Significance To systematically address problems related to the interpretation and evaluation of image logs and expand their application range in geology and engineering fields, firstly, the development history, data processing steps and interpretation process of image log acquisition series are reviewed, and then fine interpretation methods for image logs, constrained by "four levels" (direct interpretation of images, conventional logging constraints, core constraints and geological theory constraints), are proposed in this paper. Progress The image log facies models can be divided into seven categories: massive, banded, cross-stratified, spotted, convoluted, graded and symmetrical groove modes. Image logs can be used for core orientation, formation attitude acquisition, fracture evaluation, fault property analysis, in situ stress direction discrimination, sedimentary characteristic pickup, palaeocurrent direction discrimination, etc. Through the inverse calculation of Archie's formula, the image log data matrix can generate porosity spectra and apparent formation water resistivity spectra, which can be used for reservoir quality evaluation and fluid property identification, respectively. Slab images can reveal information such as sedimentary laminae, fine-grained sediment lamina textures can be identified, and high-resolution logging can be used to evaluate unconventional oil and gas reservoirs. Poro Tex image structure analysis technology can be used to identify the characteristics of reservoir spaces such as pores and fractures and to quantitatively calculate parameters such as porosity and fracture porosity. Therefore, it is widely used in comprehensive logging evaluations of highly heterogeneous carbonate reservoirs. Conclusions and Prospects These research results can help us to avoid misunderstandings in image logs interpretation and expand the application field of image logs.

Neutralization slag (NS) of copper smelting is a waste residue mainly composed of calcium sulfate, which is produced during the process of metal smelting. The large accumulation and storage of NS poses a significant threat to the environment. Therefore, it is imperative to explore a new economically and environmentally friendly approach for recycling NS. This study proposes a method for preparing a new eco-friendly geopolymer material by utilizing NS. A geopolymer with satisfactory mechanical properties was created by employing a hybrid solid activator consisting of CaO-Na2SiO3. The effects of hybrid solid alkali activators calcium oxide (CaO)/sodium silicate (Na2SiO3) ratio on the hydration properties of the NS-based geopolymer were studied, and its curing mechanism was discussed. The results show that, when the hybrid solid activator CaO/Na2SiO3 ratio was 3:1, the maximum 28 d compressive strength of the geopolymer reached 27.8 MPa. The solid activator at a suitable ratio can accelerate the hydration reaction, form higher amounts of C-A-S-H gel and ettringite, reduce the proportion of harmful macropores in the geopolymer, and improve the microstructure of the geopolymer. Toxicity characteristic leaching procedure (TCLP) test confirmed that the NS-based geopolymer had a solidification effect on heavy metals. The leaching concentration of elemental Pb from the neutralised slag feedstock was 7.241 mg/L, while the lowest concentration was 0.286 mg/L in the NS based geopolymer. As a green cementitious material, the NS-based geopolymer is expected to be applied in engineering practice.

Geophysical well logs are widely used in geological fields, however, there are considerable incompatibilities existing in solving geological issues using well log data. This review critically fills the gaps between geology and geophysical well logs, as assessed from peer reviewed papers and from the authors’ personal experiences, in the particular goal of solving geological issues using geophysical well logs. The origin and history of geophysical logging are summarized. Next follows a review of the state of knowledge for geophysical well logs in terms of type of specifications, vertical resolution, depth of investigations and demonstrated applications. Then the current status and advances in applications of geophysical well logs in fields of structural geology, sedimentary geology and petroleum geology are discussed. Well logs are used in structural and sedimentary geology in terms of structure detection, in situ stress evaluation, sedimentary characterization, sequence stratigraphy division and fracture prediction. Well logs can also be applied in petroleum geology fields of optimizing sweet spots for hydraulic fracturing in unconventional oil and gas resource. Geophysical well logs are extending their application in other fields of geosciences, and geological issues will be efficiently solved via well logs with the improvements of advanced well log suits. Further work is required in order to improve accuracy and diminish uncertainties by introducing artificial intelligence. This review provides a systematic and clear descriptions of the applications of geophysical well log data along with examples of how the data is displayed and processed for solving geologic problems.

Abstract Background Frailty contributes to adverse outcomes in older adults and places a heavy burden on healthcare resources. Dysphagia is associated with frailty, but the mechanisms by which dysphagia affects frailty in older adults are unclear. This study aimed to investigate a serial mediating effect of self-perceived oral health and self-reported nutritional status in the relationship between dysphagia and frailty among hospitalized older patients in China. Methods This cross-sectional study included 1200 patients aged ≥ 65 years in the Department of Geriatrics, Shaanxi Provincial People’s Hospital. A structured face-to-face interview was used to survey the following questionnaires: General Information Questionnaire, Tilburg Frailty Indicators (TFI), Eating Assessment Tool-10 (EAT-10), 30mL Water Swallow Test (WST), Geriatric Oral Health Assessment Index (GOHAI), and Short-Form Mini-Nutritional Assessment (MNA-SF). A total of 980 participants with complete data were included in the analysis. Statistical analysis was performed using SPSS 26.0 and Amos 28.0 software. Spearman’s correlation analysis was used for correlation analysis of study variables. The results of the multivariate linear regression analysis for frailty were used as covariates in the mediation analysis, and the structural equation model (SEM) was used to analyze the mediating effects among the study variables. Shale oil reservoirs, characterized by no productivity, are developed by horizontal drilling and volume fracturing, and it is very important to optimize shale fracable intervals based on engineering quality logging evaluation. The Paleogene Funing Formation shale in the Subei Basin is taken as a typical example in this study. A sonic scanner is used to calculate the elastic parameters, including Poisson's ratio and Young's modulus. Dynamic and static parameters are converted through core analysis data. The brittleness index and in situ stress are calculated according to Poisson's ratio and Young's modulus. In addition, a one-dimensional rock mechanics model was constructed with the shear slowness in a single well to calculate three components of in situ stress. Finally, considering the difference in the brittleness index and horizontal stress differences between different layers, brittleness index (BI) and (BI/(σH-σh)) were selected to describe the engineering quality. According to oil test data, the larger the brittleness index and the smaller the horizontal stress are, the higher the capacity after fracturing. A cross plot of the brittleness index and BI/(σH-σh) is established to divide the reservoir types. Consequently, there are two types, including I high productivity(engineering quality characterization parameters >2.2) and Ⅱ medium-low productivity (engineering quality characterization parameters < 2.2), in the Paleogene Funing Formation in the Subei Basin. High productivity produces oil after fracturing, which suggests that the classification results of sweet spots depending on the engineering quality characterization parameters are better. Logging evaluation of the engineering quality of shale oil reservoirs can provide a theoretical basis and technical guidance for optimizing favorable fracability and high productivity layers and provide scientific guidance for drilling and fracturing layer optimization of shale reservoirs.

The method and theoretical system of well logging geology have been widely used in the fields of basic geology, petroleum geology and engineering geology, but the different response sensitivity of different well logging series to geological information and the mismatching between geophysical properties of multiple well logs and geological genesis of rocks frequently result in misunderstandings in the research process of well logging geology. Therefore, it is in an urgent need to analyze the typical misunderstanding cases in the research of well logging geology and explore the corresponding scientific ideas and countermeasures. After analyzing the typical misunderstandings in the research of well logging geology, this paper investigates vertical resolution scale of various logging series and its contradiction with detection depth and illustrates the importance of the integration of different scales of data. In addition, the factor inducing “fake logging data” and its influence on interpretation evaluation are clarified and a set of ideas for well logging evaluation of geological interpretation is put forward. And the following research results are obtained. First, the typical misunderstandings in the research of well logging geology can be classified into two categories, namely geological body interpretation misunderstanding and reservoir property parameter calculation misunderstanding. Second, special geological phenomena, such as high-density and high-resistivity mudstone can lead to logging data ambiguity, so attention shall be paid to petrophysical response mechanisms during geological logging interpretation. Third, to carry out well logging evaluation of unconventional oil and gas, it is necessary to integrate new technologies of electric imaging logging, dipole acoustic logging and nuclear magnetic resonance logging, and the calibration of core data and the integration of geological ideas can improve the interpretation accuracy. Fourth, In the process of borehole structural logging analysis, sedimentary response, geostress evaluation and fracture identification, geological ideas shall be integrated to avoid the logging interpretation misunderstanding caused by the same response of different geological phenomena in well logs. In conclusion, the dialectical and systematic thinking from geology to logging and then to geology, from practice to recognition and then to practice and from “a narrow view” to “a broad view” can provide a scientific ideas for the comprehensive research of well logging geology.

Knowledge of how high can fracture porosity become in the ultra-deep burial conditions is important but remains problematic. Fracture aperture and porosity are measured using X-ray computed tomography (CT) at atmospheric pressure and then calculated by image logs. Special attention is paid to how high fracture porosity can become in ultra-deep (>6000 m) settings, and which situations will result in high fracture porosities. In situ stress magnitudes, which can be calculated using well logs, control fracture performances, and dissolution along fracture improve fracture porosity at ultra-deep burial depths. Low horizontal stress difference (Δσ 45 MPa), a high degree of dissolution along the fracture dramatically increases fracture porosity. Dissolution will result in the vuggy fracture planes and improve fracture porosity up to 2.0%. The results provide insights into the detection, characterization, and modeling of subsurface fractures.

Eosinophilia may result from three main causes: secondary (reactive), primary (clonal), and/or idiopathic. The diagnosis of idiopathic eosinophilia must be made based on excluding all reactive or clonal causes. However, some causes may be very rare so as to be misdiagnosed as idiopathic. We present the case of eosinophilia caused by aggressive systemic mastocytosis, originally recognized as idiopathic. Lymphadenopathy, dysmyelopoiesis, and hepatosplenomegaly gradually appeared and deteriorated with increasing eosinophils. This case carried KIT D816V mutation. The BCR::ABL fusion gene and the mutations in JAK2 V617F, PDGFRα , and PDGFRβ in bone marrow were all negative. PHF6, PPM1D , and TET2 mutations were demonstrable. The patient was prescribed to avapritinib. The condition was effectively controlled. However, the patient discontinued medication for economic reasons 5 months later. Disease progression happened and died 10 months after diagnosis. Our study indicates that gene mutation detection at diagnosis is helpful for patient accurate diagnosis and targeted therapy of such patients.

Bashijiqike Formation in Bozi-Dabei area develops deep and ultra deep tight sandstone reservoirs. It is urgent to reveal the main controlling factors of reservoir quality and establish a matching well logging evaluation method for high quality reservoirs. In order to quantitatively evaluate the diagenetic facies, based on the data of core, thin section, cast thin section and scanning electron microscope and etc., the petrology, physical properties, diagenesis types and intensity, diagenetic minerals and other characteristics of the tight reservoir of Bashijiqike Formation in Bozi-Dabei area were studied. Based on quantitative calculation of ompaction rate, cementation rate, dissolution pore content and comprehensive diagenetic coefficient, the diagenetic facies were classified according to characteristics of diagenesis intensity and its combination. The reservoirs can be divided into three types of diagenetic facies, i.e., medium compaction weak dissolution facies, carbonate cemented facies and compaction dense facies. Through the fitting analysis of thin section data and well logging data, the well logging calculation model of diagenetic comprehensive coefficient Cg was established, and the method of quantitative identification of reservoir diagenetic facies were established. The well logging data of Well X1, Well Y1 and etc were processed.The reliability of the model was verified by matching the identification results with physical property analysis and casting thin section. The establishment of well logging quantitative characterization method of diagenetic facies can provide guidance for "sweet spot" prediction.

Context Coronavirus disease 2019 is a global pandemic. Studies suggest that folic acid has antiviral effects. Molecular docking shown that folic acid can act on SARS-CoV-2 Nucleocapsid Phosphoprotein (SARS-CoV-2 N).Objective To identify novel molecular therapeutic targets for SARS-CoV-2.Materials and methods Traditional Chinese medicine targets and virus-related genes were identified with network pharmacology and big data analysis. Folic acid was singled out by molecular docking, and its potential target SARS-CoV-2 N was identified. Inhibition of SARS-CoV-2 N of folic acid was verified at the cellular level.Results In total, 8355 drug targets were potentially involved in the inhibition of SARS-CoV-2. 113 hub genes were screened by further association analysis between targets and virus-related genes. The hub genes related compounds were analysed and folic acid was screened as a potential new drug. Moreover, molecular docking showed folic acid could target on SARS-CoV-2 N which inhibits host RNA interference (RNAi). Objectives: Previous studies demonstrated that endometriosis and adenomyosis are closely linked to lots of adverse pregnancy outcomes while the role of endometriosis in pregnant women with adenomyosis has not been explored yet. The present study aimed to evaluate the influence of previous laparoscopic surgical and pathological diagnosis of endometriosis on pregnancy outcomes in women with adenomyosis. Methods: A total of 60 pregnant women who were diagnosed with adenomyosis before or during pregnancy were included in this study. Among them, 8 were also diagnosed with endometriosis by previous laparoscopic surgery. The demographic characteristics and pregnancy outcomes were compared between women with adenomyosis only and those with the surgical history of endometriosis. Results: Compared with women with adenomyosis only, those concomitant with the surgical history of endometriosis had significantly higher age at delivery [37.5(36.25–39.75) vs. 35(33.25–37), P ​= ​0.016] and an increased risk of postpartum hemorrhage (PPH) (adjusted OR: 5.992, 95% CI: 1.03–34.857, P ​= ​0.046) while no significant differences were found in other adverse pregnancy outcomes between these two groups. Then we further detected the risk factor of PPH in women with adenomyosis and found that the surgical history of endometriosis (OR: 6.995, 95% CI: 1.16–42.171, P ​= ​0.034) and assisted reproductive technology (ART) (OR: 5.062, 95% CI: 1.494–17.146, P ​= ​0.009) were the parameters closely associated with the occurrence of PPH. Conclusions: The history of previous laparoscopic surgical and pathological diagnosis of endometriosis in pregnant women with adenomyosis may increase the risk of PPH, which still needs to be verified by future studies with a large sample size. Besides, pregnancy through ART is also an increased risk factor for PPH in women with adenomyosis. Pregnant women with adenomyosis who conceived with the surgical history of endometriosis or by ART should be closely monitored for the reason of being at high risk of PPH.

To explore the correlation between earthquakes and the pre-earthquake ionospheric shallow frequency (ELF) electric field perturbations phenomenon, the paper investigated the pre-earthquake ionospheric perturbations phenomenon, and then the Spatio-temporal evolution characteristics of the electromagnetic field before and after the global Ms ≥6.0 strong earthquakes from 2019 to 2021 were statistically analyzed. In this paper, the power spectrum data of the ELF (19.5–250 Hz) band of ionospheric electric field observed by the China Seismo-Electromagnetic Satellite (CSES) electric field detector are preclinically processed by the C-value method. A stable background field observation model was constructed using the data from 75 to 45 days before the earthquake observed by the CSES in the range of 15° above the epicenter. Then, the amplitude of the spatial electric field disturbance over the epicenter relative to the background field is extracted. Finally, the superposition analysis method statistically analyzes the spatial and temporal evolution of the spatial electric field before and after the earthquake with different characteristics. The statistical results show that the anomalies first appear in the fourth period (15–19 days before the earthquake) and the third period (10–14 days before the earthquake) and then reach the most vital and most evident during the pro-earthquake period (4 days before the earthquake and the day of the earthquake); In terms of the intensity of the anomalies caused, the magnitude seven earthquakes are stronger than the magnitude 6.0–7.0 earthquakes, and marine earthquakes are stronger than land earthquakes; in terms of the ease of observing the anomalies, the magnitude 7.0 and above are more accessible to observe than the magnitude 6.0–7.0 earthquakes, and marine earthquakes are more accessible to observe than land earthquakes.

We investigated the therapeutic effects of superoxide dismutase (SOD) from thermophilic bacterium HB27 on chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) and its underlying mechanisms. A Sprague–Dawley rat model of CP/CPPS was prepared and then administered saline or Thermus thermophilic (Tt)-SOD intragastrically for 4 weeks. Prostate inflammation and fibrosis were analyzed by hematoxylin and eosin staining, and Masson staining. Alanine transaminase (ALT), aspartate transaminase (AST), serum creatinine (CR), and blood urea nitrogen (BUN) levels were assayed for all animals. Enzyme-linked immunosorbent assays (ELISA) were performed to analyze serum cytokine concentrations and tissue levels of malondialdehyde, nitric oxide, SOD, catalase, and glutathione peroxidase. Reactive oxygen species levels were detected using dichlorofluorescein diacetate. The messenger ribonucleic acid (mRNA) expression of tissue cytokines was analyzed by reverse transcription polymerase chain reaction (RT-PCR), and infiltrating inflammatory cells were examined using immunohistochemistry. Nuclear factor-κB (NF-κB) P65, P38, and inhibitor of nuclear factor-κBα (I-κBα) protein levels were determined using western blot. Tt-SOD significantly improved histopathological changes in CP/CPPS, reduced inflammatory cell infiltration and fibrosis, increased pain threshold, and reduced the prostate index. Tt-SOD treatment showed no significant effect on ALT, AST, CR, or BUN levels. Furthermore, Tt-SOD reduced inflammatory cytokine expression in prostate tissue and increased antioxidant capacity. This anti-inflammatory activity correlated with decreases in the abundance of cluster of differentiation 3 (CD3), cluster of differentiation 45 (CD45), and macrophage inflammatory protein 1α (MIP1α) cells. Tt-SOD alleviated inflammation and oxidative stress by reducing NF-κB P65 and P38 protein levels and increasing I-κBα protein levels. These findings support Tt-SOD as a potential drug for CP/CPPS.

The controlling factors of organic-rich shale accumulation is essential for the exploration and development of shale oil and gas resources. The sedimentary environment plays a vital role in the formation of organic-rich sediments in lacustrine facies. This article unravels the mineralogy, geochemistry, and paleoenvironmental evolution during the deposition of the Paleogene Shahejie Formation (Es3 L). It discusses the effects of paleoclimate, paleosalinity, paleoredox conditions, paleowater depth, and paleoproductivity on organic matter (OM) enrichment. Finally, the OM enrichment model was established. The results show that the mineralogical compositions are mainly composed of calcite (avg. 40.13%), quartz (avg. 21.64%) and clay minerals (avg. 24.07%), accompanied by dolomite (avg. 7.07%), feldspar (avg. 6.36%) and pyrite (avg. 2.95%). The Es3 L shale has a high abundance of OM, with total organic carbon (TOC) ranging from 1.07% to 5.12%. The organic matter type is mainly composed of type I-II1 kerogen, which is generally considered a good-quality source rock. The source of OM is a mixture of lower bacteria, algae, and plants. During the early sedimentary period, the paleoclimate was dry and cold, with high salinity, intense reducibility, and relatively low productivity. During the late sedimentary period, the climate became warmer and more humid. As a result, the salinity decreased to a level that was suitable for biological reproduction, and productivity increased gradually due to the input of terrigenous plants. Paleosalinity and paleoclimate determined the environment of the sedimentary period, in addition, paleoproductivity and paleoredox condition indicated the formation and preservation conditions of OM. The warm and humid climate, brackish water, suitable reduction conditions and high productivity are the favorable conditions for the generation and preservation of organic matter. The research results may have implications for the genetic mechanisms of organic matter accumulation. Angioimmunoblastic T-cell lymphoma (AITL) genomic abnormalities are highly disease-specific, and the ras homology family member A (RHOA) gene is one of the most recurrent mutated genes, especially for RHOA G17V mutation site. Here, we identified a rare RHOA A161E mutation in an AITL patient through gene sequencing platforms. The patient presented with persistent hypereosinophilia, asymptomatic or symptomatic mildly for over 3 years. At diagnosis, this patient manifested night sweats, weight loss, multiple lymphadenopathies, and enlargement of the liver and spleen. We performed a retrospective genetic mutation analysis by whole-exome sequencing (WES) and droplet digital PCR (ddPCR) on serial gastric, intestinal, and lymph node specimens. The genetic mutation testing result demonstrated that a rare RHOA A161E mutation was found, which was elevated significantly on diagnosis related to AITL pathogenesis. Our case confirms that genetic mutation testing is helpful for diagnostic classification in AITL and dynamic monitoring of gene mutations at multiple time points may facilitate early detection of disease diagnosis.

Abstract Background The motion capture has been used as the usual method for measuring movement parameters of human, and most of the measuring data are obtained by partial manual process based on commercial software. An automatic kinematics data process was developed by programming on MATLAB software in this paper. Methods The motion capture measurement of healthy volunteers was carried out and the MATLAB program was used for data process. Firstly, the coordinate data of markers and anatomical points on human lower limb measured by motion capture system were read and repaired through the usual and the patch program. Meantime, the local coordinate systems of human femur and tibia were established with anatomical points. Then flexion/extension, abduction/adduction and internal/external rotation of human knee tibiofemoral joint were obtained by special coordinate transformation program. Results Using the above methods, motion capture measurements and batch data processing were carried out on squatting and climbing stairs of 29 healthy volunteers. And the motion characteristics (flexion/extension, internal/external rotation and adduction/abduction) of the knee joint were obtained. For example, the maximum internal/external rotation in squatting and climbing stairs were respectively was 30.5 degrees and 14 degrees, etc. Meantime, the results of this paper also were respectively compared with the results processed by other research methods, and the results were basically consistent, thus the reliability of our research method was verified. After calibration processing, the compiled MATLAB program of this paper can directly be used for efficient batch processing and avoiding manual modeling one by one. Conclusion A novel Patch Program of this paper has been developed, which can make reasonable compensation for missing and noise signals to obtain more complete motion data. At the same time, a universal data processing program has also been developed for obtaining the relative movement of various components of the human body, and the program can be modified for detail special analysis. These motion capture technologies can be used to judge whether the human body functions are abnormal, provide a reference for rehabilitation treatment and design of rehabilitation equipment, and evaluate the effectiveness before and after surgery.

The Ni-11Mo-8Al-3Ta-2Cr-1Re spherical powders of

Deep-focus earthquakes, located in very high-pressure conditions 300 to 700 km below the Earth's surface within sinking slabs of relatively cold oceanic lithosphere, are mysterious phenomena. The largest recorded deep-focus earthquake (MW 7.9) in the Izu-Bonin slab struck on 30 May 2015 beneath the Ogasawara (Bonin) Islands, isolated from prior seismicity by over 100 km in depth, and followed by only a few small aftershocks. Globally, this is the deepest (680 km centroid depth) event with MW≥7.8 in the seismological record. Seismicity indicates along-strike contortion of the Izu-Bonin slab, with horizontal flattening near a depth of 550 km in the Izu region and rapid steepening to near-vertical toward the south above the location of the 2015 event. This event was exceptionally well-recorded by seismic stations around the world, allowing detailed constraints to be placed on the source process. Analyses of a large global data set of P, SH and pP seismic phases using short-period back-projection, subevent directivity, and broadband finite-fault inversion indicate that the mainshock ruptured a shallowly-dipping fault plane with patchy slip that spread over a distance of ~40 km with a multi-stage expansion rate (~5+ km/s down-dip initially, ~3 km/s up-dip later). During the 17 s total rupture duration the radiated energy was ~3.3×1016 J and the stress drop was ~38 MPa. The radiation efficiency is moderate (0.34), intermediate to that of the 1994 Bolivia and 2013 Sea of Okhotsk MW 8.3 deep earthquakes, indicating that source processes of very large deep earthquakes sample a wide range of behavior from dissipative, more viscous failure to very brittle failure. The isolated occurrence of the event, much deeper than the apparently thermally-bounded distribution of Bonin-slab seismicity above 600 km depth, suggests that localized stress concentration associated with the pronounced deformation of the Izu-Bonin slab and proximity to the 660-km phase transition likely played a dominant role in generating this major earthquake.

Abstract Background Lupeol exhibits novel physiological and pharmacological activities, such as anticancer and immunity-enhancing activities. However, cytotoxicity remains a challenge for triterpenoid overproduction in microbial cell factories. As lipophilic and relatively small molecular compounds, triterpenes are generally secreted into the extracellular space. The effect of increasing triterpene efflux on the synthesis capacity remains unknown. Results In this study, we developed a strategy to enhance triterpene efflux through manipulation of lipid components in Y. lipolytica by overexpressing the enzyme Δ9-fatty acid desaturase (OLE1) and disturbing phosphatidic acid phosphatase (PAH1) and diacylglycerol kinase (DGK1). By this strategy combined with two-phase fermentation, the highest lupeol production reported to date was achieved, where the titer in the organic phase reached 381.67 mg/L and the total production was 411.72 mg/L in shake flasks, exhibiting a 33.20-fold improvement over the initial strain. Lipid manipulation led to a twofold increase in the unsaturated fatty acid (UFA) content, up to 61–73%, and an exceptionally elongated cell morphology, which might have been caused by enhanced membrane phospholipid biosynthesis flux. Both phenotypes accelerated the export of toxic products to the extracellular space and ultimately stimulated the capacity for triterpenoid synthesis, which was proven by the 5.11-fold higher ratio of extra/intracellular lupeol concentrations, 2.79-fold higher biomass accumulation and 2.56-fold higher lupeol productivity per unit OD in the modified strains. This strategy was also highly efficient for the biosynthesis of other triterpenes and sesquiterpenes, including α-amyrin, β-amyrin, longifolene, longipinene and longicyclene. Conclusions In conclusion, we successfully created a high-yield lupeol-producing strain via lipid manipulation. We demonstrated that the enhancement of lupeol efflux and synthesis capacity was induced by the increased UFA content and elongated cell morphology. Our study provides a novel strategy to promote the biosynthesis of valuable but toxic products in microbial cell factories.

Abstract Fracture detection and in‐situ stress determination via well logs are important for exploration and exploitation of subsurface hydrocarbons. Cores, thin sections, and image logs are used to describe and interpret the subsurface fractures and in‐situ stress in the Paleogene Dongying sandstones in Nanpu Sag, Bohai Bay Basin, China. The maximum horizontal stress (SHmax) indicates a nearly east‐west trend according to the borehole breakouts and drilling‐induced fractures. Natural fractures in the Dongying sandstones are classified into: (1) open fractures and (2) closed fractures. The cement‐filled (closed) fractures are commonly detected by image logs as bright discontinuous sinusoidal waves due to resistive filling materials, while the open fractures are evident on image logs appearing as dark sine waves. The open fractures are of dominantly high dip angles, and the rose diagrams confirm the presences of two sets of fractures: northwest‐southeast and northwest‐southeast orientation. Fractures with strikes approximately parallel to the SHmax have good connectedness, making a significant contribution in hydrocarbon production. In addition, the oil‐bearing layers are mainly associated with the intervals with open fractures. The presence of natural fractures provides important pore spaces and fluid flow conduits. Insights can be provided into the subsurface fracture (natural and induced) detection and characterization using image logs.

Background and Aims: It’s reported that bone morphogenetic protein 9 (BMP9) played an important role in lipid and glucose metabolism, but the role of BMP9 in nonalcoholic fatty liver disease (NAFLD) is unclear. Here, we evaluated the therapeutic efficacy of recombined BMP9 in NAFLD mice and investigated the potential mechanism.Methods: The effects of recombinant BMP9 on NAFLD were assessed in HFD-induced NAFLD mice. C57BL/6 mice were administrated with high-fat diet (HFD) for 12 weeks. In the last 4 weeks, mice were treated with PBS or recombined BMP9 once daily. Insulin sensitivity was evaluated by glucose tolerance test (GTT) and insulin tolerance test (ITT) at the end of the 12th week. Then NAFLD related indicators were assessed by a variety of biological methods, including histology, western blotting, real-time PCR, RNA-seq and assay for transposase-accessible chromatin using sequencing (ATAC-seq) analyses.Results: BMP9 reduced obesity, improved glucose metabolism, alleviated hepatic steatosis and decreased liver macrophages infiltration in HFD mice. RNA-seq showed that Cers6, Cidea, Fabp4 involved in lipid and glucose metabolism and Fos, Ccl2, Tlr1 involved in inflammatory response downregulated significantly after BMP9 treatment in HFD mouse liver. ATAC-seq showed that chromatin accessibility on promoters of Cers6, Fabp4, Ccl2 and Fos decreased after BMP9 treatment in HFD mouse liver. KEGG pathway analysis of dysregulated genes in RNA-seq and integration of RNA-seq and ATAC-seq showed that TNF signaling pathway and Toll-like receptor signaling pathway decreased in BMP9 treated HFD mouse liver.Conclusion: Our data revealed that BMP9 might alleviate NAFLD via improving glucose and lipid metabolism, decreasing inflammatory response and reshaping chromatin accessibility in HFD mouse liver. BMP9 downregulate genes related to lipid metabolism, glucose metabolism and inflammation expression, at least partially via decreasing promoter chromatin accessibility of Cers6, Fabp4, Fos and Tlr1. BMP9 may also reduce the expression of liver Ccl2, thereby changing the number or composition of liver macrophages, and ultimately reducing liver inflammation. The effect of BMP9 on NAFLD might be all-round, and not limit to lipid and glucose metabolism. Therefore, the underlying mechanism needs to be studied in detail further.

Abstract Background Lateral compression type1 (LC-1) pelvic fractures represent a wide spectrum of heterogeneous injuries. These include both stable and unstable patterns; however, determining whether a LC-1 fracture is stable or unstable is a challenge, and the method used to evaluate fracture stability is complicated. Methods We prospectively collected and analyzed data from 22 patients with LC-1 pelvic fractures, who underwent ultrasonography and a pelvic compression and separation test, in order to evaluate the role of ultrasonography in determining fracture stability and assist decision-making for treatment strategy. Results Twenty-two patients (15 men and 7 women) were included in the study. Following an ultrasound examination, 10 patients were classified into the stable group and 12 into the unstable group. In total, 13 patients received conservative treatment and 9 underwent surgery. At follow-up, there were no differences in fracture healing times or fracture-related complications between the two groups. The Majeed score was comparable between the two groups and most patients recovered well. There was a moderate degree of consistency in Kappa values (Kappa = 0.571, P = 0.01) between the classification of stability and the final treatment received. In addition, the sensitivity of ultrasonography was 66.67% and the specificity was 76.92%. Conclusions In conclusion, ultrasonography is a useful tool for diagnosing the stability of LC-1 pelvic fractures and assists the determination of treatment strategy. Left-right mobility ≥ 0.3 cm may be used as the criterion for determining instability. Trial registration ChiCTR-DDD-16008722.
