PurposeTo review current evidence on the efficacy and safety outcomes of HIF-PHIs in chronic kidney disease (CKD) populations with an emphasize on the safety profile.MethodsA systematic search was conducted in the Medline, Embase, and Cochrane Central databases. Randomized controlled trials that had assessed the efficacy and safety of HIF-PHIs for anemia in CKD were included. The efficacy outcome included change of hemoglobin and the safety outcomes any adverse events, severe adverse events, major adverse cardiovascular events, and mortality. The qualities of studies were assessed using the Cochrane ROB tool.Results47 studies encompassing 55 RCTs for the study outcomes were included in this study. All six commercially available HIF-PHIs had direct comparisons to ESA and placebo, yet lacked direct comparisons among each other. The network analysis demonstrated all six HIF-PHIs were able to effectively elevate hemoglobin in the general CKD patients compared to placebo. All HIF-PHIs did not differ among each other in the efficacy of correcting anemia. Roxadustat and daprodustat had the largest number of reports in terms of adverse events. The overall risk of each safety outcome did not increase in comparison to erythropoiesis stimulating agent (ESA) or placebo, and did not differ among different types of HIF-PHIs.ConclusionHIF-PHIs can effectively elevate hemoglobin without causing higher risk of safety concerns in CKD patients with anemia. Further evidence from long-term studies and the ongoing post-market surveillance is necessary.

The surrounding environment of architectural heritage sites is integral to cultural heritage protection; plant landscapes play crucial roles in them. Controlling plant spaces and appearances is essential for preserving plant landscapes. A World Cultural Heritage Site, the Summer Palace has undergone multiple changes since the 1860s; restoring and protecting plant landscapes has been an ongoing research focus. However, data accuracy limitations have hindered analyses of the overall spatial characteristics of historical gardens. Here, the historical dynamics and unique landscape features of plants on the front hill of Longevity Hill (FLH) are explored, and conservation and renewal strategies are proposed. Geographic information system (GIS) and WSL Monoplotting Tool are used to identify historical plant spaces. Plant space types are classified, and their landscape characteristics are analyzed. On the basis of historical events, the historical plant spaces on the FLH can be divided into two major categories and six subcategories. The vegetation retention area (south side) was less affected, and the plant landscape along Kunming Lake was the most well-preserved. However, the vegetation-damaged area (north side) was impacted more in the western part than in the eastern part, with notable changes in spatial landscape characteristics, particularly regarding forest function, morphology, and structure. Strategies are proposed for reducing human intervention and adjusting retention areas; furthermore, historical images and spatial grading in damaged areas can be used to suggest landscape adjustment and restoration strategies. This study introduces a method for analyzing the historical characteristics of plant landscapes over time that can be used to protect cultural heritage sites worldwide.

Abstract An extended (3+1)-dimensional Kadomtsev–Petviashvili–Boussinesq equation is studied in this paper to construct periodic solution, n-soliton solution and folded localized excitation. Firstly, with the help of the Hirota’s bilinear method and ansatz, some periodic solutions have been derived. Secondly, taking Burgers equation as an auxiliary function, we have obtained n-soliton solution and n-shock wave. Lastly, we present a new variable separation method for (3+1)-dimensional and higher dimensional models, and use it to derive localized excitation solutions. To be specific, we have constructed various novel structures and discussed the interaction dynamics of folded solitary waves. Compared with the other methods, the variable separation solutions obtained in this paper not only directly give the analytical form of the solution u instead of its potential $$u_y$$ u y , but also provide us a straightforward approach to construct localized excitation for higher order dimensional nonlinear partial differential equation.

Background: A cytokine storm (CS) is a rapidly occurring, complex, and highly lethal systemic acute inflammatory response induced by pathogens and other factors. Currently, no clinical therapeutic drugs are available with a significant effect and minimal side effects. Given the pathogenesis of CS, natural products have become important resources for bioactive agents in the discovery of anti-CS drugs. Purpose: This study aimed to provide guidance for preventing and treating CS-related diseases by reviewing the natural products identified to inhibit CS in recent years. Methods: A comprehensive literature review was conducted on CS and natural products, utilizing databases such as PubMed and Web of Science. The quality of the studies was evaluated and summarized for further analysis. Results: This study summarized more than 30 types of natural products, including 9 classes of flavonoids, phenols, and terpenoids, among others. In vivo and in vitro experiments demonstrated that these natural products could effectively inhibit CS via nuclear factor kappa-B, mitogen-activated protein kinase, and Mammalian target of rapamycin (mTOR) signaling pathways. Moreover, the enzyme inhibition assays revealed that more than 20 chemical components had the potential to inhibit ACE2, 3CL-protease, and papain-like protease activity. The experimental results were obtained using advanced technologies such as biochips and omics. Conclusions: Various natural compounds in traditional Chinese medicine (TCM) extracts could directly or indirectly inhibit CS occurrence, potentially serving as effective drugs for treating CS-related diseases. This study may guide further exploration of the therapeutic effects and biochemical mechanisms of natural products on CS.

The genus Ogcogaster Westwood, 1847, which is endemic to the Oriental Region and contains only five species, has been recorded in India and Pakistan, but not in China. The genus was not sampled in any previous study on the phylogeny of Neuroptera and its affinity within Ascalaphinae is unclear.The owlfly species Ogcogaster segmentator (Westwood, 1847) is firstly recorded from China, based on a female specimen collected from Yunnan Province, which represents the first record of the genus Ogcogaster from China. The complete mitochondrial genome of this species is first sequenced and described. The phylogenetic analysis, based on all 13 PCGs and two rRNA genes of the owlfly mitogenomes determined so far, assigned O. segmentator into a monophyletic group with Libelloides macaronius (Scopoli, 1763) and Ascalohybris subjacens (Walker, 1853).

The option is an important derivative tool in financial market, and after decades of development, the option has emerged in various forms. This paper studies an exotic option with a proactive investment strategy. Compared with the classic option theory, we assume that the option holders continuously trade the underlying assets according to a predetermined investment strategy. Taking the European call option as an example, we first give some assumptions and define the loss function in terms of a logarithmic investment strategy. Then, the specific pricing expression of the exotic option is derived from the Black-Scholes option pricing formula. Next, numerical simulations are presented to visualize the mechanism of the exotic option in 2D and 3D forms by selecting appropriate parameters. The results indicate that the exotic option has a significant price advantage (up to 43.9% under specific parameter settings) over the classic option. Moreover, the empirical results illustrate a perfect fit between 50 ETF (issued by the Shanghai Stock Exchange) and our exotic option. The new proposed exotic option extends the Black-Scholes option theory from a no-trading condition (do not buy or sell underlying assets during the validity period) to a dynamic investment condition and has important practical significance in real life.

BackgroundAerobic glycolysis is a process that metabolizes glucose under aerobic conditions, finally producing pyruvate, lactic acid, and ATP for tumor cells. Nevertheless, the overall significance of glycolysis-related genes in colorectal cancer and how they affect the immune microenvironment have not been investigated.MethodsBy combining the transcriptome and single-cell analysis, we summarize the various expression patterns of glycolysis-related genes in colorectal cancer. Three glycolysis-associated clusters (GAC) were identified with distinct clinical, genomic, and tumor microenvironment (TME). By mapping GAC to single-cell RNA sequencing analysis (scRNA-seq), we next discovered that the immune infiltration profile of GACs was similar to that of bulk RNA sequencing analysis (bulk RNA-seq). In order to determine the kind of GAC for each sample, we developed the GAC predictor using markers of single cells and GACs that were most pertinent to clinical prognostic indications. Additionally, potential drugs for each GAC were discovered using different algorithms.ResultsGAC1 was comparable to the immune-desert type, with a low mutation probability and a relatively general prognosis; GAC2 was more likely to be immune-inflamed/excluded, with more immunosuppressive cells and stromal components, which also carried the risk of the poorest prognosis; Similar to the immune-activated type, GAC3 had a high mutation rate, more active immune cells, and excellent therapeutic potential.ConclusionIn conclusion, we combined transcriptome and single-cell data to identify new molecular subtypes using glycolysis-related genes in colorectal cancer based on machine-learning methods, which provided therapeutic direction for colorectal patients.

Background: Lysosomes are essential for the development and recurrence of cancer. The relationship between a single lysosome-related gene and cancer has previously been studied, but the relationship between the lysosome-related genes (LRGs) and colon adenocarcinoma (COAD) remains unknown. This research examined the role of lysosome-related genes in colon adenocarcinoma.Methods: 28 lysosome-related genes associated with prognosis (PLRGs) were found by fusing the gene set that is differently expressed between tumor and non-tumor in colon adenocarcinoma with the gene set that is related to lysosomes. Using consensus unsupervised clustering of PLRGs, the colon adenocarcinoma cohort was divided into two subtypes. Prognostic and tumor microenvironment (TME) comparisons between the two subtypes were then made. The PLRGs_score was constructed using the least absolute shrinkage and selection operator regression (LASSO) method to quantify each patient’s prognosis and provide advice for treatment. Lastly, Western Blot and immunohistochemistry (IHC) were used to identify MOGS expression at the protein level in colon adenocarcinoma tissues.Results: PLRGs had more somatic mutations and changes in genetic level, and the outcomes of the two subtypes differed significantly in terms of prognosis, tumor microenvironment, and enrichment pathways. Then, PLRGs_score was established based on two clusters of differential genes in the cancer genome atlas (TCGA) database, and external verification was performed using the gene expression omnibus (GEO) database. Then, we developed a highly accurate nomogram to enhance the clinical applicability of the PLRGs_score. Finally, a higher PLRGs_score was associated with a poorer overall survival (OS), a lower tumor mutation burden (TMB), a lower cancer stem cell (CSC) index, more microsatellite stability (MSS), and a higher clinical stage. MOGS was substantially elevated at the protein level in colon adenocarcinoma as additional confirmation.Conclusion: Overall, based on PLRGs, we identified two subtypes that varied significantly in terms of prognosis and tumor microenvironment. Then, in order to forecast patient prognosis and make treatment suggestions, we developed a diagnostic model with major significance for prognosis, clinical relevance, and immunotherapy. Moreover, we were the first to demonstrate that MOGS is highly expressed in colon adenocarcinoma.

ObjectiveTo compare the performance of a deep learning survival network with the tumor, node, and metastasis (TNM) staging system in survival prediction and test the reliability of individual treatment recommendations provided by the network.MethodsIn this population-based cohort study, we developed and validated a deep learning survival model using consecutive cases of newly diagnosed stage I to IV esophageal cancer between January 2004 and December 2015 in a Surveillance, Epidemiology, and End Results (SEER) database. The model was externally validated in an independent cohort from Fujian Provincial Hospital. The C statistic was used to compare the performance of the deep learning survival model and TNM staging system. Two other deep learning risk prediction models were trained for treatment recommendations. A Kaplan–Meier survival curve was used to compare survival between the population that followed the recommended therapy and those who did not.ResultsA total of 9069 patients were included in this study. The deep learning network showed more promising results in predicting esophageal cancer-specific survival than the TNM stage in the internal test dataset (C-index=0.753 vs. 0.638) and external validation dataset (C-index=0.687 vs. 0.643). The population who received the recommended treatments had superior survival compared to those who did not, based on the internal test dataset (hazard ratio, 0.753; 95% CI, 0.556-0.987; P=0.042) and the external validation dataset (hazard ratio, 0.633; 95% CI, 0.459-0.834; P=0.0003).ConclusionDeep learning neural networks have potential advantages over traditional linear models in prognostic assessment and treatment recommendations. This novel analytical approach may provide reliable information on individual survival and treatment recommendations for patients with esophageal cancer.

Amino acid metabolization is verified to be a part in the progression of cancer. However, genes related to the amino acid metabolism have not been identified in colon adenocarcinoma (COAD). A systematic prognostic model of COAD becomes a pressing need. Among genes related to the amino acid metabolism, RIMKLB, ASPG, TH, MTAP, AZIN2, PSMB2, HDC, ACMSD, and PSMA8 were identified to construct a risk model. Kaplan–Meier (K–M) analyses demonstrated that the high-risk group achieved a poor prognosis. Area under the respective ROC (AUC) values indicated the robustness of the model. To highlight its clinical value, multivariate Cox was used to obtain the optimal variables to construct a nomogram. A higher tumor mutation burden was observed in the high-risk group. However, the low-risk group had a stronger immune infiltration. Seven molecular subtypes were found by consensus cluster. Twenty-two hub genes were identified related to the ESTIMATE score using WGCNA. In brief, our research constructed a stable prognostic model related to the amino acid metabolism in COAD, revealing its connection to the immune microenvironment. The model guided the outcome of COAD and the direction of immunotherapy.

In this study, the variation of fatigue stiffness, fatigue life, and residual strength, as well as the macroscopic damage initiation, expansion, and fracture of CFRP (carbon fiber reinforced polymer) rods in bending-anchored CFRP cable, were investigated experimentally to verify the anchoring performance of the bending anchoring system and evaluate the additional shear effect caused by bending anchoring. Additionally, the acoustic emission technique was used to monitor the progression of critical microscopic damage to CFRP rods in a bending anchoring system, which is closely related to the compression-shear fracture of CFRP rods within the anchor. The experimental results indicate that after the fatigue cycles of two million, the residual strength retention rate of CFRP rod was as high as 95.1% and 76.7% under the stress amplitudes of 500 MPa and 600 MPa, indicating good fatigue resistance. Moreover, the bending-anchored CFRP cable could withstand 2 million cycles of fatigue loading with a maximum stress of 0.4 σult and an amplitude of 500 MPa without obvious fatigue damage. Moreover, under more severe fatigue-loading conditions, it can be found that fiber splitting in CFRP rods in the free section of cable and compression-shear fracture of CFRP rods are the predominant macroscopic damage modes, and the spatial distribution of macroscopic fatigue damage of CFRP rods reveals that the additional shear effect has become the determining factor in the fatigue resistance of the cable. This study demonstrates the good fatigue-bearing capacity of CFRP cable with a bending anchoring system, and the findings can be used for the optimization of the bending anchoring system to further enhance its fatigue resistance, which further promotes the application and development of CFRP cable and bending anchoring system in bridge structures.

This paper presents a multiplexable fiber optic chemical sensor with the capability of monitoring hydrogen gas concentration at high temperatures up to 750 °C. The Pd-nanoparticle infused TiO2 films coated on intrinsic Fabry–Perot interferometer (IFPI) array were used as sensory films. Strains induced upon exposure to hydrogen with varied concentrations can be monitored by IFPI sensors. The fiber sensor shows a repetitive and reversible response when exposed to a low level (1–6%) of hydrogen gas. Uniform sensory behavior across all the sensing cavities is demonstrated and reported in this paper.

Nitrate Transporter 1/Peptide Transporter Family (NPF) genes encode membrane transporters involved in the transport of diverse substrates. However, little is known about the diversity and functions of NPFs in Brassica rapa. In this study, 85 NPFs were identified in B. rapa (BrNPFs) which comprised eight subfamilies. Gene structure and conserved motif analysis suggested that BrNFPs were conserved throughout the genus. Stress and hormone-responsive cis-acting elements and transcription factor binding sites were identified in BrNPF promoters. Syntenic analysis suggested that tandem duplication contributed to the expansion of BrNPFs in B. rapa. Transcriptomic profiling analysis indicated that BrNPF2.6, BrNPF2.15, BrNPF7.6, and BrNPF8.9 were expressed in fertile floral buds, suggesting important roles in pollen development. Thirty-nine BrNPFs were responsive to low nitrate availability in shoots or roots. BrNPF2.10, BrNPF2.19, BrNPF2.3, BrNPF5.12, BrNPF5.16, BrNPF5.8, and BrNPF6.3 were only up-regulated in roots under low nitrate conditions, indicating that they play positive roles in nitrate absorption. Furthermore, many genes were identified in contrasting genotypes that responded to vernalization and clubroot disease. Our results increase understanding of BrNPFs as candidate genes for genetic improvement studies of B. rapa to promote low nitrate availability tolerance and for generating sterile male lines based on gene editing methods.

Ocean mesoscale eddies are ubiquitous in world ocean and account for 90% oceanic kinetic energy, which dominate the upper ocean flow field. Accurately predicting the variation of ocean mesoscale eddies is the key to understand the oceanic flow field and circulation system. In this article, we propose to make an initial attempt to explore spatio-temporal predictability of mesoscale eddies, employing deep learning architecture, which primarily establishes Memory In Memory (MIM) for sea level anomaly (SLA) prediction, combined with the existing mesoscale eddy detection. Oriented to the western Pacific ocean (125°−137.5°E and 15°−27.5°N), we quantitatively investigate the historic daily SLA variability at a 0.25° spatial resolution from 2000 to 2018, derived by satellite altimetry. We develop the enhanced MIM prediction strategies, equipped with Gated Recurrent Unit (GRU) and spatial attention module, in a scheduled sampling manner, which overcomes the gradient vanishing and complements to strengthen spatio-temporal features for long-term dependencies. At the early stage, the real value SLA input guides the model training process for initialization, while the scheduled sampling intentionally feeds the newly predicted value, to resolve the distribution inconsistency of inference. It has been demonstrated in our experiment results that our proposed prediction scheme outperformed the state-of-art approaches for SLA time series, with MAPE, RMSE of the 14-day prediction duration, respectively, 5.1%, 0.023 m on average, even up to 4.6%, 0.018 m for the effective sub-regions, compared to 19.8%, 0.086 m in ConvLSTM and 8.3%, 0.040 m in original MIM, which greatly facilitated the mesoscale eddy prediction. This proposed scheme will be beneficial to understand of the underlying dynamical mechanism behind the predictability of mesoscale eddies in the future, and help the deployment of ARGO, glider, AUV and other observational platforms.

BackgroundGrowing evidence demonstrates that the initiation and progression of colorectal carcinoma (CRC) is related to the presence of cancer stem cells (CSCs). However, the mechanism through which the stem cell features of CRC cells are maintained is poorly understood. In this study, we identified the oncogenic histone cluster 2 H2B family member F (HIST2H2BF) and aimed to investigate the function of upregulated HIST2H2BF expression in maintaining the stem cell features of CRC cells, which accelerate the progression of CRC.MethodsHIST2H2BF expression was quantified using real-time polymerase chain reaction, immunohistochemistry, and western blotting. The correlation between CpG island methylation status and HIST2H2BF re-expression was assessed through bisulfite sequencing polymerase chain reaction, methylation-specific polymerase chain reaction, and 5-Aza-dC treatment. Functional assays were performed on CRC cells and mice to investigate the HIST2H2BF-induced stem cell-like and cancer properties of CRC. Using the Notch pathway inhibitor FLI-06, the regulatory effect of HIST2H2BF on downstream Notch signaling was confirmed.ResultsHIST2H2BF was highly expressed in CRC tissues and cell lines. The reactivation of HIST2H2BF in CRC stems at least in part from the hypomethylated CpG islands. CRC patients with high HIST2H2BF expression have poor survival outcomes. Functional studies have shown that HIST2H2BF promotes CSC phenotype, malignancy, and liver metastasis through the activation of Notch signaling in CRC. Blockage of the Notch pathway reduced the stem cell-like and cancer properties.ConclusionOur study suggests that HIST2H2BF upregulation enhances the CSC phenotype, malignancy, and liver metastasis through the activation of Notch signaling in CRC. These results identified a new perspective on the mechanism by which the stem cell features of CRC cells are maintained and highlighted the potential novel therapeutic targets for CRC.

Macrophages can be polarized into classically activated macrophages (M1) and alternatively activated macrophages (M2) in the immune system, performing pro-inflammatory and anti-inflammatory functions, respectively. Human THP-1 and mouse RAW264.7 cell line models have been widely used in various macrophage-associated studies, while the similarities and differences in protein expression profiles between the two macrophage models are still largely unclear. In this study, the protein expression profiles of M1 and M2 phenotypes from both THP-1 and RAW264.7 macrophages were systematically investigated using mass spectrometry-based proteomics. By quantitatively analyzing more than 5,000 proteins among different types of macrophages (M0, M1 and M2) from both cell lines, we identified a list of proteins that were uniquely up-regulated in each macrophage type and further confirmed 43 proteins that were commonly up-regulated in M1 macrophages of both cell lines. These results revealed considerable divergences of each polarization type between THP-1 and RAW264.7 macrophages. Moreover, the mRNA and protein expression of CMPK2, RSAD2, DDX58, and DHX58 were strongly up-regulated in M1 macrophages for both macrophage models. These data can serve as important resources for further studies of macrophage-associated diseases in experimental pathology using human and mouse cell line models.

The complex in which scribble planar cell polarity protein (SCRIB) is located is one of the three main polar protein complexes that play an important role in maintaining epithelial polarity and affecting tumour growth. However, the role of SCRIB in colorectal cancer (CRC) remains largely unknown. This study used date from The Cancer Genome Atlas (TCGA) and clinical samples to determine the expression of SCRIB in CRC and explored its mechanism through bioinformatics analysis and in vivo and in vitro experiments. In this study, SCRIB was found to be highly expressed in CRC patients, and it was often associated with malignant characteristics, such as proliferation, apoptosis, and epithelial-mesenchymal transition (EMT). Furthermore, we found that SCRIB may interact with the Hippo signalling pathway and affect the phosphorylation of YAP and its distribution inside and outside of the nucleus. We concluded that increased expression of SCRIB is likely to inhibit the Hippo signalling pathway by promoting YAP phosphorylation. This role of SCRIB in the progression of CRC provides an important information for the treatment of CRC.

Abstract Background A suitable reference gene is an important prerequisite for guarantying accurate and reliable results in quantitative real-time PCR (qRT-PCR) analyses. However, there is no absolute universality in reference genes among different species. It’s hard to find an ideal reference gene to fit for different tissues and growth periods. Pitaya (Hylocereus) is commercially produced as a new fruit crop at a large scale in tropical and subtropical regions. To date, there is no report on the identification of the most reliable reference genes for qRT-PCR normalization in pitaya. Results In this study, six candidate reference genes i.e. Actin(1), GAPDH, UBC(1), UBC(2) EF1-α(1) and histone(1) were selected from thirty-nine typical candidate reference genes to determine the most stable reference genes for qRT-PCR normalization in different tissues, temperature stresses and fruit developmental stages of pitaya. Among the six candidate reference genes, Actin(1) and EF1-α(1) were the most stable gene according to calculations of three statistical methods (GeNorm, NormFinder and BestKeeper) while UBC(1) and UBC(2) showed the lowest expression stability. The six candidate reference genes were further validated by comparing expression profiles of key genes related to betalain biosynthesis at flesh coloration stages of Guanhuahong (Hylocereus monacanthus) and Guanhuabai (H. undatus) pitayas. Actin(1) was recommended the best reference gene for accurate normalization of qRT-PCR data. Conclusions In this study, the stability of the selected reference genes for normalizing the qRT-PCR data were identified from pitaya. Actin(1) was the most stably expressed genes in different tissues and fruit developmental stages in pitaya. The present work provides the first data of reference gene identification for pitaya and will facilitate further studies in molecular biology and gene function on Hylocereus and other closely related species.

When the thermoplastic composites reach the service limits during the service, the recovery and utilization are the key concerns. Meanwhile, the improvement of strength, toughness and durability of epoxy resin is the effective method to prolong the service life of materials and structures. In the present paper, three kinds of thermoplastic resins (polypropylene-PP, polyamide 6-PA6 and polyether-ether-ketone-PEEK) and composites (carbon fiber-PEEK, glass fiber-PA6 and glass fiber-PP) were adopted as the fillers to reinforce and toughen the epoxy resin (Ts). The mechanical, thermal and microscopic analysis were conducted to reveal the performance improvement mechanism of Ts. It can be found that adding thermoplastic resin and composite fillers at the low mass ratio of 0.5~1.0% brought about the maximum improvement of tensile strength (7~15%), flexural strength (7~15%) and shear strength (20~30%) of Ts resin. The improvement mechanism was because the addition of thermoplastic fillers can prolong the cracking path and delay the failure process through the load bearing of fiber, energy absorption of thermoplastic resin and superior interface bonding. In addition, the thermoplastic composite had better enhancement effect on the mechanical/thermal properties of Ts resin compared to thermoplastic resin. When the mass ratio was increased to 2.0~3.0%, the agglomeration and stress concentration of thermoplastic filler in Ts resin appeared, leading to the decrease of mechanical and thermal properties. The optimal addition ratios of thermoplastic resin were 0.5~1.0% (PEEK), 1.0~2.0% (PA6) and 0.5~1.0% (PP) to obtain the desirable property improvement. In contrast, the optimal mass ratios of three kinds of composite were determined to be 0.5~1.0%. Application prospect analysis indicated adding the thermoplastic resin and composite fillers to Ts resin can promote the recycling and reutilization of thermoplastic composites and improve the performance of Ts resin, which can be used as the resin matrix, interface adhesive and anti-corrosion coating.

Considering little literature investigate the influence of haze on humans in psychology field and the increasing frequency of haze hitting China, and its remarkably adverse impacts on society, this research uses two studies to explore the mixed effect of haze on donation behavior, and aims to make contribution to current literature and provide insights to haze issues. Study 1: 110 participants were included into a weather information survey in which half of them were instructed to read haze weather information, and the other half were assigned to good weather information condition. After reading and recalling experiences under the same weather condition, all participants were displayed and asked to report their attitudes on a donation program, including their donation intention with money and time, the amount of donating money, and the behavior measurement about whether they would like to leave their email addresses to the charity organization to keep in further connection. The results showed people in haze weather condition, compared with whom in good weather condition, were more likely to donate money and less likely to donate time to the donation program. There is a significant interaction effect between haze or not and donation type on donation intention. We did not find effect of haze on the amount of donation and donating behavior. Study 2: 101 participants were randomly assigned to haze weather condition or good weather condition first and then were asked to judge a donation program as study 1. After that, we measured mortality salience using three items as our mediator variable. The results showed there was a significant interaction effect between haze (vs. good) weather and donation type on donation intention which replicated the results of study 1. People in haze condition would donate more money and less time compared with people in good weather condition. Besides, we showed mortality salience was the underlying mechanism. People in haze condition perceived higher level of mortality salience, which altered their attitudes on money and time resources. Across two studies, we found convergent evidence supporting our hypotheses. Specially, haze weather can increase donation intention with money resources but decrease donation intention with time resources. This effect is mediated by mortality salience caused by haze. Based on our results, we conclude with an exploratory discussion.

Hepatocellular carcinoma (HCC) is still one of the malignant tumors with high morbidity and mortality in China and worldwide. Although alpha-fetoprotein (AFP) as well as core fucosylated AFP-L3 have been widely used as important biomarkers for HCC diagnosis and evaluation, the AFP level shows a huge variation among HCC patient populations. In addition, the AFP level has also been proved to be associated with pathological grade, progression, and survival of HCC patients. Understanding the intrinsic heterogeneities of HCC associated with AFP levels is essential for the molecular mechanism studies of HCC with different AFP levels as well as for the potential early diagnosis and personalized treatment of HCC with AFP negative. In this study, an integrated N-glycoproteomic and proteomic analysis of low and high AFP levels of HCC tumors was performed to investigate the intrinsic heterogeneities of site-specific glycosylation associated with different AFP levels of HCC. By large-scale profiling and quantifying more than 4,700 intact N-glycopeptides from 20 HCC and 20 paired paracancer samples, we identified many commonly altered site-specific N-glycans from HCC tumors regardless of AFP levels, including decreased modifications by oligo-mannose and sialylated bi-antennary glycans, and increased modifications by bisecting glycans. By relative quantifying the intact N-glycopeptides between low and high AFP tumor groups, the great heterogeneities of site-specific N-glycans between two groups of HCC tumors were also uncovered. We found that several sialylated but not core fucosylated tri-antennary glycans were uniquely increased in low AFP level of HCC tumors, while many core fucosylated bi-antennary or hybrid glycans as well as bisecting glycans were uniquely increased in high AFP tumors. The data provide a valuable resource for future HCC studies regarding the mechanism, heterogeneities and new biomarker discovery.

Background Evidences indicated that non-small-cell lung cancer (NSCLC) and small-cell lung cancer (SCLC) might originate from the same cell type, which however ended up to be two different subtypes of lung carcinoma, requiring different therapeutic regimens. We aimed to identify the differences between these two subtypes of lung cancer by using integrated proteome and genome approaches. Methods and Materials Two representative cell lines for each lung cancer subtype were comparatively analysed by quantitative proteomics, and their corresponding transcriptomics data were obtained from the Gene Expression Omnibus database. The integrated analyses of proteogenomic data were performed to determine key differentially expressed proteins that were positively correlated between proteomic and transcriptomic data. Result The proteomics analysis revealed 147 differentially expressed proteins between SCLC and NSCLC from a total of 3,970 identified proteins. Combined with available transcriptomics data, we further confirmed 14 differentially expressed proteins including six known and eight new lung cancer related proteins that were positively correlated with their transcriptomics data. These proteins are mainly involved in cell migration, proliferation, and invasion. Conclusion The proteogenomic data on both NSCLC and SCLC cell lines presented in this manuscript is complementary to existing genomic and proteomic data related to lung cancers and will be crucial for a systems biology-level understanding of the molecular mechanism of lung cancers. Abstract Post-translational methylation of histone lysine or arginine residues plays important roles in gene regulation and other physiological processes. Aberrant histone methylation caused by a gene mutation, translocation, or overexpression can often lead to initiation of a disease such as cancer. Small molecule inhibitors of such histone modifying enzymes that correct the abnormal methylation could be used as novel therapeutics for these diseases, or as chemical probes for investigation of epigenetics. Discovery and development of histone methylation modulators are in an early stage and undergo a rapid expansion in the past few years. A number of highly potent and selective compounds have been reported, together with extensive preclinical studies of their biological activity. Several compounds have been in clinical trials for safety, pharmacokinetics, and efficacy, targeting several types of cancer. This review summarizes the biochemistry, structures, and biology of cancer-relevant histone methylation modifying enzymes, small molecule inhibitors and their preclinical and clinical antitumor activities. Perspectives for targeting histone methylation for cancer therapy are also discussed.

At present, the insulation performances of electrical apparatus under lightning impulse voltages are always evaluated by the withstand test of standard lightning impulse voltage (1.2/50 µs), which is recommended by IEC Standard. However, the actual time parameters of transformer invading lightning impulse voltage present an obvious dispersibility characteristics. The standard wave shape is only representative of one particular class of lightning waves, which is far from satisfactory to act as the guidance waveform in the design of power equipment insulation. On the basis of the above research background, the relationship between breakdown characteristics of oil paper insulation and time parameters of applied lightning impulse voltage was investigated in this study. The 50% breakdown voltage characteristics of oil paper insulation under lightning impulse voltage with different time parameters were determined. The test results indicate that a short wave front time could lead to relatively low 50% breakdown voltage, while when the wave tail time varied, the 50% breakdown voltage of oil paper insulation barely changed. At last the simulation model of oil paper insulation was developed, and the current flowing through the oil paper insulating system under lightning impulse voltage was calculated. On the basis of the simulating results, the relationship between current and time parameters was analysed.

Red dragon fruit or red pitaya (Hylocereus polyrhizus) is the only edible fruit that contains betalains. The color of betalains ranges from red and violet to yellow in plants. Betalains may also serve as an important component of health-promoting and disease-preventing functional food. Currently, the biosynthetic and regulatory pathways for betalain production remain to be fully deciphered. In this study, isobaric tags for relative and absolute quantitation (iTRAQ)-based proteomic analyses were used to reveal the molecular mechanism of betalain biosynthesis in H. polyrhizus fruits at white and red pulp stages, respectively. A total of 1946 proteins were identified as the differentially expressed between the two samples, and 936 of them were significantly highly expressed at the red pulp stage of H. polyrhizus. RNA-seq and iTRAQ analyses showed that some transcripts and proteins were positively correlated; they belonged to “phenylpropanoid biosynthesis”, “tyrosine metabolism”, “flavonoid biosynthesis”, “ascorbate and aldarate metabolism”, “betalains biosynthesis” and “anthocyanin biosynthesis”. In betalains biosynthesis pathway, several proteins/enzymes such as polyphenol oxidase, CYP76AD3 and 4,5-dihydroxy-phenylalanine (DOPA) dioxygenase extradiol-like protein were identified. The present study provides a new insight into the molecular mechanism of the betalain biosynthesis at the posttranscriptional level.

The concept of chiplet was proposed in the post-Moore era. How to layout the multiple chips with different processes and sizes in the package structure is a problem that needs to be considered as different layouts may significantly affect the manufacturability during the packaging process. XDFOI-O is a 2.5D organic interposer structure with a significant coefficient of thermal expansion mismatch in it. Different layouts may cause excessive stress concentration in the package structure, as well as large wafer warpage, which can affect the normal operations of the production line. Stress accumulation on specific chiplet during the wafer thinning process is another manufacturability problem, leading to chip cracking. Prospective finite element analysis can be applied to evaluate the various layouts. In simulation work, different placement processes of dummy chips as stiffeners, as well as different chiplet thicknesses and underfill coverage, can be used as factors for simulation studies, thereby making a reference for further chiplet package design. In traditional historical research, interpreting historical documents subjectively and manually causes problems such as one-sided understanding, selective analysis, and one-way knowledge connection. In this study, we aim to use machine learning to automatically analyze and explore historical documents from a text analysis and visualization perspective. This technology solves the problem of large-scale historical data analysis that is difficult for humans to read and intuitively understand. In this study, we use the historical documents of the Qing Dynasty Hetu Dangse, preserved in the Archives of Liaoning Province, as data analysis samples. China's Hetu Dangse is the largest Qing Dynasty thematic archive with Manchu and Chinese characters in the world. Through word frequency analysis, correlation analysis, co-word clustering, word2vec model, and SVM (Support Vector Machines) algorithms, we visualize historical documents, reveal the relationships between functions of the government departments in the Shengjing area of the Qing Dynasty, achieve the automatic classification of historical archives, improve the efficient use of historical materials as well as build connections between historical knowledge.
