Objective To report the diagnosis and treatment of a patient with meningeal metastases from lung adenocarcinoma, and explore the significance of cerebrospinal fluid (CSF) gene detection for clinical treatment and the efficacy and safety of double⁃dose icotinib in the treatment of meningeal metastases from lung adenocarcinoma with EGFR gene sensitive mutation. Methods and Results A 56⁃year⁃old female patient with meningeal metastasis from lung adenocarcinoma was examined for CSF and blood double⁃sample genes. The CSF had an exon 19 deletion mutation in the EGFR gene, and no sensitive mutations were found in blood sample. Oral icotinib hydrochloride 250 mg/time (3 times/d), and chest CT and head MRI enhanced scans were re⁃examined every 8 weeks, suggesting that the lung tumor was significantly reduced, and the degree of jumbo meningeal enhancement was lessened than before. Right lateral ventricle⁃peritoneal shunt was performed to effectively reduce intracranila pressure (ICP) and relieve clinical symptoms. Up to September 21, 2019, patient had been taking double doses of icotinib for 589 d, and her condition was stable. The main adverse drug reaction is rash. She is still taking medication and is being followed up. Conclusions For patients with meningeal metastases from lung adenocarcinoma, CSF gene detection of EGFR gene sensitive mutation can guide clinical development of treatment strategies. Double⁃dose icotinib is effective in treating meningeal metastases from lung adenocarcinoma with EGFR gene sensitive mutation, and is well tolerated and safe.

Objective To report the diaynosis and afatinib treatment of a patient with leptomeningeal metastasis of lung adenocarcinoma with epidermal growth factor receptor (EGFR) exon18: G719A mutation, detected in cerebrospinal fluid. Methods Cerebrospinal fluid specimens were obtained by lumbar puncture. EGFR mutation status was detected by next⁃generation sequencing (NGS) method. Afatinib molecular targeted drug therapy was given after positive results were obtained. Results The cerebrospinal fluid EGFR gene test of the patient suggested exon18: G719A mutation. After 8 weeks of afatinib treatment, chest CT showed that the lung lesions were smaller than before, and head MRI showed no obvious intracranial changes. The review indicated that the patient's tumor condition was stable and he had taken the medicine for 55 weeks. The main adverse drug reaction was rash (Grade 1). Conclusions EGFR mutation in cerebrospinal fluid detection of patients with leptomeningeal metastasis of lung adenocarcinoma can guidethe clinical treatment strategy. Afatinib in treating leptomeningeal metastasisof lung adenocarcinoma accompanied by EGFR gene exon18: G719A mutation has definite clinical efficacy andistolerable. Traffic prediction has attracted a lot of attention in recent years. However, it is challenging due to the dynamic and heterogeneous spatial–temporal correlations. Most of the existing methods adopt the attention mechanism to capture the dynamic spatial features, but it is difficult for the attention mechanism to learn the real-time change of spatial dependencies, which restrict accurate spatial dependencies learning. Furthermore, most methods are out at elbows when solving heterogeneous spatial–temporal data. To overcome these problems, we propose a novel traffic prediction model called Dynamic spatial–temporal Heterogeneous Graph Convolution Network. Different from the existing methods, we have designed a dynamic localized graph and a corresponding adaptive localized graph convolution network, which are capable of simultaneously capturing dynamic and heterogeneous spatial correlations. We propose a gated adaptive temporal convolution network to capture the temporal heterogeneity of traffic data and enjoy global receptive fields. Finally, a global correlations fusion network is provided to incorporate the global spatial–temporal correlations. This study examined the antibacterial effect of protoporphyrin IX-ethylenediamine derivative (PPIX-ED)-mediated photodynamic antimicrobial chemotherapy (PPIX-ED-PACT) against Pseudomonas aeruginosa in vitro and in vivo. PPIX-ED potently inhibited the growth of Pseudomonas aeruginosa by inducing reactive oxygen species production via photoactivation. Atomic force microscopy revealed that PPIX-ED-PACT induced the leakage of bacterial content by degrading the bacterial membrane and wall. As revealed using acridine orange/ethidium bromide staining, PPIX-ED-PACT altered the permeability of the bacterial membrane. In addition, the antibacterial effect of PPIX-ED-PACT was demonstrated in an in vivo model of P. aeruginosa-infected wounds. PPIX-ED (100 μM) decreased the number of P. aeruginosa colony-forming units by 4.2 log10. Moreover, histological analysis illustrated that the wound healing rate was 98% on day 14 after treatment, which was 10% higher than that in the control group. According to the present findings, PPIX-ED-PACT can effectively inhibit the growth of P. aeruginosa in vitro and in vivo.

Despite the essential role of the corticospinal tract (CST) in controlling voluntary movements, successful regeneration of large numbers of injured CST axons beyond a spinal cord lesion has never been achieved. We found that PTEN/mTOR are critical for controlling the regenerative capacity of mouse corticospinal neurons. After development, the regrowth potential of CST axons was lost and this was accompanied by a downregulation of mTOR activity in corticospinal neurons. Axonal injury further diminished neuronal mTOR activity in these neurons. Forced upregulation of mTOR activity in corticospinal neurons by conditional deletion of Pten, a negative regulator of mTOR, enhanced compensatory sprouting of uninjured CST axons and enabled successful regeneration of a cohort of injured CST axons past a spinal cord lesion. Furthermore, these regenerating CST axons possessed the ability to reform synapses in spinal segments distal to the injury. Thus, modulating neuronal intrinsic PTEN/mTOR activity represents a potential therapeutic strategy for promoting axon regeneration and functional repair after adult spinal cord injury.

Spatial–temporal graph neural network has drawn more and more attention in recent years and is widely used to various real-world applications. However, learning the spatial–temporal graph neural network structure presents unique challenges including: (i) the dynamic spatial correlation; (ii) the dynamic temporal correlation. Even the existing methods take into account the spatial correlation, they still learn the static road network structure information, which cannot reflect the dynamic of road relations. Some of the works has focused on modeling the long-term time series, but the improvements have been limited tightly. To overcome these challenges, we proposed a novel approach called Multi-View Spatial–Temporal Graph Neural Network. Differ from the existing research, we designed a multi-view temporal transformer module to extract dynamic temporal correlation and enhance the expression of medium and long-term temporal features. We propose a multi-view spatial structure and a corresponding multi-view graph convolutional module, which are capable of simultaneously combining the features of static road network structure and dynamic changes.
