Background Alzheimer’s disease (AD) early diagnosis remains difficult due to limitations in clinical exams and amyloid plaque imaging. Methods In the present study, the hippocampus, cortex, and blood plasma extracellular vesicles (EVs) from 3- and 6-month-old 5xFAD mice were analysed by reliable quantitative proteomics approach. Results The 3- and 6-month-old hippocampus and cortex proteome in both the age groups showed similar features in functional annotation and canonical pathway analysis, but the significantly changed proteins were rarely overlapped. Furthermore, the plasma EVs proteome showed significantly different informatic features compared with other proteomes. Depending on the AD stage, proteomic profiles undergo drastic changes in brain subregion- and in tissue-specific manners. Notably, regulations of several canonical pathways, including PI3K/Akt signalling, were differing between the hippocampus and cortex. Furthermore, we identified eight potential biomarkers that can detect early-stage AD (integrin alpha-IIb (ITGA2B), sulfhydryl oxidase 1, phospholipid transfer protein, talin (TLN), heat shock 70 kDa protein 1 (HSPA1L), alpha-2 macroglobulin (A2M), platelet factor 4, and filamin A (FLNA)) and validated them, using plasma EVs of stage-divided patients with AD. Conclusion ITGA2B, TLN, HSPA1L, and A2M were finally selected by machine learning modelling as distinguishing biomarkers for normal and early-stage AD with 85% accuracy. The present study provides insights into AD pathogenesis and identifies novel early-stage AD biomarkers.

A 32-year-old woman with schizophrenia and persistent auditory verbal hallucinations (AVHs), which caused continuous suicidal thoughts and depression, was treated with electroconvulsive therapy (ECT) of an acute course followed by maintenance ECT (M-ECT) augmented onto clozapine for 7 years. Although the general psychopathology and AVHs initially reduced slightly with ECT and clozapine, her AVHs and suicidal thoughts did not decrease subjectively. When 3 years of M-ECT, her voices declined sharply, and improvement was maintained for 2 years thereafter. A total 91 ECT sessions were performed. The daily clozapine dose was decreased from 325 to 200 mg and plasma levels remained higher than 350 ng/ml; there were no noticeable cognitive side effects. In summary, we report a case showing a sudden sharp reduction in persistent AVHs after 3 years of long-term M-ECT.

Objective The present study aimed to report the initial seizure threshold (IST) of a brief-pulse bilateral electroconvulsive therapy (BP-BL ECT) in Korean patients with schizophrenia/schizoaffective disorder and to identify IST predictors. Methods Among 67 patients who received ECT and diagnosed with schizophrenia/schizoaffective disorder based on the criteria of the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition, Text Revision, we included 56 patients who received 1-millisecond BP-BL ECT after anesthesia with sodium thiopental between March 2012 and June 2018. Demographic and clinical information was gathered from electronic medical records, and a multiple regression analysis was conducted to identify predictors of the IST. Results The mean age of the patients was 36.9±12.0 years and 30 (53.6%) patients were male. The mean and median IST were 105.9±54.5 and 96 millicoulombs (mC), respectively. The IST was predicted by age, gender, and dose (mg/kg) of sodium thiopental. Other physical and clinical variables were not associated with the IST. Conclusion The present study demonstrated that the IST of 1-ms BP-BL ECT following sodium thiopental anesthesia in Korean patients was comparable to those reported in previous literature. The IST was associated with age, gender, and dose of sodium thiopental.

Most antidepressants, including selective serotonin reuptake inhibitors (SSRIs), initiate their drug actions by rapid elevation of serotonin, but they take several weeks to achieve therapeutic onset. This therapeutic delay suggests slow adaptive changes in multiple neuronal subtypes and their neural circuits over prolonged periods of drug treatment. Mossy cells are excitatory neurons in the dentate hilus that regulate dentate gyrus activity and function. Here we show that neuronal activity of hippocampal mossy cells is enhanced by chronic, but not acute, SSRI administration. Behavioral and neurogenic effects of chronic treatment with the SSRI, fluoxetine, are abolished by mossy cell-specific knockout of p11 or Smarca3 or by an inhibition of the p11/AnxA2/SMARCA3 heterohexamer, an SSRI-inducible protein complex. Furthermore, simple chemogenetic activation of mossy cells using Gq-DREADD is sufficient to elevate the proliferation and survival of the neural stem cells. Conversely, acute chemogenetic inhibition of mossy cells using Gi-DREADD impairs behavioral and neurogenic responses to chronic administration of SSRI. The present data establish that mossy cells play a crucial role in mediating the effects of chronic antidepressant medication. Our results indicate that compounds that target mossy cell activity would be attractive candidates for the development of new antidepressant medications.

Parkinson's disease (PD) and Alzheimer's disease exhibit common features of neurodegenerative diseases and can be caused by numerous factors. A common feature of these diseases is neurotoxic inflammation by activated microglia, indicating that regulation of microglial activation is a potential mechanism for preserving neurons in the adult brain. Recently, we reported that upregulation of prothrombin kringle-2 (pKr-2), one of the domains that make up prothrombin and which is cleaved and generated by active thrombin, induces nigral dopaminergic (DA) neuronal death through neurotoxic microglial activation in the adult brain. In this study, we show that silibinin, a flavonoid found in milk thistle, can suppress the production of inducible nitric oxide synthase and neurotoxic inflammatory cytokines, such as interleukin-1β and tumor necrosis factor-α, after pKr-2 treatment by downregulating the extracellular signal-regulated kinase signaling pathway in the mouse substantia nigra. Moreover, as demonstrated by immunohistochemical staining, measurements of the dopamine and metabolite levels, and open-field behavioral tests, silibinin treatment protected the nigrostriatal DA system resulting from the occurrence of pKr-2-triggered neurotoxic inflammation in vivo. Thus, we conclude that silibinin may be beneficial as a natural compound with anti-inflammatory effects against pKr-2-triggered neurotoxicity to protect the nigrostriatal DA pathway and its properties, and thus, may be applicable for PD therapy.

Recently, redox flow batteries (RFBs) have emerged as one of the promising candidates for safe and economical grid-scale energy storage system (ESS) that store the intermittent energies such as wind or solar energy. Unlike the conventional aqueous vanadium RFBs, the design of new molecules in nonaqueous RFB (NRFB) has been in the spotlight for the high energy density, cycling stability, and low cost. Among the redox active materials, in particular, organic molecule has various structures through functionalization, and thus its redox potential, solubility, and chemical stability can be controlled. However, there are only a few viable negative-side redox electrolyte, called a negolyte (or anolyte) to date, and even the reported negolytes have suffered from low solubility and stability, and severe crossover problems for long-term cycling. Here we show the systematic design strategies for pyridinium-based organic redox molecule to enhance the its stability and solubility and suppress the crossover rate for NRFB. A benzo[d]thiazole ring, which provides an electron-withdrawing effect, was introduced at the C4 position of pyridinium core by Pd catalyzed C-H arylation. The addition of the π-conjugation system to the pyridinium redox core was key to enhance chemical and electrochemical stability, resulting in negatively low redox potential of -1.19 ~ 1.21 V vs. Fc/Fc+. The solubility of pyridinium derivatives was significantly enhanced from 0.26 M to 1.00 M in acetonitrile by simple anion exchange from tetrafluoroborate (BF4 -) or hexafluorophosphate (PF6 -) anion to bis(trifluoromethanesulfonyl)imide (TFSI-) anion. Moreover, exchanging the functional group on N of pyridinium from methyl group to cationic 3-(trimethylammonio)propyl (TMAP) group suppressed the crossover rate with an anion-exchange membrane in the NRFB. 4-(benzo[d]thaizol-2-yl)-1-(3-(trimethylammonio)propyl)pyridin-1-ium (TMAP-BTP) led to electrochemical stability in symmetric cell, showing a capacity decay rate of 0.0083% per cycle. Contrary to the results of only 60% capacity retention after 100 cycle in full cell with 4-(benzo[d]thaizol-2-yl)-1-methylpyridin-1-ium (BTP) as negolyte, in the case of full cell with TMAP-BTP as negolyte, the capacity retention was significantly increased, showing 89.8 % after 100 cycle, which is ~0.08% capacity decay rate per cycle. Glutamatergic mossy cells (MCs) are responsible for the associational and commissural connectivity of the dentate gyrus. MCs are widely distributed along the dorsoventral axis, but potential heterogeneity within MCs is scarcely explored. Here, we showed that MCs consist of two subpopulations which differ in their neuronal properties and functions. We discovered that MCs, depending on their dorsoventral location, extend distinct axonal projections in the molecular layers. Comparative transcriptional profiling of dorsal and ventral MCs revealed different neurobiological characteristics in axon guidance and synapse assembly. Despite common activation by external stimuli, dorsal MCs, but not ventral MCs, provide net inhibitory control on granule cells across the longitudinal axis. Furthermore, dorsal MC inhibition, unlikely that of ventral MCs, increases behavioral anxiety and disables rapid contextual discrimination. Collectively, dorsoventral heterogeneity of MCs may provide a novel mechanism for functional differentiation as well as distinct association along the longitudinal extent of the hippocampus.

Pd-catalyzed C–H annulation reactions of halo- and aryl-heteroarenes were developed using readily available o-bromobiaryls and o-dibromoaryls, respectively. A variety of five-membered heteroarenes rapidly provided the corresponding phenanthrene-fused heteroarenes, which led to the identification of phenanthro-pyrazole and thiazole as new, stable −2 V redox couples. The flexible syntheses and tunability of the redox potentials of these azole-fused phenanthrenes over a wide range are expected to facilitate their application as redox-active organic functional materials.

Depression is a leading cause of disability. Current pharmacological treatment of depression is insufficient, and development of improved treatments especially for treatment-resistant depression is desired. Understanding the neurobiology of antidepressant actions may lead to development of improved therapeutic approaches. Here, we demonstrate that dopamine D1 receptors in the dentate gyrus act as a pivotal mediator of antidepressant actions in mice. Chronic administration of a selective serotonin reuptake inhibitor (SSRI), fluoxetine, increases D1 receptor expression in mature granule cells in the dentate gyrus. The increased D1 receptor signaling, in turn, contributes to the actions of chronic fluoxetine treatment, such as suppression of acute stress-evoked serotonin release, stimulation of adult neurogenesis and behavioral improvement. Importantly, under severely stressed conditions, chronic administration of a D1 receptor agonist in conjunction with fluoxetine restores the efficacy of fluoxetine actions on D1 receptor expression and behavioral responses. Thus, our results suggest that stimulation of D1 receptors in the dentate gyrus is a potential adjunctive approach to improve therapeutic efficacy of SSRI antidepressants.

Lysophosphatidic acid (LPA) has been implicated in the pathology of human ovarian cancer. This phospholipid elicits a wide range of cancer cell responses, such as proliferation, trans-differentiation, migration, and invasion, via various G-protein-coupled LPA receptors (LPARs). Here, we explored the cellular signaling pathway via which LPA induces migration of ovarian cancer cells. LPA induced robust phosphorylation of ezrin/radixin/moesin (ERM) proteins, which are membrane-cytoskeleton linkers, in the ovarian cancer cell line OVCAR-3. Among the LPAR subtypes expressed in these cells, LPA1 and LPA2, but not LPA3, induced phosphorylation of ERM proteins at their C-termini. This phosphorylation was dependent on the Gα12/13/RhoA pathway, but not on the Gαq/Ca2+/PKC or Gαs/adenylate cyclase/PKA pathway. The activated ERM proteins mediated cytoskeletal reorganization and formation of membrane protrusions in OVCAR-3 cells. Importantly, LPA-induced migration of OVCAR-3 cells was completely abolished not only by gene silencing of LPA1 or LPA2, but also by overexpression of a dominant negative ezrin mutant (ezrin-T567A). Taken together, this study demonstrates that the LPA1/LPA2/ERM pathway mediates LPA-induced migration of ovarian cancer cells. These findings may provide a potential therapeutic target to prevent metastatic progression of ovarian cancer.

The synthesis of indazoles from pyrazoles and internal alkynes is described. Instead of complex benzenoid compounds, readily available pyrazoles were used for the preparation of indazoles by reaction of the C-H bonds of the heterocyclic ring. Oxidative benzannulation was also applied to imidazoles, providing benzimidazoles. This convergent strategy enabled alteration of the photochemical properties of benzo-fused diazoles by varying the substituents at the benzene ring, thus leading to the development of tetraarylindazoles as new fluorophores.

A simple method was reported to fabricate self-cleaning and water–oil separation fabrics sprayed with hot-iron-treated graphene oxide (GO). The GO solution was prepared with a modified Hummers' method and coated on the fabrics by spraying or soaking method. A 160 °C hot iron was pressed at the surface of the fabrics to make it flat, dry, thermally reduced in part, and strongly bonded. Afterward, the fabrics were thermally reduced at 250 °C for 20 minutes in an oven. The reduced graphene oxide (rGO) coated fabrics exhibited a superhydrophobic nature with a water contact angle of 129.4°, through which water could barely permeate the fabrics, in contrast to oil and organic solvents of low polarity. Additionally, this rGO fabric presented outstanding mechanical properties as well as a reusable stability.

Elevated levels of the free fatty acid palmitate are found in the plasma of obese patients and induce insulin resistance. Skeletal muscle secretes myokines as extracellular signaling mediators in response to pathophysiological conditions. Here, we identified and characterized the skeletal muscle secretome in response to palmitate-induced insulin resistance. Using a quantitative proteomic approach, we identified 36 secretory proteins modulated by palmitate-induced insulin resistance. Bioinformatics analysis revealed that palmitate-induced insulin resistance induced cellular stress and modulated secretory events. We found that the decrease in the level of annexin A1, a secretory protein, depended on palmitate, and that annexin A1 and its receptor, formyl peptide receptor 2 agonist, played a protective role in the palmitate-induced insulin resistance of L6 myotubes through PKC-θ modulation. In mice fed with a high-fat diet, treatment with the formyl peptide receptor 2 agonist improved systemic insulin sensitivity. Thus, we identified myokine candidates modulated by palmitate-induced insulin resistance and found that the annexin A1- formyl peptide receptor 2 pathway mediated the insulin resistance of skeletal muscle, as well as systemic insulin sensitivity.

Ras homolog enriched in brain (Rheb) regulates diverse cellular functions by modulating its nucleotide-bound status. Although Rheb contains a high basal GTP level, the regulatory mechanism of Rheb is not well understood. In this study, we propose soluble αβ-tubulin acts as a constitutively active Rheb activator, which may explain the reason why Rheb has a high basal GTP levels. We found that soluble αβ-tubulin is a direct Rheb-binding protein and that its deacetylated form has a high binding affinity for Rheb. Modulation of both soluble and acetylated αβ-tubulin levels affects the level of GTP-bound Rheb. This occurs in the mitotic phase in which the level of acetylated αβ-tubulin is increased but that of GTP-bound Rheb is decreased. Constitutively active Rheb-overexpressing cells showed an abnormal mitotic progression, suggesting the deacetylated αβ-tubulin-mediated regulation of Rheb status may be important for proper mitotic progression. Taken together, we propose that deacetylated soluble αβ-tubulin is a novel type of positive regulator of Rheb and may play a role as a temporal regulator for Rheb during the cell cycle.

We report ambivalent rejection behavior of a graphene oxide membrane (GOM) having a reduced interlayer spacing. Ultrathin GOMs having a thickness of 50 nm were fabricated using a vacuum filtration method followed by subjecting the samples to thermal reduction at 162 °C. The interlayer spacing of GOMs was reduced by 1 A on thermal reduction as compared with that of the natural GOMs. The rejection rate with dye molecules was tested using dyes having three different types of charges in a dead-end filtration instrument. Rejection rate of the reduced GOM with the dyes having an opposite charge was improved up to 99.7%, indicating the dominant effect of the physical sieving diameter. In contrast, in the case of ion permeation of natural GOM, a higher rejection rate for several metal ions was observed as compared with that of GOMs having 1 A smaller interlayer spacing, indicating the dominant effect of surface charges on the GOM samples.

NHERF1/EBP50 (Na+/H+ exchanger regulating factor 1; Ezrin-binding phosphoprotein of 50 kDa) organizes stable protein complexes beneath the apical membrane of polar epithelial cells. By contrast, in cancer cells without any fixed polarity, NHERF1 often localizes in the cytoplasm. The regulation of cytoplasmic NHERF1 and its role in cancer progression remain unclear. In this study, we found that, upon lysophosphatidic acid (LPA) stimulation, cytoplasmic NHERF1 rapidly translocated to the plasma membrane, and subsequently to cortical protrusion structures, of ovarian cancer cells. This movement depended on direct binding of NHERF1 to C-terminally phosphorylated ERM proteins (cpERMs). Moreover, NHERF1 depletion downregulated cpERMs and further impaired cpERM-dependent remodeling of the cell cortex, suggesting reciprocal regulation between these proteins. The LPA-induced protein complex was highly enriched in migratory pseudopodia, whose formation was impaired by overexpression of NHERF1 truncation mutants. Consistent with this, NHERF1 depletion in various types of cancer cells abolished chemotactic cell migration toward a LPA gradient. Taken together, our findings suggest that the high dynamics of cytosolic NHERF1 provide cancer cells with a means of controlling chemotactic migration. This capacity is likely to be essential for ovarian cancer progression in tumor microenvironments containing LPA.

Phospholipases (PLC, PLD and PLA) are essential mediators of intracellular and intercellular signalling. They can function as phospholipid-hydrolysing enzymes that can generate many bioactive lipid mediators, such as diacylglycerol, phosphatidic acid, lysophosphatidic acid and arachidonic acid. Lipid mediators generated by phospholipases regulate multiple cellular processes that can promote tumorigenesis, including proliferation, migration, invasion and angiogenesis. Although many individual phospholipases have been extensively studied, how phospholipases regulate diverse cancer-associated cellular processes and the interplay between different phospholipases have yet to be fully elucidated. A thorough understanding of the cancer-associated signalling networks of phospholipases is necessary to determine whether these enzymes can be targeted therapeutically.

There is a strong possibility that skeletal muscle can respond to irregular metabolic states by secreting specific cytokines. Obesity-related chronic inflammation, mediated by pro-inflammatory cytokines, is believed to be one of the causes of insulin resistance that results in type 2 diabetes. Here, we attempted to identify and characterize the members of the skeletal muscle secretome in response to tumor necrosis factor-alpha (TNF-α)-induced insulin resistance. To conduct this study, we comparatively analyzed the media levels of proteins released from L6 skeletal muscle cells. We found 28 TNF-α modulated secretory proteins by using separate filtering methods: Gene Ontology, SignalP, and SecretomeP, as well as the normalized Spectral Index for label-free quantification. Ten of these secretory proteins were increased and 18 secretory proteins were decreased by TNF-α treatment. Using microarray analysis of Zuker diabetic rat skeletal muscle combined with bioinformatics and Q-PCR, we found a correlation between TNF-α-mediated insulin resistance and type 2 diabetes. This novel approach combining analysis of the conditioned secretome and transcriptome has identified several previously unknown, TNF-α-dependent secretory proteins, which establish a foothold for research on the different causes of insulin resistance and their relationships with each other.

Epidermal growth factor receptor (EGFR) is a representative model of receptor tyrosine kinases (RTKs), and offers a means of understanding their common principles and fundamental mechanisms. Furthermore, EGFR plays an essential role in cell proliferation and migration, and the disruption of EGFR signaling has been implicated in the development and growth of cancer. Phospholipase D (PLD) is a key mediator of EGFR function, and can be directly regulated by upstream binding partners in an EGF-dependent manner. PLD regulates downstream molecules by generating phosphatidic acid (PA), but it also dynamically interacts with a variety of intracellular molecules and these interactions spatiotemporally regulate EGFR function and serve as a hub that orchestrates signaling flow. This review summarizes the interrelationship between PLD and its binding molecules in the context of EGFR signaling, and addresses the roles of PLD in the mediation and coordination of this signaling.

Recently, the lack of clean and safe drinking water is huge challenge because of rapid population growth, severe droughts and environmental pollution. Two-dimensional Graphene oxide (GO) membrane with their ultrafast permanence, outstanding mechanical properties, high chemical stability is increasingly emerging as promising candidate for water purification process. But GO-based water purification technology still needs further investigation and optimization. Here, we fabricated ultrathin graphene oxide membrane by the vacuum filtration method, and successfully applied for the water purification. GO membranes with several tenth nm thickness prepared show well stacked layer structure, as characterized by atomic force microscopy and scanning electron microscopy. We have tried pressure-driven filtration to examine water flux, retention and rejection ability of the GO membranes in water purification process. The ultrathin graphene oxide membrane showed high retention and rejection rates for the charged dyes and moderate results for noncharged organic dye.

Objective— Aberrant regulation of the proliferation, survival, and migration of endothelial cells (ECs) is closely related to the abnormal angiogenesis that occurs in hypoxia-induced pathological situations, such as cancer and vascular retinopathy. Hypoxic conditions and the subsequent upregulation of hypoxia-inducible factor-1α and target genes are important for the angiogenic functions of ECs. Phospholipase D2 (PLD2) is a crucial signaling mediator that stimulates the production of the second messenger phosphatidic acid. PLD2 is involved in various cellular functions; however, its specific roles in ECs under hypoxia and in vivo angiogenesis remain unclear. In the present study, we investigated the potential roles of PLD2 in ECs under hypoxia and in hypoxia-induced pathological angiogenesis in vivo. Approach and Results— Pld2 knockout ECs exhibited decreased hypoxia-induced cellular responses in survival, migration, and thus vessel sprouting. Analysis of hypoxia-induced gene expression revealed that PLD2 deficiency disrupted the upregulation of hypoxia-inducible factor-1α target genes, including VEGF , PFKFB3 , HMOX-1 , and NTRK2 . Consistent with this, PLD2 contributed to hypoxia-induced hypoxia-inducible factor-1α expression at the translational level. The roles of PLD2 in hypoxia-induced in vivo pathological angiogenesis were assessed using oxygen-induced retinopathy and tumor implantation models in endothelial-specific Pld2 knockout mice. Pld2 endothelial-specific knockout retinae showed decreased neovascular tuft formation, despite a larger avascular region. Tumor growth and tumor blood vessel formation were also reduced in Pld2 endothelial-specific knockout mice. Conclusions— Our findings demonstrate a novel role for endothelial PLD2 in the survival and migration of ECs under hypoxia via the expression of hypoxia-inducible factor-1α and in pathological retinal angiogenesis and tumor angiogenesis in vivo.

Phospholipase D (PLD) is a phospholipid-hydrolyzing enzyme that generates phosphatidic acid (PA) as a lipid second messenger by hydrolyzing phosphatidyl choline (PC). Various extracellular signals have been reported to activate PLD, which acts as a key mediator of many cellular functions through the generation of PA and the interactions of PLD and PA with their binding partners. Currently, about 60 PLD-binding partners, including proteins and phospholipids, are known, and PA has been found to interact with about 50 proteins. Although the interactions of binding molecules with PLD and PA are complex and multilayered, the unique interactions between them are important for their unique intracellular functions. Here, we address the interrelationships between PLD and PA and their binding partners in several key signaling pathways, such as the EGFR–ERK signaling axis, nutrient/growth signaling axis, and cytoskeletal reorganization machinery axis. These interrelationships demonstrate dynamic interactions and cooperative regulation, which mediate special intracellular functions. Furthermore, we describe the regulation and functions of PLD in mediating normal and pathological signaling. Additionally, we summarize the roles of PLD as determined in animal studies (Drosophila, zebrafish, and mice) and changes in the PLD expression level in disease states. These findings provide new insight into the functions of PLD under pathophysiological conditions.

mTOR complex 1 (mTORC1) is a multiprotein complex that integrates diverse signals including growth factors, nutrients, and stress to control cell growth. Raptor is an essential component of mTORC1 that functions to recruit specific substrates. Recently, Raptor was suggested to be a key target of regulation of mTORC1. Here, we show that Raptor is phosphorylated by JNK upon osmotic stress. We identified that osmotic stress induces the phosphorylation of Raptor at Ser-696, Thr-706, and Ser-863 using liquid chromatography-tandem mass spectrometry. We found that JNK is responsible for the phosphorylation. The inhibition of JNK abolishes the phosphorylation of Raptor induced by osmotic stress in cells. Furthermore, JNK physically associates with Raptor and phosphorylates Raptor in vitro, implying that JNK is responsible for the phosphorylation of Raptor. Finally, we found that osmotic stress activates mTORC1 kinase activity in a JNK-dependent manner. Our findings suggest that the molecular link between JNK and Raptor is a potential mechanism by which stress regulates the mTORC1 signaling pathway.

Phospholipase D (PLD) is a phosphatidyl choline (PC)-hydrolyzing enzyme that generates phosphatidic acid (PA), a lipid second messenger that modulates diverse intracellular signaling. Through interactions with signaling molecules, both PLD and PA can mediate a variety of cellular functions, such as, growth/proliferation, vesicle trafficking, cytoskeleton modulation, development, and morphogenesis. Therefore, systemic approaches for investigating PLD networks including interrelationship between PLD and PA and theirs binding partners, such as proteins and lipids, can enhance fundamental knowledge of roles of PLD and PA in diverse biological processes. In this review, we summarize previously reported protein-protein and protein-lipid interactions of PLD and PA and their binding partners. In addition, we describe the functional roles played by PLD and PA in these interactions, and provide PLD network that summarizes these interactions. The PLD network suggests that PLD and PA could act as a decision maker and/or as a coordinator of signal dynamics. This viewpoint provides a turning point for understanding the roles of PLD-PA as a dynamic signaling hub.
