Abstract Randomness is an essential resource and plays important roles in various applications ranging from cryptography to simulation of complex systems. Certified randomness from quantum process is ensured to have the element of privacy but usually relies on the device’s behavior. To certify randomness without the characterization for device, it is crucial to realize the one-sided device-independent random number generation based on quantum steering, which guarantees security of randomness and relaxes the demands of one party’s device. Here, we distribute quantum steering between two distant users through a 2 km fiber channel and generate quantum random numbers at the remote station with untrustworthy device. We certify the steering-based randomness by reconstructing covariance matrix of the Gaussian entangled state shared between distant parties. Then, the quantum random numbers with a generation rate of 7.06 Mbits/s are extracted from the measured amplitude quadrature fluctuation of the state owned by the remote party. Our results demonstrate the first realization of steering-based random numbers extraction in a practical fiber channel, which paves the way to the quantum random numbers generation in asymmetric networks.

Abstract This study delves into the challenges posed by the low-temperature calcination of high-ferrite Portland cement (HFPC) clinker and explores the potential of boron oxide (B2O3) as a stabilizing agent. Clinker production is a major contributor to global carbon dioxide emissions, and finding sustainable solutions is paramount. At 1350 °C, the HFPC clinker exhibits severe pulverization due to the metastable nature of the C2S phase formed at low temperature. To address these challenges, various stabilizing agents, including K2O + Na2O, barium carbonate (BaCO3), calcium fluoride (CaF2), and B2O3, were investigated. K2O + Na2O, BaCO3, and B2O3 exhibited promising stabilization effects, although K2O + Na2O negatively impacted the stability, and BaCO3 resulted in significant retardation. Consequently, B2O3 was chosen as the preferred stabilizing agent for the low-temperature calcination of HFPC clinker. However, it was observed that the B2O3 content should not exceed 1% to prevent destabilization of the C3S phase, which affects early-stage strength development. This research contributes to the understanding of HFPC clinker stability under low-temperature conditions and provides a potential avenue for reducing energy consumption and carbon emissions in HFPC clinker production.

Cutaneous pseudolymphoma is a type of reactive lymphoid hyperplasia that pathologically and/or clinically mimics cutaneous lymphoma. It has a variety of pathogenic factors. However, in many cases, no etiology can be identified. The histopathological findings of cutaneous pseudolymphoma can only be used as the basis for suspected diagnosis. Clinical data, further diagnostic tests and follow-up are needed to confirm the diagnosis. This article reports three cases of cutaneous pseudolymphoma, with different etiology, clinical manifestations and treatment methods. The purpose of this article is to emphasize that the etiology and clinical manifestations of cutaneous pseudolymphoma are diverse, and targeted treatment strategies and long-term follow-up are needed.

BackgroundIn contrast to previous network meta-analysis using classical frequentist methods, we evaluated the efficacy and safety of six frequently-used biologics through a Bayesian method.MethodsWeb of Science, Scopus, CENTRAL, ClinicalTrials.gov and ICTRP were searched to collect randomized controlled trials (RCTs) in adults with moderate-to-severe Crohn’s disease, comparing Infliximab, Adalimumab, Certolizumab pegol, Ustekinumab, Risankizumab, or Vedolizumab, relative to placebo or an active comparator for induction of clinical response (two different definitions) and maintenance of clinical remission. A random-effects model was performed with rankings according to the surface under cumulative ranking curve (SUCRA) probability. Finally, we completed sensitivity and consistency analyses, and evaluated the certainty of evidence through GRADE working group guidance.ResultsWe identified 22 and 20 RCTs for induction and maintenance therapy, respectively. Infliximab combined with azathioprine was most effective for inducing clinical response in TNF (tumor necrosis factor) antagonist-naïve patients. For TNF antagonist-experienced patients, Ustekinumab (SUCRA 86.19) and Risankizumab (SUCRA 62.56) have the largest SUCRA in induction of clinical response. Risankizumab has the lowest risk of adverse events (SUCRA 84.81), serious adverse events (SUCRA 94.23), and serious infections (SUCRA 79.73) in induction therapy. Adalimumab and the 10 mg/kg regimen of Infliximab rank highest for maintaining clinical remission.ConclusionThis analysis suggests that Infliximab in combination with azathioprine may be preferred biologic agents for induction therapy in TNF antagonist-naïve patients. For TNF antagonist-experienced patients, Ustekinumab and Risankizumab may be preferred biologic agents for induction therapy. Risankizumab potentially has the lowest safety risk worth exploring in induction therapy. Abstract Despite the prevalent of hexagonal, tetragonal, and triangular pore structures in two-dimensional covalent organic frameworks (2D COFs), the pentagonal pores remain conspicuously absent. We herein present the Cairo pentagonal tessellated COFs, achieved through precisely chosen geometry and metrics of the linkers, resulting in unprecedented mcm topology. In each pentagonal structure, porphyrin units create four uniform sides around 15.5 Å with 90° angles, while tetrabiphenyl unit establish a bottom edge about 11.6 Å with 120° angles, aligning precisely with the criteria of Cairo Pentagon. According to the narrow bandgap and strong near-infrared (NIR) absorbance, as-synthesized COFs exhibit the efficient singlet oxygen (1O2) generation and photothermal conversion, resulting in NIR photothermal combined photodynamic therapy to guide cancer cell apoptosis. Mechanistic studies reveal that the good 1O2 production capability upregulates intracellular lipid peroxidation, leading to glutathione depletion, low expression of glutathione peroxidase 4, and induction of ferroptosis. The implementation of pentagonal Cairo tessellations in this work provides a promising strategy for diversifying COFs with new topologies, along with multimodal NIR phototherapy.

Hereditary factors contribute to the pathogenesis of pediatric leukemia. However, few studies have reported gene mutation pathopoeias. This paper reports genetic mutations associated with hereditary acute lymphoblastic leukemia. We reported a case of siblings diagnosed with acute lymphoblastic leukemia when aged 3 and 7 years, both siblings are alive after chemotherapy, and whole exome sequencing analysis was performed on the siblings and their parents. It was observed that both siblings had diheterozygous mutations in PML gene ( PML , NM_033250, exon7, c.2170A > G, p.S724G; PML , NM_033250, exon7, c.2195G > T, p.G732V), and their parents had heterozygous mutations in one mutation site of PML gene, respectively, suggesting that the diheterozygous mutations of PML gene might be causal genetic genes for the occurrence of acute lymphoblastic leukemia.

In this study, WC-12Co cemented carbides were prepared by laser powder bed fusion (LPBF) on a WC-20Co substrate. The effects of the scanning speed and the substrate on the microstructure, mechanical properties and wear characteristics of the WC-12Co cemented carbide were investigated. The results showed that an alternating distribution of coarse and fine WC grains is observed in the LPBF-prepared WC-12Co cemented carbides. As the laser scanning speed decreases or the laser energy density increases, the WC phase gradually decomposes, accompanied by the loss of C and the formation of ƞ phases. WC-12Co cemented carbides produced at a lower scanning speed exhibit a higher density of thermal cracks, while those produced at a higher scanning speed have a larger number of pores. As the laser scanning speed increases, the transverse rupture strength (TRS) first increases to a maximum value of 823.13 MPa and then decreases. The variation of TRS is mainly attributed to the evolution of the brittle ƞ-phase and metallurgical defects. For WC-12Co cemented carbides formed at low scanning speeds, the brittle η-phase may fracture and cut through the matrix during the friction process, leading to an increase in coefficient of friction (COF) and wear mass loss. For samples formed at high scanning speeds, the formation and detachment of the shear layer could accelerate the friction process and undermine the wear resistance of the WC-12Co cemented carbide. Therefore, the sample prepared at the scanning speed of 400 mm/s shows the best wear performance.

The trade-off between growth and defense is common in plants. We previously demonstrated that BnaA03.WRKY28 weakened resistance strength but promoted shoot branching in Brassica napus (rapeseed). However, the molecular mechanism by which WRKY28 promotes branching formation is still obscure. In this study, we found that BnaA01.BRC1, BnaC01.BRC1, and BnaC03.BRC1 are mainly expressed in the leaf axils and contained W-box cis-acting elements in the promoter regions. BnaA03.WRKY28 directly bound to the promoter regions of these three copies and inhibited their expression. The brc1 mutants, the BnaA01.BRC1, BnaC01.BRC1 BnaA03.BRC1 and BnaC03.BRC1 were simultaneously knocked out, mediated by CRISPR/Cas9, and exhibited excessive branching. The expression level of the ABA biosynthesis encoding gene NCED3 was significantly reduced in the mutant compared to that in the WT. Instead, the expression level of the ABA catabolism encoding gene CYP707A3 was significantly higher than that in WT. These results suggest that the excessive branching of the brc1 mutant may be caused by the release of ABA-mediated bud dormancy. This study provides direct evidence for the potential mechanism of the WRKY28-BRC1 transcription factor module contributing to shoot branching in rapeseed.

Brassica napus is one of the most important oil crops in the world. Breeding oilseed rape with colorful flowers can greatly enhance the ornamental value of B. napus and thus improve the economic benefits of planting. As water-soluble flavonoid secondary metabolites, anthocyanins are very important for the synthesis and accumulation of pigments in the petals of plants, giving them a wide range of bright colors. Despite the documentation of over 60 distinct flower shades in B. napus, the intricacies underlying flower color variation remain elusive. Particularly, the mechanisms driving color development across varying flower color backgrounds necessitate further comprehensive investigation. This research undertook a comprehensive exploration through the integration of transcriptome and metabolome analyses to pinpoint pivotal genes and metabolites underpinning an array of flower colors, including beige, beige-red, yellow, orange-red, deep orange-red, white, light-purple, and purple. First, we used a two-way BLAST search to find 275 genes in the reference genome of B. napus Darmor v10 that were involved in making anthocyanins. The subsequent scrutiny of RNA-seq outcomes underscored notable upregulation in the structural genes F3H and UGT, alongside the MYB75, GL3, and TTG1 transcriptional regulators within petals, showing anthocyanin accumulation. By synergizing this data with a weighted gene co-expression network analysis, we identified CHS, F3H, MYB75, MYB12, and MYB111 as the key players driving anthocyanin synthesis in beige-red, orange-red, deep orange-red, light-purple, and purple petals. By integrating transcriptome and weighted gene co-expression network analysis findings with anthocyanin metabolism data, it is hypothesized that the upregulation of MYB75, which, in turn, enhances F3H expression, plays a pivotal role in the development of pigmented oilseed rape flowers. These findings help to understand the transcriptional regulation of anthocyanin biosynthesis in B. napus and provide valuable genetic resources for breeding B. napus varieties with novel flower colors.

Abstract The programmed cell death protein 1 (PD-1) is an inhibitory receptor on T cells and plays an important role in promoting cancer immune evasion. While ubiquitin E3 ligases regulating PD-1 stability have been reported, deubiquitinases governing PD-1 homeostasis to modulate tumor immunotherapy remain unknown. Here, we identify the ubiquitin-specific protease 5 (USP5) as a bona fide deubiquitinase for PD-1. Mechanistically, USP5 interacts with PD-1, leading to deubiquitination and stabilization of PD-1. Moreover, extracellular signal-regulated kinase (ERK) phosphorylates PD-1 at Thr234 and promotes PD-1 interaction with USP5. Conditional knockout of Usp5 in T cells increases the production of effector cytokines and retards tumor growth in mice. USP5 inhibition in combination with Trametinib or anti-CTLA-4 has an additive effect on suppressing tumor growth in mice. Together, this study describes a molecular mechanism of ERK/USP5-mediated regulation of PD-1 and identifies potential combinatorial therapeutic strategies for enhancing anti-tumor efficacy.

Background: Osteosarcoma (OS) is the most common primary malignancy in bone tissues, and effective therapeutics remain absent in clinical practice. Traditional Chinese medicines (TCM) have been used for thousands of years, which provide great insights into OS management. Gallic acid (GA) is a natural phenolic acid enriched in various foods and herbs. Several pharmacological activities of GA such as anti-oxidation and anti-inflammation have been well-established. However, its biological function in OS remains not fully understood. Methods: The potential anti-cancer properties of GA were evaluated in 143 ​B, U2OS and MG63 ​cells. Its effects on cell growth, cell cycle, apoptosis and migration were examined in these OS cells. The lncRNA H19 and Wnt/β-catenin signaling were detected by qPCR, luciferase activity and Western blotting assays. The in vivo effect of GA on tumor growth was investigated using an orthotopic mouse model. Results: In the present study, GA was found to suppress the tumor growth in vitro via inducing cell cycle arrest and apoptosis in OS cells, and inhibit the invasion and metastasis as well. Using the orthotopic animal model, GA was also found to suppress tumorigenesis in vivo. Long noncoding RNA (lncRNA) H19 was demonstrated to be down-regulated by GA, and thus disrupted the canonical Wnt/β-catenin signaling in OS cells. Furthermore, the ectopic expression of H19 rescued the GA-induced suppressive effects on tumor growth and metastasis, and partially reversed the inactivation of Wnt/β-catenin signaling. Conclusions: Taken together, our results indicated that GA inhibited tumor growth through an H19-mediated Wnt/β-catenin signaling regulatory axis in OS cells. The translational potential of this article: The information gained from this study provides a novel underlying mechanism of GA mediated anti-OS activity, suggesting that GA may be a promising drug candidate for OS patients.

A 64-year-old man presented with painful rash on the trunk and extremities for more than half a month. Nephrotic syndrome had been diagnosed 2 years ago and had been regularly treated with glucocorticoids and immunosuppressants. He developed herpes zoster 1 year ago. Laboratory investigation demonstrated varicella-zoster virus DNA (VZV-DNA), varicella-zoster virus IgM (VZV-IgM), and varicella-zoster virus antibody IgG (VZV-IgG) were all positive. After the clinical diagnosis of relapsed disseminated cutaneous herpes zoster had been confirmed, systemic therapy with brivudine 125 mg everyday was started.

Increase in air pollution caused by acidic gases, such as nitrogen dioxide, call for the need to study potential effects on the structure of textile materials. Wool is a natural protein fiber which is easily damaged due to rich reaction positions and cross-linked bonds in the fiber. This study reports, for the first time, effect of nitrogen dioxide, as a model acid gas pollutant, on the structure of wool. On the macro level, wool fabric undergoes strength loss after incubation in nitrogen dioxide for 6 h. Furthermore, although there was no obvious difference on the surface morphology as shown by SEM analysis, FTIR and Raman tests showed that the probable damage mechanism is by hydrolysis of amino bond, and oxidation of tyrosine residues and disulfide bonds, which was ascertained using XPS and amino acid analysis. In addition, crystallinity degree and α-helix structure content decreased after nitrogen dioxide treatment as shown by XRD and NMR results. These findings show that nitrogen dioxide has significant effect on the structure of wool. The study offers a novel approach for investigation of the interaction between gas pollutants and protein fiber, and provide information for protection of textiles against similar pollutants.

In this study, WC-12Co cemented carbides were fabricated by selective laser melting (SLM) and subsequently subjected to sintering. Effects of the scanning speed on the microstructure, mechanical properties and friction and wear performance of the prepared WC-12Co cemented carbides were investigated. Based on the XRD spectra, η phase appeared after the SLM process, confirming a loss of carbon during the preparation of WC-12Co. The WC average grain size increased with improving the scanning speed, which fluctuated in the range of 1.5 μm–2.5 μm. The relatively small WC average grain size was associated with the lamellar structure (alternating layers of coarse and fine WC grains) caused by the uneven temperature distribution of SLM. Co evaporation was quantitatively revealed by EBSD phase maps. Relationships between hardness, transverse rupture strength (TRS) and scanning speeds were analyzed. The comprehensive performance of the samples reached the best at the scanning speed of 400 mm/s, which the hardness, TRS was 81 HRA, and 650 MPa, respectively. Furthermore, after process optimization, the friction and wear properties of the material were tested, the friction coefficient was about 0.2 at room temperature, while 0.5–0.7 at 200°C–600°C.

Auxin is essential for plant growth and development, also closely related to plant architecture. YUCCA (YUC) proteins catalyse the rate-limiting step for endogenous auxin biosynthesis. In this study, we identified 57 BnYUC genes in Brassica napus (rapeseed) genome, located on 17 chromosomes. Among them, A09 chromosome contained the most BnYUCs as 6. Phylogenetic tree showed that all BnYUC members were divided into 2 evolutionary clusters, including 7 sub-groups. Among the sub-groups, CladeⅠ-C consisted 20 members, evolved independently from Arabidopsis thaliana, and motif 10 was unique. Genomic collinearity analysis showed that YUC collinear gene pairs widely existed in Brassica species, indicating that this family has a high retention rate in the evolution. Expression profile suggested that the BnYUC family genes tended to express in specific growth/development stages as well as suffering stress/hormone treatments. It suggested that dominant expression of specific members existed with each development stage or treatment, indicating that each BnYUC might have a clear division of work, and play a given biological function. This work was expected to build a basis for further study of the involvement of BnYUC in growth, development and stress response.

The regulatory mechanisms of PD-L1 posttranslational modifications are not completely understood. Here the authors show that USP8 negatively regulates PD-L1 protein abundance by removing the K63-linked ubiquitination of PD-L1; while USP8 inhibition increases MHC-I expression and triggers anti-tumour immune responses through activating NF-κB signalling.

A patient presented with erythema and oedema on the right eyelid, malar region and left thigh with pruritus and pain. She was diagnosed with erysipelas and had been treated with anti-inflammatory drugs, but the symptoms did not improve. Laboratory investigation and biopsy showed that she was consistent with lupus erythematosus tumidus. It's a typical and rare diease relatively.

Aberrant expression of long non-coding RNAs (lncRNAs) has been reported in multiple cancers. However, the underlying mechanisms mediated by super-enhancers remain elusive. Here we sought to define the role of a novel lncRNA termed lncRNA-DAW in tumorigenesis. Our results revealed that lncRNA-DAW was driven by a liver-specific super-enhancer and transcriptionally activated by HNF4G, leading to frequent elevation in hepatocellular carcinoma (HCC) specimens. Ectopic expression of lncRNA-DAW promoted both in vivo and in vitro tumor growth. By using RNA sequencing, Wnt2 was screened out as a downstream effector of lncRNA-DAW. We next found that lncRNA-DAW physically interacted with EZH2, a negative regulator of Wnt2. This interplay subsequently potentiated CDK1-EZH2 interaction, leading to the phosphorylation and ubiquitination of EZH2. The lncRNA-DAW-mediated EZH2 degradation facilitated the de-repression of Wnt2 transcription, which eventually activated the Wnt/β-catenin pathway. Furthermore, we verified that Wnt2 potentiated in vitro and in vivo cancer cell growth by activating the Wnt/β-catenin pathway. Finally, Wnt2 amplification was confirmed as a common event in liver cancer, and the expression of lncRNA-DAW was positively correlated with Wnt2 in HCC specimens. Collectively, we are the first to identify lncRNA-DAW as a novel candidate oncogene in liver cancer, and this lncRNA may serve as a novel clinical diagnosis biomarker for liver cancer.

Objective To explore the relationship between the serum concentration of IL-17 and the pathogenesis of psoriasis. Methods ampere database was established through literature search, and meta-analysis was used to evaluate the effect indicator of the included literatures. Results 5 articles were included in the study, and the quality evaluation score was 6–7 points by the Newcastle-Ottawa Scale (NOS) criteria. There was heterogeneity in the standardized mean difference (SMD) between the two groups (I 2 = 98.63%). The random effect model analysis showed that the combined SMD = 2.11, 95%CI (0.79, 3.43), the effect size between studies was statistically significant (z = 3.13, p = 0.00). Conclusion There was a statistical difference in serum IL-17 concentration between the case group and the control group. The serum concentration of IL-17 in the case group was higher than that in the control group.

The siRNA-loaded lipid nanoparticles have attracted much attention due to its significant gene silencing effect and successful marketization. However, the in vivo distribution and release of siRNA still cannot be effectively monitored. In this study, based on the fluorescence resonance energy transfer (FRET) principle, a fluorescence dye Cy5-modified survivin siRNA was conjugated to nanogolds (Au-DR-siRNA), which were then wrapped with lipid nanoparticles (LNPs) for monitoring the release behaviour of siRNA in vivo. The results showed that once Au-DR-siRNA was released from the LNPs and cleaved by the Dicer enzyme to produce free siRNA in cells, the fluorescence of Cy5 would change from quenched state to activated state, showing the location and time of siRNA release. Besides, the LNPs showed a significant antitumor effect by silencing the survivin gene and a CT imaging function superior to iohexol by nanogolds. Therefore, this work provided not only an effective method for monitoring the pharmacokinetic behaviour of LNP-based siRNA, but also a siRNA delivery system for treating and diagnosing tumors.

Acquisition of CDK4/6i resistance represents a major clinical challenge. Here, the authors report that inhibition of CK1ε can prevent acquisition of CDK4/6i resistance, potentiating the therapeutic efficacy of CDK4/6i in human breast cancer.

Objective: To analyze the clinicopathological characteristics of skin basal cell carcinoma, so as to improve the coincidence rate of clinical diagnosis and provide basis for early diagnosis, treatment and prognosis. Methods: The clinical and pathological features of 611 patients with basal cell carcinoma diagnosed by primary biopsy in Dermatology Hospital, Southern Medical University from March 2014 to February 2021 were retrospectively analyzed. Results: There were 268 males and 343 females in 611 patients, the ratio of male to female was 1 ∶1.28; the onset age was 19~92 (64.18±13.90) years old. There were 296 cases (48.45%) with mass, nodule and tumor, 248 cases (40.59%) with patch and papule, and 67 cases (10.97%) with erosion or ulcer, scar and cyst. Pathological classification: nodular type 377 cases (61.71%), infiltrative type 85 cases (13.92%), superficial type 69 cases (11.30%), other types 80 cases (13.10%). Clinical misdiagnosis rate was 16.86%. Conclusions: The main clinical manifestation is mass, and the main pathological type is nodule. It is important to improve the diagnosis, treatment and prognosis of BCC by improving the rate of skin biopsy and strengthening the understanding of easily misdiagnosed lesions.

Based on previous researches, we further investigated the multi-silique trait in rapeseed (Brassica napus L.) line zws-ms. In this study, we used a relatively comprehensive list of flowering related genes in rapeseed and compared them between zws-ms and its near-isogenic line (NIL) zws-217. Genes were studied on genome, transcriptome and proteome levels and then we focused on genes with non-synonymous single nucleotide polymorphism (SNP) or frame-shift insertion-deletion (InDel), finding some genes on the list which changes their sequences. Then, combined with their annotation and the information of their orthologs, certain genes such as BnaA09g05900D, ortholog of AGAMOUS-LIKE 42 (AGL42), which encodes an MADS-box protein, were assumed as probably responsible for the multi-silique trait. Also, we analyzed the Differentially Accumulated Proteins (DAPs) between zws-ms and zws-217, revealing some genes involved in homologous recombination and mismatch repair pathways. Since the development of flowers/siliques is crucial to crops and it influences the yield of rapeseed, this study paved a way to deeply understand the mechanism of the multi-pistil flower formation, which may facilitate researches on rapeseed production in future.

Blackleg is a serious disease in Brassica plants, causing moderate to severe yield losses in rapeseed worldwide. Although China has not suffered from this disease yet (more aggressive Leptosphaeria maculans is not present yet), it is crucial to take provisions in breeding for disease resistance to have excellent blackleg-resistant cultivars already in the fields or in the breeding pipeline. The most efficient strategy for controlling this disease is breeding plants with identified resistance genes. We selected 135 rapeseed accessions in Sichuan, including 30 parental materials and 105 hybrids, and we determined their glucosinolate and erucic acid content and confirmed 17 double-low materials. A recently developed single-nucleotide polymorphism (SNP) marker, SNP_208, was used to genotype allelic Rlm1/rlm1 on chromosome A07, and 87 AvrLm1-resistant materials. Combined with the above-mentioned seed quality data, we identified 11 AvrLm1-resistant double-low rapeseed accessions, including nine parental materials and two hybrids. This study lays the foundation of specific R gene-oriented breeding, in the case that the aggressive Leptosphaeria maculans invades and establishes in China in the future and a robust and less labor consuming method to identify resistance in canola germplasm.

ObjectiveTo investigate the correlation between gene mutations and glucocorticoid resistance in pediatric acute lymphoblastic leukemia (ALL).MethodsA total of 71 children with ALL admitted to our center between September 2019 and September 2021 were enrolled. DNA obtained from bone marrow or peripheral blood samples at initial diagnosis was used for genetic testing via whole exome sequencing. Meanwhile, patient clinical information was collected. Subsequently, the correlations of gene mutations with clinical features and glucocorticoid resistance were analyzed.ResultsOf the 71 children enrolled, 61 (85.9%) had B-cell ALL (B-ALL) and 10 (14.1%) had T-cell ALL (T-ALL). The five genes with the highest mutation frequency in B-ALL were TTN (24.4%), FLT3 (14.6%), TP53 (14.6%), MUC16 (9.8%), and EPPK1 (9.8%). In contrast, those with the highest frequency in T-ALL were NOTCH1 (54.5%), FBXW7 (27.3%), TTN (27.3%), MUC16 (27.3%), and PHF6 (18.2%). Upon statistical analysis, TTN and NOTCH1 mutations were found to be associated with prednisone resistance. Further, TTN and MUC16 mutations were associated with a lower age at diagnosis, and NOTCH1 mutations were associated with T-ALL in female patients. Leukocyte counts and LDH levels did not differ based on the presence of any common gene mutation, and no association between these gene mutations and overall survival was observed.ConclusionsOur study is the first to demonstrate the association between TTN mutation and glucocorticoid resistance in ALL. Our findings could guide strategies for overcoming drug resistance and aid in the development of drug targets.

Abstract Background The expression of long non‐coding RNA (lncRNA) prostate cancer‐associated ncRNA transcripts 1 (PCAT1) is increased in non‐small cell lung cancer (NSCLC). It stimulates tumour growth and metastasis, but its role in the radioimmune responses remain unknown. We aimed to explore the impacts of PCAT1 on tumorigenesis and radioimmune responses and the underlying molecular mechanisms in NSCLC. Methods Comprehensive bioinformatics analysis was performed to identify immunosuppressive lncRNAs involved with tumour invasion in NSCLC. The expression levels of PCAT1 were analysed by in situ hybridisation in 55 paired NSCLC tissues and adjacent normal tissues. Both loss‐ and gain‐of‐function assays were performed to examine the effects of PCAT1 and SOX2 on NSCLC cell behaviours in vivo and in vitro. Bioinformatic analyses, chromatin isolation by RNA purification (ChIRP) and dual‐luciferase reporter assays were applied to validate the regulatory effects of PCAT1 on SOX2 expression. Chromatin immunoprecipitation, luciferase and rescue assays were utilised to identify the relationship between SOX2 and the cGAS/stimulator of interferon genes (STING) signalling. Results PCAT1 was immunosuppressive and related with NSCLC invasion. Increased PCAT1 was negatively correlated with immune cell infiltration in NSCLC. PCAT1 knockdown restrained proliferation, increased apoptosis, and repressed cell metastasis in vivo and in vitro. PCAT1 activated SOX2 that accelerated tumorigenesis and immunosuppression. SOX2 promoted tumour growth through inhibiting cytotoxic T‐cell immunity. Moreover, SOX2 restrained cGAS transcription and hampered downstream type I interferon (IFN)‐induced immune responses. Inhibition of PCAT1/SOX2 in collaboration with radiation further inhibited tumour growth, and initiated the cGAS/STING signalling pathway, which enhanced the immune responses of radiotherapy in NSCLC. Conclusions PCAT1/SOX2 axis promoted tumorigenesis and immunosuppression through inhibition of cGAS/STING signalling‐mediated T‐cell activation. Inhibition of PCAT1 and SOX2 synergised with radiotherapy to activate the immune response and could serve as potential therapeutic targets.

Bacillus amyloliquefaciens is a generally recognized as safe (GRAS) microorganism that presents great potential for the production of heterologous proteins. In this study, we performed genomic and comparative transcriptome to investigate the critical modular in B. amyloliquefaciens on the production of heterologous alkaline proteases (AprE). After investigation, it was concluded that the key modules affecting the production of alkaline protease were the sporulation germination module (Module I), extracellular protease synthesis module (Module II), and extracellular polysaccharide synthesis module (Module III) in B. amyloliquefaciens. In Module I, AprE yield for mutant BA ΔsigF was 25.3% greater than that of BA Δupp. Combining Module I synergistically with mutation of extracellular proteases in Module II significantly increased AprE production by 36.1% compared with production by BA Δupp. In Module III, the mutation of genes controlling extracellular polysaccharides reduced the viscosity and the accumulation of sediment, and increased the rate of dissolved oxygen in fermentation. Moreover, AprE production was 39.6% higher than in BA Δupp when Modules I, II and III were engineered in combination. This study provides modular engineering strategies for the modification of B. amyloliquefaciens for the production of alkaline proteases.

There has been sufficient evidence for the relationship between lifestyle and insomnia in the general population, but for individuals who already suffer from insomnia, it is not clear whether a healthy lifestyle can also pose similar benefits. The present study investigated the roles of different aspects of lifestyle in the development of individual insomnia by tracking insomnia symptoms of college students during the COVID-19 lock-down. Two surveys were conducted on 65,200 college students in the process of home isolation in Guangdong Province of China, at the pandemic outbreak period (T1) and the initial remission period (T2), respectively. Given the objectives of the present study, a total of 1,702 college students with clinical insomnia from T1 were selected as subjects. Insomnia symptoms were assessed using the Youth Self Rating Insomnia Scale (YSIS), while demographic information, epidemic exposure, and lifestyle were all measured by self-developed questionnaire, through network survey. The 1,702 college students (mean age ± standard deviation, 20.06 ± 1.46, range 16–25; 71.9% females) with insomnia symptoms were divided into three trajectory groups: recovery group, remission group, and chronic insomnia group according to their insomnia scores in T2 phase. The results showed that there was no significant difference in demographic backgrounds or epidemic exposure among the three groups, however, there were significant differences in food intake, exercise, and Internet use. The regression results further showed that both the recovery group and the remission group adopted more regular food intake than the chronic group. The recovery group exhibited better daily exercise habits than both the remission group and the chronic group. The duration of Internet use was significantly shorter for the recovery group than for the chronic group. These findings indicate a strong relationship between the lifestyle and the recovery of insomnia for college students isolated at home during the epidemic period. Significance of the different aspects of lifestyle on the recovery of insomnia are discussed.

The sequential occurrence of Langerhans cell histiocytosis and acute leukemia in only one individual has been reported previously; however, it is rarely observed that Langerhans cell histiocytosis can transform into acute lymphoblastic leukemia, and the underlying mechanisms remain unclear. In this report, we have analyzed a case of acute lymphoblastic leukemia converted from Langerhans cell histiocytosis using high-throughput sequencing method, and found that mitogen-activated protein kinase gene mutation, which can act as a marker for poor prognosis, might be involved in disease transformation. This is the first description about acute lymphoblastic leukemia B-cell type after Langerhans cell histiocytosis diagnosis and therapy in China.

In this study, seven 316L stainless steel (316L SS) bulks with different angles (0°, 15°, 30°, 45°, 60°, 75°, and 90°) relative to a build substrate were built via selective laser melting (SLM). The influences of different angles on the metallography, microstructure evolution, tensile properties, and corrosion resistance of 316L SS were studied. The 0° sample showed the morphology of corrugated columnar grains, while the 90° sample exhibited equiaxed grains but with a strong texture. The 60° sample had a good strength and plasticity: the tensile strength with 708 MPa, the yield strength with 588 MPa, and the elongation with 54.51%. The dislocation strengthening and grain refinement play a vital role in the mechanical properties for different anisotropy of the SLM-fabricated 316L SS. The 90° sample had greater toughness and corrosion resistance, owing to the higher volume fraction of low-angle grain boundaries and finer grains.

Continuous optimization of source, grid and load factors is needed to solve the renewable energy curtailment problem. This article focuse d on the decomposition algorithm of contribution to renewable energy accommodation under the combined effect of source-grid-load. From the point of view of the planning, development scenarios were established under different source-grid-load factors, based on production simulation. Source-grid-load contributions to renewable energy accommodation were calculated by sensitivity analysis method and Shapley value method, respectively. The sensitivity analysis calculate contribution by judgment factors according to a specific scenario. It is suitable to give a list of measures for an established stage of development. The Shapley value method is a comprehensive weighting for the contribution of all development scenarios. It is more suitable for examining the whole process of development from the present situation to the level of planning.

Abstract Background Although rapeseed (Brassica napus L.) mutant forming multiple siliques was morphologically described and considered to increase the silique number per plant, an important agronomic trait in this crop, the molecular mechanism underlying this beneficial trait remains unclear. Here, we combined bulked-segregant analysis (BSA) and whole genome re-sequencing (WGR) to map the genomic regions responsible for the multi-silique trait using two pools of DNA from the near-isogenic lines (NILs) zws-ms (multi-silique) and zws-217 (single-silique). We used the Euclidean Distance (ED) to identify genomic regions associated with this trait based on both SNPs and InDels. We also conducted transcriptome sequencing to identify differentially expressed genes (DEGs) between zws-ms and zws-217. Results Genetic analysis using the ED algorithm identified three SNP- and two InDel-associated regions for the multi-silique trait. Two highly overlapped parts of the SNP- and InDel-associated regions were identified as important intersecting regions, which are located on chromosomes A09 and C08, respectively, including 2044 genes in 10.20-MB length totally. Transcriptome sequencing revealed 129 DEGs between zws-ms and zws-217 in buds, including 39 DEGs located in the two abovementioned associated regions. We identified candidate genes involved in multi-silique formation in rapeseed based on the results of functional annotation. Conclusions This study identified the genomic regions and candidate genes related to the multi-silique trait in rapeseed.

Abstract Background Tendons are a crucial component of the musculoskeletal system and responsible for transmission forces derived from muscle to bone. Patients with tendon injuries are often observed with decreased collagen production and matrix degeneration, and healing of tendon injuries remains a challenge as a result of limited understanding of tendon biology. Recent studies highlight the contribution of miR-378a on the regulation gene expression during tendon differentiation. Methods We examined the tendon microstructure and tendon repair with using miR-378a knock-in transgenic mice, and the tendon-derived stem cells were also isolated from transgenic mice to study their tenogenic differentiation ability. Meanwhile, the expression levels of tenogenic markers were also examined in mouse tendon-derived stem cells transfected with miR-378a mimics during tenogenic differentiation. With using online prediction software and luciferase reporter assay, the binding target of miR-378a was also studied. Results Our results indicated miR-378a impairs tenogenic differentiation and tendon repair by inhibition collagen and extracellular matrix production both in vitro and in vivo. We also demonstrated that miR-378a exert its inhibitory role during tenogenic differentiation through binding at TGFβ2 by luciferase reporter assay and western blot. Conclusions Our investigation suggests that miR-378a could be considered as a new potential biomarker for tendon injury diagnosis or drug target for a possible therapeutic approach in future clinical practice.

Until now, Streptoverticillium mobaraense transglutaminase (TG) is the only commercialized TG, but limited information is known about its selection tendency on crosslinking sites at the protein level, restricting its application in the food industry. Here, four recombinant Bacillus TGs were stable in a broad range of pH (5.0–9.0) and temperatures (B. cereus (BCETG) demonstrated the maximal specific activity of 177 U/mg. A structural analysis indicated that the Ala147-Ala156 region in the substrate tunnel of BCETG played a vital role in catalytic activity. Furthermore, bovine serum albumin, as well as nearly all protein ingredients in soy protein isolate and whey protein, could be cross-linked by BCETG, and the internal crosslinking paths of three protein substrates were elucidated. This study demonstrated Bacillus TGs are a candidate for protein crosslinking and provided their crosslinking mechanism at the protein level for applications in food processing.

In this study, a novel low molecular weight of acetylaminoglucan (AGA) was obtained and its antitumor activity on H22 tumor-bearing mice was investigated. The results of UV, HPLC and FT-IR showed that AGA present high purity with low molecular weight of 2.76 × 103 Da. Animal experiments showed that AGA could inhibit the proliferation of tumor cells in H22 tumor-bearing mice by protecting the immune organs, enhancing the phagocytosis ability of macrophages, killing activity of NK cells and proliferation capacity of lymphocytes, improving the levels of cytokines in vivo and regulating the distribution of lymphocyte subsets, and the tumor inhibition rate reached to 52.74% (50 mg/kg). Cell cycle determination further indicated that AGA could induce apoptosis of tumor cells and arrests it in S phase. These results will provide a data basis for the potential application of AGA in pharmaceutical industry.

In higher plants, the structure of a flower is precisely controlled by a series of genes. An aberrance flower results in abnormal fruit morphology. Previously, we reported multi-silique rapeseed (Brassica napus) line zws-ms. We identified two associated regions and investigated differentially expressed genes (DEGs); thus, some candidate genes underlying the multi-silique phenotype in warm area Xindu were selected. However, this phenotype was switched off by lower temperature, and the responsive genes, known as thermomorphogenesis-related genes, remained elusive. So, based on that, in this study, we further investigated the transcriptome data from buds of zws-ms and its near-isogenic line zws-217 grown in colder area Ma’erkang, where both lines showed normal siliques only, and the DEGs between them analyzed. We compared the 129 DEGs from Xindu to the 117 ones from Ma’erkang and found that 33 of them represented the same or similar expression trends, whereas the other 96 DEGs showed different expression trends, which were defined as environment-specific. Furthermore, we combined this with the gene annotations and ortholog information and then selected BnaA09g45320D (chaperonin gene CPN10-homologous) and BnaC08g41780D [Seryl-tRNA synthetase gene OVULE ABORTION 7 (OVA7)-homologous] the possible thermomorphogenesis-related genes, which probably switched off the multi-silique under lower temperature. This study paves a way to a new perspective into flower/fruit development in Brassica plants.

Herpes zoster (HZ), caused by the varicella-zoster virus, is an infectious skin disease that rarely recurs after initial presentation. The mechanism underlying HZ recurrence is currently under investigation. In this article, we report a case of HZ relapse within 1 month. Analysis of patient’s clinical manifestations, histopathological features, and flow cytometry results indicated that the absolute and percentage values of B cells were below the lower limit. We hypothesized that the patient had abnormal humoral immune function, which may be one reason leading to the HZ relapse within 1 month. The findings of this case will serve as useful reference for HZ recurrence for clinicians. This case was impactful and added to the literature on HZ recurrence.

Leaf is an important organ for higher plants, and the shape of it is one of the crucial traits of crops. In this study, we investigated a unique aberrant leaf morphology trait in a mutational rapeseed material, which displayed ectopic blade-like outgrowths on the adaxial side of leaf. The abnormal line 132000B-3 was crossed with the normal line 827-3. Based on the F2:3 family, we constructed two DNA pools (normal pool and abnormal pool) by the bulked segregant analysis (BSA) method and performed whole genome re-sequencing (WGR), obtaining the single-nucleotide polymorphism (SNP) and insertion/deletion (InDel) data. The SNP-index method was used to calculate the Δ(SNP/InDel-index), and then an association region was identified on chromosome A10 with a length of 5.5 Mbp, harboring 1048 genes totally. Subsequently, the fine mapping was conducted by using the penta-primer amplification refractory mutation system (PARMS), and the associated region was narrowed down to a 35.1-kbp segment, containing only seven genes. These seven genes were then analyzed according to their annotations and finally, BnA10g0422620 and BnA10g0422610, orthologs of LATE MERISTEM IDENTITY1 (LMI1) gene from Arabidopsis and REDUCED COMPLEXITY (RCO) gene from its relative Cardamine hirsuta, respectively, were identified as the candidate genes responding to this blade-like outgrowth trait in rapeseed. This study provides a novel perspective into the leaf formation in Brassica plants.

Background Flower and fruit development are vital stages of the angiosperm lifecycle. We previously investigated the multi-silique trait in the rapeseed (Brassica napus) line zws-ms on a genomic and transcriptomic level, leading to the identification of two genomic regions and several candidate genes associated with this trait. However, some events on the transcriptome level, like alternative splicing, were poorly understood. Methods Plants from zws-ms and its near-isogenic line (NIL) zws-217 were both grown in Xindu with normal conditions and a colder area Ma’erkang. Buds from the two lines were sampled and RNA was isolated to perform the transcriptomic sequencing. The numbers and types of alternative splicing (AS) events from the two lines were counted and classified. Genes with AS events and expressed differentially between the two lines, as well as genes with AS events which occurred in only one line were emphasized. Their annotations were further studied. Results From the plants in Xindu District, an average of 205,496 AS events, which could be sorted into five AS types, were identified. zws-ms and zws-217 shared highly similar ratios of each AS type: The alternative 5′ and 3′ splice site types were the most common, while the exon skipping type was observed least often. Eleven differentially expressed AS genes were identified, of which four were upregulated and seven were downregulated in zws-ms. Their annotations implied that five of these genes were directly associated with the multi-silique trait. While samples from colder area Ma’erkang generated generally reduced number of each type of AS events except for Intron Retention; but the number of differentially expressed AS genes increased significantly. Further analysis found that among the 11 differentially expressed AS genes from Xindu, three of them maintained the same expression models, while the other eight genes did not show significant difference between the two lines in expression level. Additionally, the 205 line-specific expressed AS genes were analyzed, of which 187 could be annotated, and two were considered to be important. Discussion This study provides new insights into the molecular mechanism of the agronomically important multi-silique trait in rapeseed on the transcriptome level and screens out some environment-responding candidate genes.

Abstract Background Florfenicol is a derivative of chloramphenicol that is used only for the treatment of animal diseases. A key resistance gene for florfenicol, floR, can spread among bacteria of the same and different species or genera through horizontal gene transfer. To analyze the potential transmission of resistance genes between animal and human pathogens, we investigated floR in Klebsiella pneumoniae isolates from patient samples. floR in human pathogens may originate from animal pathogens and would reflect the risk to human health of using antimicrobial agents in animals. Methods PCR was used to identify floR-positive strains. The floR genes were cloned, and the minimum inhibitory concentrations (MICs) were determined to assess the relative resistance levels of the genes and strains. Sequencing and comparative genomics methods were used to analyze floR gene-related sequence structure as well as the molecular mechanism of resistance dissemination. Results Of the strains evaluated, 20.42% (67/328) were resistant to florfenicol, and 86.96% (20/23) of the floR-positive strains demonstrated high resistance to florfenicol with MICs ≥512 μg/mL. Conjugation experiments showed that transferrable plasmids carried the floR gene in three isolates. Sequencing analysis of a plasmid approximately 125 kb in size (pKP18–125) indicated that the floR gene was flanked by multiple copies of mobile genetic elements. Comparative genomics analysis of a 9-kb transposon-like fragment of pKP18–125 showed that an approximately 2-kb sequence encoding lysR-floR-virD2 was conserved in the majority (79.01%, 83/105) of floR sequences collected from NCBI nucleotide database. Interestingly, the most similar sequence was a 7-kb fragment of plasmid pEC012 from an Escherichia coli strain isolated from a chicken. Conclusions Identified on a transferable plasmid in the human pathogen K. pneumoniae, the floR gene may be disseminated through horizontal gene transfer from animal pathogens. Studies on the molecular mechanism of resistance gene dissemination in different bacterial species of animal origin could provide useful information for preventing or controlling the spread of resistance between animal and human pathogens.

Setting sights on the problem of input-output constraints in most industrial systems, an implicit generalized predictive control algorithm based on an improved particle swarm optimization algorithm (PSO) is presented in this paper. PSO has the advantages of high precision and fast convergence speed in solving constraint problems. In order to effectively avoid the problems of premature and slow operation in the later stage, combined with the idea of the entropy of system (SR), a new weight attenuation strategy and local jump out optimization strategy are introduced into PSO. The velocity update mechanism is cancelled, and the algorithm is adjusted respectively in the iterative process and after falling into local optimization. The improved PSO is used to optimize the performance index in predictive control. The combination of PSO and gradient optimization for rolling-horizon improves the optimization effect of the algorithm. The simulation results show that the system overshoot is reduced by about 7.5% and the settling time is reduced by about 6% compared with the implicit generalized predictive control algorithm based on particle swarm optimization algorithm (PSO-IGPC).

Abstract Distraction osteogenesis (DO) is a unique technique for promoting bone formation in clinical practice. However the underlying mechanism remains elusive. As epigenetic mediators, microRNAs have been reported to play important roles in regulating osteogenesis. In this study, after successfully established the DO model of rats, a microRNA microarray was performed to find molecular targets for DO. Total 100 microRNAs were identified as differently expressed, with miR-503 being one of the most significantly up-regulated miRNAs in DO. The further investigation also showed that miR-503 was upregulated during osteogenesis in mesenchymal stem cells of rats, and overexpression of miR-503 significantly promoted osteogenesis in vitro and accelerated mineralization in DO process in vivo. By using bioinformatic investigations and luciferase activities, we successfully demonstrated that Smurf1, a negative regulator of osteogenesis, was a real target of miR-503. Furthermore, Smurf1 knockdown promoted osteogenesis and antagomir-503 abolished the promotive effect, suggesting that miR-503 mediated osteogenic differentiation via suppressing Smurf1 expression. To sum up, these findings indicated that miR-503 promoted osteogenesis and accelerated bone formation, which may shed light on the development for a potential therapeutic target for bone repair.

ObjectiveSince December 2019, an increasing number of cases of the 2019 novel coronavirus disease (COVID-19) infected by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have been identified in Wuhan, Hubei Province, China. Now, more cases have been reported in 200 other countries and regions. The pandemic disease not only affects physical health who suffered it, but also affects the mental health of the general population. This study aims to know about the impact of the COVID-19 epidemic on the health-related quality of life (HRQOL) of living using EQ-5D in general population in China.MethodsAn online-based survey was developed and participants were recruited via social media. The questionnaires included demographic and socioeconomic data, health status, the condition epidemic situation and EQ-5D scale. The relationships of all factors and the scores of EQ-5D were analyzed. Logistic regression model were used to the five health dimensions.ResultsThe respondents obtained a mean EQ-5D index score of 0.949 and a mean VAS score of 85.52.The most frequently reported problem were pain/discomfort (19.0%) and anxiety/depression (17.6%). Logistic regression models showed that the risk of pain/discomfort and anxiety/depression among people with aging, with chronic disease, lower income, epidemic effects, worry about get COVID-19 raised significantly.ConclusionThe article provides important evidence on HRQOL during the COVID-19 pandemic. The risk of pain/discomfort and anxiety/depression in general population in China raised significantly with aging, with chronic disease, lower income, epidemic effects, worried about get COVID-19 during the COVID-19 pandemic. The results from each categorical data can be used for future healthcare measures among general population.

Control loop performance assessment (CPA) is essential in the operation of industrial systems. In this paper, the shortcomings of existing performance assessment methods and indicators are summarized firstly, and a novel evaluation method based on generalized correntropy criterion (GCC) is proposed to evaluate the performance of non-Gaussian stochastic systems. This criterion could characterize the statistical properties of non-Gaussian random variables more fully, so it can be directly used as the assessment index. When the expected output of the given system is unknown, generalized correntropy is used to describe the similarity of two random variables in the joint space neighborhood controlled and take it as the criterion function of the identification algorithms. To estimate the performance benchmark more quickly and accurately, a hybrid-EDA (H-EDA) combined with the idea of “wading across the stream algorithm” is proposed to obtain the system parameters and disturbance noise PDF. Through the simulation of a single loop feedback control system under different noise disturbances, the effectiveness of the improved algorithm and new indexes are verified.

In the anti-wrinkle finishing of cotton fabrics, the decreased dyeability of the finished fabrics has always been a difficult problem. A new anti-wrinkle finishing mode was developed to solve this problem by changing the finishing sequence of fabric dyeing and anti-wrinkle. In this research, the partial oxidization of raffinose with sodium periodate generated multiple aldehydes, which acted as multifunctional cross-linkers and endowed cotton fabrics with anti-wrinkle and hydrophilic properties. The structural characteristics of oxyRa were analyzed by FTIR and 13C-NMR. Through response surface methodology (RSM), the finishing model of oxyRa was established from the influencing factors of catalyst concentration, pH, curing temperature and time, and the optimized finishing process: the catalyst concentration was 20.12 g/L, pH was 4.32, curing temperature was 150 °C and curing time was 120 s. Under this condition, the predicted wrinkle recovery angle (WRA) of the finished fabric was up to 249.76°, Tensile strength (TS) was 75.62%, Whiteness index (WI) was 70.69. Importantly, comparing the anti-wrinkle and dyeing performance of the fabric with anti-wrinkle and then dyeing and anti-wrinkle after dyeing, the oxyRa-treated fabrics showed better dyeing properties compared with previously reported dimethyldihydroxyethylene urea (DMDHEU), glutaraldehyde (GA), and 1,2,3,4-butanetetracarboxylic acid (BTCA). Analysis of the combined mechanism of different finishing agents and cellulose, demonstrated the reason why oxyRa can be used to change the order of dyeing and anti-wrinkle finishing.
