Abstract Cryptosporidium parvum and C. hominis are parasites that cause life-threatening diarrhea in children and immunocompromised people. There is only one approved treatment that is modestly effective for children and ineffective for AIDS patients. Here, screening 278 compounds from the Merck KGaA, Darmstadt, Germany collection and accelerated follow-up enabled by prior investigation of the compounds identifies a series of pyrazolopyrimidine human phosphodiesterase (PDE)-V (hPDE-V) inhibitors with potent anticryptosporidial activity and efficacy following oral administration in C. parvum-infected male mice. The lead compounds affect parasite host cell egress, inhibit both C. parvum and C. hominis, work rapidly, and have minimal off-target effects in a safety screening panel. Interestingly, the hPDE-V inhibitors sildenafil and the 4-aminoquinoline compound 7a do not affect Cryptosporidium. C. parvum expresses one PDE (CpPDE1) continuously during asexual growth, the inhibited life stage. According to homology modeling and docking, the lead compounds interact with CpPDE1. Bulkier amino acids (Val900 and His884) in the CpPDE1 active site replace alanines in hPDE-V and block sildenafil binding. Supporting this, sildenafil kills a CRISPR-engineered Cryptosporidium CpPDE1 V900A mutant. The CpPDE1 mutation also alters parasite susceptibility to pyrazolopyrimidines. CpPDE1 is therefore a validated pyrazolopyrimidine molecular target to exploit for target-based optimization for improved anticryptosporidial development.

IntroductionA promising approach to optimizing recovery in youth football has been the use of machine learning (ML) models to predict recovery states and prevent mental fatigue. This research investigates the application of ML models in classifying male young football players aged under (U)15, U17, and U19 according to their recovery state. Weekly training load data were systematically monitored across three age groups throughout the initial month of the 2019–2020 competitive season, covering 18 training sessions and 120 observation instances. Outfield players were tracked using portable 18-Hz global positioning system (GPS) devices, while heart rate (HR) was measured using 1 Hz telemetry HR bands. The rating of perceived exertion (RPE 6–20) and total quality recovery (TQR 6–20) scores were employed to evaluate perceived exertion, internal training load, and recovery state, respectively. Data preprocessing involved handling missing values, normalization, and feature selection using correlation coefficients and a random forest (RF) classifier. Five ML algorithms [K-nearest neighbors (KNN), extreme gradient boosting (XGBoost), support vector machine (SVM), RF, and decision tree (DT)] were assessed for classification performance. The K-fold method was employed to cross-validate the ML outputs.ResultsA high accuracy for this ML classification model (73–100%) was verified. The feature selection highlighted critical variables, and we implemented the ML algorithms considering a panel of 9 variables (U15, U19, body mass, accelerations, decelerations, training weeks, sprint distance, and RPE). These features were included according to their percentage of importance (3–18%). The results were cross-validated with good accuracy across 5-fold (79%).ConclusionThe five ML models, in combination with weekly data, demonstrated the efficacy of wearable device-collected features as an efficient combination in predicting football players’ recovery states.

OBJECTIVES Type 2 diabetes (T2D) is considered one of the most prevalent diseases worldwide. The aim of this study was to analyse the risk of developing T2D in a higher education community of Bragança, North-Eastern Portugal. METHODS The observational, cross-sectional and prospective sample comprised 3021 individuals, aged between 18 and 64 years old, of whom 1759 were women (mean age 24.16 ± 8.80 years) and 1263 were men (mean age 23.37 ± 8.70 years). The Finnish Diabetes Risk Score (FINDRISC) questionnaire was employed to estimate the risk of developing T2D over the next 10 years. RESULTS The results indicated that approximately 37 (1%) participants in the sample exhibited a high risk of developing T2D, 117 (4%) participants exhibited a moderate risk, 660 (22%) participants exhibited a slight risk, and 2206 (73%) participants exhibited a low risk. A higher T2D risk was observed in the staff group in comparison with the other groups (p < 0.001). Furthermore, body mass index (BMI) and waist circumference (WC) were the variables that most contributed to the increased risk of developing T2D (X2 = 1034.5; p < 0.001). CONCLUSIONS In conclusion, the study reported a low T2D risk for the higher education community of Bragança in the Portuguese North-Eastern region. However, the risk of T2D was found to be higher in the staff group than in the student group. The FINDRISK questionnaire may be applied for the early diagnosis of T2D risk factors.

Alzheimer's dementia leads to a progressive decline in the individual's cognitive capacity, interfering with activities of daily living. Applying cognitive stimuli in the early stages can add value to slowing cognitive decline. In this process, can the use of digital technologies help to avoid the use of medication and delay the institutionalization of Alzheimer's patients? Therefore, the general aim of this article was to identify their perceptions of the use of digital technologies in the cognitive stimulation of Alzheimer's patients with formal careers. To this end, an action research methodology was applied, researching the state-of-the-art digital technologies that could be used in Alzheimer's disease from the perspective of formal careers. Sixty-two formal careers participated in this study, 74.2% of whom were female and the rest (25.8%) male. All those interviewed can identify signs of Alzheimer's dementia in patients. Regarding working with digital technologies with Alzheimer's dementia patients, a large percentage of the interviewees (73.6 percent) replied that they only use them sometimes, and 26.4 percent said they never use them. The data suggest that there is still resistance or a lack of confidence in the use of digital technologies on the part of formal careers working with patients with Alzheimer's dementia. However, even if they have never received specific training in this area, especially in cognitive stimulation, we can admit this is understandable.

Background: Childhood obesity presents a significant public health concern globally, with implications for cardiovascular health and metabolic syndrome. In Portugal, approximately 31.6% of children are affected, highlighting the urgency for intervention strategies. This study aimed to assess the prevalence of overweight and obesity in Portuguese school-aged children, with a focus on sex and age differences. Methods: Anthropometric measurements were conducted on 1564 children aged 6–10 years, including weight, height, and skinfold thickness. Body Mass Index (BMI) and the percentage of body fat were calculated using established methods. Results: The results revealed significant differences in BMI (≤0.001) and body fat percentage (≤0.001) among different BMI categories, with a notable prevalence of overweight and obesity, particularly among boys. A total of 37% of the studied population is overweight or obese, among which 40.1% and 33.9% are boys and girls, respectively. Conclusions: This study highlights statistically significant differences in BMI and body fat percentage for both sexes in different BMI categories. A large proportion of the population is overweight or obese, with a greater prevalence in boys. In short, childhood obesity has a negative impact on body composition and is associated with significant differences in anthropometric parameters, emphasizing the importance of preventative and intervention strategies to address this health problem.

Introduction: The purpose of this study was to investigate the ideal training load to be applied during periods of fixture congestion to ensure an adequate dose-response effect for performance maintenance.Methods: Match performance data and corresponding pre-match training load sessions (both N = 498 match performance cases and training-block session cases) were collected (with the catapult system, VECTOR7) from 36 male professional soccer players (23.5 ± 5.2 years; 178 ± 4 cm; 75.5 ± 6.0 kg) belonging to the Brazilian First Division team during the 2022 season. The following data were collected in match and training sessions: jump, acceleration, deceleration, and change of direction (COD); running distance producing metabolic power at different intensities (>20, >20–35, >35–45, >45–55, and >55 W kg−1), total distance (m), relative distance (m/min), running distance at different speeds (>20, >25, and >30 km/h), number of sprints (running >25 km/h), and maximum speed (km/h). Mixed linear model (MLM), decision tree regression (DTR), and cluster K means model (SPSS v.26) approach were performed to identify the most critical variables (and their respective load) in the training sessions that could explain the athlete’s match performance.Results: MLM and DTR regression show that training load significantly affects game performance in a specific way. According to the present data, an interference phenomenon can occur when a high load of two different skills (running in a straight line vs COD, deceleration, and jumping) is applied in the same training block of the week. The cluster approach, followed by a chi-squared test, identified significant associations between training load and athlete match performance in a dose-dependent manner.Discussion: The high load values described here have a beneficial effect on match performance, despite the interference between stimuli discussed above. We present a positive training load from a congested season from the Brazilian First Division team. The study suggests that an interference effect occurs when high physical training loads are applied to different specific physical skills throughout the season.

The aim of this study was to analyze the match-to-match variation in high-intensity demands from one Portuguese professional football team according to playing positions. Twenty-three male outfield professional football players were observed during eighteen matches of the Portuguese Second League. Time–motion data were collected using Global Positioning System (GPS) technology. Match running performance was analyzed based on the following three playing positions: defenders (DF), midfielders (MF), and forwards (FW). Repeated measures ANOVA was utilized to compare match running performance within each position role, and seasonal running variation. Practical differences were assessed using the smallest worthwhile change (SWC), coefficient of variation (CV), and twice the coefficient of variation (2CV). Significant differences were found among playing positions in total distance covered (F = 15.45, p < 0.001, η2 = 0.33), average speed (F = 12.79, p < 0.001, η2 = 0.29), high-speed running (F = 16.93, p < 0.001, η2 = 0.36), sprinting (F = 13.49, p < 0.001, η2 = 0.31), accelerations (F = 4.69, p = 0.001, η2 = 0.132), and decelerations (F = 12.21, p < 0.001, η2 = 0.284). The match-to-match running performance encompassed TD (6.59%), AvS (8.67%), HSRr (37.83%), SPR (34.82%), ACC (26.92%), and DEC (27.85%). CV values for total distance covered ranged from 4.87–6.82%, with forwards and midfielders exhibiting the greatest and smallest variation, respectively. Midfielders demonstrated the highest match-to-match variation for all other analyzed variables (8.12–69.17%). All playing positions showed significant variation in high-demanding variables (26.94–37.83%). This study presents the initial analysis of match-to-match variation in high-intensity demands within a Portuguese professional football team. Thus, the position’s specificity and context can provide a helpful strategy for evaluating match-to-match running performance, and for recommending individualized training exercises based on the peak and high-intensity demands for each player’s role within the game.

The aim of this study was to test a machine learning (ML) model to predict high-intensity actions and body impacts during youth football training. Sixty under-15, -17, and -19 sub-elite Portuguese football players were monitored over a 6-week period. External training load data were collected from the target variables of accelerations (ACCs), decelerations (DECs), and dynamic stress load (DSL) using an 18 Hz global positioning system (GPS). Additionally, we monitored the perceived exertion and biological characteristics using total quality recovery (TQR), rating of perceived exertion (RPE), session RPE (sRPE), chronological age, maturation offset (MO), and age at peak height velocity (APHV). The ML model was computed by a feature selection process with a linear regression forecast and bootstrap method. The predictive analysis revealed that the players’ MO demonstrated varying degrees of effectiveness in predicting their DEC and ACC across different ranges of IQR. After predictive analysis, the following performance values were observed: DEC (x¯predicted = 41, β = 3.24, intercept = 37.0), lower IQR (IQRpredicted = 36.6, β = 3.24, intercept = 37.0), and upper IQR (IQRpredicted = 46 decelerations, β = 3.24, intercept = 37.0). The player’s MO also demonstrated the ability to predict their upper IQR (IQRpredicted = 51, β = 3.8, intercept = 40.62), lower IQR (IQRpredicted = 40, β = 3.8, intercept = 40.62), and ACC (x¯predicted = 46 accelerations, β = 3.8, intercept = 40.62). The ML model showed poor performance in predicting the players’ ACC and DEC using MO (MSE = 2.47–4.76; RMSE = 1.57–2.18: R2 = −0.78–0.02). Maturational concerns are prevalent in football performance and should be regularly checked, as the current ML model treated MO as the sole variable for ACC, DEC, and DSL. Applying ML models to assess automated tracking data can be an effective strategy, particularly in the context of forecasting peak ACC, DEC, and bodily effects in sub-elite youth football training.

IntroductionThe application of muscle force is a determinant of football success as it is inherent to the motor control and sport. The aims of this study are: (1) to describe force variables Isometric Maximal Force (IMF), Concentric Peak Force (CPPF), and Dynamic Strength Index (DSI) in football goalkeepers from different age groups; (2) to compare these variables’ behavior between those groups.MethodsThe sample was formed by 19 youth players (15.97 ± 1.55 years old) from a first-division Brazilian football team. Applying appropriate training loads in accordance with the defined objectives promotes optimal physical and physiological adaptations, reduces the likelihood of illness and injury and therefore increases the possibility of success during the Rugby. The aim of this review was to compile and systematize the information in the literature on the association between training load variables (internal and external) and performance outcomes in Rugby. As such, the main objective will be to conduct a systematic review of the published literature to identify the physical and physiological performance variables in Rugby sport to monitor the training load. Following the preferred reporting item for systematic reviews and meta-analyses (PRISMA) and PICOS approach, the search was conducted systematically only in the PubMed database, which, in itself, also restricts the search spectrum of the paper, thus conferring a limitation to the present academic work. The search included specific terms - namely, Rugby; Training Load; - throughout the possible temporal spectrum in PubMed, since there is still little robustness in the literature of the content in question in the sport of Rugby. Articles were selected using pre-defined selection criteria, namely observational studies, randomized clinical studies, and clinical trial studies. The initial electronic search with the variables "Training Load", "Rugby" led to a set of 114 articles, of which only 96 articles were selected after the selection of variables for presenting observational study designs and/or randomized clinical trials. A further screening was carried out, and based on the inclusion criteria of the papers, the result of the analysis of the relevance of the studies was only 19 articles. Three duplicates were eliminated, therefore, the final set of analysis resulted in 16 articles. From the studies compiled in this review, it seems that there is a strong correlation between the perceived exertion (RPE) and the prescription and definition of the training load applied in Rugby athletes. The RPE reflects the most applied and analyzed variable throughout all the studies. Several articles reflect that there is a strong relationship between the training load, and the inter-individual capacity of each athlete and their tolerance to the load (player load).

IntroductionA multivariate training program could be a pedagogical choice to improve physical and tactical performance in a team sport taught during physical education classes at different levels of education. Thus, the aim of this study was to verify the effects of applying a multivariate training program on physical fitness and tactical performance during the teaching of a basketball didactic unit in basic and secondary education.MethodsSeventy-five students from a Portuguese school, with an average age of 15.02 ± 1.31 years, included forty-two students from basic school and thirty-three students from secondary school. The FITescola® test battery was used to assess physical fitness (i.e., sit-ups, push-ups, horizontal impulse, shuttle test, 40 m sprint, agility 4 × 10 m). The Game Performance Assessment Instrument (GPAI) was used to assess students' tactical performance for each player's game performance during a 20-minute 3 vs. 3 match. The GPAI variables were decision making index (DMI), skill execution index (SEI), support actions index (SI), and adaptability index (AI). During a basketball didactic unit teaching, the students were randomly divided into two groups, a control group that will not carry out the training program and an experimental group that will carry out a strength training program, high intensity explosive exercises and activities based on small-sided games (SSG) for 6 weeks. The two groups were evaluated in two moments: before the application of the training program and after the application of the training program regarding changes in physical fitness and tactical performance. Asthma is one of the most prevalent chronic diseases worldwide, with a considerable increase, especially in children. It is considered the main cause of childhood morbidity, school absenteeism, and limitations in sports practice. The causes are multifactorial, and their prevalence varies from region to region, thus verifying a great disparity in the estimates of the prevalence of asthma. In this sense, the objective of this study is to investigate the prevalence of asthma, its control, as well as the frequency of associated symptoms, in adolescents who attended the 3rd cycle of basic education and secondary education in schools in the municipalities of Paços de Ferreira, Paredes, and Penafiel. The sample consisted of 1222 (587 males and 635 females) (p = 0.17) aged between 12 and 17 years. The instruments used to diagnose asthma-associated symptoms were the standard questionnaire of the “International Study of Asthma and Allergies in Childhood—ISAAC” and to check whether asthma was controlled, the “Test for Asthma Control” questionnaire was used. The results reveal a high prevalence of adolescents with asthma (8.9%) with a significant percentage that did not have the disease under control (38%). There was also a considerable percentage of adolescents who, despite not having asthma, have many symptoms associated with the disease. These results may be associated with environmental factors.

Medical ethicists have questioned the use of no-treatment controlled studies (placebo and sham procedure) of new therapies when safe and effective standard therapies are available for use as an active or “equivalence” control. Currently, ethical and conduct principles for biomedical research specifically prohibit projects that do not make or deny the “best-proven diagnosis and therapeutic treatment” to any participant in a clinical trial, including individuals who consent to randomisation into a control group. Studies of psychophysiological therapies are often criticised for not having a placebo or sham treatment control group. In sports science research, particularly in the case of clinical exercise, the use of control groups also raises ethical questions. This article briefly reviews the problem and discusses the ethical standards governing human research derived from the Nuremberg Code and the Declaration of Helsinki.

Changing the date of the FIFA World Cup Qatar 2022 may represent a factor to consider for the expected performance of participating players. This was due to fixture congestion at the start of the season and expected weather conditions during the competition. Thus, the main purpose of this brief report was to critically analyze the potential impact of changing the competition date and weather conditions on players’ performance. In addition, a brief description about the performance during the World Cup is also provided. For the research, the Web of Science, PubMed and SPORTDiscus databases were accessed using the primary keywords FIFA World Cup and World Soccer Cup associated with the secondary keywords match running performance, fixture congestion, fatigue and weather conditions. After applying inclusion and exclusion criteria, 52 articles were considered for analysis. The results seem to indicate that although changes were expected due to the modifications made (i.e., the competition date and scheduling congestion), the performance of the players seems not to have been affected in terms of the analyzed indicators. Furthermore, it seems possible to identify some patterns in the behavior of the teams that reached the most advanced stages of the competition.

Background and Objectives: The effect of the blood flow restriction technique (BFR) on delayed onset muscular soreness (DOMS) symptoms remains unclear. Since there is no consensus in the literature, the aim of the present study is to systematically identify and appraise the available evidence on the effects of the BFR technique on DOMS, in healthy subjects. Materials and Methods: Computerized literature search in the databases Pubmed, Google Scholar, EBSCO, Cochrane and PEDro to identify randomized controlled trials that assessed the effects of blood flow restriction on delayed onset muscular soreness symptoms. Results: Eight trials met the eligibility criteria and were included in this review, presenting the results of 118 participants, with a mean methodological rating of 6/10 on the PEDro scale. Conclusions: So far, there is not enough evidence to confirm or refute the influence of BFR on DOMS, and more studies with a good methodological basis are needed, in larger samples, to establish protocols and parameters of exercise and intervention. Data analysis suggests a tendency toward the proinflammatory effect of BFR during high restrictive pressures combined with eccentric exercises, while postconditioning BFR seems to have a protective effect on DOMS. The aim of this study was two-fold: (1) to analyze the influence of season phase (i.e., the start of the in-season and mid-in-season) on match running performance in a Portuguese professional football team; (2) to determine and model the main factor influencing match running performance during the in-season in this specific football team. Eighteen matches were collected by an 18 Hz global positioning system (GPS) from a professional Portuguese football team during the start of the in-season and mid-in-season. The match running performance was analyzed according to season phases, presenting significant differences in total distance (TD) (tlowerbound = 4.71, p < 0.001; tupperbound = −2.22, p = 0.002), average speed (AvS) (tlowerbound = 359.45, p < 0.001; tupperbound = −359.87, p < 0.001), and relative high speed running (rHSR) (tlowerbound = 13.10, p < 0.001; tupperbound = −10.21, p < 0.001). The logistic regression showed TD (β = −1.59, z = −2.84, p = 0.005) and AvS (β = 2.68, z = −2.84, p = 0.007) as the major factors influencing match running performance during seasonal variation. Sprints and accelerations showed no significance for predicting match running performance during the season phases (β = −0.05 to 1.07, z = −0.95 to 1.07, p = 0.29 to 0.72). Current research confirms that lower and upper bounds should be used to quantify seasonal differences on match running performance. TD and AvS were described as the main factors influencing match running performance during the in-season phase. Thus, it is important to highlight the pace and volume of the game to maximize match running performance.

Physical Education plays a fundamental role in promoting healthy habits and lifestyles, as well as in the development of individual and cognitive skills. To date, several investigations have reported positive effects on indicators of physical fitness, motor proficiency, and creativity as a result of specific training programs during Physical Education classes. However, the effects of multivariate training programs on the improvement of the aforementioned skills remain unclear in the literature. Through this brief review, the benefit of applying multivariate training programs during Physical Education classes on indicators of physical fitness, motor proficiency and creativity was critically analyzed. A narrative approach was applied to summarize the availed research as following: (i) theoretical background; (ii) research gaps/issues; (iii) subject explanation about multivariate training programs in Physical Education; and (iv) practical application and further research. The evidence reported in this regard may be useful for the development of multivariate training programs that simultaneously enable the improvement of indicators of physical fitness, motor proficiency and creativity. However, there is still no consensus in the literature on the best strategies (i.e., type of program, duration, intensity) to enhance motor proficiency and creativity in the context of Physical Education classes using multivariate training programs.

ABSTRACT Invasion of the colon wall by Entamoeba histolytica during amoebic dysentery entails migration of trophozoites through tissue layers that are rich in extracellular matrix. Transcriptional silencing of the E. histolytica surface metalloprotease EhMSP-1 produces hyperadherent less-motile trophozoites that are deficient in forming invadosomes. Reversible protein phosphorylation is often implicated in regulation of cell motility and invadosome formation. To identify such intermediaries of the EhMSP-1-silenced phenotype, here we compared the phosphoproteomes of EhMSP-1-silenced and vector control trophozoites by using quantitative tandem mass spectrometry-based proteomics. Six proteins were found to be differentially phosphorylated in EhMSP-1-silenced and control cells, including EhCoactosin, a member of the ADF/cofilin family of actin-binding proteins, which was more frequently phosphorylated at serine 147. Regulated overexpression of wild-type, phosphomimetic, and nonphosphorylatable EhCoactosin variants was used to test if phosphorylation functions in control of E. histolytica actin dynamics. Each of the overexpressed proteins colocalized with F-actin during E. histolytica phagocytosis. Nonetheless, trophozoites overexpressing an EhCoactosin phosphomimetic mutant formed more and poorly coordinated cell membrane protrusions compared to those in control or cells expressing a nonphosphorylatable mutant, while trophozoites overexpressing nonphosphorylatable EhCoactosin were significantly more motile within a model of mammalian extracellular matrix. Therefore, although EhCoactosin’s actin-binding ability appeared unaffected by phosphorylation, EhCoactosin phosphorylation helps to regulate amoebic motility. These data help to understand the mechanisms underlying altered adherence and motility in EhMSP-1-silenced trophozoites and lay the groundwork for identifying kinases and phosphatases critical for control of amoebic invasiveness. IMPORTANCE Invasive amoebiasis, caused by the intestinal parasite Entamoeba histolytica, causes life-threatening diarrhea and liver abscesses, but, for unknown reasons, only approximately 10% of E. histolytica infections become symptomatic. A key requirement of invasion is the ability of the parasite to migrate through tissue layers. Here, we systematically looked for differences in protein phosphorylation between control parasites and a previously identified hyperadherent E. histolytica cell line that has reduced motility. We identified EhCoactosin, an actin-binding protein not previously known to be phosphoregulated, as one of the differentially phosphorylated proteins in E. histolytica and demonstrated that EhCoactosin phosphorylation functions in control of cell membrane dynamics and amoebic motility. This and the additional differentially phosphorylated proteins reported lay the groundwork for identifying kinases and phosphatases that regulate tissue invasiveness.

Background and objectives: Starting the multicomponent training sessions with aerobic-based exercises or resistance-based exercises may have different effects on functional fitness and body composition. Thus, the aim of this study was to assess the effects of the order of exercises in elderly women’s physical fitness and body composition by multicomponent training. Materials and Methods: A sample of 91 elderly females, aged between 60 and 81, were randomly divided into three groups (A, B, C). Each group performed the following order of exercises: Group A consisted of warm-up followed by aerobic training, strength training, stretching and cool down; Group B consisted of warm-up followed by strength training, aerobic training, stretching and cool down; while the control group (C) did not perform any exercise. Functional fitness and body composition were assessed at 3 moments of the 32 weeks (baseline and after each 16-week) intervention. One-way ANOVA for comparison between groups, ANOVA for repeated measures and multiple linear regression were used for statistical analysis. Results: The results showed that the functional fitness and body composition varied over the 32 weeks of multicomponent training. However, group A seems to show higher improvements in more variables. Conclusion: In the current study, group A obtained better results in most of the evaluated parameters. Thus, to improve functional fitness, warm-up, followed by aerobic training, strength training and relaxation may be the most suitable training for elderly women.

The aim of this study was to analyze the effects of match location, quality of opposition and match outcome on match running performance according to playing position in a Portuguese professional football team. Twenty-three male professional football players were monitored from eighteen Portuguese Football League matches during the 2019–2020 season. Global positioning system technology (GPS) was used to collect time-motion data. The match running performance was obtained from five playing positions: central defenders (CD), fullbacks (FB), central midfielders (CM), wide midfielders (WM) and forwards (FW). Match running performance was analyzed within specific position and contextual factors using one-way analysis of variance (ANOVA) for repeated measures, standardized (Cohen) differences and smallest worthwhile change. CM and WM players covered significantly greater total distance (F = 15.45, p = 0.000, η2 = 0.334) and average speed (F = 12.79, p < 0.001, η2 = 0.294). WM and FB players covered higher distances at high-speed running (F = 16.93, p = 0.000, η2 = 0.355) and sprinting (F = 13.49; p < 0.001, η2 = 0.305). WM players covered the highest number of accelerations (F = 4.69, p < 0.001, η2 = 0.132) and decelerations (F = 12.21, p < 0.001, η2 = 0.284). The match running performance was influenced by match location (d = 0.06–2.04; CI: −0.42–2.31; SWC = 0.01–1.10), quality of opposition (d = 0.13–2.14; CI: –0.02–2.60; SWC = 0.01–1.55) and match outcome (d = 0.01–2.49; CI: −0.01–2.31; SWC = 0.01–0.35). Contextual factors influenced the match running performance with differential effects between playing positions. This study provides the first report about the contextual influence on match running performance in a Portuguese professional football team. Future research should also integrate tactical and technical key indicators when analyzing the match-related contextual influence on match running performance.

Monitoring the training load in football is an important strategy to improve athletic performance and an effective training periodization. The aim of this study was two-fold: (1) to quantify the weekly training load and recovery status variations performed by under-15, under-17 and under-19 sub-elite young football players; and (2) to analyze the influence of age, training day, weekly microcycle, training and playing position on the training load and recovery status. Twenty under-15, twenty under-17 and twenty under-19 players were monitored over a 2-week period during the first month of the 2019–2020 competitive season. Global positioning system technology (GPS) was used to collect external training loads: total distance covered, average speed, maximal running speed, relative high-speed running distance, high metabolic load distance, sprinting distance, dynamic stress load, accelerations and decelerations. Internal training load was monitored using ratings of perceived exertion (RPE) and session rating of perceived exertion (sRPE). Recovery status was obtained using the total quality recovery (TQR) scale. The results show an age-related influence for external training load (p ≤ 0.001; d = 0.29–0.86; moderate to strong effect), internal training load (p ≤ 0.001, d = 0.12–0.69; minimum to strong effect) and recovery status (p ≤ 0.001, d = 0.59; strong effect). The external training load presented differences between training days (p < 0.05, d = 0.26–0.95; moderate to strong effect). The playing position had a minimum effect on the weekly training load (p < 0.05; d = 0.06–0.18). The weekly microcycle had a moderate effect in the TD (p < 0.05, d = 0.39), RPE (p < 0.05; d = 0.35) and sRPE (p < 0.05, d = 0.35). Interaction effects were found between the four factors analyzed for deceleration (F = 2.819, p = 0.017) and between inter-day, inter-week and age for total covered distance (F = 8.342, p = 0.008). This study provided specific insights about sub-elite youth football training load and recovery status to monitor training environments and load variations. Future research should include a longer monitoring period to assess training load and recovery variations across different season phases.

Here, the authors provide a panel of medium-throughput assays to test potential drug candidates against different life cycle stages of Cryptosporidium with the goal to support a drug development pipeline that contains compounds with diverse molecular mechanisms of action.

The apicomplexan parasite Cryptosporidium is a leading cause of childhood diarrhea in developing countries. Current treatment options are inadequate and multiple preclinical compounds are being actively pursued as potential drugs for cryptosporidiosis. Unlike most apicomplexans, Cryptosporidium spp. sequentially replicate asexually and then sexually within a single host to complete their lifecycles. Anti-cryptosporidial compounds are generally identified or tested through in vitro phenotypic assays that only assess the asexual stages. Therefore, compounds that specifically target the sexual stages remain unexplored. In this study, we leveraged the ReFRAME drug repurposing library against a newly devised multi-readout imaging assay to identify small-molecule compounds that modulate macrogamont differentiation and maturation. RNA-seq studies confirmed selective modulation of macrogamont differentiation for 10 identified compounds (9 inhibitors and 1 accelerator). The collective transcriptomic profiles of these compounds indicates that translational repression accompanies Cryptosporidium sexual differentiation, which we validated experimentally. Additionally, cross comparison of the RNA-seq data with promoter sequence analysis for stage-specific genes converged on a key role for an Apetala 2 (AP2) transcription factor (cgd2_3490) in differentiation into macrogamonts. Finally, drug annotation for the ReFRAME hits indicates that an elevated supply of energy equivalence in the host cell is critical for macrogamont formation.
