Two-dimensional layered structure of rGO@Bi2O2CO3@NiCo-LDHs as positive material of supercapacitor is fabricated using the simple hydrothermal reaction, freeze-drying technology, and heat treatment process. The corresponding electrode exhibits a maximum specific capacity of 1147 F/g at 0.5 A/g and 82.2% capacity retention after 1000 times charging and discharging at 10 A/g. Additionally, negative material of rGO film is also prepared, which has a higher specific capacity of 354 F/g than that of other carbon materials. To obtain a high performance of supercapacitor device, an asymmetric supercapacitor of rGO//rGO@Bi2O2CO3@NiCo-LDHs is assembled based on rGO@Bi2O2CO3@NiCo-LDHs, rGO film, and 2 M KOH electrolyte. The testing data show that the maximum specific capacitance and energy density of asymmetric supercapacitor are 85 F/g and 23.14 Wh/kg, respectively. Simultaneously, it also shows the good electrochemical stability, remaining 80% capacity after 3000 cycles. This means that rGO@Bi2O2CO3@NiCo-LDHs could be regarded as a promising electrode material for energy storage devices.

BACKGROUND: The concordance between mutations detected from plasma and tissue is critical for treatment choices of patients with advanced lung adenocarcinoma. METHODS: We prospectively analyzed the association of the serum tumor markers with the concordance between blood and tissue genomic profiles from 185 patients with advanced lung adenocarcinoma. The concordance was defined according to 3 criteria. Class 1 included all targetable driver mutations in 8 genes; class 2 included class 1 mutations plus mutations in KRAS, STK11, and TP53; class 3 included class 2 mutations plus tumor mutation burden (TMB) status. RESULTS: Collectively, 150 out of 185 patients had mutations in both tissue and plasma samples, while one patient was mutation-negative for both, resulting a concordance of 81.6%. The concordance rate for class 1 mutations was 80%, and 65% and 69% for class 2 and class 3, respectively. Carbohydrate antigen 19-9 (CA19-9) or cytokeratin 19 (CYFRA21-1) levels higher than the normal upper limit predicted the concordance of tissue and blood results in class 1 (P=0.005, P=0.011), class 2 (P=0.011, P0.99, P>0.99). When the sum score for 4 serum tumor markers was greater than 35, both class 1, class 2, and class 3 reached a predictive threshold of 90%. CONCLUSIONS: Serum tumor markers can be used as easy and practical clinical predictors of concordance in mutation profiles between blood and tissue samples from patients with advanced lung adenocarcinoma.

Although the concepts of loyalty, service quality and customer satisfaction have frequently been discussed by scholars, their connections with emotional factors have rarely been investigated in the field of museum tourism. The Anthropology Museum of Guangxi focuses on unique ethnic minorities, which is used as a case study in this paper. The key question is whether visitors are driven by certain emotional factors. This paper examines the relationships among the factors that influence visitors ’ emotional attachment, satisfaction, conative loyalty and perceived quality. Structural equation modelling (PLS-SEM) is used to model the relations among these four constructs, which include formative and reflective constructs. The structural model was tested on a sample of 184 visitors to the Anthropology Museum of Guangxi, and empirical study of the conceptual model supports all of the paper ’ s hypotheses. The results show positive influences among emotional attachment, visitor satisfaction and perceived quality, which may indeed affect visitors ’ conative loyalty to a specific museum. The practical applications are suggested based on the finding that emotional attachment plays an important role in predicting visitor loyalty.

A tunable frequency-multiplying optoelectronic oscillator (OEO) based on a dual-parallel Mach-Zehnder modulator (DPMZM) is proposed and experimentally demonstrated. In the proposed system, the tunable fundamental microware signal is generated by a tunable optoelectronic oscillator incorporating a phase-shifted fiber Bragg grating (PS-FBG). By adjusting the DC bias of the DPMZM, the frequency-doubled microwave signal with a tunable frequency range from 11 GHz to 20 GHz and the frequency-quadrupled microwave signal with a tunable frequency range from 22.5 GHz to 26 GHz are generated. The phase noises of the fundamental, frequency-doubled and frequency-quadrupled signals at 10 kHz offset frequency are −105.9 dBc/Hz, −103.3 dBc/Hz and −86.2 dBc/Hz, respectively.

Rearing rodents in an enriched environment (EE), with increased sensory stimulations and social interactions, is a well-established model for naturally increasing neural activity. It is well-known that EE-rearing of rodents from adolescence or during adulthood leads to extensive biochemical, morphological, electrophysiological and behavioral changes. Here, we examine the effects of EE-rearing from birth on adult behavior. Through a battery of assays, we found that mice EE-reared from birth had better acquisition and consolidation of memory, in both aversive-based fear conditioning and reward-based contextual association tasks. Moreover, EE-reared mice showed reduced anxiety in novel environments and enhanced social interactions. Together, these results demonstrated that EE-rearing from birth significantly improved motor ability, learning and memory and sociability, while reducing anxiety. A better understanding of how early environmental influences affect behavior is not only important for understanding neural circuit wiring, but also provides insight into developing more effective intervention programs for neurodevelopmental disorders.

In this paper, the application of chemometrics coupled with high-performance liquid chromatography-diode array detection (HPLC–DAD) for the rapid and sensitive quantitative analysis of seven flavonoids in the raw and purified propolis samples was investigated. The objective of this paper was to receive a successful resolution of the complex propolis matrix with minimum experimental time (3.5 min) when appearing unknown interferences, highly overlapping peaks, time and baseline drifts among chromatograms. First, the validation samples contained seven flavonoids at five levels, helped to understand and interpret the features of the second-order calibration method based on the alternating trilinear decomposition (ATLD) algorithm. Second, the experimental data were studied for the simultaneous determination of flavonoids in the raw and purified propolis samples using the function of "mathematical separation" of ATLD algorithm. Except for myricetin (MY), the average recoveries of other analytes were 95.79–105.82% for the raw propolis samples, 94.41–108.06% for the purified propolis samples. In addition, the limit of detection (LOD, 0.01–0.20 μg mL−1), limit of quantification (LOQ, 0.03–0.60 μg mL−1), sensitivity (SEN, 15.97–184.31 mL μg−1), selectivity (SEL, 0.16–0.76) and root-mean-squared error of prediction (RMSEP, 0.29–1.64 μg mL−1) of the proposed method, demonstrated that all the target analytes have good sensitivity, low detection limit. According to the experimental results, the total content of flavonoids in purified propolis is about twice that in raw propolis, which is consistent with the actual situation. This analytical strategy will be extremely promising for the identification and quantification of analytes studied in the complex samples.

BACKGROUND: SOXF family genes (SOX7, SOX17, SOX18) have been reported to involved in tumorigenesis and development in previous articles, separately. But data sources, analysis contents and criteria are not same. Here, we focused on SOXF genes to analyze the regulatory mechanisms and diagnostic value at the same standards. METHODS: This study analyzed functions, expressions, methylations, and mutations of SOXF genes through public databases including Metascape, Gene Expression Profiling Interactive Analysis (GEPIA), cBioPortal, Tumor IMmune Estimation Resource (TIMER), and Kaplan-Meier Plotter. TIMER applies a deconvolution method to infer the abundance of tumor-infiltrating immune cells (TIICs) from gene expression profiles. Metascape combines several biological functions and over 40 independent knowledge bases within one integrated portal. GEPIA analyses RNA sequencing expression data from the The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) projects. The cBioPortal visualizes and analyses genetic data from cancer studies. RESULTS: This study found that SOXF genes had low expressions in multiple types of cancer, such as lung cancer and breast cancer (ANOVA differential methods, |log2FC| cutoff: 1, q value cutoff: 0.01). The lung adenocarcinoma (LUAD) patients with high expression of SOX7 [HR =0.72 (0.61–0.85), logrank P=8.1e-05) and SOX17 [HR =0.54 (0.45–0.64), logrank P=1.7e-12] had a higher overall survival (OS) rate. Expression of SOX7 was significantly related to the copy number variation (CNV) (P=3.02e-8) and promoter methylation level (P=5.33e-14), while SOX17 was only related to the promoter methylation level (P=3.32e-12). The expression of SOXF genes was positively correlated with CD4(+) T cell infiltration (SOX7: P=8.32e-07, SOX17: P=4.93e-06, SOX18: P=1.61e-11). The AUC for cg07660671 site of SOX7, cg15377283 site of SOX17, and cg24199599 site of SOX18 in distinguishing between normal and tumor in LUAD, intestinal cancer, and breast cancer reached 0.9. SOXF genes were mainly involved in transcriptional regulation, and the Wnt signaling pathway and low expression of SOXF genes in tumor tissue had a strong negative correlation with tumor hypoxia (correlation: −0.35, P≤0.001). CONCLUSIONS: This study implied that the expression of SOX7 and SOX17 are potential prognosis markers for patients with Lung cancer and the SOXF genes methylation is potential biomarkers for pan-cancer screening. The SOX7 and SOX17 might modulate the Wnt signaling pathway and the expression of SOXF family genes was significantly negatively correlated with tumor hypoxia.

Since the beginning of this century, three types of coronavirus have widely transmitted and caused severe diseases and deaths, which strongly indicates that severe infectious diseases caused by coronavirus infection are not accidental events. Coronavirus-infected diseases are mainly manifested by respiratory symptoms, with multiple organ dysfunctions. Precisely investigating the pathological process, characteristics and pathogenesis of coronavirus-infected diseases will be beneficial for us to understand clinical manifestations and provide targeted suggestions on prophylaxis and treatment. This paper briefly reviews the pathological findings of three known coronavirus-infected diseases, and attempts to construct the pathological spectrum of coronavirus-infected diseases, aiming to provide reference and thinking for autopsy, histopathological examination and animal infection model study of coronavirus-infected diseases.21世纪以来，已经有3种冠状病毒在人群间广泛传播并造成严重的疾病与死亡，强烈提示冠状病毒感染引发的高传染性疾病并非偶发事件。冠状病毒感染性疾病以呼吸系统症状为主要表现，但同时存在多器官功能障碍。精准探明冠状病毒感染性疾病的病理过程、特征和发生机制，有助于认识其临床特征，为探寻针对性的防治策略提供病理学依据。本文简要回顾已知的冠状病毒感染性疾病的病理学研究发现，尝试构建冠状病毒感染性疾病的病变谱，旨在为开展冠状病毒感染性疾病的尸体解剖、组织病理学检验与动物感染模型研究等提供借鉴与思考。.

Two-dimensional (2D) molybdenum diselenide (MoSe2) as one of the ultrathin transition metal dichalcogenides (TMDs) has attracted considerable attention because of its potential applications in thermoelectric and nano-electronic devices. Here, the thermal conductivity of monolayer MoSe2 and its responses to simulated size and defects are studied by nonequilibrium molecular dynamics simulations. With the increase of sample length, the thermal conductivity of monolayer MoSe2 nanoribbons exhibits an enhancement whereas it is insensitive to the width. At room temperature, the thermal conductivities of monolayer MoSe2 along armchair and zigzag directions are 17.758 and 18.932 W (m K)−1, respectively, which are consistent with previous results. The impact of defects on thermal conductivity has also been studied by varying the concentration of the vacancy from 0.1% to 0.5%. The results show that an increase of the defect concentration will greatly suppress the thermal conductivity. The 0.5% defect concentration with a Mo vacancy can result in a thermal conductivity reduction of ∼43%. Such a study would provide a good insight into the tunable thermal transport for potential applications of not only monolayer MoSe2, but also many other TMDs.

Rapid and personalized single-cell drug screening testing plays an essential role in acute myeloid leukemia drug combination chemotherapy. Conventional chemotherapeutic drug screening is a time-consuming process because of the natural resistance of cell membranes to drugs, and there are still great challenges related to using technologies that change membrane permeability such as sonoporation in high-throughput and precise single-cell drug screening with minimal damage. In this study, we proposed an acoustic streaming-based non-invasive single-cell drug screening acceleration method, using high-frequency acoustic waves (>10 MHz) in a concentration gradient microfluidic device. High-frequency acoustics leads to increased difficulties in inducing cavitation and generates acoustic streaming around each single cell. Therefore, single-cell membrane permeability is non-invasively increased by the acoustic pressure and acoustic streaming-induced shear force, which significantly improves the drug uptake process. In the experiment, single human myeloid leukemia mononuclear (THP-1) cells were trapped by triangle cell traps in concentration gradient chips with different cytarabine (Ara-C) drug concentrations. Due to this dual acoustic effect, the drugs affect cell viability in less than 30 min, which is faster than traditional methods (usually more than 24 h). This dual acoustic effect-based drug delivery strategy has the potential to save time and reduce the cost of drug screening, when combined with microfluidic technology for multi-concentration drug screening. This strategy offers enormous potential for use in multiple drug screening or efficient drug combination screening in individualized/personalized treatments, which can greatly improve efficiency and reduce costs.

To explore the effects of MTA, iRoot SP and AH Plus on periodontal ligament stem cells.The periodontal ligament stem cells were cloned by limiting dilution culture method. The effects of MTA, iRoot SP and AH Plus on proliferation and apoptosis of periodontal ligament stem cells were detected by MTT and Annexin-V-FITC/PI double staining. Alizarin and qRT-PCR were used to evaluate the effect of MTA, iRoot SP and AH plus on osteogenesis of periodontal ligament stem cells. SPSS 21.0 software package was used for statistical analysis.MTA showed mild toxicity at 24 and 48 hours, AH Plus showed mild toxicity at 24 h. iRoot SP was the least (P0.05) compared to MTA and AH Plus. The effect of three kinds of materials on apoptosis of periodontal ligament stem cells gradually decreased with the prolongation of time. Compared with the control group, the three kinds of materials were toxic at 3 d, the toxicity of MTA was the strongest and the toxicity of iRoot SP was the lowest(P0.05). Mineralization nodules in MTA and iRoot SP group were significantly higher than those in AH Plus and control group. The expression of OC, RUNX2, COL1A and ALP gene was higher at 7, 14, 21 d than in the control group and the expression of iRoot SP mineralization was the greatest(P0.05).The hardened iRoot SP is non-toxic to human periodontal ligament stem cells. Osteogenic ability and mineralization capacity of hardened iRoot SP on human periodontal ligament stem cells are better than MTA.

Bilayer transition metal dichalcogenide heterostructures obtained by vertical stacking have attracted considerable attention because of their potential applications in thermoelectric and optoelectronics devices. The thermal transport behavior plays a pivotal role in assessing their functional performance. Here, we systematically investigate the thermal transport properties of the MoS2/MoSe2 bilayer heterostructure (MoS2/MoSe2-BH) by combining first-principles calculations and Boltzmann transport theory (BTE). The results show that the thermal conductivity of MoS2/MoSe2-BH at room temperature is 25.39 W m−1 K−1, which is in-between those of monolayer MoSe2 and MoS2. According to our calculated orbital-resolved phonon dispersion curves, Gruneisen parameters, phonon group velocity and relaxation time, we find that the acoustic and low-frequency optical branches below 172.65 cm−1 have strong coupling and contribute mainly to the lattice thermal conductivity. Compared with free standing monolayer MoS2 and MoSe2, the lattice thermal conductivity of MoS2/MoSe2-BH is influenced by the weak van der Waals interlayer interactions.

To investigate correlation between abnormal replicative senescence of endometrial gland epithelial cells (EGECs) in shedding and non-shedding endometria and endometriosis cyst during menstruation. Musashi-1 expression, β-catenin expression, and EGECs ultrastructure in shedding and non-shedding endometrium when menstruation were observed through real-time PCR and transmission electron microscopy technologies. (1) Musashi-1 and β-catenin exhibited a high expression in shedding and non-shedding endometria in experimental group, showing a positive correlation between each other; and were significantly higher than that in control group. However; there was no correlation between these two in control group. (2) Transmission electron microscopy results: In experimental group, organelles in EGECs in shedding endometrium obtained were abundant on the first day of menstruation, nuclei were irregular, double nucleoli could be observed, and chromatin was rich. Furthermore, morphology of EGECs in non-shedding endometrium was irregular, organelles were abundant, basement membrane was irregular with abnormal curvature, and a large amount of collagenic fibers were found in intercellular spaces. On the fifth day of menstruation, the cilia and microvilli on secretory cells in endometrium increased and prolongated, and organelles became extremely rich. EGECs have potentials of division, proliferation, invasion and migration; and is associated with formation of endometriosis cysts.摘要 本研究目的是探究月经期间脱落和未脱落子宫内膜腺上皮细胞（endometrial gland epithelial cells, EGECs）的异常复制衰老与子宫内膜异位囊肿的相关性。通过实时PCR和透射电子显微镜技术观察月经期间脱落和未脱落子宫内膜中Musashi-1和β-catenin的表达以及EGECs的超微结构。（1）Musashi-1和β-catenin在实验组脱落和未脱落子宫内膜中存在高表达, 两者呈正相关, 并且显著高于对照组。然而, 在对照组中这两者之间无相关性。（2）透射电镜结果：实验组在月经第一天获得的脱落子宫内膜中EGECs所含的细胞器丰富, 细胞核不规则, 可见双核仁, 染色质丰富。此外, 在未脱落的子宫内膜中EGECs形态不规则, 细胞器丰富, 基底膜不规则, 曲率异常, 细胞间隙存在大量胶原纤维。在月经第5天, 子宫内膜分泌细胞上的纤毛和微绒毛增多并延长, 细胞器变得极为丰富。EGECs具有分裂、增殖、侵袭和迁移的潜力, 并且与子宫内膜异位囊肿的形成有关。.

Compound eyes are ubiquitous natural biosensors that possess high temporal resolution and large fields of view (FOVs). While for solid materials based artificial imaging systems, flexible zooming ability while keeping the constant FOV is still challenging, as well as the low-cost fabrication. Herein, liquid compound eyes with natural structures are presented that synthesize optofluidics and bionics in a non-trivial manner, which enables the deformation-free zooming and flexible cell fluorescence sensing. Experimental results indicate that the innovatively manufactured bionic template possesses low roughness and uniform lens configuration with more than two thousands units, which endows the eyes with high-quality and low aberration imaging ability. Besides, digital controlled miscible liquids switching enables the focus of ommatidia simultaneously be adjusted from 150 μm to 5 mm with 100° view angle, and without bending the microlens curvature, to avoid FOV changing and image aberration. Due to large FOV and tunable ability, large-area cell fluorescence signal arrays and dynamic cell motion are imaged using this liquid compound eyes. This work presents novel strategy for compound lens manufacture at low-cost, and proposes deformation-free and continuous focus-tuning strategy, offering potentials for numerous applications, including biomedical sensing and adaptive imaging with large FOV.

The first-principles projector augmented-wave (PAW) method within the framework of density functional theory (DFT) is applied to study the equilibrium lattice parameters, mechanical stability, tensile and shear properties, lattice dynamical stability and vibration modes of the Th-Sn system. The optimized equilibrium lattice parameters and equilibrium cell volume reproduce perfectly the available experimental data. The individual elastic constants and polycrystalline aggregates of Pm-3m-ThSn3, Cmcm-ThSn2, P63/mcm-Th5Sn3, and P63/mcm-Th5Sn4 have been obtained, the results show that four Th-Sn systems are mechanically stable according to the elastic stability criteria, also, the calculated elastic constants have allowed us to evaluate their elastic anisotropy. Additionally, the Density Functional Perturbation Theory (DFPT) is used to determine the lattice dynamical stability and phonon modes of the series of the Th-Sn compounds, by the vibrational frequencies, it is predicted that four Th-Sn systems are also dynamically stable. Pm-3m-ThSn3 is mechanical and dynamical stable up to 35GPa. The microscopic symmetry properties for optical modes at the center of the Brillouin zone of ThSn3, ThSn2, Th5Sn3, and Th5Sn4 have also been estimated by using group theory.

MicroRNAs are known to be important in a variety of cancer types. The specific expression and roles of miR-338-3p in the context of gastric cancer, however, remains largely unknown. In this study, we found that miR-338-3p was expressed significantly lower in established/primary human gastric cancer cells than that in human gastric epithelial cells; miR-338-3p is also decreased in human gastric cancer tissues and was positively associated with the worse prognosis of patients with gastric cancer. Enforced expression of miR-338-3p could inhibit cell growth, survival, and proliferation, while inducing cell apoptosis. In addition, miR-338-3p negatively regulated SOX5 expression through directly binding to the 3′-untranslated region of SOX5, and an inverse correlation was found between miR-338-3p and SOX5 messenger RNA expression in gastric cancer tissues. Furthermore, miR-338-3p-induced inactivation of Wnt/β-catenin signaling was greatly abrogated by SOX5 upregulation. Finally, we found that hypoxic conditions were linked with reduced miR-338-3p expression in the context of gastric cancer. In conclusion, miR-338-3p acts as a tumor suppressor in gastric cancer, possibly by directly targeting SOX5 and blocking Wnt/β-catenin signaling. These findings might provide novel therapeutic targets for gastric cancer.

The chemometric second-order multivariate calibration method was applied to the qualitative and quantitative analysis of seven phenolic acids in honey samples with five different floral origins. First, based on the “second-order advantage,” reasonable resolutions and satisfactory quantitative results were obtained even in the presence of strong background interference and overlapping peaks in complex real honey samples. Average spike recoveries for the seven target analytes were in the range of 89.34–118.81%, with high correlation coefficient (r ≥ 0.9997), low limit of quantification (1.00–20.20 ng mL−1) and high sensitivity (0.135–0.375 mL ng−1). Second, the quantitative data of seven target analytes were analyzed by principal component analysis (PCA) on MATLAB software to classify honey of five nectar sources. These results indicated that there is a strong association between the content of major phenolic acids and the flower sources of honey. Third, the quantitative composition-antioxidant activity relationship analysis was evaluated by partial least squares (PLS) analysis and studied to provide reliable analysis of multi-marker antioxidant data. The results enable the continued exploitation and utilization of honey from different floral origins and further contribute to honey market development and choices in available honey for consumption.

This paper focuses on the research about selecting the best college sports coaches during the last century. Firstly, we build a reasonable evaluation index system, taking basketball for instance, we choose 9 evaluation indexes which are the coaching years, rate of winning, times of attending major competitions, etc. Because of too many indexes, it brings difficulty in building judgment matrix. Therefore, using the Principal Component Analysis, we get not only a fewer of indexes which almost explain the original indexes, but also the corresponding weight coefficients. Furthermore, based on the results, we give the evaluation scores of basketball coaches, and then rank them from top to bottom. AnalyticalHierarchyProcess and Comprehensive Evaluation are integrated to test the ranking result. Finally, this evaluation model is applied to football and hockey fields to select the best coaches.

The first mixed alkali metal and alkaline earth metal compound with 3D anion framework consists of zinc-oxygen and tellurium-oxygen polyhedron, Na1.7Li0.3BaZn7(TeO6)3, has been synthesized by high-temperature solution method, which crystallizes in the centrosymmetric hexagonal space group P63/m (No. 176). The [Te6Zn13O57] units in Na1.7Li0.3BaZn7(TeO6)3 is very similar to that of in classic polyoxometalate, Keggin. The UV–vis-NIR diffuse reflectance spectrum, thermal gravimetric results, differential scanning calorimetry curves and infrared spectrum of Na1.7Li0.3BaZn7(TeO6)3 were studied. In addition, the band structures, and the partial density of states have been discussed through first-principles calculations.

We investigated the crystal structure, electronic structure, elastic stiffness constants, responses to tensile and shear deformations, lattice vibrational modes as well as vibration heat capacity and vibration entropy of the orthorhombic YSi compound using ab initio approach. The relaxed ground-state lattice constants a, b and c of YSi exhibit fair agreement with the available experimental and theoretical data. Nine independent elastic stiffness constants have been computed by increasingly varying minute strains based on “energy-strain” approach, which confirm that the orthorhombic YSi is mechanically stable. Some polycrystalline moduli involving bulk modulus, Young's modulus and shear modulus, Poisson's ratio, brittle/ductile characteristic have also been systematically derived, and YSi can be classified as brittle compound. Both three-directional Young's modulus anisotropy and bulk modulus anisotropy are calculated in detail, and the calculations indicate that the orthorhombic YSi takes on significant elastic anisotropy. We further studied the theoretical tensile and shear strengths under different loads. Also, the phonon spectra, total and partial phonon densities of states are researched by employing Density Functional Perturbation Theory (DFPT) and the finite-displacement method, and the infrared-active and Raman active vibration modes at the center of Brillouin zone are classified using the group theory. Moreover, we predicted the constant volume heat capacity of YSi under high temperature relying on the calculated phonon densities of states as well as quasi-harmonic approximation (QHA) and Debye model.

e21537 Background: Due to its non-invasiveness and advantage in overcoming tumor heterogeneity, plasma-based genomic profiling is widely used in advanced NSCLC patients for treatment guidance. However, whether the mutational profile derived from ctDNA comprehensively represents that in tumor tissue is the major concern for the application. It has been reported that a variety of factors influence the consistency of mutational profile between plasma and tissue samples. This study explores clinical features which predict the concordance between tissue and plasma samples. Methods: Paired tissue and plasma samples were collected from 79 (stage III and IV) NSCLC patients. Samples were profiled using a 520-cancer-related gene panel. Serum markers such as CEA, CA199, SCC and NSE were collected. Results: All tissue samples except for 1 (98.7%) had mutations detected from this panel. 68 plasma samples (86%) had mutations detected from this panel. The overall concordance of mutation between plasma and tissue samples was 43%; and the concordances for point mutation and copy number variations were 60.2% and 15%, respectively. The concordance for 8 classic lung cancer driver genes and druggable mutations were 77.2% and 34.2%, respectively. Next, we explore the correlation of serum biomarkers and the concordance rate. Univariate analyses revealed that CA19-9 (P < 0.001) and CEA (P = 0.01) were significantly associated with the concordance rate of actionable mutations. Using CEA > 5ng/ml as a cutoff, 43.1% patients having higher CEA level harbored an identical mutation profile between plasma and tissue vs only 20.8% patients with lower CEA level had an identical mutation profile (p = 0.01). Furthermore, 75% patients with CA 19-9 > 39U/ml harbored an identical mutation profile between plasma and tissue vs 22.8% patients with CA19-9 £ 39U/ml (P < 0.001). Combining both CEA and CA19-9 can effectively predict the concordance (p < 0.001); 85.7% of patients with CEA and CA19-9 levels above the cutoff have identical mutation profile and only 34.2% patients with either one or both below the cutoff have identical mutation profile (p < 0.001). Conclusions: We identified the clinical and molecular factors which showed promising value in predicting the consistency of mutational profile between plasma and tissue in NSCLC patients. Plasma samples of patients with high CEA ( > 5ng/ml) and CA19-91 ( > 39U/ml) can comprehensively reflect the mutation profile of tumor samples. Tissue biopsy is not necessary for such patients.
