Abstract Rangelands, covering half of the global land area, are critically degraded by unsustainable use and climate change. Despite their extensive presence, global assessments of rangeland condition and sustainability are limited. Here we introduce a novel analytical approach that combines satellite big data and statistical modeling to quantify the likelihood of changes in rangeland conditions. These probabilities are then used to assess the effectiveness of management interventions targeting rangeland sustainability. This approach holds global potential, as demonstrated in Mongolia, where the shift to a capitalist economy has led to increased livestock numbers and grazing intensity. From 1986 to 2020, heavy grazing caused a marked decline in Mongolia’s rangeland condition. Our evaluation of diverse management strategies, corroborated by local ground observations, further substantiates our approach. Leveraging globally available yet locally detailed satellite data, our proposed condition tracking approach provides a rapid, cost-effective tool for sustainable rangeland management.

Abstract Parental or ancestral environments can induce heritable phenotypic changes, but whether such environment-induced heritable changes are a common phenomenon remains unexplored. Here, we subject 14 genotypes of Arabidopsis thaliana to 10 different environmental treatments and observe phenotypic and genome-wide gene expression changes over four successive generations. We find that all treatments caused heritable phenotypic and gene expression changes, with a substantial proportion stably transmitted over all observed generations. Intriguingly, the susceptibility of a genotype to environmental inductions could be predicted based on the transposon abundance in the genome. Our study thus challenges the classic view that the environment only participates in the selection of heritable variation and suggests that the environment can play a significant role in generating of heritable variations.

Abstract Objective We attempted to identify abnormal immune cell components and signaling pathways in lupus nephritis (LN) and to identify potential therapeutic targets. Methods Differentially expressed genes (DEGs) between LN and normal kidney tissues were identified from bulk transcriptome data, and functional annotation was performed. The phenotypic changes in macrophages and aberrant intercellular signaling communications within immune cells were imputed from LN scRNA-seq data using trajectory analysis and verified using immunofluorescence staining. Finally, lentivirus-mediated overexpression of LGALS9, the gene encoding Galectin 9, in THP-1 cells was used to study the functional effect of this gene on monocytic cells. Results From bulk transcriptome data, a significant activation of interferon (IFN) signaling was observed, and its intensity showed a significantly positive correlation with the abundance of infiltrating macrophages in LN. Analysis of scRNA-seq data revealed 17 immune cell clusters, with macrophages showing the highest enrichment of intercellular signal communication in LN. Trajectory analysis revealed macrophages in LN undergo a phenotypic change from inflammatory patrolling macrophages to phagocytic and then to antigen-presenting macrophages, and secrete various pro-inflammatory factors and complement components. LGALS9 was found significantly upregulated in macrophages in LN, which was confirmed by the immunofluorescence assay. Gene functional study showed that LGALS9 overexpression in THP-1 cells significantly elicited pro-inflammatory activation, releasing multiple immune cell chemoattractants. Conclusion Our results present an important pathophysiological role for macrophages in LN, and our preliminary results demonstrate significant pro-inflammatory effects of LGALS9 gene in LN macrophages.

The Gross Domestic Product (GDP) per capita is one of the most widely used socioeconomic indicators, serving as an integral component for climate change impact analysis. However, a national scale assessment may induce considerable bias because it conceals any internal variations within a country. The lack of a long-term sub-national scale GDP data is a substantive hinderance. Leveraging the close relationship between nighttime lights and GDP, we address this gap by developing a novel methodological framework in two steps. First, under the modeling philosophy of spatial statistics, we developed a novel approach based on deep and machine learning techniques to establish a complex mapping between two inconsistent nighttime lights (NTL) datasets: the Defense Meteorological Satellite Program’s Operational Linescan System (DMSP) and the National Polar-Orbiting Partnership’s Visible Infrared Imaging Radiometer Suite (VIIRS). The models achieve accuracies ranging from 0.945 to 0.980 (correlation coefficients). By taking the estimations ensemble of the two techniques, the time series of DMSP data was extended to 2021. Next, a novel modeling strategy based on multi-layer perceptron was developed to derive the non-linear relationship between NTL and GDP per capita at sub-national scale to alleviate scale effects at this granularity, while explicitly capturing regional heterogeneity effect. The trained models achieve average accuracies of 0.967, 0.959, and 0.959 on the training, validation, and test sets, respectively. We evaluate the developed dataset at the global, national, and sub-national scales from various perspective, and the results offer solid evidence on the reliability of the estimated economic data. By linking to historical global climate change data, we quantify global economic losses attributed to extreme heat to demonstrate how the estimated GDP data can be useful in the climate change impact analysis.

As a pathological hallmark of type 2 diabetes mellitus (T2DM), islet amyloid is formed by the aggregation of islet amyloid polypeptide (IAPP). Endoplasmic reticulum (ER) stress interacts with IAPP aggregates and has been implicated in the pathogenesis of T2DM. To examine the role of ER stress in T2DM, we cloned the hIAPP promoter and analyzed its promoter activity in human β-cells. We found that ER stress significantly enhanced hIAPP promoter activity and expression in human β-cells via triggering X-box binding protein 1 (XBP1) splicing. We identified a binding site of XBP1 in the hIAPP promoter. Disruption of this binding site by substitution or deletion mutagenesis significantly diminished the effects of ER stress on hIAPP promoter activity. Blockade of XBP splicing by MKC3946 treatment inhibited ER stress-induced hIAPP up-regulation and improved human β-cell survival and function. Our study uncovers a link between ER stress and IAPP at the transcriptional level and may provide novel insights into the role of ER stress in IAPP cytotoxicity and the pathogenesis of T2DM.

A thorough comprehension of the trade-offs/synergies (TOS) among ecosystem services (ESs) and their underlying drivers are vital for promoting regional sustainable development and making informed decisions. In this study, we explored the spatial and temporal dynamics of six ESs and their driving factors at both the grid and county scales in the Central-southern Liaoning urban agglomeration (CSLUA) utilizing various analytical techniques such as Pearson correlation analysis, geographically weighted regression, self-organized mapping analysis, and Redundancy Analysis. Our results revealed that: (1) Between 2000 and 2020, water yield (WY), soil retention (SR), landscape aesthetics (LA), and food production (FP) exhibited an overall upward trend, while habitat quality (HQ) and carbon storage (CS) demonstrated a decreasing trend; (2) From grid scale to county scale, we found concentration effects of TOS relationships was weaken, mainly between supply services (WY, FP) and other services. The significant relationship between some ES pairs has become neutral. The geospatial heterogeneity of TOS relationships for all ESs was also weakened at county scale. More details of geographical features can be captured at grid scale than at county scale. (3) At both scales of analysis, we identified five ES bundles with different ecological functional characteristics and spatial patterns. We found that the Potential High-value Synergistic Bundle would shift to High-value Synergistic Bundle. Although the areas of the LA bundle increased from the grid scale to the county scale, there is less continuity. (4) The direction of the influence of the driving factor on ES is not constant in time, and the degree of the drivers’ impacts on the six ESs varies on the two scales. But NDVI was the main driver for most of the ESs. This study contributes to expanding our understanding of the relationships between ESs at different scales and could inform sustainable ecosystem management at multiple scales.

Abstract Artificial intelligence has gained significant attention for exploiting optical scattering for optical encryption. Conventional scattering media are inevitably influenced by instability or perturbations, and hence unsuitable for long-term scenarios. Additionally, the plaintext can be easily compromised due to the single channel within the medium and one-to-one mapping between input and output. To mitigate these issues, a stable spin-multiplexing disordered metasurface (DM) with numerous polarized transmission channels serves as the scattering medium, and a double-secure procedure with superposition of plaintext and security key achieves two-to-one mapping between input and output. In attack analysis, when the ciphertext, security key, and incident polarization are all correct, the plaintext can be decrypted. This system demonstrates excellent decryption efficiency over extended periods in noisy environments. The DM, functioning as an ultra-stable and active speckle generator, coupled with the double-secure approach, creates a highly secure speckle-based cryptosystem with immense potentials for practical applications.

Abstract Inflammasomes are large protein complexes that play a major role in sensing inflammatory signals and triggering the innate immune response. Each inflammasome complex has three major components: an upstream sensor molecule that is connected to a downstream effector protein such as caspase-1 through the adapter protein ASC. Inflammasome formation typically occurs in response to infectious agents or cellular damage. The active inflammasome then triggers caspase-1 activation, followed by the secretion of pro-inflammatory cytokines and pyroptotic cell death. Aberrant inflammasome activation and activity contribute to the development of diabetes, cancer, and several cardiovascular and neurodegenerative disorders. As a result, recent research has increasingly focused on investigating the mechanisms that regulate inflammasome assembly and activation, as well as the potential of targeting inflammasomes to treat various diseases. Multiple clinical trials are currently underway to evaluate the therapeutic potential of several distinct inflammasome-targeting therapies. Therefore, understanding how different inflammasomes contribute to disease pathology may have significant implications for developing novel therapeutic strategies. In this article, we provide a summary of the biological and pathological roles of inflammasomes in health and disease. We also highlight key evidence that suggests targeting inflammasomes could be a novel strategy for developing new disease-modifying therapies that may be effective in several conditions.

Given the ever-growing availability of remote sensing data (e.g., Gaofen in China, Sentinel in the EU, and Landsat in the USA), multimodal remote sensing techniques have been garnering increasing attention and have made extraordinary progress in various Earth observation (EO)-related tasks. The data acquired by different platforms can provide diverse and complementary information. The joint exploitation of multimodal remote sensing has been proven effective in improving the existing methods of land-use/land-cover segmentation in urban environments. To boost technical breakthroughs and accelerate the development of EO applications across cities and regions, one important task is to build novel cross-city semantic segmentation models based on modern artificial intelligence technologies and emerging multimodal remote sensing data. This leads to the development of better semantic segmentation models with high transferability among different cities and regions. The Cross-City Semantic Segmentation contest is organized in conjunction with the 13th Workshop on Hyperspectral Image and Signal Processing: Evolution in Remote Sensing (WHISPERS).

Introduction: Primary pleural epithelial angiosarcoma (EAS) is an extremely rare tumor with no specific clinical symptoms. Clinical data on primary pleural EAS are limited, and misdiagnosis often occurs. Case Presentation: The present study reports the case of a 31-year-old patient diagnosed with primary pleural EAS with lung and bone metastases. The patient presented with persistent right chest pain for 5 months and dyspnea for 2 months. Chest computed tomography (CT) scan revealed right hydropneumothorax, diffuse thickening of the right pleura, passive atelectasis, and scattered nodules in the left lung. A medical thoracoscopic pleural biopsy revealed a vasogenic tumor. To further confirm the diagnosis, positron emission tomography/CT (PET/CT) examination was recommended to determine the biopsy site after multidisciplinary discussion. Increased 18F-FDG uptake in the right pleura and hypermetabolic nodules in the right chest wall, first lumbar vertebrae, second sacral vertebrae, and bilateral iliac crest was detected via PET/CT. CT-guided chest wall and lung biopsies were performed. Immunohistochemistry of specific markers was performed according to remote consultation with a pathologist, and tumor cells with strong positive expression of CD31, CD34, and ETS-related genes led to the final diagnosis of primary pleural EAS. Conclusion: Primary pleural EAS should be considered for hydropneumothorax of an unknown cause. PET/CT can accurately locate the lesion. The pathological examination is the basis for primary pleural EAS diagnosis. Moreover, multidisciplinary discussion and remote expert consultation can improve the diagnosis rate of primary pleural EAS.

The imperative to address traditional energy crises and environmental concerns has accelerated the need for energy structure transformation. However, the variable nature of renewable energy poses challenges in meeting complex practical energy requirements. To address this issue, the construction of a multifunctional large-scale stationary energy storage system is considered an effective solution. This paper critically examines the battery and hydrogen hybrid energy storage systems. Both technologies face limitations hindering them from fully meeting future energy storage needs, such as large storage capacity in limited space, frequent storage with rapid response, and continuous storage without loss. Batteries, with their rapid response (90 ​%), excel in frequent short-duration energy storage. However, limitations such as a self-discharge rate (>1 ​%) and capacity loss (∼20 ​%) restrict their use for long-duration energy storage. Hydrogen, as a potential energy carrier, is suitable for large-scale, long-duration energy storage due to its high energy density, steady state, and low loss. Nevertheless, it is less efficient for frequent energy storage due to its low storage efficiency (∼50 ​%). Ongoing research suggests that a battery and hydrogen hybrid energy storage system could combine the strengths of both technologies to meet the growing demand for large-scale, long-duration energy storage. To assess their applied potentials, this paper provides a detailed analysis of the research status of both energy storage technologies using proposed key performance indices. Additionally, application-oriented future directions and challenges of the battery and hydrogen hybrid energy storage system are outlined from multiple perspectives, offering guidance for the development of advanced energy storage systems.

Abstract Background To evaluate outcomes of panretinal photocoagulation (PRP) plus intravitreal conbercept (IVC) for diabetic retinopathy (DR) in real world and explore risk factors for patients with poor reactivity and presence of vision-threatening complications after combination treatment. Methods Retrospective review of DR patients received PRP plus IVC over 6 months. The main outcome was improvement ≥ 2 steps in ETDRS diabetic retinopathy severity scale (DRSS) levels. To determine the paclitaxel-related substances in a paclitaxel injection (albumin-bound), it is necessary to remove human blood albumin from the sample via solid-phase extraction and subsequently perform high-performance liquid chromatography analysis. The pre-processing operation is complicated, the duration of analysis is long, and much human power and material resources are needed in the sample testing process. Thus, the purpose of this study was to develop a quick two-dimensional liquid chromatography method that requires less consumables and possesses high sensitivity. Samples were directly injected into one-dimensional solid-phase extraction columns to remove human blood albumin. Paclitaxel and its related substances in the solid-phase extraction columns were then eluted into two-dimensional analytical columns through valve switching technology. The sample pretreatment step can be omitted in this method, which allows for the analysis of paclitaxel and its related substances to be completed more quickly, within only 15 min, under the developed two-dimensional elution conditions. The established method had good linearity (R2 = 0.9994), limit of detection (0.25 μg·mL−1), limit of quantification (0.5 μg·mL−1), precision (RSD = 0.5%), and accuracy (102.53–107.97%).

Type 2 diabetes mellitus (T2DM) is a common metabolic disease that adversely impacts patient health. In this study, a T2DM model was established in ICR mice through the administration of a high-sugar and high-fat diet combined with the intraperitoneal injection of streptozotocin to explore the hypoglycemic effect of polysaccharides from Physalis alkekengi L. After six weeks of treatment, the mice in the high-dosage group (800 mg/kg bw) displayed significant improvements in terms of fasting blood glucose concentration, glucose tolerance, serum insulin level, insulin resistance, and weight loss (p < 0.05). The polysaccharides also significantly regulated blood lipid levels by reducing the serum contents of total triglycerides, total cholesterol, and low-density lipoproteins and increasing the serum content of high-density lipoproteins (p < 0.05). Furthermore, they significantly enhanced the hepatic and pancreatic antioxidant capacities, as determined by measuring the catalase and superoxide dismutase activities and the total antioxidant capacity (p < 0.05). The results of immunohistochemistry showed that the P. alkekengi polysaccharides can increase the expression of GPR43 in mice colon epithelial cells, thereby promoting the secretion of glucagon-like peptide-1. In summary, P. alkekengi polysaccharides can help to regulate blood glucose levels in T2DM mice and alleviate the decline in the antioxidant capacities of the liver and pancreas, thus protecting these organs from damage.

We present a unique case of left atrial (LA) dissection in a 46-year-old man following aortic dissection surgery. The LA dissection was attributed to coronary sinus catheter–related injury. This report highlights the importance of recognizing this rare complication and the crucial role of transesophageal echocardiography in its diagnosis. We discuss the pathogenesis, diagnostic criteria, and management strategies for LA dissection.

Zinc oxide (ZnO) serves as a photocatalyst, but it faces challenges due to its high band gap energy, limiting photoactivation to ultraviolet irradiation and rendering it scarcely recoverable after degradation. This work focuses on enhancing the photocatalytic efficiency of ZnO through cobalt doping and immobilization onto magnetic iron oxide (Fe3O4) via co-precipitation. The concentration of the cobalt precursor (0–10 mol %) and the molar ratio between Co–ZnO and Fe3O4 (5:1–15:1) were varied to achieve the optimal degradation efficiency and magnetic properties. Characterization tools such as the scanning electron microscope (SEM), energy dispersive X-ray analysis (EDX), attenuated total reflectance – Fourier-transform infrared spectroscopy (ATR-FTIR), and X-ray diffraction analysis (XRD) were utilized to evaluate the effect of cobalt doping on the physicochemical properties of the developed photocatalyst. Tauc plots indicated that 5 mol % Co–ZnO (Co5–ZnO) had the smallest band gap energy (2.79 eV), resulting in the highest photodegradation of methylene blue (88.72 %), compared to pristine ZnO (74.88 %) with a band gap energy of 3.39 eV. Co–ZnO/Fe3O4 showed a slight degradation efficiency decline, reaching a minimum of 84.53 % for the 5:1 molar ratio. These photocatalysts exhibited magnetic properties, with recovery efficiency ranging between 69.8 % and 72 %. Active radicals trapping experiments confirmed the involvement of holes, hydroxyl, and superoxide radicals in the degradation process, with superoxide radicals playing the most significant roles. In summary, this study successfully demonstrated that the synthesized Co–ZnO photocatalysts gained visible light sensitivity, and the incorporation of Fe3O4 imparted magnetic separability to the composite, thereby facilitating photocatalyst recovery.

Abstract Bacteroides fragilis, one of the potential next-generation probiotics, but its protective mechanism is not yet known. We aimed to characterize the anti-inflammatory effect of B. fragilisATCC25285 and to elucidate its mechanism through in vivo and in vitro experiments. An in vitro model of inflammation by induction of colonic cells with TNF-a, and co-cultured with B. fragilis to detect cell viability, apoptosis and invasive capacity. Furthermore, critical proteins of the TLR/NF-κB pathway and the inflammatory cytokines were measured. For animal trials, C57BL/6 J male mice were orally administered B. fragilis or PBS once daily for 21 days. Colitis was induced by drinking 2.5% DSS from days 0 to 7. The mice were weighed daily and rectal bleeding, stool condition and blood in the stool were recorded. We found that B. fragilis treatment alone was harmless and had no effect on cell viability or apoptosis. While predictably TNF-α decreased cell viability and increased apoptosis, B. fragilis attenuated this deterioration. The NF-κB pathway and inflammatory cytokines IL-6 and IL-1β activated by TNF-α were also blocked by B. fragilis. Notably, the metabolic supernatant of B. fragilis also has an anti-inflammatory effect. Animal studies showed that live B. fragilis rather than dead strain ameliorated DSS-induced colitis, as evidenced by weight loss, shortened colon length and enhanced barrier function. The colonic tissue levels of inflammatory cytokines (TNF-α, IL-1β, IL-6) were decreased and IL-10 was increased as a result of B. fragilis administration. In conclusion, B. fragilis ATCC25285 exhibited anti-inflammatory effects whether in vivo or in vitro, and it may be a potential probiotic agent for improving colitis.

Abstract Advances in sequencing technologies have empowered epitranscriptomic profiling at the single-base resolution. Putative RNA modification sites identified from a single high-throughput experiment may contain one type of modification deposited by different writers or different types of modifications, along with false positive results because of the challenge of distinguishing signals from noise. However, current tools are insufficient for subtyping, visualization, and denoising these signals. Here, we present iMVP, which is an interactive framework for epitranscriptomic analysis with a nonlinear dimension reduction technique and density-based partition. As exemplified by the analysis of mRNA m5C and ModTect variant data, we show that iMVP allows the identification of previously unknown RNA modification motifs and writers and the discovery of false positives that are undetectable by traditional methods. Using putative m6A/m6Am sites called from 8 profiling approaches, we illustrate that iMVP enables comprehensive comparison of different approaches and advances our understanding of the difference and pattern of true positives and artifacts in these methods. Finally, we demonstrate the ability of iMVP to analyze an extremely large human A-to-I editing dataset that was previously unmanageable. Our work provides a general framework for the visualization and interpretation of epitranscriptomic data.

Objective: A method for simultaneous determination of 17 β-receptor agonists in meat was established by QuEChERS EMR-Lipid with isotope dilution-ultra performance liquid chromatography-tandem mass spectrometry. Methods: The samples were added to ammonium acetate buffer solution (pH5.2) and enzymatic hydrolyzed by β-glucuronidase and arysulfatase. The hydrolysates were extracted by acetonitrile containing 5% formic acid, and purified by QuEChERS EMR-Lipid. The analyses were separated by CORTECS™ UPLC® C18 chromatographic column with gradient elution using 0.1% formic acid and methanol as the mobile phase. The target compounds were monitored in scheduled MRM mode. Then the internal standard method was used for quantitative analysis. Results: The linear regression correlation coefficients for the 17 β-receptor agonists were all higher than 0.99 in the range from 0 ng/mL to 20 ng/mL. The limits of detection were 0.01~0.09 μg/kg and the limits of quantification were 0.05~0.31 μg/kg. The average recoveries at three spiked levels of 0.5, 2.0 and 5.0 μg/kg ranged from 81.7% to 111.8% with the relative standard deviations of 0.8%~10.3% (n=6). No β-receptor agonists were detected in 100 batches of fresh meat. Conclusion: The method has the advantages of simple pretreatment steps and good purification effect. It shortened enzymolysis time, improved sample detection efficiency. The internal standard method was accurate and reliable. It was suitable for simultaneous determination of β-receptor agonists in a large number of meat samples.

Abstract Aims/Introduction The triglyceride‐glucose (TyG) index is a simple and reliable indicator of insulin resistance, and is associated with the development and poor outcomes of cardiovascular disease. Subclinical left ventricular dysfunction (SLVD) is frequently detected in approximately one‐third of diabetes patients, but it has not been established whether the TyG index correlates with SLVD. We carried out this research to evaluate the relationship between the TyG index and SLVD in type 2 diabetes mellitus patients. Materials and Methods This was a cross‐sectional and observational study of 183 type 2 diabetes mellitus inpatients at Nanjing Drum Tower Hospital, Nanjing, China. The TyG index and homeostasis model assessment 2 estimates for insulin resistance (HOMA2‐IR) were calculated from biochemical measurements, and speckle‐tracking echocardiography was carried out. Abstract Growing evidence indicates that the epithelial to mesenchymal (E/M) hybrid state plays a key role in tumorigenesis. Importantly, a hybrid mesenchymal to epithelial transition (MET) state in which individual cells express both epithelial and mesenchymal markers was recently identified in vivo, further strengthening the bonds between the hybrid EMT state and cancer progression. However, the role and the molecular mechanisms by which the hybrid MET state is maintained in triple-negative breast cancer cells (TNBC) remain elusive. Here, we find that loss of ZHX2 expression results in the hybrid MET phenotype in mesenchymal TNBC cells. Mechanistically, through directly binding to the CDH1 promoter, depletion of ZHX2 specifically reactivates expression of CDH1 encoding E-cadherin, an epithelial marker that is crucial for maintaining epithelial phenotype. Functionally, loss of ZHX2 expression enriches the hybrid MET cells and inhibits the migration and dissemination of TNBC cells or organoids, which could be reversed by restoration of E-cadherin. Moreover, depletion of ZHX2 suppresses lung metastasis in preclinical models of TNBC. In patients with TNBC, ZHX2 expression was amplified and negatively correlated with the expression of E-cadherin. These findings suggest that loss of ZHX2 promotes the hybrid MET state to impair TNBC progression.

Abstract Background Intradialytic hypotension (IDH) is frequently accompanied by symptoms of nausea, dizziness, fatigue, muscle spasm, and arrhythmia, which can adversely impact the daily lives of patients who undergo hemodialysis and may lead to decreased quality of life (QoL). This study employed the KDQOL™-36 scale to evaluate the impact of frequent IDH, based on the definition determined by predialysis blood pressure (BP) and nadir systolic blood pressure (SBP) thresholds, on the QoL of patients. Methods This is a single center retrospective cohort study involving 160 hemodialysis patients. Abstract Aim To evaluate the expression profile of long non-coding RNAs (lncRNAs) in calcific aortic valve disease (CAVD) and explore their potential mechanism of action. Methods The gene expression profiles (GSE153555, GSE148219, GSE199718) were downloaded from the Gene Expression Omnibus (GEO) database and FastQC was run for quality control checks. After filtering and classifying candidate lncRNAs by differentially expressed genes (DEGs) and weighted co-expression networks (WGCNA) in GSE153555, we predicted the potential cis- or trans-regulatory target genes of differentially expressed lncRNAs (DELs) by using FEELnc and established the competitive endogenous RNA (ceRNA) network by miRanda, more over functional enrichment was analyzed using the ClusterProfiler package in R Bioconductor. The hub cis- or trans-regulatory genes were verified in GSE148219 and GSE199718 respectively. Results There were 340 up-regulated lncRNAs identified in AS group compared with the control group (|log2Fold Change| ≥ 1.0 and Padj ≤ 0.05), and 460 down-regulated lncRNAs. Based on target gene prediction and co-expression network construction, twelve Long non-coding RNAs (CDKN2B-AS1, AC244453.2, APCDD1L-DT, SLC12A5-AS1, TGFB3, AC243829.4, MIR4435-2HG, FAM225A, BHLHE40-AS1, LINC01614, AL356417.2, LINC01150) were identified as the hub cis- or trans-regulatory genes in the pathogenesis of CAVD which were validated in GSE148219 and GSE19971. Additionally, we found that MIR4435-2HG was the top hub trans-acting lncRNA which also plays a crucial role by ceRNA pattern. Conclusion LncRNAs may play an important role in CAVD and may provide a new perspective on the pathogenesis, diagnosis, and treatment of this disease. Further studies are required to illuminate the underlying mechanisms and provide potential therapeutic targets.

Objective: MicroRNA-29a-3p has been reported in a variety of cancers, but its role in hypopharyngeal cancer remains unclear. This study was to determine the role of microRNA-29a-3p in the occurrence and development of hypopharyngeal cancer. Methods: 40 patients with hypopharyngeal cancer who underwent surgery in the Affiliated Hospital of Jining Medical University from April 2013 to November 2017 were selected for this study. The cancer tissue samples of the patients were collected, and the patients were followed up for three years. The expression of microRNA-29a-3p in tissue samples was detected by in situ hybridization with fluorescent probe, and the relationships among microRNA-29a-3p and clinicopathological factors, postoperative recurrent-metastasis, survival time were studied. Immunohistochemical was used to detect the expression of Ki67 and E-cadherin in tissue samples. Results: Combined with HE staining results showed that microRNA-29a-3p expression was relatively high in non-cancer tissue cells (red blood cells and fibroblasts in tumor interstitial vessels), but was relatively low in cancer tissue and cells. Pumpkin polysaccharides (PPe) have a variety of bioactive effects and our previous research showed the acid hydrolysate (PPe-S, a mixture) from PPe had an antioxidative capacity both in vitro and in viro. The aim of this study was to purify PPe-S and investigate the antioxidant stress effects of 2 purified components (PPe-S-1 and PPe-S-2) using Caenorhabditis elegans as model organism. The results showed that PPe-S-2 had a notable antioxidant effect, and could significantly enhance the activities of antioxidant enzymes including superoxide dismutase (SOD) (P < 0.01), catalase (CAT) (P < 0.01) and glutathione reductase (GR) (P < 0.05), and increase the level of glutathione (GSH) (P < 0.01), and decreased the content of malondialdehyde (MDA) (P < 0.05). PPe-S-2 could significantly extend the survival time of C. elegans (P < 0.01), which were stress-induced by hydrogen peroxide and methyl viologen. PPe-S-2 was a heteropolys accharide composed of glucose, arabinose, rhamnose and galactose with the molar ratio of 1.00:0.03:0.02:0.14. The molecular weight of PPe-S-2 was 0.73 kDa detected by high performance liquid chromatography.These studies demonstrated that PPe-S-2 obtained by the acid hydrolysis of PPe had a prominent protective effect to the damage induced by the intracellular free radical generating agents.

Humic acid (HA) is a major component in dissolved natural organic matter (NOM) that is commonly found in natural water sources such as surface water and soil. Although HA is non-toxic, it is a precursor of carcinogenic and mutagenic disinfection by-products that will be generated when chlorine and chloramine are applied to disinfect water during the chlorination process. Hence, researchers have been investigating various strategies to remove HA from water sources and nanoparticles stood out as one of the preferred materials for the removal. However, owing to the tiny size of nanoparticles, the recycling, and removal of nanoparticles through sedimentation and centrifugation method is often time and energy-consuming. Therefore, this work set out to immobilize iron oxide nanoparticles (nanoFe3O4) onto fabric material to create a flexible catalyst that is feasible in degrading HA. The immobilization of nanoFe3O4 onto woven and non-woven fabrics was successfully done through in situ co-precipitation method. The flexible catalyst was found to be responsive to magnetic pull, which is one of the properties of nanoFe3O4 itself. On the other hand, scanning electron microscopy (SEM) images have verified the attachment of nanoFe3O4 was in an irregular pattern across the heterogeneous surface and it was grown on the fabric’s filament instead of being trapped between the pores of the fabric. Subsequently, the as-made flexible catalysts were tested and found to be feasible as it can degrade HA completely in 24 to 36 h. More importantly, the flexible catalyst can be removed easily in an instant with a negligible detachment of nanoparticles from the fabric material. While this preliminary result is promising, it is suggested that further study should be carried out to optimize the efficiency of this novel flexible catalyst on the degradation of HA.

BackgroundCardiovascular MRI is advantageous in transcatheter aortic valve implantation (TAVI) planning. This study aimed to evaluate the feasibility of comprehensive non-contrast MRI [relaxation-enhanced angiography without contrast and triggering (REACT)] combined with a three-dimensional whole-heart MRI protocol for preprocedural planning of TAVI vs. computed tomography angiography (CTA).MethodsThirty patients with severe aortic stenosis were prospectively enrolled. The anatomical properties of the aortic root anatomy, including the perimeter and area of the virtual aortic valve annulus and coronary heights, were determined from 3D whole-heart MRI and cardiac CTA (CCTA) images, respectively. The diameters of the aorta (thoracic and abdominal aorta) and iliofemoral arteries were measured from REACT and aortic CTA (ACTA) images, respectively. A paired t-test was used to compare these two modalities. Bland–Altman plots were used to assess cardiovascular MRI and CTA measurements. Transcatheter heart valve (THV) sizing was performed based on CCTA measurements and compared with 3D whole-heart MRI measurements. The extent of annular calcification on 3D whole-heart MRI images was evaluated by a four-point grading scale and compared with CCTA data.ResultsAll 30 patients completed CTA and cardiovascular MRI examinations, with the TAVI procedure being administered in 25 patients. The mean acquisition time of the comprehensive MRI protocol was 18 ± 3.2 min. There were no significant differences between ACTA and REACT data in regard to the diameters of aortic and iliofemoral arteries, including the ascending thoracic aorta (37 ± 4.6 mm vs. 37.7 ± 5.2 mm, p = 0.085), descending thoracic aorta (24.3 ± 2.8 mm vs. 24.3 ± 2.8 mm, p = 0.832), abdominal aorta (20.9 ± 2.5 mm vs. 20.8 ± 2.5 mm, p = 0.602), bilateral common iliac arteries (right: 8.36 ± 1.44 mm vs. 8.42 ± 1.27 mm, p = 0.590; left: 8.61 ± 1.71 mm vs. 8.86 ± 1.46 mm, p = 0.050), and bilateral femoral arteries (right: 6.77 ± 1.06 mm vs. 6.87 ± 1.00 mm, p = 0.157; left: 6.75 ± 1.02 mm vs. 6.90 ± 0.80 mm, p = 0.142). Both modalities showed similar aortic valve morphology and semi-quantitative valve calcification (all, p's > 0.05). Overall agreement for implanted THV was found in all 25 (100%) patients assessed with both modalities.ConclusionREACT combined with 3D whole-heart MRI enables reliable measurements of aortic root anatomy, annular calcification, and aorta and iliofemoral access in patients under evaluation for TAVI.

Malignant tumors of the biliary tract exhibit a high degree of malignancy and heterogeneity with a poor overall prognosis. Immunotherapy has limited benefits for patients with cholangiocarcinoma. Radiation therapy can change the tumor microenvironment, but its effect heavily depends on radiation dose and fraction. We report a case of advanced intrahepatic cholangiocarcinoma in a 43-year-old male patient, with a huge liver mass of 16.5 cm in diameter, with bone and liver metastases at the first diagnosis. First-line treatment with chemotherapy and PD1 inhibitor was sustained only for 8 months. In second-line treatment, radiotherapy was administered, with 5 Gy in 5 fractions administered to the entire tumor area and 25 Gy in 5 fractions to the solid lesions of the tumor. After the completion of radiotherapy, programmed cell death 1 inhibitor combined with tyrosine kinase inhibitor was maintained. The patient achieved a progression-free-survival time of 12 months and an overall survival time of 25 months. The success of our case suggests that mixed low- and high-dose radiation can significantly improve tumor control and survival time. In clinical practice, based on the characteristics of the tumor and existing treatment options, the rational combination of existing treatment regimens can improve the prognosis of cholangiocarcinoma.

BackgroundTranscatheter aortic valve replacement (TAVR) potentially may be significantly simplified by using the single artery access (SA) technique, which does not require a secondary artery access. Nevertheless, the safety and efficacy of this technique remains unclear. Our goal was to determine if single artery access TAVR (without upgrading the sheath size) is a feasible, minimally invasive procedure.MethodsPatients with symptomatic severe aortic stenosis who underwent TAVR via the femoral artery were consecutively enrolled in this study. Eligible individuals were divided into 2 groups: the SA group and the dual artery access (DA) group. The primary end point was device success (defined by the valve academic research consortium 3, VARC 3). A 6-month follow-up and propensity score matching analyses were performed.ResultsAfter propensity score matching analysis, a total of 130 patients were included: 65 in the SA group and 65 in the DA group. The SA procedure achieved similar device success (95.4% vs. 87.7%; P = 0.115) compared with the DA procedure. The SA procedure shortened the operating time (102 min vs. 125 min; P = 0.001) but did not increase the x-ray time or dose. Both a 20 Fr and a 22 Fr sheath (without upgrading the sheath size) could be used for the SA procedure. There was no major vascular complication occurred in both groups. The incidence of minor main vascular and access complications in the SA group was comparable to those of the DA procedure (0.0% vs. 3.1%; P = 0.156).ConclusionsThe SA access procedure is a promising minimally invasive TAVR technique with a low incidence of vascular complications and a high incidence of device success. It is safe and possibly applicable in all TAVR procedures.

BackgroundClinical evidence of transcatheter aortic valve replacement in patients with type-0 bicuspid aortic valve was relatively scarce.AimsOur goal was to explore determinants of device success after transcatheter aortic valve replacement in patients with type-0 bicuspid aortic valve morphology.MethodsIn this retrospective multicenter analysis, we included 59 patients with symptomatic severe aortic stenosis with type-0 bicuspid aortic valve morphology who underwent transcatheter aortic valve replacement. Type-0 bicuspid aortic valve was identified with multidetector computed tomography scans. The technical success rate was 89.8%, and the device success rate was 81.4%. Patients were divided into a device success group and a device failure group according to Valve Academic Research Consortium- 3 criteria.ResultsWhen we compared the two groups, we found that the ellipticity index of the aortic root and the presence of bulky calcifications at the commissure were statistically different (ellipticity index 35.7 ± 1.7 vs. 29.7 ± 1.1, p = 0.018; bulky calcification at the commissure, 54.5% vs. 4.5%, p

Turbidity source apportionment is essential but complex for precise environmental rehabilitation of worldwide urban lakeside river networks impaired by turbidity. A new turbidity source analysis framework including in-situ sampling, vessel monitoring, remote sensing, and hydrodynamic modelling was proposed, where the critical processes of flow reversal from the turbid lake, navigation, and urban non-point source in urban lakeside river networks were explicitly considered. The case study results showed that the turbidity problem (mean value was 211.3 nephelometric turbidity units (NTU)) was primarily led by suspended inorganic matter (58%), followed by suspended organic matter (19%) and other matter. The Tiaoxi River and busy shipping contributed most to the annual total suspended solids load of the target river, with the same percentage of 34%. The turbid Lake Taihu provided 64,663 tonnes and 31% of the total suspended solids load, focusing on the autumn and winter months. Tiaoxi River load was highly concentrated on the storm events, which accounted for 71.4% of the annual Tiaoxi River source (209846 tonnes). The large water volume of reverse flow and the high TSS concentration of Lake Taihu in the autumn and winter months accounted for the seasonal change in Lake Taihu’s TSS load. The coronavirus disease lockdown reduced the mean monthly total suspended solids concentration of 152.4 mg/L in November and December 2019 to 49.2 mg/L in January and February 2020, by limiting the waterway transportation and the market demand for building materials. The river estuary’s vegetation interception belt, shipping restriction and rerouting, and integrated catchment management were advised to mitigate the urban river network turbidity.

Abstract Background Age‐associated sarcopenia is characterized of progressed loss of skeletal muscle power, mass, and function, which affects human physical activity and life quality. Besides, accompanied with sarcopenia, aged population also faces a series of metabolic dysfunctions. Irisin, the cleaved form of fibronectin type III domain‐containing protein 5 (FNDC5), is a myokine induced by exercise and has been shown to exert multiple beneficial effects on health. The goal of the study is to investigate the alterations of Fndc5/irisin in skeletal muscles during ageing and whether irisin administration could ameliorate age‐associated sarcopenia and metabolic dysfunction. Methods The mRNA and protein levels of FNDC5/irisin in skeletal muscle and serum from 2‐ and 24‐month‐old mice or human subjects were analysed using qRT‐PCR and western blot. FNDC5/irisin knockout mice were generated to investigate the consequences of FNDC5/irisin deletion on skeletal muscle mass, as well as morphological and molecular changes in muscle during ageing via histological and molecular analysis. To identify the therapeutic effects of chronic irisin treatment in mice during ageing, in vivo intraperitoneal administration of 2 mg/kg recombinant irisin was performed three times per week in ageing mice (14‐month‐old) for 4 months or in aged mice (22‐month‐old) for 1 month to systematically investigate irisin's effects on age‐associated sarcopenia and metabolic performances, including grip strength, body weights, body composition, insulin sensitivity, energy expenditure, serum parameters and phenotypical and molecular changes in fat and liver. Results We showed that the expression levels of irisin, as well as its precursor Fndc5, were reduced at mRNA and protein expression levels in muscle during ageing. Multispectral image (MSI) denoising is a crucial preprocessing step for various subsequent image processing tasks, including classification, recognition, and unmixing. This article proposes a novel image denoising model that integrates both noise modeling and image prior knowledge modeling. Specifically, to account for the complexity and nonuniformity of noise, a nonindependent identically distributed mixture of Gaussian model is employed for noise modeling, and a weighted loss function is obtained. The weights used in the loss function are adaptively learned from noisy MSI and employed to adjust the denoising strength of each pixel. In additionally, the model leverages the prior knowledge of the image by utilizing a nonlocal low-rank matrix model that captures the spatial–spectral correlation and nonlocal spatial similarity priors of the image. Moreover, our model adopts the weighted spatial–spectral TV model to encode the local smoothness prior of the image. Both prior models are translated into regularization terms in the denoising model. The efficacy of the proposed method is demonstrated through both simulated and real image experiments.

The attention mechanism is one of the most influential ideas in the deep learning community, which has shown excellent efficiency in various computer vision tasks. Thus, this article proposes the convolution neural network method with the attention mechanism to enhance the feature extraction of light detection and ranging (LiDAR) data. Meanwhile, our elaborately designed cascaded block contains a short path architecture beneficial for multistage information exchange. With the full exploitation of elevation information from LiDAR data and efficient utilization of the spatial-spectral information underlying hyperspectral data, our method provides a novel solution for multimodal feature fusion. Experiments are conducted on the LiDAR and hyperspectral dataset provided by the 2013 IEEE GRSS Data Fusion Contest and multisource Trento dataset to demonstrate the effectiveness of the proposed method. The experimental results have shown the superior results of the proposed method on both LiDAR and multimodality remote sensing data in comparison with several popular baselines.

Abstract Automatic skin lesion segmentation is the most critical and relevant task in computer‐aided skin cancer diagnosis. Methods based on convolutional neural networks (CNNs) are mainly used in current skin lesion segmentation. The requirement of huge pixel‐level labels is a significant obstacle to achieve semantic segmentation of skin lesion by CNNs. In this paper, a novel weakly supervised framework for skin lesion segmentation is presented, which generates high‐quality pixel‐level annotations and optimizes the segmentation network. A hierarchical image segmentation algorithm can predict a boundary map for training images. Then, the optimal regions of candidate hierarchical levels are selected. Afterward, Superpixels‐CRF built on the optimal regions is guided by spot seeds to propagate information from spot seeds to unlabeled regions, resulting in high‐quality pixel‐level annotations. Using these high‐quality pixel‐level annotations, a segmentation network can be trained and segmentation masks can be predicted. To iteratively optimize the segmentation network, the predicted segmentation masks are refined and the segmentation network are retrained. Comparative experiments demonstrate that the proposed segmentation framework reduces the gap between weakly and fully supervised skin lesion segmentation methods, and achieves state‐of‐the‐art performance while reducing human labeling efforts.

ObjectiveTo elucidate the clinicopathological characteristics and prognostic factors of poorly differentiated thyroid carcinoma.MethodA total of 24912 thyroid carcinoma patients admitted to the First Medical Center of Chinese People’s Liberation Army General Hospital from 2005 to 2020 were retrospectively reviewed. A total of 94 patients (39 males and 55 females, a male-female ratio of 1:1.4) fulfilled the selection criteria. Of these, 73 patients had undergone surgery. The clinical and pathological data were collected from each enrolled patient. Univariate and multivariate Cox regression analyses were performed to determine independent prognostic factors. All analyses were performed with the SPSS version 26.0 and R version 1.2.5033 in the R Studio environment.ResultsThe specimens included 20 cases of poorly differentiated thyroid carcinoma complicated with papillary thyroid carcinoma, 17 cases complicated with follicular thyroid carcinoma, 34 cases complicated with other pathological types and 23 with a separate entity. The patient demonstrated a large age span, median age was 57 years (range 8–85 years, average 55.20 ± 15.74 years). Abstract. Background:. The study aimed to describe the aortic valve morphology in Chinese patients underwent transcatheter aortic valve replacement (TAVR) for symptomatic severe aortic stenosis (AS), and the impact of sizing strategies and related procedural outcomes. Methods:. Patients with severe AS who underwent TAVR were consecutively enrolled from 2012 to 2019. The anatomy and morphology of the aortic root were assessed. “Downsize” strategy was preformed when patients had complex morphology. The clinical outcomes of patients who performed downsize strategy were compared with those received annular sizing strategy. The primary outcome was device success rate, and secondary outcomes included Valve Academic Research Consortium-3 clinical outcomes variables based on 1-year follow-up. Results:. A total of 293 patients were enrolled. Among them, 95 patients (32.4%) had bicuspid aortic valve. The calcium volume (Hounsfield Unit-850) of aortic root was 449.90 (243.15–782.15) mm3. Calcium is distributed mostly on the leaflet level. Downsize strategy was performed in 204 patients (69.6%). Compared with the patients who performed annular sizing strategy, those received downsize strategy achieved a similar device success rate (82.0% [73] vs. 83.3% [170], P = 0.79). Aortic valve gradients (downsize strategy group vs. annular sizing group, 11.28 mmHg vs. 11.88 mmHg, P = 0.64) and percentages of patients with moderate or severe paravalvular regurgitation 2.0% (4/204) vs. 4.5% (4/89), P = 0.21) were similar in the two groups at 30 days after TAVR. These echocardiographic results were sustainable for one year. Conclusions:. Chinese TAVR patients have more prevalent bicuspid morphology and large calcium volume of aortic root. Calcium is distributed mostly on the leaflet level. Compare with annular sizing strategy, downsize strategy provided a non-inferior device success rate and transcatheter heart valve hemodynamic performance in self-expanding TAVR procedure.

Abstract Background Success rate of transcatheter aortic valve replacement (TAVR) in aortic regurgitation (AR) patients is relatively low on account of the absence of calcified anchoring structures. Morphological classification and corresponding TAVR strategies for AR are lacking yet. Methods The AURORA study is a prospective, multicenter, single-arm cohort study to evaluate the safety and efficacy of transfemoral TAVR for severe AR in patients with high or prohibitive risk for surgery. Patients who are ≥ 65 years and diagnosed with severe pure AR as defined by the Echocardiographic Core Laboratory will be consecutively enrolled for further multidetector computed tomography (MDCT) scanning and multiplanar analyses. Based on a new anatomical classification and dual anchoring theory, patients will be classified into 4 types according to the level of the anchoring area. Types 1, 2 and 3 (at least 2 anchoring areas) will undergo the TAVR procedure with a domestic Chinese self-expanding valve (VitaFlow Valve, MicroPort, Shanghai, China), whereas type 4 (0 or 1 anchoring area) patients will be considered unsuitable for TAVR and will receive medical treatment. Our goal is to recruit 100 patients to account for 10% missing data or loss of patients to follow-up. Procedural, 30-day, 6-month and 12-month outcomes will be assessed according to Valve Academic Research Consortium-3 criteria. Discussion The AURORA study will establish a new AR anatomical classification based on dual anchoring theory through MDCT multiplanar measurement and assess the safety and efficacy of TAVR guided by this new classification and strategy in AR patients. BackgroundArsenic is recognized as a kind of developmental toxicant, which can pass through the placenta barrier and induce health defects in offspring. However, the effects of environmental levels of arsenic exposure during gestation on the reproductive system of adult male offspring remain unclear.ObjectiveTo investigate the impact of environmental levels of arsenic exposure during gestation on testosterone synthesis and sperm quality in F1 adult male rats.MethodsForty sexually mature Wistar female rats were randomly divided into four groups according to body weight, namely control group, low-dose sodium arsenite (NaAsO2) group, middle-dose NaAsO2 group, and high-dose NaAsO2 group. They were mated with sexually mature Wistar male rats in a ratio of 2:1, and the day with presence of a vaginal plug or spermatozoa in the vaginal smear was designated as gestational day 0 (GD0). Pregnant rats were provided drinking water containing 0, 1, 5,, or 25 mg·L−1 NaAsO2 until delivery. At postnatal day 70, the F1 male rats were euthanized. Anogenital distance was measured, testis and epididymis were weighed, and associated organ coefficients were calculated. Epididymal sperm quality was evaluated. The histological changes of testis were observed. The levels of testosterone and estradiol in serum were determined with ELISA kit. The testicular mRNA relative expression levels of key steroidogenic enzymes were determined by quantitative real-time PCR. Abstract Solid contamination existing as solid particles in power fluid transmission systems may lead to transmission performance reduction, system failures, and component damage. The hydraulic reservoir will deposit the contamination and store hydraulic fluid. To investigate its purification ability for solid contamination, experiments and simulations for the motion and deposition status of the typical hydraulic system particles are carried out to reveal the interaction of particles and fluid in hydraulic water reservoirs. The results show that the CFD-DEM coupling method could predict the accurate deposition position of iron particles and sand particles when ignoring the small-scale turbulence effect in the flow field. Besides, the particle motion traces and deposition patterns in the reservoir illustrate that the flow development on the bottom surface results in the particles turning, and particles tend to settle in the low flow energy position. The motion of particles is also linked to particles Stokes number, and the same-size sand particles are easily driven by the fluid. The contribution of this paper could provide a guide for predicting the particle motion and deposition pattern in the hydraulic reservoir.

Amomi fructus is rich in volatile components and valuable as a medicine and edible spice. However, the quality of commercially available A. fructus varies, and issues with mixed sources and adulteration by similar products are common. In addition, due to incomplete identification methods, rapid detection of the purchased A. fructus quality is still an issue. In this study, we developed qualitative and quantitative evaluation models to assess the variety and quality of A. fructus using GC, electronic tongue, and electronic nose to provide a rapid and accurate variety and quality evaluation method of A. fructus. The models performed well; the qualitative authenticity model had an accuracy of 1.00 (n = 64), the accuracy of the qualitative origin model was 0.86 (n = 44), and the quantitative model was optimal on the sensory fusion data from the electronic tongue and electronic nose combined with borneol acetate content, with R2 = 0.7944, RMSEF = 0.1050, and RMSEP = 0.1349. The electronic tongue and electronic nose combined with GC quickly and accurately evaluated the variety and quality of A. fructus, and the introduction of multi-source information fusion technology improved the model prediction accuracy. This study provides a useful tool for quality evaluation of medicine and food.

Geocomputation and geospatial artificial intelligence (GeoAI) have essential roles in advancing geographic information science (GIS) and Earth observation to a new stage. GeoAI has enhanced traditional geospatial analysis and mapping, altering the methods for understanding and managing complex human–natural systems. However, there are still challenges in various aspects of geospatial applications related to natural, built, and social environments, and in integrating unique geospatial features into GeoAI models. Meanwhile, geospatial and Earth data are critical components in geocomputation and GeoAI studies, as they can effectively reveal geospatial patterns, factors, relationships, and decision-making processes. This editorial provides a comprehensive overview of geocomputation and GeoAI applications in mapping, classifying them into four categories: (i) buildings and infrastructure, (ii) land use analysis, (iii) natural environment and hazards, and (iv) social issues and human activities. In addition, the editorial summarizes geospatial and Earth data in case studies into seven categories, including in-situ data, geospatial datasets, crowdsourced geospatial data (i.e., geospatial big data), remote sensing data, photogrammetry data, LiDAR, and statistical data. Finally, the editorial presents challenges and opportunities for future research.

This paper focuses on determining the authenticity and identifying the species of Fritillariae cirrhosae using electronic nose, electronic tongue, and electronic eye sensors, near infrared and mid-level data fusion. 80 batches of Fritillariae cirrhosae and its counterfeits (including several batches of Fritillaria unibracteata Hsiao et K.C. Hsia, Fritillaria przewalskii Maxim, Fritillaria delavayi Franch and Fritillaria ussuriensis Maxim) were initially identified by Chinese medicine specialists and by criteria in the 2020 edition of Chinese Pharmacopoeia. After obtaining the information from several sensors we constructed single-source PLS-DA models for authenticity identification and single-source PCA-DA models for species identification. We selected variables of interest by VIP value and Wilk’s lambda value, and we subsequently constructed the three-source fusion model of intelligent senses and the four-source fusion model of intelligent senses and near-infrared spectroscopy. We then explained and analyzed the four-source fusion models based on the sensitive substances detected by key sensors. The accuracies of single-source authenticity PLS-DA identification models based on electronic nose, electronic eye, electronic tongue sensors and near-infrared were respectively 96.25%, 91.25%, 97.50% and 97.50%. The accuracies of single-source PCA-DA species identification models were respectively 85%, 71.25%, 97.50% and 97.50%. After three-source data fusion, the accuracy of the authenticity identification of the PLS-DA identification model was 97.50% and the accuracy of the species identification of the PCA-DA model was 95%. After four-source data fusion, the accuracy of the authenticity of the PLS-DA identification model was 98.75% and the accuracy of the species identification of the PCA-DA model was 97.50%. In terms of authenticity identification, four-source data fusion can improve the performance of the model, while for the identification of the species the four-source data fusion failed to optimize the performance of the model. We conclude that electronic nose, electronic tongue, electronic eye data and near-infrared spectroscopy combined with data fusion and chemometrics methods can identify the authenticity and determine the species of Fritillariae cirrhosae. Our model explanation and analysis can help other researchers identify key quality factors for sample identification. This study aims to provide a reference method for the quality evaluation of Chinese herbs.

BackgroundClinical values of metagenomic next-generation sequencing (mNGS) in patients with severe pneumonia remain controversial. Therefore, we conduct this meta-analysis to evaluate the diagnostic performance of mNGS for pathogen detection and its role in the prognosis of severe pneumonia.MethodsWe systematically searched the literature published in PubMed, Embase, Cochrane Library, Web of Science, Clinical Trials.gov, CNKI, Wanfang Data, and CBM from the inception to the 28th September 2022. Relevant trials comparing mNGS with conventional methods applied to patients with severe pneumonia were included. The primary outcomes of this study were the pathogen-positive rate, the 28-day mortality, and the 90-day mortality; secondary outcomes included the duration of mechanical ventilation, the length of hospital stay, and the length of stay in the ICU.ResultsTotally, 24 publications with 3220 patients met the inclusion criteria and were enrolled in this study. Compared with conventional methods (45.78%, 705/1540), mNGS (80.48%, 1233/1532) significantly increased the positive rate of pathogen detection [OR = 6.81, 95% CI (4.59, 10.11, P < 0.001]. The pooled 28-day and 90-day mortality in mNGS group were 15.08% (38/252) and 22.36% (36/161), respectively, which were significantly lower than those in conventional methods group 33.05% (117/354) [OR = 0.35, 95% CI (0.23, 0.55), P < 0.001, I2 = 0%] and 43.43%(109/251) [OR = 0.34, 95% CI (0.21, 0.54), P < 0.001]. Meanwhile, adjusted treatment based on the results of mNGS shortened the length of hospital stay [MD = -2.76, 95% CI (− 3.56, − 1.96), P < 0.001] and the length of stay in ICU [MD = -4.11, 95% CI (− 5.35, − 2.87), P < 0.001].ConclusionThe pathogen detection positive rate of mNGS was much higher than that of conventional methods. Adjusted treatment based on mNGS results can reduce the 28-day and 90-day mortality of patients with severe pneumonia, and shorten the length of hospital and ICU stay. Therefore, mNGS advised to be applied to severe pneumonia patients as early as possible in addition to conventional methods to improve the prognosis and reduce the length of hospital stay.

Abstract Background To evaluate changes in corneal biomechanical properties after long-term orthokeratology (OK) treatment and the factors affecting treatment outcomes. Methods Twenty-four myopic teenagers who wore OK lenses for more than 1 year were included. Twenty-three individuals of the same age and with the same spherical equivalent wearing single-vision spectacles (SVS) were enrolled as controls. After routine eye examinations, corneal biomechanical properties and axial length were measured. Parameters were compared between groups. Results Less axial elongation (AE) occurred in the OK group (P = 0.021). The OK group experienced a statistically significant decrease in the A1 deformation amplitude (P = 0.02), whole eye movement maximum (P = 0.026), and Ambrósio’s relational thickness to the horizontal profile (ARTh) (P < 0.001), and a statistically significant increase in the pachyslope (P < 0.001) and Corvis biomechanical index (P < 0.001). Smaller ARTh and a larger highest concavity deflection area resulted in a better refractive state. The inhibitory effect of AE was better for older patients with smaller ARTh. Conclusions Long-term OK treatment slowed myopia progression by reshaping the cornea. Smaller ARTh after OK lens wear indicated a better refractive state and slower AE and could predict OK lens treatment outcomes.

Introduction: Accumulated studies have suggested that hepatitis C virus (HCV) infection is one of the leading causes for hepatocellular carcinoma (HCC). However, the mechanisms underlying the effect of HCV on the occurrence of HCC are still poorly understood. Methods: HCV infection datasets (GSE82177 and GSE17856) and HCC datasets (The Cancer Genome Atlas Liver Hepatocellular Carcinoma and GSE89377) were downloaded from Gene Expression Omnibus or TCGA for analysis. The common differentially expressed genes in the above four datasets were identified by R software. The expression of ubiquitin D (UBD) in HCV-infected HepG2 cells was detected by RT-qPCR and Western blot, respectively. The interaction between NS3 and p53 was detected by co-immunoprecipitation. The influence of UBD on the proliferation and migration ability of HepG2 cells was evaluated by CCK-8 and wound healing assay, respectively. Results: UBD was upregulated in both HCV-infected samples and HCC samples. HCV NS3 interacted with p53 and inhibited its expression. HCV NS3-induced UBD promoted the proliferation and migration of HepG2 cells. Conclusion: Our results suggest that HCV NS3-induced UBD is positively correlated with the development of HCV-related HCC during HCV infection. Targeting UBD could be a potential strategy for preventing and treating HCV-induced HCC.

Abstract With the increasing demand to reduce emissions and save energy, hydraulic reservoirs require new architecture to optimize their weight, space, and volume. Conventional open reservoirs are large, heavy, and easily polluted, and threaten the operation of hydraulic systems. A closed reservoir provides the advantages of small volume and light weight, compared to open reservoirs. In this study, a non-metallic pressure reservoir with variable volume is designed and manufactured for closed-circuit hydraulic systems. The reservoir housing is made of rubber, and the Mooney-Rivlin model is chosen based on the rubber strain properties. The FEA simulation for the reservoir is performed using ANSYS Workbench to obtain the structural stiffness. The major contribution is the establishment of mathematical models for this reservoir, including the volume equation changing with height, flow equation, and force balance equation, to explore the output characteristics of this reservoir. Based on these results, simulation models were built to analyze the output characteristics of the reservoir. Moreover, the test rig of a conventional hydraulic system was transformed into a closed-circuit asymmetric hydraulic system for the reservoir, and preliminary verification experiments were conducted on it. The results demonstrate that the designed reservoir can absorb and discharge oil and supercharge pump inlet to benefit system operation. The changes in the volume and pressure with displacements under different volume ratios and frequencies were obtained, which verified the accuracy of the mathematical models. Owing to its lightweight design and small volume, the reservoir can replace conventional open reservoirs, and this lays a foundation for future theoretical research on this reservoir.

Abstract Obesity induced by antipsychotics have plagued more than 20 million people worldwide. However, no drug is available to eliminate the obesity induced by antipsychotics. Here we examined the effect and potential mechanisms of a gold nanoclusters (AuNCs) modified by N-isobutyryl-L-cysteine on the obesity induced by olanzapine, the most prescribed but obesogenic antipsychotics, in a rat model. Our results showed that AuNCs completely prevented and reversed the obesity induced by olanzapine and improved glucose metabolism profile in rats. Further mechanism investigations revealed that AuNCs exert its anti-obesity function through inhibition of olanzapine-induced dysfunction of histamine H1 receptor and proopiomelanocortin signaling therefore reducing hyperphagia, and reversing olanzapine-induced inhibition of uncoupling-protein-1 signaling which increases thermogenesis. Together with AuNCs’ good biocompatibility, these findings not only provide AuNCs as a promising nanodrug candidate for treating obesity induced by antipsychotics, but also open an avenue for the potential application of AuNCs-based nanodrugs in treating general obesity.

Abstract Background FBXL8 is a conserved F‐box protein, belonging to the ubiquitin ligase complex, which promotes the development and progression of tumours. However, the regulation function and mechanism of FBXL8's involvement in colorectal cancer (CRC) remain unclear. Methods RT–PCR is used to detect gene expression levels. Protein levels were determined by western blotting and flow cytometry. The bindings of FBXL8 and p53 and ubiquitination levels were detected by cell transfection and immunoprecipitation. The transwell assay was used to measure the ability of cells to migrate and invade. Animal studies were used to verify the function of FBXL8 in vivo. Results The expression of FBXL8 was up‐regulated in CRC tissues, and its overexpression was associated with poor prognosis in CRC patients. The up‐regulation of FBXL8 promoted the proliferation, invasion and migration of CRC tumour cells and maintained the stem‐cell characteristics of colorectal tumour cells. Further analysis demonstrated that FBXL8 targeted p53 and reduced its stability through ubiquitination. Knockout of FBXL8 down‐regulated the proliferation, migration and stem‐like properties of tumour cells. CRC mouse xenograft tumour model confirmed that FBXL8 gene knockout inhibited tumour formation and liver metastasis. Conclusion FBXL8 was highly expressed in CRC. Mechanism studies have shown that FBXL8 degraded tumour suppressor gene p53 by ubiquitination. FBXL8 knockout inhibited the proliferation and stem characteristics of CRC cells, so SCF‐FBXL8‐TP53 has potential to be used as a therapeutic target for CRC in subsequent studies.

It is widely acknowledged that synergetic supplies of multiple ecosystem services (ESs) can improve the efficiency of multifunctional landscape management. However, even though the satisfaction of demands for ESs clearly and comprehensively reflects their efficiency, little is known about the efficiency of co-benefits for multiple ESs when both the supply and demand sides are considered. For the first time, we analyzed the co-benefits of the supply of and demand for multiple ESs in urban areas according to the degrees of synergy found among five typical urban ESs. We found that although supplies of multiple ESs were synergetic, and the degrees of synergy of single ES supply–demand were positively correlated with each other, the co-benefits of supply–demand for multiple ESs were not substantial. We also found that the demand for ESs critically affects ES efficiency, which should therefore be assessed from the perspectives of both supply and demand. Any initiative that focuses on just one aspect cannot bring about genuine synergy; nor can it prompt the re-entry of the natural ecosystem in urban areas into a positive cycle. We elucidated the different patterns of synergy that foster a balance in the supply and demand of multiple ESs, which can provide a basis for their differentiation and for grouped management approaches to enhance the co-benefits of the supply of and demand for multiple ESs. Evidently, the supply side of ESs requires attention, especially the key aspect of ecological processes that affect the synergy of ES supplies. Consequently, an in-depth exploration of the key ecological process affecting ES supplies and their synergy is warranted in the future.
