There is a need to improve fishing methods to select for certain sizes and species while excluding others. Experiments are conducted to quantify selectivity of fishing gears and how variables such as gear design (e.g., mesh size, mesh shape), environmental parameters (e.g., light, turbidity, substrate) or biological parameters (e.g., fish condition) alter selectivity; the resulting data need to be analyzed using specialized statistical methods in many cases. Here, we present a new tool for analyzing this type of data: an R package named 'selfisher'. It allows estimating multiple fixed effects (e.g., fish length, total catch weight, environmental variables) and random effects (e.g., haul). A bootstrapping procedure is also provided. We demonstrate its use via four case studies, including (A) covered codend analyses of four gears, (B) a paired gear study with numerous covariates, (C) a catch comparison study of unpaired hauls of gillnets and (D) a catch comparison study of paired hauls using polynomials and splines. This software will make it easier to model selectivity, teach statistical methods, and make analyses more repeatable. l‐(+)‐Muscarine (1)‐producing mushrooms pose a severe threat to human health as ingestion can result in circulatory collapse or even death. However, their metabolic profile is surprisingly poorly understood, including knowledge of poison release and potentially toxic congeners. In the mycelium of the 1‐producing fool's funnel mushroom Clitocybe rivulosa, we identified 4'‐phosphomuscarine (2) as the major natural product. Its structure was elucidated by high‐resolution mass spectrometry, nuclear magnetic resonance spectroscopy and by comparison with a synthesized reference. We also detected this previously overlooked phosphorylated compound in the fiber cap mushrooms Pseudosperma spectrale and Inocybe nitidiuscula. Studies on the activation of the muscarinic acetylcholine receptor M3 indicate only weak affinity of 2 to this target. Furthermore, we present biological evidence that muscaridine (3), a quaternary amine congener related to and co‐occurring with 1, does not activate the muscarinic acetylcholine receptor M3 on human embryonic kidney cells. Our work provides important insight into the metabolic profile and the pharmacology of some of the most poisonous mushrooms. As the harmless 2 can liberate the potentially fatal 1 by unspecific enzymatic ester cleavage, these results are highly relevant for emergency medicine to estimate the true toxicity of these mushrooms. A library of degradable poly(2-alkyl-2-oxazoline) analogues (dPOx) with different length of the alkyl substituents was characterized in detail by gradient elution liquid chromatography. The hydrophobicity increased with increased side chain length as confirmed by a hydrophobicity row, established by reversed-phase liquid chromatography. Those dPOx were cytocompatible and formed colloidally stable nanoparticle (NP) formulations with positive zeta potential. Dynamic light scattering (DLS) revealed that dPOx with increased hydrophobicity tended to form NPs with increased sizes. NPs created from the most hydrophobic polymer, degradable poly(2-nonyl-2-oxazoline) (dPNonOx), showed tendency for aggregation at pH 5.0, and in the presence of protease in solution, in particular for NPs formulated without surfactant. Liquid chromatography revealed enzymatic degradation of dPNonOx NPs, clearly demonstrating the disappearance of polymer signals and the appearance of hydrophilic degradation products eluting close to the chromatographic void time. The degradation process was confirmed by 1H NMR spectroscopy. dPNonOx NPs containing the anti-inflammatory drug BRP-201 as payload reduced 5-lipoxygenase activity in human neutrophils. Thereby, composition analysis of the resultant NPs, including drug quantification, was also enabled by liquid chromatography. The results indicate the importance of a detailed analysis of the final polymer-based NP formulations by a multimethod approach, including, next to standard applied techniques such as DLS/ELS, the underexplored potential of liquid chromatography. The latter is demonstrated to resolve a fine structure of solution composition, together with an assessment of possible degradation pathways and is versatile in determining hydrophobicity/hydrophilicity of polymer materials. Gliotoxin (GT), a secondary metabolite and virulence factor of the fungal pathogen Aspergillus fumigatus, suppresses innate immunity and supports the suppression of host immune responses. Recently, we revealed that GT blocks the formation of the chemotactic lipid mediator leukotriene (LT)B4 in activated human neutrophils and monocytes, and in rodents in vivo, by directly inhibiting LTA4 hydrolase. Here, we elucidated the impact of GT on LTB4 biosynthesis and the entire lipid mediator networks in human M1‐ and M2‐like monocyte‐derived macrophages (MDMs) and in human tissue‐resident alveolar macrophages. In activated M1‐MDMs with high capacities to generate LTs, the formation of LTB4 was effectively suppressed by GT, connected to attenuated macrophage phagocytic activity as well as human neutrophil movement and migration. In resting macrophages, especially in M1‐MDMs, GT elicited strong formation of prostaglandins, while bacterial exotoxins from Staphylococcus aureus evoked a broad spectrum of lipid mediator biosynthesis in both MDM phenotypes. We conclude that GT impairs functions of activated innate immune cells through selective suppression of LTB4 biosynthesis, while GT may also prime the immune system by provoking prostaglandin formation in macrophages. BackgroundInfluenza causes substantial morbidity and mortality despite available treatments. Anecdotal reports suggest that plasma with high antibody titres to influenza might be of benefit in the treatment of severe influenza.MethodsIn this randomised, open-label, multicentre, phase 2 trial, 29 academic medical centres in the USA assessed the safety and efficacy of anti-influenza plasma with haemagglutination inhibition antibody titres of 1:80 or more to the infecting strain. Hospitalised children and adults (including pregnant women) with severe influenza A or B (defined as the presence of hypoxia or tachypnoea) were randomly assigned to receive either two units (or paediatric equivalent) of anti-influenza plasma plus standard care, versus standard care alone, and were followed up for 28 days. The primary endpoint was time to normalisation of patients' respiratory status (respiratory rate of ≤20 breaths per min for adults or age-defined thresholds of 20-38 breaths per min for children) and a room air oxygen saturation of 93% or more. This study is registered with ClinicalTrials.gov, number NCT01052480.FindingsBetween Jan 13, 2011, and March 2, 2015, 113 participants were screened for eligibility and 98 were randomly assigned from 20 out of 29 participating sites. Of the participants with confirmed influenza (by PCR), 28 (67%) of 42 in the plasma plus standard care group normalised their respiratory status by day 28 compared with 24 (53%) of 45 participants on standard care alone (p=0·069). The hazard ratio (HR) comparing plasma plus standard care with standard care alone was 1·71 (95% CI 0·96-3·06). Six participants died, one (2%) from the plasma plus standard care group and five (10%) from the standard care group (HR 0·19 [95% CI 0·02-1·65], p=0·093). Participants in the plasma plus standard care group had non-significant reductions in days in hospital (median 6 days [IQR 4-16] vs 11 days [5-25], p=0·13) and days on mechanical ventilation (median 0 days [IQR 0-6] vs 3 days [0-14], p=0·14). Fewer plasma plus standard care participants had serious adverse events compared with standard care alone recipients (nine [20%] of 46 vs 20 [38%] of 52, p=0·041), the most frequent of which were acute respiratory distress syndrome (one [2%] vs two [4%] patients) and stroke (one [2%] vs two [4%] patients).InterpretationAlthough there was no significant effect of plasma treatment on the primary endpoint, the treatment seemed safe and well tolerated. This study examines the impact of sludge manure adoption on household farm production, focusing on matooke (bananas), maize, beans, sweet potatoes, coffee and cassava. We employ a propensity score matching methodology and estimate the average treatment effect on the treated (ATT), and we report the results from the nearest neighbor algorithm and test for robustness using the kernel algorithm. The study uses household survey data collected from the central districts of Uganda between January and February 2023. Our key findings indicate increased and significant yields of bananas, maize, coffee and cassava. These results are similar to those of different estimation algorithms. From a policy perspective, our results suggest that the design of agricultural productivity enhancing programmes, especially for farm households, requires leveraging organic technologies to promote agricultural production.
