Phylogenetic analyses of partial DNA sequences of internal transcribed spacer (ITS) and nuclear ribosomal large subunit (LSU), using Maximum-Likelihood and Bayesian Inference, reveal the taxonomic placement of Neoveronaea within the Herpotrichiellaceae, in which it forms a lineage distinct from other genera.

The current paper examines whether resilience-promoting interventions lead to increased self-reported resilience relative to a control group. The literature search was conducted using terms “resilience intervention” OR “promoting resilience” OR “promoting resiliency” OR “resilience-based intervention”. A total of 83 studies were included in the meta-analysis and systematic review. The quality of studies was evaluated based on sample size, appropriateness of statistical analyses, and protocol for intervention adoption. Moderation models examined effects of study characteristics (study rigour), delivery characteristics (mode of intervention [e.g., self-guided], mode of delivery [e.g., in-person] and format [e.g., group-based]), and resilience characteristics (type of resilience [e.g., coping] and resilience process) on outcomes. To unravel the geochemical heterogeneity and its origin in different terranes of North China, we conducted geochronological and geochemical analyses of the meta-mafic rocks from the Luliang–Zhongtiao rift zone (Shanxi Province). LA-ICP-MS zircon U–Pb dating yielded mostly End-Neoarchean to Proterozoic ages for the basement rocks (Sushui Complex: 2516 ± 26 Ma; Meta-mafic rocks: 2494 ± 31 Ma), Jiangxian Group (~ 2213 Ma), Zhongtiao Group (2077 ± 29 Ma), Jiehekou Group (1998 ± 23 Ma), and Luliang Group (2152 ± 52 Ma). Petrographic characteristics show that the meta-mafic rocks from the Neoarchean–Paleoproterozoic Zhongtiaoshan (Sushui Complex) have similar geochemical characteristics to the overlying Jiangxian and Zhongtiao Groups. The Paleoproterozoic Luliang and Yejishan Group meta-mafic rocks from Luliangshan also have similar geochemical characteristics but are geochemically different from similar-age rocks from Zhongtiaoshan. This shows that the late-stage rocks have a geochemical inheritance from the early-stage rocks in the same region and that the geochemical heterogeneity of rocks from different areas was originated from the inherited heterogeneity of the magma source.

Five new species of Neopodoconis, N. jiangxiensis, N. meilingensis, N. obclavata, N. saprophyticus and N. sinensis are described and illustrated from specimens collected on dead branches of unidentified plants in China. Phylogenetic analysis of partial DNA sequences of nuclear ribosomal small subunit (SSU) and nuclear ribosomal large subunit (LSU), using Maximum-Likelihood (ML) and Bayesian Inference (BI), supported the establishment of the five new species, and indicated a close relationship to Torulaceae in Pleosporales, Dothideomycetes. Sporidesmioides and Rostriconidium are synonymised with Neopodoconis based on morphological and molecular phylogenetic data, and the new combinations Neopodoconis aquaticum, N. cangshanense, N. pandanicola and N. thailandica are proposed. A synoptic table to Neopodoconis species are provided.

Objective To discuss the irrational drug use in clinic and its improvement measures. Methods To select 132 prescriptions of Western medicine from the pharmacy department of our hospital as the research object, prescribed from July 2017 to September 2018. According to the prescription time, the safety and irrational use of drugs were observed according to the principle of clinical rational use of drugs. They were divided into experimental group and control group, in which the control group did not include the improvement measures, the experimental group included the improvement measures and each group had sixty-six prescriptions. Results The incidence of repeated drug use, improper drug use combination, improper use of antibiotics, the abuse of antibiotics and other irrational drug uses in the experiment was lower than that in the control group and there was significant difference between the two groups (P < 0.05). Conclusion In the clinical drugs administration of Western medicine, it is necessary to strengthen the observation of irrational drug use and give timely improvement measures to ensure the safety of clinical drugs.

The interactions between Emiliania huxleyi and E. huxleyi virus (EhV) regulate marine carbon and sulfur biogeochemical cycle and play a prominent role in global climate change. As a large DNA virus, EhVs have developed a novel "virocell metabolism" model to meet their higher metabolic needs. However, the regulatory mechanism of this metabolic model is still largely unclear. MicroRNAs (miRNAs) can regulate biological pathways through targeting hub genes in the metabolic processes. Here, we performed high-throughput small RNA sequencing to analyse miRNA expression in EhV99B1 infected E. huxleyi BOF92. A total of 26 miRNAs (including 2 virus-derived miRNAs) were identified, including four up-regulated and one down-regulated miRNAs. These results were further validated through quantitative real-time PCR. Functional enrichment analysis showed that five differentially-expressed miRNAs might be involved in the regulation of carbohydrate metabolism, lipid metabolism and amino acid metabolism. Moreover, the expression levels of differentially-expressed miRNAs were negatively correlated with that of several lipid metabolism-related genes, such as ACC-1, SPT, ACOX, ACAT, CERS and ACADS, indicating that these miRNAs might play an important regulatory role in virus-mediated lipid metabolism.海洋颗石藻病毒-宿主互作是影响海洋碳、硫生物地化循环及全球气候变化的重要环节。作为大的双链DNA病毒,颗石藻病毒进化出一种“病毒细胞代谢”模式,通过重编程宿主代谢途径以满足其代谢需求,但对这一代谢模式的调控机制尚缺乏足够的认识。MicroRNA (miRNA)作为一种基因表达调控的重要因子,能够通过调控代谢过程中的靶基因表达,从而调节相关代谢通路。本研究采用small RNA测序技术分析病毒感染颗石藻差异表达的miRNA及其靶基因功能,鉴定出26条成熟miRNA (包括2条病毒来源的miRNA),均来自23条新的miRNA前体序列,其中5条miRNA显著差异表达,包括4条上调,1条下调。实时荧光定量PCR (quantitative real-time PCR, qRT-PCR)验证结果与miRNA-seq结果基本一致。功能富集分析显示,5个差异表达的miRNA可能参与调节糖代谢、脂代谢和氨基酸等代谢。此外,差异表达miRNA的表达水平与脂质代谢相关靶基因如ACC-1、SPT、ACOX、ACAT、CERS、ACADS等的表达水平呈负相关,说明这些miRNA可能在病毒感染过程中对宿主的脂质代谢发挥重要的调控作用。.

Therapeutic targeting of the nuclear enzyme poly (ADP-ribose) polymerase 1 (PARP1) with PARP inhibitors (PARPis) in patients with a homologous recombination (HR)- deficient phenotype based on the mechanism of synthetic lethality has been shown tremendous success in cancer therapy. With the clinical use of various PARPis, emerging evidence has shown that some PARPis offer hope for breakthroughs in triple-negative breast cancer (TNBC) therapy, regardless of HR status. However, similar to other conventional cytotoxic drugs, PARPis are also subject to the intractable problem of drug resistance. Notably, acquired resistance to PARPis caused by point mutations in the PARP1 protein is hard to overcome with current strategies. To explore modalities to overcome resistance and identify patients who are most likely to benefit from PARP1-targeted therapy, we developed a proteolysis-targeted chimaera (PROTAC) to degrade mutant PARP1 in TNBC. Here, we investigated a PARP1 PROTAC termed "NN3″, which triggered ubiquitination and proteasome-mediated degradation of PARP1. Moreover, NN3 degraded PARP1 with resistance-related mutations. Interestingly, compared with other reported PARP1 degraders, NN3 exhibited a unique antitumor mechanism in p53-positive breast cancer cells that effectively promoted ferroptosis by downregulating the SLC7A11 pathway. Furthermore, NN3 showed potent activity and low toxicity in vivo. In conclusion, we propose PROTAC-mediated degradation of PARP1 as a novel strategy against mutation-related PARPi resistance and a paradigm for targeting breast cancer with functional p53 via ferroptosis induction.

To observe the moxibustion sensation and the clinical therapeutic effect of heat-sensitive moxibustion of two different suspension moxibustion methods and imitation moxibustion apparatus on mild to moderate knee osteoarthritis (KOA), and compare the therapeutic effect of different moxibustion methods.A total of 90 patients with mild to moderate KOA were randomized into a hand-held group (30 cases, 1 case dropped off), an imitation moxibustion apparatus group (30 cases) and a moxibustion shelf group (30 cases, 1 case dropped off).The compositions of moxibustion sensation and numbers of moxibustion sensation types in individuals in the hand-held group and the moxibustion shelf group were richer, the moxibustion sensation intensity was higher than that in the imitation moxibustion apparatus group (Heat-sensitive moxibustion can effectively treat knee osteoarthritis, while the different suspension moxibustion methods have an influence on clinical therapeutic effect, hand-held suspension moxibustion has the best efficacy.

Background: Colorectal cancer (CRC) has been a major public health problem. Tumor microenvironment (TME) greatly contributes to the heterogeneity of CRC and is crucial for the regulation of CRC progression. The authors' study wanted to develop a robust prognostic signature for CRC patients based on TME-related genes. Materials and Methods: Gene expression data and clinicopathologic information of CRC patients were collected from Gene Expression Omnibus and The Cancer Genome Atlas databases. TME-related genes with prognostic value were identified by Cox regression and bootstrap method. Then, the authors used the prognostic genes to construct robust prognostic model through least absolute shrinkage and selection operator (LASSO) regression method. The immune and stromal cell abundance of CRC samples were estimated by microenvironment cell populations-counter method. Results: Based on a training set that comprised 893 CRC samples and 4775 TME-related genes, they established a prognostic model consisting of 25 TME-related genes. With specific risk score formulae, the prognostic model divided CRC patients into high-risk and low-risk subgroups with significantly different survival, which were further confirmed in validation cohorts consisting of other 473 CRC cases or subpopulation of specific stages. The result of time-dependent receiver operating characteristic analysis demonstrated strong predictive accuracy of the prognostic model both in training and validation cohorts. Multivariate Cox regression analysis showed that the 25-gene signature was an independent prognostic factor for overall survival, which was validated through clinical subgroups analysis. Further analysis revealed that CRC samples of high-risk group was abundant of stromal-relevant processes and had a significantly higher proportion of fibroblasts and endothelial cells infiltration. Conclusion: The authors established a robust prognostic signature of 25 TME-related genes. It may be an effective tool for prognostic prediction and CRC patient stratification, which helps in treatment decisions.

Metastasis is the primary cause of cancer-related mortality in colorectal cancer (CRC) patients. How to improve therapeutic options for patients with metastatic CRC is the core question for CRC treatment. However, the complexity and diversity of stromal context of the tumor microenvironment (TME) in liver metastases of CRC have not been fully understood, and the influence of stromal cells on response to chemotherapy is unclear. Here we performed an in-depth analysis of the transcriptional landscape of primary CRC, matched liver metastases and blood at single-cell resolution, and a systematic examination of transcriptional changes and phenotypic alterations of the TME in response to preoperative chemotherapy (PC). Based on 111,292 single-cell transcriptomes, our study reveals that TME of treatment-naïve tumors is characterized by the higher abundance of less-activated B cells and higher heterogeneity of tumor-associated macrophages (TAMs). By contrast, in tumors treated with PC, we found activation of B cells, lower diversity of TAMs with immature and less activated phenotype, lower abundance of both dysfunctional T cells and ECM-remodeling cancer-associated fibroblasts, and an accumulation of myofibroblasts. Our study provides a foundation for future investigation of the cellular mechanisms underlying liver metastasis of CRC and its response to PC, and opens up new possibilities for the development of therapeutic strategies for CRC.

Background Psychological distress, isolation, feelings of powerlessness, and limited social support are realities faced by temporary migrant live-in caregivers in Canada. Furthermore, they experience multiple barriers in accessing mental health services due to their long work hours, limited knowledge of health resources, precarious employment, and immigration status. Objective The Women Empowerment - Caregiver Acceptance & Resilience E-Learning (WE2CARE) project is a pilot intervention research project that aims to promote the mental well-being and resiliency of migrant live-in caregivers. The objectives include exploring the effectiveness of this program in achieving the following: (1) reducing psychological distress (depression, anxiety, and stress); (2) promoting committed actions of self-care; and (3) building mutual support social networks. Further, participants’ satisfaction with the intervention and their perceived barriers to and facilitators of practicing the self-care strategies embedded in WE2CARE will be examined. Methods A total of 36 live-in caregivers residing in the Greater Toronto Area will be recruited and randomly assigned to either the intervention or waitlist control group. The intervention group will receive a 6-week web-based psychosocial intervention that will be based on Acceptance and Commitment Therapy (ACT). Standardized self-reported surveys will be administered online preintervention, postintervention, and at 6 weeks postintervention to assess mental distress (Depression, Anxiety and Stress Scale), psychological flexibility (Acceptance and Action Questionnaire), mindfulness (Cognitive and Affective Mindfulness Scale – Revised), and resilience (Multi-System Model of Resilience Inventory). In addition, two focus groups will be held with a subset of participants to explore their feedback on the utility of the WE2CARE program. Results WE2CARE was funded in January 2019 for a year. The protocol was approved by the research ethics boards of Ryerson University (REB 2019-036) and the University of Toronto (RIS37623) in February and May 2019, respectively. Data collection started upon ethics approval and was completed by May 2020. A total of 29 caregivers completed the study and 20 participated in the focus groups. Data analyses are in progress and results will be published in 2021. Conclusions WE2CARE could be a promising approach to reducing stress, promoting resilience, and providing a virtual space for peer emotional support and collaborative learning among socially isolated and marginalized women. The results of this pilot study will inform the adaptation of an ACT-based psychological intervention for online delivery and determine its utility in promoting mental health among disadvantaged and vulnerable populations. Two new cephalochromin derivatives, prenylcephalochromin A (1), prenylcephalochromin B (2), along with cephalochromin (3) were isolated from the Alternaria sp. ZG22 obtained from a Dasymaschalon rostratum collected from the Hainan. The structures of two new compounds were elucidated by comprehensive spectroscopic methods. Compounds 1-3 showed ��-glucosidase inhibitory activity.

Background YAP activation is crucial for cancer development including colorectal cancer (CRC). Nevertheless, it remains unclear whether N6-Methyladenosine (m6A) modified transcripts of long noncoding RNAs (lncRNAs) can regulate YAP activation in cancer progression. We investigated the functional link between lncRNAs and the m6A modification in YAP signaling and CRC progression. Methods YAP interacting lncRNAs were screened by RIP-sequencing, RNA FISH and immunofluorescence co-staining assays. Interaction between YAP and lncRNA GAS5 was studied by biochemical methods. MeRIP-sequencing combined with lncRNA-sequencing were used to identify the m6A modified targets of YTHDF3 in CRC. Gain-of-function and Loss-of-function analysis were performed to measure the function of GAS5-YAP-YTHDF3 axis in CRC progression in vitro and in vivo. Results GAS5 directly interacts with WW domain of YAP to facilitate translocation of endogenous YAP from the nucleus to the cytoplasm and promotes phosphorylation and subsequently ubiquitin-mediated degradation of YAP to inhibit CRC progression in vitro and in vivo. Notably, we demonstrate the m6A reader YTHDF3 not only a novel target of YAP but also a key player in YAP signaling by facilitating m6A-modified lncRNA GAS5 degradation, which profile a new insight into CRC progression. Clinically, lncRNA GAS5 expressions is negatively correlated with YAP and YTHDF3 protein levels in tumors from CRC patients. Conclusions Our study uncovers a negative functional loop of lncRNA GAS5-YAP-YTHDF3 axis, and identifies a new mechanism for m6A-induced decay of GAS5 on YAP signaling in progression of CRC which may offer a promising approach for CRC treatment.

A series of d10 based metal-organic frameworks (MOFs), namely, [Cd2(dpc)(bipy)(H2O)3]n (Ⅰ), {[Cd2(dpc)(bib)(H2O)]·H2O}n (Ⅱ), {[Cd2(dpc)(bitb)1.5(H2O)2]·H2O}n (Ⅲ) (H4dpc = 2-(3′,4′-dicarboxylphenoxy)isophthalic acid, bipy = 4,4′-dipyridyl, bib = 1,4-bis (1-imidazolyl)benzene, bitb = 1,4-bis (imidazol-1-ylmethyl)benzene) have been designed and successfully synthesized using a flexible tetrabasic carboxylic acid along with three ancillary ligands under hydrothermal conditions, which exhibit structural diversities of targeted Cd-MOFs. Single-crystal X-ray diffraction analysis reveals that they contain building units of {CdN1O6} and {CdN1O5} (for I), {CdN1O5} (for II), as well as {CdN2O4} and {CdN5O1} (for Ⅲ), based on which to assemble microporous three-dimensional (3D) frameworks via covalent bonds and/or hydrogen bonds using multiple coordination modes of H4dpc ligand and auxiliary linkers. The solid-state luminescence performances of these Cd-MOFs were investigated at ambient temperature. Furthermore, luminescent sensing responses of Ⅰ-Ⅲ to amino acids and metal cations were systematically explored. Notably, Ⅰ and II may act as potential dual-functional luminescent sensors for Aspartic acid and Cu2+ cations as well as Aspartic acid and Fe3+ cations with high selectivity and sensitivity in aqueous solutions, respectively. The mechanisms of the quenching effect and sensing properties of the as-synthesized Cd-MOFs have also been discussed in this report.

Three metal-organic frameworks (MOFs), namely, [Mn(CPP)2(H2O)]n (I), [Cd(CPP)2(H2O)]n (II) and {[Zn2(CPP)2(CO3)]·(ClO4)·(H2O)2}n (III) (HCPP = 4'-(4-(4-carboxylphenoxy)-2,2':6′,2′-terpyridine), were successfully achieved under hydrothermal conditions. The as-synthesized coordination polymers have been characterized by IR spectroscopy, elemental analyses and single crystal X-ray diffraction. Single-crystal X-ray diffraction analyses indicate that compounds Ⅰ and Ⅱ display isomorphous and isostructural, in which the central metal ions present distorted octahedral configurations. Ⅰ and Ⅱ feature fascinating 3D frameworks based on π⋯π and C H⋯π interactions between the pyridine rings appertaining CPP− ligands, while compound Ⅲ possesses a 3D net interlinked via C-H⋯π interactions between the phenyl rings belonging to the CPP− ligands. In their 3D frameworks, the {MnN3O3}, {CdN3O3} and {ZnO2N3}–{ZnO2N3} building units are observed, respectively. Luminescent properties of coordination polymer Ⅱ and Ⅲ demonstrate that they may be acted as fluorescent sensors toward Cu2+ cation and Tryptophan, respectively. Furthermore, investigations on magnetic property reveal that coordination polymer Ⅰ presents antiferromagnetic coupling.

Past research has often conceptualized stress from a deficit-oriented approach. This approach is unbalanced, often associating stress with negative events and outcomes. The current study examined stress from the Transactional Model, and proposed alternative ways to conceptualize stress. In addition, both psychosocial and physiological stressors were utilized to induce stress. Study aims were threefold: (1) to examine beliefs about stress and effects of framing on changes to perceptions of stress, (2) to examine the comparative effects of two different stress-induction methods, and (3) to explore the interactive effects of framing and stress-induction on subjective perceptions of stress and measures of stress reactivity. Results confirmed a deficit-orientation of stress within the sampled population. Comparative effects of both stressors highlighted differential stress responses based on task demands and appraisal. Finally, interactive effects of framing and stress-induction provided support for alternative conceptualizations of stress in adaptive coping.

Background Chinese students are extremely vulnerable to developing mental illness. The stigma associated with mental illness presents a barrier to seeking help for their mental health. Objective The Linking Hearts—Linking Youth and ‘Xin’ (hearts) project is an implementation science project that seeks to reduce mental illness stigma and promote the mental health of university students in Jinan, China. The Linking Hearts project consists of 3 components. In this paper, we outline the protocol for the first component, that is, the contextual assessment and analysis of the mental health needs of university students as the first step to inform the adaptation of an evidence-based intervention to be implemented in Jinan, China. Methods Six local universities will participate in the Linking Hearts project. A total of 100 students from each university (n=600) will engage in the contextual assessment through self-report surveys on depression, anxiety, stress, mental health knowledge, and mental health stigma. Quantitative data will be analyzed using several descriptive and inferential analyses via SPSS. A small number of participants (144 students and 144 service providers) will also be engaged in focus groups to assess the socio-environmental contexts of university students’ health and availability of mental health resources. Qualitative data will be transcribed verbatim and NVivo will be used for data management. Social network analysis will also be performed using EgoNet. Results Linking Hearts was funded in January 2018 for 5 years. The protocol of Linking Hearts and its 3 components was approved by the research ethics boards of all participating institutions in China in November 2018. Canadian institutions that gave approval were Ryerson University (REB2018-455) in January 2019, University of Alberta (Pro00089364), York University (e2019-162) in May 2019, and University of Toronto (RIS37724) in August 2019. Data collection took place upon ethics approval and was completed in January 2020. A total of 600 students were surveyed. An additional 147 students and 138 service providers took part in focus groups. Data analysis is ongoing. Results will be published in 2021. Conclusions Findings from this contextual assessment and analysis will generate new knowledge on university students’ mental health status, mental health knowledge, and resources available for them. These findings will be used to adapt and refine the Acceptance and Commitment to Empowerment-Linking Youth N’ Xin intervention model. Reactive oxygen species (ROS) are mitochondrial respiration byproducts, the accumulation of which may cause oxidative damage and is associated with several chronic health problems. As an essential unsaturated fatty acid, eicosapentaenoic acid (EPA) provides various physiological functions; however, its exact regulatory role remains elusive. The current study aimed to address how EPA regulates cellular antioxidant capacity and the possible mechanisms of action. Upon 48 h of EPA treatment, the ROS levels of HepG2 cells were reduced by at least 40%; the total cellular antioxidant capacity was increased by approximately 50–70%, accompanied by enhanced activities and expression of major antioxidant enzymes. Furthermore, the mitochondrial membrane potential and the mitochondrial biogenesis were dramatically improved in EPA-treated cells. These data suggest that EPA improves cellular antioxidant capacity by enhancing mitochondrial function and biogenesis, which sheds light on EPA as a dietary complement to relieve the oxidative damage caused by chronic diseases.

Parkes-Weber syndrome (PWS) is a rare congenital vascular syndrome consisting of capillary, venous, lymphatic, and arteriovenous malformation. There are many complications of PWS, such as ulceration, bleeding, infection, and cardiac failure. Among them, skin ulceration is one of the thorniest problems in PWS, requiring multidisciplinary approaches for the management. In this article, we presented the case of an elderly patient with refractory ulceration who received numerous treatments with no effect and finally underwent a major amputation to improve the quality of life. Moreover, we reviewed 23 previously reported cases to improve our understanding of the management for PWS patients with ulceration.
