This study developed an efficient method using hyperspectral camera for detecting diesel content in soils with spectral indices. Over 70 days of the experiment, clean soils were saturated with diesel, and 186 measurements were taken to monitor the evaporation rate and spectral variation. The diesel evaporation followed a logarithmic pattern, where the diesel volatility decreased from 1.57% per day during the initial period to 0.06% per day during the late period. Using the hull-quotient reflectance at 2236 nm, the diesel content prediction model derived from a stepwise multiple linear regression (SMLR) achieved satisfactory accuracy with sufficient statistical significance (R2 = 0.89, RPD = 2.52). This spectral band was well visualized for diesel presence in hyperspectral images as the band infers variations in two absorptions (CH/AlOH and CH) concurrently. Additionally, this study presented an age estimation model based on the diesel evaporation rate using the same spectral band. Given the fact that this study is based on the largest number of samples with the longest observation period and models were developed excluding atmospheric absorption bands, the simple form of the spectral index makes it applicable to large-scale diesel pollution detection with hyperspectral scanners or narrow-band multispectral cameras in real-world cases.

Background After coronary stent implantation, prolonged dual antiplatelet therapy (DAPT) increases bleeding risk, requiring personalization of DAPT duration. The aim of this study was to develop and validate a machine learning model to predict optimal DAPT duration after contemporary drug‐eluting stent implantation in patients with coronary artery disease. Methods and Results The One‐Month DAPT, RESET (Real Safety and Efficacy of 3‐Month Dual Antiplatelet Therapy Following Endeavor Zotarolimus‐Eluting Stent Implantation), and IVUS‐XPL (Impact of Intravascular Ultrasound Guidance on Outcomes of Xience Prime Stents in Long Lesion) trials provided a derivation cohort (n=6568). Using the X‐learner approach, an individualized DAPT score was developed to determine the therapeutic benefit of abbreviated (1–6 months) versus standard (12‐month) DAPT using various predictors. The primary outcome was major bleeding; the secondary outcomes included 1‐year major adverse cardiac and cerebrovascular events and 1‐year net adverse clinical events. The risk reduction with abbreviated DAPT (3 months) in the individualized DAPT–determined higher predicted benefit group was validated in the TICO (Ticagrelor Monotherapy After 3 Months in the Patients Treated With New Generation Sirolimus‐Eluting Stent for Acute Coronary Syndrome) trial (n=3056), which enrolled patients with acute coronary syndrome treated with ticagrelor. The validation cohort comprised 1527 abbreviated and 1529 standard DAPT cases. Major bleeding occurred in 25 (1.7%) and 45 (3.0%) patients in the abbreviated and standard DAPT groups, respectively. The individualized DAPT score identified 2582 (84.5%) participants who would benefit from abbreviated DAPT, which was significantly associated with a lower major bleeding risk (absolute risk difference [ARD], 1.26 [95% CI, 0.15–2.36]) and net adverse clinical events (ARD, 1.59 [95% CI, 0.07–3.10]) but not major adverse cardiac and cerebrovascular events (ARD, 0.63 [95% CI, −0.34 to 1.61]), compared with standard DAPT in the higher predicted benefit group. Abbreviated DAPT had no significant difference in clinical outcomes of major bleeding (ARD, 1.49 [95% CI, −1.74 to 4.72]), net adverse clinical events (ARD, 2.57 [95% CI, −1.85 to 6.99]), or major adverse cardiac and cerebrovascular events (ARD, 1.54 [95% CI, −1.26 to 4.34]), compared with standard DAPT in the individualized DAPT–determined lower predicted benefit group. Conclusions Machine learning using the X‐learner approach identifies patients with acute coronary syndrome who may benefit from abbreviated DAPT after drug‐eluting stent implantation, laying the groundwork for personalized antiplatelet therapy.

Hand and gaze pointers are effective visual communication cues for remote collaboration. In this paper, we studied the effect of combining them when two basic visual cues (hand gestures and sketches) are available. We conducted a user study with 24 participants, performing two tasks (Tangram and Origami) under four experimental conditions with two independent variables (added hand and gaze pointers) when hand gesture and sketch cues were available as the baseline. The results demonstrated that the added hand and gaze pointer cues improved the co-presence and reduced the task load for remote experts. Additionally, the added gaze pointer cue was effective in fostering behavioral interdependence and message understanding. Participants most preferred the condition of combining all cues, followed by the condition with added gaze pointer, which resulted in quicker task completion and more accurate communication between collaborators. These findings suggest that adding hand and gaze pointer cues can significantly enhance the user experience in remote collaboration.

Aim: To determine the trends in the usage of antimicrobial drugs by patients with pneumonia with prescriptions from long-term care (LTC) hospitals in the Republic of Korea. Method: This retrospective study was conducted from 2011 to 2022 using the National Health Insurance Review and Assessment Service claim data in Korea. We calculated antibiotic usage expressed as a daily defined dose (DDD) per 1000 patients per day (DID). Results: The number of patients with pneumonia in LTC hospitals increased by 2.7 times, from 30,000 in 2011 to 79,000 in 2022. Furthermore, antibiotic consumption per episode by patients with pneumonia in LTC hospitals increased from 17.14 DDD in 2011 to 18.11 DDD in 2022. Abstract Heat networks play a vital role in the energy sector by offering thermal energy to residents in certain countries. Effective management and optimization of heat networks require a deep understanding of users' heat usage patterns. Irregular patterns, such as peak usage periods, can exceed the design capacities of the system. However, previous work has mostly neglected the analysis of heat usage profiles or performed on a small scale. To close the gap, this study proposes a data-driven approach to analyze and predict heat load in a district heating network. The study uses data from over eight heating seasons of a cogeneration DH plant in Cheongju, Korea, to build analysis and forecast models using supervised machine learning (ML) algorithms, including support vector regression (SVR), boosting algorithms, and multilayer perceptron (MLP). The models take weather data, holiday information, and historical hourly heat load as input variables. The performance of these algorithms is compared using different training sample sizes of the dataset. The results show that boosting algorithms, particularly XGBoost, are more suitable ML algorithms with lower prediction errors than SVR and MLP. Finally, different explainable artificial intelligence approaches are applied to provide an in-depth interpretation of the trained model and the importance of input variables.

We investigate when and how the relations of galaxy morphology and star-forming activity with clustercentric radius become evident in galaxy clusters. We identify 162 galaxy clusters with total mass ${{M}}_{\mathrm{tot}}^{\mathrm{cl}}\gt 5\times {10}^{13}{{M}}_{\odot }$ at z = 0.625 in the Horizon Run 5 cosmological hydrodynamical simulation and study how the properties of the galaxies with stellar mass M _* > 5 × 10 ^9 M _⊙ near the cluster main progenitors have evolved in the past. Galaxies are classified into disk, spheroid, and irregular morphological types according to the asymmetry and Sérsic index of their stellar mass distribution. We also classify galaxies into active and passive ones depending on their specific star formation rate. We find that the morphology–clustercentric radius relation (MRR) emerges at z ≃ 1.8 as the fraction of spheroidal types exceeds 50% in the central region ( d ≲ 0.1 R _200 ). Galaxies outside the central region remain disk-dominated. Numerous encounters between galaxies in the central region seem to be responsible for the morphology transformation from disks to spheroids. We also find that the star formation activity–clustercentric radius relation emerges at an epoch different from that of MRR. At z ≃ 0.8, passive galaxies start to dominate the intermediate radius region (0.1 ≲ d / R _200 ≲ 0.3), and this “quenching region” grows inward and outward thereafter. The region dominated by early-type galaxies (spheroids and passive disks) first appears at the central region at z ≃ 1.8, expands rapidly to larger radii as the population of passive disks grows in the intermediate radii, and clusters are dominated by early types after z ≃ 0.8.

We propose a new method for finding galaxy protoclusters that is motivated by structure formation theory and also directly applicable to observations. We adopt the conventional definition that a protocluster is a galaxy group whose virial mass M _vir < M _cl at its epoch, where M _cl = 10 ^14 M _⊙ , but would exceed that limit when it evolves to z = 0. We use the critical overdensity for complete collapse at z = 0 predicted by the spherical top-hat collapse model to find the radius and total mass of the regions that would collapse at z = 0. If the mass of a region centered at a massive galaxy exceeds M _cl , the galaxy is at the center of a protocluster. We define the outer boundary of a protocluster as the zero-velocity surface at the turnaround radius so that the member galaxies are those sharing the same protocluster environment and showing some conformity in physical properties. We use the cosmological hydrodynamical simulation Horizon Run 5 ( HR5 ) to calibrate this prescription and demonstrate its performance. We find that the protocluster identification method suggested in this study is quite successful. Its application to the high-redshift HR5 galaxies shows a tight correlation between the mass within the protocluster regions identified according to the spherical collapse model and the final mass to be found within the clusters at z = 0, meaning that the regions can be regarded as the bona fide protoclusters with high reliability. We also confirm that the redshift-space distortion does not significantly affect the performance of the protocluster identification scheme.

The bovine coronavirus (BCoV) KBR-1 strain, obtained from calf diarrhea samples collected in 2017, belongs to group GIIa. To attenuate this strain, it was subcultured continuously (up to 79 times) in HRT-18 cells, followed by 80–120 passages in MDBK cells. The KBR-1-p120 strain harvested from MDBK cells at passage 120 harbored 13 amino acid mutations in the spike gene. Additionally, the KBR-1-p120 strain showed a high viral titer and cytopathogenic effects in MDBK cells. Seven-day-old calves (negative for BCoV antigen and antibodies) that did not consume colostrum were orally inoculated with the attenuated candidate strain (KBR-1-p120), or with KBR-1 passaged 10 times (KBR-1-p10) in HRT-18 cells. Calves inoculated with KBR-1-p10 had a low diarrhea score, and BCoV RNA was detected at 3–7 days post-inoculation (DPI). The virus was also present in the duodenum, jejunum, and ileum at autopsy; however, calves inoculated with KBR-1-p120 had low levels of BCoV RNA in feces at 4–6 DPI, and no diarrhea. In addition, an extremely small amount of BCoV RNA was present in the jejunum and ileum at autopsy. The small intestines of calves inoculated with KBR-1-p120 were emulsified and used to infect calves two more times, but pathogenicity was not recovered. Therefore, the KBR-1-p120 strain has potential as a live vaccine candidate.

We study the formation and evolution of dark galaxies using the IllustrisTNG cosmological hydrodynamical simulation. We first identify dark galaxies with stellar-to-total mass ratios, M _* / M _tot , smaller than 10 ^−4 , which differ from luminous galaxies with M _* / M _tot ≥ 10 ^−4 . We then select the galaxies with a dark matter halo mass of ∼10 ^9 h ^−1 M _⊙ for mass completeness and compare their physical properties with those of luminous galaxies. We find that, at the present epoch ( z = 0), dark galaxies are predominantly located in void regions without star-forming gas. We also find that dark galaxies tend to have larger sizes and higher spin parameters than luminous galaxies. In the early universe, dark and luminous galaxies show small differences in the distributions of spin and local environment estimates, and the difference between the two samples becomes more significant as they evolve. Our results suggest that, unlike luminous galaxies, dark galaxies tend to be initially formed in less dense regions and could not form stars because of heating from cosmic reionization and few interactions and mergers with other systems containing stars. This study based on numerical simulations can provide important hints for validating dark galaxy candidates in observations and for constraining galaxy formation models.

One intriguing approach for studying the dynamical evolution of galaxy clusters is to compare the spatial distributions among various components such as dark matter, member galaxies, gas, and intracluster light (ICL). Utilizing the recently introduced weighted overlap coefficient (WOC), we analyze the spatial distributions of components within 174 galaxy clusters ( M _tot > 5 × 10 ^13 M _⊙ , z = 0.625) at varying dynamical states in the cosmological hydrodynamical simulation Horizon Run 5. We observe that the distributions of gas and the combination of ICL with the brightest cluster galaxy (BCG) closely resembles the dark matter distribution, particularly in more relaxed clusters, characterized by the half-mass epoch. The similarity in spatial distribution between dark matter and BCG+ICL mimics the changes in the dynamical state of clusters during a major merger. Notably, at redshifts >1, BCG+ICL traced dark matter more accurately than the gas. Additionally, we examined the one-dimensional radial profiles of each component, which show that the BCG+ICL is a sensitive component revealing the dynamical state of clusters. We propose a new method that can approximately recover the dark matter profile by scaling the BCG+ICL radial profile. Furthermore, we find a recipe for tracing dark matter in unrelaxed clusters by including the most massive satellite galaxies together with the BCG+ICL distribution. Combining the BCG+ICL and the gas distribution enhances the dark matter tracing ability. Our results imply that the BCG+ICL distribution is an effective tracer for the dark matter distribution, and the similarity of the spatial distribution may be a useful probe of the dynamical state of a cluster.

We explore the formation of the intragroup light and intracluster light, representing diffuse lights within groups and clusters, from the point that z = 1.5. For this, we perform multiresolution cosmological N -body simulations using the “galaxy replacement technique” and identify the progenitors in which the diffuse light stars existed when they fell into the groups or clusters. Our findings reveal that typical progenitors contributing to diffuse lights enter the host halo with massive galaxies containing a stellar mass of $10\lt \mathrm{log}{M}_{\mathrm{gal}}\,[{M}_{\odot }]\lt 11$ , regardless of the mass or dynamical state of the host halos at z = 0. In cases where the host halos are dynamically unrelaxed or more massive, diffuse lights from massive progenitors with $\mathrm{log}{M}_{\mathrm{gal}}\,[{M}_{\odot }]\gt 11$ are more prominent, with over half of them already preprocessed before entering the host halo. Additionally, we find that the main formation mechanism of diffuse lights is the stripping process of satellites, and a substantial fraction (40%–45%) of diffuse light stars are linked to the merger tree of the brightest cluster galaxy. Remarkably, all trends persist for groups and clusters at higher redshifts. The fraction of diffuse lights in the host halos with a similar mass decreases as the redshift increases, but they are already substantial at z = 1.5 (∼10%). However, it is crucial to acknowledge that detection limits related to the observable radius and faint-end surface brightness may obscure numerous diffuse light stars and even alter the main formation channel of diffuse lights.

During cluster assembly, a cluster’s virialization process leaves behind signatures that can provide information on its dynamical state. However, no clear consensus yet exists on the best way to achieve this. Therefore, we attempt to derive improved recipes for classifying the dynamical states of clusters in observations using cosmological simulations. The cluster halo mass and their subhalos’ mass are used to 10 ^14 M _⊙ h ^−1 and 10 ^10 M _⊙ h ^−1 to calculate five independent dynamical state indicators. We experiment with recipes by combining two to four indicators for detecting specific merger stages, like recent and ancient mergers. These recipes are made by plotting merging clusters and a control sample of relaxed clusters in multiple-indicator parameter space, then applying a rotation matrix method to derive the best way to separate mergers from the control sample. The success of the recipe is quantified using the success rate and overlap percentage of the merger and control histograms along the newly rotated x -axis. This provides us with recipes using different numbers of combined indicators and for different merger stages. Among the recipes, the stellar mass gap and center offset are the first and second most dominant of the indicators, and using more indicators improves the effectiveness of the recipe. When applied to observations, our results show good agreement with literature values of cluster dynamical states.

We test whether Lyα emitters (LAEs) and Lyman-break galaxies (LBGs) can be good tracers of high- z large-scale structures, using the Horizon Run 5 cosmological hydrodynamical simulation. We identify LAEs using the Ly α emission line luminosity and its equivalent width, and LBGs using the broadband magnitudes at z ∼ 2.4, 3.1, and 4.5. We first compare the spatial distributions of LAEs, LBGs, all galaxies, and dark matter around the filamentary structures defined by dark matter. The comparison shows that both LAEs and LBGs are more concentrated toward the dark matter filaments than dark matter. We also find an empirical fitting formula for the vertical density profile of filaments as a binomial power-law relation of the distance to the filaments. We then compare the spatial distributions of the samples around the filaments defined by themselves. LAEs and LBGs are again more concentrated toward their filaments than dark matter. We also find the overall consistency between filamentary structures defined by LAEs, LBGs, and dark matter, with the median spatial offsets that are smaller than the mean separation of the sample. These results support the idea that the LAEs and LBGs could be good tracers of large-scale structures of dark matter at high redshifts.

Abstract This study probed the market share of generic medicines by patients’ age and sex, type of medical facilities, route of administration, number of generic brands, market size (drug expenditure), and therapeutic class and investigated factors associated with a high market share of generic medicines in Korea. We analyzed using national health insurance data between 2010 and 2019. The dependent variable was the generic medicine market share, measured as the number of prescriptions and expenditures. Multivariable regression analysis was conducted using the numbers of generic brands, market size by each ingredient and therapeutic class, relative price, the number of prescriptions, and therapeutic class. Total pharmaceutical expenditures have increased due to the high use of single‐source drugs. The number of prescriptions and expenditures for generic medicines were 0.3 billion prescriptions and $7.8 billion, respectively, accounting for 46.5% and 46% of the total market. Multivariate analysis showed that the number of prescriptions (>20 thousand) and the market size of main active ingredients (>$1 million) were associated with an increased market share, whereas the number of generic brands (compared to

A chimeric pestivirus (KD26_E2LOM) was prepared by inserting the E2 gene of the classical swine fever virus (CSFV) LOM strain into the backbone of the bovine viral diarrhea virus (BVDV) KD26 strain. KD26_E2LOM was obtained by transfecting the cDNA pACKD26_E2LOM into PK-15 cells. KD26_E2LOM chimeric pestivirus proliferated to titers of 106.5 TCID50/mL and 108.0 TCID50/mL at 96 h post-inoculation into PK-15 cells or MDBK cells, respectively. It also reacted with antibodies specific for CSFV E2 and BVDV Erns, but not with an anti-BVDV E2 antibody. Piglets (55–60 days old) inoculated with a high dose (107.0 TCID50/mL) of KD26_E2LOM produced high levels of CSFV E2 antibodies. In addition, no co-habiting pigs were infected with KD26_E2LOM; however, some inoculated pigs excreted the virus, and the virus was detected in some organs. When pregnant sows were inoculated during the first trimester (55–60 days) with a high dose (107.0 TCID50/mL) of KD26_E2LOM, anti-CSFV E2 antibodies were produced at high levels; chimeric pestivirus was detected in one fetus and in the ileum of one sow. When 5-day-old calves that did not consume colostrum received a high dose (107.0 TCID50/mL) of KD26_E2LOM, one calf secreted the virus in both feces and nasal fluid on Day 2. A high dose of KD26_E2LOM does not induce specific clinical signs in most animals, does not spread from animal to animal, and generates CSFV E2 antibodies with DVIA functions. Therefore, chimeric pestivirus KD26_E2LOM is a potential CSFV live marker vaccine.

From late 2013–2022, 1131 cases of porcine epidemic diarrhea (PED) were reported to the Korean Animal Health Integrated System (KAHIS). There were four major outbreaks from winter to spring (2013–2014, 2017–2018, 2018–2019, and 2021–2022), with the main outbreaks occurring in Chungnam (CN), Jeonbuk (JB), and Jeju (JJ). Analysis of the complete spike (S) gene of 140/1131 KAHIS PEDV cases nationwide confirmed that 139 belonged to the G2b genotype and 1 to the G2a genotype. Among them, two strains (K17GG1 and K17GB3) were similar to an S INDEL isolated in the United States (strain OH851), and 12 strains had deletions (nucleotides (nt) 3–99) or insertions (12 nt) within the S gene. PEDVs in JJ formed a regionally independent cluster. The substitution rates (substitutions/site/year) were as follows: 1.5952 × 10−3 in CN, 1.8065 × 10−3 in JB, and 1.5113 × 10−3 in JJ. A Bayesian skyline plot showed that the effective population size of PEDs in JJ fell from 2013–2022, whereas in CN and JB it was maintained. Genotyping of 340 Korean PEDV strains, including the 140 PEDVs in this study and 200 Korean reference strains from GenBank, revealed that only the highly pathogenic non-INDEL type (G2b) was dominant from 2020 onwards. Therefore, it is predicted that the incidence of PED will be maintained by the G2b (non-INDEL) genotype.

Abstract The COVID-19 pandemic is an unprecedented threat to humanity that has provoked global health concerns. Since the etiopathogenesis of this illness is not fully characterized, the prognostic factors enabling treatment decisions have not been well documented. Accurately predicting the progression of the disease would aid in appropriate patient categorization and thus help determine the best treatment option. Here, we have introduced a proteomic approach utilizing data-independent acquisition mass spectrometry (DIA-MS) to identify the serum proteins that are closely associated with COVID-19 prognosis. Twenty-seven proteins were differentially expressed between severely ill COVID-19 patients with an adverse or favorable prognosis. Ingenuity Pathway Analysis revealed that 15 of the 27 proteins might be regulated by cytokine signaling relevant to interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF), and their differential expression was implicated in the systemic inflammatory response and in cardiovascular disorders. We further evaluated practical predictors of the clinical prognosis of severe COVID-19 patients. Subsequent ELISA assays revealed that CHI3L1 and IGFALS may serve as highly sensitive prognostic markers. Our findings can help formulate a diagnostic approach for accurately identifying COVID-19 patients with severe disease and for providing appropriate treatment based on their predicted prognosis.

This study aimed to investigate the genetic diversity of G- and P-type bovine RVAs (BoRVAs) prevalent in Vietnam. Between 2017 and 2018, the prevalence of BoRVAs detected in diarrhea samples from 8 regions was as low as 1.9% (11/582). The prevalence of the G-type was 45.5% for G6 and 18.2% for G10; however, 36.3% remain unidentified. Interestingly, all BoRVAs were investigated as P[11], and there was no diversity within this P-type. Geographically, the G6 and G10 types were not identified in any specific area; rather, they occurred in both Northern and Southern Vietnam. G6P[11] and G10P[11], which are combined G- and P-types, were identified in 71.4% and 28.6% of BoRVA-positive samples, respectively. Phylogenetic tree analysis revealed that the G6-type detected in Vietnamese cows is similar to strains derived from China, Japan, and Korea, whereas the G10 type is closely related to the Chinese strain. In addition, the P11 strain detected in Vietnamese cows is similar to the Spanish and Chinese strains. The BoRVA-positive rate was higher in cows aged less than 2 months (3.2%, 3/94) than in those aged 2 months or more (1.6%, 8/488). In summary, we detected the presence of G6P11 and G10P11 BoVRAs on Vietnamese cow farms, and found that they were more predominant in young calves than in older cows.

In South Korea in 2013, the G1-based vaccine failed to prevent an outbreak of G2b-type porcine epidemic diarrhea virus (PEDV), which is more pathogenic than the traditional G1-type strain, thereby allowing the virus to spread. In 2017 and 2018, field samples were cultured sequentially on Vero cells to isolate HS (virulent) and SGP-M1 (partially attenuated) strains, respectively, of the G2b type. The HS strain harbors a single amino acid (aa) change and two aa deletions in the N-terminal domain of S1 (55I56G57E→55K56Δ57Δ). The SGP-M1 strain harbors a seven aa deletion in the C-terminal domain of S2 (1380~1386ΔFEKVHVQ). By co-infecting various animal cells with these two strains (HS and SGP-M1), we succeeded in cloning strain HSGP, which harbors the mutations present in the two original viruses. The CPE pattern of the HSGP strain was different from that of the HS and SGP-M1 strains, with higher viral titers. Studies in piglets showed attenuated pathogenicity of the HSGP strain, with no clinical symptoms or viral shedding, and histopathologic lesions similar to those in negative controls. These findings confirm that deletion of specific sequences from the S gene attenuates the pathogenicity of PEDV. In addition, HSGP strains created by combining two different strains have the potential for use as novel attenuated live vaccine candidates.

Abstract PTEN is one of the most frequently altered tumor suppressor genes in malignant tumors. The dominant-negative effect of PTEN alteration suggests that the aberrant function of PTEN mutation might be more disastrous than deletion, the most frequent genomic event in glioblastoma (GBM). This study aimed to understand the functional properties of various PTEN missense mutations and to investigate their clinical relevance. The genomic landscape of PTEN alteration was analyzed using the Samsung Medical Center GBM cohort and validated via The Cancer Genome Atlas dataset. Several hotspot mutations were identified, and their subcellular distributions and phenotypes were evaluated. We established a library of cancer cell lines that overexpress these mutant proteins using the U87MG and patient-derived cell models lacking functional PTEN. PTEN mutations were categorized into two major subsets: missense mutations in the phosphatase domain and truncal mutations in the C2 domain. We determined the subcellular compartmentalization of four mutant proteins (H93Y, C124S, R130Q, and R173C) from the former group and found that they had distinct localizations; those associated with invasive phenotypes (‘edge mutations’) localized to the cell periphery, while the R173C mutant localized to the nucleus. Invasive phenotypes derived from edge substitutions were unaffected by an anti-PI3K/Akt agent but were disrupted by microtubule inhibitors. PTEN mutations exhibit distinct functional properties regarding their subcellular localization. Further, some missense mutations (‘edge mutations’) in the phosphatase domain caused enhanced invasiveness associated with dysfunctional cytoskeletal assembly, thus suggesting it to be a potent therapeutic target.

We investigate the formation channels of the intracluster light (ICL) and the brightest cluster galaxy (BCG) in clusters at z = 0. For this, we perform multi-resolution cosmological N -body simulations using the “galaxy replacement technique.” We study the formation channels of the ICL and BCG as a function of distance from the cluster center and the dynamical state of the clusters at z = 0. To do this, we trace back the stars of the ICL and BCG, and identify the stellar components in which they existed when they first fell into the clusters. We find that the progenitors of the ICL and BCG in the central region of the cluster fell earlier and with a higher total mass ratio of the progenitors to the cluster compared to the outer region. This causes a negative radial gradient in the infall time and total mass ratio of the progenitors. Although stellar mass of the progenitors does not show the same radial gradient in all clusters, massive galaxies ( M _gal > 10 ^10 M _⊙ h ^−1 ) are the dominant formation channel of the ICL and BCG for all clusters, except for our most relaxed cluster. For clusters that are dynamically more unrelaxed, we find that the progenitors of the ICL and BCG fall into their clusters more recently, and with a higher mass and mass ratio. Furthermore, we find that the diffuse material of massive galaxies and group-mass halos that is formed by preprocessing contributes significantly to the ICL in the outer region of the unrelaxed clusters.

We investigate the impact of the surface-brightness (SB) limit on the galaxy stellar mass functions (GSMFs) using galaxy catalogs generated from the Horizon Run 5 ( HR5 ) simulation. We compare the stellar-to-halo-mass relation, GSMF, and size–stellar mass relation of the HR5 galaxies with observational data and other cosmological simulations. The mean SB of simulated galaxies are computed using their effective radii, luminosities, and colors. To examine the cosmic SB dimming effect, we compute k -corrections from the spectral energy distributions of individual simulated galaxy at each redshift, apply the k -corrections to the galaxies, and conduct mock surveys based on the various SB limits. We find that the GSMFs are significantly affected by the SB limits at the low-mass end. This approach can ease the discrepancy between the GSMFs obtained from simulations and observations at 0.6 ≲ z ≤ 2. We also find that a redshift survey with an SB selection limit of ${\left\langle {\mu }_{r}\right\rangle }^{e}=$ 25 mag arcsec ^−2 will miss 20% of galaxies with ${M}_{\star }^{g}={10}^{9}\,{M}_{\odot }$ at z = 0.625. The missing fraction of low-surface-brightness galaxies increases to 35%, 55%, and 80% at z = 0.9, 1.1, and 1.9, respectively, at the same SB limit.

We investigate the fraction of quenched satellite galaxies in host galaxy groups and clusters using TNG300 in the IllustrisTNG cosmological magnetohydrodynamical simulations. The simulations show that most satellites are quenched after they fall into their final hosts, and that post-processing is a more dominant mechanism of galaxy quenching than pre-processing. We find that the fraction of quenched satellites at z = 0 increases with host mass, which implies that more massive hosts have higher quenching efficiency because they have more massive groups infalling. Furthermore, we find that hosts that have many early-infall satellites show a higher fraction of quenched satellites at z = 0 than those that have many late-infall satellites, which results in a scatter of the quenched fraction of satellites in a given mass range of hosts at z = 0. Our results highlight the significance of the mass of hosts and the different infall times of satellites in understanding galaxy quenching.

The purpose of this study was to investigate annual changes in BoRVA strains by examining the VP4 and VP7 genes of rotaviruses in Korean calves. Between 2014 and 2018, 35 out of 138 samples of calf diarrhea feces collected nationwide were positive for BoRVA. Further genetic characterization of the VP7 and VP4 genes of 35 BoRVA isolates identified three different G-genotypes (G6, G8, and G10) and two different P genotypes (P[5] and P[11]). The G6 genotype was most common (94.3%) in BoRVA-positive calves, followed by the P[5] genotype (82.9%). Four genotypes comprised combinations of VP4 and VP7: 80% were G6P[5], 14.2% were G6P[11], 2.9% were G8P[5], and 2.9% were G10P[11]. Susceptibility to infection was highest in calves aged < 10 days (35%) and lowest in calves aged 30–50 days (15.4%). The data presented herein suggest that the G6P[5] genotype is the main causative agent of diarrhea in Korean calves. In addition, it is predicted that G6P[5] will continue to act as a major cause of diarrhea in Korean calves.

Porcine group A rotavirus (PoRVA; family, Reovirideae) strains cause acute viral gastroenteritis in piglets (especially suckling and weaned pigs), resulting in significant economic losses. In this study, we analyzed the VP7 and VP4 genes of PoRVA isolated between 2014 and 2018 from domestic pigs in South Korea to investigate the prevalence of predominant circulating genotypes (G and P types). The prevalence of the PoRVA antigen in the diarrheic fecal samples was 14.1% (53/377). Further genetic characterization of the VP7 and VP4 genes of 53 PoRVA isolates identified six different G-genotypes and five different P genotypes. The G4 and G9 genotypes were the most common (each 39.6%) in PoRVA-positive pigs, followed by P[7] and P[6] (33.9% and 30.1%, respectively). Because the G5 and G9 genotype vaccines are currently mainly used in South Korea, this result provides valuable epidemiological information about the genetic characteristics of PoRVA circulating on domestic pig farms. Development of a novel PoRVA vaccine that targets the current strains circulating in South Korea may be required for more effective virus control on pig farms.

Classical swine fever virus (CSFV) is one of the major pathogens that causes severe economic damage to the swine industry. Circulation of CSFV in wild boars carries the potential risk of reintroducing the virus into CSFV-free pig farms. This study carried out a genetic analysis of CSFV isolates from wild boars and analyzed the mtDNA haplotypes of the wild boars. Blood samples (n = 2140) from wild Korean boars captured in 2020 were subjected to qRT-PCR to detect CSFV, which was classified as subgenotype 2.1d based on phylogenetic analysis. CSFV had been detected in wild boars only in northern regions (Gangwon and Gyeonggi) of South Korea between 2011 and 2019. However, CSFV was identified in wild boars in the more southern regions (Chungbuk and Gyeongbuk) in 2020. Based on mitochondrial DNA analysis, all wild boars with CSFV were haplotype 01 (H01). Thus, we presume that the H01 haplotype is more susceptible to CSFV. In the future, infection of wild boars by CSFV is expected to occur intermittently every year, and we predict that most wild boars infected with CSFV will be haplotype H01.

Bovine coronavirus (BCoV) causes severe diarrhea in neonatal calves, winter dysentery in adult cattle, and respiratory disease in feedlot cattle, resulting in economic losses. A total of 16/140 calf diarrheic feces samples collected in South Korea between 2017 and 2018 were positive for BCoV. Phylogenetic analysis of the complete spike and hemagglutinin/esterase genes revealed that the 16 Korean BCoV strains belonged to group GIIa along with Korean strains isolated after 2000, whereas Korean BCoV strains isolated before 2000 belonged to group GI. Mice and goats inoculated with an inactivated KBR-1 strain (isolated from this study) generated higher antibody titers (96 ± 13.49 and 73 ± 13.49, respectively) when mixed with the Montanide01 adjuvant than when mixed with the Carbopol or IMS1313 adjuvants. Viral antigens were detected in the large intestine, jejunum, and ileum of calves inoculated with inactivated KBR-1 vaccine (104.0 TCID50/mL) at 14 days of post-challenge (DPC). However, no viral antigens were detected in calves vaccinated with a higher dose of inactivated KBR-1 strain (106.0 TCID50/mL) at 14 DPC, and they had high antibody titers and stable diarrhea scores. Currently, the group GIIa is prevalent in cows in South Korea, and although further research is needed in the future, the recently isolated KBR-1 strain has potential value as a new vaccine candidate.

This paper describes a community effort to improve earlier versions of the full-text corpus of Genomics & Informatics by semi-automatically detecting and correcting PDF-to-text conversion errors and optical character recognition errors during the first hackathon of Genomics & Informatics Annotation Hackathon (GIAH) event. Extracting text from multi-column biomedical documents such as Genomics & Informatics is known to be notoriously difficult. The hackathon was piloted as part of a coding competition of the ELTEC College of Engineering at Ewha Womans University in order to enable researchers and students to create or annotate their own versions of the Genomics & Informatics corpus, to gain and create knowledge about corpus linguistics, and simultaneously to acquire tangible and transferable skills. The proposed projects during the hackathon harness an internal database containing different versions of the corpus and annotations.

Human Norwalk viruses (HuNoVs), the most common etiological agents of acute gastroenteritis, are genetically diverse RNA viruses that frequently cause mass food poisoning internationally. Although nucleic acid detection methods, such as reverse transcription–quantitative polymerase chain reaction (RT-qPCR), are the gold standard for the diagnosis of norovirus infection, alternative methods are needed for the specific and sensitive viral protein detection for rapid diagnosis and surveillance. In this study, we developed a robust and high-throughput targeted proteomic assay workflow to directly detect the VP1 major capsid protein of HuNoVs. A parallel reaction monitoring (PRM) assay using a high-resolution mass spectrometer was used to detect representative peptides derived from VP1 in six different HuNoV genotypes. An optimized protocol using synthesized heavy isotope-labeled peptides as internal standards was also used to simultaneously genotype and quantify the VP1 protein in human stool specimens. This method is expected to become a new tool for studying the molecular epidemiology of HuNoV and to shed new light on targeted proteomics in clinical practice.

On Jeju Island, South Korea, pigs have not been vaccinated against classical swine fever (CSF) since 1999. Analysis of bovine viral diarrhea virus (BVDV) isolated from pigs on Jeju Island between 2009 and 2019 identified five BVDV-1a strains and one BVDV-1b strain. These BVDV types were shown to be the same types as BVDV strains isolated from neighboring cow farms. BVDV antibody-positive pigs (both BVDV-1 and -2) were also detected at 54 of 168 pig farms during this period. In pig infection experiments using BVDV-1a and -2a strains isolated from neighboring cow farms, BVDV-1a was detected in the blood of one of four pigs infected at both 6 and 35 days post-infection (dpi) and in the blood of two of the four pigs at 28 dpi. Pigs showed higher anti-BVDV-1 titers (5.5 ± 1.5 log2) at 35 dpi. BVDV-2a was detected in the blood of one of four pigs infected with this virus at 28 dpi only, and lower antibody titers (2.75 ± 0.75 log2) were seen in these pigs at 35 dpi. While BVDV infection is not particularly pathogenic in pigs, it is still important to monitor porcine BVDV infections due to a differential diagnosis of CSFV.

Abstract Despite the wide use of mesenchymal stromal cells (MSCs) for paracrine support in clinical trials, their variable and heterogeneous supporting activity pose major challenges. While three-dimensional (3D) MSC cultures are emerging as alternative approaches, key changes in cellular characteristics during 3D-spheroid formation remain unclear. Here, we show that MSCs in 3D spheroids undergo further progression towards the epithelial-mesenchymal transition (EMT), driven by upregulation of EMT-promoting microRNAs and suppression of EMT-inhibitory miRNAs. The shift of EMT in MSCs is associated with widespread histone modifications mimicking the epigenetic reprogramming towards enhanced chromatin dynamics and stem cell-like properties, but without changes in their surface phenotype. Notably, these molecular shifts towards EMT in 3D MSCs caused enhanced stem cell niche activity, resulting in higher stimulation of hematopoietic progenitor self-renewal and cancer stem cell metastasis. Moreover, miRNA-mediated induction of EMT in 2D MSCs were sufficient to mimic the enhanced niche activity of 3D spheroid MSCs. Thus, the molecular hierarchy in the EMT gradient among phenotypically indistinguishable MSCs revealed the previously unrecognized functional parameters in MSCs, and the EMT-enhanced “naïve” mesenchymal state represents an ‘activated mesenchymal niche’ in 3D spheroid MSCs.

To prevent diarrhea in suckling piglets infected by porcine epidemic diarrhea virus (PEDV), porcine epidemic diarrhea (PED) vaccines are administered mainly through intramuscular (IM) or oral routes. We found that growing pigs vaccinated with an inactivated PEDV vaccine via the intradermal (ID) route had higher neutralizing antibody titers and cytokine (IFN-γ, IL-4, and IL-10) levels than non-vaccinated pigs. In addition, suckling piglets acquired lactogenic immunity from pregnant sows inoculated with an ID PED vaccine. We evaluated the efficacy of vaccination via this route, along with subsequent protection against virulent PEDV. At six days post-challenge, the survival rate of suckling piglets exposed to virulent PEDV was 70% for the ID group and 0% for the mock group (no vaccine). At necropsy, villi length in the duodenum and ileum of piglets with lactogenic immunity provided by ID-vaccinated sows proved to be significant (p < 0.05) when compared with those in piglets from mock group sows. Thus, vaccination using an inactivated PED vaccine via the ID route provides partial protection against infection by virulent PEDV.

Herein, we compared the productivity of pigs inoculated with one of two classical swine fever (CSF) vaccines (low virulent of Miyagi (LOM) or Flc-LOM-BErns) plus the swine erysipelothrix rhusiopathiae (SE) vaccine. The feed intake and weight increase of the pigs inoculated with Flc-LOM-BErns + SE were normal. However, the feed intake of the pigs inoculated with LOM + SE dropped sharply from four days post-vaccination (dpv). In addition, the slaughter date was an average of eight days later than that of the pigs inoculated with Flc-LOM-BErns + SE. All pigs inoculated with the Flc-LOM-BErns + SE vaccine were completely differentiated at 14 days against CSF Erns antibody and at approximately 45 days against the bovine viral diarrhea virus (BVDV) Erns antibody; the titers were maintained until slaughter. Leucopenia occurred temporarily in the LOM + SE group, but not in the Flc-LOM-BErns + SE group. Expression of tumor necrosis factor (TNF)-α and IFN-γ was significantly (p < 0.05) higher in the LOM + SE group than in the mock (no vaccine) group. When conducting the same experiment on a breeding farm, the results were similar to those of the laboratory experiments. In conclusion, the biggest advantage of replacing the CSF LOM vaccine with the Flc-LOM-BErns vaccine is improved productivity.

In the information age, the role of data in any industry is getting more and more important. In the construction industry, however, experts’ experience and intuition are still key determinants in decision-making, while other industries achieve tangible improvement in paradigm shifts by adopting cutting-edge information technology. Cost and schedule controls, which are closely connected, are decisive in the success of construction project execution. While a vast body of research has developed methodologies for cost-schedule integration over 50 years, there is no method used in practice; it remains a significant challenge in the construction industry. This study aims to propose a data processing algorithm for integrated cost-schedule data management employing big data technology. It is designed to resolve the main obstacle to the practicality of existing methods by providing integrity and flexibility in integrating cost-schedule data and reducing time on building and changing databases. The proposed algorithm is expected to transform field engineers’ current perception regarding information management as one of the troublesome tasks in a data-friendly way.

Here, we investigated the protective efficacy provided by passive immunity induced by a classical swine fever (Flc-LOM-BErns) vaccine with the newly developed DIVA (Differentiating Infected from Vaccinated Animals) function. Ten pigs (aged 40–60 days) with maternally derived antibodies (MDAs) obtained from sows inoculated with the Flc-LOM-BErns vaccine were challenged with virulent classical swine fever virus (CSFV). Pigs with an MDA titer of 6 log2 induced by the Flc-LOM-BErns vaccine were fully protected against virulent CSFV challenge but not the pigs with an MDA titer under 5 log2. In addition, Flc-LOM-BErns vaccine-derived MDAs successfully differentiated vaccinated pigs by bovine viral diarrhea virus (BVDV) Erns/CSFV Erns antibody detection, functioning as a DIVA.

Here, we examined the pathogenicity and genetic differences between classical swine fever viruses (CSFV) isolated on pig farms in North Vietnam from 2014−2018. Twenty CSFV strains from 16 pig farms were classified as genotype 2 (sub-genotypes 2.1b, 2.1c, and 2.2). The main sub-genotype, 2.1c, was classified phylogenetically as belonging to the same cluster as viruses isolated from the Guangdong region in South China. Strain HY58 (sub-genotype 2.1c), isolated from pigs in Vietnam, caused higher mortality (60%) than the Vietnamese ND20 strain (sub-genotype 2.2). The Vietnamese strain of sub-genotype 2.1b was estimated to have moderate virulence; indeed, genetic analysis revealed that it belongs to the same cluster as Korean CSFV sub-genotype 2.1b. Most CSFVs circulating in North Vietnam belong to sub-genotype 2.1c. Geographical proximity means that this genotype might continue to circulate in both North Vietnam and Southern China (Guangdong, Guangxi, and Hunan).

In Jeju island of South Korea, a classical swine fever (CSF) non-vaccinated region, many pig farmers insisted on abortion and stillbirth in pregnant sows and high mortality of suckling/weaning piglets by circulating CSF virus from 2014 to 2018. We investigated whether CSF viruses isolated from pigs in Jeju Island (Jeju LOM) have recovered their pathogenicity by conducting experiments using pregnant sows and specific pathogen-free (SPF) pigs. The CSF modified live LOM vaccine (MLV-LOM) and Jeju LOM strains induced abortion and stillbirth in pregnant sows. Viral antigens were detected in the organs of fetuses and stillborn piglets in the absence of specific pathological lesions associated with the virulent CSF virus in both groups (MLV-LOM and Jeju LOM strain). However, antigen was detected in one newborn piglet from a sow inoculated with a Jeju LOM strain, suggesting that it may cause persistent infections in pigs. SPF pigs inoculated with the MLV-LOM or Jeju LOM strains were asymptomatic, but virus antigen was detected in several organ and blood samples. Virus shedding in both groups of animals was not detected in the feces or saliva until 21 days post inoculation. The serum concentration of the three major cytokines, IFN-α, TNF-α, and IL-10, known to be related to lymphocytopenia, were similar in both groups when the MLV-LOM or Jeju LOM strains were inoculated into SPF pigs. In conclusion, Jeju LOM strains exhibited most of the characteristics of the MLV-LOM in pigs and resulted in the same adverse effects as the MLV-LOM strain.

Non-small-cell lung cancer (NSCLC) constitutes approximately 80% of all diagnosed lung cancers, and diagnostic markers detectable in the plasma/serum of NSCLC patients are greatly needed. In this study, we established a pipeline for the discovery of markers using 9 transcriptome datasets from publicly available databases and profiling of six lung cancer cell secretomes. Thirty-one out of 312 proteins that overlapped between two-fold differentially expressed genes and identified cell secretome proteins were detected in the pooled plasma of lung cancer patients. To quantify the candidates in the serum of NSCLC patients, multiple-reaction-monitoring mass spectrometry (MRM-MS) was performed for five candidate biomarkers. Finally, two potential biomarkers (BCHE and GPx3; AUC = 0.713 and 0.673, respectively) and one two-marker panel generated by logistic regression (BCHE/GPx3; AUC = 0.773) were identified. A validation test was performed by ELISA to evaluate the reproducibility of GPx3 and BCHE expression in an independent set of samples (BCHE and GPx3; AUC = 0.630 and 0.759, respectively, BCHE/GPx3 panel; AUC = 0.788). Collectively, these results demonstrate the feasibility of using our pipeline for marker discovery and our MRM-MS platform for verifying potential biomarkers of human diseases.

Mass spectrometric (MS) data of human cell secretomes are usually run through the conventional human database for identification. However, the search may result in false identifications due to contamination of the secretome with fetal bovine serum (FBS) proteins. To overcome this challenge, here we provide a composite protein database including human as well as 199 FBS protein sequences for MS data search of human cell secretomes. Searching against the human-FBS database returned more reliable results with fewer false-positive and false-negative identifications compared to using either a human only database or a human-bovine database. Furthermore, the improved results validated our strategy without complex experiments like SILAC. We expect our strategy to improve the accuracy of human secreted protein identification and to also add value for general use.
