Abstract The Staphylococcus aureus (S. aureus) SaeRS two-component system (TCS) regulates over 20 virulence factors. While its impact on chronic infection has been thoroughly discussed, its role in the early stage of infection remains elusive. Since macrophages serve as the primary immune defenders at the onset of infection, this study investigates the influence of SaeRS on macrophage functions and elucidates the underlying mechanisms. Macrophage expression of inflammatory and chemotactic factors, phagocytosis, and bactericidal activity against S. aureus were assessed, along with the evaluation of cellular oxidative stress. SaeRS was found to impair macrophage function. Mechanistically, SaeRS inhibited NF-κB pathway activation via toll-like receptor 2 (TLR2). Its immune-modulating effect could partially be explained by the strengthened biofilm formation. More importantly, we found SaeRS compromised macrophage immune functions at early infection stages even prior to biofilm formation. These early immune evasion effects were dependent on bacterial clumping as cytokine secretion, phagocytosis, and bactericidal activity were repaired when clumping was inhibited. We speculate that the bacterial clumping-mediated antigen mask is responsible for SaeRS-mediated immune evasion at the early infection stage. In vivo, ΔsaeRS infection was cleared earlier, accompanied by early pro-inflammatory cytokines production, and increased tissue oxidative stress. Subsequently, macrophages transitioned to an anti-inflammatory state, thereby promoting tissue repair. In summary, our findings underscore the critical role of the SaeRS TCS in S. aureus pathogenicity, particularly during early infection, which is likely initiated by SaeRS-mediated bacterial clumping.

In indoor visible light communication (VLC) and visible light positioning (VLP) systems, the performance of conventional orthogonal frequency-division multiplexing (OFDM) schemes is often compromised due to the nonlinear characteristics and limited modulation bandwidth of light-emitting diodes, the multipath effect in enclosed indoor environments, and the relative positions of transmitters and receivers. This paper proposes an OFDM scheme based on the fractional Fourier transform (FRFT) to address these issues, demonstrating promising results when applied to indoor VLC and VLP systems. The FRFT, a generalization of the conventional Fourier transform (FT) in the fractional domain, captures information in both the time and frequency domains, offering greater flexibility than the FT. In this paper, we first introduce the computation method of the reality-preserving FRFT for an intensity modulation/direct detection VLC system and integrate it with OFDM to optimize system performance. By adopting FRFT-OFDM under the optimal fractional order, we enhance both the bit error ratio (BER) performance and positioning accuracy. Simulation results reveal that the FRFT-OFDM scheme with an optimized fractional order significantly improves the BER and positioning accuracy compared to the FT-OFDM scheme across most receiver positions within the indoor observation plane. For communication, the FRFT-OFDM scheme achieves over 6 dB Eb/N0 gain compared to the FT-OFDM scheme at a BER of 3×10−4 when the receiver is positioned at most locations in the room. For positioning, the FRFT-OFDM scheme enhances positioning accuracy by more than 1 cm relative to the FT-OFDM scheme at most locations in the room. Notably, both systems maintain the same computational complexity and spectral efficiency.

Drought stress is a major abiotic stress that poses serious threats to the yield and quality of rapeseed. Understanding the mechanisms underlying the drought tolerance of rapeseed is of great importance for the breeding of drought-resistant rapeseed varieties. Here, we compared the phenotypic characteristics, photosynthetic physiology, specific metabolites and related metabolic pathways between the drought-tolerant genotype (Q2) and drought-sensitive genotype (Q8) of rapeseed under drought stress using gas chromatography and liquid chromatography–mass spectrometry. The results indicated that drought stress decreased the total biomass and photosynthesis of both genotypes, particularly those of Q8, and the reduction was substantially lower in the root surface area, root volume and root diameter of Q2 than in those of Q8 under drought stress. The adverse effect on Q2 could be improved by increasing the specific leaf area, root length, and non-photochemical quenching. Furthermore, four metabolic pathways (galactose metabolism, TCA cycle, aminoacyl-tRNA biosynthesis as well as nicotinate and nicotinamide metabolisms) were greatly activated in Q2 compared with those in Q8 under drought stress. Metabolic profiling revealed 17 differential metabolites between the two genotypes, including carbohydrates, amino acids and organic acids and the increase in tagatose was accompanied by the accumulation of fumaric acid in the roots of Q2 under drought stress. The drought resistance of Q2 could be attributed to the accumulation of more metabolites, such as galactose, tagatose, glycerone and fumaric acid. Our results provide valuable insights into the growth characteristics, physiological changes and metabolic regulation mechanism of rapeseed under drought stress as well as a theoretical basis for the breeding of drought-resistant rapeseed varieties.

Classical frequency-stabilized lasers have achieved high-frequency stability and reproducibility; however, their extensive wavelength spacing limits their utility in various scenarios. This study introduces a novel frequency-stabilized laser scheme that integrates a Fabry-Perot etalon (FPE) with digital control technology and wavelength modulation techniques. The FPE, characterized by multiple transmission peaks at minimal frequency intervals, provides stable frequency references for different lasers, thereby enhancing the system’s flexibility and adaptability. An error signal is derived from the first-order differentiation of the FPE’s transmission curve. A 180° phase difference was observed in the feedback output signal when the laser’s central frequency diverged from the reference, determining that the direction of the frequency control was accordingly determined.Employing feedback control, the laser’s output frequency is stabilized at the transmission peak frequency of the FPE. Experimental results demonstrate that this stabilization scheme effectively locks the laser’s output wavelength to different transmission peak frequencies of the FPE, achieving 25 GHz wavelength spacing. The frequency stability is improved by two orders of magnitude on a second-level timescale, maintained within hundreds of kHz, equating to a frequency stability level of 10−10.

Abstract Background This study assessed the distribution characteristics of pathogens isolated from cases of orthopedic infections and focused on the antimicrobial susceptibility of the main pathogens. Methods This retrospective study involved patients with orthopedic infection in a tertiary medical center located in Shanghai, China, from 2008 to 2021.Pathogen information and the basic information of patients were identified from clinical microbiology laboratory data and the institutional medical record system. Results In total, the pathogen information of 2821 patients were enrolled in the study. S. aureus (37.71%) was the main causative pathogen responsible for orthopedic infection. A microwave photonic frequency multiplier with low phase noise based on an optoelectronic oscillator (OEO) is proposed and experimentally demonstrated. In this scheme, a dual-parallel Mach–Zehnder modulator (DPMZM) is employed to generate the third-harmonic frequency of the input radio frequency (RF) signal, while the oscillation frequency of the OEO is also three times the RF signal frequency. By adjusting the bias voltages of different arms in the DPMZM, a triple-frequency signal with a high side-mode suppression ratio of 64.8 dB can be obtained. The experimental results indicate that the output of the frequency-multiplier has a better single-sideband phase noise value, for instance, −126 dBc/Hz@10 kHz at 20.019 GHz. It has improvements of 34 dB and 43.5 dB compared with the input RF signal and the simulated electrical frequency tripler module, respectively.

The classical 1.5 μm band frequency-stabilized laser using acetylene gas saturated absorption can achieve high frequency stability and reproducibility, but its system design is complex and bulky. For some practical applications, a simple, compact system containing anti-interference abilities is preferred. In this study, a low-cost and simple-structured 1.5 μm frequency-stabilized laser is constructed using digital control methods, wavelength modulation technology, and acetylene gas absorption. The fiber input and output optical devices of the system significantly simplify the optical path and reduce the volume of the system. The error signal is obtained by the first-order differential method, and a combination of the high-speed comparator circuit and the microcontroller unit (MCU) is used to detect the error signal. Through the feedback control method of coarse temperature adjustment and fine current adjustment, the second-level frequency stability of the laser is stabilized within 100 kHz, that is, the frequency stability reaches 10​−10. The designed system achieved continuous and stable operation for more than 6 h, and the long-term frequency stability reached 10​−9.

In this paper, an optoelectronic oscillator (OEO) with ultra-low phase noise and high stability based on the injection-locked and phase-locked loop is proposed. In theory, the injection-locked frequency range of the injection signal is studied based on the phase dynamics equation, and the phase noise performance of the injection-locked OEO is analyzed. The role of the phase-locked loop on the frequency stability of the OEO is analyzed based on the phase-locked loop transfer function. In addition, this paper builds an injection-locked OEO based on a phase-locked loop. The injection-locked signal is the high-frequency output of the multiplication crystal oscillator (MCO). At the same time, this MCO synchronously outputs a low-frequency signal, which is used as the reference signal of the phase-locked loop. The experimental results show that the proposed OEO output frequency is 10 GHz, and the phase noise is −89.25 dBc/Hz@100 Hz, −121.71 dBc/Hz@1 kHz, and −145.39 dBc/Hz@10 kHz; the side-mode suppression ratio is 80 dB; the frequency stability is 2.06 × 10−11@1 s, 9.03 × 10−11@10 s, 1.03 × 10−10@100 s, and 3.03 × 10−10@1000 s. Consistent with the theoretical analysis results, the solution takes into account the frequency stability, side-mode suppression ratio, and phase noise performance. The simple structure is more advantageous in practical applications.

Based on astrometric measurements and spectral analysis from Gaia DR3, two quiescent black hole (BH) binaries, Gaia BH1 and BH2, have been identified. Their origins remain controversial, particularly for Gaia BH1. By considering a rapidly rotating ( ω / ω _crit = 0.8) and strongly magnetized ( B _0 = 5000 G) merger product, we find that, at typical Galactic metallicity, the merger product can undergo efficient chemically homogeneous evolution. This results in the merger product having a significantly smaller radius during its evolution compared to that of a normally evolving massive star. Under the condition that the initial triple stability is satisfied, we use the Multiple Stellar Evolution code and the MESA code to identify an initial hierarchical triple that can evolve into Gaia BH1. It initially consists of three stars with masses of 9.03 M _⊙ , 3.12 M _⊙ , and 1 M _⊙ , with inner and outer orbital periods of 2.21 days and 121.92 days, and inner and outer eccentricities of 0.41 and 0.45, respectively. This triple initially experiences triple evolution dynamics instability (TEDI) followed by Roche lobe overflow (RLOF). During RLOF, the inner orbit shrinks, and tidal effects gradually suppress the TEDI. Eventually, the inner binary undergoes a merger through contact (or collision). Finally, using models of rapidly rotating and strongly magnetic stars, along with standard core-collapse supernova (SN) or failed supernova (FSN) models, we find that a postmerger binary (PMB) consisting of an 12.11 M _⊙ merger product and a 1 M _⊙ companion star (originally an outer tertiary) can avoid RLOF. After an SN or FSN with a low ejected mass of ∼0.22 M _⊙ and a low kick velocity ( ${46}_{-33}^{+25}\,\mathrm{km}\,{{\rm{s}}}^{-1}$ or ${9}_{-8}^{+16}\,\mathrm{km}\,{{\rm{s}}}^{-1}$ ), the PMB can form Gaia BH1 in the Galactic disk.

ABSTRACTThe virulence factors of Staphylococcus aureus are tightly controlled by two-component systems (TCSs) and small RNA (sRNA). TCSs have been well studied over the past several decades, but our understanding of sRNA functions lags far behind that of TCS functions. Here, we studied the biological role of sRNA from 506 S. aureus RNA-seq datasets using independent component analysis (ICA). We found that a previously neglected sRNA, Sau-41, functions in the Agr system. Sau-41 is located within the PSMα operon and controlled by the Agr system. It was predicted to share 22-base complementarity with RNAIII, a major regulator of S. aureus virulence. The EMSA results demonstrated that Sau-41 directly binds to RNAIII. Furthermore, our results found that Sau-41 is capable of repressing S. aureus haemolysin activity by downregulating α-haemolysin and δ-toxin. The repression of α-haemolysin was attributed to the competition between the 5’ UTR of hla and Sau-41 for binding RNAIII. We observed that Sau-41 mitigated S. aureus virulence in an orthopaedic implant infection mouse model and alleviated osteolysis. Together, our results indicate that Sau-41 is a virulence-regulating RNA and suggest that Sau-41 might be involved in a negative feedback mechanism to control the Agr system. This work is a demonstration of using ICA in sRNA identification by mining high-throughput data and could be extended to other organisms as well.

Abstract Background This study aimed to delineate the cell heterogeneity in the bone-implant interface and investigate the fibroblast responses to implant-associated S. aureus infection. Methods Single-cell RNA sequencing of human periprosthetic tissues from patients with periprosthetic joint infection (PJI, n = 3) and patients with aseptic loosening (AL, n = 2) was performed. Cell type identities and gene expression profiles were analyzed to depict the single-cell landscape in the periprosthetic environment. In addition, 11 publicly available human scRNA-seq datasets were downloaded from GSE datasets and integrated with the in-house sequencing data to identify disease-specific fibroblast subtypes. Furthermore, fibroblast pseudotime trajectory analysis and Single-cell regulatory network inference and clustering (SCENIC) analysis were combined to identify transcription regulators responsible for fibroblast differentiation. Immunofluorescence was performed on the sequenced samples to validate the protein expression of the differentially expressed transcription regulators. Results Eight major cell types were identified in the human bone-implant interface by analyzing 36,466 cells. Meta-analysis of fibroblasts scRNA-seq data found fibroblasts in the bone-implant interface express a high level of CTHRC1. We also found fibroblasts could differentiate into pro-inflammatory and matrix-producing phenotypes, each primarily presented in the PJI and AL groups, respectively. Furthermore, NPAS2 and TFEC which are activated in PJI samples were suggested to induce pro-inflammatory polarization in fibroblasts, whereas HMX1, SOX5, SOX9, ZIC1, ETS2, and FOXO1 are matrix-producing regulators. Meanwhile, we conducted a CMap analysis and identified forskolin as a potential regulator for fibroblast differentiation toward matrix-producing phenotypes. Conclusions In this study, we discovered the existence of CTHRC1 + fibroblast in the bone-implant interface. Moreover, we revealed a bipolar mode of fibroblast differentiation and put forward the hypothesis that infection could modulate fibroblast toward a pro-inflammatory phenotype through NPAS2 and TFEC.

Abstract Background Actin cytoskeleton is connected with the processes of cell proliferation and migration in colorectal cancer (CRC). However, it is unknown how to accomplish these adjustments in CRC by actin cytoskeleton genes (ACGs) and here we investigated the role of hub prognosis‐related ACGs‐Diaphanous‐related formin 3 (DIAPH3) in CRC, as a potential, novel target. Methods The ACGs gene set from the Kyoto Encyclopedia of Genes and Genomes (KEGG) was used to group CRC patients and select prognosis‐related ACGs by univariate and multivariate Cox regression for constructing prognostic model. Next, we tested hub prognosis‐related ACGs‐ DIAPH3 expression in CRC and clarified the role of DIAPH3 by shRNA constructs in KM12 and SW480. Activation of EGFR was analyzed by western blot and immunofluorescence. Results The results showed that actin cytoskeleton function is a significant prognostic factor for CRC patients and related to clinicopathological characteristics such as T stage and lymph node metastasis. A prognostic model constructed by four prognosis‐related ACGs has a moderate intensity to 1‐year Survival (AUC = 0.71). And hub prognosis‐related ACGs DIAPH3 is downregulated in CRC. Knockdown of DIAPH3 could promote the proliferation and migration capacity of CRC. In addition, DIAPH3‐silenced cells increase EGFR phosphorylation by inhibiting EGFR transportation to lysosome. Conclusions ACGs play a significant role in tumor invasion and have the potential to predict the prognosis of CRC. Prognosis‐related ACGs DIAPH3 might be a new prognostic biomarker and DIAPH3 could inhibit CRC progression through maintaining EGFR degradation.

In Hilbert space, the geometry of the quantum state is identified by the quantum geometric tensor (QGT), whose imaginary part is the Berry curvature and whose real part is the quantum metric tensor. Here, we experimentally realize a complete Bloch-state tomography to directly measure the eigenfunction of an optical Raman lattice for ultracold atoms. Through the measured eigenfunction, the distribution of the complete QGT in the Brillouin zone is reconstructed, with which the topological invariants are extracted by the Berry curvature and the distances of quantum states in momentum space are measured by the quantum metric tensor. Furthermore, we experimentally test a predicted inequality between the Berry curvature and the quantum metric tensor, which reveals a deep connection between the topology and geometry.

The capacity performance of visible light communication (VLC) systems can be improved by using sparse code multiple access (SCMA). Since SCMA is a codebook-based multiple access scheme, the design of the codebook is crucial. SCMA codebook performance is severely affected by shot noise. Although several codebook design metrics have been proposed to compensate for the constellation distortion caused by shot noise, the improvement in performance is limited. In order to solve this problem, we use a separable codebook design structure to derive the analytical expression of the symbol error rate (SER) for the SCMA-VLC system under shot noise. According to the SER expression, we formulate an optimization problem and obtain a multi-user superimposed codebook to achieve a minimum SER for the SCMA-VLC system. The simulation results show that the proposed codebook significantly improves the performance of the SCMA-VLC system, especially in the case of high shot noise.

Objective To evaluate the diagnostic efficacy of triple‐phase bone scanning and the temporal impact of prosthesis implantation on the diagnostic efficacy of triple‐phase bone scanning for periprosthetic joint infection (PJI). Methods Patients who were admitted to our hospital for joint pain and dysfunction after total joint arthroplasty between 2014 and 2020 were retrospectively included. Triple‐phase bone scanning was performed, and the blood pool images were evaluated to obtain the semi‐quantitative criteria. The patients were then grouped into six groups according to the time interval from index primary arthroplasty to triple‐phase bone scanning. We examined whether there were significant differences in sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy between the groups. Results Overall, 66 patients who underwent total hip arthroplasty (THA) and 74 patients who underwent total knee arthroplasty (TKA) were analyzed. No significant differences were observed between visual analysis and semi‐quantitative measurement in terms of sensitivity, specificity, PPV, NPV, and accuracy. For patients with a time interval from prosthesis implantation to bone scanning of >1 year, visual analysis had a higher PPV (100%) in patients who underwent THA and the use of semi‐quantitative criteria had a higher NPV (85.7%) in patients who underwent TKA. Conclusion The semi‐quantitative criteria showed no advantages in the diagnosis of PJI. In addition, triple‐phase bone scanning demonstrated good clinical diagnostic efficacy when the time interval from prosthesis implantation to bone scanning was >1 year.

Wolf–Rayet (W-R) stars are very important massive stars. However, their origin and the observed binary fraction within the entire W-R population are still debated. We investigate some possible merger channels for the formation of W-R stars, including main sequence (MS)/Hertzsprung gap (HG) + MS, He + HG/giant branch (GB). We find that many products produced via binary merger can evolve into W-R stars, the MS/HG + MS merger channel can explain W-R stars with luminosities higher than ∼10 ^5.4 L _⊙ , while the He + HG/GB merger channel can explain low-luminosity W-R stars in the range of 10 ^4.7 L _⊙ ∼ 10 ^5.5 L _⊙ . In the population synthesis analysis of W-R stars, we assume an initial binary fraction ( f _ini,bin ) of 50% and 100% for massive stars. We also assume that MS/HG + MS merger products are nonrotating or rapidly rotating ( ω / ω _crit = 0.8). In different cases, the calculated single fractions of W-R stars range from 22.2%–60.6% in the Milky Way (MW) and from 8.3%–70.9% in the Large Magellanic Cloud (LMC). The current observations fall within the range of our calculations. When the merger product of MS/HG + MS rotates rapidly, we estimate that there are approximately 1015–1396 W-R stars in the MW and 128–204 W-R stars in the LMC. Our model also roughly reproduces the observed single-peak luminosity distribution of W-R stars in the MW. However, the weak bimodal luminosity distribution observed in the LMC is not reproduced in our model. We assess that this may be due to the model underestimating the mass-loss rate in the LMC. In conclusion, we consider that the binary merger is a significant formation channel for W-R formation, and can explain the observed high fraction of the single W-R stars in the total population.

The source of the Galactic lithium (Li) has long been a puzzle. With the discovery of Li in novae, extensive research has been conducted. However, there still exists a significant disparity between the observed abundance of Li in novae and the existing theoretical predictions. Using the Modules for Experiments in Stellar Astrophysics, we simulate the evolution of novae with element diffusion and appropriately increase the amount of ^3 He in the mixtures. Element diffusion enhances the transport efficiency between the nuclear reaction zone and the convective region on the surface of the white dwarf (WD) during nova eruptions, which results in more ^7 Be being transmitted to the WD surface and ultimately ejected. Compared to the previous predictions, the abundance of ^7 Be in novae simulated in our model significantly increases. The result is able to explain almost all observed novae. Using the method of population synthesis, we calculate Li yield in the Galaxy. We find that the Galactic occurrence rate of novae is about 130 yr ^−1 , and about 110 M _⊙ Li produced by nova eruption is ejected into the interstellar medium (ISM). About 73% of Li in the Galactic ISM originates from novae and approximately 15%–20% of the entire Galaxy. This means that novae are the important source of Li in the Galaxy.

Under the conventional scheme to generate an optical frequency comb (OFC) using an electro-optic modulator (EOM), the frequency interval of the OFC is determined via the frequency of the fundamental tone of the radio frequency (RF) driving signals. In this work, we use two harmonics without the fundamental tone to drive two EOMs, where the frequency interval of the generated flat OFC is the frequency of the fundamental tone. The orders of the two harmonics are coprime. Specifically, one harmonic drives the first branch of the dual-parallel Mach–Zehnder modulator (DPMZM) only, and the other harmonic drives the phase modulator (PM). The flatness of the OFC is achieved by adjusting the amplitude and phase of the RF driving harmonics as well as the bias of the EOM. Both a simulation and an experiment were carried out to verify the effectiveness of the proposed scheme. When the second harmonic drives the DPMZM and the third harmonic drives the PM, an 11-comb line OFC is generated, where the flatness of the OFC was 0.63 dB and 0.65 dB under the simulation and experiment, respectively. When the third harmonic drives the DPMZM and the second harmonic drives the PM, a 13-comb line OFC is generated, where the flatness of the OFC was 0.62 dB and 0.95 dB under the simulation and experiment, respectively. We also investigate the performance of the generated OFC when one harmonic drives two branches of the DPMZM and the other harmonic drives the PM. The comparison of the OFCs’ performance demonstrates the effectiveness of the proposed scheme.

Abstract Background Gastrointestinal mucormycosis (GIM) is a rare, opportunistic fungal infection with poor prognosis. Clinically, it is difficult to diagnose GIM owing to its nonspecific clinical symptoms and poor suspicion. The estimated incidence of GIM is inaccurate, and most cases are diagnosed accidentally during surgery or upon postmortem examination. GIM usually occurs in patients with immune deficiencies or diabetes. Here, we report two cases of immunocompetent young patients with GIM who had good prognosis after treatment. Compared to other case reports on GIM, our cases had unusual infection sites and no obvious predisposing factors, which make it important to highlight these cases. Case presentation The first case was that of a 16-year-old immunocompetent boy who was admitted with gastrointestinal bleeding and perforation due to a gastric ulcer. Strategies used to arrest bleeding during emergency gastroscopy were unsuccessful. An adhesive mass was then discovered through laparoscopy. The patient underwent type II gastric resection. Pathological examination of the mass revealed bacterial infection and GIM. The second case was of a 33-year-old immunocompetent woman with a recent history of a lower leg sprain. The patient subsequently became critically ill and required ventilatory support. After hemodynamic stabilization and extubation, she presented with hematemesis due to exfoliation and necrosis of the stomach wall. The patient underwent total gastrectomy plus jejunostomy. The pathology results revealed severe bacterial infection and fungal infection that was confirmed as GIM. The patient fully recovered after receiving anti-infective and antifungal treatments. Conclusions Neither patient was immunosuppressed, and both patients presented with gastrointestinal bleeding. GIM was confirmed via pathological examination. GIM is not limited to immunocompromised patients, and its diagnosis mainly relies on pathological examination. Early diagnosis, timely surgical treatment, and early administration of systemic drug treatment are fundamental to improving its prognosis.

Abstract In China, after the revision of the renewable energy policy, transactions among the main parties involved in the electricity market are becoming increasingly complicated. Nevertheless, the impact of the Renewable Energy Portfolio Standard (RPS) on market equilibrium has not been studied so far. Hence, in this study, a noncooperative multiagent game model for the electricity market was developed on the basis of the recently released RPS and by considering various factors, such as green certificate transactions and the changing of the selling entities. The improved firefly algorithm was used to solve the model, and problematic gaming was analyzed empirically. Once the new RPS is implemented, the price of bound electricity will become higher than that of nonbound electricity, and the trading volume of bound electricity will increase. Because fossil energy producers will no longer be responsible for the electricity quotas, their electricity trading volumes will now be more related to the market demand than they were earlier. Renewable energy companies will increase their revenues by reducing their bound electricity sales. If the RPS increases the electricity quota ratios, the costs incurred by electricity sales companies and large power users will also increase, leading the electricity sales companies to reduce their electricity trading volumes. The study findings will help analyze the impacts of implementing the RPS and provide a basis and a reference for policymakers to make improvements to the standard in the future.

In the visible light positioning (VLP) system, the received signal strength (RSS) algorithm has a better signal noise ratio performance than the time difference of arrival (TDOA) algorithm, while the RSS algorithm needs to work under the condition that the transmitter and receiver are strictly parallel. However, the receiver is prone to tilt due to environmental disturbances, which reduces the accuracy of the RSS algorithm. For the tilted receiver, the TDOA algorithm has a higher positioning accuracy than the RSS algorithm. In order to take full advantage of the two algorithms, we propose a hybrid positioning algorithm to locate the tilted receiver by using a Gaussian process (GP). The scheme separately uses RSS and the distance difference as the inputs of the GP model to estimate the position of the receiver. Then, according to the proposed positioning selection strategy, the more credible estimated position in our opinion is selected as the final estimated position. In addition, RSS information in the hybrid algorithm is extracted from the TDOA signal, which allows the hybrid algorithm to prevent an increase in the complexity of the VLP system. During the training and testing, RSS is normalized to meet the order-of-magnitude requirements of the GP model on the input data. Simulation results validate the hybrid algorithm based on a two-dimensional positioning system for the tilted receiver. When the standard deviations of the azimuth angle and elevation angle are 1°, the positioning accuracy of the hybrid algorithm is 53.7% higher than that of the RSS algorithm using an artificial neural network, and 49.9% higher than that of the RSS algorithm using a GP. The localization error under 1° standard deviations of azimuth and elevation angles is 20.2% lower than that under 20° standard deviations of the two angles.

Biofilm formation of Staphylococcus aureus is the major cause of implant-associated infections (IAIs). Antimicrobial treatment is one of the most effective therapeutic options for S. aureus infections. However, it can also lead to adaptive transcriptomic changes due to extreme selective pressure, which may increase the risk of antimicrobial resistance. To study the transcriptional changes in S. aureus upon exposure to antimicrobial agents, we obtained expression profiles of S. aureus treated with six antimicrobials (flucloxacillin, vancomycin, ciprofloxacin, clindamycin, erythromycin, and linezolid, n = 6 for each group). We also included an untreated control group (n = 8) downloaded from the Gene Expression Omnibus (GEO) database (GSE70043, GSE56100) for integrated bioinformatic analyses. We identified 82 (44 up, 38 down) and 53 (17 up, 36 down) differentially expressed genes (DEGs) in logarithmic and stationary phases, respectively. When exposed to different antimicrobial agents, we found that manganese import system genes and immune response gene sbi (immunoglobulin G-binding protein Sbi) were upregulated in S. aureus at all stages. During the logarithmic phase, we observed adaptive transcriptomic changes in S. aureus mainly in the stability of protein synthesis, adhesion, and biofilm formation. In the stationary phase, we observed a downregulation in genes related to amino biosynthesis, ATP synthesis, and DNA replication. We verified these results by qPCR. Importantly, these results could help our understanding of the molecular mechanisms underlying the proliferation and antimicrobial resistance of S. aureus.

Spectral clustering makes use of the spectrum of an input affinity matrix to segment data into disjoint clusters. The performance of spectral clustering depends heavily on the quality of the affinity matrix. Commonly used affinity matrices are constructed by either the Gaussian kernel or the self-expressive model with sparse or low-rank constraints. A technique called diffusion which acts as a post-process has recently shown to improve the quality of the affinity matrix significantly, by taking advantage of the contextual information. In this paper, we propose a variant of the diffusion process, named Self-Supervised Diffusion, which incorporates clustering result as feedback to provide supervisory signals for the diffusion process. The proposed method contains two stages, namely affinity learning with diffusion and spectral clustering. It works in an iterative fashion, where in each iteration the clustering result is utilized to calculate a pseudo-label similarity so that it can aid the affinity learning stage in the next iteration. Extensive experiments on both synthetic and real-world data have demonstrated that the proposed method can learn accurate and robust affinity, and thus achieves superior clustering performance.

Abstract Aberrant cholesterol homeostasis is implicated in the pathogenesis of amyotrophic lateral sclerosis (ALS), a fatal neuromuscular disease that is due to motor neuron (MN) death. Cellular toxicity from excess cholesterol is averted when it is enzymatically oxidized to oxysterols and bile acids (BAs) to promote its removal. In contrast, the auto oxidation of excess cholesterol is often detrimental to cellular survival. Although oxidized metabolites of cholesterol are altered in the blood and CSF of ALS patients, it is unknown if increased cholesterol oxidation occurs in the SC during ALS, and if exposure to oxidized cholesterol metabolites affects human MN viability. Here, we show that in the SOD1G93A mouse model of ALS that several oxysterols, BAs and auto oxidized sterols are increased in the lumbar SC, plasma, and feces during disease. Similar changes in cholesterol oxidation were found in the cervical SC of sporadic ALS patients. Notably, auto-oxidized sterols, but not oxysterols and BAs, were toxic to iPSC derived human MNs. Thus, increased cholesterol oxidation is a manifestation of ALS and non-regulated sterol oxidation likely contributes to MN death. Developing therapeutic approaches to restore cholesterol homeostasis in the SC may lead to a treatment for ALS.

Background: Tumor dormancy is an important way to develop drug resistance. This study aimed to identify the characteristics of colorectal cancer (CRC) cell dormancy.Methods: Based on the CRC cohorts, a total of 1,044 CRC patients were included in this study, and divided into a dormant subgroup and proliferous subgroup. Non-negative matrix factorization (NMF) was used to distinguish the dormant subgroup of CRC via transcriptome data of cancer tissues. Gene Set Enrichment Analysis (GSEA) was used to explore the characteristics of dormant CRC. The characteristics were verified in the cell model, which was used to predict key factors driving CRC dormancy. Potential treatments for CRC dormancy were also examined.Results: The dormant subgroup had a poor prognosis and was more likely to relapse. GSEA analysis showed two defining characteristics of the dormant subgroup, a difference in energy metabolism and synergistic effects of cancer-associated fibroblasts (CAFs), which were verified in a dormant cell model. Transcriptome and clinical data identified LMOD1, MAB21L2, and ASPN as important factors associated with cell dormancy and verified that erlotinib, and CB-839 were potential treatment options.Conclusion: Dormant CRC is associated with high glutamine metabolism and synergizes with CAFs in 5-FU resistance, and the key effectors are LMOD1, MAB21L2, and ASPN. Austocystin D, erlotinib, and CB-839 may be useful for dormant CRC.

Staphylococcus aureus (S. aureus) biofilm is the major cause of failure of implant infection treatment that results in heavy social and economic burden on individuals, families, and communities. Planktonic S. aureus attaches to medical implant surfaces where it proliferates and is wrapped by extracellular polymeric substances, forming a solid and complex biofilm. This provides a stable environment for bacterial growth, infection maintenance, and diffusion and protects the bacteria from antimicrobial agents and the immune system of the host. Macrophages are an important component of the innate immune system and resist pathogen invasion and infection through phagocytosis, antigen presentation, and cytokine secretion. The persistence, spread, or clearance of infection is determined by interplay between macrophages and S. aureus in the implant infection microenvironment. In this review, we discuss the interactions between S. aureus biofilm and macrophages, including the effects of biofilm-related bacteria on the macrophage immune response, roles of myeloid-derived suppressor cells during biofilm infection, regulation of immune cell metabolic patterns by the biofilm environment, and immune evasion strategies adopted by the biofilm against macrophages. Finally, we summarize the current methods that support macrophage-mediated removal of biofilms and emphasize the importance of considering multi-dimensions and factors related to implant-associated infection such as immunity, metabolism, the host, and the pathogen when developing new treatments.

Background: Alcohol intake is a well-known lifestyle risk factor for CRC, and an increasing number of studies have revealed that alcohol intake is also tightly associated with CRC metastasis. However, the effect of alcohol on CRC metastasis and its underlying mechanism remain unclear. Methods: A retrospective cohort study was performed to investigate the characteristics of patients with alcohol-related CRC. The effects of ethanol on the biological behaviours of CRC cells were assessed through in vivo and in vitro assays using the Lieber-DeCarli ethanol liquid diet and ethanol, respectively. The ethanol-mediated signalling pathway and downstream factors were screened through ELISA, western blot, immunofluorescence and co-immunoprecipitation. Findings: Most patients with alcohol-related CRC, particularly those with tumour metastasis, were characterized by a notably higher circulating ethanol level and a lower systemic acetaldehyde level. Moreover, CRC cells accumulated in ethanol, but not acetaldehyde, to notably higher levels compared with adjacent normal cells. Alcohol intake significantly promoted CRC metastasis via the ethanol-mediated TGF-β/Smad/Snail axis, and ethanol induced the cytoplasmic mislocalization of RUNX3 and further promoted the aggressiveness of CRC by targeting Snail. Pirfenidone (PFD) significantly eliminated the effects of ethanol on CRC metastasis by specifically blocking TGF-β signalling. Interpretation: Alcohol intake plays a vital role in CRC metastasis via the ethanol-mediated TGF-β/RUNX3/Snail axis, and PFD might be a novel therapeutic management strategy for CRC. Biofilms formed by Staphylococcus aureus are one of the predominant causes of implant-associated infections (IAIs). Previous studies have found that S. aureus nucleases nuc1 and nuc2 modulate biofilm formation. In this study, we found low nuc1/nuc2 expression and high biofilm-forming ability among IAI isolates. Furthermore, in a mouse model of exogenous IAIs, Δnuc1/2 exhibited higher bacterial load on the surface of the implant than that exhibited by the other groups (WT, Δnuc1, and Δnuc2). Survival analysis of the hematogenous IAI mouse model indicated that nuc1 is a virulence factor related to mortality. We then detected the influence of nuc1 and nuc2 on biofilm formation and immune evasion in vitro. Observation of in vitro biofilm structures with scanning electron microscopy and evaluation of bacterial aggregation with flow cytometry revealed that both nuc1 and nuc2 are involved in biofilm structuring and bacterial aggregation. Unlike nuc1, which is reported to participate in immune evasion, nuc2 cannot degrade neutrophil extracellular traps. Moreover, we found that nuc1/nuc2 transcription is negatively correlated during S. aureus growth, and a possible complementary relationship has been proposed. In conclusion, nuc1/nuc2 are complementary genes involved in biofilm formation in exogenous IAIs. However, nuc2 contributes less to virulence and is not involved in immune evasion.

A switching neural network control scheme, consisting of the adaptive neural network controller and sliding mode controller, is proposed for underactuated formation reconfiguration in elliptic orbits with the loss of either the radial or in-track thrust. By using the inherent coupling of system states, the switching neural network technique is then adopted to estimate the unmatched disturbances and design the underactuated controller to achieve underactuated formation reconfiguration with high precision. The adaptive neural network controller works in the active region, and the disturbances composed of linearization errors and external perturbations are approximated by radial basis function neural networks. The adaptive sliding mode controller works outside the active region, and the upper bound of the approximation errors is estimated by the adaptation law. The stability of the closed-loop control system is proved via the Lyapunov-based approach. The numerical simulation results have demonstrated the rapid, high-precision and robust performance of the proposed controller compared with the linear sliding mode controller.

This work aims to achieve the 6000t compression-shear test machine frame design with the lightweight. The force condition of the compression-shear test machine frame under limited working conditions is first analyzed, and the static analysis of the compression-shear test machine frame is performed using ABAQUS. Then, taking the volume of the frame of the compression-shear testing machine as the constraint condition, the topology optimization of the compression-shear testing machine frame is performed using the variable density method of topology optimization, and the model is reconstructed accordingly. Finally, not only the static characteristics of the frame before and after optimization but also the modal characteristics of the frame before and after optimization and the dynamic characteristics after sudden unloading are compared and analyzed. The results show that the weight of the frame decreases by 14.5% after optimization, and the maximum static stress of the frame is still less than the yield strength of the material; the maximum displacement is still less than the allowable maximum displacement, which meets the requirements of static strength and stiffness. The natural frequency of each mode is much greater than the working frequency, which meets the requirements of dynamic stiffness. After sudden unloading, the maximum dynamic stress of beams, columns, and base of the frame are less than the yield strength of materials, which meets the requirements of dynamic strength.

Staphylococcus aureus is one of the predominant causes of periprosthetic joint infections (PJIs). Bacterial adhesion and biofilm formation are important factors in the pathogenesis of PJIs. S. aureus biofilm formation is regulated by several factors, including S. aureus regulator A (SarA). Previous studies have found that SarA mutants have limited ability to develop biofilms. In this study, we identified a SarA-targeting antibiofilm compound, ZINC00990144, and evaluated its efficacy and toxicity. According to static biofilm assay, the antibiofilm ability of the compound was concentration dependent. ZINC00990144 reduced biofilm in multiple strains by 40–86% at a concentration of 11.5 μM. Additionally, ZINC00990144 inhibited biofilm formation on different orthopedic implant materials including Titanium and UHMWPE disc. Furthermore, quantitative polymerase chain reaction results demonstrated that ZINC00990144 upregulated the expression of S. aureus exoproteases to inhibit the formation of biofilms. Moreover, ZINC00990144 prevented biofilm formation when exposed to sub-inhibitory doses of vancomycin, which is known to promote biofilm formation. CCK-8 results demonstrated ZINC00990144 has no significant effect on cell viability at concentration of 11.5 μM or below. Finally, we verified the antibiofilm function of the compound in vivo using implant infection mice model with/without exposure to sub-inhibitory vancomycin. In conclusion, ZINC00990144 acts by modulating between biofilm and planktonic state of S. aureus instead of being bactericidal. Therefore, it has the potential to be used in combination with other antibiotics to prevent PJIs.

In recent years, bariatric surgery has emerged as a promising treatment for type 2 diabetes. Bariatric surgery is known to cause alterations in the relative abundance and composition of gut microbiota, which may lead to alterations in the levels of Short-Chain Fatty Acids (SCFAs) that are produced during fermentation by gut microbes. However, little is known about the mechanism of improved glucose metabolism mediated by gut microbiota following bariatric surgery. The aim of our study was to explore whether changes in gut microbiota and in fecal SCFA could be detected following single-anastomosis duodenal jejunal bypass (DJB-sa) surgery, a type of bariatric surgery, and whether these alterations might be related to the improvement of glucose metabolism. To this end, we performed DJB-sa or SHAM surgery on Goto-Kakisaki (GK) rats. We then compared the glucose metabolism as well as changes in gut microbiota and SCFAs levels between both groups. Our results showed that DJB-sa surgery was associated with a significant decrease in fasting blood glucose (FBG), intraperitoneal glucose tolerance test (IPGTT), and fasting serum insulin (FSI). And, DJB-sa led to a change in the composition of gut microbiota including an increase in the relative abundance of SCFA-producing bacteria (Bifidobacterium and Subdoligranulum). Moreover, the levels of six SCFAs in feces, as well as the intestinal expression of SCFA receptors including G-protein-coupled receptor 41 (GPR41), G-protein-coupled receptor 43 (GPR43), and G-protein-coupled receptor 109A (GPR109A), and the expression of Glucagon-like peptide-1 (GLP-1) displayed a significant increase following DJB-sa compared with the Sham group. Thus, the gut microbiota may contribute to the improvement of glucose metabolism in type 2 diabetes following DJB-sa. In conclusion, our study shows that DJB-sa improves glucose metabolism by modulating gut microbiota and by increasing short-chain fatty acid production.

Constellation configuration design is a prerequisite and critical step in the construction of a mega-constellation system in low Earth orbit. However, the huge number of satellites and the intricate changes in relative positions among them make the configuration design the most challenging part. In this paper, we propose a configuration design scheme for mega-constellations considering collision-avoidance constraints with the objective of uniform global coverage. In this design scheme, the constellation is made up of multiple Walker constellations with the same orbital altitude and different orbital inclination. Moreover, the analytical expression for the minimum distance between any two satellites in the same orbital altitude is derived, and the constellation internal collision-avoidance constraint is established accordingly. Finally, a permanent inter-satellite link design scheme without dynamic reconstruction is presented based on the mega-constellation configuration. Simulation results show that the mega-constellation design scheme introduced in this paper can achieve relatively uniform global coverage (its N Asset Coverage ranges from 18 to 25). The mixed Walker constellation is capable of providing a greater number of N Asset Coverage for most of the world than the Walker constellation of the same satellite order of magnitude. In addition, the inter-satellite link scheme designed in this paper can ensure continuous and stable communication between any satellite nodes.

The bit error ratio (BER) performance of a non-orthogonal multiple access (NOMA) visible light communication (VLC) system is poor due to the unequal distances between adjacent points in the superposition constellation (SC). In this paper, we propose a novel scheme to improve the BER performance by adjusting parameters to change the shape of SC at the transmitter and by adjusting the parameters of successive interference cancellation (SIC) decoding at the receiver simultaneously, which is called a SC and SIC adjustment (SC-SIC-A) scheme. For multi-user NOMA VLC system, we derive the closed-form BER expression for each user, where the modulation format is four-quadrature amplitude modulation. According to the derived BER expressions, we formulate an optimization problem that minimizes the average BER for all users by adjusting the obtained parameters of SC and SIC decoding via differential evolution algorithm. The improvement of capacity performance is investigated consequently. In order to verify the feasibility and effectiveness of the proposed SC-SIC-A scheme, we carried out theoretical analysis, Monte Carlo simulation and experiments of two-user and three-user NOMA VLC systems. Results show that the SC-SIC-A scheme outperforms the existing schemes in NOMA VLC system, where the signal-to-noise ratio (SNR) reductions to achieve BER of 10−3 are 1.3 dB and 0.8 dB for both users in the two-user NOMA VLC system, respectively, and the SNR reductions to achieve BER of 10−3 are 5.7 dB, 4.3 dB and 4.6 dB for all users in the three-user NOMA VLC system, respectively.

The development of non-Hermitian topological band theory has led to observations of novel topological phenomena in effectively classical, driven, and dissipative systems. However, for open quantum many-body systems, the absence of a ground state presents a challenge to define robust signatures of non-Hermitian topology. We show that such a signature is provided by crossings in the time evolution of the entanglement spectrum. These crossings occur in quenches from the trivial to the topological phase of a driven-dissipative Kitaev chain that is described by a Markovian quantum master equation in Lindblad form. At the topological transition, which can be crossed either by changing parameters of the Hamiltonian of the system or by increasing the strength of dissipation, the timescale at which the first entanglement spectrum crossing occurs diverges with a dynamical critical exponent of ε=1/2. We corroborate these numerical findings with an exact analytical solution of the quench dynamics for a spectrally flat postquench Liouvillian. This exact solution suggests an interpretation of the topological quench dynamics as a fermion parity pump. Our work thus reveals signatures of non-Hermitian topology that are unique to quantum many-body systems and cannot be emulated in classical simulators of non-Hermitian wave physics.

Intestinal injury has long been considered to play a crucial role in the pathophysiology of sepsis and has even been characterized as the “motor” of it. Thus, we explored the effects of connexin43 (Cx43) on sepsis-induced intestinal injury in order to provide potential therapeutic strategies. Rat cecal ligation and puncture (CLP) models in vivo and cell models (IEC-6 cells) pretreated with LPS in vitro were used in the current study. Firstly, different methods, such as Cx43 inhibitors (18-α-GA and oleamide) or siRNA targeting Cx43 and N-acetyl cysteine (NAC) (a kind of ROS scavenger), were used to observe the effects of Cx43 channels mediating ROS transfer on intestinal injury. Secondly, the influence of ROS content on the activity of the JNK1/Sirt1/FoxO3a signaling pathway was explored through the application of NAC, sp600125 (a JNK1 inhibitor), and nicotinamide (a Sirt1 inhibitor). Finally, luciferase assays and ChIP were used to determine the direct regulation of FoxO3a on proapoptotic proteins, Bim and Puma. The results showed that sepsis-induced intestinal injury presented a dynamic change, coincident with the alternation of Cx43 expression. The inhibition of Cx43 attenuated CLP-induced intestinal injury in vivo and LPS-induced IEC-6 injury in vitro. The changes of Cx43 channel function regulated ROS transfer between the neighboring cells, which mediated the activation of the JNK1/Sirt1/FoxO3a signaling pathway. FoxO3a directly affected its downstream target genes, Bim and Puma, which are responsible for cell or tissue apoptosis. In summary, our results suggest that Cx43 inhibition suppresses ROS transfer and inactivates the JNK1/Sirt1/FoxO3a signaling pathway to protect against sepsis-induced intestinal injury.

In this paper, a terahertz (THz) wireless communication system at 400 GHz with various modulation formats [on-off keying (OOK), quadrature phase-shift keying (QPSK), 16-quadrature amplitude modulation (16-QAM), and 32-quadrature amplitude modulation (32-QAM)] is experimentally demonstrated based on photonic generation of highly pure THz carriers. The experimental THz wireless photonic transmission system is enabled by the ultrawideband behavior of an antenna-integrated unitraveling-carrier-photodiode-based transmitter and a Schottky mixer-based THz receiver. In the experiment, a phase-correlated optical frequency comb (OFC) is created for photomixing generation of the desired THz carrier frequencies with low phase noise. The OFC allows for the generation of flexibly tunable THz carrier frequencies. The performance of the generated THz carriers is experimentally characterized in terms of phase noise, spectrum purity, tunability, and long-term stability. In the case of generating 400 GHz carrier, the measured timing jitter, linewidth, and long-term stability in the experiment are 51.5 fs, less than 2 Hz, and less than ±1 Hz with 3 hours, respectively. We also theoretically analyze the phase noise of photonically generated THz beat-notes when phase correlation of two optical comb tones is damaged due to their path-length difference. In addition, we demonstrate THz wireless transmission of various modulation formats, including OOK, QPSK, 16-QAM, and 32-QAM at beyond 10 Gb/s in such a system, and the measured bit error rate (BER) performance for all the signals after 0.5 m free-space delivery is below the hard decision forward error correction threshold of 3.8 × 10-3. Furthermore, the influence of THz carrier purity on the system performance is experimentally analyzed with respect to the BER of the THz communication signals.

A novel scheme of a highly stable dual-wavelength short optical pulse source (SOPS) based on a dual-loop optoelectronic oscillator (OEO) with two wavelengths is demonstrated. The structure of the dual-loop OEO with two wavelengths can suppress the random beating noise effectively and generate the microwave signal with low phase noise (-122.4 dBc/Hz at 10 kHz) . It consists of two directly modulated lasers and a phase modulator (PM) . The light that is directly modulated by the large signal generated from the OEO is injected into the PM to achieve remarkable chirp. Then, by optimizing the length of the dispersion compensating fiber, the optical pulses are further compressed. In the experiment, the SOPS with dual wavelengths is generated simultaneously. The repetition rate, pulse width, and timing jitter of the SOPS are 10 GHz, 2.9 ps, and 12.5 fs, respectively. In particular, the loop drift of the OEO is effectively compensated by fiber stretchers using phase-locked-loop technology. The frequency drift of the microwave signal is less than ±73.3 mHz for a long-term measurement with a duration of 1 h. Therefore, the long-term stability of SOPS is guaranteed.

Background/Aims: MicroRNAs (miRNAs) are known to produce post-transcriptional repression of gene expression. In light of the ability of decoy oligodeocynucleotides (ODNs) to sequestrate transcription factors (TFs) and the similar double-stranded structure between decoy ODNs and miRNAs, we proposed that miRNAs might act as endogenous decoy molecules to produce transcriptional regulation of gene expression. Methods: Quantitative real-time RT-PCR analysis was used to measure the changes of miRNA and mRNA expression. Luciferase reporter gene activity assay was used to investigate the functional interaction between miRNAs and TFs. Electrophoresis mobility shift assay (EMSA) and modified chromatin immunoprecipitation assay (ChIP) were utilized to investigate the physical interactions between miRNAs and TFs. MTT cell viability assay and cellular DNA fragmentation ELISA were used to study apoptotic cell death. Results: We presented here that miRNAs could regulate, either negatively or positively, gene expression at the transcriptional level through its decoy-like actions and this mechanism operates under physiological conditions to produce cellular functions. We identified the putative cis-elements for transcriptional factors NF-κB and NFAT in the mature miR-939 and miR-376a, respectively. We experimentally established the ability of these miRNAs to physically bind their respective target TFs, using EMSA and ChIP methods. We then utilized the luciferase reporter gene assay to characterize the specific regulation of luciferase gene activities by miR-939/pre-miR-939:NF-κB or miR-376a/pre-miR-376a:NFAT interactions. Moreover, miR-939 and miR-376a produced transcriptional regulation of endogenous genes Bcl-xL and FasL/miR-26 that are the transcriptional targets for NF-kB and NFAT, respectively, but are not post-transcriptional targets for these two miRNAs. Finally, interference of these miRNAs with NF-κB and NFAT demonstrated clear phenotypes at the cellular level as manifested by the regulation of neuroblastoma cell death by miR-939 and miR-376a. Conclusion: Our study identified a novel non-canonical mechanism of miRNAs and suggests that when considering the cellular function of miRNAs the decoy-like mechanism for transcriptional regulation (activation or repression) should be taken into account.

Calibration transfer is an important field for near-infrared (NIR) spectroscopy in practical applications. However, most transfer methods are constructed with standard samples, which are expensive and difficult to obtain. Taking this problem into account, this paper proposes a calibration transfer method based on affine invariance without transfer standards (CTAI). Our method can be utilized to adjust the difference between two instruments by affine transformation. CTAI firstly establishes a partial least squares (PLS) model of the master instrument to obtain score matrices and predicted values of the two instruments, and then the regression coefficients between each of the score vectors and predicted values are computed for the master instrument and the slave instrument, respectively. Next, angles and biases are calculated between the regression coefficients of the master instrument and the corresponding regression coefficients of the slave instrument, respectively. Finally, by introducing affine transformation, new samples are predicted based on the obtained angles and biases. A comparative study between CTAI and the other five methods was conducted, and the performances of these algorithms were tested with two NIR spectral datasets. The obtained experimental results show clearly that, in general CTAI is more robust and can also achieve the best Root Mean Square Error of test sets (RMSEPs). In addition, the results of statistical difference with the Wilcoxon signed rank test show that CTAI is generally better than the others, and at least statistically the same.

In order to enable the calibration model to be effectively transferred among multiple instruments and correct the differences between the spectra measured by different instruments, a new feature transfer model based on partial least squares regression (PLS) subspace (PLSCT) is proposed in this paper. Firstly, the PLS model of the master instrument is built, meanwhile a PLS subspace is constructed by the feature vectors. Then the master spectra and the slave spectra are projected into the PLS subspace, and the features of the spectra are also extracted at the same time. In the subspace, the pseudo predicted feature of the slave spectra is transferred by the ordinary least squares method so that it matches the predicted feature of the master spectra. Finally, a feature transfer relationship model is constructed through the feature transfer of the PLS subspace. This PLS-based subspace transfer provides an efficient method for performing calibration transfer with only a small number of standard samples. The performance of the PLSCT was compared and assessed with slope and bias correction (SBC), piecewise direct standardization (PDS), calibration transfer method based on canonical correlation analysis (CCACT), generalized least squares (GLSW), multiplicative signal correction (MSC) methods in three real datasets, statistically tested by the Wilcoxon signed rank test. The obtained experimental results indicate that PLSCT method based on the PLS subspace is more stable and can acquire more accurate prediction results.

We have demonstrated in this paper a novel tunable microwave frequency multiplication by injecting a weakly phase-modulated optical signal into a DFB laser diode. Signals with multiple weak sidebands are generated by cross-phase modulation of a continuous wave (CW) with short pulses from mode-locked fiber laser. Then, frequency multiplication is achieved by injection and phase locking a commercially available DFB laser to one of the harmonics of the phase modulated signal. The multiplication factor can be tuned by changing the frequency difference between the CW and the free oscillating wavelength of the DFB laser. The experimental results show that, with an original signal at a repetition rate of 1 GHz, a microwave signal with high spectral purity and stability is generated with a multiplication factor up to 60. The side-mode suppression ratio over 40 dB and phase noise lower than -90 dBc/Hz at 10 kHz are demonstrated over a continuous tuning range from 20 to 40.

Tunable all-optical wavelength conversion (AOWC) of a 40-Gb/s RZ-OOK data signal based on cascaded second-harmonic generation (SHG) and difference-frequency generation (DFG) in a Ti:PPLN waveguide is demonstrated. Error-free performances with negligible power penalty are achieved for the wavelength-converted signal at 1535, 1538, 1541, 1551, 1554, and 1557 nm, respectively.

We experimentally demonstrate simultaneous 3R regeneration of 4 $\times$ 40-Gbit/s WDM channels, which is based on the four-wave mixing (FWM) in a single dispersion-shifted fiber. The multichannel 3R regeneration performance is validated by bit-error rate (BER) measurements. Receiver powers at a BER of $10^{-9}$ are improved by 5.2 dB, 4.8 dB, 3.3 dB, and 3.9 dB for the four WDM channels, respectively. Mitigation of the interchannel nonlinearities is achieved mainly through an interchannel 0.5-bit slot time delay, which is introduced by differential group delay. Bidirectional propagation and polarization multiplexing are also applied to relieve the interchannel FWM.

Utilizing the Fermi gas microscope, recently the MIT group has measured the spin transport of the Fermi Hubbard model (FHM) starting from a spin-density-wave state, and the Princeton group has measured the charge transport of the FHM starting from a charge-density-wave state. Motivated by these two experiments, we prove a theorem that shows under certain conditions, the spin and charge transports can be equivalent to each other. The proof makes use of the particle-hole transformation of the FHM and a recently discovered symmetry protected dynamical symmetry. Our results can be directly verified in future cold atom experiment with the Fermi gas microscope.

Dissemination of HIV in the host involves transit of the virus and virus-infected cells across the lymphatic endothelium. HIV may alter lymphatic endothelial permeability to foster dissemination, but the mechanism is largely unexplored. Using a primary human lymphatic endothelial cell model, we found that HIV-1 envelope protein gp120 induced lymphatic hyperpermeability by disturbing the normal function of Robo4, a novel regulator of endothelial permeability. HIV-1 gp120 induced fibronectin expression and integrin α₅β₁ phosphorylation, which led to the complexing of these three proteins, and their subsequent interaction with Robo4 through its fibronectin type III repeats. Moreover, pretreatment with an active N-terminus fragment of Slit2, a Robo4 agonist, protected lymphatic endothelial cells from HIV-1 gp120-induced hyperpermeability by inhibiting c-Src kinase activation. Our results indicate that targeting Slit2/Robo4 signaling may protect the integrity of the lymphatic barrier and limit the dissemination of HIV in the host.
