IntroductionLung cancer is the most commonly diagnosed and the main cause of cancer death, usually related to cigarette smoking. Furthermore, the microbiota of people exposed to cigarette smoke can be modified, making it difficult to eliminate opportunistic microorganisms. The leaves of Eugenia pyriformis are a by-product of fruit production and, to date, there have been no studies addressing the antiproliferative, anti-inflammatory, and antimicrobial activities.ObjectiveInvestigate the antimicrobial, Nitric Oxide (NO)-production inhibition, and antiproliferative activities of the essential oil from E. pyriformis leaves and its possible effect on the treatment and prevention of damage caused by tobacco.MethodsThe essential oil (EO) was obtained by hydrodistillation (3 h). Its chemical composition was investigated by GC-MS. It was proposed to investigate antiproliferative activity against human tumor cell lines, namely, breast adenocarcinoma (MCF-7), lung (NCI-H460), cervical (HeLa), and hepatocellular (HepG2) carcinomas. A non-tumor primary culture from pig liver (PLP2) was also tested. The EO capacity to inhibit nitric oxide (NO) production was evaluated by a lipopolysaccharide stimulated murine macrophage cell line. Antibacterial and antifungal activities against opportunistic pathogens were investigated against seven strains of bacteria and eight fungi.ResultsThe results indicated the presence of 23 compounds in the essential oil, the majority were spathulenol (45.63%) and β-caryophyllene oxide (12.72%). Leaf EO provided 50% inhibition of nitric oxide production at a concentration of 92.04 µg mL−1. The EO also demonstrated antiproliferative activity against all human tumor cell lines studied, with GI50 values comprised between 270.86 and 337.25 µg mL−1. The essential oil showed antimicrobial potential against the bacteria Listeria monocytogenes (Murray et al.) Pirie (NCTC 7973) and Salmonella Typhimurium ATCC 13311 (MIC 1870 µg mL−1) and fungi Aspergillus versicolor ATCC 11730, Aspergillus ochraceus ATCC 12066, Penicillium ochrochloron ATCC 90288, Penicillium verrucosum var. cyclopium (Westling) Samson, Stolk & Hadlok (food isolate) (MIC 1870 µg mL−1) and Trichoderma viride Pers. IAM 5061 (1,400 µg mL–1).ConclusionThe demonstrated anti-inflammatory, antiproliferative, and antimicrobial activities in the leaves of E. pyriformis can add value to the production chain of this plant, being a possible option for preventing and combating cancer, including lung cancer.

The chemical composition of extracts (CEs) and essential oils (EOs) from Tetradenia riparia leaves, flower buds, and stems was analyzed. Antiproliferative activity against tumor cell lines, NO production inhibition, and antioxidant and antiviral activities were assessed. The CEs contained flavonoids, phenolic acids, coumarins, and saturated fatty acids. The EOs included monoterpenes, oxygenated sesquiterpenes, and diterpenes. NO production inhibition ranged from 76 to 247 µg mL−1, and antiproliferative activity exhibited GI50 between 20 and >204 µg mL−1, with low cytotoxicity (SI: 1.08 to 4.75). Reactive oxygen species inhibition ranged from 45 to 82%. Antioxidant activity varied when determined by the 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay (IC50: 0.51 to 8.47 mg mL−1) and ferric reducing antioxidant power (0.35 to 0.81 µM ferrous sulfate per mg). The reduction in β-carotene–linoleic acid co-oxidation varied between 76.13 and 102.25%. The total phenolic content of CEs and EOs was 10.70 to 111.68 µg gallic acid mg−1. Antiviral activity against herpes simplex virus type 1 (HSV-1) showed an EC50 between 9.64 and 24.55 µg mL−1 and an SI between 8.67 and 15.04. Leaf EOs exhibited an EC50 of 9.64 µg mL−1 and an SI of 15.04. Our study unveils the diverse chemical composition and multifaceted pharmacological properties of T. riparia, demonstrating its potential as a valuable source of bioactive compounds for therapeutic applications.

ABSTRACT Many industrially synthesized human - therapeutic agents, agrochemicals, and additives used by industries are heterocyclic compounds. Many of these contribute to increased environmental contamination in localized and diffuse sources of water bodies, reflecting soil quality, communities, and human health. This concern led us to develop this research to evaluate the presence of organic compounds in the surface waters of the Pirapó river basin, Paraná, Brazil, influenced by the soybean and safflower corn crops that are cultivated in adjacent areas around the river. The water samples were collected from October 2017 to January 2019, at three collection points. Organic compounds were analyzed by GC-MS, after the solid phase extraction (SPE). Two organic compounds were analyzed: azetidine and sarcosine that are used in the pharmaceutical industry. Organic compounds existing in drugs and/or contaminants that were observed in this study reveal the importance of further investigation into their origins and the consequences for the health of the biota and the population.

Tetradenia riparia (Hochst.) Codd (Lamiaceae) is a shrub, commonly known as ginger bush or false myrrh, and several studies have shown that T. riparia exhibits a variety of biological properties. This study aimed to determine the chemical composition of T. riparia essential oil and its fractions, investigate their anticholinesterase activity, and assess their larvicidal activity against the cattle tick Rhipicephalus microplus and the mosquito Aedes aegypti. Eleven essential oil fractions were obtained by fractionation and analyzed by gas chromatography/mass spectrometry. Larvicidal activity against R. microplus and third-instar A. aegypti was assessed using a larval packet test and a larval immersion test, respectively. Anticholinesterase activity was determined by a bioautographic method. Forty-nine compounds were identified in the essential oil, of which the major classes were oxygenated sesquiterpenes (45.95%) and sesquiterpene hydrocarbons (35.20%) and the major components were isospathulenol (17.40%), β-caryophyllene (15.61%), 14-hydroxy-9-epi-caryophyllene (10.07%), 14-hydroxy-α-muurolene (8.32%), and 9β,13β-epoxy-7-abietene (5.53%). Bioassays showed that T. riparia essential oil (LC50 = 1.56 µg/mL) and FR3 (LC50 = 0.30 µg/mL) were the most active against R. microplus and A. aegypti larvae, respectively. The essential oil and FR1, FR2, and FR3 exhibited acetylcholinesterase inhibitory activity. These results indicate that T. riparia essential oil and its fractions hold promise in the development of novel, environmentally safe agents for the control of R. microplus and A. aegypti larvae.

Abstract Around the world, the main problems of livestock are caused by ectoparasites, however, commercial acaracide are toxic to the environment and detrimental to One Health. Therefore, research has increasingly focused on development of natural products as alternatives for tick control. The purpose of this study was to evaluate the larvicidal effect on Rhipicephalus (Boophilus) microplus, through use of essential oils (EOs) extracted from the leaves, flower buds and stems of Tetradenia riparia. The chemical composition of these EOs was determined through gas chromatography coupled to mass spectrometry (GC-MS). They were tested on larvae at concentrations of 100.000 to 40 µg/mL, using the larval packet test and under semi-natural conditions. The main class of compounds in the chemical composition was sesquiterpenes (both oxygenates and hydrocarbons), whereas the predominant compounds in the leaves, flower buds and stems were 14-hydroxy-9-epi-caryophyllene, T-cadinol and 6-7-dehydroroyleanone, respectively. The leaves proved to be the most effective, with highest larvicidal activity (LC99.9 = 83.53 µg/mL). When tested under semi-natural conditions, the oils obtained efficiency above 98% in all compound tests. The results indicated that these EOs were effective against R. (B.) microplus larvae in vitro and ex-situ, proving that this plant has bioactive molecules with significant larvicidal activity.

Abstract Tetradenia riparia (Hochst.) Codd (Lamiaceae) is a species native to the African continent and used as an insect repellent. The objective of the study was to evaluate the larvicidal potential of essential oils (EOs) from the leaves, flower buds, and stem of T. riparia, collected in winter against Aedes aegypti larvae. The EOs were extracted by hydrodistillation (3 h) and identified by GC/MS. The EOs were tested against larvae of A. aegypti at concentrations ranging from 12500 to 1.5 µg/mL for 24 h. The insecticide activity was evaluated by probit analysis, and the anticholinesterase activity was determined by bioautographic method. The results of the class projection indicated sesquiterpenes as the majority class, corresponding to 60.66% (leaves), 64.70% (flower buds) and 83.99% (stem), and the bioassays on A. aegypti larvae indicated LC50 of 1590, 675 and 665 µg/mL, respectively. The anticholinesterase activity indicated that the EO of the leaves inhibited the enzyme at a concentration of 780 µg/mL, and those from the flower buds and stem inhibited up to 1560 µg/mL. The results indicated weak activity of essential oils against A. aegypti larvae.

Com o desenvolvimento urbano e social das cidades, os moradores que residem nas proximidades de riachos e vales passam a sofrer as consequências do lançamento contínuo de esgoto. O objetivo deste trabalho foi avaliar a qualidade da água do Ribeirão Floriano, localizado no município de Maringá-PR. Para tanto, quatro pontos, denotados por P1, P2, P3 e P4, foram escolhidos. Para a análise de DQO todas as coletas realizadas apresentaram a mesma concentração, sendo inferior a 15 mg L-1, porém, em relação ao DBO5 no período do verão os pontos P2, P3 e P4 mostraram-se dentro do limite esperado, 2.0 mg L-1, para águas superficiais de Classe II, de acordo com a legislação. Em relação a análise da contagem de Escherichia coli os maiores valores encontrados, para a contaminação biológica do ribeirão, foram nos pontos P2 e P3, equivalentes a 2000 UFC/mL e 424 UFC/mL, respectivamente, no período da primavera. Estes dois pontos estão localizados próximos a lavanderias industriais que podem estar associados a lançamento de efluentes advindos destas empresas. Nas análises, foi determinado concentrações elevadas de cobre (Cu), alumínio (Al) e níquel (Ni), evidenciando uma distribuição equitativa da ação antrópica em todo o curso do ribeirão. Estes resultados sugerem a necessidade da implantação de programas de monitoramento ambiental em águas superficiais nos rios do município.

Gallesia integrifolia, a notable species in the Atlantic Forest, has been traditionally employed in folk medicine for treating rheumatism, asthma, and worms. This study investigated the cellular antioxidant, antiproliferative, and anti-inflammatory activities of the essential oils (EOs) and crude extracts (CEs) from G. integrifolia flowers, fruits, and leaves. The chemical identification of EOs was performed by GC–MS and CEs by UHPLC–MS. Cellular antioxidant and anti-inflammatory activities were assessed through mouse macrophage cell culture. In addition, the antiproliferative potential was evaluated in gastric, colorectal, breast, and lung tumor cell lines and non-tumor VERO cells. EOs predominantly contained organosulfur compounds in flowers (96.29%), fruits (94.94%), and leaves (90.72%). We found the main compound is 2,2′-Disulfanediyldiethanethiol in the EOs of flowers (47.00%), leaves (41.82%), and fruits (44.39%). Phenolic compounds were identified in CEs. The EOs and CEs demonstrated potential against the tumor cell lines tested (GI50 between 51 and 230 µg/mL). The selectivity index values were greater than 1.0 (1.01 to 3.37), suggesting a relative safety profile. Moreover, the anti-inflammatory activity IC50 ranged from 36.00 to 268 µg/mL, and the cellular oxidation inhibition ranged from 69% to 82%. The results suggest that oils and extracts derived from G. integrifolia have potential for use in various industrial sectors.

Iron is a fundamental microelement for human life; however, deficiencies or excesses of these metal ions can cause severe complications and mortality. Chelators are compounds that bind and inhibit iron. Ultraviolet-visible (UV-vis) spectrophotometric methods are key analytical tools in the identification of chemical entities, with the benefits of having good precision and accuracy, and the equipment being easily available as well as quick and simple to implement. In this study, we aimed to provide an alternative, cheaper method for the quantification of iron ion chelation by substituting ferrozine for gallic acid and validating its use with UV-vis according to official ANVISA and ICH guidelines. The parameters assessed were specificity, linearity, precision, accuracy, robustness, and finally, the percentage of iron ions chelating was calculated. The results demonstrated that this method was accurate, simple, specific, selective, precise, and reproducible, and was successfully validated for the determination of iron ions chelating. The percentage of iron ions chelating, promoted by the standard chelator EDTA, was 45% and 47% for Fe2+ and Fe3+, respectively. It is concluded that this new method is beneficial in terms of its simplicity, rapidness, low cost, and the fact that it produces very low levels of dangerous residues.

Resumo A ocorrência de eventos extremos de precipitação (EEP) no litoral do Paraná eleva o potencial de erosividade dos rios da região, além de causar inundações. Neste contexto, o objetivo deste estudo é analisar a tendência da intensidade e frequência dos EEP diários, além de analisar o ciclo horário dos extremos para a região costeira do Paraná e o ambiente sinótico. Coletaram-se os dados de precipitação e divergência do fluxo de umidade. Consideraram-se EEP diários acima do percentil 95 (EEP95). A análise de tendência linear através do teste Mann-Kendall, análise do ciclo diário e a distribuição espacial do ambiente sinótico foram estudados. Os resultados mostram que os EEP95 não apresentaram tendência significativa. Apesar do estudo limitar-se a uma única localidade, áreas de máxima convergência de umidade destacam o padrão da Zona de Convergência de umidade. Os resultados mostram que o limiar para um EEP95 diário é de 46,40 mm.dia−1; apenas durante a madrugada e na estação chuvosa, o ciclo diário dos EEP95 apresentou taxas horárias que atingiram o percentil 95. O período de janeiro a março, concentrou cerca de 50% EEP95, com a frequência de EEP95 diminuindo em anos de La Niña e aumentando em anos de El Niño e neutros.

ABSTRACT: Brazil has one of the largest commercial cattle herds in the world, which naturally coexist with an enormous number of parasitic species. Southern cattle tick, Rhipicephalus (Boophilus) microplus, is among these species, interfering with animal productivity and causing losses to the beef and dairy cattle sector. The use of chemical acaricides in the control of this mite has resulted in the emergence of resistant populations. In this sense, alternative control measures using plants as sources of botanical acaricides have shown to be effective. Eugenia pyriformis Cambess is a Brazilian plant with antioxidant and antimicrobial activity; however, there are no reports on its acaricidal activity in the literature. The present study aimed to evaluate the acaricidal and larvicidal potential of E. pyriformis leaf essential oil (EO) on southern cattle tick at different stages of the reproductive cycle. E. pyriformis leaves were collected and dried, and had their EO extracted by hydrodistillation (3h) using a modified Clevenger apparatus. Chemical analysis was performed by gas chromatography coupled with mass spectrometry (GC/MS), and 32 compounds belonging to the sesquiterpene class were identified: hydrocarbons (17.98%) and oxygenated forms (81.96%), with spathulenol (43.65%) and caryophyllene oxide (12.17%) as the most common. The EO was evaluated by the Adult Immersion Test at the concentrations (500.00 to 3.12mg/mL) in which the following parameters were measured: mortality of females (%), hatchability of eggs (%), and product efficiency (%). Larvae were assessed by the Larval Packet Test at concentrations ranging from 25.00 to 0.00004mg/mL. Lethal concentrations (LC) required for killing 50 and 99.9% of adult females and larvae were determined using Probit analysis. LC50 and LC99.9 of EO were 0.06 and 24.60mg/mL and 1,208.80 and 2,538mg/mL for larvae and adult females, respectively. Action of the EO in the free-living cycle of R. (B.) microplus larvae was another parameter assessed. To this end, the larvae were deposited in pots containing Brachiaria decumbens and, after migration to the leaf apex, a solution containing LC99.9 (24.60mg/mL) of the EO was sprayed. After 24h, 72.25% of the larvae had died, indicating stability of the EO when subjected to uncontrolled temperature and humidity conditions. The mechanism of action through which the EO killed the larvae and adult females was investigated by the Bioautographic Method, which showed inhibition of 3.15mg/mL of the EO on the acetylcholinesterase (AChE) enzyme. The results found in the present experiment indicate that E. pyriformis essential oil is an alternative in the control of southern cattle tick in the larval (parasitic) and free-living cycle (non-parasitic) stages under field conditions.

Plant tissue culture has emerged as an important tool to produce bioactive compounds from various plant species, including the sustainable production of limonoids that are receiving considerable attention due to the benefits associated with human health such as anticancer activities. The purpose of the present study was to evaluate the capacity of limonoids aglycone production from callus culture from sweet orange cv. Pera (Citrus sinensis) seeds and identify the compounds produced in this cell line. Callus induction occurred in Murashige and Skoog medium supplemented with 2,4-dichlorophenoxyacetic (2,4-D), malt extract, agar and coconut water. For the analysis and identification of the limonoids, CG-MS-EI ion-positive mode and UPLC-QTOF-ESI were used operating in positive and negative mode. An intense peak corresponding to limonin appeared in the callus extracts at a retention time of 58.1 min. in CG-MS-EI and four major limonoids aglycone by positive ion mode UPLC-QTOF-ESI: limonin, nomilin, deacetylnomilin, and nomilinic acid. The culture medium was efficient at the bioproduction of limonoids aglycone in callus cultures of C. sinensis seeds. Therefore, data obtained from UPLC-QTOF-ESI proved its importance at identifying new compounds that benefit human health, and may assist future work in the identification of known or new limonoids in Citrus species and related genera.

Abstract Purpose To analyze, in aged obese patients, the weight loss, comorbidity control, and safety postoperative complications of bariatric surgery by Roux-en-Y gastric bypass technique. Methods Twenty-seven patients who underwent laparoscopic weight-reducing gastroplasty with Roux-en-Y gastric bypass to treat obesity were included. All patients were ≥ 60 years old at the time of surgery. The Wilcoxon test was used for statistical analysis, and a p-value ≤0.05it was considered significant. Results Ten (90.9%) patients with dyslipidemia were cured (p < 0.001). Nine (81.8%) patients with type 2 diabetes mellitus had total improvement and 2 (18.2%) had partial improvement (p = 0.003). In 23 patients with systemic arterial hypertension, 9 (39.1%) achieved total improvement and 14 (60.9%) partial improvement (p = 0.140). PURPOSE:Investigation of the antibacterial action of aqueous extracts of Bidens sulphurea, Bidens pilosa, and Tanacetum vulgare, species of Asteraceae family that are popularly used for the treatment of genito-urinary infection. METHODS:The minimum inhibitory concentration (MIC) and minimal bacterial concentration (MBC) of the extracts against standard strains of Staphylococcus aureus (ATCC25923), Enterococcus faecalis (ATCC29212), Escherichia coli (ATCC25922), and Pseudomonas aeruginosa (ATCC27853) and against bacteria that were isolated from cultures of vaginal secretions and urine from menopausal women with a diagnosis of recurrent urinary tract infections (rUTI) were determined by broth microdilution. RESULTS:The MIC values of the three extracts against Gram-positive and Gram-negative standard bacterial strains ranged from 7.81 to 125.00 mg ml-1, and the MBC values ranged from 7.81 to 500.00 mg ml-1. However, B. sulphurea was more efficient. In the urine samples, the three extracts inhibited the growth of coagulase-negative Staphylococcus spp., and the B. pilosa was the most active extract against E. coli compared with the other ones. For the vaginal secretion samples, no significant differences in the inhibition of coagulase-positive Staphylococcus spp. and P. mirabilis were found among the extracts. T. vulgare and B. sulphurea were more effective in inhibiting coagulase-negative Staphylococcus spp. compared with B. pilosa. E. coli was more susceptible to the B. sulphurea extract compared with the B. pilosa and T. vulgare extracts. CONCLUSION:The present results suggested the potential medicinal use of Asteraceae species, especially B. sulphurea, as therapeutic agents against rUTI-related bacteria.

This work describes the sol-gel mixed oxide SiO 2 /TiO 2 property, ST, as prepared, and submitted to heat treatment a 773 K, STC. SEM and EDS images show, within magnification used, a uniform distribution of the TiO 2 particles in SiO 2 /TiO 2 matrix. Both, ST and STC adsorb hydrogen peroxide on the surface and through EPR and UV-Vis diffuse reflectance spectra, it was possible to conclude that the species on the surface is the peroxide molecule attached to the Lewis acid site of titanium particle surface, α Ti(H 2 O 2 ) + . As the material is very porous, presumably the hydrogen peroxide molecule is confined in the matrix pores on the surface, a reason why the adsorbed species presents an exceptional long lived stability.

Anisakid nematode helminths have great importance in public health, with several reports of human infections caused by larvae of these parasites in several countries of the world. These parasites are common in freshwater and marine fish, with man as an accidental host. Contamination occurs when humans feed on raw fish in the form of sushi, sashimi, and ceviche, which are infected by the larval stages of these nematodes. In Brazilian rivers there are records of these larvae in Astyanax altiparanae (lambaris) and Geophagus brasiliensis (pearl cichlid), which are species widely distributed in the Neotropical region, especially in Brazil. These diseases are little-known by Brazilian health professionals because their frequency is still low, mainly due to the population’s low fish consumption and the difficulties medical professionals have in diagnosing them. The present study aimed to evaluate the larvicidal action of ginger essential oil (Zingiber officinale) on larvae of Contracaecum sp. removed from the visceral cavity of pearl cichlid and lambaris collected from the lake at Ingá Park, Maringá, PR, in the year 2016. Two hundred and thirty-four fish specimens were necropsied, and their larvae were tested to verify the larvicidal effect of ginger. Of these fish, 123 were parasitized, equivalent to 52.5%, of which 181 larvae were collected, all from the coelomic cavity. Ginger essential oil proved to be a promising larvicide, promoting larval death in vitro. This larvicidal action was observed at different times and concentrations, showing the efficacy of ginger oil in combating this zoonosis. It is suggested that new natural products be tested for this purpose, as the number of people who consume raw fish in Brazil continues to increase.

Este trabalho tem por objetivo estudar um evento de tombamento de torre de suporte de Linha de Transmissão de Energia Elétrica (LT) ocorrido no dia 12 de julho de 2016 em Cascavel – PR devido a ocorrência de uma microexplosão. Para o estudo foram utilizados dados de anemômetros sônicos instalados em quatro níveis de altura (10m, 22m, 33m e 44m) da torre de transmissão número 37 da LT Toledo-Cascavel na região oeste do Paraná. Para análise espacial das rajadas e das assinaturas de mesoescala foram utilizados dados de velocidade radial e refletividade do radar meteorológico banda S Doppler de dupla polarização do Simepar instalado em Cascavel. As análises da assinatura de mesoescala principalmente da velocidade radial do radar Doppler indicaram a presença de uma microexplosão a qual gerou rajadas próximas de 45 ms-1. O registro no anemômetro na torre situada a 300m de distância registrou uma rajada de 32,6 m.s-1. A microexplosão foi gerada por uma supercélula que se formou no Paraguai e se propagou zonalmente até o oeste do Paraná durante 5 horas com velocidade de propagação de 18,5 ms-1. The pet industry is currently expanding and specializing mainly in the field of domestic felines. Problems related to antimicrobial resistance are frequent, and the use of essential oils (EOs) in animal feed has become a novel treatment strategy. Thus, the objective of this study was to assess the bacteriostatic activity of Brazilian lemon balm (Lippia alba), lemon grass (Cymbopogon citratus), and oregano (Origanum vulgare) in bacterial isolates from 12 samples of cat food sold in bulk. The EOs from fresh leaves of crops were obtained from the Medicinal Garden of Paranaense University, Umuarama, Paraná. Cat food samples were processed for identification of gram-positive and gram-negative microorganisms. The determination of the bacteriostatic activity of the EOs was performed by determination of the minimum inhibitory concentration (MIC) at dilutions of 2.5, 1.25, and 0.62 mg/mL. The diffusion disc technique was used to evaluate the resistance profile to the main antimicrobials used in the feline clinic and to analyze the effect of the association of these antimicrobials with the EOs studied. A total of 23 isolates were obtained, of which 16 were gram-negative and seven were gram-positive. As for the oil composition for L. alba, C. citratus, and O. vulgare, 40, 24, and 44 compounds were identified, respectively, with the major ones being geranial, geranial/?-citral, and carvacrol, respectively. Regarding MIC, no differences were found for any EOs tested. The lowest MIC value was obtained for the C. citratus EO (0.83 mg/mL) for two bacteria (coagulase-negative Staphylococcus and Corynebacterium kutscheri). The means of the inhibition halos for the 10 antimicrobials tested in association or not with one of the EOs for Klebsiella aerogenes, Proteus vulgaris, and Serratia rubidaea showed that, for S. rubidaea, the inhibition halo diameter (12.4 mm) was greater (p < 0.05) when amoxicillin was associated with the O. vulgare EO than the association of the same antibiotic with the C. citratus EO (11.0 mm). For K. aerogenes and P. vulgaris, there was no difference in inhibition halo diameter when EOs were included. In conclusion, L. alba, C. citratus, and O. vulgare EOs are effective in inhibiting the growth of gram-positive and gram-negative bacteria and can be added to cat food to replace chemical antimicrobials.

Several technologies have been developed to control Aedes aegypti, mainly studies on isolated plant molecules. The Schinus terebinthifolius (Raddi) (Anacardiaceae), popularly known as pink pepper is a plant widely used in reforestation of degraded areas and its fruits are used as condiments. The objective of this work was to investigate the potential of essential oils (EOs) and fractions (FRs) obtained from fresh fruits and leaves of S. terebinthifolius. The EOs were obtained by hydrodistillation (2 hours), fractionated on a chromatographic column using as the stationary phase silica gel 60 (0.063-0.2mm), mobile phases: n-hexane, dichloromethane, ethyl acetate and methanol and chemically evaluated by gas chromatography coupled to mass spectrometer (GC/MS). EOs and FRs were tested against larvae of the third stage and pupae of Ae. aegypti by Immersion Test at concentrations ranging from 500.00 to 0.003 mg mL-1 (v/v). The hexane FRs obtained from fruits and leaves were the ones that showed the greatest activity on the larvae (LC99.9= 0.60 mg mL-1 and LC99.9 0.64 mg mL-1, respectively) and pupae (LC99,9 = 2.51 mg mL-1 and 2.61 mg mL-1, respectively). These results were confirmed by the anticholinesterase activity where the hexane (fruit and leaf) FRs presented the highest inhibitory potential on the acetylcholinesterase enzyme (0.156 mg mL-1 and 0.312 mg mL-1, respectively), suggesting the likely mechanism of action. The larvicidal potential can be explained by the presence of the major compounds bicyclogermacrene and germacrene D in the hexane FRs, indicating in this way that they may replace or even act in synergisms with conventional chemical larvicides. In this way the present study opens the field for new researches, aiming the development of products with the compounds bicyclogermacrene and germacrene D, as an alternative in the control of this culicide.

In this study the potential bioinseticide of the essential oil (OE) extracted from the rhizomes of the species Curcuma zedoaria (Zingiberaceae) was evaluated. The rhizomes were collected during dormancy (winter) and budding (summer). The EO was obtained by hydrodistillation (2h) and identified by GC/MS. In addition, a multivariate exploratory analysis was done to determine the analysis of the major compounds (PCA). The EO yield in dormancy was 0.61± 0.07 (%) and in budding 0.55 ± 0.08 (%). The bioassays on Aedes aegypti larvae and pupae were done by immersion test at different EO concentrations which ranged from 500.00 to 0.003 mg mL-1 (v/v). The results on the larvae and pupae indicated LC99.9 of (0.01 and 1.38 mg mL-1) for EO in dormancy, and (0.08 and 2.63 mg mL-1) for EO during budding, respectively. The action mechanism of EOs in both periods was determined by autobiographic method evaluating the inhibitory potential on the acetylcholinesterase enzyme, indicating greater inhibition of the EO enzyme during dormancy (0.039 mg mL-1) when compared to the EO during budding (0.156 mg mL-1). The projection representation of the EO chemical classes in both evaluated periods indicated that oxygenated sesquiterpenes are the major compound class (46.99% in dormancy) and (43.59% in budding). The projection of major chemical compounds of EOs presented three compounds with greater mass flow distancing: epicurzerenone (18.20% and 12.10%); 1.8 cineole (15.76% and 12.10%) and β-elemene (4.43 and 0.01%) that are found in greater amounts in the dormancy EO when compared to budding, respectively. These results corroborate with the greater potential on Ae. aegypti larvae and pupae found for the dormancy EO. The results are promising because they show in which vegetative cycle phase C. zedoaria EO presents greater bioinsecticide potential.

IntroductionVancomycin is widely used to treat infections caused by Gram positive bacteria, mostly methicillin-resistant strains. Despite its therapeutic effectiveness, vancomycin is a nephrotoxic drug that has been associated with the occurrence of acute kidney injury (AKI). In this study, we sought to evaluate the variability of serum trough concentrations of vancomycin and to determine the incidence and risk factors of vancomycin-associated nephrotoxicity (VAN) in non-critically ill patients.MethodsThis was a prospective cohort including Brazilian public hospital inpatients from April 2017 to June 2018. The participants received intravenous vancomycin therapy for at least 48 hours for any suspected or confirmed infection by Gram positive bacteria. Demographic, clinical and laboratory data were collected. Information on vancomycin therapy and concomitant use of other nephrotoxic drugs were also recorded. Patients were followed up until discontinuation of vancomycin treatment or death, whatever occurred first. The primary outcome was the occurrence of AKI. We performed a Poisson regression to determine risk factors for AKI.ResultsOverall, 98 participants were included in the study. Median age was 55.9 (interquartile range [IQR] 40.6-66.8) years and 58 (59.2%) were men. Most of them showed subtherapeutic (20mg/L) trough levels of vancomycin; 42.9% and 15.3%, respectively. A total of 19 (19.4%) patients developed AKI. Poisson regression showed that male sex (odds ratio [OR] 2.90; confidence interval [CI] 95% 1.28-6.53; p = 0.011), concomitant use of piperacillin-tazobactam (OR 4.66; CI 95% 2.26-9.58; p ConclusionsOur study showed that usual doses of vancomycin did not reach recommended therapeutic serum trough levels of vancomycin in non-critically ill patients. Besides that, nephrotoxicity was common in this population, being associated with male sex, concomitant use of piperacillin-tazobactam and supra-therapeutic trough serum levels of vancomycin.

Baccharis dracunculifolia DC (Asteraceae) is a Brazilian native bush tree, and its leaf essential oil has been reported to possess some biological activities, but the antimicrobial activity of its aerial part essential oil at the flowering period is unknown or little studied, mainly against agents that cause foodborne diseases. Thus, this study aimed to determine the chemical composition and evaluate the antimicrobial activity of the essential oil of B. dracunculifolia aerial part at flowering period. This essential oil was obtained by hydro distillation and its chemical composition was determined by gas chromatography coupled with mass spectrometry (GC–MS). The minimum inhibitory concentration, minimum bactericidal concentration, and minimum fungicidal concentration of the essential oil were evaluated against eight bacteria and eight fungi using 96-well microtiter plates. The essential oil yield was 1.8 ± 0.07%, and spathulenol (27%) and trans-nerolidol (23%), both oxygenated sesquiterpenes, were the major compounds found among 30 chemical constituents identified. The essential oil presented bacteriostatic and bactericidal activities, mainly against Staphylococcus aureus, Bacillus cereus and Pseudomonas aeruginosa, and also fungistatic and fungicidal activities. However, its antibacterial activity was more effective than the antifungal one by using the essential oil at lower concentrations. Essential oil of B. dracunculifolia may be a potential alternative for food applications in order to reduce synthetic chemicals in a more sustainable food industry.

Objective: The aim of this report is to describe a case of a patient with synchronous gastric tumors and demonstrate the importance of identifying this coexistence before surgery, considering that synchronicity is generally detected only after the surgery. Methods: The medical records of patients from Servidor Público Estadual Hospital of São Paulo were analyzed. Results: We identified a case of a 69-year-old male patient who presented with symptoms of dyspepsia, weight loss, and asthenia. The patient underwent serum laboratory tests, abdominal computed tomography, and upper digestive endoscopy. Analysis of the results revealed that the symptomatology was related to the association of two different gastric lesions, gastric adenocarcinoma and a nearby gastrointestinal stromal tumor (GIST), although without collision. Conclusion: The patient was submitted to a total gastrectomy with a Y-en-Roux reconstruction. Importance of the problem: Gastric cancer is the major cause of mortality of neoplastic nature, considering its high frequency and presentation of few symptoms. Although gastric adenocarcinoma and GIST are extremely different in their histopathological aspects, they can occur synchronously in the stomach. Preoperative identification of the synchronicity of these tumors is significantly important to improve surgical planning. Comments: Currently, there is a lack of a specific examination for screening GISTs, which explains the difficulty in diagnosing this tumor before the surgical procedure. In the present case, the diagnosis of adenocarcinoma was made despite the fact that two distinct gastric lesions were detected in the radiological imaging examination, and the final diagnosis of GIST was made only through anatomopathological analysis.

O aproveitamento de resíduos da indústria na fabricação de tijolos de solo-cimento é uma alternativa ambientalmente sustentável e possibilita a valorização destes resíduos, em vez de simplesmente lançá-los na natureza. Neste trabalho, analisou-se a influência da incorporação de cinza de bagaço de cana, pó de serra e PET nas características físicas e mecânicas de tijolos de solo-cimento. Os resultados obtidos mostraram que a incorporação dos resíduos (cinza de bagaço de cana e pó de serra) ao tijolo de solo-cimento manteve as propriedades mecânicas gerais, possibilitando a produção de tijolos prensados mais eficiente na relação custo-benefício, constituindo-se, portanto, numa excelente alternativa para o aproveitamento destes materiais.

BackgroundThe impact on quality of life attributed to treatment for rectal cancer remains high. Deterioration of the urinary function is a relevant complication within that context.ObjectiveTo detect the presence of urinary dysfunction and its risk factors among individuals underwent surgical treatment for rectal cancer.MethodsThe present prospective study analyzed 42 patients from both genders underwent surgical treatment for rectal adenocarcinoma with curative intent. The version of the International Prostatic Symptom Score (IPSS) questionnaire validated for the Portuguese language was applied at two time-points: immediately before and 6 months after surgery. CONTEXT: Living donor liver transplantation has become an alternative to reduce the lack of organ donation. OBJECTIVE: To identify factors predictive of early graft loss in the first 3 months after living donor liver transplantation. METHODS: Seventy-eight adults submitted to living donor liver transplantation were divided into group I with 62 (79.5%) patients with graft survival longer than 3 months, and group II with 16 (20.5%) patients who died and/or showed graft failure within 3 months after liver transplantation. The variables analyzed were gender, age, etiology of liver disease, Child-Pugh classification, model of end-stage liver disease (MELD score), pretransplantation serum sodium level, and graft weight-to-recipient body weight (GRBW) ratio. The GRBW ratio was categorized into < 0.8 and MELD score into >18. The chi-square test, Student t-test and uni- and multivariate analysis were used for the evaluation of risk factors for early graft loss. RESULTS: MELD score 135 mEq/L (P = 0.03) were higher in group II than in group I. In the multivariate analysis MELD scores > 18 (P18 and GRBW < 0.8 ratios are associated with higher probability of graft failure after living donor liver transplantation.

PURPOSE: To study the functional behavior of the allograft with immunosuppression of pancreatic islets in the spleen. METHODS: Five groups of 10 Mongrel dogs were used: Group A (control) underwent biochemical tests; Group B underwent total pancreatectomy; Group C underwent total pancreatectomy and pancreatic islet autotransplant in the spleen; Group D underwent pancreatic islet allograft in the spleen without immunosuppressive therapy; Group E underwent pancreatic islet allograft in the spleen and immunosuppression with cyclosporine. All of the animals with grafts received pancreatic islets prepared by the mechanical-enzymatic method - stationary collagenase digestion and purification with dextran discontinuous density gradient, implanted in the spleen. RESULTS: The animals with autotransplant and those with allografts with immunosuppression that became normoglycemic showed altered results of intravenous tolerance glucose (p < 0.001) and peripheral and splenic vein plasmatic insulin levels were significantly lower (p < 0.001) in animals that had allografts with immunosuppression than in those with just autotransplants. CONCLUSIONS: In the animals with immunosupression with cyclosporine subjected to allograft of pancreatic islets prepared with the mechanical-enzymatic preparation method (stationary collagenase digestion and purification with dextran discontinuous density gradient), the production of insulin is decreased and the response to intravenous glucose is altered.OBJETIVO: Avaliar o comportamento funcional do alotransplante com imunossupressão de ilhotas pancreáticas no baço. O controle de qualidade realizado nas farmácias de manipulação e homeopáticas é de suma importância para assegurar a qualidade microbiológica e físico-química das matériasprimas e produtos acabados, garantindo eficácia, segurança e credibilidade dos medicamentos dispensados à população. O presente trabalho teve como objetivos realizar uma pesquisa sobre o número de farmácias de manipulação e homeopáticas da cidade de Maringá, Estado do Paraná,que estão realizando o controle de qualidade de acordo com o estabelecido pela Vigilância Sanitária na Resolução de Diretoria Colegiada (RDC33); diagnosticar quais os testes mínimos realizados; observar a adequação das técnicas empregadas e quais as maiores dificuldades encontradas pelas farmácias para implantar o controle de qualidade. Com base nas diretrizes da RDC33 foi formulado um questionário, o qual foi utilizado como suporte para a realização dapesquisa direta em 25 farmácias de manipulação e homeopáticas, que aceitaram participar do estudo, de um total de 30 farmácias da cidade de Maringá. A partir das análises realizadas, concluiu-se que a maioria das farmácias de manipulação e homeopáticas da cidade de Maringá está se adequando às exigências estabelecidas pela Anvisa. Porém, encontram dificuldades como recursos financeiros e humanos para realizarem o controle de qualidade mínimo necessário paraa manutenção da eficácia, estabilidade e segurança dos medicamentos manipulados e matériasprimas utilizadas.The quality control in compounding and homeopathic pharmacies is of highest importance to assure the microbiological and physicalchemicalquality of drugs and compounding preparations, guaranteeing effectiveness, safety and credibility of the released medicines to the population. The objective of the present work was to verify the number of compounding and homeopathic pharmacies of the city ofMaringá, Paraná State, which are doing the quality control in agreement with the determinations of the Sanitary Surveillance in RDC33. It was observed which minimum tests, were realized and if they were adequate as well the largest difficulties found by the drugstoresto implant the quality control. Using as guideline the RDC33 a questionnaire was formulated, which was used as support for the accomplishment of the direct research in 25 compounding and homeopathic pharmacies from a total of 30 drugstores of the city of Maringá. Based analyses of the results it was concluded that most of the compounding and homeopathic pharmacies of the city of Maringá are trying to adaptate to the established demands of ANVISA. However they have difficulties such as financial and human resources to do theminimum quality control necessary for the maintenance of the effectiveness, stability and safety of the manipulated medicines and raw materials used.

Essential oils from fresh Piperaceae leaves were obtained by hydrodistillation and analyzed by gas chromatography mass spectrometry (GC–MS), and a total of 68 components were identified. Principal components analysis results showed a chemical variability between species, with sesquiterpene compounds predominating in the majority of species analyzed. The composition of the essential oil of Piper mosenii was described for the first time. The cytotoxicity of the essential oils was evaluated in peritoneal macrophages and the oils of P. rivinoides, P. arboretum, and P. aduncum exhibited the highest values, with cytotoxic concentration at 50% (CC50) > 200 µg/mL. Both P. diospyrifolium and P. aduncum displayed activity against Leishmania amazonensis, and were more selective for the parasite than for the macrophages, with a selectivity index (SI) of 2.35 and >5.52, respectively. These SI values were greater than the 1 for the standard drug pentamidine. The antileishmanial activity of the essential oils of P. diospyrifolium and P. aduncum was described for the first time. P. rivinoides, P. cernuum, and P. diospyrifolium displayed moderate activity against the Mycobacterium tuberculosis H37Rv bacillus, with a minimum inhibitory concentration (MIC) of 125 µg/mL. These results are relevant and suggests their potential for therapeutic purposes. Nevertheless, further studies are required to explain the exact mechanism of action of these essential oils.

O receio de graves complicações pós-operatórias tem inibido muitas das indicações cirúrgicas da litíase biliar no paciente idoso sintomático. A finalidade principal deste trabalho foi averiguar a extensão real desse problema no Serviço de Gastroenterologia Cirúrgica do HSPE-FMO. Foram estudados 185 idosos portadores de litíase biliar com idade média de 73,0 ± 6,2 anos, no período de seis anos (1990-1995). Os pacientes foram divididos em dois grupos de acordo com a idade: I - com 118 (63,8%) pacientes de 60 a 74 anos e o II - com 67 (36,2%) pacientes entre 75 e 90 anos. OBJETIVO: Comparar os níveis séricos de CA19-9 e CEA e a expressão tecidual do CA19-9 e relacioná-los com os aspectos morfológicos do carcinoma colorretal. MÉTODOS: Quarenta e cinco pacientes com carcinoma colorretal foram operados com coleta de CEA e CA19-9 séricos pré-operatórios. Valores séricos de CEA = 5,0ng/mL e de CA19-9 = 37UI/mL foram considerados aumentados. A avaliação da imunoexpressão do CA19-9 no tecido neoplásico foi realizada por meio de estudo imunoistoquímico com anticorpo monoclonal anti-CA19-9. A intensidade de expressão do CA19-9 no tecido neoplásico foi semiquantificada em leve(+/+++), moderada(++/+++), intensa(+++/+++) e ausente. RESULTADOS: Os valores do CA19-9 sérico foram progressivamente maiores conforme o aumento da expressão do CA19-9 no tecido neoplásico, porém sem significância (p=0,06). OBJETIVO: O propósito deste estudo foi determinar a probabilidade de ocorrência de coledocolitíase através do estudo da associação de indicadores clínicos e laboratoriais desta doença em dois momentos do pré-operatório de colecistectomia. MÉTODO: Entre março de 2001 e março de 2002, 48 pacientes consecutivos com colelitíase foram submetidos a colecistectomia e colangiografia intra-operatória (CIO). Os pacientes foram divididos em dois grupos, sendo o grupo A constituído por 13 pacientes com coledocolitíase e o grupo B por 35 pacientes sem esta doença. Os pacientes foram investigados quanto aos indicadores clínicos e laboratoriais da coledocolitíase, analisados em dois períodos, tomando como ponto de corte as 48 horas que precederam a cirurgia. Posteriormente, estes indicadores pré-operatórios foram associados na equação da regressão logística em diferentes combinações. RESULTADOS: Utilizando a equação da regressão logística, constatou-se que a associação de dois indicadores clínicos em ambos os períodos (icterícia e sinal de Murphy) e dois laboratoriais ( nível de corte da gama glutamil transpeptidase e bilirrubina direta 48 horas antes da cirurgia) foi a mais adequada para a predição da coledocolitíase. Os valores obtidos por esta equação mostraram concordância com os grupos A e B, de 95,6%, e discordância de 4,4% (p= 0,0000007 e k = 0,89). Esta equação mostrou sensibilidade de 92,3%, especificidade de 97,0%, valor preditivo positivo de 92,3% e valor preditivo negativo de 97%. Estes valores foram próximos aos obtidos pela CIO, que mostrou concordância com os grupos estudados de 95,8%, e discordância de 4,2% (k = 0,90). CONCLUSÃO: Considerando os resultados obtidos, recomenda-se a associação de indicadores da coledocolitíase na equação da regressão logística para estabelecer a probabilidade de ocorrer coledocolitíase associada à colelitíase. A utilização desta equação pode orientar melhor a conduta diagnóstica e terapêutica nesta doença.

ABSTRACT An asymptomatic 79-year-old woman, with incidental finding on abdominal ultrasound of a solid nodule in the tail of the pancreas. Magnetic resonance imaging showed a 12mm solid tumor. The suggested diagnosis was pancreatic neuroendocrine tumor. The pathological examination showed an intrapancreatic splenic tissue. This is a rare ectopic location of spleen tissue and it should be considered in the differential diagnosis of pancreatic solid tumors.

ABSTRACT Epiphrenic diverticulum is a rare disease associated with esophageal motor disorders that is usually asymptomatic and has a well-established surgical indication. We report a case of giant epiphrenic diverticulum in a 59-year-old symptomatic woman who was diagnosed after underwent complementary exams. Because of her symptoms, the surgical treatment was chosen, and esophageal diverticulectomy was performed along with laparoscopic cardiomyotomy and anterior partial fundoplication.
