Abstract Novel haptics technologies are urgently needed to bridge the gap between entirely physical world and fully digital environment to render a more realistic and immersive human–computer interaction. Current virtual reality (VR) haptic gloves either deliver limited haptic feedback or are bulky and heavy. The authors develop a haptic glove or HaptGlove, an untethered and lightweight pneumatic glove, that allows users to “physically” interact in a VR environment and enables both kinesthetic and cutaneous sensations naturally and realistically. Integrated with five pairs of haptic feedback modules and fiber sensors, HaptGlove provides variable stiffness force feedback and fingertip force and vibration feedback, allowing users to touch, press, grasp, squeeze, and pull various virtual objects and feel the dynamic haptic changes. Significant improvements in VR realism and immersion are observed in a user study with participants achieving 78.9% accuracy in sorting six virtual balls of different stiffnesses. Importantly, HaptGlove facilitates VR training, education, entertainment, and socialization in a reality–virtuality continuum.

As the impact of global warming on climate change becomes noticeable, the importance of energy efficiency for reducing greenhouse gas emissions grows immense. To this end, a platform, solution, and mobile apps are developed as part of the European Union’s Horizon 2020 research and innovation program to support energy optimization in residences. However, to ensure long-term energy optimization, it is crucial to keep users engaged with the apps. Since augmented reality (AR) and a virtual animal companion positively influenced user engagement, we designed an AR companion that represented the user’s residence states; thereby making the user aware of indoor information. We conducted user evaluations to determine the effect of the AR companion on user engagement and perceived usability in the context of energy management. We identified that the user interface (UI) with AR (ARUI) barely affected user engagement and perceived usability compared to the traditional UI without AR (TUI); however, we found that the ARUI positively affected one of the user engagement aspects. Our results show AR companion integration’s potential benefits and effects on energy management mobile apps. Furthermore, our findings provide insights into UI design elements for developers considering multiple interaction modalities with AR.

In this work, a lightweight compliant glove that detects scratching using data from microtubular stretchable sensors on each finger and an inertial measurement unit (IMU) on the palm through a machine learning model is presented: the SensorIsed Glove for Monitoring Atopic Dermatitis (SIGMA). SIGMA provides the user and clinicians with a quantifiable way of assaying scratch as a proxy to itch. With the quantitative information detailing scratching frequency and duration, the clinicians would be able to better classify the severity of itch and scratching caused by atopic dermatitis (AD) more objectively to optimise treatment for the patients, as opposed to the current subjective methods of assessments that are currently in use in hospitals and research settings. The validation data demonstrated an accuracy of 83% of the scratch prediction algorithm, while a separate 30 min validation trial had an accuracy of 99% in a controlled environment. In a pilot study with children (n = 6), SIGMA accurately detected 94.4% of scratching when the glove was donned. We believe that this simple device will empower dermatologists to more effectively measure and quantify itching and scratching in AD, and guide personalised treatment decisions.

BackgroundThe older population needs solutions for independent living and reducing the burden on caregivers while maintaining the quality and dignity of life. ObjectiveThe aim of this study was to design, develop, and evaluate an older adult health care app that supports trained caregivers (ie, formal caregivers) and relatives (ie, informal caregivers). We aimed to identify the factors that affect user acceptance of interfaces depending on the user’s role. MethodsWe designed and developed an app with 3 user interfaces that enable remote sensing of an older adult’s daily activities and behaviors. We conducted user evaluations (N=25) with older adults and their formal and informal caregivers to obtain an overall impression of the health care monitoring app in terms of user experience and usability. In our design study, the participants had firsthand experience with our app, followed by a questionnaire and individual interview to express their opinions on the app. Through the interview, we also identified their views on each user interface and interaction modality to identify the relationship between the user’s role and their acceptance of a particular interface. The questionnaire answers were statistically analyzed, and we coded the interview answers based on keywords related to a participant’s experience, for example, ease of use and usefulness. ResultsWe obtained overall positive results in the user evaluation of our app regarding key aspects such as efficiency, perspicuity, dependability, stimulation, and novelty, with an average between 1.74 (SD 1.02) and 2.18 (SD 0.93) on a scale of −3.0 to 3.0. The overall impression of our app was favorable, and we identified that “simple” and “intuitive” were the main factors affecting older adults’ and caregivers’ preference for the user interface and interaction modality. We also identified a positive user acceptance of the use of augmented reality by 91% (10/11) of the older adults to share information with their formal and informal caregivers. ConclusionsTo address the need for a study to evaluate the user experience and user acceptance by older adults as well as both formal and informal caregivers regarding the user interfaces with multimodal interaction in the context of health monitoring, we designed, developed, and conducted user evaluations with the target user groups. Our results through this design study show important implications for designing future health monitoring apps with multiple interaction modalities and intuitive user interfaces in the older adult health care domain.

Summary: The papain-like protease (PLpro) of coronaviruses is an attractive antiviral target to inhibit both viral replication and interference of the host immune response. We have identified and characterized three novel classes of small molecules, thiophene, cyanofuran, and triazoloquinazoline, as PLpro inhibitors. Thiophene inhibited the PLpro of two major coronaviruses, Middle East respiratory syndrome coronavirus (MERS-CoV) and severe acute respiratory syndrome coronavirus (SARS-CoV) including SARS-CoV-2, while cyanofuran and triazoloquinazoline more selectively inhibited MERS-CoV PLpro. Unlike GRL0617, a known PLpro inhibitor, all three compounds contain no naphthyl group but like GRL0617 were predicted to fit on the cleft near the BL2 loop. Docking studies further revealed that the location and direction of the binding determined their specificity to different coronaviruses. Together, our work demonstrates that the BL2 loop and nearby regions are outstanding druggable targets, and our three inhibitors can be applicable to the development of therapeutics for coronavirus infection.

Abstract Background Alopecia refers to a condition developed by gradual reduction of hair loss by various abnormal causes such as endocrine system, genetic factors, and stress. Stromal vascular fraction (SVF) isolated from the fat is one of the latest innovative solutions in the field of regeneration therapy. We focused on presenting effectiveness of clinical cases to improve AGA through transplantation of autologous SVF into the scalp. Objective To confirm the efficacy of the autologous SVF usage to the patients with AGA. Methods Nine patients (age range 43–64 years; 4 men, grade IV to V and 5 women, grade I to III), who are suffering from androgenic alopecia (AGA), were treated with single transplantation of autologous SVF in the upper scalp. Autologous SVF was isolated and characterized prior to the injection of live 7–9 × 106 cells into the patients’ treatment site. The hair loss improvement effect was assessed by three test criteria: hair skin quality, hair thickness and hair density 3 and 6 months after post-injection compared to pre-injection status. Results Hair density of SVF-treated side was significantly increased after 3 and 6 months of transplantation compared to non-treated side (P = 0.01 and P = 0.009 per each). And significant improvement in the score of the keratin on the scalp was seen in the injected area as compared to the non-injected area 6 months after transplantation (P = 0.032). Although thickness increase was observed at 3 and 6 months after transplantation, there was no statistical significance (P = 0.142 and 0.155, respectively). Conclusions One transplantation of autologous SVF for the AGA patients, hair density and score for the keratin were significantly increased within 6 months. This study shows that SVF is a very effective way to treat hair loss and most of subjects are satisfied with the result after treatment.

Objective To compare the convenience and effectiveness of the existing lumbosacral orthoses (LSO) (classic LSO and Cybertech) and a newly developed LSO (V-LSO) by analyzing postoperative data. Methods This prospective cohort study was performed from May 2019 to November 2019 and enrolled and analyzed 88 patients with degenerative lumbar spine disease scheduled for elective lumbar surgery. Three types of LSO that were provided according to the time of patient registration were applied for 6 weeks. Patients were randomized into the classic LSO group (n=31), Cybertech group (n=26), and V-LSO group (n=31). All patients were assessed using the Oswestry Disability Index (ODI) preoperatively and underwent plain lumbar radiography (anteroposterior and lateral views) 10 days postoperatively. Lumbar lordosis (LS angle) and frontal imbalance were measured with and without LSO. At the sixth postoperative week, a follow-up assessment with the ODI and orthosis questionnaire was conducted. Results No significant differences were found among the three groups in terms of the LS angle, frontal imbalance, ODI, and orthosis questionnaire results. When the change in the LS angle and frontal imbalance toward the reference value was defined as a positive change with and without LSO, the rate of positive change was significantly different in the V-LSO group (LS angle: 41.94% vs. 61.54% vs. 83.87%; p=0.003). Conclusion The newly developed LSO showed no difference regarding its effectiveness and compliance when compared with the existing LSO, but it was more effective in correcting lumbar lordosis.

Abstract Aerial images obtained from autonomous aerial vehicles have lots of small and densely distributed objects because of the capture distance. This paper proposes a deep neural network architecture and training/inference techniques for robust detection of objects in the aerial images. Based on cascade R‐CNN, the proposed model adopts the recursive feature pyramid and switchable atrous convolution for robust detection of dense objects. A patch‐level division and multi‐scale inference techniques are applied to effectively detect small objects. The results show that the proposed approach achieves the highest performance on the VisDrone test‐dev dataset, in the official ECCV VisDrone2020‐DET challenge.

Quantifiable erectile dysfunction (ED) diagnosis involves the monitoring of rigidity and tumescence of the penile shaft during nocturnal penile tumescence (NPT). In this work, we introduce Erectile Dysfunction SENsor (EDSEN), a home-based wearable device for quantitative penile health monitoring based on stretchable microtubular sensing technology. Two types of sensors, the T- and R-sensors, are developed to effectively measure penile tumescence and rigidity, respectively. Conical models mimicking penile shaft were fabricated with polydimethylsiloxane (PDMS) material, using different base to curing agent ratios to replicate the different hardness properties of a penile shaft. A theoretical buckling force chart for the different penile models is generated to determine sufficiency criteria for sexual intercourse. An average erect penile length and circumference requires at least a Young’s modulus of 179 kPa for optimal buckling force required for satisfactory sexual intercourse. The conical penile models were evaluated using EDSEN. Our results verified that the circumference of a penile shaft can be accurately measured by T-sensor and rigidity using the R-sensor. EDSEN provides a private and quantitative method to detect ED within the comfortable confines of the user’s home.

Background/Aims: Leptin is a hormone expressed by adipose tissue that regulates body energy homeostasis and weight loss by activating leptin receptors in the hypothalamus. Leptin receptors are also expressed in astrocytes. An anti-apoptosis effect of leptin in brain has recently been reported. However, the anti-apoptosis mechanism of leptin in the brain is unknown. Methods: To investigate whether leptin exerts protective effects against glutamate-induced apoptosis in astrocytes, we performed cell viability assays and apoptosis assays using rat primary astrocytes. Intracellular signaling pathways involved in anti-apoptosis effects of leptin were analyzed by immunoblotting together with a leptin mutant (S120A/T121A) with antagonist function and pharmacological inhibitors. Results: We found that glutamate-induced apoptosis in rat primary astrocytes was significantly decreased by treatment with leptin. Leptin inhibited glutamate-induced phosphorylation of ERK1/2 in astrocytes. The leptin S120A/T121A mutant did not inhibit glutamate-induced ERK1/2 phosphorylation and ERK1/2-mediated apoptosis. Conclusions: Collectively, our results provide initial evidence that leptin exerts an anti-apoptotic effect against glutamate toxicity through activation of intracellular signaling pathways which reverse glutamate-induced ERK1/2 phosphorylation in primary astrocytes. Therefore, our findings suggest that leptin might be considered a candidate for potential therapeutic applications in glutamate-induced brain excitotoxicity.

This study demonstrates that p-type copper-doped lanthanum manganite (LaMnO3) film on aluminum plate substrate is a stable and efficient photo-absorber for hydrogen collection from water. Compared to using photocatalyst powder, thin-film reactor is a convenient method for industrial applications as no further filtration process is required. The copper-doped LaMnO3 semiconductor films were readily deposited on aluminum plate substrates via sol-gel approach as well as spin-coating technology using photoelectrochemical cathodic current under AM 1.5 G illumination (100 mW/cm2). The influences of copper loading on LaMnO3 films in terms of surface and internal pore structure, morphology, optical and photoelectrochemical properties were comprehensively investigated. The bandgap energy and carrier density of the copper-doped LaMnO3 films were found to range between 2.670–2.677 eV and 4.80 × 1015–6.18 × 1015 cm−3, respectively. The flat band potentials of these films lied in the range of −0.424 to +0.067 V vs. Ag/AgCl electrode (or −0.217 to +0.232 V vs. NHE) based on the Mott-Schottky measurements. When the molar ratio of copper in the reaction solution was higher than 0.6, the copper-doped LaMnO3 film possessed the characteristics of a p-type semiconductor and yielded the best photo-performance when compared to n-type semiconductor films. Technology developments have expanded the diversity of interaction modalities that can be used by an agent (either a human or machine) to interact with a computer system. This expansion has created the need for more natural and user-friendly interfaces in order to achieve effective user experience and usability. More than one modality can be provided to an agent for interaction with a system to accomplish this goal, which is referred to as a multimodal interaction (MI) system. The Internet of Things (IoT) and augmented reality (AR) are popular technologies that allow interaction systems to combine the real-world context of the agent and immersive AR content. However, although MI systems have been extensively studied, there are only several studies that reviewed MI systems that used IoT and AR. Therefore, this paper presents an in-depth review of studies that proposed various MI systems utilizing IoT and AR. A total of 23 studies were identified and analyzed through a rigorous systematic literature review protocol. The results of our analysis of MI system architectures, the relationship between system components, input/output interaction modalities, and open research challenges are presented and discussed to summarize the findings and identify future research and development avenues for researchers and MI developers.

On March 25th 2019, the Belle II detector recorded the first collisions delivered by the SuperKEKB accelerator. This marked the beginning of the physics run with vertex detector. The vertex detector was aligned initially with cosmic ray tracks without magnetic field simultaneously with the drift chamber. The alignment method is based on Millepede II and the General Broken Lines track model and includes also the muon system or primary vertex position alignment. To control weak modes, we employ sensitive validation tools and various track samples can be used as alignment input, from straight cosmic tracks to mass-constrained decays. With increasing luminosity and experience, the alignment is approaching the target performance, crucial for the first physics analyses in the era of Super-BFactories. We will present the software framework for the detector calibration and alignment, the results from the first physics run and the prospects in view of the experience with the first data.

The Belle II experiment features a substantial upgrade of the Belle detector and will operate at the SuperKEKB energy-asymmetric e+e− collider at KEK in Tsukuba, Japan. The accelerator completed its first phase of commissioning in 2016, and the Belle II detector saw its first electron-positron collisions in April 2018. Belle II features a newly designed silicon vertex detector based on double-sided strip layers and DEPFET pixel layers. A subset of the vertex detector was operated in 2018 to determine background conditions (Phase 2 operation). The collaboration completed full detector installation in January 2019, and the experiment started full data taking. This paper will report on the final arrangement of the silicon vertex detector part of Belle II with a focus on online monitoring of detector conditions and data quality, on the design and use of diagnostic and reference plots, and on integration with the software framework of Belle II. Data quality monitoring plots will be discussed with a focus on simulation and acquired cosmic and collision data.

This study compares the efficacy of a soaking pretreatment with an alkaline solution for banana pseudostem prior to enzymatic hydrolysis. Banana pseudostem was pretreated by soaking in sodium hydroxide solutions at various concentrations and durations. The pretreatment more than doubled delignification but retained 82.09% of the holocellulose content and 73.74% of the cellulose content. The enzymatic (Trichoderma reesei) digestibility of pretreated banana pseudostem was found to have been enhanced by 44.41% as compared to initial biomass. This was evidenced by higher enzymatic activities (endoglucanase, exoglucanase, and β-glucosidase) on the treated sample. Meanwhile, glucose yield showed a proportional relationship with incubation time and enzyme loading throughout the hydrolysis process.

Hepatitis C virus (HCV) infection is characterized by a high frequency of chronic cases owing to the impairment of innate and adaptive immune responses. The modulation of natural killer (NK) cell functions by HCV leads to an impaired innate immune response. However, the underling mechanisms and roles of HCV proteins in this immune evasion are controversial, especially in the early phase of HCV infection. To investigate the role of HCV nonstructural proteins especially NS3 in the impairment of NK functions, NK cells were isolated from the PBMCs by negative selection. To assess the direct cytotoxicity and IFN-γ production capability of NK cells, co-cultured with uninfected, HCV-infected, HCV-NS3 DNA-transfected Huh-7.5, or HCV-NS replicon cells. To determine the effect of an NS3 serine protease inhibitor, HCV-infected Huh-7.5 cells were treated with BILN-2061. Then, NK cells were harvested and further co-cultured with K-562 target cells. NK cell functions were analyzed by flow cytometry and enzyme-linked immunosorbent assay. When co-cultured with HCV-infected Huh-7.5 cells, the natural cytotoxicity and IFN-γ production capability of NK cells were significantly reduced. NK cell functions were inhibited to similar levels upon co-culture with HCV-NS replicon cells, NS3-transfected cells, and HCV-infected Huh-7.5 cells. These reductions were restored by BILN-2061-treatment. Furthermore, BILN-2061-treatment significantly increased degranulation against K-562 target cells and IFN-γ productivity in NK cells. Consistent with these findings, the expression levels of activating NK cell receptors, such as NKp46 and NKp30, were also increased. In HCV-infected cells, the serine protease NS3 may play a role in the abrogation of NK cell functions in the early phase of infection through downregulation of NKp46 and NKp30 receptors on NK cells. Together, these results suggest that NS3 represents a novel drug target for the treatment of HCV infections.

Phytochemical investigation of the methanol (MeOH) extract of Pueraria lobata roots, known as “kudzu”, combined with liquid chromatography/mass spectrometry (LC/MS)-based analysis, resulted in the identification of four norlignans (1−4), including three new norlignans, lobatamunsolides A−C (1−3), and five known isoflavonoids (5−9). The structures of the new compounds were elucidated by a combination of spectroscopic methods, including 1D and 2D nuclear magnetic resonance (NMR) and high resolution (HR)-electrospray ionization mass spectrometry (ESIMS), and their absolute configurations were determined by chemical reaction and quantum chemical electronic circular dichroism (ECD) calculations. The isolated compounds (1−9) were evaluated for their inhibitory effects on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Compound 9 displayed the strongest NO inhibitory effect and compound 2 showed a weak effect. The potential mechanism of the effect of compound 9 was investigated by analysis of its molecular docking on the active site of inducible nitric oxide synthase (iNOS), which showed the potential interactions of compound 9 with key amino acid residues and the heme cofactor of iNOS. The mechanism as the inhibition of transcriptional iNOS protein expression was confirmed by western blotting experiments.

Currently, delay procedures remain a reliable method of maximizing flap survival but the necessity of additional procedures is a persistent disadvantage. Botulinum toxin A (BTXA) is considered to exert the most powerful neuromuscular blockade. In this study, BTXA is used to demonstrate the usefulness of a chemical delay technique. Thirty rats were subdivided into three groups of 10. In the group 1(control), no procedure was performed before the transverse rectus abdominis musculocutaneous (TRAM) flap elevation. In the group 2 (surgical delay), TRAM flaps were delayed surgically one week before formal elevation. And, in the group 3 (BTXA delay) rats, 4U BTXA was injected into the under surface of TRAM flaps two weeks before surgical elevation. The TRAM flaps were designed in rectangular shape, on the rat abdomen. On the seventh day after operation, the results were evaluated and compared in terms of flap survival area, vessel counts and vascular endothelial growth factor (VEGF) expression. The mean percentages of the flap survival area in group 2 (71.76+/-.86%) and group 3 (73.92+/-.70%) were significantly higher than those in group 1 (34.60+/-.14%) (p

Gynecomastia is enlargement of the male breast caused by parenchymal hypertrophy or a cutaneous distortion of breast skin covering or both. It can be classified by the breast size and severity of breast ptosis. If there is slight hypertrophy alone with no ptosis, then liposuction or subcutaneous mastectomy is usually sufficient. However severe gynecomastia, when hypertrophy accompanying with breast ptosis, is not enough to be corrected by liposuction or subcutaneous mastectomy, or both together. We cannot obtain a good cosmetic result by performing that method. The patient is a 18 year old man, having used steroid for 5 years from when he was five years old. He is slightly corpulent, BMI 33.9. And Simon's gynecomastia classification was grade 3. We used horizontal excision ellipse of the breast's redundant skin and deep excess tissue and a superior pedicle flap carrying the areola-nipple complex to its new site on the chest wall. We could get flat, male muscular-looking breast and symmetric areola-nipple complex position without hematoma, infection and nipple sensory loss. In treating severe gynecomastia, the horizontal excision ellipse method is good surgical technique that can draw good results in cosmetic appearance with little complication.

Zoonotic influenza A viruses constantly pose a health threat to humans as novel strains occasionally emerge from the avian population to cause human infections. Many past epidemic as well as pandemic strains have originated from avian species. While most viruses are restricted to their primary hosts, zoonotic strains can sometimes arise from mutations or reassortment, leading them to acquire the capability to escape host species barrier and successfully infect a new host. Phylogenetic analyses and genetic markers are useful in tracing the origins of zoonotic infections, but there are still no effective means to identify high risk strains prior to an outbreak. Here we show that distinct host tropism protein signatures can be used to identify possible zoonotic strains in avian species which have the potential to cause human infections. We have discovered that influenza A viruses can now be classified into avian, human, or zoonotic strains based on their host tropism protein signatures. Analysis of all influenza A viruses with complete proteome using the host tropism prediction system, based on machine learning classifications of avian and human viral proteins has uncovered distinct signatures of zoonotic strains as mosaics of avian and human viral proteins. This is in contrast with typical avian or human strains where they show mostly avian or human viral proteins in their signatures respectively. Moreover, we have found that zoonotic strains from the same influenza outbreaks carry similar host tropism protein signatures characteristic of a common ancestry. Our results demonstrate that the distinct host tropism protein signature in zoonotic strains may prove useful in influenza surveillance to rapidly identify potential high risk strains circulating in avian species, which may grant us the foresight in anticipating an impending influenza outbreak.

Human leukocyte antigen (HLA) class I molecules are critical components of the cell-mediated immune system that bind and present intracellular antigenic peptides to CD8(+) T cell receptors. To understand the interaction mechanism underlying human leukocyte antigen (HLA) class I specificity in detail, we studied the structural interaction characteristics of 16,393 nonameric peptides binding to 58 HLA-A and -B molecules. Our analysis showed for the first time that HLA-peptide intermolecular bonding patterns vary among different alleles and may be grouped in a superfamily dependent manner. Through the use of these HLA class I 'fingerprints', a high resolution HLA class I superfamily classification schema was developed. This classification is capable of separating HLA alleles into well resolved, non-overlapping clusters, which is consistent with known HLA superfamily definitions. Such structural interaction approach serves as an excellent alternative to the traditional methods of HLA superfamily definitions that use peptide binding motifs or receptor information, and will help identify appropriate antigens suitable for broad-based subunit vaccine design.

Abstract Knowing and recognizing various types of catheters placed in patients as well as their appropriate and inappropriate positioning is important knowledge to medical students and residents. Acquiring such knowledge can be more effective and efficient with a comprehensive learning resource. The Lines and Tubes Radiology Learning Module was designed to provide a systematic review of correct and incorrect line and tube placements, and is intended for clinical (third- and fourth-year) medical students, interns, and junior radiology residents. The module is a hyperlinked PowerPoint presentation with over 600 slides. The initial compilation of radiographs with line placements was acquired through PACS from one of the author's collections. Digital photographs of the lines alone and those positioned in simulation humanoid models were taken for correlation. Lines and tubes dicussed in the module include: central venous lines, Swan-Ganz catheters, endotracheal tubes, nasogastric tubes, Dobhoff tubes, Foley catheters, pacemakers, PICC, tracheostomy tubes, umbilical catheters, and misplaced guide wires. Images may be accessed by line type, or randomly. Each tube/line type includes normal and abnormal placements, as well as text describing potential complications. Images can be viewed with and without annotation illustrating the tube/line path. The module is designed to be self-administered over several sessions. Overall positive feedback was received, and it was generally perceived as a useful educational tool. A web-based version is being developed.

After two decades of quiescence, epidemic resurgence of Chikungunya fever (CHIKF) was reported in Africa, several islands in the Indian Ocean, South-East Asia and the Pacific causing unprecedented morbidity with some cases of fatality. Early phylogenetic analyses based on partial sequences of Chikungunya virus (CHIKV) have led to speculation that the virus behind recent epidemics may result in greater pathogenicity. To understand the reasons for these new epidemics, we first performed extensive analyses of existing CHIKV sequences from its introduction in 1952 to 2009. Our results revealed the existence of a continuous genotypic lineage, suggesting selective pressure is active in CHIKV evolution. We further showed that CHIKV is undergoing mild positive selection, and that site-specific mutations may be driven by cell-mediated immune pressure, with occasional changes that resulted in the loss of human leukocyte antigen (HLA) class I-restricting elements. These findings provide a basis to understand Chikungunya virus evolution and reveal the power of post-genomic analyses to understand CHIKV and other viral epidemiology. Such an approach is useful for studying the impact of host immunity on pathogen evolution, and may help identify appropriate antigens suitable for subunit vaccine formulations.

Allergy is a major health problem in industrialized countries. The number of transgenic food crops is growing rapidly creating the need for allergenicity assessment before they are introduced into human food chain. While existing bioinformatic methods have achieved good accuracies for highly conserved sequences, the discrimination of allergens and non-allergens from allergen-like non-allergen sequences remains difficult. We describe AllerHunter, a web-based computational system for the assessment of potential allergenicity and allergic cross-reactivity in proteins. It combines an iterative pairwise sequence similarity encoding scheme with SVM as the discriminating engine. The pairwise vectorization framework allows the system to model essential features in allergens that are involved in cross-reactivity, but not limited to distinct sets of physicochemical properties. The system was rigorously trained and tested using 1,356 known allergen and 13,449 putative non-allergen sequences. Extensive testing was performed for validation of the prediction models. The system is effective for distinguishing allergens and non-allergens from allergen-like non-allergen sequences. Testing results showed that AllerHunter, with a sensitivity of 83.4% and specificity of 96.4% (accuracy = 95.3%, area under the receiver operating characteristic curve AROC = 0.928+/-0.004 and Matthew's correlation coefficient MCC = 0.738), performs significantly better than a number of existing methods using an independent dataset of 1443 protein sequences. We consider the problem of throughput-optimal scheduling in wireless networks subject to interference constraints. We model the interference using a family of -hop interference models, under which no two links within a -hop distance can successfully transmit at the same time. For a given , we can obtain a throughput-optimal scheduling policy by solving the well-known maximum weighted matching problem. We show that for , the resulting problems are NP-Hard that cannot be approximated within a factor that grows polynomially with the number of nodes. Interestingly, for geometric unit-disk graphs that can be used to describe a wide range of wireless networks, the problems admit polynomial time approximation schemes within a factor arbitrarily close to 1. In these network settings, we also show that a simple greedy algorithm can provide a 49-approximation, and the maximal matching scheduling policy, which can be easily implemented in a distributed fashion, achieves a guaranteed fraction of the capacity region for "all ." The geometric constraints are crucial to obtain these throughput guarantees. These results are encouraging as they suggest that one can develop low-complexity distributed algorithms to achieve near-optimal throughput for a wide range of wireless networks.

Abstract Background In vivo proton magnetic resonance spectroscopy (1H-MRS) studies of HIV-infected humans have demonstrated significant metabolic abnormalities that vary by brain region, but the causes are poorly understood. Metabolic changes in the frontal cortex, basal ganglia and white matter in 18 SIV-infected macaques were investigated using MRS during the first month of infection. Results Changes in the N-acetylaspartate (NAA), choline (Cho), myo-inositol (MI), creatine (Cr) and glutamine/glutamate (Glx) resonances were quantified both in absolute terms and relative to the creatine resonance. Most abnormalities were observed at the time of peak viremia, 2 weeks post infection (wpi). At that time point, significant decreases in NAA and NAA/Cr, reflecting neuronal injury, were observed only in the frontal cortex. Cr was significantly elevated only in the white matter. Changes in Cho and Cho/Cr were similar across the brain regions, increasing at 2 wpi, and falling below baseline levels at 4 wpi. MI and MI/Cr levels were increased across all brain regions. Conclusion These data best support the hypothesis that different brain regions have variable intrinsic vulnerabilities to neuronal injury caused by the AIDS virus.

Background: Exercise and dietary nutrition are considered crucial in human immunodeficiency virus (HIV)/ acquired immunodeficiency syndrome (AIDS) treatment protocols and people living with HIV/AIDS (PLWHA) rehabilitation care. However, there is no well-studied research evaluating the effects of combined interventions on the fitness and immune systems of PLWHA. Therefore, this study aimed to analyze the effects of exercise and dietary intervention on physical fitness, quality of life and immune response in PLWHA., Methods: This was an experimental study, with a sample of 25 male PLWHA divided into two groups: the intervention group (IG: 12 participants) and the control group (CG: 13 participants). All participants have not had any exercise habits and nutritional supplements in the past six months. The participants in the IG completed 45 min of exercise (60-80% HRmax) 4 times per week for 4 weeks. The exercise was in the form of brisk walking or running. They were also given a nutritional dietary supplement 3 times a day for 4 weeks. The 13 individuals in the CG continued their normal daily life (physical activity and diet). The following parameters were evaluated before and after the intervention: body composition, physical fitness, immune response, quality of life (QoL), stress, dietary behavior, dietary habits, exercise motivation, and physical self-efficacy., Results: The significant changes were observed in burnout of stress variables and physical efficiency index (PEI) of physical fitness in the IG (p =.023). Moreover, in the saliva samples, sal-T levels significantly increased only after the intervention in the IG (p =.012). Additionally, regarding the analysis of the interaction (group × time), there was a significant improvement in the reaction speed (p =.001) and grip strength (left: p =.002, right: p =.030) and a significant difference in physical satisfaction in QoL (p =.001), stress burnout (p =.043), self-confidence in physical efficacy (p =.045), external display (p =.008), and fulfillment (p =.047) in exercise motivation. Moreover, the significant effect of the intervention on emotional eating in dietary behavior was shown in the comparison of the IG before and after intervention (p =.001) and in the comparison of the IG group with the CG after the experiment (p =.013). However, there was no significant effect of time or interaction between the condition and time on body composition., Conclusions: In conclusion, exercise training and diet therapy caused changes in physical fitness and Sal-T levels, which had positive effects on the health promotion of PLWHA., (© 2024. The purpose of this study is to analyze the effect of proprioceptive exercise training on soccer skills and physical fitness in young soccer players. Nineteen young soccer players participated in the study. Participants were divided into proprioceptive exercises training (PT) and control (Cont) groups. The physical fitness and soccer skill performance test assessed at pre- and posttraining. Balance ability and power significantly improved in both groups after 8 weeks of training ( P <0.05). Dribble in the PT group was similar between pre- and posttraining, with a statistically significant difference in the Cont group ( P <0.05). There were no significant differences within and between groups according to training in the left and right feet for juggling. The PT and Cont groups did not differ significantly regarding short passes posttraining. However, a significant improvement in long passes was observed in the PT group after training period ( P <0.05). Shooting was no different between and within the groups. In conclusion, these inconsistent findings are thought to be due to the applied exercise method, training duration, and the specificity of the physical growth period of the young players. Therefore, it is necessary to increase the frequency and time of proprioceptive exercise training and apply factors for cognitive ability improvement to training programs for young soccer players in future studies. The purpose of this study is to analyze the effects of the combined recovery method of cold water immersion (CWI) and protein supplement intake after eccentric exercise that causes muscle fatigue in elite soccer players. Eleven semiprofessional soccer players participated in this study. Participants were divided into CWI group, combined protein and CWI group (PCWI), and passive resting group (CON). The participants completed the eccentric exercise for one hour and performed one of three recovery methods. The muscle strength of the quadriceps and hamstring muscles significantly decreased at 48-hr postexercise compared to before exercise in all recovery groups ( P <0.05), with no significantly different between the recovery groups. The time required to sprint 40 m was significantly longer in all groups at 24 hr and 48 hr after exercise than before exercise ( P <0.05). The vertical jump height was significantly decreased at 48 hr after exercise compared to before exercise in the CON and CWI groups ( P <0.05). The muscle soreness values were higher at 6 hr, 24 hr, and 48 hr after exercise than before exercise in all groups ( P <0.001). The perceived recovery quality was reduced after exercise in the PCWI ( P <0.01) and CON groups ( P <0.001) compared to before exercise; it was unchanged in the CWI group. The recovery quality decreased at 6 hr, 24 hr, and 48 hr after exercise in all recovery groups ( P <0.01). The aim of this study was to analyze the daily physical activity and the relationship between the physical activity levels and psychological variables of Koreans with HIV. Twenty-two people with HIV participated in this study. The participants completed questionnaires, and we assessed the daily physical activity for 2 weeks. The daily physical activity level of most of the participants was low-intensity; high-intensity activities were only performed for approximately 1 minute. The participants had unhealthy dietary habits, such as eating 2 meals a day, eating irregularly, and skipping breakfast. Psychological well-being and environmental satisfaction were higher in the high-intensity group than in the medium- and low-intensity groups (P < .05). The overall stress level among the groups was statistically significantly different (P < .05), and the stress level was lower in the high-intensity group than in the low- and medium-intensity groups. Restraint eating was higher in the low-intensity group than in the medium- and high-intensity groups (P < .05). However, among the groups, the external eating variable was highest in the high-intensity group (P < .05). It is necessary to understand movement characteristics of elite female soccer players during a match to develop effective training program. The purpose of this study is to analyze the physical demands of Korean female professional soccer players during a competitive season. Twenty-four female professional players (age: 27.8±3.9 years; height: 165.7± 5.1 cm) from a team, belonging to the women's professional soccer league in South Korea participated in the study. The players participated in 11 home matches and 10 away matches from April 26 to November 9 of the 2011 season. Body weight and body mass index decreased during the first half-season compared to the preseason ( P <0.05), and the decreased values were maintained until the second half-season. Total distance covered by the players was more than 9.5 km per match. No difference was found in the total distance, movement distance by exercise intensities, number of sprints and accelerations, and maximum speed between the first half- and second half-seasons. Midfielders covered the most distance during a match compared to other positions ( P <0.05). Wing forward covered the most distance of high-intensity exercise. Maximum speed during the match was higher among wing forward and forward than in other positions ( P <0.05). There were no significant differences between home and away matches in all variables ( P >0.05). Exercise training involving exercises of optimal intensity and duration improves psychological and medical variables in relative leisure-deprived people living with HIV/AIDS. This study aimed to analyze associated psychological variables and the effect of exercise intensity and duration on immune responses in relative leisure-deprived people infected with HIV. The participants completed different moderate-intensity exercises (30 min (60−80% HRmax) and 45 min (60−80% HRmax)) and high-intensity exercise for 10 min (>80% HRmax). Levels higher than “normal” were rated for relative leisure deprivation, indicating relative deprivation of leisure among participants. The overall level of quality of life was “normal”, indicating that quality of life was not considered high. The stress level was psychologically considered low. Time had a significant effect on cortisol levels (p < 0.05). Compared to pre-exercise, cortisol level was significantly decreased immediately after moderate exercise for 45 min and 3 h post-exercise after high-intensity exercise for 10 min (p < 0.05). However, time and the interaction of condition and time had no significant effect on IL-6 and sIgA levels (p > 0.05). Despite the small sample size of this pilot study, the results demonstrate that moderate-intensity exercise can be recommended to improve the health and quality of life of people infected with HIV.

The purpose of this study was to analyze the characteristics of injuries in female national soccer players. The injuries data were recorded by Korea Football Association medical staff using the Daily Medical Report provided by the Asian Football Confederation during a total 21 training camps and friendly and official matches from January 2014 to December 2018. A total of 483 players in the national team training camps and matches participated in the study, of which 377 players experienced more than one injury. Sports injuries accounted for 89% of the total 1,019 injuries; general diseases accounted for the remaining 11%. The number of injuries per 1,000 exposure hours was higher in matches than in training sessions. Most injuries that occurred during training and matches involved the lower extremities (79%). The most common injury diagnosed during training and matches was muscle rigidity (45%), followed by contusions/hematomas/bruises (22%) and sprain (15%). The largest proportions of injuries in terms of absence from soccer play were minimal injury (67%), followed by mild (29%), moderate (3.5%), and severe (1%). As conclusion, the characteristics of injuries during training sessions and matches were different. Exercise performance is reduced in hot environments due to physiological responses caused by increased body temperature. A proper residential environment is important for improving the performance and maintaining physical condition of soccer players in the summer. The purpose of this study was to determine the effect of indoor temperature of the resting space during the summer on the fitness and condition of soccer players. A total of 12 K-3 League semiprofessional players without serious injuries in the last 3 months voluntarily participated in the study. Participants performed speed (10 m, 20 m, and 30 m), soccer-specific coordination skill (dribbling), agility, repeated sprints, Yo-Yo intermittent level 2, vertical jump, and questionnaire (fatigue, sleep quality, muscle soreness, stress, and mood) after staying indoor temperature at 20°C, 26°C, and 30°C for one night, respectively. There was no difference among groups in physical fitness (speed, agility, jump, coordination, Yo-Yo intermittent level 2, and repeated sprints). The differences in fatigue and sleep quality were not statistically significant among groups, but they tended to be different. Muscle soreness was similar among all groups. Significant differences were observed between the 20°C and 30°C groups in stress and mood levels. The present study concluded that, while the physical fitness did not differ among groups, the 30°C residential environment was shown to have a negative psychological effect. There may be an optimal period of time to maximize the improvement of physical fitness during adolescence. The aim of this study was to examine the magnitude of changes in physical fitness after 8 weeks of preseason training according to chronological ages after the age at peak high velocity. Thirty male young football players from an elite football team (U-16, n=10; U-17, n=10; U-18, n=10) participated in the study. The players completed an 8-week general preseason football training and participated in the pre- and posttests to measure physical fitness. The 8-week preseason training improved the power of all young players ( P <0.05). The 20-m sprint performance was improved by training in U-16 and U-18 ( P <0.05), but no changes were found in the U-17 group ( P >0.05). Significant differences were found in the arrowhead left in U-16 and U-18 ( P <0.05) after training; however, no difference was observed in U-17 ( P >0.05). Coordination was enhanced further in U-16 and U-17 ( P <0.05) compared with that in U-18 ( P >0.05). The performance of repeated sprints and Yo-Yo intermittent recovery test level 1 (Yo-Yo IR1) were similar between pre- and posttraining in all age groups ( P >0.05). Collectively, the results emphasized the importance of systematic and scientific training methods to improve the fitness levels of young football players in the preseason training period. As a new class of cancer therapeutic agents, oncolytic viruses (OVs) have gained much attention not only due to their ability to selectively replicate in and lyse tumor cells, but also for their potential to stimulate antitumor immune responses. As a result, there is an increasing need for in vitro modeling systems capable of recapitulating the 3D physiological tumor microenvironment. Here, we investigated the potential of our recently developed microphysiological system (MPS), featuring a vessel-like channel to reflect the in vivo tumor microenvironment and serving as culture spaces for 3D multicellular tumor spheroids (MCTSs). The MCTSs consist of cancer A549 cells, stromal MRC5 cells, endothelial HUVECs, as well as the extracellular matrix. 3D MCTSs residing in the MPS were infected with oncolytic VSV expressing GFP (oVSV-GFP). Post-infection, GFP signal intensity increased only in A549 cells of the MPS. On the other hand, HUVECs were susceptible to virus infection under 2D culture and IFN-β secretion was quite delayed in HUVECs. These results thus demonstrate that OV antitumoral characteristics can be readily monitored in the MPS and that its behavior therein somewhat differs compared to its activity in 2D system. In conclusion, we present the first application of the MPS, an in vitro model that was developed to better reflect in vivo conditions. Immune reactions are controlled by the delicate spatiotemporal orchestration of multiple cells communicating by cytokines. Studies of cytokines that began with the discovery of IFN focused on positive regulatory mechanisms that induce secretion in response to harmful stimuli. However, there is a growing awareness that negative regulatory mechanisms that stop secretion of cytokines at specific times and spaces are also important for a successful immune reaction. Type I IFN is the primary cytokine in innate immunity. Although its induction is a prerequisite for the consequent adaptive immune reaction, its oversecretion can cause destructive tissue damage. IFN regulatory factor 7 (IRF7) is a master transcription factor of type I IFN, and multiple observations indicate the key role of IRF7 and the importance of its negative regulation. In this study, we found that the inducible heat shock protein 70 (HSP70) regulated the early type I IFN response by using mice knockout for HSP70. HSP70 dampened IRF7 activation; the inhibitory effect of HSP70 over IKKε-mediated IRF7 activation originated from simple competitive binding. Replicable oncolytic viruses (OVs) induce tumor cell lysis and release viral progeny. The released progeny virions and cell debris can spread within surrounding tumor cells or blood vessels. These released molecules may also induce bystander damage in additional tumor cells through spreading within surrounding tumor cells or blood vessels. However, this effect has not been clearly demonstrated due to the difficulty of direct observation. Here, the bystander infection of OVs by vessel delivery and selective infection in 3D multicellular tumoroids (MCTs) in an in vitro microphysiological system (MPS) with integrated medium flow is demonstrated. This study uses replicable vesicular stomatitis virus (VSV)-green fluorescence protein (GFP) to identify the location of infection in 3D MCTs. Using this MPS, the oncoselective infection by VSV-GFP and the spreading by delivery of OVs through flow via block-to-block linkage of the primary infected MPS with uninfected 3D MCTs in an integrated MPS is observed. This MPS enables real-time monitoring and various analysis for the bystander infection of OVs. The purpose of this study was to identify the frequency and characteristics of injuries that occurred during the training camps and matches of the national soccer teams (under-20 [U-20], under-23 [U-23], A men's national teams). The research data were based on injuries that occurred from January 2016 to December 2017. The athletic trainer, a member of the Korea Football Association, recorded injuries based on the recording sheet of the Asian Football Confederation Daily Medical Report. A total of 1,689 (984 in 2014; 705 in 2017) injuries were reported for the 2 years. Most injuries were sports injuries (95%, sprains, etc.), and other conditions such as respiratory, digestive, and ocular, comprised only 5% of cases. The most injured areas of the national team players were the lower limbs (85%, 1,366). The frequency of the head/trunk and upper extremity injuries were relatively low (8.4%, 135; 6.5%, 105). The frequency of injuries according to positions were as follows: midfielder (40.3%, 648), defender (36.6%, 688), forward (17.8%, 286), and goalkeeper (5.2%, 84). The most common types of injuries were contusions (37.3%, 599), followed by muscle rigidity (29.0%, 466) and sprains (16.8%, 270). The characteristics of injuries amongst national soccer teams, according to age, were similar, but the injury characteristics for each position were different. Joo, CH and Jee, H. Activity profiles of top-class players and referees and accuracy in foul decision-making during Korean national league soccer games. J Strength Cond Res 33(9): 2530-2540, 2019-The aims of this study were to compare the activity profiles between the top-class referees and players and elucidate the factors related to foul decision-making. Three hundred thirty-five elite-level players and referees were analyzed for distance covered during 20 matches of nationally held 2016 Korean league competitions. Distance covered by the players and referees was analyzed for the activity zones (slow walking, walking, jogging, running, high-intensity running, and sprinting) and 15-minute match periods. Mean distance between foul play and referee locations, foul plays, and 15-minute match periods were compared with the foul decision errors. Foul play and decision error rates (%) were also analyzed per segmented pitch zone. Although the total distance covered during a match and distances covered by jogging, running, and sprinting were significantly different between the players and referees, differences were within 1%. Significant differences in the distance covered before and after halftime were observed. The greatest distance between the foul play and referee locations, number of foul plays, and number of foul decision errors were observed at the 75-minute match period. Finally, the greater number of foul plays was observed in the neutral and attacking zones, and the foul decision errors were observed in the right defensive and left attacking zones 1. In conclusion, although the activity profiles may be different, referees should maintain certain level of physical fitness to match that of the players. To reduce the number of foul decision errors, factors such as match time, foul occurring location, and distance between foul play and referee locations should be considered.

The airway is the major entry route of pathogens due to continuous gas exchange with the environment. In particular, the nasal epithelial layer is the common site of airborne mucotropic virus infections. The inflammatory response to such infections must be tightly controlled due to its non-specific nature. Unrestrained inflammation breaks down the physiological mucosal defense system and leads to secondary bacterial or fungal infections. Chronic rhinosinusitis (CRS) is a prevalent inflammatory disease that compromises quality of life. In spite of its importance in the initiation of inflammation, the role of interferon signaling in nasal airway epithelial cells is largely unknown. We analyzed the expression of interferon signaling genes using clinical lavage specimens and nasal airway epithelial cells collected from CRS patients and controls. Basal expression of IFNAs, IKBKE, STAT1, and some CXC chemokines was elevated in samples from CRS patients. In subsequent in vitro studies, we found IKKε to be the key molecule and augmented CXCL10 secretion. Based on our findings and review of the literature, we hypothesized that high levels of IKKε might induce intractable inflammation via CXCL10. We tested the hypothesis in an animal model and found not only that IKKε induced severe eosinophilic inflammation with CXCL10 over-production, but also that inhibition of IKKε resolved the inflammation., (Copyright © 2019 Elsevier Inc. Purpose: The aim of this study was to examine the effects of aerobic high-intensity training with reduced volume and training cessation on body composition and physical fitness after the end of season and the time required to recapture physical fitness with intensified retraining following two weeks of detraining in elite soccer players., Method: Twenty male semi-professional soccer players participated in this study. The soccer players were assigned to either a group that completed high-intensity aerobic training (HAT, n = 10) or to a detraining and retraining group (DHAT, n = 10) for a 5-week period immediately after the end of the season. The first 2 weeks of the period, members of the HAT group performed high-intensity aerobic exercise (80-90% of HRmax, 12 min × 3, three times per week), whereas members of the DHAT group abstained from any physical activity. During the subsequent 3 weeks, members of both the HAT and DHAT groups completed high-intensity aerobic exercise. Exercise performance testing and body composition analysis were performed before; after 2 weeks of detraining; and at 1, 2 and 3 weeks of retraining., Results: Intensified high-intensity training for 5 weeks maintained the performance in the Yo-Yo Intermittent Recovery level 2 test (Yo-Yo IR2) and repeated sprints at any time point (P > 0.05). However 2 weeks of detraining resulted in significant decreases in the performance on the Yo-Yo IR2 (P < 0.01) and repeated sprints test (P < 0.05). Performance on the Yo-Yo IR2 enhanced after 2 weeks of retraining and was maintained up to 3 weeks after retraining, with no significant differences between conditions (P > 0.05). In addition, repeated sprint performance markedly decreased after the detraining period (P < 0.05) and was continuously lower compared to the baseline at 2 weeks after retraining (P < 0.05). Furthermore, this value reached baseline level at the end of the experimental period (P > 0.05). There were no significant differences between conditions in body composition, performance of agility, or sprint ability throughout the 5-week experimental period (P > 0.05)., Conclusions: The present data suggest that short-term detraining after the competitive season can markedly decrease performances in the Yo-Yo IR2 test and repeated sprints. To return to a previous level of ability on the Yo-Yo IR2 and/or sprint test with retraining through high-intensity aerobic training after a period of detraining, a similar or longer period of retraining is required. However, the high-intensity training with reduced amount of training after competitive season can prevent reductions in physical fitness.

The aims of this study were to examine the differences in technical aspects and physical demands between small-size games (SSG; 8 vs. 8) and regular-size games (RSG; 11 vs. 11) in young Korean soccer players. Seventy-nine young soccer players from 6 teams (U-12) volunteered to participate in the study. The players completed 4 games (2 SSG, 62×51 m, and 2 RSG, 80×54 m) in 2 days. Each game was filmed to evaluate technical actions. Physical demand variables were measured using global positioning system technology. SSG showed significantly greater numbers of technical plays among 17 variables when compared to RSG ( P <0.05). The players covered significantly greater total distance during low-, moderate-, and high-speed running and sprinting in SSG than in RSG ( P <0.05). Higher numbers of high-intensity activities (repeated high-intensity efforts, explosive efforts, decelera-tions, accelerations, and sprinting) were observed in SSG compared to RSG ( P <0.05). Mean heart rate was also higher in SSG than in RSG ( P <0.05). Despite the greater physical demands during SSG, the exercise intensity was similar to that reported in previous studies. The purpose of this study was to compare performance factors of youth soccer players according to position. A total of 101 high school soccer players were selected and were classified into goalkeeper (n=7), defense (n=37), midfield (n=39), and forward (n=18) positions. All subjects were subjected to the Wingate test for anaerobic capacity, shuttle run test for aerobic capacity, and pass, kick, dribble, and shooting tests for soccer skills. There was no significant difference in aerobic capacity according to position. However, anaerobic capacity was significantly higher in defenders than midfielders ( P <0.05), and soccer skills were significant lower in goalies than in other positions ( P <0.01). The results show significant differences in anaerobic capacity and soccer skills according to position in youth soccer players. Therefore, we suggest that middle and high school soccer players should improve aerobic, an-aerobic capacity, and soccer skills irrespective position to achieve high-level soccer performance.

Purpose: We tested the hypothesis that both post-exercise and passive cold water immersion (CWI) increases PGC-1α and VEGF mRNA expression in human skeletal muscle., Method: Study 1 Nine males completed an intermittent running protocol (8 × 3-min bouts at 90 % [Formula: see text], interspersed with 3-min active recovery (1.5-min at 25 % and 1.5-min at 50 % [Formula: see text]) before undergoing CWI (10 min at 8 °C) or seated rest (CONT) in a counterbalanced, randomised manner. Study 2 Ten males underwent an identical CWI protocol under passive conditions., Results: Study 1 PGC-1α mRNA increased in CONT (~3.4-fold; P < 0.001) and CWI (~5.9-fold; P < 0.001) at 3 h post-exercise with a greater increase observed in CWI (P < 0.001). VEGFtotal mRNA increased after CWI only (~2.4-fold) compared with CONT (~1.1-fold) at 3 h post-exercise (P < 0.01). Study 2 Following CWI, PGC-1α mRNA expression was significantly increased ~1.3-fold (P = 0.001) and 1.4-fold (P = 0.0004) at 3 and 6 h, respectively. Similarly, VEGF165 mRNA was significantly increased in CWI ~1.9-fold (P = 0.03) and 2.2-fold (P = 0.009) at 3 and 6 h post-immersion., Conclusions: Data confirm post-exercise CWI augments the acute exercise-induced expression of PGC-1α mRNA in human skeletal muscle compared to exercise per se. Additionally CWI per se mediates the activation of PGC-1α and VEGF mRNA expression in human skeletal muscle.
