Auscultation methods enable non-invasive diagnosis of diseases, e.g. of the heart, based on heartbeat sounds. Regular, early examinations using machine learning techniques could help to detect diseases at an early stage to prevent serious health conditions and then provide optimal therapy through continuous monitoring. There is already a lot of work on human data using AI algorithms to detect patterns in signals or images. However, there is hardly no work on detecting heart murmurs with digital such as Myxomatous Mitral Valve Disease. In this paper, we present a canine auscultation project that aims to provide a tool to establish a baseline of classification parameters from audio signals that could be used to monitor canine health status by analyzing deviations from this baseline. In the future, data analysis could also lead to prediction and early detection of other diseases.

ZusammenfassungIn einem Schweinemastbetrieb in Süddeutschland konnten über einen Zeitraum von 4 Wochen Hinterhandlähmungen bei 2 Altersgruppen (50 kg sowie 60 kg) beobachtet werden. Die Morbidität bei den beiden Altersgruppen war mit 3,3 % niedrig, der Großteil der betroffenen Tiere musste jedoch aufgrund einer fortschreitenden Paralyse der Hintergliedmaßen euthanasiert werden. Im Rahmen der pathomorphologischen Untersuchung von 2 Mastschweinen konnte eine hochgradige und diffuse lymphohistiozytäre Meningoenzephalomyelitis und Vaskulitis festgestellt werden. Immunhistochemisch wurde in allen Arealen des zentralen Nervensystems und in mehreren Zelltypen (Neuronen, Gliazellen, Endothelzellen, mononukleäre Zellen) Porzines Teschovirus-Antigen nachgewiesen. Die Untersuchung von Rückenmarksproben mittels PCR ergab den Nachweis von Porzinen Teschoviren. Der anschließend durchgeführten phylogenetischen Analyse zufolge wiesen die PCR positiven Proben die höchste Identität (88 % Vollgenomsequenz) zum Porzinen Teschovirus A11 Stamm „Dresden“ auf. Andere differentialdiagnostisch in Frage kommende Pathogene konnten mittels PCR, bakteriologischer Untersuchung und Sequenzierung nicht nachgewiesen werden. In dem Mastbestand traten nach ca. 4 Wochen keine weiteren Fälle von Lähmungserscheinungen mehr auf.

ZusammenfassungIn einem Jungsauenerzeugerbetrieb in Niederösterreich traten über mehrere Jahre hinweg gehäuft Atemwegserkrankungen bei Jungsauen aus Eigenremontierung nach Einstallung in die Altsauenherde auf. Im Herbst 2019 wurden zudem Fruchtbarkeitsstörungen in Form von Spätaborten und Umrauschen beobachtet. Bei der Untersuchung von Nasentupfern mittels PCR auf Influenza-A-Virus (IAV) konnte bei 3 Jungsauen mit respiratorischer Symptomatik und Fieber der IAV-Subtyp H1avN1 nachgewiesen werden. Die Untersuchung der Serumproben dieser Tiere an 2 Zeitpunkten im Abstand von 3 Wochen ergab jedoch keinen Nachweis von Antikörpern im Hämagglutinationshemmtest (HAH), der standardmäßig im Labor verwendet wurde. Auch bei der Untersuchung von Sauen weiterer Altersgruppen waren keine eindeutigen Antikörpertiter gegenüber H1avN1 detektierbar. Nach Erweiterung des diagnostischen Panels des HAH um 7 H1avN1-Testantigene konnte eine Serokonversion um bis zu 3 Titerstufen bei den PCR-positiven Sauen gegenüber 2 verschiedenen H1avN1-Isolaten gemessen werden. Darüber hinaus ließen sich auch bei der Mehrzahl der restlichen untersuchten Altersgruppen hohe Antikörpertiter gegen diese beiden H1avN1-Teststämme nachweisen. Nach Anwendung des europaweit zugelassenen trivalenten Influenzaimpfstoffs konnte das klinische Erscheinungsbild in der Herde deutlich verbessert werden. Der Fallbericht verdeutlicht, dass für eine zielgerichtete Influenzadiagnostik der direkte und der indirekte Erregernachweis kombiniert verwendet werden sollten. Zudem wurde gezeigt, dass die kontinuierliche Anpassung von Testantigenen an die im Feld zirkulierenden Isolate überaus entscheidend für die Aussagekraft des HAH wäre.

ZusammenfassungDer Porcine Respiratory Disease Complex (PRDC) beschreibt eine klinische Kondition, die sich in Form einer häufig therapieresistenten Atemwegsinfektion bei Mastschweinen manifestiert. Die multifaktorielle Ätiologie beinhaltet infektiöse und nicht infektiöse Faktoren. Bei Entstehung und Verlauf des PRDC spielen neben Management und Hygiene v. a. virale und bakterielle Erreger eine bedeutende Rolle. Das Virus des Porzinen Reproduktiven und Respiratorischen Syndroms (PRRSV), das porzine Circovirus Typ 2 (PCV2), Influenza-A-Virus (IAV) und Mycoplasma (M.) hyopneumoniae stellen die relevantesten Erreger dar. Das klinische Bild und die zugrundeliegenden pathomorphologischen Veränderungen können je nach Erregerbeteiligung variieren. Die Komplexität des PRDC erschwert die Diagnose und auch die Prävention auf Bestandsebene. Der Übersichtsartikel gibt einen Einblick in die Pathomorphologie, Pathogenese sowie Inter-Erreger-Interaktionen und zielt darauf ab, praktizierende Tierärztinnen und Tierärzte bei der Diagnose, Befundinterpretation und Prävention des PRDC zu unterstützen.

Synchrotron processes, the radiative processes associated with the interaction of energetic charged particles with magnetic field, are of interest in many areas in astronomy, from the interstellar medium to extreme environments near compact objects. Consequently, observations of synchrotron sources carry information on the physical properties of the sources themselves and those of their close vicinity. In recent years, novel observations of such sources with multi-wavelength collaborations reveal complex features and peculiarities, especially near black holes. Exploring the nature of these sources in more detail necessitates numerical tools complementary to analytical one-zone modelling efforts. In this paper, we introduce an open-source Python package tailored to this purpose, FLAREMODEL. The core of the code consists of low-level utility functions to describe physical processes relevant to synchrotron sources, which are written in C for performance and parallelised with OpenMP for scalability. The Python interface provides access to these functions and built-in source models are provided as a guidance. At the same time, the modular design of the code and the generic nature of these functions enable users to build a variety of source models applicable to many astrophysical synchrotron sources. We describe our methodology and the structure of our code along with selected examples demonstrating capabilities and options for future modelling efforts.

We present a large ${\sim 30" \times 30"}$ spectroscopic survey of the Galactic Center using the SINFONI IFU at the VLT. Combining observations of the last two decades we compile spectra of over $2800$ stars. Using the Bracket-$\gamma$ absorption lines we identify $195$ young stars, extending the list of known young stars by $79$. In order to explore the angular momentum distribution of the young stars, we introduce an isotropic cluster prior. This prior reproduces an isotropic cluster in a mathematically exact way, which we test through numerical simulations. We calculate the posterior angular momentum space as function of projected separation from Sgr~A*. We find that the observed young star distribution is substantially different from an isotropic cluster. We identify the previously reported feature of the clockwise disk and find that its angular momentum changes as function of separation from the black hole, and thus confirm a warp of the clockwise disk ($p \sim 99.2\%$). At large separations, we discover three prominent overdensities of angular momentum. One overdensity has been reported previously, the counter-clockwise disk. The other two are new. Determining the likely members of these structures, we find that as many as $75\%$ of stars can be associated with one of these features. Stars belonging to the warped clockwise-disk show a top heavy K-band luminosity function, while stars belonging to the larger separation features do not. Our observations are in good agreement with the predictions of simulations of in-situ star formation, and argue for common formation of these structures., Comment: Accepted for Publication in APJL

A combination of biochemical preparation methods with microscopic, spectroscopic, and mass spectrometric analysis techniques as contemplating state of the art application, was used for direct visualization, localization, and chemical identification of europium in plants. This works illustrates the chemical journey of europium (Eu(III)) through winter rye (Secale cereale L.), providing insight into the possibilities of speciation for Rare Earth Elements (REE) and trivalent f-elements. Kinetic experiments of contaminated plants show a maximum europium concentration in Secale cereale L. after four days. Transport of the element through the vascular bundle was confirmed with Scanning Electron Microscopy (SEM) and Energy Dispersive X-ray analysis (EDS). For chemical speciation, plants were grown in a liquid nutrition medium, whereby Eu(III) species distribution could be measured by mass spectrometry and luminescence measurements. Both techniques confirm the occurrence of Eu malate species in the nutrition medium, and further analysis of the plant was performed. Luminescence results indicate a change in Eu(III) species distribution from root tip to plant leaves. Microscopic analysis show at least three different Eu(III) species with potential binding to organic and inorganic phosphate groups and a Eu(III) protein complex. With plant root extraction, further europium species could be identified by using Electrospray Ionization Mass Spectrometry (ESI MS). Complexation with malate, citrate, a combined malate-citrate ligand, and aspartate was confirmed mostly in a 1:1 stoichiometry (Eu:ligand). The combination of the used analytical techniques opens new possibilities in direct species analysis, especially regarding to the understanding of rare earth elements (REE) uptake in plants. This work provides a contribution in better understanding of plant mechanisms of the f-elements and their species uptake.

Pregravid obesity is one of the major risk factors for pregnancy complications such as gestational diabetes mellitus (GDM) and an increased risk of cardiovascular events in children of affected mothers. However, the biological mechanisms that underpin these adverse outcomes are not well understood. High-density lipoproteins (HDLs) are antiatherogenic by promoting the efflux of cholesterol from macrophages and by suppression of inflammation. Functional impairment of HDLs in obese and GDM-complicated pregnancies may have long-term effects on maternal and offspring health. In the present study, we assessed metrics of HDL function in sera of pregnant women with overweight/obesity of the DALI lifestyle trial (prepregnancy BMI ≥ 29 kg/m2) and women with normal weight (prepregnancy BMI < 25 kg/m2), as well as HDL functionalities in cord blood at delivery. We observed that pregravid obesity was associated with impaired serum antioxidative capacity and lecithin–cholesterol acyltransferase activity in both mothers and offspring, whereas maternal HDL cholesterol efflux capacity was increased. Interestingly, functionalities of maternal and fetal HDL correlated robustly. GDM did not significantly further alter the parameters of HDL function and metabolism in women with obesity, so obesity itself appears to have a major impact on HDL functionality in mothers and their offspring.

In a gilt producing farm in Lower Austria, respiratory diseases occurred over the previous years in self-reared gilts after being introduced into the sow herd. In addition, fertility disorders in terms of late abortions and re-breeders were observed in the fall of 2019. Nasal swabs of 3 gilts with respiratory signs and fever were tested positive for influenza A virus (IAV) subtype H1avN1 by PCR. However, examination of serum samples from these animals at 2 different time points did not detect antibodies using the standard hemagglutination inhibition (HI) test of the laboratory. Examination of additional age groups likewise failed to detect H1avN1 antibody titers. In consequence to the extension of the diagnostic panel of the HI test by 7 additional H1avN1 test antigens, a clear seroconversion of the PCR positive sows against 2 different H1avN1 isolates could be measured. In addition, high antibody titers against these 2 H1avN1 strains were also detectable in the majority of the remaining age groups tested. Following the administration of the trivalent influenza vaccine, which has been approved throughout Europe, a significant improvement of the clinical presentation in the herd was achieved. The present case report illustrates that direct and indirect pathogen detection should be used in combination for targeted influenza diagnostics. Human induced pluripotent stem cell (iPSC) lines are a powerful tool for studying development and disease, but the considerable phenotypic variation between lines makes it challenging to replicate key findings and integrate data across research groups. To address this issue, we sub-cloned candidate human iPSC lines and deeply characterized their genetic properties using whole genome sequencing, their genomic stability upon CRISPR-Cas9-based gene editing, and their phenotypic properties including differentiation to commonly used cell types. These studies identified KOLF2.1J as an all-around well-performing iPSC line. We then shared KOLF2.1J with groups around the world who tested its performance in head-to-head comparisons with their own preferred iPSC lines across a diverse range of differentiation protocols and functional assays. On the strength of these findings, we have made KOLF2.1J and its gene-edited derivative clones readily accessible to promote the standardization required for large-scale collaborative science in the stem cell field.

Fully automated proteomic pipelines have the potential to achieve deep coverage of cellular proteomes with high throughput and scalability. However, it is important to evaluate performance, including both reproducibility and ability to provide meaningful levels of biological insight. Here, we present an approach combining high field asymmetric waveform ion mobility spectrometer (FAIMS) interface and data independent acquisition (DIA) proteomics approach developed as part of the induced pluripotent stem cell (iPSC) Neurodegenerative Disease Initiative (iNDI), a large-scale effort to understand how inherited diseases may manifest in neuronal cells. Our FAIMS-DIA approach identified more than 8000 proteins per mass spectrometry (MS) acquisition as well as superior total identification, reproducibility, and accuracy compared to other existing DIA methods. Next, we applied this approach to perform a longitudinal proteomic profiling of the differentiation of iPSC-derived neurons from the KOLF2.1J parental line used in iNDI. This analysis demonstrated a steady increase in expression of mature cortical neuron markers over the course of neuron differentiation. We validated the performance of our proteomics pipeline by comparing it to single cell RNA-Seq datasets obtained in parallel, confirming expression of key markers and cell type annotations. In summary, we report an extensively optimized and validated proteomic pipeline that will be suitable for large-scale studies such as iNDI.

Reproductive disorders induced by porcine reproductive and respiratory syndrome virus (PRRSV) cause high economic losses in the pig industry worldwide. In this study, we aimed to phenotypically characterize a virulent PRRSV-1 subtype 1 isolate (AUT15-33) in a reproductive model. Furthermore, the protective effect of a heterologous modified live virus vaccine (ReproCyc® PRRS EU) was evaluated. In addition, PRRSV AUT15-33 was genotypically compared to other well-characterized isolates. Sixteen gilts were equally divided into four groups: a vaccinated and infected group (V–I), a vaccinated and non-infected group (V–NI), a non-vaccinated and infected group (NV–I), and a non-vaccinated and non-infected (NV–NI) group. After PRRSV infection on gestation day 84, all gilts were clinically examined on a daily basis, and blood samples were taken at five timepoints. Necropsy was performed 3 weeks after infection. The fetal preservation status was assessed, and PRRSV RNA concentrations were measured in the blood and tissue samples from all gilts and fetuses. After infection, all four gilts in the NV–I group were viremic throughout 17 days post-infection (dpi), whereas two gilts in the V–I group were viremic at only one timepoint at 6 dpi. The viral load was significantly higher in gilt serum, tracheobronchial lymph nodes, uterine lymph nodes, maternal endometrium, and fetal placenta of NV–I gilts compared to the V–I ones (p < 0.05). Moreover, the preservation status of the fetuses derived from NV–I gilts was significantly impaired (55.9% of viable fetuses) compared to the other groups (p < 0.001). Upon comparison with other known isolates, the phylogenetic analyses revealed the closest relation to a well-characterized PRRSV-1 subtype 1 field isolate from Belgium. In conclusion, the high virulence of AUT15-33 was phenotypically confirmed in an experimental reproductive model. The vaccination of the gilts showed promising results in reducing viremia, fetal damage, and transplacental transmission of the PRRSV-1 strain characterized in this study.

Background The prognosis of patients with brain metastases (BM) is poor despite advances in our understanding of the underlying pathophysiology. The high incidence of thrombotic complications defines tumor progression and the high mortality rate. We, therefore, postulated that von Willebrand factor (VWF) promotes BM via its ability to induce platelet aggregation and thrombosis. Methods We measured the abundance of VWF in the blood and intravascular platelet aggregates of patients with BM, and determined the specific contribution of endothelial and platelet-derived VWF using in vitro models and microfluidics. The relevance for the brain metastatic cascade in vivo was demonstrated in ret transgenic mice, which spontaneously develop BM, and by the intracardiac injection of melanoma cells. Results Higher levels of plasma VWF in patients with BM were associated with enhanced intraluminal VWF fiber formation and platelet aggregation in the metastatic tissue and peritumoral regions. Platelet activation triggered the formation of VWF multimers, promoting platelet aggregation and activation, in turn enhancing tumor invasiveness. The absence of VWF in platelets, or the blocking of platelet activation, abolished platelet aggregation, and reduced tumor cell transmigration. Anticoagulation and platelet inhibition consistently reduced the number of BM in preclinical animal models. Conclusions Our data indicate that platelet-derived VWF is involved in cerebral clot formation and in metastatic growth of melanoma in the brain. Targeting platelet activation with low-molecular-weight heparins represents a promising therapeutic approach to prevent melanoma BM.

We report the time-resolved spectral analysis of a bright near-infrared and moderate X-ray flare of Sgr A*. We obtained light curves in the $M$-, $K$-, and $H$-bands in the mid- and near-infrared and in the $2-8~\mathrm{keV}$ and $2-70~\mathrm{keV}$ bands in the X-ray. The observed spectral slope in the near-infrared band is $\nu L_\nu\propto \nu^{0.5\pm0.2}$; the spectral slope observed in the X-ray band is $\nu L_\nu \propto \nu^{-0.7\pm0.5}$. We tested synchrotron and synchrotron self-Compton (SSC) scenarios. The observed near-infrared brightness and X-ray faintness, together with the observed spectral slopes, pose challenges for all models explored. We rule out a scenario in which the near-infrared emission is synchrotron emission and the X-ray emission is SSC. A one-zone model in which both the near-infrared and X-ray luminosity are produced by SSC and a model in which the luminosity stems from a cooled synchrotron spectrum can explain the flare. In order to describe the mean SED, both models require specific values of the maximum Lorentz factor $\gamma_{max}$, which however differ by roughly two orders of magnitude: the SSC model suggests that electrons are accelerated to $\gamma_{max}\sim 500$, while cooled synchrotron model requires acceleration up to $\gamma_{max}\sim5\times 10^{4}$. The SSC scenario requires electron densities of $10^{10}~\mathrm{cm^{-3}}$ much larger than typical ambient densities in the accretion flow, and thus require in an extraordinary accretion event. In contrast, assuming a source size of $1R_s$, the cooled synchrotron scenario can be realized with densities and magnetic fields comparable with the ambient accretion flow. Its multifactorial etiology includes infectious and non-infectious factors. Besides management and hygiene conditions, particularly viral and bacterial pathogens contribute to the development and course of PRDC. Porcine circovirus type 3 (PCV3) is regularly reported in association with various clinical presentations, including porcine dermatitis and nephropathy syndrome (PDNS)-like lesions, respiratory signs, congenital tremor, and reproductive disorders. To investigate the epidemiology of PCV3 in a boar stud, we analysed fresh boar semen and matching sera from 181 boars from a German stud supplying semen for artificial insemination (AI) to approximately 740 breeder farms for PCV3 DNA. PCV3 DNA was detected in 1.7% semen samples and 24.3% sera. Spearman rho correlation demonstrated a significant positive correlation between boar age and quantitative DNA (by PCR quantification cycles [Cq] values) in serum samples (r = 0.636; P0.001). Sera from boars up to 12 months of age had higher viral loads (P0.001) and were PCV3-positive significantly more often (P0.01) than older boars. Detection of PCV3 DNA was not associated with breed (P0.05). PCV3 DNA was detected sporadically in fresh boar semen. Based on the assumption that processing fresh semen reduces viral load in semen used for AI, it is likely that the risk of sexual transmission of PCV3 during AI in is low. However, young boars may contribute to the maintenance of PCV3 infection in boar studs.

The impact of dark matter-neutrino interactions on the measurement of the cosmological parameters has been investigated in the past in the context of massless neutrinos exclusively. Here we revisit the role of a neutrino-dark matter coupling in light of ongoing cosmological tensions by implementing the full Boltzmann hierarchy for three massive neutrinos. Our tightest 95% CL upper limit on the strength of the interactions, parameterized via $u_\chi =\frac{\sigma_0}{\sigma_{Th}}\left(\frac{m_\chi}{100 \text{GeV}}\right)^{-1}$, is $u_\chi\leq3.34 \cdot 10^{-4}$, arising from a combination of Planck TTTEEE data, Planck lensing data and SDSS BAO data. This upper bound is, as expected, slightly higher than previous results for interacting massless neutrinos, due to the correction factor associated with neutrino masses. We find that these interactions significantly relax the lower bounds on the value of $\sigma_8$ that is inferred in the context of $\Lambda$CDM from the Planck data, leading to agreement within 1-2$\sigma$ with weak lensing estimates of $\sigma_8$, as those from KiDS-1000. These protoplanetary disks in T Tauri stars play a central role in star and planet formation. We spatially resolve at sub-au scales the innermost regions of a sample of T Tauri's disks to better understand their morphology and composition. We extended our homogeneous data set of 27 Herbig stars and collected near-IR K-band observations of 17 T Tauri stars, spanning effective temperatures and luminosities in the ranges of ~4000-6000 K and ~0.4-10 Lsun. We focus on the continuum emission and develop semi-physical geometrical models to fit the interferometric data and search for trends between the properties of the disk and the central star. The best-fit models of the disk's inner rim correspond to wide rings. We extend the Radius-luminosity relation toward the smallest luminosities (0.4-10 Lsun) and find the R~L^(1/2) trend is no longer valid, since the K-band sizes measured with GRAVITY are larger than the predicted sizes from sublimation radius computation. No clear correlation between the K-band half-flux radius and the mass accretion rate is seen. Having magnetic truncation radii in agreement with the K-band GRAVITY sizes would require magnetic fields as strong as a few kG, which should have been detected, suggesting that accretion is not the main process governing the location of the half-flux radius of the inner dusty disk. Our measurements agree with models that take into account the scattered light. The N-to-K band size ratio may be a proxy for disentangling disks with silicate features in emission from disks with weak and/or in absorption silicate features. International audience; The spin of the supermassive black hole that resides at the Galactic Center can, in principle, be measured by accurate measurements of the orbits of stars that are much closer to Sgr A* than S2, the orbit of which recently provided the measurement of the gravitational redshift and the Schwarzschild precession. The GRAVITY near-infrared interferometric instrument combining the four 8m telescopes of the VLT provides a spatial resolution of 2–4 mas, breaking the confusion barrier for adaptive-optics-assisted imaging with a single 8–10m telescope. We used GRAVITY to observe Sgr A* over a period of six months in 2019 and employed interferometric reconstruction methods developed in radio astronomy to search for faint objects near Sgr A*. This revealed a slowly moving star of magnitude 18.9 in the K-band within 30 mas of Sgr A*. The position and proper motion of the star are consistent with the previously known star S62, which is at a substantially greater physical distance, but in projection passes close to Sgr A*. Observations in August and September 2019 detected S29 easily, with K-magnitude of 16.6, at approximately 130 mas from Sgr A*. Using general relativistic magnetohydrodynamic simulations of accreting black holes, we show that a suitable subtraction of the linear polarization per pixel from total intensity images can enhance the photon ring features. We find that the photon ring is typically a factor of $\simeq 2$ less polarized than the rest of the image. This is due to a combination of plasma and general relativistic effects, as well as magnetic turbulence. When there are no other persistently depolarized image features, adding the subtracted residuals over time results in a sharp image of the photon ring. We show that the method works well for sample, viable GRMHD models of Sgr A* and M87*, where measurements of the photon ring properties would provide new measurements of black hole mass and spin, and potentially allow for tests of the "no-hair" theorem of general relativity., Comment: 13 pages, 12 figures, accepted for publication in MNRAS

The formation and evolution of planetary systems impact the primordial accretion disk. HD141569 is the only known pre-main sequence star characterized by a hybrid disk. Observations probed the outer-disk structure showing a complex system of rings and interferometric observations attempted to characterize its inner 5 au region, but derived limited constraints. The goal of this work was to explore with new high-resolution interferometric observations the properties of the dust and gas in the internal regions of HD141569. We observed HD141569 on mas scales with GRAVITY/VLTI in the near-infrared at low and high spectral resolution. We interpreted the visibilities and spectral energy distribution with geometrical models and radiative transfer techniques to constrain the dust emission. We analyzed the high spectral resolution quantities to investigate the properties of the Br-Gamma line emitting region. Thanks to the combination of three different epochs, GRAVITY resolves the inner dusty disk in the K band. Data modeling shows that an IR excess of about 6% is spatially resolved and that the origin of this emission is confined in a ring of material located at a radius of 1 au from the star with a width smaller than 0.3 au. The MCMax modeling suggests that this emission could originate from a small amount of QHPs, while large silicate grain models cannot reproduce at the same time the observational constraints on the properties of near-IR and mid-IR fluxes. The differential phases in the Br-Gamma line clearly show an S-shape that can be best reproduced witha gas disk in Keplerian rotation, confined within 0.09 au. This is also hinted at by the double-peaked Br-Gamma emission line shape. The modeling of the continuum and gas emission shows that the inclination and position angle of these two components are consistent with a system showing relatively coplanar rings on all scales., Comment: Accepted for publication in A&A; 25 pages, 15 figures, 5 tables

GCIRS 7, the brightest star in the Galactic central parsec, formed $6\pm2$ Myr ago together with dozens of massive stars in a disk orbiting the central black-hole. It has been argued that GCIRS 7 is a pulsating body, on the basis of photometric variability. We present the first medium-resolution ($R=500$), K-band spectro-interferometric observations of GCIRS 7, using the GRAVITY instrument with the four auxiliary telescopes of the ESO VLTI. We looked for variations using two epochs, namely 2017 and 2019. We find GCIRS 7 to be moderately resolved with a uniform-disk photospheric diameter of $\theta^*_\text{UD}=1.55 \pm 0.03$ mas ($R^*_\text{UD}=1368 \pm 26$ $R_\odot$) in the K-band continuum. The narrow-band uniform-disk diameter increases above 2.3 $\mu$m, with a clear correlation with the CO band heads in the spectrum. This correlation is aptly modeled by a hot ($T_\text{L}=2368\pm37$ K), geometrically thin molecular shell with a diameter of $\theta_\text{L}=1.74\pm0.03$ mas, as measured in 2017. The shell diameter increased ($\theta_\text{L}=1.89\pm0.03$ mas), while its temperature decreased ($T_\text{L}=2140\pm42$ K) in 2019. In contrast, the photospheric diameter $\theta^*_\text{UD}$ and the extinction up to the photosphere of GCIRS 7 ($A_{\mathrm{K}_\mathrm{S}}=3.18 \pm 0.16$) have the same value within uncertainties at the two epochs. In the context of previous interferometric and photo-spectrometric measurements, the GRAVITY data allow for an interpretation in terms of photospheric pulsations. The photospheric diameter measured in 2017 and 2019 is significantly larger than previously reported using the PIONIER instrument ($\theta_*=1.076 \pm 0.093$ mas in 2013 in the H band). The parameters of the photosphere and molecular shell of GCIRS 7 are comparable to those of other red supergiants that have previously been studied using interferometry., Comment: 12 pages, 11 figures, 3 tables. During the past years, the VLTI-instrument GRAVITY has made spectacular discoveries with phase-referenced interferometric imaging with milliarcsecond resolution and ten microarcsecond astrometry. Here, we report on the upgrade of the GRAVITY science spectrometer with two new grisms in October 2019, increasing the instrument throughput by a factor > 2. This improvement was made possible by using a high refractive index Germanium substrate, which reduces the grism and groove angles, and by successfully applying an anti-reflection coating to the ruled surface to overcome Fresnel losses. We present the design, manufacturing, and laboratory testing of the new grisms, as well as the results from the re-commissioning on sky.

Human induced pluripotent stem cell (iPSC) lines are a powerful tool for studying development and disease, but the considerable phenotypic variation between lines makes it challenging to replicate key findings and integrate data across research groups. To address this issue, we sub-cloned candidate iPSC lines and deeply characterised their genetic properties using whole genome sequencing, their genomic stability upon CRISPR/Cas9-based gene editing, and their phenotypic properties including differentiation to commonly-used cell types. These studies identified KOLF2.1J as an all-around well-performing iPSC line. We then shared KOLF2.1J with groups around the world who tested its performance in head-to-head comparisons with their own preferred iPSC lines across a diverse range of differentiation protocols and functional assays. On the strength of these findings, we have made KOLF2.1J and hundreds of its gene-edited derivative clones readily accessible to promote the standardization required for large-scale collaborative science in the stem cell field.SummaryThe authors of this collaborative study deeply characterized human induced pluripotent stem cell (iPSC) lines to rationally select a clonally-derived cell line that performs well across multiple modalities. KOLF2.1J was identified as a candidate reference cell line based on single-cell analysis of its gene expression in the pluripotent state, whole genome sequencing, genomic stability after highly efficient CRISPR-mediated gene editing, integrity of the p53 pathway, and the efficiency with which it differentiated into multiple target cell populations. Since it is deeply characterized and can be readily acquired, KOLF2.1J is an attractive reference cell line for groups working with iPSCs.Graphical abstract

Aims.We aim to demonstrate that the presence and mass of an exoplanet can now be effectively derived from the astrometry of another exoplanet.Methods.We combined previous astrometry ofβPictoris b with a new set of observations from the GRAVITY interferometer. The orbital motion ofβPictoris b is fit using Markov chain Monte Carlo simulations in Jacobi coordinates. The inner planet,βPictoris c, was also reobserved at a separation of 96 mas, confirming the previous orbital estimations.Results.From the astrometry of planet b only, we can (i) detect the presence ofβPictoris c and (ii) constrain its mass to 10.04−3.10+4.53MJup. If one adds the astrometry ofβPictoris c, the mass is narrowed down to 9.15−1.06+1.08MJup. The inclusion of radial velocity measurements does not affect the orbital parameters significantly, but it does slightly decrease the mass estimate to 8.89−0.75+0.75MJup. With a semimajor axis of 2.68 ± 0.02 au, a period of 1221 ± 15 days, and an eccentricity of 0.32 ± 0.02, the orbital parameters ofβPictoris c are now constrained as precisely as those ofβPictoris b. The orbital configuration is compatible with a high-order mean-motion resonance (7:1). The impact of the resonance on the planets’ dynamics would then be negligible with respect to the secular perturbations, which might have played an important role in the eccentricity excitation of the outer planet.
