be hazardous alcohol users compared to women in the lower three quartiles (P< .05). However, there were no significant associations between these phenotypes in men. After adjusting for age and squared age, the heritability estimates were 0.26 in men and 0.34 in women for the state score; for the trait score, 0.35 in men and 0.31 in women; for the AUDIT score, 0.32 in men and 0.37 in women (P< .001). After adjusting for age and squared age, there was a significant genetic correlation between the trait score and the AUDIT score, and a significant non-genetic correlation between the state score and the AUDIT score in women, while there were no significant genetic or non-genetic correlations between these phenotypes in men. The STAI and AUDIT scores are heritable in Koreans and the relationships between these phenotypes may be inconsistent by sex.

We report on the growth and the characterization of magnetite thin films grown on Si(111) substrates by using pulsed laser deposition at different substrate temperatures (TS = 350, 450 and 550 ◦C) by performing X-ray absorption spectroscopy (XAS) and X-ray magnetic circular dichroism (XMCD) experiments. The O K− and Fe L3,2−edge XAS spectra reveal that the couplings of the O 2p with Fe 3d orbitals highly rely on the growth processes. The XMCD data of well-characterized thin films exhibit characteristic contributions from Fe A ions in tetrahedral sites and Fe 2+ B and Fe B ions in octahedral sites and have spectral features similar to those of the Fe3O4 single crystal. The temperature dependence of the XMCD line shape features remain unchanged below and above the Verwey transition. These investigations demonstrate the experimental conditions for controlled growth of Fe3O4 thin films on Si(111) substrates and suggest that the Fe3O4/Si(111) might a promising spintronic materials for future technology.
