1. Computational investigations of the electronic structure of molecular actinide compounds

Abstract

In this PhD thesis the electronic structure of a range of actinide compounds has been investigated using density functional theory. The reason for using DFT instead of other methods is mainly due to the size of the compounds which makes multireference calculations prohibitively expensive, but also to make comparisons with previously calculated DFT results. The first chapter presents the basic concepts of electronic structure theory and the chemical properties of the actinides and lanthanides. The theoretical foundation of DFT and the consequences of relativity are also introduced. In the second chapter the bonding in mixed MUCl6, MUCl8 2-, NpReCl8 2- and PuOsC NpReCl8 2- (M = Mo, W) systems is investigated and compared with previous work on the M2Cl6, M2C NpReCl8 2- U2Cl6 and U2C NpReCl8 2- systems. The study shows that the total bonding energy in the mixed compounds is the average of the two “pure” compounds. The third chapter deals with systems of plenary or lacunary Keggin phosphomolybdate coordination to actinide (Th), lanthanide (Ce, La, Lu) and transition metal (Hf, Zr) cations: [PMo12O40]3-, [PMo11O39]2 14-, [PMo12O40]2 6- and [PMo11O39][PMo12O40]10-. These large, highly anionic systems proved to be very challenging computationally. The main result of the study confirms that the bonding is ionic and that there are few differences in the behaviour of the transition metals. In the fourth chapter the electronic spectrum of NpO2 2+, NpO2Cl4 2- and NpO2(OH)4 2- is calculated using time dependent DFT. TDDFT has proved adequate for the uranium analogues of these systems and this extends previous work on f0 systems to f1 systems. The results show that TDDFT is in poor agreement with both experimental results and multireference calculations for these compounds. In chapter five, group 15 and 16 uranyl analogues have been investigated. For the UE2 (E = O, S, Se, Te) analogues the geometry bends for all chalcogens heavier than O. The UE2 2+ analogues remain linear all the way down group 16. In U(NCH3)2 2+ the formation of a {pi} “back bone” along the axis of the molecule was noted. The {sigma}-bonding valence MOs stabilize while the {pi} MOs are destabilized down group 15 and 16. Chapter six is a summary of the results in this thesis and an outlook on potential future work.

Abstract

The reproducibility of findings is a compelling methodological problem that the neuroimaging community is facing these days. The lack of standardized pipelines for image processing, quantification and statistics plays a major role in the variability and interpretation of results, even when the same data are analysed. This problem is well-known in MRI studies, where the indisputable value of the method has been complicated by a number of studies that produce discrepant results. However, any research domain with complex data and flexible analytical procedures can experience a similar lack of reproducibility. In this paper we investigate this issue for brain PET imaging. During the 2018 NeuroReceptor Mapping conference, the brain PET community was challenged with a computational contest involving a simulated neurotransmitter release experiment. Fourteen international teams analysed the same imaging dataset, for which the ground-truth was known. Despite a plurality of methods, the solutions were consistent across participants, although not identical. These results should create awareness that the increased sharing of PET data alone will only be one component of enhancing confidence in neuroimaging results and that it will be important to complement this with full details of the analysis pipelines and procedures that have been used to quantify data.

Abstract

In longitudinal positron emission tomography (PET), the presence of volumetric changes over time can lead to an overestimation or underestimation of the true changes in the quantified PET signal due to the partial volume effect (PVE) introduced by the limited spatial resolution of existing PET cameras and reconstruction algorithms. Here, a 3D-printed anthropomorphic brain phantom with attachable striata in three sizes was designed to enable controlled volumetric changes. Using a method to eliminate the non-radioactive plastic wall, and manipulating BP levels by adding different number of events from list-mode acquisitions, we investigated the artificial volume dependence of BP due to PVE, and potential bias arising from varying BP. Comparing multiple reconstruction algorithms we found that a high-resolution ordered-subsets maximization algorithm with spatially variant point-spread function resolution modeling provided the most accurate data. For striatum, the BP changed by 0.08% for every 1% volume change, but for smaller volumes such as the posterior caudate the artificial change in BP was as high as 0.7% per 1% volume change. A simple gross correction for striatal volume is unsatisfactory, as the amplitude of the PVE on the BP differs depending on where in the striatum the change occurred. Therefore, to correctly interpret age-related longitudinal changes in the BP, we must account for volumetric changes also within a structure, rather than across the whole volume. The present 3D-printing technology, combined with the wall removal method, can be implemented to gain knowledge about the predictable bias introduced by the PVE differences in uptake regions of varying shape.

Abstract

Background: Oxygen therapy has been used routinely in normoxemic patients with suspected acute myocardial infarction (AMI) despite limited evidence supporting a beneficial effect. AMI is associated with a systemic inflammation. Here, we hypothesized that the inflammatory response to AMI is potentiated by oxygen therapy.Methods: The DETermination of the role of Oxygen in suspected Acute Myocardial Infarction (DETO2X-AMI) multicentre trial randomized patients with suspected AMI to receive oxygen at 6 L min-1 for 6-12 h or ambient air. For this prespecified subgroup analysis, we recruited patients with confirmed AMI from two sites for evaluation of inflammatory biomarkers at randomization and 5-7 h later. Ninety-two inflammatory biomarkers were analysed using proximity extension assay technology, to evaluate the effect of oxygen on the systemic inflammatory response to AMI.Results: Plasma from 144 AMI patients was analysed whereof 76 (53%) were randomized to oxygen and 68 (47%) to air. Eight biomarkers showed a significant increase, whereas 13 were decreased 5-7 h after randomization. The inflammatory response did not differ between the two treatment groups neither did plasma troponin T levels. After adjustment for increase in troponin T over time, age and sex, the release of inflammation-related biomarkers was still similar in the groups.Conclusions: In a randomized controlled setting of normoxemic patients with AMI, the use of supplemental oxygen did not have any significant impact on the early release of systemic inflammatory markers. Abstract

BACKGROUND: Current fibrinolytic therapies fail to achieve optimum reperfusion in many patients. Low-molecular-weight heparins and platelet glycoprotein IIb/IIIa inhibitors have shown the potential to improve pharmacological reperfusion therapy. We did a randomised, open-label trial to compare the efficacy and safety of tenecteplase plus enoxaparin or abciximab, with that of tenecteplase plus weight-adjusted unfractionated heparin in patients with acute myocardial infarction. METHODS: 6095 patients with acute myocardial infarction of less than 6 h were randomly assigned one of three regimens: full-dose tenecteplase and enoxaparin for a maximum of 7 days (enoxaparin group; n=2040), half-dose tenecteplase with weight-adjusted low-dose unfractionated heparin and a 12-h infusion of abciximab (abciximab group; n=2017), or full-dose tenecteplase with weight-adjusted unfractionated heparin for 48 h (unfractionated heparin group; n=2038). The primary endpoints were the composites of 30-day mortality, in-hospital reinfarction, or in-hospital refractory ischaemia (efficacy endpoint), and the above endpoint plus in-hospital intracranial haemorrhage or in-hospital major bleeding complications (efficacy plus safety endpoint). Analysis was by intention to treat. FINDINGS: There were significantly fewer efficacy endpoints in the enoxaparin and abciximab groups than in the unfractionated heparin group: 233/2037 (11.4%) versus 315/2038 (15.4%; relative risk 0.74 [95% CI 0.63-0.87], p=0.0002) for enoxaparin, and 223/2017 (11.1%) versus 315/2038 (15.4%; 0.72 [0.61-0.84], p

Abstract

Despite political efforts, diffuse pollution from agriculture continues to be the single largest source of nitrogen (N) emissions into the aquatic environment in many countries and regions. This fact, and the recent enactment of a new Swedish environmental code, led to the design of a study targeted at the evaluation of new N pollution abatement strategies. An actor game was chosen as the key component of the study, with a focus on four major goals: to test the implementability of legally binding environmental quality standards for nitrate concentration in groundwater and N transport to the sea, to find sets of agriculturally feasible and cost-effective measures to decrease N loads, to investigate the possibilities for collective action through negotiated and institutionalized actor cooperation, and to investigate the role of mathematical modeling in environmental N management. Characteristics from the agriculturally dominated catchment of Genevadsan (224 km(2)) on the southwest coast of Sweden served as the playing field for the actor game. The most noteworthy result from the study was that it appeared to be possible to meet ambitious environmental N standards with less economically drastic measures than anticipated by most of the participants. The actor game was shown to be a good method for learning about the new Swedish environmental code and its application and for gaining deeper insight into the issues of N management. Abstract

In this paper, we present a 3D geometric flow designed to segment the main core of fiber tracts in diffusion tensor magnetic resonance images. The fundamental assumption of our fiber segmentation technique is that adjacent voxels in a tract have similar properties of diffusion. The fiber segmentation is carried out with a front propagation algorithm constructed to fill the whole fiber tract. The front is a 3D surface that evolves with a propagation speed proportional to a measure indicating the similarity of diffusion between the tensors lying on the surface and their neighbors in the direction of propagation. We use a level set implementation to assure a stable and accurate evolution of the surface and to handle changes of topology of the surface during the evolution process. The fiber tract segmentation method does not need a regularized tensor field since the surface is automatically smoothed as it propagates. The smoothing is done by an intrinsic surface force, based on the minimal principal curvature. This segmentation can be used for obtaining quantitative measures of the diffusion in the fiber tracts and it can also be used for white matter registration and for surgical planning.
