1. Baseline patient demographics for TETRIS: a prospective, noninterventional study to characterize the use of triple therapy for COPD in Germany

Abstract: Background: Evidence on how decisions regarding escalation to triple therapy and de- or re-escalation are taken and the rationale on which these decisions are based is currently limited in Germany. Objectives: The TETRIS study aims to elucidate influences on treatment decisions surrounding triple therapy in a real-world practice setting in Germany. Design: TETRIS is an ongoing, multicenter, prospective, observational cohort study recruiting patients with chronic obstructive pulmonary disease (COPD) with or without asthma who have already been treated with triple therapy for 2–48 weeks. Methods: For better representation of the treatment reality in Germany, patients are recruited from general practitioners and pulmonologists. Data are collected in two parts. Part 1 involves cross-sectional phenotyping of patients at enrollment. Part 2 involves a 2-year longitudinal follow-up period to monitor/document all visits by the patients during the 24-month observation period per routine clinical practice. Here, we report the demographic and baseline characteristics of 1213 eligible patients recruited to part 1 of the study. Results: The mean patient age was 66.4 years overall, and 29.3% (356/1213) of patients had no comorbidities. The mean CAT score was 19.4; the number of exacerbations and hospitalizations due to exacerbations in the past 3 years before starting triple therapy was 0.6 and 0.1, respectively. Dual bronchodilation with a long-acting muscarinic antagonist (LAMA) plus a long-acting β-2 agonist (LABA) was the most common therapy for COPD before initiation of triple therapy in 58.3% of patients. Conclusion: In this real-world setting in Germany, patients with COPD have a relatively low reported exacerbation rate but high symptom burden, and over 70% are multimorbid. Triple therapy is initiated in patients who are primarily highly symptomatic despite being on LAMA + LABA. Future prospective studies in patients with multimorbidity are warranted to better understand the treatment landscape across the disease spectrum. Abstract: The detection of small size air showers at high altitude allows the search for γ-ray point sources at few hundreds GeV energy threshold. The ARGO-YBJ experiment, currently under construction at the Yangbajing Cosmic Ray Laboratory (4300 m a.s.l.), has been designed to meet these requirements by exploiting the RPC technology and a dedicated DAQ system based on a custom high-speed read-out architecture. About 1600 m2 of detector have been instrumented and put into operation for calibration runs devoted to test the performance of the individual components and their integrated operation. Events collected with a ’shower mode’ trigger have been recorded to check the consistency of physics data. We present the current status of the experiment and report on future prospects.

Abstract: The ARGO-YBJ experiment has been designed to detect air shower events over a large size scale and with an energy threshold of a few hundreds GeV. The building blocks of the ARGO-YBJ detector are single-gap Resistive Plate Counters (RPCs). The trigger logic selects the events on the basis of their hit multiplicity. Inclusive triggers as well as dedicated triggers for specific physics channels or calibration purposes have been developed. This paper describes the architecture and the main features of the trigger system.

Abstract: A resistive plate counters (RPCs) carpet of similar to50 m(2) has been put in operation in the Yangbajing Laboratory (Tibet, P.R. China) at 4300 in a.s.l., in order to study the RPCs performance at high altitude and the detector capability of imaging the EAS disc. This test has been performed in view of an enlarged use of RPCs for the ARGO-YBJ experiment. This experiment will be devoted to a wide range of fundamental issues in cosmic rays and astroparticle physics, including in particular gamma-ray astronomy and gamma-ray bursts physics at energies greater than or equal to 100 GeV. In this paper we present and discuss the procedures adopted to calibrate the detector and reconstruct the shower direction. Results concerning many shower features as the angular distribution, the density spectrum, the time profile of the shower front, are found well consistent with the expectation. Abstract: The characteristics of a 50m(2) RPC carpet(prototype of YBJ-ARGO experiment)was analyzed using its test run data. A correction method of the systematic time error is suggested, and nonuniform azimuthal angle distribution possibly due to direction reconstruction error on an asymmetric carpet is reported.

Abstract: Context. Primary cosmic rays experience multiple deflections in the non-uniform galactic and heliospheric magnetic fields which may generate anisotropies. Aims. A study of anisotropies in the energy range between 100 and 500 GeV is performed. This energy range is not yet well explored.Methods. The L3 detector at the CERN electron-positron collider, LEP, is used for a study of the angular distribution of atmospheric muons with energies above 20 GeV. This distribution is used to investigate the isotropy of the time-dependent intensity of the primary cosmic-ray flux with a Fourier analysis.Results. A small deviation from isotropy at energies around 200 GeV is observed for the second harmonics at the solar frequency. No sidereal anisotropy is found at a level above . Abstract: Aims. Several experiments have reported observations on possible correlations between the flux of high energy muons and intense solar flares. If confirmed, these observations would have significant implications for acceleration processes in the heliosphere able to accelerate protons and other ions to energies of at least tens of GeV. Methods. The solar flare of the 14 of July 2000 offered a unique opportunity for the L3+C experiment to search for a correlated enhancement in the flux of muons using the L3 precision muon spectrometer. Its capabilities for observing a directional excess in the flux of muons above 15 GeV (corresponding to primary proton energies above 40 GeV) are presented along with observations made on the 14th of July 2000. Results. We report an excess which appeared at a time coincident with the peak increase of solar protons observed at lower energies. The probability that the excess is a background fluctuation is estimated to be 1 %. No similar excess of the muon flux was observed up to 1.5 h after the solar flare time. © ESO 2006.

Abstract: Bakelite RPCs, assembled according to the ARGO design, have been operated in the high altitude Laboratory of YBJ using dedicated electronics to pick-up the streamer signal. Here we report on the results concerning absorbed current, single counting rate, efficiency and time resolution. Abstract: A resistive plate counters (RPCs) carpet of ∼50 m 2 has been put in operation in the Yangbajing Laboratory (Tibet, P.R. China) at 4300 m a.s.l., in order to study the RPCs performance at high altitude and the detector capability of imaging the EAS disc. This test has been performed in view of an enlarged use of RPCs for the ARGO-YBJ experiment. This experiment will be devoted to a wide range of fundamental issues in cosmic rays and astroparticle physics, including in particular γ-ray astronomy and γ-ray bursts physics at energies ⩾100 GeV. In this paper we present and discuss the procedures adopted to calibrate the detector and reconstruct the shower direction. Results concerning many shower features as the angular distribution, the density spectrum, the time profile of the shower front, are found well consistent with the expectation.

15. Avian malaria parasite infections do not affect personality in the chestnut thrush ( Turdus rubrocanus ) on the Qinghai-Tibet Plateau.

Abstract: Personality traits, the consistent individual behavioral differences, are currently gaining much attention in studies of natural bird populations. However, associations between personality traits and parasite infections are not often investigated. Even less attention has been given to studies of birds in the high-elevation region such as the Tibetan plateau. This research aims to examine the relationship between avian malaria parasites and two personality traits in a population of the Chestnut Thrush ( Turdus rubrocanus ) breed in the Tibetan plateau. Our results revealed no evidence of sex bias in malaria parasite prevalence. Furthermore, we found no effect of infection status on two personality scores: activity and boldness. Additionally, no effects on the activity level or boldness were observed for different parasite lineages of Haemoproteus , Leucocytozoon , the sex of the birds, or their interactions. Similarly, we did not find any relationship between activity level and boldness with nestling numbers, sex, or their interactions. Notably, individuals with a larger number of offspring tended to display greater boldness. 16. A novel risk prediction model of pyroptosis-related genes for the prognosis and immunotherapy response of endometrial cancer.

Abstract: Objective: This study aims to develop a risk prediction model of pyroptosis-related genes based on its impact on immunotherapy sensitivity of uterine corpus endometrial carcinoma (UCEC), one of the most common and threatening gynecological malignancies., Patients and Methods: Through multiple bioinformatics analysis, we obtained raw counts of RNA-sequencing data and corresponding clinical information related to UCEC from The Cancer Genome Atlas (TCGA) and Gene Expression Profiling Interactive Analysis (GEPIA) to investigate the potential mechanisms of differentially expressed pyroptosis-related genes (DEPRGs), including the correlation between DEPRGs and prognosis, tumor immune microenvironment and the immunotherapy sensitivity of UCEC patients. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) Enrichment Analysis were used to figure out the functional differences. Furthermore, a mRNA-miRNA-lncRNA network was constructed to identify potential impact of pyroptosis on tumor progression., Results: In this study, we achieved six DEPRGs (CASP3, GPX4, GSDMD, NOD2, PYCARD and TIRAP) and constructed a 6-gene signature which classified UCEC patients in the TCGA cohort into a low-risk group or a high-risk group. Patients in the low-risk group showed significantly longer survival time (p=0.000373). The risk score was also confirmed as an independent prognostic factor combining with the clinical characteristics. GO and KEGG functional analysis revealed the possible molecular mechanisms by which six DEPRGs influence anti-tumor immunity in UCEC patients. In addition, we found that two DEPRGs (GPX4, TIRAP) were not only significantly associated with tumor mutational burden (TMB) or microsatellite Instability (MSI), but also involved in regulating the number and function of CD8+ cells., Conclusions: Upon comprehensive bioinformatics analysis, it was concluded that pyroptosis-related genes (PRGs) could predict the prognosis of EC patients and be affected in modulating the anti-tumor immune responses for patients with EC.

Abstract: Flos Chrysanthemi indici , an important medicinal and aromatic plant in China, is considered to have many different preservative and pharmacological properties. Considering the capability of essential oils (EOs), the present study is conducted to compare different extraction methods in order to improve yield and biological activities. Hydro-distillation (HD), steam-distillation (SD), solvent-free microwave extraction (SFME), and supercritical fluid extraction (SFE) are employed to prepare EOs from Flos Chrysanthemi indici . A total of 71 compounds are assigned by gas chromatography/mass spectrometry (GC-MS) in comparison with retention indices. These include 32 (HD), 16 (SD), 31 (SFME) and 38 (SFE) compounds. Major constituents of EOs differ according to the extraction methods were heptenol, tricosane, camphor, borneol, and eucalyptol. EOs extracted by SFME exhibit higher antioxidant activity. All EOs show varying degrees of antimicrobial activity, with minimum inhibitory concentration (MIC) ranging from 0.0625 to 0.125 mg/mL and SFME and SFE prove to be efficient extraction methods. EOs alter the hyphal morphology of Alternaria alternata , with visible bumps forming on the mycelium. Overall, these results indicate that the extraction method can significantly influence the composition and biological activity of EOs and SFME and SFE are outstanding methods to extract EOs with high yield and antimicrobial activity.

18. Optimization of Ultrasonic-Assisted Extraction of Flavonoid Compounds and Antioxidants from Alfalfa Using Response Surface Method.

Abstract: Ultrasonic-assisted extraction (UAE) was used to extract flavonoid-enriched antioxidants from alfalfa aerial part. Response surface methodology (RSM), based on a four-factor, five-level central composite design (CCD), was employed to obtain the optimal extraction parameters, in which the flavonoid content was maximum and the antioxidant activity of the extracts was strongest. Radical scavenging capacity of the extracts, which represents the amounts of antioxidants in alfalfa, was determined by using 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) methods. The results showed good fit with the proposed models for the total flavonoid extraction (R² = 0.9849), for the antioxidant extraction assayed by ABTS method (R² = 0.9764), and by DPPH method (R² = 0.9806). Optimized extraction conditions for total flavonoids was a ratio of liquid to solid of 57.16 mL/g, 62.33 °C, 57.08 min, and 52.14% ethanol. The optimal extraction parameters of extracts for the highest antioxidant activity by DPPH method was a ratio of liquid to solid 60.3 mL/g, 54.56 °C, 45.59 min, and 46.67% ethanol, and by ABTS assay was a ratio of liquid to solid 47.29 mL/g, 63.73 °C, 51.62 min, and 60% ethanol concentration. Our work offers optimal extraction conditions for total flavonoids and antioxidants from alfalfa.

Abstract: The aim of this study was to compare the effect of DL-selenomethionine (SM) with 2 routinely used Se sources, sodium selenite (SS) and seleno-yeast (SY), on relative bioavailability based on antioxidant activity and tissue Se content. Six hundred thirty 131-day-old brown laying hens were randomly assigned to 7 treatments for 168 d (24 wks) with 6 replicates of 15 hens per replicate. The SS and SY animals were supplemented a cornmeal and soybean diet that supplied a total Se 0.3 mg/kg whereas SM was added at 4 different levels to the total Se at 0.1, 0.3, 0.5 and 0.7 mg/kg. All hens fed the Se-supplemented diet showed higher glutathione peroxidase (GSH-Px) activity (P < 0.01), higher superoxide dismutase (SOD) activity (P < 0.05), lower malondialdehyde (MDA) content (P < 0.05) in plasma, and greater Se contents in egg yolks, albumen, leg muscle, breast muscle, liver, and plasma compared with those fed the control diet (P < 0.01). The organic sources (SY and SM) exhibited a greater ability to increase the GSH-Px activity (P < 0.01) and Se content in albumen (P < 0.01), leg, and breast muscles (P = 0.0099 and P = 0.0014, respectively) than the SS that was added at 0.3 mg Se/kg. The higher SM added levels increased the GSH-Px activity until the dose of 0.5mg Se/kg (P < 0.01).The greater Se concentrations in albumen, muscle and liver appeared in the higher SM-added level, as well as above the dose of 0.1 mg Se/kg (P < 0.01). In addition, hens fed the diet with SM accumulated more Se in albumen, leg, and breast muscle than those fed diets with SY (P < 0.05). These results confirmed the higher ability of organic Se sources to increase the antioxidant activity and Se deposition in egg albumen, leg, and breast muscles compared with SS, and demonstrated a significantly better efficiency of SM compared with SY for albumen and muscle Se enrichment., (© 2015 Poultry Science Association Inc.)

Abstract: Objective: To determine the interference of antithyroglobulin (TgAb) on the measurement of serum thyroglobulin (Tg) in patients with differentiated thyroid carcinomas (DTC)., Methods: The Tg positive serum and TgAb positive serum were taken from the patients with total thyroidectomy and thyroid remnant ablation by radioiodine. The Tg positive serum contained high level of Tg (>10 microg/L) and nondetectable TgAb. The TgAb positive serum contained high level of TgAb (>115 IU/mL) and nondetectable Tg. We incubated a constant amount of Tg with increasing volumes of Tg-free autoantibodies and allowed them to equilibrate at room temperature for 12 hours before measuring Tg and TgAb. The thyroglobulin and TgAb values were determined by the Roche electrode induce chemiluminescent emission system with monoclonal antibody sandwich analysis. The correlation between TgAb and serum Tg values was analysed., Results: TgAb led to underestimation of serum Tg values by 15%-50% in 8 patients with DTC. TgAb was correlated with the magnitude of underestimated serum Tg values in 3 patients with DTC, but not in the other 5 patients with DTC., Conclusion: TgAb underestimated serum Tg values in a dose-dependent manner in some patients with DTC, but not in others.

Abstract: Objective: To study the relationship between the DNA content in follicular epithelial cells of the human thyroid and postmortem interval (PMI)., Methods: Changes of the DNA content in thyroid follicular epithelial cells at different PMI were determined by Methyl Green-Pyronin (MGP) stain combined with an image analysis technique., Results: The average DNA content in the thyroid follicular epithelial continued to decrease with increased PMI. The coefficient of determination for DNA content with average gray, aimed area, aimed area ratio and positive index in follicular epithelial cells were 0.960, 0.987, 0.988 and 0.990, respectively., Conclusion: There is an apparent correlation between the average DNA content of follicular epithelial cells and the PMI. MGP stain combined with an image analysis technique seems to be a useful means to study the degradation of DNA in thyroid follicular epithelial cells.
