Concerns regarding human health and food safety have generated interest in developing simple, accurate, and cost-effective strategies for evaluating organophosphorus pesticide (OP) residues. In this study, nitrogen-doped carbon dots (N-CDs) were synthesized from a common low-cost precursor via a simple pyrolysis process. The fluorescence of the N-CDs can be suppressed by p-nitrophenol, which is the hydrolysis product of the p-nitrophenyl phosphate salt catalyzed by alkaline phosphatase (ALP). A fluorescent turn-on assay for the inhibitory effect of glyphosate was developed with a low detection limit and wide linear range. Moreover, the feasibility of visualizing OPs in vegetables was demonstrated by the fluorescence imaging of glyphosate on cabbage leaves. This research not only offers a facile method for the synthesis of highly fluorescent CDs but also inspires the development of effective multi-mode sensing platforms that include fluorescent testing and imaging for monitoring pesticide residues.

Abstract Cognitive dysfunction caused by diabetes has become a serious global medical issue. Diabetic kidney disease (DKD) exacerbates cognitive dysfunction in patients, although the precise mechanism behind this remains unclear. Here, we conducted an investigation using RNA sequencing data from the Gene Expression Omnibus (GEO) database. We analyzed the differentially expressed genes in DKD and three types of neurons in the temporal cortex (TC) of diabetic patients with cognitive dysfunction. Through our analysis, we identified a total of 133 differentially expressed genes (DEGs) shared between DKD and TC neurons (62 up-regulated and 71 down-regulated). To identify potential common biomarkers, we employed machine learning algorithms (LASSO and SVM-RFE) and Venn diagram analysis. Ultimately, we identified 8 overlapping marker genes (ZNF564, VPS11, YPEL4, VWA5B1, A2ML1, KRT6A, SEC14L1P1, SH3RF1) as potential biomarkers, which exhibited high sensitivity and specificity in ROC curve analysis. Functional analysis using Gene Ontology (GO) revealed that these genes were primarily enriched in autophagy, ubiquitin/ubiquitin-like protein ligase activity, MAP-kinase scaffold activity, and syntaxin binding. Further enrichment analysis using Gene Set Enrichment Analysis (GSEA) and Gene Set Variation Analysis (GSVA) indicates that these biomarkers may play a crucial role in the development of cognitive dysfunction and diabetic nephropathy. Building upon these biomarkers, we developed a diagnostic model with a reliable predictive ability for DKD complicated by cognitive dysfunction. To validate the 8 biomarkers, we conducted RT-PCR analysis in the cortex, hippocampus and kidney of animal models. The results demonstrated the up-regulation of SH3RF1 in the cortex, hippocampus and kidney of mice, which was further confirmed by immunofluorescence and Western blot validation. Notably, SH3RF1 is a scaffold protein involved in cell survival in the JNK signaling pathway. Based on these findings, we support that SH3RF1 may be a common gene expression feature that influences DKD and cognitive dysfunction through the apoptotic pathway.

The requirement for high-speed and high-precision contouring with free-form surfaces in the CNC machining process is increasing. The contour error is an essential criterion for the quality of CNC machining. For the problem of contour error control, the present research is mainly based on position tracking, and the control method directly aiming at contour control has not been well studied. This paper proposed a new type of coordinate transformation-based direct contour control method (CT-DCC) for free-form surface machining trajectories. By real-time monitoring of the contour error and the foot point of the free-form parametric trajectory, the contouring task is decomposed into the contour error control task in the normal direction and the velocity control problem in the feed direction. Using coordinate transformation, the output commands of the two tasks are converted to the axial motion command. CT-DCC completely eliminates position tracking in the traditional control to achieve direct control of the contour error, which provides a new solution for the contour control problem of free-form surface machining. Both the axial velocity control and the axial torque control-based CT-DCC scheme were studied, and the two-axis and three-axis direct contour controllers were tested. The simulation and experiment results show the feasibility of the proposed method.

The running speed of high-speed trains in the tunnel is as high as 350 km, which is very sensitive to the construction disturbance of the new shield tunnel. Therefore, it is of positive significance to study the influence of shield tunneling on existing high-speed railway lines and tunnel structures and control standards. Combined with centrifuge test and three-dimensional numerical simulation, the dynamic response of shield tunnel undercrossing existing high-speed railway tunnel is studied, and the influence of settlement joint and steel pipe pile reinforcement on existing tunnel is analyzed. Studies have shown that the existence of existing tunnels will reduce the surface settlement caused by tunnel excavation, but this shielding effect will be reduced if the influence of construction joints is considered. Therefore, if the construction joint is not considered in the numerical calculation, the ground deformation will be underestimated and the mechanical performance of the existing tunnel structure will be overestimated. In addition, steel pipe piles can effectively control the settlement of existing tunnels.

This paper presents a 14-bit hybrid column-parallel compact analog-to-digital converter (ADC) for the application of digital infrared focal plane arrays (IRFPAs) with compromised power and speed performance. The proposed hybrid ADC works in two phases: in the first phase, a 7-bit successive approximation register (SAR) ADC performs coarse quantization; in the second phase, a 7-bit single-slope (SS) ADC performs fine quantization to complete the residue voltage conversion. In this work, the number of unit capacitors is reduced to 1/128th of that of a conventional 14-bit SAR ADC, which is beneficial for the application of small pixel-pitch IRFPAs. In this work, a tradeoff segmented thermometer-coded digital-to-analog converter (DAC) is adopted in the first 7-bit coarse quantization process: the lower 3-bit is binary coded, and the upper 4-bit is thermometer coded. A thermometer-coded DAC can improve the linearity of ADC. Capacitor array matching can be incredibly relaxed compared with a binary-weight 14-bit SAR ADC, resulting in a noncalibration feature. Moreover, by sharing DAC and comparator analog circuits between the SAR ADC and the SS ADC, the power consumption and layout area are consequently reduced. The proposed hybrid ADC was fabricated using a 180 nm CMOS process. The measurement results show that the proposed ADC has a differential nonlinearity of −0.61/+0.84 LSB and a sampling rate of 120 kS/s. The developed ADC achieves a temporal noise of 1.7 LSBrms at a temperature of 77 K. In addition, the SNDR is 72.9 dB, and the ENOB is 11.82 bit, respectively. Total power consumption is 71 μW from supply voltages of 3.3 V (analog) and 1.8 V (digital).

Exploring an efficient electromagnetic protection strategy for buildings is of great significance to solve the problems caused by increasing electromagnetic pollution, as the rapid progress of technology continues. In this work, FeCo alloy/carbon–carbon nanotube (FeCo/C–CNT) nanocomposites, with significant microwave absorption performance, were successfully synthesized using a simple pyrolysis method involving FeCo–ZIF MOFs precursors and added to epoxy resin to prepare a novel electromagnetic wave absorption (EWA) coating. The minimum reflection loss (RLmin) of the coating applied on the surface of the ceramic tiles was −23.89 dB at 11.37 GHz and the effective absorption bandwidth (EAB) reached 8.85 GHz. Through microscopic characterization and analysis of the electromagnetic parameters of the FeCo/C–CNT nanocomposites, it was found that the EWA coating has an ultrabroad band wave absorption effect, mainly due to the comprehensive advantages of the polarization loss from CNTs, impedance matching, the dual loss synergy effect, and multiple reflection between the FeCo alloys, the carbon layer, and the CNTs. This study has successfully developed high-performance EWA materials and demonstrated the feasibility of an EWA coating applied to building surfaces, contributing to the improvement of electromagnetic protection functions of buildings.

This paper addresses the risk for environmental transmission of pathogenic microorganisms in confined spaces and the serious health hazards for personnel, and research on efficient eradication methods for the pathogenic microorganisms was carried out to provide technical support for ensuring the health of personnel in confined spaces. A series of graphene-MnO2 (G-MnO2) catalytic materials was prepared by hydrothermal and precipitation methods, and processing parameters such as the graphene doping method, the raw material ratio and the plasma action time were optimized. It was shown that G-MnOX-P/HAC prepared by a one-step precipitation method and with a graphene doping ratio of 10% had the best bactericidal effect in a dielectric barrier discharge (DBD) reactor after 4 min of reaction. The eradication rates for Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), coronavirus and Aspergillus niger were all greater than 99.9%. The characterization techniques TEM, SEM, XRD, XPS, BET and FT-IR showed that the G-MnOX-P samples prepared by the one-step precipitation method had larger specific surface areas with more oxygen vacancies and functional groups on the surfaces, which was conducive to decomposition of the ozone generated by the dissociated plasma and formation of reactive oxygen species (ROS) for the microbial eradication process. Finally, by comparing the ozone-decomposition activity with the plasma co-catalytic performance, it was verified that efficient decomposition of the ozone facilitated the eradication of microorganisms. Based on this, an analysis of the mechanism for efficient eradication was carried out.

In order to study the ground disturbance and the influence relationship between the two tunnels during the construction of the new shield tunnel undercrossing the existing high-speed railway tunnel, the centrifuge test was used to simulate the construction of the parallel shield tunnel undercrossing the high-speed railway tunnel, and the variation law of the internal force, segment deformation and surface settlement of the existing high-speed railway tunnel undercrossing the shield was studied. It is found that the adverse effects caused by the later tunnel are less than those caused by the first tunnel excavation. For the existing tunnels without settlement joints, the longitudinal settlement of the inverted arch and the vault is U-shaped and anti-U-shaped respectively. The settlement value of the ground surface and the existing tunnel is increased by more than 100%. When the shield passes through the high-speed railway tunnel, the transverse bending strain is larger than the longitudinal, and special attention should be paid at the corner.

Background Micropercutaneous nephrolithotomy (microperc) is the least invasive among percutaneous nephrolithotripsy (PCNL) procedures. Although microperc has a high stone-free rate and certain advantages over other methods, modifications may be needed to improve the technique. We describe our experience performing microperc using a self-assembled visual needle and ureteral access sheath (UAS). Methods Between June 2016 and April 2019, the data of 30 patients with kidney stones undergoing microperc with our self-assembled 4.8 Fr visual needle combined with a UAS was retrospectively analyzed. Patients were placed in an obilique spine lithotomy position. Results Two cases were excluded: one due to conversion to mini PCNL and the other required flexible ureteroscopy during microperc. The remaining 28 cases included 18 men and 10 women, age 38.4 ± 7.5 years, stone size 1.7 ± 0.4 cm, and stone density on CT 969 ± 233 HU. Operative time was 47 ± 9.9 minutes, visual analogue scale score of tract pain on postoperative day 1 was 2.5 ± 1.0, hemoglobin decrease was 6.4 ± 1.0 g/L, and hospital stay was 3.1 ± 0.8 days. There was 1 case of fever and urinary infection, 2 cases of hematuria, and 1 case of flank pain. All symptoms resolved after conservative or antibiotic treatment. On postoperative day 1, 12 (42.9%) caseswere stone-free. The stone-free rates at postoperative month 1 and 3 were 92.9% (26/28) and 100% (28/28), respectively. Conclusions Our self-assembled visual needle and UAS instrument is effective for microperc. Use of the UAS may improve the operative outcome.

High-strength alloyed steel has been widely used in engineering equipment because of its exceptional strength and toughness, particularly at low temperatures. However, the performance of high-strength alloy steel has not been fully developed, and it is necessary to further optimize the microstructure and mechanical properties. Therefore, the focus of this study is on the phase transition and corresponding mechanical properties of high-strength low-carbon alloyed steels. Three experimental steels were austenitized at 900 °C for 1 h, followed by water quenching, and were then tempered at 570, 600, and 630 °C. They were denoted as QT570, QIT600, and QIT630, respectively. The results show that appropriate intercritical tempering of QIT600 steel significantly increases the proportion of retained austenite and promotes VC precipitation within tempered martensite in comparison to QT570 and QIT630 steels. The enrichment of multiple alloys improved the thermal stability of retained austenite, which was further demonstrated with low-temperature insulation tests. Meanwhile, QIT600 steel with 18 vol.% of retained austenite achieved a superior yield strength of 1025 MPa, an elongation of 21%, and a cryogenic impact energy of 1.25 MJ/m2. The plasticity induced by the transformation of the retained austenite significantly enhanced the strain-hardening rate and postponed necking, thereby increasing elongation. The retained austenite enhanced cryogenic toughness by significantly arresting crack growth and increasing the ability of plastic deformation.

Abstract Backgrounds: Previous studies have demonstrated that excretion of urinary extracellular vesicles (EVs) from different nephron segments differs between kidney stone formers and non-stone formers (NSFs), and could reflect pathogenic mechanisms of urinary stone disease. In this study we quantified selected populations of specific urinary EVs carrying protein markers of immune cells and calcium/phosphorus physiology in calcium oxalate stone formers (CSFs) compared to non-stone formers (NSFs). Methods Biobanked urine samples from CSFs (n = 24) undergoing stone removal surgery and age- and sex- matched NSFs (n = 21) were studied. Urinary EVs carrying proteins related to renal calcium/phosphorus physiology (phosphorus transporters (PiT1 and PiT2), Klotho, and fibroblast growth factor 23 (FGF23); markers associated with EV generation (anoctamin-4 (ANO4) and Huntington interacting protein 1 (HIP1)), and markers shed from activated immune cells were quantified by standardized and published method of digital flow cytometry. ABSTRACT: Wulong geese are renowned for their egg-laying and reproductive abilities. This work investigated the potential of using body size traits in the selection and breeding of high-yielding Wulong geese. A total of forty 479day-old female geese (high-yielding geese, n = 20; low-yielding geese, n = 20) were selected to evaluate the relationship between body weight, body size trait indicators, serum reproductive hormones, and biochemical indicators. The results showed that serum estradiol (E2), glucose (GLU), and triglyceride (TG) concentrations, together with pubic spacing and abdominal circumference were significantly higher in high-yielding geese (P < 0.01), whereas the opposite was true for neck circumference, neck length, and tibial circumference. In addition, the serum testosterone (T) concentration and body weight were higher in high-yielding geese (P < 0.05). Neck circumference and neck length were negatively correlated with E2 and TG (P < 0.01); while pubic spacing and abdominal circumference were positively correlated with E2, GLU, and TG (P < 0.01), the highest correlation coefficient was 0.777 between TG and pubic spacing; T was also strongly associated with neck circumference (P < 0.01). In conclusion, high-yielding Wulong geese can be selected through neck circumference, neck length, pubic spacing, and abdominal circumference.

The deformation mechanism is complex in the hot rolling process of clad plates, and head bending is a common defect. In this paper, an analytical computational mechanical model of a metal plate was established by the classical elastic mechanics method, and the relationship between uneven thickness extension and warpage was obtained. The hot rolling bonding process of dissimilar bimetallic plates of Ti/Steel was investigated. On this basis, the mechanical origins of the plate bending defects and the influence of various factors on the evolution of the plate bending defects of the metal plates were revealed. The results indicated that the rolling forces increased with the increase in reduction ratio and thickness ratio. As the total reduction ratios of the clad plates increased, the reduction ratio of each layer increased. Furthermore, it was found that the thickness reduction ratios of steel were larger than those of Ti at a certain total reduction ratio, which could reach up to 59.6%. When the reduction ratios were 0.4 and 0.45, the bending degree increased with the thickness ratio of the upper and lower plates increasing gradually. The maximum warpage could reach 0.349 m. The clad plate shape was better when the roll speed ratio was 1.02 and the reduction rate was 0.4. The present numerical results provide a valuable insight into the deformation behaviors and mechanisms involved in the hot rolling of clad plates.

The dynamic of droplet spreading on a free-slip surface was studied experimentally and numerically, with particularly interest in the impacts under relatively small droplet inertias (We≤30). Our experimental results and numerical predictions of dimensionless droplet maximum spreading diameter βmax agree well with those of Wildeman et al.’s widely-used model at We>30. The “1/2 rule” (i.e., approximately one half of the initial kinetic energy Ek0 finally transferred into surface energy) was found to break down at small Weber numbers (We≤30) and droplet height is non-negligible when the energy conservation approach is employed to estimate βmax. As We increases, surface energy and kinetic energy alternately dominates the energy budget. When the initial kinetic energy is comparable to the initial surface energy, competition between surface energy and kinetic energy finally results in the non-monotonic energy budget. In this case, gas viscous dissipation contributes the majority of the dissipated energy under relatively large Reynolds numbers. A practical model for estimating βmax under small Weber numbers (We≤30) was proposed by accounting for the influence of impact parameters on the energy budget and the droplet height. Good agreement was found between our model predictions and previous experiments.

With the rapid development of more-electric and all-electric aircraft, the role of power supply systems in aircraft is becoming increasingly prominent. However, due to the complex coupling within the power supply system, a fault in one component often leads to parameter abnormalities in multiple components within the system, which are termed associated faults. Compared with conventional faults, the diagnosis of associated faults is difficult because the fault source is hard to trace and the fault mode is difficult to identify accurately. To this end, this paper proposes a graph-matching approach for the associated fault diagnosis of power supply systems based on a deep residual shrinkage network. The core of the proposed approach involves supplementing the incomplete prior fault knowledge with monitoring data to obtain a complete cluster of associated fault graphs. The association graph model of the power supply system is first constructed based on a topology with characteristic signal propagation and the associated measurements of typical components. Furthermore, fault propagation paths are backtracked based on the Warshall algorithm, and abnormal components are set to update and enhance the association relationship, establishing a complete cluster of typical associated fault mode graphs and realizing the organic combination and structured storage of knowledge and data. Finally, a deep residual shrinkage network is used to diagnose the associated faults via graph matching between the current state graph and the historical graph cluster. The comparative experiments conducted on the simulation model of an aircraft power supply system demonstrate that the proposed method can achieve high-precision associated fault diagnosis, even under circumstances where there are an insufficient number of samples and missing parameters.

Heavy metal contamination in herbal medicines is a global threat to human beings especially at levels above known threshold concentrations. The concentrations of five heavy metals cadmium (Cd), lead (Pb), arsenic (As), mercury (Hg) and copper (Cu) were investigated using Inductively Coupled Plasma Optical Mass Spectrometry (ICP-MS) with 1773 samples around the world. According to Chinese Pharmacopoeia, 30.51% (541) samples were detected with at least one over-limit metal. The over-limit ratio for Pb was 5.75% (102), Cd at 4.96% (88), As at 4.17% (74), Hg at 3.78% (67), and of Cu, 1.75% (31). For exposure assessment, Pb, Cd, As, and Hg have resulted in higher than acceptable risks in 25 kinds of herbs. The maximal Estimated Daily Intake of Pb in seven herbs, of Cd in five, of Hg in four, and As in three exceeded their corresponding Provisional Tolerable Daily Intakes. In total 25 kinds of herbs present an unacceptable risk as assessed with the Hazard Quotient or Hazard Index. Additionally, the carcinogenic risks were all under acceptable limits. Notably, As posed the highest risk in all indicators including Estimated Daily Intake, Hazard Index, and carcinogenic risks. Therefore further study on enrichment effect of different states of As and special attention to monitoring shall be placed on As related contamination.

Abstract Background Cancer surgery can promote tumour metastases and worsen prognosis, however, the effect of perioperative complications on metastatic disease remains unclear. In this study we sought to evaluate the effect of common perioperative complications including perioperative blood loss, hypothermia, and sepsis on tumour metastases in a murine model. Methods Prior to surgery, pulmonary metastases were established by intravenous challenge of CT26LacZ colon cancer cells in BALB/c mice. Surgical stress was generated through partial hepatectomy (PH) or left nephrectomy (LN). Sepsis was induced by puncturing the cecum to express stool into the abdomen. Hemorrhagic shock was induced by removal of 30% of total blood volume (i.e. stage 3 hemorrhage) via the saphenous vein. Hypothermia was induced by removing the heating apparatus during surgery and lowering core body temperatures to 30 °C. Lung tumour burden was quantified 3 days following surgery. Results Surgically stressed mice subjected to stage 3 hemorrhage or hypothermia did not show an additional increase in lung tumour burden. In contrast, surgically stressed mice subjected to intraoperative sepsis demonstrated an additional 2-fold increase in the number of tumour metastases. Furthermore, natural killer (NK) cell function, as assessed by YAC-1 tumour cell lysis, was significantly attenuated in surgically stressed mice subjected to intraoperative sepsis. Both NK cell-mediated cytotoxic function and lung tumour burden were improved with perioperative administration of polyI:C, which is a toll-like receptor (TLR)-3 ligand. Conclusions Perioperative sepsis alone, but not hemorrhage or hypothermia, enhances the prometastatic effect of surgery in murine models of cancer. Understanding the cellular mechanisms underlying perioperative immune suppression will facilitate the development of immunomodulation strategies that can attenuate metastatic disease.

A two-dimensional calibration technique is proposed to correct the spatial nonuniformity in infrared imaging systems adapting to different integration time and time-varying offset with one-time calibration. Differing from traditional calibration-based nonuniformity correction, this method calibrates nonuniformity with two-dimensional information from two integration time besides different irradiance, which conquers drawbacks of traditional calibration-based correction. First, it eliminates the dependence on integration time in calibration process and dramatically suppresses fixed pattern noise by a large attenuation factor. In addition, time-varying offset is real-time canceled by the subtraction of images integrated with normal and short time. Correction error of two-point correction and the proposed method are analyzed in detail. In experiments with cooled infrared camera, the proposed method provides enhanced uniformity even for seven-time variation of integration time using same correction coefficients. Both quantitative and qualitative comparisons to two-point correction demonstrate its superiority. The one-time calibration and shutterless correction scheme avoids interruption of the normal operation for real scene, extending the application range in practical engineering for infrared imaging systems with low complexity of computation and hardware.

During late August and early September 2011, stem rot symptoms were observed on adzuki bean plants (Vigna angularis) growing in fields located in Beijing and Hebei Province, China, respectively. In this study, four isolates were obtained from infected stems of adzuki bean plants. Based on their morphology, and sequence and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analyses of the ribosomal DNA internal transcribed spacers (rDNA-ITS) region, the four isolates were identified as Rhizoctonia solani in anastomosis group (AG) 4 HGI. Pathogenicity tests showed that all isolates were strongly pathogenic to adzuki bean and resulted in serious wilt symptoms which was similar to observations in the fields. Additionally, the isolates infected several other crops and induced related rot on the roots and basal stems. To our knowledge, this is the first report of Rhizoctonia solani AG 4 HGI causing stem rot on adzuki bean.

Introduction: Strangulation of the penis and scrotum by a constricting object has been rarely reported. Aim: To describe a man with penoscrotal strangulation caused by a steel ring and its successful removal. Methods: A 28-year-old man presented to the emergency department with a 7-hour history of a steel ring lodged at the base of his penis and scrotum. Removal was accomplished with the assistance of fire brigade personnel who used their hydraulic cable cutter to shear the ring. During the removal, there were no complications. Results: The hydraulic cable cutter avoided thermal injury and shortened removal time compared with procedures described in the literature. The patient's recovery was uneventful, with erectile function restored after 1 week. Conclusion: Genital incarceration is an urgent clinical situation requiring prompt treatment. However, suitable tools for removing the foreign object are not readily available in emergency and urology departments. Anti-tumor CD8+ T cells are a key determinant for overall survival in patients following surgical resection for solid malignancies. Using a mouse model of cancer vaccination (adenovirus expressing melanoma tumor-associated antigen (TAA)-dopachrome tautomerase (AdDCT) and resection resulting in major surgical stress (abdominal nephrectomy), we demonstrate that surgical stress results in a reduction in the number of CD8+ T cell that produce cytokines (IFNγ, TNFα, Granzyme B) in response to TAA. This effect is secondary to both reduced proliferation and impaired T cell function following antigen binding. In a prophylactic model, surgical stress completely abrogates tumor protection conferred by vaccination in the immediate postoperative period. In a clinically relevant surgical resection model, vaccinated mice undergoing a positive margin resection with surgical stress had decreased survival compared to mice with positive margin resection alone. Preoperative immunotherapy with IFNα significantly extends survival in surgically stressed mice. Importantly, myeloid derived suppressor cell (MDSC) population numbers and functional impairment of TAA-specific CD8+ T cell were altered in surgically stressed mice. Our observations suggest that cancer progression may result from surgery-induced suppression of tumor-specific CD8+ T cells. Preoperative immunotherapies aimed at targeting the prometastatic effects of cancer surgery will reduce recurrence and improve survival in cancer surgery patients.

Resistance to Phytophthora sojae isolate PsMC1 was evaluated in 102 F2∶3 families derived from a cross between the resistant soybean cultivar Wandou 15 and the susceptible cultivar Williams and genotyped using simple sequence repeat (SSR) markers. The segregation ratio of resistant, segregating, and susceptible phenotypes in the population suggested that the resistance in Wandou 15 was dominant and monogenic. Twenty-six polymorphic SSR markers were identified on soybean chromosome 17 (Molecular linkage group D2; MLG D2), which were linked to the resistance gene based on bulked segregation analysis (BSA). Markers Sattwd15-24/25 and Sattwd15-47 flanked the resistance gene at a distance of 0.5 cM and 0.8 cM, respectively. Two cosegregating markers, Sattwd15-28 and Sattwd15-32, were also screened in this region. This is the first Rps resistance gene mapped on chromosome 17, which is designated as Rps10. Eight putative genes were found in the mapped region between markers Sattwd15-24/25 and Sattwd15-47. Among them, two candidate genes encoding serine/threonine (Ser/Thr) protein kinases in Wandou 15 and Williams were identified and sequenced. And the differences in genomic sequence and the putative amino acid sequence, respectively, were identified within each candidate gene between Wandou 15 and Williams. This novel gene Rps10 and the linked markers should be useful in developing soybean cultivars with durable resistance to P. sojae.

Myxoma virus (MYXV) is a well-established oncolytic agent against different types of tumors. MYXV is also known for its immunomodulatory properties in down-regulating major histocompatibility complex (MHC) I surface expression (via the M153R gene product, a viral E3-ubiquitin ligase) and suppressing T cell killing of infected target cells. MHC I down-regulation, however, favors NK cell activation. Brain tumors including gliomas are characterized by high MHC I expression with impaired NK activity. We thus hypothesized that MYXV infection of glioma cells will promote NK cell-mediated recognition and killing of gliomas. We infected human gliomas with MYXV and evaluated their susceptibility to NK cell-mediated cytotoxicity. MYXV enhanced NK cell-mediated killing of glioma cells (U87 cells, MYXV vs. Mock: 51.73% vs. 28.63%, P = .0001, t test; U251 cells, MYXV vs. Mock: 40.4% vs. 20.03%, P .0007, t test). Using MYXV M153R targeted knockout (designated vMyx-M153KO) to infect gliomas, we demonstrate that M153R was responsible for reduced expression of MHC I on gliomas and enhanced NK cell-mediated antiglioma activity (U87 cells, MYXV vs. vMyx-M153KO: 51.73% vs. 25.17%, P = .0002, t test; U251 cells, MYXV vs. vMyx-M153KO: 40.4% vs. 19.27, P = .0013, t test). Consequently, NK cell-mediated lysis of established human glioma tumors in CB-17 SCID mice was accelerated with improved mouse survival (log-rank P = .0072). These results demonstrate the potential for combining MYXV with NK cells to effectively kill malignant gliomas.
