Objetivo: Realizar el diseño y la validación del cuestionario “Nivel de autocuidado en la prevención del pie diabético” en términos de su contenido, constructo, confiabilidad y sensibilidad. Material y método: Estudio psicométrico desarrollado en tres fases: diseño del instrumento, desde las categorías teóricas conceptualizadas de 8 entrevistas a personas con diabetes mellitus; validación aparente y de contenido, contando con la participación de 20 jueces expertos; y validez de constructo, confiabilidad y sensibilidad, contando con 158 mediciones del instrumento. Resultados: El proceso de teorización generó la versión inicial del cuestionario, el cual en la validación facial y de contenido requirió mejorías en redacción y retirar un ítem, esto bajo considerable concordancia entre expertos -Índice de Validez de Contenido de 0.88 y Kappa de Fleiss de 0.7606(p=0.000); se generó así la versión 2 del cuestionario NAPD.V2 con 22 ítems agrupados en 5 dimensiones: automanejo de la diabetes, percepción sobre el pie diabético, impacto emocional de la enfermedad, el cuidado rutinario de los pies y la percepción sobre el soporte social. El análisis factorial exploratorio mostró adecuada validez de constructo, la confiabilidad evidenció buen rendimiento -Alfa de Cronbach de 0,942- y sensibilidad significativa -prueba Wilcoxon p=0.000-. Conclusiones: Tras el diseño y la validación se obtiene el cuestionario NAPD.V2 con adecuadas propiedades psicométricas para ser usado en la población con diabetes mellitus colombiana.

Background Metabolic programs in cancer cells are influenced by genotype and the tissue of origin. We have previously shown that central carbon metabolism is rewired in pancreatic ductal adenocarcinoma (PDA) to support proliferation through a glutamate oxaloacetate transaminase 1 (GOT1)-dependent pathway. Methods We utilized a doxycycline-inducible shRNA-mediated strategy to knockdown GOT1 in PDA and colorectal cancer (CRC) cell lines and tumor models of similar genotype. These cells were analyzed for the ability to form colonies and tumors to test if tissue type impacted GOT1 dependence. Additionally, the ability of GOT1 to impact the response to chemo- and radiotherapy was assessed. Mechanistically, the associated specimens were examined using a combination of steady-state and stable isotope tracing metabolomics strategies and computational modeling. Statistics were calculated using GraphPad Prism 7. One-way ANOVA was performed for experiments comparing multiple groups with one changing variable. Student’s t test (unpaired, two-tailed) was performed when comparing two groups to each other. Metabolomics data comparing three PDA and three CRC cell lines were analyzed by performing Student’s t test (unpaired, two-tailed) between all PDA metabolites and CRC metabolites. Results While PDA exhibits profound growth inhibition upon GOT1 knockdown, we found CRC to be insensitive. In PDA, but not CRC, GOT1 inhibition disrupted glycolysis, nucleotide metabolism, and redox homeostasis. These insights were leveraged in PDA, where we demonstrate that radiotherapy potently enhanced the effect of GOT1 inhibition on tumor growth. Conclusions Taken together, these results illustrate the role of tissue type in dictating metabolic dependencies and provide new insights for targeting metabolism to treat PDA.

Background Diffuse Midline Glioma (DMG) with the H3K27M mutation is a lethal childhood brain cancer, with patients rarely surviving 2 years from diagnosis. Methods We conducted a multi-site Phase 1 trial of the imipridone ONC201 for children with H3K27M-mutant glioma (NCT03416530). Patients enrolled on Arm D of the trial (n = 24) underwent serial lumbar puncture for cell-free tumor DNA (cf-tDNA) analysis and patients on all arms at the University of Michigan underwent serial plasma collection. We performed digital droplet polymerase chain reaction (ddPCR) analysis of cf-tDNA samples and compared variant allele fraction (VAF) to radiographic change (maximal 2D tumor area on MRI). Results Change in H3.3K27M VAF over time (“VAF delta”) correlated with prolonged PFS in both CSF and plasma samples. Nonrecurrent patients that had a decrease in CSF VAF displayed a longer progression free survival (P = .0042). Decrease in plasma VAF displayed a similar trend (P = .085). VAF “spikes” (increase of at least 25%) preceded tumor progression in 8/16 cases (50%) in plasma and 5/11 cases (45.4%) in CSF. In individual cases, early reduction in H3K27M VAF predicted long-term clinical response (>1 year) to ONC201, and did not increase in cases of later-defined pseudo-progression. Conclusion Our work demonstrates the feasibility and potential utility of serial cf-tDNA in both plasma and CSF of DMG patients to supplement radiographic monitoring. Patterns of change in H3K27M VAF over time demonstrate clinical utility in terms of predicting progression and sustained response and possible differentiation of pseudo-progression and pseudo-response.

Cancer metabolism is rewired to support cell survival in response to intrinsic and environmental stressors. Identification of strategies to target these adaptions is an area of active research. We previously described a cytosolic aspartate aminotransaminase (GOT1)-driven pathway in pancreatic cancer used to maintain redox balance. Here, we sought to identify metabolic dependencies following GOT1 inhibition to exploit this feature of pancreatic cancer and to provide additional insight into regulation of redox metabolism. Using pharmacological methods, we identify cysteine, glutathione, and lipid antioxidant function as metabolic vulnerabilities following GOT1 withdrawal. We demonstrate that targeting any of these pathways triggers ferroptosis, an oxidative, iron-dependent form of cell death, in GOT1 knockdown cells. Mechanistically, we reveal that GOT1 inhibition represses mitochondrial metabolism and promotes a catabolic state. Consequently, we find that this enhances labile iron availability through autophagy, which potentiates the activity of ferroptotic stimuli. Overall, our study identifies a biochemical connection between GOT1, iron regulation, and ferroptosis., The aspartate aminotransaminase GOT1 is important for maintaining redox balance. Here, the authors show that inhibition of GOT1 in pancreatic cancer cells leads to cell death via ferroptosis.

Background: Physical activity may improve quality of life in patients with chronic kidney disease. Aim: To assess the relationship between physical activity and quality of life in patients with Chronic Kidney Disease. Material and Methods: The Kidney Disease Quality of Life-36 (KDQOL-36) and the International Physical Activity questionnaire were answered by 130 patients with chronic kidney disease (74 women, 80 receiving renal substitution therapy). Sociodemographic variables were recorded. Results: Patients on renal substitution therapy with a time lapse since diagnosis of 0 to 6 months had higher levels of physical activity than those with longer time lapses (51.4 ± 12.5 and 34.6 ± 8.1 minutes respectively). Disease burden scores were lower among patients with renal substitution therapy. There was a direct correlation between levels of vigorous and moderate physical activity and the physical functioning dimension in the quality of life questionnaire for patients with more than 19 months of disease. The dimension general physical health was significantly associated with physical activity in women and patients with 7 to 18 months of diagnosis. The dimension disease burden was associated with physical activity in women, patients not receiving substitution therapy and those with 7 to 18 months of diagnosis. Conclusions: Moderate and vigorous physical activity is directly related to the dimensions physical functioning, the general perception of physical health and inversely related with the dimension burden of disease.

El objetivo del estudio fue identificar los aspectos asociados a la permanencia estudiantil como resultado de la conducta resiliente en un grupo de estudiantes de educación superior. Se espera ampliar el conocimiento en torno a la permanencia estudiantil como resultado de la evolución de los estudios sobre deserción. Participaron 20 estudiantes de los niveles tecnológico, profesional y maestría, quienes fueron identificados como resilientes. Se hizo uso del método de la Teoría Anclada o Grounded Theory y, a partir de la información recogida por medio de grupos focales, se identificaron dos súper familias o categorías, denominadas Impulsores y Frenos de la permanencia; estas agrupan en sí mismas los aspectos que potencializan o desestimulan la permanencia de los estudiantes desde la perspectiva de la resiliencia. Simultáneamente, estas categorías se clasificaron de acuerdo a cuatro fuentes identificadas: Institución Educativa (I), Persona (P), Empresa (E) y Familia (F). Finalmente, se identificaron 21 aspectos que, en forma coherente con el modelo interaccionista, se asocian a la resilencia educativa y promueven la permanencia estudiantil en Educación Superior.

The tumor microenvironment (TME) in pancreatic ductal adenocarcinoma (PDA) restricts vascularization and, consequently, access to blood-derived nutrients and oxygen, which impacts tumor growth. Intracellular redox imbalance is another restraint on cellular proliferation, yet it is unknown if the TME contributes to the maintenance of redox homeostasis in PDA cells. Here, we demonstrate that the loss of mitochondrial glutamate-oxaloacetate transaminase 2 (GOT2), a component in the malate-aspartate shuttle, disturbs redox homeostasis and halts proliferation of PDA cells in vitro. In contrast, GOT2 inhibition has no effect on in vivo tumor growth or tumorigenesis in an autochthonous model. We propose that this discrepancy is explained by heterocellular pyruvate exchange from the TME, including from cancer associated fibroblasts. More broadly, pyruvate similarly confers resistance to inhibitors of mitochondrial respiration. Genetic or pharmacologic inhibition of pyruvate uptake or metabolism abrogated pyruvate-mediated alleviation of reductive stress from NADH buildup. In sum, this work describes a potential resistance mechanism mediated by metabolic crosstalk within the pancreatic TME. These findings have important implications for metabolic treatment strategies since several mitochondrial inhibitors are currently in clinical trials for PDA and other cancers.

SummaryPancreatic ductal adenocarcinoma (PDA) is one of the deadliest solid malignancies, with a 5-year survival rate at ten percent. PDA have unique metabolic adaptations in response to cell-intrinsic and environmental stressors, and identifying new strategies to target these adaptions is an area of active research. We previously described a dependency on a cytosolic aspartate aminotransaminase (GOT1)-dependent pathway for NADPH generation. Here, we sought to identify metabolic dependencies induced by GOT1 inhibition that could be exploited to selectively kill PDA. Using pharmacological methods, we identified cysteine, glutathione, and lipid antioxidant function as metabolic vulnerabilities following GOT1 withdrawal. Targeting any of these pathways was synthetic lethal in GOT1 knockdown cells and triggered ferroptosis, an oxidative, non-apoptotic, iron-dependent form of cell death. Mechanistically, GOT1 inhibition promoted the activation of autophagy in response to metabolic stress. This enhanced the availability of labile iron through ferritinophagy, the autolysosome-mediated degradation of ferritin. Desde una perspectiva crítico-social, este artículo da cuenta de un proyecto investigativo en curso que tiene como objetivo, fortalecer la comprensión lectora a través de la pregunta, partiendo de las vivencias, la realidad, la experiencia personal, social y familiar de 26 estudiantes: 16 niños y 10 niñas, de la Institución Educativa “Gimnasio Gran Colombiano”, de la ciudad de Tunja, del grado segundo de básica primaria, así como de las dos docentes investigadoras. Desde la Investigación acción educativa, se busca generar alternativas de trabajo y cambio en las prácticas pedagógicas dentro del aula, para mejorar la comprensión lectora a partir de la intervención con la pregunta. Se asume como referentes de este trabajo investigativo a Solé (2011), Larrosa (2010), Lipman (1998), Splitter y Sharp (1995) y Freire (1989). Como avances del proceso investigativo se registran aportes en cuanto a la revisión de antecedentes, así mismo, se presentan avances en torno a cuatro categorías de análisis emergentes: la comprensión lectora, la pregunta, el diálogo y la experiencia.

Diffuse midline glioma (DMG) with the H3K27M mutation is a lethal childhood brain cancer, with patients rarely surviving 2 years from diagnosis. We conducted a multi-site Phase 1 trial of the imipridone ONC201 for children with H3K27M-mutant glioma ({"type":"clinical-trial","attrs":{"text":"NCT03416530","term_id":"NCT03416530"}}NCT03416530). Patients enrolled on Arm D of the trial (n=24) underwent serial lumbar puncture (baseline, 2, 6 months) for cell-free tumor DNA (cf-tDNA) analysis at time of MRI. Additionally, patients on all arms of the trial at the University of Michigan underwent serial plasma collection. CSF collection was feasible in this cohort, with no procedural complications. We collected 96 plasma samples and 53 CSF samples from 29 patients, including those with H3F3A (H3.3) (n=13), HIST13HB (H3.1) (n= 4), and unknown H3 status/not biopsied (n=12) [range of 0–8 CSF samples and 0–10 plasma samples]. We performed digital droplet polymerase chain reaction (ddPCR) analysis and/or amplicon-based electronic sequencing (Oxford Nanopore) of cf-tDNA samples and compared variant allele fraction (VAF) to radiographic change (maximal 2D tumor area on MRI). Preliminary analysis of samples demonstrates a correlation between changes in tumor size and H3K27M cf-tDNA VAF, when removing samples with concurrent bevacizumab. In multiple cases, early reduction in CSF cf-tDNA predicts long-term clinical response (>1 year) to ONC201, and does not increase in cases of later-defined pseudo-progression (radiation necrosis). For example, a now 9-year old patient with thalamic H3K27M-mutant DMG underwent treatment with ONC201 after initial radiation and developed increase in tumor size at 4 months post-radiation (124% baseline) of unclear etiology at the time. The aim of this study was to establish the relationship between personal values and organizational commitment of a group of officials from an institution of higher education in Bogotá. This research is part of the four-factor model which is proposed by Schwartz (2006) and the three-components model of organizational commitment of Meyer et al (1993). This research was conducted by a correlational study with a sample of 128 officials from executive, professional and assistant levels. In order to collect the data the questionnaires used were Portrait of Schwartz and the Questionnaire of organizational commitment of Meyer et al (1993). Both tools were adapted culturally to ensure linguistic, semantic and cultural equivalence and a pilot study was made with a group of 101 people. The correlation coefficients did not show an association between the variables proposed. The structural equations model showed a very low percentage of variance of the data. An alternative model was found to best explain the relationship of the constructs; in this alternative model three variables emerged, which were analyzed based on different groups of values within two factors: individualism and collectivism. It was concluded that while some authors have identified the relationship between the proposed constructs, this associations complex, because it involves many more variables, and could be more appropriate to analyze the consistency of the values of the subjects and the declared values of the company.

For 30 years, Hermosillo has been wondering whether it has a future after Ford. Until the early 1980s, the city relied mainly on agricultural activity. When the multinational motor company arrived in this northwestern Mexican city in 1986, it changed the history of a region that up until that point had relied heavily on agriculture. The assembly plant was established and many auto parts suppliers sprang up, triggering industrialization and increasing the complexity of its economy, its productivity, and wages. Intensive manufacturing development turned Hermosillo into the fifth richest metropolitan area in Mexico in 1998. Broadly speaking, this growth trajectory was maintained. In 2015, Hermosillo was in the top 5% of wealthiest municipalities, with poverty levels and informal employment rates significantly lower than in the rest of the country. But the economy of this city in Sonora State has clearly lost its dynamism over the past few years. Even in Mexico’s low-growth context during the period 2005-2015, growth in per capita gross domestic product in Hermosillo (1.3%) fell below the federal average (1.4%). Hermosillo’s relative performance during this decade was not uniform. Between 2005 and 2010, it grew at a rate of 1.3%, placing it in the 66th percentile (among the top 34% for growth rate) of all municipalities in Mexico. In the second half of the decade, Hermosillo barely reached 1.2% growth, falling to the 47th percentile (53% of Mexican municipalities grew more). The situation worsened in the years 2013-2015, when output per worker fell by 7.2%. What happened in Hermosillo? Can the current economic structure sustain the municipality’s high wages and guarantee future growth? What policy interventions are needed? Seeking answers to these questions, the Growth Lab at Harvard's Center for International Development joined forces with the Inter-American Development Bank or IDB, in particular with its Emerging and Sustainable Cities Program (ESC). This program is a technical assistance initiative that provides support to regional governments to develop and execute urban sustainability projects. ESC’s goal is to contribute to priority urban interventions to provide the sustainable, harmonious growth of cities. This part of its vision is aligned with the idea of promoting inclusive growth and prosperity that guides the Growth Lab's research.

En respuesta a la necesidad de generar alternativas que permitan asegurar la calidad y diferenciación del café de Nariño, se realizó esta investigación con el objetivo de evaluar la calidad física y sensorial del café variedad Colombia, fruto rojo, sembrado a plena exposición solar por caficultores del programa Nespresso AAA 2015, en diferentes altitudes y variaciones en el beneficio, en el Municipio de la Unión, Nariño. En cada finca se cosecharon diez kilos de café maduro con color rojo intenso, se procesaron con beneficio húmedo en la misma finca, se secaron con diferentes tiempos de exposición solar y se llevaron al laboratorio de Almacafé para evaluar sus características físicas y organolépticas como: aroma, fragancia, acidez, cuerpo, sabor, sabor residual, balance, taza limpia, dulzor y puntaje del catador. Igualmente se analizaron las relaciones de los factores climáticos y agronómicos de cada finca con la calidad de taza. Toda la información se analizó mediante análisis multivariado de componentes principales (ACP) y análisis de correspondencias múltiples (ACM). Se concluyó que las condiciones de altitud, no están asociadas particularmente con los parámetros de calidad de la bebida del café; menores contenidos de fósforo en el suelo, menor tiempo de fermentación (16 - 17h) y secado (20 - 25h de sol), se asociaron a mayores puntajes de calidad. La compleja interacción de todas las variables asociadas a la producción del café, es la que define al final, la calidad de taza.

PurposeThe purpose of this paper is to analyze gender disparities in labor earnings for a comprehensive set of 64 countries.Design/methodology/approachUsing the methodological approach proposed by Ñopo, socio‐demographic characteristics are used to match males and females such that gender earnings disparities are computed only among individuals with the same observable characteristics.FindingsDisparities are partially attributed to gender differences in observable socio‐demographic and job characteristics. After matching males and females with the same characteristics, the authors found that the earnings gap falls within a range between 8 per cent and 48 per cent of average females’ earnings, being more pronounced in South Asia and Sub‐Saharan Africa. The unexplained earnings gaps are more pronounced among part‐time workers and those with low education levels.Originality/valueThis paper presents a comprehensive view of gender earnings gaps in the world, simultaneously exploring many of the issues highlighted in the related literature. It adds value by exploring gender gaps in a comparative perspective, applying the same methods for several different countries.

Este documento evalúa el comportamiento y la magnitud de los diferenciales salariales en Colombia entre trabajadores rurales y urbanos, y jóvenes (18-24 años) y mayores (25-65 años), durante 2002-2009. La aplicación de pruebas de raíz unitaria y la metodología matching de descomposición de brechas salariales concluyen que si bien existen diferencias en los niveles de salario de los grupos de interés, no hay divergencia cuando se comparan individuos con iguales características. Los diferenciales salariales encontrados son de -50 por ciento para trabajadores rurales y -40 por ciento para jóvenes, de los cuales alrededor de 14 y 19 puntos porcentuales, respectivamente, permanecen no explicados después de introducir controles demográficos y laborales.

En este documento se presenta una evaluacion de la Oficina de Evaluacion y Supervision (OVE) del Programa de Pais del Banco Interamericano de Desarrollo (BID o el Banco) con Colombia durante el periodo 2011-2014. De acuerdo con el Protocolo para las Evaluaciones de los Programas de Pais (documento RE-348-3) del Banco, la principal funcion de estas evaluaciones es "proporcionar informacion sobre el desempeno del Banco a nivel de pais que sea creible y util y que permita incorporar lecciones y recomendaciones a fin de aumentar la efectividad en el desarrollo de la estrategia y el programa generales de la asistencia del Banco a un pais". Al igual que en otras evaluaciones de programas de pais, se examina el apoyo del Banco al pais con dos finalidades: fortalecer la rendicion de cuentas y facilitar el aprendizaje.

La Economía Solidaria se ha convertido en la actualidad en una de las puntas de lanza de aquellos que acuerdan que el sistema de producción capitalista dominante se encuentra en una crisis profunda que amenaza la estabilidad y continuidad de la civilización actual en todos sus ámbitos, económico, social, político, ambiental y cultural. Esta propuesta es una más de las que surgen o se plantean como alternativa al capitalismo de mercado en la búsqueda de otro tipo de sociedad, fundamentada en principios diferentes a la acumulación de capital y maximización de ganancias. En el caso de la Economía Solidaria los principios que podrían ser los fundamentales para su análisis y comprensión radican en la solidaridad, ciudadanía y comunidad puesto que desde tiempos inmemorables han estado asociados a la búsqueda y reivindicación de los derechos de los excluidos en la sociedad. No obstante, existen posiciones a favor y en contra. Los que están a favor a su vez se dividen en aquellos que miran a la Economía Solidaria como una forma de benevolencia y caridad hacia los más desposeídos, y aquellos que la conciben como un reemplazo al capitalismo o al menos como una economía para atender a los sectores que no son rentables para el mercado, ni para el Estado debido a la escasez de recursos públicos. Mientras que para los que están en contra no se trataría más que de una ambigüedad relevante de una concepción ideológica equivocada que intenta fusionar conceptos que por principio son irreconciliables. La presente investigación pretende develar la esencia de este planteamiento a través del análisis de sus bases conceptuales y de esta manera poder realizar un entendimiento real de la viabilidad de esta propuesta, y las diversas interpretaciones para su aplicación, concretamente en el caso del Ecuador. Resumen: En este artículo se busca profundizar el debate sobre el tema de la Economía Social y Solidaria y su particular adaptación a la realidad ecuatoriana encarnada en la Ley de Economía Popular y Solidaria y su Reglamento que surge como producto de la Constitución de 2008, la cual declara al sistema económico del Ecuador como social y solidario. Se realiza un entendimiento de sus bases conceptuales: economía, solidaridad, comunidad; y se utiliza el enfoque de la teoría crítica como método de análisis para precisar los equívocos así como la viabilidad o no de esta propuesta. Finalmente, se exponen las conclusiones que resaltan tanto las dificultades de su aplicación, sus potencialidades, así como nuevas interrogantes susceptibles de investigaciones posteriores. Abstract: This article seeks to deepen the debate on the issue of Social and Solidarity Economy and its particular adaptation to Ecuadorian reality embodied in the Law of Social and Solidarity Economy and theirs Regulations that arises as a result of the 2008 Constitution, which document state Ecuador’s economic system social and solidarity. It takes an understanding of the conceptual basis: the economy, solidarity, community; and used the approach of critical theory as a method of analysis to clarify the misunderstandings such as the feasibility or otherwise of this proposal. Finally, are exposed the conclusions highlight both the difficulties of their application, their potentialities, as well as new questions susceptible of further research.

In the present redetermination of the complex cis-tetracarbonylbis(tricyclohexylphosphine)molybdenum(0), (I), [Mo(C(18)H(33)P)(2)(CO)(4)] or cis-eta(1)-[P(C(6)H(11))(3)](2)Mo(CO)(4), the Mo atom has a distorted octahedral geometry with a large P-Mo-P angle of 104.8 (1) degrees. A strong trans influence on the carbonyls in (I) is seen in a shortening of the Mo-C and a lengthening of the C-O distances opposite the phosphines compared with those that are cis. This influence is greatly diminished in the complex pentacarbonyl(tricyclohexylphosphine)molybdenum(0), (II), [Mo(C(18)H(33)P)(CO)(5)] or eta(1)-[P(C(6)H(11))(3)]Mo(CO)(5), the core of which has a slightly distorted C(4v) geometry.
