Objective. To investigate the relationship between expression levels of platelet miRNAs and severe pneumonia (SP) in children. Methods. A randomized controlled trial was conducted in 129 children with SP hospitalized from May 2018 to May 2020. All children joined the study group and were divided into the mild infection group, moderate infection group, and severe infection group according to the diagnostic criteria, 43 cases in each group. Besides, 129 healthy children were selected as the control group. The expression levels of platelet miR-223 and miR-192 were detected by real-time quantitative polymerase chain reaction (qPCR). The prothrombin time (PT), activated partial thromboplastin time (APTT), and fibrinogen (FIB) were detected by the Sysmex CA-1500 System (Sysmex Corporation, Japan). The Pearson analysis was conducted to evaluate the correlation between coagulation function and the levels of miR-223 and miR-192. Results. Compared with the control group, miR-223 in the study group was significantly higher and miR-192 was significantly lower ( P < 0.05 ). Compared with the mild infection group, miR-223 was significantly higher and miR-192 was significantly lower in the moderate infection group and severe infection group ( P < 0.05 ). Compared with the control group, PT and APTT were significantly lower and FIB was significantly higher in the study group ( P < 0.05 ). Pearson correlation analysis revealed that miR-223 was positively correlated with PT and APTT ( P < 0.05 ) and negatively correlated with FIB ( P < 0.05 ); miR-192 was negatively correlated with PT and APTT ( P < 0.05 ) and positively correlated with FIB ( P < 0.05 ). Conclusion. miR-223 and miR-192 can reflect coagulation function in children with SP, which can provide a certain reference basis for clinical guidance and treatment and prognosis.

Introduction: Carnosine show robust neuroprotective activity against neurodegenerative diseases. Here, we report that carnosine ameliorates diabetes-associated cognitive decline in vivo through the modulation of autophagy. Methods: A high-fat diet (HFD) and one intraperitoneal injection of 30 mg/kg streptozotocin (STZ) were used to induce type 2 diabetes mellitus in Sprague–Dawley rats. The rats were randomly divided into five groups: Control (CON), HFD/STZ, and three intragastric carnosine treatment groups over 12 weeks. Body weight, blood glucose levels, and cognitive function were continuously monitored. From excised rat hippocampi, we determined SOD activity and MDA levels; carnosine concentration; protein expressions of Akt, mTOR, and the autophagy markers LC3B and P62; and performed histopathological evaluations of the CA1 region. Results: The HFD/STZ groups showed increased blood glucose levels and decreased body weight compared to the CON group. However, there were no significant differences in body weight and blood glucose levels between carnosine-treated and -untreated HFD-STZ-induced diabetic rats. Diabetic animals showed obvious learning and memory impairments in the Morris water maze test compared to the CON group. Compared to those in the HFD/STZ group, carnosine increased SOD activity and decreased MDA levels, increased hippocampal carnosine concentration, increased p-Akt and p-mTOR expression, decreased LC3B and P62 expression, alleviated neuronal injuries, and improved cognitive performance in a dose-dependent manner. Conclusion: Independent of any hyperglycemic effect, carnosine may improve mild cognitive impairments by mitigating oxidative stress, activating the Akt/mTOR pathway, and modulating autophagy in the hippocampus of type 2 diabetic rats.

Working memory (WM) has been a major focus of cognitive science and neuroscience for the past 50 years. While most WM research has centered on the mechanisms of objects, there has been a lack of investigation into the cognitive and neural mechanisms of events, which are the building blocks of our experience. Employing confirmatory factor analysis and resting-state and task fMRI, our study demonstrated for the first time that events have an independent storage space within WM, known as the event cache, with distinct neural correlates compared to object storage in WM. The cerebellar network was found to be the most essential network for event cache, with the left cerebellum Crus I being particularly involved in encoding and maintaining events. Our findings shed critical light on the neuropsychological mechanism of WM by revealing event cache as an independent sub-component of WM and encourage the reconsideration of theoretical models for WM.

In many types of cancer, tumor cells prefer to use glycolysis as a major energy acquisition method. Here, we found that the 18fluoro-deoxyglucose (FDG) positron emission tomography (PET)/computed tomography (CT)-based markers were positively associated with the expression of programmed cell death ligand 1 (PD-L1), pyruvate kinase M2 (PKM2), both of which indicate poor prognosis in patients with pancreatic ductal adenocarcinoma (PDAC). However, the regulatory mechanism of PD-L1 remains elusive. In this study, we confirmed that transforming growth factor-beta1 (TGF-β1) secreted by tumor-associated macrophages (TAMs) was a key factor contributing to the expression of PD-L1 in PDAC cells by inducing the nuclear translocation of PKM2. Using co-immunoprecipitation and chromatin immunoprecipitation assays, we demonstrated that the interaction between PKM2 and signal transducer and activator of transcription 1 (STAT1) was enhanced by TGF-β1 stimulation, which facilitated the transactivation of PD-L1 by the binding of PKM2 and STAT1 to its promoter. In vivo, PKM2 knockdown decreased PD-L1 expression in PDAC cells and inhibited tumor growth partly by promoting natural killer cell activation and function, and the combination of PD-1/PD-L1 blockade with PKM2 knockdown limited tumor growth. In conclusion, PKM2 significantly contributes to TAM-induced PD-L1 overexpression and immunosuppression, providing a novel target for immunotherapies for PDAC.

Neutral nanoparticles (NPs) of copper (Cu), iron (Fe) and zinc (Zn) are widely used in agriculture. Polymer seed coating with different metal NPs may supply important nutrients during plant growth and consequently enhances yields. In this research, three kinds of metal NPs were conducted to optimize the optimal concentration through seed coating for improving plant growth and productivity of tomato. Seeds of Venice tomato cultivars were coated by polymer‐based mixture with different concentrations of Cu, Fe and Zn NPs, respectively. At harvest, seed germination, internode length, average weight of single fruit, yield and fruit shape index were measured. When compared with control, the internode length increased by 7.3% and 6.8% with low concentration of Fe NPs and Zn NPs, respectively. The average weight per fruit improved over control by 10.2% and 7.5% with low concentration of Cu NPs and Fe NPs, respectively. The yield with low concentration of Cu NPs and Fe NPs increased the yield by 10.7% and 6.5% compared with control. These results indicated that polymer seed coating with low concentration of metal NPs may promote the uptake of some nutrient and thus improve the productivity of tomato.

Type 3 innate lymphoid cells (ILC3s) are essential for host defense against infection and tissue homeostasis. However, their role in the development of HCC has not been adequately confirmed. In this study, we investigated the immunomodulatory role of short-chain fatty acids (SCFAs) derived from intestinal microbiota in ILC3 regulation.We report that Lactobacillus reuteri was markedly reduced in the gut microbiota of mice with HCC, accompanied by decreased SCFA levels, especially acetate. Additionally, transplantation of fecal bacteria from wild-type mice or L. reuteri could promote an anticancer effect, elevate acetate levels, and reduce IL-17A secretion in mice with HCC. Mechanistically, acetate reduced the production of IL-17A in hepatic ILC3s by inhibiting histone deacetylase activity, increasing the acetylation of SRY (sex-determining region Y)-box transcription factor 13 (Sox13) at site K30, and decreasing expression of Sox13. Moreover, the combination of acetate with programmed death 1/programmed death ligand 1 blockade significantly enhanced antitumor immunity. Consistently, tumor-infiltrating ILC3s correlated with negative prognosis in patients with HCC, which could be functionally mediated by acetate.These findings suggested that modifying bacteria, changing SCFAs, reducing IL-17A-producing ILC3 infiltration, and combining with immune checkpoint inhibitors will contribute to the clinical treatment of HCC.

Deformable boring bar is the executive component of embedded giant magnetostrictive actuator (GMA), which plays a key role in the output performance of embedded GMA in precision machining of non-cylindrical piston pinhole. In this paper, a multi-parametric coupling design method was presented for deformable boring bar and giant magnetostrictive material. Firstly, the dynamic model of deformable boring bar was built. Second, the performance index of length-diameter ratio was introduced, and the problem of multi-parametric coupling design was solved by using the idea of nonlinear programming. The first-order natural frequency, the end output displacement and the output force of deformable boring bar were taken as the evaluation indexes to ensure the performance requirements of embedded GMA. Finally, according to project requirements and proposed method, an embedded GMA with high frequency response and large output displacement was further designed, which met the performance requirements of displacement and stiffness in precision machining of non-cylindrical piston pinholes and also verified the validity of the design method.

This study is aimed to explore the capacity of metal nanoparticles (NPs) iron, zinc, copper and their combinations introduced in the Murashige–Skoog (MS) nutrient medium (NM) to affect the growth and development of tomato plants (Solanum lycopersicum L.). NPs were prepared by a flow‐levitation method. Metal NPs were characterised by transmission and scanning electron microscopy, X‐ray phase analysis. Average NPs diameters were: iron – 27.0 nm, zinc – 54.0 nm, copper – 79.0 nm. MS NM was modified by substitution of common metal sulphates by neutral metal NPs instead of salts. Tomato seedlings cultivation on NM MS with NPs instead of salts assures improved seedling parameters (root length and root activity) in comparison with plants grown on standard MS. Venice cultivar tomato seedlings grown on NM with metal NPs demonstrated an increase in: seed germination by 10–180%, root length by 10–20%, and root activity by 10 –125%. After 45 days of cultivation, tomato seedlings were transplanted in a greenhouse and were grown up to the harvest. Effects in seed germination and increase of crop mass depended on metal nature and NPs concentration.

BackgroundThe significance of the relationship between the microbiota and diseases is increasingly being recognized. However, the characterization of tumor microbiome and their precise molecular mechanisms through which microbiota promotes hepatocellular carcinoma (HCC) development are still unclear.MethodsThe intrahepatic microbiota was investigated from tumor, normal adjacent tissues in 46 patients with HCC and normal hepatic tissues in 33 patients with hemangioma by 16S rRNA gene sequencing. Taxonomic composition differences in patients were evaluated using Linear discriminant analysis Effect Size (LefSe) and Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt) to predict microbial functional pathways. Associations between the most relevant taxa and clinical characteristics of HCC patients were analyzed by Spearman rank correlations. The effects of microbe on hepatic stellate cells (HSCs) activation and HCC progression were examined.ResultsWe observed intrahepatic microbiota disturbances by reduced microbial diversity in HCC. The tumor microbiota of the HCC patients with cirrhosis showed higher abundance of Stenotrophomonas maltophilia (S. maltophilia). S. maltophilia provoked senescence-associated secretory phenotype (SASP) in HSCs by activating TLR-4-mediated NF-κB signaling pathway, which in turn induced NLRP3 inflammasome complex formation and secreted various inflammatory factors in the liver, thus facilitating HCC progression in mice. Moreover, signs of SASP were also observed in the HSCs in the area of HCC with higher S. maltophilia enrichment arising in patients with cirrhosis.ConclusionsOur analysis of the hepatic microbiota revealed for the first time that patients with HCC exhibited a dysbiotic microbial community with higher S. maltophilia abundance, which induced the expression SASP factors of HSCs and cirrhosis in the liver, concurring in the process of hepatocarcinogenesis.

BackgroundImmune-checkpoint inhibitors (ICIs) remain ineffective in a large group of non-small cell lung cancer (NSCLC) patients. Mucosal-associated invariant T (MAIT) cells, a population of unconventional innate-like T lymphocytes abundant in the human body, play important roles in human malignancies. Little is known about the immune characteristics of MAIT cells in NSCLC and correlation with prognosis and response rate of ICIs treatment.MethodsTo investigate the distribution, activation status, and function of MAIT cells in NSCLC patients and their correlations with anti-PD-1 immunotherapy, MAIT cells in peripheral blood, tumor and paratumor samples from NSCLC patients with or without anti-PD-1 immunotherapy were analyzed using flow cytometry and single-cell RNA-sequencing.ResultsMAIT cells were enriched in the tumor lesions of NSCLC patients migrating from peripheral blood via the CCR6-CCL20 axis. Both peripheral and tumor-infiltrating MAIT cells displayed an exhausted phenotype with upregulated PD-1, TIM-3, and IL-17A while less IFN-γ. Anti-PD-1 therapy reversed the function of circulating MAIT cells with higher expression of IFN-γ and granzyme B. Subcluster MAIT-17s (defined as cells highly expressing exhausted and Th17-related genes) mainly infiltrated in the non-responsive tissues, while the subcluster MAIT-IFNGRs (cells expressing genes related to cytotoxic function) were mainly enriched in responsive tissues. Moreover, we found predictive value of circulating MAIT cells for anti-PD-1 immunotherapy in NSCLC patients.ConclusionsMAIT cells shifted to an exhausted tumor-promoting phenotype in NSCLC patients and the circulating MAIT subset could be a predictor for patients who respond to anti-PD-1 immunotherapy.

Sour or wild jujube fruits and dried seeds are popular food all over the world. In this study, we reported a high-quality genome assembly of sour jujube (Ziziphus jujuba Mill. var. spinosa), with a size of 406 Mbp and scaffold N50 of 30.3 Mbp, which experienced only γ hexaploidization event, without recent genome duplication. Population structure analysis identified four jujube subgroups (two domesticated ones, i.e., D1 in West China and D2 in East/SouthEast China, semi-wild, and wild), which underwent an evolutionary history of a significant decline of effective population size during the Last Glacial Period. The respective selection signatures of three subgroups were discovered, such as strong peaks on chromosomes #3 in D1, #1 in D2, and #4 in wild. Genes under the most significant selection on chromosomes #4 in wild were confirmed to be involved in fruit variations among jujube accessions, in transcriptomic analysis. Our study offered novel insights into the jujube population structure and domestication and provided valuable genomic resources for jujube improvement in stress response and fruit flavor in the future.

In this study, a series of novel compounds were synthesized by adding azo functional groups and chlorine substituent to the central bent-cores to form a 4-chloro-1,3-dizaophenylene bent-core. The structure, mesogenic properties, and photosensitivity of these synthesized compounds were evaluated experimentally. The results show that these compounds exhibit a broad temperature window up to 63.8 °C for nematic phase and fast trans – cis photoisomerization in seconds with 80% conversion. Quantum mechanics modeling confirms that using azos as the central linkages can effectively reduce the molecular dipole moment, which appears to promote favorable mesogenic behaviors and photonic characteristics. Moreover, varying the carbon number in the terminal alkyl chains can alter molecular dipole, especially the polarizability anisotropy, of which the variation is strongly correlated with the phase transition temperature and temperature range of nematic phase. These findings suggest that reducing molecular dipole can promote favorable mesogenic and photonic properties.

In this study, the anatomical and ultrastructural responses of Capsicum annuum to iron nanoparticles (Fe NPs) were determined. The results showed that the bio-effects of Fe NPs on plants could be positive or negative, depending on the additive concentrations. Low concentrations of Fe NPs were found to promote plant growth. Light and electron microscope analyses showed that the Fe NPs promoted plant growth by altering the leaf organization, and increasing the chloroplast number and grana stacking, as well as regulating the development of vascular bundles. Meanwhile, it was found that the Fe NPs could be absorbed in the roots, and then transported to the central cylinder in bio-available forms, where they were translocated and utilized by the leaves and stems. In contrast, high concentrations of Fe NPs appeared to be harmful to the plants, and the majority of Fe NPs were aggregated into cell walls and transported via the apoplastic pathway in the roots, which may potentially block the transfer of iron nutrients. Taken together, the aforementioned data showed that the rational use of Fe NPs could alleviate iron deficiency, and Fe NPs could be an ideal supply for Fe2+ ions fertilizers in agriculture.

Osteosarcoma is the most common primary malignancy of bone in children and the elderly. Recently, more and more researches have demonstrated that Ginsenoside Rg3 (Rg3) is involved in chemotherapy resistance in many cancer, making it a promising Chinese herbal monomer for oncotherapy. In this study, we investigated the efficacy of Rg3 in human osteosarcoma cell lines (MG-63, U-2OS, and SaOS-2). Cell proliferation was measured by CCK8 assay. The migration of cells was examined using the scratch assay method. Quantification of apoptosis was assessed further by flow cytometry. In addition, the expression of apoptosis-related genes (caspase9, caspase3, Bcl2, and Bax) were investigated using RT-PCR. We further investigated the protein level expression of Bcl 2, cleaved-caspase3, and PI3K/AKT/mTOR signaling pathway factors by Western blot assay. Our results revealed that Rg3 inhibited the proliferation and migration of human osteosarcoma cells and induced apoptosis in a concentration- and time-dependent manner. Western blot results showed that Rg3 reduced the protein expression of Bcl2 and PI3K/AKT/mTORbut increased the levels of cleaved-caspase3. Therefore, we hypothesized Rg3 inhibits the proliferation of osteosarcoma cell line and induces their apoptosis by affecting apoptosis-related genes (Bcl2, caspase3) as well as the PI3K/AKT/mTOR signaling pathway. To conclude, Rg3 is a new therapeutic agent against osteosarcoma.

Plant disease control and prevention in microgravity are critical for space plant cultivation. This study investigated the effects of a biocontrol bacterium Pseudochrobactrum kiredjianiae A4 on growth development and antifungal potential of wheat seedlings under simulated microgravity. The growth, antioxidant status and plant immune hormone of both non-infected and infected wheat seedlings were detected before and after inoculation of A4 strains under simulated microgravity condition (μG) and ground condition (1G). Our results showed that bacteria A4 promoted wheat growth by increasing root length and biomass accumulation and meanwhile enhancing fungal disease resistance through improving the antioxidant enzyme activities and plant hormone secretion. Moreover, A4 exhibited a weaker promotion ability on wheat biomass accumulation and disease resistance under μG condition compared to that under 1G. These results not only expand our understanding of the impact of microgravity on plant-microbe interaction, but also provide valuable insights into using plant beneficial microbes for plant cultivation and crop protection in space.

Furan-2-carbohydrazide-5-(2-thienyl)-salicylaldehyde Schiff base (1) was designed and synthesized for the detection of Zn(II) in aqueous solution at neutral pH. Based on its essential excited state intramolecular proton transfer (ESIPT) process, 1 exhibited aggregation-induced emission (AIE) characteristic with a fluorescence emission at 567 nm. Ratiometric fluorescence changes could be observed in H2O/EtOH solutions with different water fraction. After binding to Zn(II) with a molar ratio of 1:1, 1 showed a new fluorescence emission peak at 493 nm and exhibited a 70-fold fluorescence enhancement. Meanwhile, a significant emission color change from orange-red to yellow was observed, enabling detecting Zn(II) by naked eye under illumination with a UV lamp. In addition, chemosensor 1 could distinguish Zn(II) from most of the common metal ions effectively. A linear range of 0.10-10.00 μmol L−1 and a detection limit of 77 nmol L−1 Zn(II) were achieved.

One of the important questions in space biology is the mechanisms underlying plant responses to an outer space environment, i.e., how gene expression is altered in space. In this study, the transcriptome of Arabidopsis thaliana seedlings was analyzed as a part of Germany SIMBOX (science in microgravity box) spaceflight experiment on Shenzhou 8 spacecraft. This experiment involved the following treatments: spaceflight with microgravity (F♣µg), spaceflight with 1g centrifugal force (F 1g), and ground 1g control (G 1g). Gene chips were used to screen gene expression differences in Arabidopsis thaliana seedlings among these treatments. Microarray analysis revealed that 62 1♣g enes were differentially expressed in samples F♣µg vs. G 1g, 249 genes in samples F 1g vs. G 1g, and 368 genes in samples F♣µg vs. F 1g. Gene ontology analysis indicated that the genes were involved in metabolism of stress response, gravitropic response, and DNA damage and repair, suggesting that plants adjust these metabolic pathways to space environmental stress, microgravity, and radiation.

To improve the output performance of embedded giant magnetostrictive actuators (GMAs) for non-circular hole precision machining and to describe the transient nonlinear hysteresis behavior of giant magnetostrictive material (GMM), the magnetostrictive process of GMM is analyzed in detail in this paper. Based on the J–A model and the Gibbs free energy model, a transient multi-field coupling model of GMM is developed by considering the eddy current effect, compression stress variation, and ΔE effect. The simulation results show that the hysteresis loop area increases with increasing driving frequency. The strain of GMM increases first and then decreases with increasing preloading stress. If the stiffness of the deformable bar is too large, the stress of GMM will increase rapidly, thus hindering the elongation of GMM. The simulation process combines the magneto-mechanical coupling model and the dynamic model of embedded GMAs. The simulation results at different excitation frequencies are basically consistent with the experimental data, indicating that the proposed model can predict the output displacement well and provide a theoretical basis for the optimized design of magneto-mechanical coupling for high-performance embedded GMAs.

MicroRNA (miRNA) is a non-coding small RNA composed of 20 to 24 nucleotides that influences plant root development. This study analyzed the miRNA expression in Arabidopsis root tip cells using Illumina sequencing and real-time PCR before (sample 0) and 15 min after (sample 15) a 3-D clinostat rotational treatment was administered. After stimulation was performed, the expression levels of seven miRNA genes, including Arabidopsis miR160, miR161, miR394, miR402, miR403, miR408, and miR823, were significantly upregulated. Illumina sequencing results also revealed two novel miRNAsthat have not been previously reported, The target genes of these miRNAs included pentatricopeptide repeat-containing protein and diadenosine tetraphosphate hydrolase. An overexpression vector of Arabidopsis miR408 was constructed and transferred to Arabidopsis plant. The roots of plants over expressing miR408 exhibited a slower reorientation upon gravistimulation in comparison with those of wild-type. This result indicate that miR408 could play a role in root gravitropic response.

Controlled ecological life support systems (CELSS) will be an important feature of long-duration space missions of which higher plants are one of the indispensable components. Because of its pivotal role in enabling plants to cope with environmental stress, interplant communication might have important implications for the ecological stability of such CELSS. However, the manifestations of interplant communication in microgravity conditions have yet to be fully elucidated. To address this, a well-established Arabidopsis thaliana co-culture experimental system, in which UV-C-induced airborne interplant communication is evaluated by the alleviation of transcriptional gene silencing (TGS) in bystander plants, was placed in microgravity modeled by a two-dimensional rotating clinostat. Compared with plants under normal gravity, TGS alleviation in bystander plants was inhibited in microgravity. Moreover, TGS alleviation was also prevented when plants of the pgm-1 line, which are impaired in gravity sensing, were used in either the UV-C-irradiated or bystander group. In addition to the specific TGS-loci, interplant communication-shaped genome-wide DNA methylation in bystander plants was altered under microgravity conditions. These results indicate that interplant communications might be modified in microgravity. Time course analysis showed that microgravity interfered with both the production of communicative signals in UV-C-irradiated plants and the induction of epigenetic responses in bystander plants. This was further confirmed by the experimental finding that microgravity also prevented the response of bystander plants to exogenous methyl jasmonate (JA) and methyl salicylate (SA), two well-known airborne signaling molecules, and down-regulated JA and SA biosynthesis in UV-C-irradiated plants.

Controlled ecological life support systems provide food, air, water, and other basic living resources for crew members on long-duration spaceflight missions. Plants are an important basic requirement of these systems and their biological characteristics in space have very high research value. Based on experiments of spaceflight in Shenzhou 8 spacecraft and simulating microgravity effects on three-dimensional (3-D) clinostat, the biological characteristics of tomatos leaf cell sub-microstructure and antioxidant enzyme activities were studied and compared in this work. Results showed that leaf cell sub-microstructure of the tomato samples experiencing spaceflight had more changes than that of the samples processed by simulated microgravity effects, and both peroxidase (POD) and superoxide dismutase (SOD) activities increase obviously in both the environments.

Space particles have an inevitable impact on organisms during space missions; radio-adaptive response (RAR) is a critical radiation effect due to both low-dose background and sudden high-dose radiation exposure during solar storms. Although it is relevant to consider RAR within the context of microgravity, another major space environmental factor, there is no existing evidence as to its effects on RAR. In the present study, we established an experimental method for detecting the effects of gamma-irradiation on the primary root growth of Arabidopsis thaliana, in which RAR of root growth was significantly induced by several dose combinations. Microgravity was simulated using a two-dimensional rotation clinostat. It was shown that RAR of root growth was significantly inhibited under the modeled microgravity condition, and was absent in pgm-1 plants that had impaired gravity sensing in root tips. These results suggest that RAR could be modulated in microgravity. Time course analysis showed that microgravity affected either the development of radio-resistance induced by priming irradiation, or the responses of plants to challenging irradiation. After treatment with the modeled microgravity, attenuation in priming irradiation-induced expressions of DNA repair genes (AtKu70 and AtRAD54), and reduced DNA repair efficiency in response to challenging irradiation were observed. In plant roots, the polar transportation of the phytohormone auxin is regulated by gravity, and treatment with an exogenous auxin (indole-3-acetic acid) prevented the induction of RAR of root growth, suggesting that auxin might play a regulatory role in the interaction between microgravity and RAR of root growth.

Differences of both amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) polymorphisms were compared between the 60-d-old rice (Oryza sativa L. cv. DH7) and F3 rice plants (SP3) derived from seed, which endured a 7-d-space flight in March 2002. Total leaf AFLP DNA bands amplified from 22 primer pairs were 537 in DH7, whereas 562 in SP3. From the total 267 SSR DNA bands generated by 267 primer pairs, 39 were polymorphic with 22 larger (56 %) or 17 smaller (44 %) fragment size bands. The greatest numbers of AFLP DNA bands were amplified by primer E1M1 in DH7 (33) and E3M1 in SP3 (35), whilst the least by E4M3 in DH7 (14) and E5M2 in SP3 (16).

Subcellular changes and enzymes activities were compared between Earth-grown plants from the original tomato seeds, 6-year long-term flown in the Mir Station and 27-day short-term flown in the satellite. In some first generation Mir-flown plants, the lamellae’s structure of some chloroplasts became curved and loose, and some mitochondrial outer membranes were broken. In some satellite-flown plants, the number of mitochondria increased, the lamellae’s structure of some chloroplasts became curved and loose, and some mitochondrial cristae disappeared. The number of starch grains per chloroplast in first generation, second generation, third generation Mir-flown plants and satellite-flown plants increased significantly compared with the ground control, but the difference between the number of starch grains per chloroplast in Mir-flown plants (include three generations) and satellite-flown plants was not significant. The number of chloroplasts per leaf cell in Mir-flown plants and satellite-flown plants increased significantly compared with the ground control. The activities of three defense enzymes supperoxide dismutase (SOD), peroxidase dismutase (POD) and catalase (CAT) in the satellite-flown plants increased significantly as compared with those of the ground controls and the Mir-flown plants, but the content of malondialdehyde (MDA) decreased significantly. Coefficients of variation of the activities of SOD, POD, CAT and the content of MDA in the satellite-flown plants were lest, and those in the Mir-flown plants were maximum. Coefficient of variation of the activities of SOD, POD, and CAT in the first, second and third generation Mir-flown plants were more than that in the ground control. The activities of defense enzyme in some Mir-flown plants increased significantly.

Both space radiation and microgravity have been demonstrated to have inevitable impact on living organisms during space flights and should be considered as important factors for estimating the potential health risk for astronauts. Therefore, the question whether radiation effects could be modulated by microgravity is an important aspect in such risk evaluation. Space particles at low dose and fluence rate, directly affect only a fraction of cells in the whole organism, which implement radiation-induced bystander effects (RIBE) in cellular response to space radiation exposure. The fact that all of the RIBE experiments are carried out in a normal gravity condition bring forward the need for evidence regarding the effect of microgravity on RIBE. In the present study, a two-dimensional rotation clinostat was adopted to demonstrate RIBE in microgravity conditions, in which the RIBE was assayed using an experimental system of root-localized irradiation of Arabidopsis thaliana (A. thaliana) plants. The results showed that the modeled microgravity inhibited significantly the RIBE-mediated up-regulation of expression of the AtRAD54 and AtRAD51 genes, generation of reactive oxygen species (ROS) and transcriptional activation of multicopy P35S:GUS, but made no difference to the induction of homologous recombination by RIBE, showing divergent responses of RIBE to the microgravity conditions. The time course of interaction between the modeled microgravity and RIBE was further investigated, and the results showed that the microgravity mainly modulated the processes of the generation or translocation of the bystander signal(s) in roots.
