Respiratory function is modulated by circulating steroid hormones. In female rats, steroid hormones fluctuate across the normal estrous cycle and decline with age, similar to human menopause. To determine the influence of steroid hormones, and mimic aspects of age-related reductions in hormones, the ovariectomy model is often employed. Ovariectomy (OVX) induces an immediate and persistent decrease in steroid hormones. The current study aimed to interrogate whether the OVX model of hormone reduction impacted specific aspects of respiratory function [chemosensitivity and the hypoxic ventilatory response (HVR)] in a manner consistent with natural age-related declines in hormones. Using barometric plethysmography, three experimental groups of female rats were assessed for HVR, chemosensitivity, and respiratory neural drive during progressive hypoxic challenges (FIO2: 0.15, 0.12, and 0.09): young (3–5 mos. old; in proestrus; n = 10), young OVX (3–5 mos. old; n = 10), and aged (>20 mos. old; n = 10). Our findings indicted that sex hormone loss did not appear to impact chemosensitivity or neural drive. Natural aging, but not OVX, resulted in decreased HVR as well as reduced magnitude in ventilatory output during stepwise hypoxia. Differences in metabolism were important to the interpretation of these results. Collectively, these data support the concept that aging impacts female respiratory function in complex and unique ways that differ from OVX.

Alterations in the function of K+ channels such as the voltage- and Ca2+-activated K+ channel of large conductance (BKCa) reportedly promote breast cancer (BC) development and progression. Underlying molecular mechanisms remain, however, elusive. Here, we provide electrophysiological evidence for a BKCa splice variant localized to the inner mitochondrial membrane of murine and human BC cells (mitoBKCa). Through a combination of genetic knockdown and knockout along with a cell permeable BKCa channel blocker, we show that mitoBKCa modulates overall cellular and mitochondrial energy production, and mediates the metabolic rewiring referred to as the ‘Warburg effect’, thereby promoting BC cell proliferation in the presence and absence of oxygen. Additionally, we detect mitoBKCa and BKCa transcripts in low or high abundance, respectively, in clinical BC specimens. Together, our results emphasize, that targeting mitoBKCa could represent a treatment strategy for selected BC patients in future.

Introduction: The usefulness of contrast enhanced mammography (CEM) for assessing architectural distortion (AD) has not been defined. This study aimed to determine the diagnostic accuracy of AD enhancement on CEM for predicting malignancy and its histopathologic correlation. Material and methods: Patients with AD detected on 2D mammography, tomosynthesis (DBT), and/or ultrasound with BI-RADS categories 4 and 5 and supplementary evaluation with CEM were included. Histopathologic confirmation of the diagnosis or 24-month follow-up imaging studies to determine stability were reported. The risk of malignancy with AD enhancement was assessed with odds ratios (OR) with 95% confidence intervals (CI). Sensitivity, specificity, PPV, NPV, and accuracy of AD enhancement for predicting malignancy were calculated. Results: Forty-nine patients with 50 AD lesions (one patient had two AD lesions) were included. Twenty-nine (58.0%) were benign AD lesions, 5 (10.0%) were benign with upgrade potential (BWUP) lesions, and 16 (32.0%) were malignant. AD enhancement on CEM was observed in all breast malignancy cases (n = 16, 100%), compared to 12 (41.4%) of 29 benign ADs (p < 0.001). The risk of malignancy with AD enhancement on CEM was OR = 23.54 (95% CI, 2.77 to 199.9). AD enhancement on CEM had a sensitivity of 95.2% (95% CI, 76.2 to 99.9) and a specificity of 58.6% (95% CI 38.9 to 76.5) for predicting malignancy. The PPV was 69.7 (95% CI, 59.6 to 78.2), the NPV was 92.5% (95% CI, 63.9 to 98.8), and diagnostic accuracy was 76.9% (95% CI, 62.8 to 87.6). Conclusion: AD enhancement on CEM has high diagnostic accuracy for predicting breast malignancy. This study is the first in Mexico to present the experience with CEM in the evaluation of AD.

Abstract Inflammatory cytokines and non-esterified fatty acids (NEFAs) are obesity-linked factors that disturb insulin secretion. The aim of this study was to investigate whether pancreatic adipose tissue (pWAT) is able to generate a NEFA/cytokine overload within the pancreatic environment and as consequence to impact on insulin secretion. Pancreatic fat is a minor fat depot, therefore we used high-fat diet (HFD) feeding to induce pancreatic steatosis in mice. Relative Adipoq and Lep mRNA levels were higher in pWAT of HFD compared to chow diet mice. Regardless of HFD, Adipoq and Lep mRNA levels of pWAT were at least 10-times lower than those of epididymal fat (eWAT). Lipolysis stimulating receptors Adrb3 and Npr1 were expressed in pWAT and eWAT, and HFD reduced their expression in eWAT only. In accordance, HFD impaired lipolysis in eWAT but not in pWAT. Despite expression of Npr mRNA, lipolysis was stimulated solely by the adrenergic agonists, isoproterenol and adrenaline. Short term co-incubation of islets with CD/HFD pWAT did not alter insulin secretion. In the presence of CD/HFD eWAT, glucose stimulated insulin secretion only upon isoproterenol-induced lipolysis, i.e. in the presence of elevated NEFA. Isoproterenol augmented Il1b and Il6 mRNA levels both in pWAT and eWAT. These results suggest that an increased sympathetic activity enhances NEFA and cytokine load of the adipose microenvironment, including that of pancreatic fat, and by doing so it may alter beta-cell function.

Fires in photovoltaic (PV) electrical systems are a real and serious problem because this phenomenon can have severe consequences for the safety of people and the environment. In some cases, fires result from a lack of maintenance or improper installation of PV modules. It is essential to consider prevention and continuous monitoring of the electrical parameters to minimize these risks, as these factors increase the temperature of the photovoltaic modules. The use of thermal analysis techniques can prevent hotspots and fires in photovoltaic systems; these techniques allow detecting and correcting problems in the installation, such as shadows, dirt, and poor-quality connections in PVs. This paper presents a case study of the implementation of thermal analysis in an installation of photovoltaic modules connected to a solar pumping system to identify the formation of hotspots through thermal images using an unmanned aerial vehicle (UAV). Here, a novel methodology is proposed based on the comparison of temperature increases concerning the values of short circuit current, open circuit voltage, and real efficiency of each PV module. In addition, an electrical safety methodology is proposed to design a photovoltaic system that prevents fires caused by hotspots, contemplating critical parameters such as photovoltaic power, number of photovoltaic modules, DC:AC conversion ratio, electrical conductor selection, control devices, and electrical protection; the performance power expected was obtained using standard power test conditions, including irradiance factor, photovoltaic module (PVM) temperature factor, and power reduction factor.

The integration of healthy management alternatives continues to be a challenge in the organic production of aromatic and medicinal plants, including of basil (Ocimum basilicum L.). The objective of this work was to evaluate the effects of three beneficial microorganisms (1) Trichoderma harzianum (TH), (2) Bacillus subtilis (BS), (3) Glomus cubense (GC) and their combinations on the growth of basil. A completely randomised design was used with a control and seven treatments with six repetitions. The control (1) was with no microorganism inoculation and the seven treatments were inoculations with the single or the combined microorganisms as follows: (2) TH, (3) BS, (4) GC, (5) TH+BS, (6) TH+GC, (7) BS+GC and (8) TH+BS+GC. Three harvests of fresh biomass were made and a number of growth variables were recorded: fresh and dry biomass, leaf area, number of commercial stems, stem length and thickness, Leaf length and width, relative chlorophyll concentration (SPAD readings) and the levels of N, P, K, Ca and Mg. Overall growth increased by 58% with TH+GC compared with the control and by 55% compared with the single inoculations (TH, BS and GC) and with the triple inoculation (TH+BS+GC). A growth increase of 51% was obtained with BS+GC compared with the control and of 38% compared with the other treatments. These results indicate co-inoculation of TH+GC or of BS+GC are useful alternative managements to increase greenhouse production of basil.

The effect of pulsing on the postharvest behavior of cut tuberose was evaluated in the cultivars ‘Mexican Single’ and ‘The Pearl’. Spikes of both were harvested with 1 or 2 of their basal buds open and treated with 20% sucrose and 200 mg of hydroxyquinoline citrate (sucrose + HQC). Pulsing increased the relative fresh weight of spikes, although water uptake in ‘The Pearl’ was greater regardless. After 5 days, visual quality was scored as ‘excellent’ and ‘good’ in ‘Mexican Single’ and ‘The Pearl’, respectively, and the greatest number of open and of senescent flowers occurred in the latter independent of pulsing. CIE L*C*h values and total sugars increased more noticeably among pulsed florets in ‘The Pearl’, whereas higher rates of respiration occurred in the unpulsed spikes of ‘Mexican Single’. Catalase activity was constant in ‘The Pearl’, although superoxide dismutase and peroxidase activities were higher and similar to, respectively, those of ‘Mexican Single’. These differences in physical, physiological and enzymatic behavior are important to consider when developing postharvest conservation and management strategies that are specific for each variety.
