Background: the purpose of this study was to analyze the reliability of a field-based fitness test battery in young trained football players, according to biological age. Methods: 197 young trained football players (12–19 years old) participated in the study. We measured anthropometric measurements (i.e., height, sitting height, length leg, and body mass), a bilateral vertical jumping test (CMJ), a progressive loading test of squats and hip thrust, acceleration and speed tests (10 m and 30 m sprint tests), a change-of-direction ability test (V–cut test), and a cardiorespiratory fitness test (30–15 intermittent fitness test). Statistical data are shown as the mean ± standard deviation by PHV group in tests and retests. Test reliability was assessed through the intraclass correlation coefficient (ICC), with an ICC above 0.9 being considered high. To evaluate accuracy and repeatability, standard error of measurement, coefficient of variation, and minimum detectable change at 90% were determined and Bland–Altman diagrams were used, establishing a statistical significance of p < 0.05. Results: All of the tests showed non-significant differences between the test and retest in the pooled sample (p > 0.05). Furthermore, all of them presented a trivial effect size (0.9), which indicates the high reproducibility of the tests, despite some of them presenting a significant difference between trials (i.e., the CMJ, 10 m sprint, V–cut, and squat tests). Low measurement errors were found for all tests (coefficient of variation [CV] = 4.39–9.39), except for the CMJ and the progressive loading test for squat and hip thrust exercises (CV = 12.2–21.11). Similar results were found irrespective of biological age group. Conclusions: All tests were reliable for the pooled sample as well as for the biological age groups.

Recent improvements in smartphone video technology may provide sufficient accuracy for estimation of jump height via flight time determined from video recordings of vertical-jump tests. The aim of this study is to evaluate the accuracy of jump height estimated from videos at different frame rates. Highdefinition videos of 10 young adults (6 males, 4 females) performing 5 countermovement jumps were recorded at a frame rate of 1000 Hz and transcoded to frame rates of 120, 240, and 480 Hz. Flight time in the videos was assessed independently by three observers at each of the four frame rates with MyJump. Flight time and jump height were analyzed with mixed models for estimation of means and of standard deviations representing technical error of measurement (free of within-subject jump-to-jump variability) at each frame rate. The four frame rates and three observers produced practically identical estimates of mean jump height. The technical errors at 120, 240, 480 and 1000 Hz were respectively 3.4, 1.8, 1.2 and 0.8 ms for flight time, and 1.4%, 0.7%, 0.5% and 0.3% for jump height. Assessed relative to either differences in jump height between elite football players (standard deviation of ~12%) or the smallest expected test-retest variability (typical error of ~3%), the technical error was substantial at 120 Hz but negligible at 240 Hz or higher. In conclusion, use of frame rates above 240 Hz to estimate jump height with MyJump will not improve accuracy substantially.

CanAssist Breast (CAB), a prognostic test uses immunohistochemistry (IHC) approach coupled with artificial intelligence-based machine learning algorithm for prognosis of early-stage hormone-receptor positive, HER2/neu negative breast cancer patients. It was developed and validated in an Indian cohort. Here we report the first blinded validation of CAB in a multi-country European patient cohort. FFPE tumor samples from 864 patients were obtained from-Spain, Italy, Austria, and Germany. IHC was performed on these samples, followed by recurrence risk score prediction. The outcomes were obtained from medical records. The performance of CAB was analyzed by hazard ratios (HR) and Kaplan Meier curves. CAB stratified European cohort (n = 864) into distinct low- and high-risk groups for recurrence (P 50 years (HR: 2.93 (1.44–5.96), P = 0.0002). This work gives an overview of the state-of-the-art in modelling of short-cut processes for nitrogen removal in mainstream wastewater treatment and presents future perspectives for directing research efforts in line with the needs of practice. The modelling status for deammonification (i.e., anammox-based) and nitrite-shunt processes is presented with its challenges and limitations. The importance of mathematical models for considering N2O emissions in the design and operation of short-cut nitrogen removal processes is considered as well. Modelling goals and potential benefits are presented and the needs for new and more advanced approaches are identified. Overall, this contribution presents how existing and future mathematical models can accelerate successful full-scale mainstream short-cut nitrogen removal applications. HIGHLIGHTS Models for mainstream short-cut N removal processes are reviewed by considering their current practice and limitations.; Modelling goals and potential benefits are presented from a modeller perspective to facilitate successful applications.; More advanced modelling approaches are presented to overcome the addressed challenges and limitations.;

Immunotherapy has transformed advanced non‐small cell lung cancer (NSCLC) treatment strategies and has led to unprecedented long‐lasting responses in some patients. However, the molecular determinants driving these long‐term responses remain elusive. To address this issue, we performed an integrative analysis of genomic and transcriptomic features of long‐term immune checkpoint inhibitors (ICIs)‐associated responders. We assembled a cohort of 47 patients with NSCLC receiving ICIs that was enriched in long‐term responders [>18 months of progression‐free survival (PFS)]. We performed whole‐exome sequencing from tumor samples, estimated the tumor mutational burden (TMB), and inferred the somatic copy number alterations (SCNAs). We also obtained gene transcription data for a subset of patients using Nanostring, which we used to assess the tumor immune infiltration status and PD‐L1 expression. Our results indicate that there is an association between TMB and benefit to ICIs, which is driven by those patients with long‐term response. Additionally, high SCNAs burden is associated with poor response and negatively correlates with the presence of several immune cell types (B cells, natural killers, regulatory T cells or effector CD8 T cells). Also, CD274 (PD‐L1) expression is increased in patients with benefit, mainly in those with long‐term response. In our cohort, combined assessment of TMB and SCNAs burden enabled identification of long‐term responders (considering PFS and overall survival). Notably, the association between TMB, SCNAs burden, and PD‐L1 expression with the outcomes of ICIs treatment was validated in two public datasets of ICI‐treated patients with NSCLC. Thus, our data indicate that TMB is associated with long‐term benefit following ICIs treatment in NSCLC and that TMB, SCNAs burden, and PD‐L1 are complementary determinants of response to ICIs.

Background Immune checkpoint inhibitors (ICIs) targeting the programmed cell death 1/programmed death-ligand 1 axis have transformed the management of advanced non-small cell lung cancer (NSCLC). However, many patients do not benefit from this type of treatment, and thus several molecular biomarkers of benefit have been explored. The value of somatic copy number alterations (SCNAs) burden remains elusive.Patients and methods We assembled a cohort of 109 patients with NSCLC treated with ICIs and available tumor samples. We performed shallow whole-genome sequencing on 89 patients to determine genome-wide SCNAs and targeted gene expression analysis on 63 patients to study immune infiltration. We analyzed SCNAs burden in different ways (ie, the fraction of the genome altered or number of events) and studied their association with ICIs benefit based on survival analysis. We correlated SCNAs burden and immune infiltration on 35 patients of our cohort and on patients with lung adenocarcinoma from The Cancer Genome Atlas (TCGA).Results High SCNAs burden, computed in diverse ways, is negatively associated with ICIs progression-free survival (PFS), with the fraction of the genome altered (FGA) by arm and chromosome events showing the strongest association with PFS (p=0.002) (n=77). Nevertheless, we found differences in SCNAs across some clinicopathological features (sample site origin). A multivariate analysis adjusted for relevant characteristics showed that the FGA of arm and chromosome alterations was strongly associated with PFS (HR=2.21, p=3.3 x 10−5). Finally, we confirmed that SCNAs burden negatively correlates with tumor immune infiltration (n=35), although this correlation was not found for the males studied. Similar results were observed in the TCGA cohort.Conclusions SCNAs burden is a potential biomarker of benefit to ICIs in patients with NSCLC, although there appear to be some nuances worth consideration. Further studies will be needed to establish its role as a biomarker of benefit to ICIs.

Objective: Ki67 is a prognostic and predictive marker in breast cancer (BC). However, manual scoring (MS) by visual assessment suffers from high inter-observer variability which limits its clinical use. Here, we developed a new digital image analysis (DIA) workflow, named KiQuant for automated scoring of Ki67 and investigated its equivalence with standard pathologist's assessment. Methods: Sequential immunohistochemistry of Ki67 and cytokeratin, for precise tumor cell recognition, were performed in the same section of 5 tissue microarrays containing 329 tumor cores from different breast cancer subtypes. Slides were digitalized and subjected to DIA and MS for Ki67 assessment. The intraclass correlation coefficient (ICC) and Bland-Altman plot were used to evaluate inter-observer reproducibility. The Kaplan-Meier analysis was used to determine the prognostic potential. Results: KiQuant showed an excellent correlation with MS (ICC:0.905,95%CI:0.878–0.926) with satisfactory inter-run (ICC:0.917,95%CI:0.884–0.942) and inter-antibody reproducibilities (ICC:0.886,95%CI:0.820–0.929). The distance between KiQuant and MS increased with the magnitude of Ki67 measurement and positively correlated with analyzed tumor area and breast cancer subtype. Agreement rates between KiQuant and MS within the clinically relevant 14% and 30% cut-off points ranged from 33% to 44% with modest interobserver reproducibility below the 20% cut-off (0.606, 95%CI:0.467–0.727). High Ki67 by KiQuant correlated with worse outcome in all BC and in the luminal subtype (P = 0.028 and P = 0.043, respectively). For MS, the association with survival was significant only in 1 out of 3 observers. Conclusions: KiQuant represents an easy and accurate methodology for Ki67 measurement providing a step toward utilizing Ki67 in the clinical setting.

Abstract Background Earth Observation ‘EO’ remote sensing technology development enables original insights into vegetation function and health at ever finer temporal, spectral and spatial resolution. Research sites equipped with monitoring infrastructure such as flux towers operate at a key bridging scale between satellite platform measurements and on-the-ground leaf-level processes. Results This paper presents the technical details of the design and operation of a proximal observation system ‘THEMS’ that generates unattended long-term high quality thermal and hyperspectral images of a forest canopy on a short (sub-daily) timescale. The primary purpose of the system is to measure canopy temperature, spectral reflectance and radiance coincident with a highly instrumented flux tower site for benchmarking purposes. Basic system capability is demonstrated through low level data product descriptions of the high-resolution multi-angular imagery and ancillary data streams. The system has been successfully operational for more than 2 years with little to no intervention. Conclusions These data can then be used to derive remotely sensed proxies of canopy and ecosystem function to study temporal forest dynamics over a wide range of wavelengths, spatial scales (individual trees to canopy), and temporal scales (minutes to multiple years). The multi-purpose system is intended to provide unprecedented spatio-temporal ecophysiological insight and to underpin upscaling of remotely sensed dynamic ecosystem water, CO2, and energy exchange processes.

Vertical jump performance is a commonly used test to measure lower-limb muscle power that is carried out with several types of equipment. The aim of this study was to validate an open-source jump mat (Chronojump Boscosystems) against a proprietary jump mat (Globus Ergo Tester). Sixty-three active sportsmen (age 23.3 ± 2.4 years) completed 8 maximal-effort countermovement jumps (CMJ). The heights of the 504 CMJ were measured from the two jump mats simultaneously. Reliability was examined with intra-class correlation coefficients (ICC), paired samples t-tests, coefficient of variation (CV) and Cronbach’s α. Bivariate Pearson’s correlation coefficient (r) was used to examine validity. Effects were evaluated using non-clinical magnitudebased inference. BackgroundType 1 diabetes (T1D) affects more than 165,000 individuals younger than 20 years in the United States of America. The transition from parent management to parent-child team management, with the child taking on increased levels of self-care, can be stressful and is associated with a deterioration in self-management behaviors. Therefore, a mobile app intervention, MyT1DHero, was designed to facilitate diabetes-specific positive parent-adolescent communication and improve diabetes-related outcomes. The MyT1DHero intervention links an adolescent with T1D and their parent through 2 separate app interfaces and is designed to promote positive communication regarding T1D management. ObjectiveThe aim of this pilot study was to determine (1) the initial efficacy of the MyT1DHero intervention in improving diabetes outcomes in adolescents, specifically the hemoglobin A1c (HbA1c) levels, diabetes care adherence, and quality of life, and (2) the adolescents’ overall satisfaction with this intervention. MethodsThis pilot study included 30 adolescent-parent pairs who used the MyT1DHero app in a 12-week single-arm clinical trial. Participants were recruited from the local pediatric endocrinology subspecialty clinic via snowball sampling. HbA1c levels, diabetes care adherence, quality of life, family conflict, and satisfaction levels were measured and analyzed using paired sample two-sided t tests and linear regression analyses. ResultsThe final analysis included 25 families. The mean age of the adolescents was 12.28 (SD 1.62) years. Half of the participants (13/25) reported a diabetes diagnosis of less than 5 years. After 12 weeks of the intervention, diabetes care adherence significantly improved (before the study: mean 3.87 [SD 0.59]; after the study: mean 4.19 [SD 0.65]; t21=–2.52, P=.02, d=0.52) as did quality of life (before the study: mean 4.02 [SD 0.84]; after the study: mean 4.27 [SD 0.73]; t24=2.48, P=.01, d=0.32). HbA1c levels (before the study: mean 8.94 [SD 1.46]; after the study: mean 8.87 [SD 1.29]; t24=0.67, P=.51, d=0.04) and family conflict (before the study: mean 2.45 [SD 0.55]; after the study: mean 2.61 [SD 0.45]; t23=0.55, P=.14, d=0.32) changed in the hypothesized direction, but the change was not significant. Study Design Retrospective, descriptive case series study. Purpose To investigate the frequency, bone pattern, and associated lesions to the spine of gunshot wounds. Overview of Literature Gunshot wounds are penetrating in nature and are caused by a bullet or projectile. These are becoming more common and are associated with significant sequelae, requiring long and costly multidisciplinary treatment. Associated spinal cord injuries (SCIs) in the civilian population represent 13%–17% of all spinal traumas. Spinal gunshot wounds are commonly thought to be stable; however, there is potential acute and chronic spinal instability if the bullet passes transversely, fracturing either both pedicles or both facet joints. Methods We obtained data from the clinical files of patients with spinal gunshot wounds treated by spine surgeons. We performed a statistical analysis to obtain the pattern and frequency of the injuries. Results We included 54 patients (48 men [89%] and six women [11%]). Eight patterns of spinal gunshot wounds were identified; 61% (33 patients) had complete SCI. The thoracic spine was most frequently affected, with 88% of patients having SCI; 75%, hemopneumothorax; 28%, pulmonary lesions; and 16%, hepatic lesions. The presence of pedicle fractures had an odds ratio of 3.64 for SCI. Conclusions The two main bone patterns are the vertebral body and posterior arch with pedicle fractures as modifiers, given that they are at a high risk for SCI. The spinal pattern and associated lesions were related to the bullet’s path, with the vertebral body having the highest frequency of associated lesions. Vertebral body burst fracture was the main indication for instrumentation.

This study deals with Transformation-Induced Plasticity (TRIP) observed in the martensitic transformation of 35NCD16 ferritic steel. In this study, TRIP tests were carried out for two different cases: First, after only free dilatometric (FD) tests, which is used as the reference test for the considered applied stress; second, with TRIP tests being performed similarly to the first case (same thermal cycle, same applied stress) but with pre-thermomechanical loading histories applied. Such histories may be FD tests, TRIP tests, elastoplastic history, etc. The comparison between the results of TRIP test (a) and TRIP test (b) indicates if TRIP holds the memory of the applied loading histories. The current obtained results tell us that TRIP does not hold any significant memory. During the martensite → austenite transformation, the material may present recovery from strain hardening. Waiting for more details about the physical phenomena responsible for the absence of TRIP memory, one can point out the importance of this result as it enables one to use the same specimen for several TRIP tests. However, this result must be validated using other combinations of loading histories (such as multiaxial and cyclic, among others).

Inaccurate estimates of animal populations may lead to flawed management interventions, therefore, it is essential to understand the status and population trend of a species in order to plan its management efficiently. Aerial surveys are considered a useful method for estimating the population size of large conspicuous animals inhabiting large areas, but raw count data from aerial surveys usually underestimate population sizes due to imperfect detection. The use of N-mixture models with aerial count data provides a useful tool to estimate the population sizes while taking detection probability into account. As a study case we used aerial surveys conducted for monitoring black rhinoceros (Diceros bicornis) in Madikwe Game Reserve and Pilanesberg Nature Reserve (South Africa) during 1999–2015, and we analysed data with a dynamic extension of the N-mixture model. We estimated 0.078–0.098 and 0.139–0.142 individuals/km2, respectively, and we found evidence for density dependence in both reserves with a carrying capacity of 0.122 (0.102–0.142) individuals/100 km2. Based on simulations used to assess precision of the estimates, root-mean-square error model (RMSE) estimates was significantly smaller than those for the raw maximum counts.The N-mixture models provide a promising approach to estimate population size, trends and demographic characteristics of large conspicuous mammals such as black rhinoceroses. Such analysis can provide estimates that are more accurate than raw counts. In addition, use of model covariates that affect a species' population parameters can provide useful information for their conservation and management. In this article, the parameter identification of a brushless DC motor (BLDC) is presented. The approach here presented is based on a direct identification considering a three-phase line-to-line voltage electromagnetic torque as function of the electric currents and rotor speed. The estimation is divided into two stages. First, the electrical parameters are estimated by well-known no-load and DC tests. Consequently, estimation of mechanical parameters is performed using a recursive Least Square Algorithm. The proposed approach is validated by comparing model responses to motor real time responses. Additionally, the design, digital simulation and real time implementation of a PI rotor speed controller, based on the estimated model, validate the identification proposal presented here.

In this work we study the initial-value problem for the fifth-order Korteweg-de Vries equation $$ \partial_{t}u+\partial_{x}^{5}u+u^k\partial_{x}u=0, \quad x,t\in \mathbb{R}, \; k=1,2, $$ in weighted Sobolev spaces $H^s(\mathbb{R})\cap L^2(\langle x \rangle^{2r}dx)$. We prove local and global results. For the case $k=2$ we point out the relationship between decay and regularity of solutions of the initial-value problem.

The achievements made in genomic technology in recent decades are yet to be matched by fast and accurate crop phenotyping methods. Such crop phenotyping methods are required for crop improvement efforts to meet expected demand for food and fibre in the future. This review evaluates the role of proximal remote sensing buggies for field-based phenotyping with a particular focus on the application of currently available sensor technology for large-scale field phenotyping. To illustrate the potential for the development of high throughput phenotyping techniques, a case study is presented with sample data sets obtained from a ground-based proximal remote sensing buggy mounted with the following sensors: LiDAR, RGB camera, thermal infra-red camera and imaging spectroradiometer. The development of such techniques for routine deployment in commercial-scale breeding and pre-breeding operations will require a multidisciplinary approach to leverage the recent technological advances realised in computer science, image analysis, proximal remote sensing and robotics.

Plant breeding trials are extensive (100s to 1000s of plots) and are difficult and expensive to monitor by conventional means, especially where measurements are time-sensitive. For example, in a land-based measure of canopy temperature (hand-held infrared thermometer at two to 10 plots per minute), the atmospheric conditions may change greatly during the time of measurement. Such sensors measure small spot samples (2 to 50 cm2), whereas image-based methods allow the sampling of entire plots (2 to 30 m2). A higher aerial position allows the rapid measurement of large numbers of plots if the altitude is low (10 to 40 m) and the flight control is sufficiently precise to collect high-resolution images. This paper outlines the implementation of a customized robotic helicopter (gas-powered, 1.78-m rotor diameter) with autonomous flight control and software to plan flights over experiments that were 0.5 to 3 ha in area and, then, to extract, straighten and characterize multiple experimental field plots from images taken by three cameras. With a capacity to carry 1.5 kg for 30 min or 1.1 kg for 60 min, the system successfully completed >150 flights for a total duration of 40 h. Example applications presented here are estimations of the variation in: ground cover in sorghum (early season); canopy temperature in sugarcane (mid-season); and three-dimensional measures of crop lodging in wheat (late season). Together with this hardware platform, improved software to automate the production of ortho-mosaics and digital elevation models and to extract plot data would further benefit the development of high-throughput field-based phenotyping systems.

Vicilins (7S globulins) are seed storage proteins and constitute the main protein family in legume seeds, particularly in narrow-leafed lupin (Lupinus angustifolius L.; NLL), where seven vicilin genes, called β1- to β7-conglutin have been identified. Vicilins are involved in germination processes supplying amino acids for seedling growth and plant development, as well as in some cases roles in plant defense and protection against pathogens.The roles of NLL -conglutins in plant defense are unknown. Here the potential role of five NLL -conglutin family members in protection against necrotrophic fungal pathogens was investigated and it was demonstrated that recombinant purified 6xHis-tagged β1- and 6-conglutin proteins exhibited the strongest in vitro growth inhibitory activity against a range of necrotrophic fungal pathogens compared to β2, β3 and β4 conglutins. To examine activity in vivo, two representative necrotrophic pathogens, the fungus Sclerotinia sclerotiorum and oomycete Phytophthora nicotianae were used. Transient expression of β1- and β6-conglutin proteins in Nicotiana benthamiana leaves demonstrated in vivo growth suppression of both of these pathogens, resulting in low percentages of hyphal growth and elongation in comparison to control treated leaves. Cellular studies using β1- and β6-GFP fusion proteins showed these conglutins localized to the cell surface including plasmodesmata. Analysis of cellular death following S. sclerotiorum or P. nicotianae revealed both β1- and β6-conglutins suppressed pathogen induced cell death in planta and prevented pathogen induced suppression of the plant oxidative burst as determined by protein oxidation in infected compared to mock-inoculated leaves.

Three different Land Surface Models have been used in three high resolution climate simulations performed with the mesoscale model MM5 over the Iberian Peninsula. The main difference among them lies in the soil moisture treatment, which is dynamically modelled by only two of them (Noah andPleim & Xiu models), while in the simplest model ( Simple Five-Layers ) it is fixed to climatological values. The simulated period covers 1958-2002, using the ERA40 reanalysis data as driving conditions. Focusing on near-surface air temperature, this work evaluates the skill of each simulation in reproducing mean values and temporal variability, by comparing the simulations with observed temperature series. When the simplest simulation was analyzed, the greatest discrepances were observed for the summer season, when both, the mean values and the temporal variability of the temperature series, were badly underestimated. These weaknesses are largely overcome in the other two simulations (performed by coupling a more advanced soil model to MM5), and there was greater concordance between the simulated and observed spatial patterns. The influence of a dynamic soil moisture parameterization and, therefore, a more realistic simulation of the latent and sensible heat fluxes between the land and the atmosphere, helps to explain these results.

Peritoneal dialysis (PD) is complicated by peritonitis episodes that cause loss of mesothelium and eventually sclerosing peritonitis. An improved understanding of the molecular contributors to peritoneal injury and defense may increase the therapeutic armamentarium to optimize peritoneal defenses while minimizing peritoneal injury. There is no information on the expression and function of the cytokine TWEAK and its receptor Fn14 during peritoneal injury. Fn14 expression and soluble TWEAK levels were measured in human PD peritoneal effluent cells or fluids with or without peritonitis. Fn14 expression was also analyzed in peritoneal biopsies from PD patients. Actions of intraperitoneal TWEAK were studied in mice in vivo. sTWEAK levels were increased in peritoneal effluent in PD peritonitis. Effluent sTWEAK levels correlated with the number of peritoneal macrophages (r=0.491, p=0.002). Potential TWEAK targets that express the receptor Fn14 include mesothelial cells and macrophages, as demonstrated by flow cytometry of peritoneal effluents and by analysis of peritoneal biopsies. Peritoneal biopsy Fn14 correlated with mesothelial injury, fibrosis and inflammation, suggesting a potential deleterious effect of TWEAK/Fn14. In this regard, intraperitoneal TWEAK administration to mice promoted peritoneal inflammation characterized by increased peritoneal effluent MCP-1, Fn14 and Gr1+ macrophages, increased mesothelial Fn14, MCP-1 and CCL21 expression and submesothelial tissue macrophage recruitment. Taken together these data suggest that the TWEAK/Fn14 system may promote inflammation and tissue injury during peritonitis and PD.

Previous studies have shown that the A-IV-347Ser polymorphism is associated with the variability in low density lipoprotein (LDL)-cholesterol response to dietary therapy. The present study was designed to evaluate the association of this polymorphism with the individual variability observed in postprandial lipemic response. This polymorphism was characterized in 50 healthy male subjects homozygous for the apolipoprotein (apo)E3 allele. All subjects were subjected to a vitamin A-fat load test. Blood was drawn at time 0 and every hour over a period of 11 hours. Cholesterol and triglycerides (TG) in plasma and lipoprotein fractions of CH, TG, and retinyl palmitate (RP) were determined. Data from the postprandial lipemia revealed that subjects with the A-IV-347Ser allele (n = 14) have a lower postprandial response in total TG (P < 0.025), large triglyceride rich lipoproteins (TRL) TG (P < 0.02), and small-TRL TG levels (P < 0.007), and a higher postprandial response in large-TRL apoA-IV (P < 0.006) and apoB-100 (P < 0.041) levels than subjects homozygous for the A-IV-347Thr subjects (n = 36). In conclusion, the modifications observed in postprandial lipoprotein metabolism associated with this polymorphism within the apoA-IV gene locus may be involved in the variability in LDL-CH response observed in subjects consuming high saturated fat diets.— Ostos, M. Conjugation is the main mode of horizontal gene transfer that spreads antibiotic resistance among bacteria. Strategies for inhibiting conjugation may be useful for preserving the effectiveness of antibiotics and preventing the emergence of bacterial strains with multiple resistances. Filamentous bacteriophages were first observed to inhibit conjugation several decades ago. Here we investigate the mechanism of inhibition and find that the primary effect on conjugation is occlusion of the conjugative pilus by phage particles. This interaction is mediated primarily by phage coat protein g3p, and exogenous addition of the soluble fragment of g3p inhibited conjugation at low nanomolar concentrations. Our data are quantitatively consistent with a simple model in which association between the pili and phage particles or g3p prevents transmission of an F plasmid encoding tetracycline resistance. We also observe a decrease in the donor ability of infected cells, which is quantitatively consistent with a reduction in pili elaboration. Since many antibiotic-resistance factors confer susceptibility to phage infection through expression of conjugative pili (the receptor for filamentous phage), these results suggest that phage may be a source of soluble proteins that slow the spread of antibiotic resistance genes.
