Maximizing protein expression levels poses a major challenge in bioengineering. To increase protein expression levels, numerous factors, including codon bias, codon context bias, hidden stop codons, homologous recombination, suitable guanine-cytosine ratio, and hairpin loop structure, are crucial and quantified by six objective functions: CAI, CPB, HSC, HD, GC3, and SL. Optimizing these six objectives simultaneously constitutes a multi-objective optimization problem, aiming to identify the favorable Pareto solutions rather than a singular optimal solution. However, achieving satisfactory solutions requires numerous cycles and solutions, thus leading to a large number of functional evaluations. While there are frameworks for multi-objective optimization problems, they often lack efficient support for objective function computation in protein encoding. In this paper, we proposed a method to design a set of coding sequences (CDSs) based on non-dominated sorting genetic algorithm III (NSGA-III), accelerated using NVIDIA graphical processing units (GPUs). Experimental results indicated that our method is 15,454 times faster than the Pymoo framework and is evaluated using 100 solutions and 100 cycles. Since our GPU implementation facilitated the use of larger solutions and more cycles, we were able to design a superior set of CDSs by increasing solutions to 400 and cycles to 12,800. In addition, our NSGA-III-based method consistently surpassed the NSGA-II approach when the number of cycles exceeded 3200 by utilizing 100 solutions. Finally, we observed that a gradual reduction of the mutation probability as the number of cycles increased yielded better quality results than maintaining a fixed mutation probability.

Abstract Background Yukgunja-tang (YGJ) is an herbal prescription used to treat the symptoms of gastroesophageal reflux disease (GERD). Although many preclinical and clinical studies on YGJ have been conducted on GERD, there is a lack of evidence from blinded studies to exclude placebo effects. Therefore, this protocol proposes a clinical trial that is single-centered, randomized, double-blinded, double-dummy to objectively evaluate the efficacy and safety of co-administered YGJ and rabeprazole (RPZ) in patients with GERD previously treated with proton pump inhibitors (PPIs) and still experiencing symptoms. Methods A total of 86 participants with refractory GERD (rGERD) will be randomized in a 1:1 ratio to the treatment [YGJ and RPZ (10 mg/d)] and control groups [double-dose RPZ (20 mg/d)] for 4 weeks of treatment (weeks 0–4) followed by 4 weeks of follow-up (weeks 4–8). The Frequency Scale for the Symptoms of GERD will be analyzed for the primary endpoint. Reflux Disease Questionnaire, Reflux Symptom Score, GERD-Health Related Quality of Life, Overall Treatment Evaluation, Spleen Qi Deficiency Questionnaire, Damum Questionnaire, and dyspepsia Visual Analogue Scale will be used to evaluate treatment effects on GERD related symptoms and quality of life and to compare treatment effects by subgroups. Safety tests will be analyzed by investigating adverse events. Discussion This clinical trial will be the first rigorous double-blind, double-dummy, placebo-controlled study to precisely evaluate the efficacy and safety of the combination of YGJ and PPIs in the treatment of rGERD. The results of this study will provide a reliable clinical basis for selecting botanical drug treatments for patients with rGERD. Background: The impact of cerebral edema on brain cells and ventricles in cardiac arrest patients can manifest as effacement of cortical sulci, diminished ventricle size, altered gray matter to white matter ratio (GWR), and increased optic nerve sheath diameter (ONSD) in brain CT scans. However, a complete investigation of GWR in whole lobes, quantitative cistern size, and comprehensive comparison of various brain CT parameters has not been conducted. This study aimed to comprehensively compare various early brain CT parameters along with conventional significant variables in relation to poor neurological outcome and diffuse cortical necrosis. Methods: This retrospective study included 86 adult patients with cardiac arrest who underwent brain CT/MRI. GWRs, the distance of the posterior ambient cistern, and ONSD in early brain CT and regions of interest (ROIs) in brain MRI were measured and analyzed along with clinical characteristics. Results: ROIs in the putamen and parietal white matter showed significant differences (p = 0.05, p = 0.022, respectively). The distance of the posterior ambient cistern and the GWR of the putamen and parietal white matter were newly developed predictors that were not used previously and demonstrated a significant correlation with the presence of diffuse cortical necrosis (OR 0.4, p = 0.006, AUC 0.637; OR 0.478, p = 0.02, AUC 0.603, respectively) or poor neurological outcomes (AUC 0.637, AUC 0.603, respectively), but were not more significant than pupil reflex (OR 0.06, p

Introduction: Motilin (MLN) is a gastrointestinal (GI) hormone produced in the upper small intestine. Its most well understood function is to participate in Phase III of the migrating myoelectric complex component of GI motility. Changes in MLN availability are associated with GI diseases such as gastroesophageal reflux disease and functional dyspepsia. Furthermore, herbal medicines have been used for several years to treat various GI disorders. We systematically reviewed clinical and animal studies on how herbal medicine affects the modulation of MLN and subsequently brings the therapeutic effects mainly focused on GI function.Methods: We searched the PubMed, Embase, Cochrane, and Web of Science databases to collect all articles published until 30 July 2023, that reported the measurement of plasma MLN levels in human randomized controlled trials and in vivo herbal medicine studies. The collected characteristics of the articles included the name and ingredients of the herbal medicine, physiological and symptomatic changes after administering the herbal medicine, changes in plasma MLN levels, key findings, and mechanisms of action. The frequency patterns (FPs) of botanical drug use and their correlations were investigated using an FP growth algorithm.Results: Nine clinical studies with 1,308 participants and 20 animal studies were included in the final analyses. Herbal medicines in clinical studies have shown therapeutic effects in association with increased levels of MLN, including GI motility regulation and symptom improvement. Herbal medicines have also shown anti-stress, anti-tumor, and anti-inflammatory effects in vivo. Various biochemical markers may correlate with MLN levels. Markers may have a positive correlation with plasma MLN levels included ghrelin, acetylcholine, and secretin, whereas a negative correlation included triglycerides and prostaglandin E2. Markers, such as gastrin and somatostatin, did not show any correlation with plasma MLN levels. Based on the FP growth algorithm, Glycyrrhiza uralensis and Paeonia japonica were the most frequently used species.Conclusion: Herbal medicine may have therapeutic effects mainly on GI symptoms with involvement of MLN regulation and may be considered as an alternative option for the treatment of GI diseases. In synthetic biology, it is a challenge to increase the production of target proteins by maximizing their expression levels. In order to augment expression levels, we need to focus on both homologous recombination and codon adaptation, which are estimated by three objective functions, namely HD (Hamming distance), LRCS (length of repeated or common substring) and CAI (codon adaptation index). Optimizing these objective functions simultaneously becomes a multi-objective optimization problem. The aim is to find satisfying solutions that have high codon adaptation and a low incidence of homologous recombination. However, obtaining satisfactory solutions requires calculating the objective functions multiple times with many cycles and solutions. In this paper, we propose an approach to accelerate the method of designing a set of CDSs (CoDing sequences) based on NSGA-II (non-dominated sorting genetic algorithm II) on NVIDIA GPUs. The implementation accelerated by GPUs improves overall performance by 187.5$ \times $ using $ 100 $ cycles and $ 128 $ solutions. Our implementation allows us to use larger solutions and more cycles, leading to outstanding solution quality. The improved implementation provides much better solutions in a similar amount of time compared to other available methods by 1.22$ \times $ improvements in hypervolume. Furthermore, our approach on GPUs also suggests how to efficiently utilize the latest computational resources in bioinformatics. Finally, we discuss the impacts of the number of cycles and the number of solutions on designing a set of CDSs.

Purpose: This study aims to examine the performance of early warning scoring systems regarding adverse events of unanticipated clinical deterioration in complementary and alternative medicine hospitals. Methods: A medical record review of 500 patients from 5-year patient data in two traditional Korean medicine hospitals was conducted. Unanticipated clinical deterioration events included unexpected in-hospital mortality, cardiac arrest, and unplanned transfers to acute-care conventional medicine hospitals. Scores of the Modified Early Warning Score (MEWS), National Early Warning Score (NEWS), and National Early Warning Score 2 (NEWS2) were calculated. Their performance was evaluated by calculating areas under the receiver-operating characteristic curve for the event occurrence. Multiple logistic regression analyses were performed to determine the factors associated with event occurrence. Results: The incidence of unanticipated clinical deterioration events was 1.1% (225/21101). The area under the curve of MEWS, NEWS, and NEWS2 was .68, .72, and .72 at 24 hours before the events, respectively. NEWS and NEWS2, with almost the same performance, were superior to MEWS (p = .009). After adjusting for other variables, patients at low-medium risk (OR = 3.28; 95% CI = 1.02–10.55) and those at medium and high risk (OR = 25.03; 95% CI = 2.78–225.46) on NEWS2 scores were more likely to experience unanticipated clinical deterioration than those at low risk. Other factors associated with the event occurrence included frailty risk scores, clinical worry scores, primary medical diagnosis, prescribed medicine administration, acupuncture treatment, and clinical department. Conclusions: The three early warning scores demonstrated moderate-to-fair performance for clinical deterioration events. NEWS2 can be used for early identification of patients at high risk of deterioration in complementary and alternative medicine hospitals. Additionally, patient, care, and system factors need to be considered to improve patient safety.

Abstract G-protein coupled receptors (GPCRs) and ion channels serve as key molecular switches through which extracellular stimuli are transformed into intracellular effects, and it has long been postulated that ion channels are direct effector molecules of the alpha subunit of G-proteins (Gα). However, no complete structural evidence supporting the direct interaction between Gα and ion channels is available. Here, we present the cryo-electron microscopy structures of the human transient receptor potential canonical 5 (TRPC5)-Gαi3 complexes with a 4:4 stoichiometry in lipid nanodiscs. Remarkably, Gαi3 binds to the ankyrin repeat edge of TRPC5 ~ 50 Å away from the cell membrane. Electrophysiological analysis shows that Gαi3 increases the sensitivity of TRPC5 to phosphatidylinositol 4,5-bisphosphate (PIP2), thereby rendering TRPC5 more easily opened in the cell membrane, where the concentration of PIP2 is physiologically regulated. Our results demonstrate that ion channels are one of the direct effector molecules of Gα proteins triggered by GPCR activation–providing a structural framework for unraveling the crosstalk between two major classes of transmembrane proteins: GPCRs and ion channels.

This paper deals with the torque vectoring control problem for uncertain in-wheel motor vehicles. The main objective of this study is to enhance the cornering performance of vehicles at high speed driving and this is done by generating possibly different motor torques applied to wheels. The proposed controller consists of a model predictive control based outer-loop controller and a disturbance observer based inner-loop controller. The outer-loop controller finds the optimal longitudinal tire force considering state and input constraints. The inner-loop controller estimates the difference between the nominal tire model that is used by the outer-loop controller and the unmodeled disturbance, and compensates the disturbance so that the actual vehicle follows the nominal model. The proposed controller is validated via numerical experiments for circular driving, double lane change, and skidpad scenarios and it is shown that the state tracking performance is improved by using the proposed controller.

Introduction Postoperative nausea and vomiting (PONV) is a distressing symptom that patients often complain of even after less invasive surgery such as laparoscopic surgery (LS). If PONV is not well managed, patient recovery and postoperative quality of life are adversely affected. Although various drugs have been administered to prevent PONV, their effectiveness is limited, and adverse effects are numerous. Although herbal medicines have been widely used to manage various gastrointestinal symptoms, including nausea and vomiting, scientific evidence of their effects is lacking. This protocol is intended for a systematic review to analyse the efficacy and safety of Chinese herbal medicines for PONV after LS through a meta-analysis.Methods and analysis Randomised controlled trials, reported until June 2022, will be retrieved from electronic databases such as Medline, EMBASE and Cochrane Library. We will compare the effects of herbal medicine in patients presenting with PONV after LS with those of Western medicine, placebo and no treatment. If sufficient studies are identified, we will evaluate the combined effects of herbal and Western medicine. The incidence of nausea and vomiting will be considered the primary outcome. Secondary outcomes will include the intensity of complaints, quality of life and incidence of adverse events. Two independent reviewers will collect data based on the Preferred Reporting Items for Systematic Review and Meta-Analyses statement, evaluate the quality of each study using the Cochrane risk-of-bias tool and synthesise the results via meta-analysis, if possible.Ethics and dissemination Ethical approval is not required for this review. Background: The demand for complementary and alternative medicine for the management of functional dyspepsia (FD) is increasing due to the insufficient efficacy of conventional treatment options. In Asia, the Chinese herbal medicine formula Banxia-xiexin tang (BXT) has been used to treat FD.Methods: We searched 11 digital medical databases on 1 September 2021. Randomized controlled trials (RCTs) that investigated the efficacy of BXT or combination therapy (BXT plus Western medicines) for FD were selected. The outcome parameters were total clinical efficacy rate (TCE), motilin level, symptom checklist-90-revised (SCL-90-R), and visual analog scale (VAS) for dyspepsia and adverse events. Cochrane risk of bias tool 2.0 (RoB 2) was used for the quality assessment of included studies.Results: The meta-analysis comprised 57 RCTs with 5,525 participants. BXT was more efficacious, with a higher TCE than Western medicine. Combination therapy (BXT plus Western medicine) also resulted in a higher TCE than Western medicine. Combination therapy improved motilin levels and psychological symptoms to a greater extent than Western medicine, evidenced by a higher SCL-90-R score. However, no significant difference in VAS scores was observed between the BXT and placebo groups. BXT and combination therapy were associated with fewer adverse events than Western medicine or placebo.Conclusion: Our findings suggest that BXT and its combination therapy may be an effective and safe alternative treatment for FD. Background: Current treatment and management options for functional dyspepsia (FD) often fail to alleviate symptoms. Naesohwajung-tang (NHT) is a herbal formula frequently used to treat functional dyspepsia in traditional Korean medicine. However, few animal and case reports on the use of Naesohwajung-tang for functional dyspepsia treatment exist, and the clinical evidence remains deficient.Objectives: This study aimed to evaluate the efficacy of Naesohwajung-tang in patients with functional dyspepsia.Methods: We enrolled 116 patients with functional dyspepsia at two study sites in this 4 weeks, randomized, double-blind, placebo-controlled trial and randomly assigned them to either the Naesohwajung-tang or placebo group. To evaluate the efficacy of Naesohwajung-tang, the primary endpoint was a score on the total dyspepsia symptom (TDS) scale after treatment. The overall treatment effect (OTE), single dyspepsia symptom (SDS) scale, food retention questionnaire (FRQ), Damum questionnaire (DQ), functional dyspepsia-related quality of life (FD-QoL) questionnaire, and gastric myoelectrical activity measured using electrogastrography were evaluated as secondary outcomes. Laboratory tests were performed to confirm the safety of the intervention.Results: The 4 weeks administration of Naesohwajung-tang granules demonstrated a significantly higher reduction in the total dyspepsia symptom (p < 0.05) and a higher degree of improvement in the total dyspepsia symptom (p < 0.01) than the placebo group. Patients who underwent Naesohwajung-tang had a significantly higher overall treatment effect and a greater increase in the degree of improvement in scores such as epigastric burning, postprandial fullness, early satiation, functional dyspepsia-related quality of life, and Damum questionnaire (p < 0.05). Additionally, the Naesohwajung-tang group showed a greater effect in preventing a decrease in the percentage of normal gastric slow waves after meals than the placebo group. As a result of subgroup analyses using the degree of improvement in total dyspepsia symptom, Naesohwajung-tang was found to be more effective than placebo in female, younger patients (

Objective: Although Xiaoyao-san (XYS) is a popular herbal remedy for indigestion, there is insufficient evidence to recommend it as a treatment option for functional dyspepsia (FD). This review aimed to assess the safety and efficacy of XYS in patients with FD, compared to conventional Western medicine (WM).Methods: Two independent reviewers searched for randomized controlled trials (RCTs) using 11 electronic databases, including Medline and Embase, to evaluate therapeutic effects of XYS on FD up to 31 January 2023. The primary outcome was the total clinical efficacy rate (TCE), and secondary outcomes included scores of dyspepsia-related symptoms (DSS) and incidence of adverse events (AEs). The risk of bias was evaluated using the Cochrane collaboration tool, and data synthesis and subgroup analyses were performed using the Review Manager program.Results: Six studies involving 707 participants were included in the meta-analysis. XYS significantly improved TCE compared to WM (RR = 1.15, 95% CI: 1.05, 1.26, p = 0.002) with high heterogeneity (I2 = 59%, p = 0.06). Combination therapy also showed higher TCE than WM alone (RR = 1.22, 95% CI: 1.05, 1.41, p = 0.008), and the heterogeneity was low (I2 = 0%, p = 0.86). The results showed a greater reduction in DSS in the XYS and combination therapy groups than in the WM alone group (SMD = −0.72, 95% CI: −0.90, −0.53, p < 0.00001) with low heterogeneity (I2 = 44%, p = 0.15), especially for abdominal distension and upper abdominal pain. AEs occurred less frequently in the XYS and combination therapy groups than in the WM alone group (RR = 0.20, 95% CI: 0.07, 0.63, p = 0.006), and the heterogeneity was low (I2 = 45%, p = 0.18). The certainty of the evidence for each outcome was rated from “very low” to “high.”Conclusion: This review suggests that XYS is effective and safe for reducing complaints in patients with FD. Dialogue relation extraction identifies semantic relations between entity pairs in dialogues. This research explores a methodology harnessing the potential of prompt-based fine-tuning paired with a trigger-generation approach. Capitalizing on the intrinsic knowledge of pre-trained language models, this strategy employs triggers that underline the relation between entities decisively. In particular, diverging from the conventional extractive methods seen in earlier research, our study leans towards a generative manner for trigger generation. The dialogue-based relation extraction (DialogeRE) benchmark dataset features multi-utterance environments of colloquial speech by multiple speakers, making it critical to capture meaningful clues for inferring relational facts. In the benchmark, empirical results reveal significant performance boosts in few-shot scenarios, where the availability of examples is notably limited. Nevertheless, the scarcity of ground-truth triggers for training hints at potential further refinements in the trigger-generation module, especially when ample examples are present. When evaluating the challenges of dialogue relation extraction, combining prompt-based learning with trigger generation offers pronounced improvements in both full-shot and few-shot scenarios. Specifically, integrating a meticulously crafted manual initialization method with the prompt-based model—considering prior distributional insights and relation class semantics—substantially surpasses the baseline. However, further advancements in trigger generation are warranted, especially in data-abundant contexts, to maximize performance enhancements.

When X-ray energy above 8 MV is used, photoneutrons are generated by the photonuclear reaction, which activates the components of linear accelerator (linac). Safely managing the radioactive material, when disposing linac or replacing components, is difficult, as the standards for the radioactive material management are not clear in Korea. We surveyed the management status of radioactive components occurred from medical linacs in Korea. And we also measured the activation of each part of the discarded Elekta linac using a survey meter and portable High Purity Germanium (HPGe) detector. We found that most medical institutions did not perform radiation measurements when disposing of radioactive components. The radioactive material was either stored within the institution or collected by the manufacturer. The surface dose rate measurements showed that the parts with high surface dose rates were target, primary collimator, and multileaf collimator (MLC). 60Co nuclide was detected in most parts, whereas for the target, 60Co and 184Re nuclides were detected. Results suggest that most institutions in Korea did not have the regulations for disposing radioactive waste from linac or the management procedures and standards were unclear. Further studies are underway to evaluate short-lived radionuclides and to lay the foundation for radioactive waste management from medical linacs.

A 64-channel scintillation fiber-based real-time monitoring system (SFRMS) was developed and evaluated to verify the dwell position and dwell time of the radioactive source in high-dose-rate brachytherapy. The measurement system consists of 64 sensors installed around a tandem applicator. The distal end of the readout system was fiber-optically connected to the measurement system. The data readout system consisted of a photomultiplier tube and a measurement and automation explorer. The SFRMS accuracy was evaluated in terms of dwell position, output linearity, and dwell time using an afterloader with a 192Ir source. Additionally, a Gafchromic EBT3 film was inserted into the applicator to compare fiber measurements. Furthermore, a clinical treatment plan and silicon-based anthropomorphic phantom were utilized to evaluate the accuracy of the dwell positions and dwell times of the source in a situation mimicking a real treatment. The measured output signals from each sensor were fit with Gaussian and inverse-square functions to determine the exact source dwell positions. The difference in dwell position between the plans and the estimated data was −0.63 ± 0.25 mm for the Gaussian fitting and 0.72 ± 0.26 mm for the inverse-square fitting. The difference in dwell time between the plans and the estimated data was within 0.1 sec. For the measurements of the clinical plan on the silicon-based anthropomorphic phantom, the difference in dwell position between the plans and the estimated data was less than 0.88 ± 0.29 mm for the Gaussian fitting and less than 0.85 ± 0.36 mm for the inverse-square fitting. The SFRMS can accurately detect source dwell positions and dwell times and provide real-time output.

Abstract In recent years, natural gas is increasingly being used in the heating and power generation sectors as a clean fuel with an aim to reduce air pollution. In this study, a standard test method was used to measure air pollutants and identify emission characteristics for gas turbines and small domestic boilers, which use LNG as fuel. For gas turbines, the air pollutants were measured at 14 sites, whereas for small domestic boilers, six of them were installed in a laboratory to run tests due to limitations in on-site measuring and testing. However, the small domestic boilers were all new machines and were operated for long consecutive hours for testing, meaning that the results could vary from that of on-site boilers. The results show that gas turbines and small domestic boilers not only emit PM2.5, but also particulate matters larger than PM2.5. According to the measurements, the average concentration level of PMtotal, PM10, and PM2.5 generated from gas turbines are 51.8, 38.5, and 28.1 µg/m3 (@O2 15%), respectively. Those generated from small domestic boilers were 31.3, 26.2, and 20.0 µg/m3 (@O2 4%), respectively. The NOx concentration levels complied with the emission limits. Especially where a NOx control device was in place, both the NOx and CO concentration levels were relatively low. However, the NOx and CO concentration levels were generated from small domestic boilers were relatively high, since the emission limits were not applied. VOCs were measured at 10 facilities where 28 samples were collected. The compounds that were identified were Aromatics, Oxygenated VOCs, Alkanes, in that order, which were consistent across the samples. Aromatics consisted mostly of toluene, o,m,p-xylenes, benzene, and ethylbenzene. Among oxygenated VOCs, ethyl acetate, vinyl acetate, and isopropyl alcohol, etc. were identified. In other words, gas turbines generated a wider range and higher concentration levels of VOCs compared to small domestic boilers. The emission factors of gas turbines and small domestic boilers were derived from the measurements, and then compared with the standard emission factors of other countries (NAER, U.S. EPA AP-42, EMEP/EEA). PM emission factors calculated in this study were lower than that of existing emission factors and the calculated NOx emission factors (uncontrolled) for the small boilers were also lower. The CO emission factor for gas turbines was lower than that of existing emission factors, but higher for the small domestic boilers. Emission factors of benzene, toluene, and xylenes, which are hazardous air pollutants, were lower than those of U.S. EPA AP-42.

Purpose Dedicated breast CT is an emerging volumetric X-ray imaging modality for diagnosis that does not require any painful breast compression. To improve the detection rate of weakly enhanced lesions, an adaptive image rescaling (AIR) technique was proposed. Materials and Methods Two disks containing five identical holes and five holes of different diameters were scanned using 60/100 kVp to obtain single-energy CT (SECT), dual-energy CT (DECT), and AIR images. A piece of pork was also scanned as a subclinical trial. The image quality was evaluated using image contrast and contrast-to-noise ratio (CNR). The difference of imaging performances was confirmed using student’s t test. Results Total mean image contrast of AIR (0.70) reached 74.5% of that of DECT (0.94) and was higher than that of SECT (0.22) by 318.2%. Total mean CNR of AIR (5.08) was 35.5% of that of SECT (14.30) and was higher than that of DECT (2.28) by 222.8%. A similar trend was observed in the subclinical study. Conclusion The results demonstrated superior image contrast of AIR over SECT, and its higher overall image quality compared to DECT with half the exposure. Therefore, AIR seems to have the potential to improve the detectability of lesions with dedicated breast CT.

The herbal medicine Yukgunja-tang has been widely used for the treatment of functional dyspepsia (FD) in the clinical setting of East Asian traditional medicine. This paper presents a protocol for a randomized, assessor-blind, controlled, multicenter, three-arm parallel clinical trial comparing the effectiveness, safety, and cost-effectiveness of Yukgunja-tang with Pyeongwi-san and usual care. A total of 140 participants with Rome IV-diagnosed FD will be randomly assigned to either the Yukgunja-tang (n = 56), Pyeongwi-san (n = 56), or usual care (n = 28) groups. All participants will be educated on dietary guidelines for FD patients. Participants in the Yukgunja-tang and Pyeongwi-san groups will take investigational products for 6 weeks. All participants will be assessed for clinical parameters at weeks 0, 3, 6, 9, and 24. The primary outcome will be measured on the total dyspepsia symptom scale, and the secondary outcome will include the single dyspepsia symptom scale, overall treatment effect, the visual analog scale for dyspepsia, FD-related quality of life, hospital anxiety and depression scale, EuroQol-5 dimension, pattern identification, and serum levels of acyl-ghrelin and deacyl-ghrelin. Adverse events and laboratory tests will be monitored for safety assessment. The results will provide evidence of the effectiveness, safety, and cost-effectiveness of Yukgunja-tang in the treatment of FD.

Gastrointestinal motility disorder (GMD) is a disease that causes digestive problems due to inhibition of the movement of the gastrointestinal tract and is one of the diseases that reduce the quality of life of modern people. Smilacis Glabrae Rhixoma (SGR) is a traditional herbal medicine for many diseases and is sometimes prescribed to improve digestion. As a network pharmacological approach, we searched the TCMSP database for SGR, reviewed its constituents and target genes, and analyzed its relevance to gastrointestinal motility disorder. The effects of the SGR extract on the pacemaker activity in interstitial cells of Cajal (ICC) and gastric emptying were investigated. In addition, using the GMD mouse model through acetic acid (AA), we investigated the locomotor effect of SGR on the intestinal transit rate (ITR). As a result of network pharmacology analysis, 56 compounds out of 74 candidate compounds of SGR have targets, the number of targets is 390 targets, and there are 904 combinations. Seventeen compounds of SGR were related to GMD, and as a result of comparing the related genes with the GMD-related genes, 17 genes (active only) corresponded to both. When looking at the relationship network between GMD and SGR, it was confirmed that quercetin, resveratrol, SCN5A, TNF, and FOS were most closely related to GMD. In addition, the SGR extract regulated the pacemaker activity in ICC and recovered the delayed gastric emptying. As a result of feeding the SGR extract to AA-induced GMD mice, it was confirmed that the ITR decreased by AA was restored by the SGR extract. Through network pharmacology, it was confirmed that quercetin, resveratrol, SCN5A, TNF, and FOS were related to GMD in SGR, and these were closely related to intestinal motility. Based on these results, it is suggested that SGR in GMD restores digestion through the recovery of intestinal motility.

Background: Irritable bowel syndrome (IBS) is a common disorder with abdominal pain and bowel habits changes. Herbal medicines (HMs) are frequently used in the treatment of IBS. Therefore, several systematic reviews (SRs) have been conducted to assess the efficacy and safety of HM in IBS patients. This study aimed to investigate the methodology and quality of evidence of SRs, and to describe the current state of research and evidence for the treatment of IBS with HM.Methods: SRs published up to January 2022 were searched using six electronic databases. SRs and/or meta-analyses on the use of HMs for IBS were included. The effects of placebo, conventional medicine (CM), and probiotics were compared with those of HMs. Two investigators independently extracted the data and assessed methodological quality using the Measure Tool to Assessment System Reviews 2 (AMSTAR 2). Grading of Recommendations, Assessment, Development, and Evaluation (GRADE) was used to evaluate the quality of evidence for each main finding.Results: Eighteen SRs were included in this overview. Among them, eight SRs reported only specific subtypes of IBS: six SRs reported patients with diarrhea-predominant IBS, and two SRs reported patients with constipation-predominant IBS. In terms of total efficacy, HM was more effective than placebo, CM, or probiotics. HM showed a more significant effect than CM in relieving independent IBS symptom score (abdominal pain score, diarrhea score, abdominal distension score, stool frequency score, etc.) and recurrence rate. The rate of adverse events was significantly lower with HM compared to CM, and no serious adverse events were reported with HM treatment. According to AMSTAR 2, the methodological quality of the included SRs was extremely low. Furthermore, the quality of evidence for total efficacy was considered low or very low according to the GRADE tool.Conclusion: HM can be considered as an effective and safe treatment for IBS. However, the methodological quality of the included SRs and the quality of evidence was generally low. Therefore, well-designed randomized controlled trials are needed in the future so that a high-quality SR can be used to better assess the safety and efficacy of HM in the treatment of IBS.Systematic Review Registration:https://osf.io/nt6wz, identifier 10.17605/OSF.IO/NT6WZ.

In this study, we present methods of optimization-based power system state estimation over sensor networks. By minimizing a composite loss function while ensuring that the state, disturbance, and measurement noise constraints are satisfied, the best or better state estimates are iteratively computed. The proposed distributed computational methods for power system state estimation are based on operator splitting. Our methods are computationally decomposable over sensor networks, so distributed and parallel computing can be applied. They can systematically handle the constraints of the state variables and noise as well as disturbances, such that the negative effects of bad data and parametric model uncertainty can automatically be reduced in the estimation. For demonstration, the IEEE 118-bus power system dynamic state estimation problem is considered. The results are compared to the ones obtained from a distributed extended Kalman filter. It is shown that compared with a distributed extended Kalman filter, the proposed method achieves improved robustness against adversarial data defection.

Tokenization is a significant primary step for the training of the Pre-trained Language Model (PLM), which alleviates the challenging Out-of-Vocabulary problem in the area of Natural Language Processing. As tokenization strategies can change linguistic understanding, it is essential to consider the composition of input features based on the characteristics of the language for model performance. This study answers the question of “Which tokenization strategy enhances the characteristics of the Korean language for the Named Entity Recognition (NER) task based on a language model?” focusing on tokenization, which significantly affects the quality of input features. We present two significant challenges for the NER task with the agglutinative characteristics in the Korean language. Next, we quantitatively and qualitatively analyze the coping process of each tokenization strategy for these challenges. By adopting various linguistic segmentation such as morpheme, syllable and subcharacter, we demonstrate the effectiveness and prove the performance between PLMs based on each tokenization strategy. We validate that the most consistent strategy for the challenges of the Korean language is a syllable based on Sentencepiece.

For the medical linear accelerators (linac) that utilize more than 10 MV of photon energy, components inside the linac head become radioactivate during the 10–15-year operating cycle. Prior to disposal, radioactive waste must be evaluated for activity, and the same procedure should be followed for medical linacs. In the Republic of Korea, regulation and methodology for the radioactivity evaluation for the medical linac is not established yet. In this study, we employed gamma spectroscopy and a survey meter for evaluating the radioactivity of medical linac components. The components of the Siemens linac considered in this study were classified after decommissioning, and dose rates were measured to up to a 5 cm distance from the component surfaces by using a survey meter. Radionuclides from components were detected using an in situ HPGe detector. Based on the type of radionuclides and dose rate, we estimated the radioactivity of the components. We studied the feasibility of the methodology for disposing of radioactive components by using the in situ HPGe detector.

In the forthcoming era of IoT, where everything will be connected, mobile devices will play a key role in providing data sharing and user-centric services between devices. In such a service environment, if a mobile application is vulnerable to security threats and exposed to malicious behavior, malware can spread to hundreds of millions of connected devices. In particular, it is important to isolate and respond quickly to malicious mobile code. This requires the prediction of malicious behavior. Currently, security risk assessment schemes based on the permission use the description of the application or user review, but these schemes mostly offer a subjective evaluation, which inevitably reduces accuracy. In this paper, we thus propose a scheme for assessing security risk of Android mobile applications by analyzing their application programming interfaces (APIs) using machine learning. The key idea of the proposed scheme is to extract the APIs from the execution code of the application with reverse engineering analysis, such that each API can be compared with the malicious API database built from the existing malware dataset. Instead of simply judging the applications as malicious or benign, our scheme shows their risk as a score. To do this quantitative evaluation, we use an ensemble of tree boosting machine learning algorithms. To prove the practicality of the proposed scheme, we experiment with a set of benign and malicious real world samples, and compare our results with existing schemes. Experimental results show better performance and accuracy than conventional schemes based on Naive Bayes and simple ensemble algorithms. Our proposed scheme is expected to significantly contribute in responding rapidly to ever-more-intelligent malware of the future.

This article presents an economic nonlinear hybrid model predictive control strategy for optimal energy management of parallel hybrid electric vehicles. Hybrid electric vehicles are controlled for operation in various driveline modes and the associated optimal control problem involves both continuous and discrete control variables. To solve the resultant mixed-integer nonlinear optimal control problem, we propose a hierarchical supervisory control architecture that consists of demand prediction, driveline mode determination, and real-time optimization. These three modules are designed independently and connected in series to perform computer-aided control. The demand prediction module uses a times series model to forecast the mechanical traction power requests of the driver over a prediction horizon based on vehicle speed, road grade, acceleration pedal scale, brake pedal scale, and past and current power demands. For a given forecasted power demand profile, the mode determination module decides a sequence of driveline modes that are presumed to be operated over the prediction horizon. The model-based real-time optimization corresponding to nonlinear model predictive control computes the optimal motor power over a prediction horizon, and the receding horizon scheme as feedback control is applied to repeat the processes of the three control modules. A dedicated case study with real driving data obtained from Hyundai IONIQ PHEV 2018 is presented to demonstrate the effectiveness in fuel economy and emission reduction offered by the proposed optimal energy management strategy. The proposed hierarchical real-time predictive optimization-based strategy is competitive with any exiting power management strategies such as dynamic programming and equivalent consumption minimization strategy in fuel economy and emission reduction while showing better charge-sustaining capability. This trade-off between fuel economy and charge-sustainability can be further improved by tuning the hyper-parameters in the proposed optimal control problem.

The pathophysiology of functional gastrointestinal disorders (FGIDs) is still unclear and various complex mechanisms have been suggested to be involved. In many cases, improvement of symptoms and quality of life (QoL) in patients with FGIDs is difficult to achieve with the single-targeted treatments alone and clinical application of these treatments can be challenging owing to the side effects. Herbal preparations as complementary and alternative medicine can control multiple treatment targets of FGIDs simultaneously and relatively safely. To date, many herbal ingredients and combination preparations have been proposed across different countries and together with a variety of traditional medicine. Among the herbal therapies that are comparatively considered to have an evidence base are iberogast (STW-5) and peppermint oil, which have been mainly studied and used in Europe, and rikkunshito and motilitone (DA-9701), which are extracted from natural substances in traditional medicine, are the focus of this review. These herbal medications have multi-target pharmacology similar to the etiology of FGIDs, such as altered intestinal sensory and motor function, inflammation, neurohormonal abnormality, and have displayed comparable efficacy and safety in controlled trials. To achieve the treatment goal of refractory FGIDs, extensive and high quality studies on the pharmacological mechanisms and clinical effects of these herbal medications as well as efforts to develop new promising herbal compounds are required.

The development of treatment for neurodegenerative diseases (NDs) such as Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, and amyotrophic lateral sclerosis is facing medical challenges due to the increasingly aging population. However, some pharmaceutical companies have ceased the development of therapeutics for NDs, and no new treatments for NDs have been established during the last decade. The relationship between ND pathogenesis and risk factors has not been completely elucidated. Herein, we review the potential involvement of transient receptor potential (TRP) channels in NDs, where oxidative stress and disrupted Ca2+ homeostasis consequently lead to neuronal apoptosis. Reactive oxygen species (ROS) -sensitive TRP channels can be key risk factors as polymodal sensors, since progressive late onset with secondary pathological damage after initial toxic insult is one of the typical characteristics of NDs. Recent evidence indicates that the dysregulation of TRP channels is a missing link between disruption of Ca2+ homeostasis and neuronal loss in NDs. In this review, we discuss the latest findings regarding TRP channels to provide insights into the research and quests for alternative therapeutic candidates for NDs. As the structures of TRP channels have recently been revealed by cryo-electron microscopy, it is necessary to develop new TRP channel antagonists and reevaluate existing drugs.

PurposeRestricted shoulder mobility is a major upper extremity dysfunction associated with lower quality of life and disability after breast cancer surgery. We hypothesized that a poloxamer and sodium alginate mixture (Guardix-SG®) applied after axillary lymph node dissection (ALND) would significantly improve shoulder range of motion (ROM) in patients with breast cancer.MethodsWe conducted a double-blind, randomized, prospective study to evaluate the clinical efficacy and safety of Guardix-SG® for the prevention of upper extremity dysfunction after ALND. The primary outcome measure was shoulder ROM at baseline (T0) and 3 (T1), 6 (T2), and 12 months (T3) after surgery. Secondary outcome measures were the Disabilities of the Arm, Shoulder, and Hand score(DASH), pain associated with movement, which was assessed using a numeric rating scale, and lymphedema assessed using body composition analyzer.ResultsA total of 83 women with breast cancer were randomly assigned to either the Guardix-SG® group or the control group. In the Guardix-SG® group (n = 37), Guardix-SG® was applied to the axillary region after ALND. In the control group (n = 46), ALND was performed without using Guardix-SG®. Comparing ROM for shoulder flexion before surgery (178.2°) and 12 months after surgery (172.3°), that was restored 12 months after surgery in the Guardix-SG® group, and there was no statistically significant difference between that at before surgery and 12 months after surgery (p = 0.182). No adverse effect was observed in either group.ConclusionsThe results of this study have shown that Guardix-SG® help improve shoulder ROM without causing adverse effects in patients who underwent breast cancer surgery. However, there was no statistically significant difference from the control group. BackgroundThis study investigated the effect of accumulated doses on radio-photoluminescence glass dosimeters (RPLGDs) from measurements involving mega-voltage photons.MethodsForty-five commercially available RPLGDs were irradiated to estimate their dose responses. Photon beams of 6, 10, and 15 MV were irradiated onto the RPLGDs inside a phantom, which were divided into five groups with different doses and energies. Groups 1 and 2 were irradiated at 1, 5, 10, 50, and 100 Gy in a sequential manner; Group 3 was irradiated 10 times with a dose of 10 Gy; and Groups 4 and 5 followed the same method as that of Group 3, but with doses of 50 Gy and 100 Gy, respectively. Each device was subjected to a measurement reading procedure each time irradiation.ResultsFor the annealed Group 1, RPLGD exhibited a linearity response with variance within 5%. For the non-annealed Group 2, readings demonstrated hyperlinearity at 6 MV and 10 MV, and linearity at 15 MV. Following the 100 Gy irradiation, the readings for Group 2 were 118.7 ± 1.9%, 112.2 ± 2.7%, and 101.5 ± 2.3% at 6, 10, and 15 MV, respectively. For Groups 3, 4, and 5, the responsiveness of the RPLGDs gradually decreased as the number of repeated irradiations increased. The percentage readings for the 10th beam irradiation with respect to the readings for the primary beam irradiation were 84.6 ± 1.9%, 87.5 ± 2.4%, and 93.0 ± 3.0% at 6 MV, 10 MV, and 15 MV, respectively.ConclusionsThe non-annealed RPLGD response to dose was hyperlinear for the 6 MV and 10 MV photon beams but not for the 15 MV photon beam. Additionally, the annealed RPLGD exhibited a fading phenomenon when the measurement was repeated several times and demonstrated a relatively large fading effect at low energies than at high energies.

In a cone-beam computed tomography (CT) system, the Feldkamp, Davis, and Kress (FDK) algorithm produces cone-beam artifacts due to insufficient object sampling in the missing cone region of frequency space. While total variation minimization-based iterative reconstruction (TV-IR) may reduce cone-beam artifacts by filling in the missing cone region, it introduces image blurring or noise increase depending on the regularization parameter. In this paper, we propose a method to reduce cone-beam artifacts through an optimal combination of FDK and TV-IR images. The method utilizes FDK (TV-IR) data outside (inside) the missing cone region, which enables to keep the original image quality of the FDK image and preserve the advantages of the TV-IR image for cone-beam artifact reduction. To validate the proposed method, we used numerical phantoms composed of Defrise disks, vertical plates, and star objects and assessed the image quality of FDK, TV-IR, and the proposed method using the mean squared error, contrast to noise ratio, and structural similarity with different TV-IR regularization parameters. Experimental validation was also conducted using a spine phantom with a bench-top cone-beam CT system. The results showed that the performance of the cone-beam artifacts reduction in TV-IR depended on the value of the regularization parameter, which often produced suboptimal image quality for different imaging tasks. However, the proposed method provided good image quality regardless of the regularization parameter values.

Due to their extraordinary electrical and physical properties, two-dimensional (2D) transition metal dichalcogenides (TMDs) are considered promising for use in next-generation electrical devices. However, the application of TMD-based devices is limited because of the Schottky barrier interface resulting from the absence of dangling bonds on the TMDs’ surface. Here, we introduce a facile phase-tuning approach for forming a homogenous interface between semiconducting hexagonal (2H) and semi-metallic monoclinic (1T′) molybdenum ditelluride (MoTe2). The formation of ohmic contacts increases the charge carrier mobility of MoTe2 field-effect transistor devices to 16.1 cm2 V−1s−1 with high reproducibility, while maintaining a high on/off current ratio by efficiently improving charge injection at the interface. The proposed method enables a simple fabrication process, local patterning, and large-area scaling for the creation of high-performance 2D electronic devices.

Abstract Background In the treatment of functional dyspepsia, the placebo effect has been reported to be high, and the influence of the patient-practitioner relationship may be a major component of this effect. The specific and non-specific effects of acupuncture cannot be easily distinguished, and the patient-practitioner relationship may influence the total therapeutic effect in clinical practice. There have been no studies that investigate the influence of patient-practitioner relationship on acupuncture treatment for patients with functional dyspepsia. Methods Patients with postprandial distress syndrome, a functional dyspepsia subtype, will be recruited at three hospitals (two in Korea and one in USA) for an international, multi-center, randomized, patient/assessor-blinded, clinical trial. The total anticipated sample size is 88. The participants will be randomly allocated into two groups: an augmented interaction group and a limited interaction group. Acupuncture, with total 12 acupoints, will be performed twice weekly for 4 weeks in both groups. Trained practitioners will provide an “augmented” or “limited” interaction context, as determined by random allocation. The primary outcome measure is the proportion of responders, the proportion of participants who answer “yes” to more than half of the adequate relief questions during the study. Secondary outcome measures include questionnaires for quality of life and symptoms of dyspepsia, and maximum tolerable volume of nutrient drink test. Data will be collected at baseline and following 4 weeks of acupuncture. Discussion This study will evaluate the influence of the patient-practitioner interaction on clinical effects of acupuncture in patients with functional dyspepsia. ObjectivesWith the increased use of computed tomography (CT) in clinics, dose reduction is the most important feature people seek when considering new CT techniques or applications. We developed an intensity-weighted region-of-interest (IWROI) imaging method in an exact half-fan geometry to reduce the imaging radiation dose to patients in cone-beam CT (CBCT) for image-guided radiation therapy (IGRT). While dose reduction is highly desirable, preserving the high-quality images of the ROI is also important for target localization in IGRT.MethodsAn intensity-weighting (IW) filter made of copper was mounted in place of a bowtie filter on the X-ray tube unit of an on-board imager (OBI) system such that the filter can substantially reduce radiation exposure to the outer ROI. In addition to mounting the IW filter, the lead-blade collimation of the OBI was adjusted to produce an exact half-fan scanning geometry for a further reduction of the radiation dose. The chord-based rebinned backprojection-filtration (BPF) algorithm in circular CBCT was implemented for image reconstruction, and a humanoid pelvis phantom was used for the IWROI imaging experiment.ResultsThe IWROI image of the phantom was successfully reconstructed after beam-quality correction, and it was registered to the reference image within an acceptable level of tolerance. Dosimetric measurements revealed that the dose is reduced by approximately 61% in the inner ROI and by 73% in the outer ROI compared to the conventional bowtie filter-based half-fan scan.ConclusionsThe IWROI method substantially reduces the imaging radiation dose and provides reconstructed images with an acceptable level of quality for patient setup and target localization. The proposed half-fan-based IWROI imaging technique can add a valuable option to CBCT in IGRT applications.

In most cancer patients, chemotherapy-induced oral mucositis (OM) is a frequent side effect, leading to low quality of life and delay in therapy. The aim of this study was to evaluate the effects of Onchung-eum , a well-known herbal prescription in traditional medicine comprising 8 herbs that has long been used for skin diseases, on 5-fluorouracil (5-FU)–induced OM in human pharyngeal cells and golden Syrian hamsters. DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, and reactive oxygen species production were measured in vitro. The effects of Onchung-eum on OM of hamster cheek pouches induced by 5-FU were evaluated histologically and using TUNEL assay. In addition, the expression of nuclear factor-κB, caspase-3, and pro-inflammatory cytokines were measured by immunoblotting and immunohistochemistry. Significantly increased cell viability was observed in the Onchung-eum –treated groups compared with the 5-FU–treated control group. In 500 and 1000 mg/kg Onchung-eum –treated groups, the damaged epithelial layers in the cheek pouches of hamsters were significantly recovered. Moreover, at all concentrations, cell death in the cheek pouches of hamsters in the Onchung-eum –treated groups significantly decreased. The expression of pro-inflammatory cytokines, nuclear factor-κB, and caspase-3 also significantly decreased in Onchung-eum –treated groups at 500 and 1000 mg/kg. In conclusion, this study revealed that Onchung-eum can be used to treat chemotherapy-induced OM. However, further studies are required to understand the underlying mechanisms.

In this study, the performance evaluation was performed by adding a polymer aqueous (PA) solution as a new additive of the red clay binder for use in the rammed-earth construction method. The evaluation items were compressive strength, water erosion, shrinkage, crystal structure, and microstructure. As a result of the experiment, the binder was improved by efficiently bonding the silica particles by the polymerized polymer. It was confirmed that adding a PA solution to red clay enhances the compressive strength, which is further improved when 5 wt% poly(Acrylic acid(AA)-co-Acrylamide(AM)) is added to the PA solution. Microstructural analysis indicated that the addition of a PA solution facilitates effective bonding of the silica particles of red clay to form hydrogen bonding with poly(AA-co-AM) and encourages aggregate formation. Therefore, the study confirmed that PA solution can be applied to satisfy the performance requirements of the rammed-earth construction by improving the durability and strength of the binder.

In this study, we examined the effects of cement fineness, SO3 content, an accelerating agent, and chemical admixtures mixed with unit weights of cement on concrete early strength using concrete mixtures. C24 (characteristic value of concrete, 24 MPa) was used in the experiment conducted. Ordinary Portland cement (OPC), high fineness and SO3 OPC (HFS_OPC), and Early Portland cement (EPC) were selected as the study materials. The unit weights of cement were set to OPC 330, 350, and 380. Further, a concrete mixture was prepared with a triethanolamine (TEA)-based chemical admixture to HFS. A raw material analysis was conducted, and the compressive strength, temperature history, and maturity (D∙h) were examined. Then, the vertical formwork removal time was evaluated according to the criterion of each country. Finally, the time required to develop concrete strength of 5 MPa was estimated. Results showed that the early strength of concrete mixed with HFS and EPC was greater than that exhibited by concrete with an increased unit weight of cement with OPC. In addition, when HFS was used with EPC, its strength developed early, similar to the trend exhibited by EPC, even at low temperatures.

This paper describes an approach for identifying and appending interior design style information stochastically with reference images and a deep-learning model. In the field of interior design, design style is a useful concept and has played an important role in helping people understand and communicate interior design. Previous studies have focused on how the interior design style categories can be defined. On the other hand, this paper focuses on how stochastically recognizing the design style of given interior design reference images using a deep learning-based data-driven approach. The proposed method can be summarized as follows: (1) data preparation based on a general design style definition, (2) implementing an interior design style recognition model using a pre-trained VGG16 model, (3) training and evaluating the trained model, and (4) demonstration of stochastic detection of interior design styles for reference images. The result shows that the trained model automatically recognizes the design styles of given interior images with probability values. The recognition results, model, and trained image set contribute to facilitating the management and utilization of an interior design references database.

Existing rammed earth construction methods have disadvantages such as increased initial costs for manufacturing the large formwork and increased labor costs owing to the labor-intensive construction techniques involved. To address the limitations of the existing rammed earth construction methods, an autonomous rammed earth construction method was introduced herein. When constructing an autonomous rammed-earth construction method, an alternative means of assuring the performance at the initial age of the binder in terms of materials is needed. In this study, in order to satisfy the performance of the red clay binder, epoxy emulsion was added to analyze the compressive strength, water loosening, shrinkage, rate of mass change, and microstructure in the range of the initial age. As a result of the analysis, the applicability of the epoxy emulsion was confirmed as a new additive for application to an autonomous rammed-earth construction method.

The study of the structure−function relationship of ion channels has been one of the most challenging goals in contemporary physiology. Revelation of the three-dimensional (3D) structure of ion channels has facilitated our understanding of many of the submolecular mechanisms inside ion channels, such as selective permeability, voltage dependency, agonist binding, and inter-subunit multimerization. Identifying the structure−function relationship of the ion channels is clinically important as well since only such knowledge can imbue potential therapeutics with practical possibilities. In a sense, recent advances in the understanding of the structure−relationship of transient receptor potential canonical (TRPC) channels look promising since human TRPC channels are calcium-permeable, non-selective cation channels expressed in many tissues such as the gastrointestinal (GI) tract, kidney, heart, vasculature, and brain. TRPC channels are known to regulate GI contractility and motility, pulmonary hypertension, right ventricular hypertrophy, podocyte injury, seizure, fear, anxiety-like behavior, and many others. In this article, we tried to elaborate recent findings of Cryo-EM (cryogenic-electron microscopy) based structural information of TRPC 4 and 5 channels and domain-specific functions of the channel, such as G-protein mediated activation mechanism, extracellular modification of the channel, homo/hetero-tetramerization, and pharmacological gating mechanisms.

Bifunctional Al2O3/Cu/ZnO catalysts with Al composition of between 30 mol% and 80 mol% were prepared by sequential precipitation (SP) for the conversion of CO2 into dimethyl ether (DME). In the SP synthesis, the concentration of a precipitation agent managed to be high enough to induce the complete precipitation of Al3+. The prepared precipitates were composed of zincian malachite and amorphous AlO(OH). Furthermore, the calcined mixed metal oxide materials of 60% and 80% Al exhibited a higher acidity than commercial Al2O3 and the H2-reduced catalysts showed the similar Cu dispersion of 6%−7% at all Cu loadings. In the activity test at 573 K and 50 bar, the SP-derived catalyst of 80% Al (SP-80) displayed the best performance corresponding to CO2 conversion of 25% and DME selectivity of 75% that are close to equilibrium values. In order to overcome the thermodynamic limitation, a dual-bed catalyst system was made up of SP-80 in the first layer and zeolite ferrierite in the next. This approach enabled DME selectivity to be enhanced to 90% while CO2 conversion increased a little. Consequently, the studied catalyst system based on the SP-derived catalysts can contribute greatly to selective DME production from CO2.

The majority of laboratory animals were transported from commercial breeders to a research facility by ground transportation. During the transportation, many biological functions and systems can be affected by stress. In this experiment, the change of body weight during the transportation was measured and the recovery periods from the transportation stress established based on the body weight changes. Total 676 laboratory animals which were aged between 3 to 9 wk old were studied. The transportation time taken from container packing to unpacking the container was approximately 24 h. Inflammatory bowel disease (IBD) is a chronically relapsing inflammatory disorder of the gastrointestinal tract. Most IBD treatments are unsatisfactory; therefore, various dietary supplements have emerged as promising interventions. Laminaria japonica (LJ) is an edible seaweed used to regulate digestive symptoms. Probiotics have been reported to improve digestive problems and their simultaneous administration with seaweeds has been shown to produce synergistic therapeutic effects. Here, we investigated the effect of LJ combination with probiotics on dextran sodium sulfate-induced colitis model in mice. Aqueous LJ extracts (LJE) at doses from 100 to 300 mg/kg and probiotics at a dose of 300 mg/kg were orally administered for 7 days. Body weight, colon length, histological score, macroscopic damage, and the levels of cytokines IFN-γ, IL-1β, IL-6, IL-10, IL-12 (P40), IL-12 (P70), IL-17, and TNF-α were assessed. LJE alone caused a significant improvement of colitis signs such as colon length, histological score, and IL-1β and IL-6 production. LJE and probiotics demonstrated a synergistic effect by the histological score and levels of IL-1β, IL-6, and IL-12 (P40) but not IFN-γ, IL-10, and IL-12 (P70). In conclusion, LJE was effective in inducing protection against colitis in mice and acted synergistically with probiotics.
