The combination of lower diet quality and increased metabolic rates is assumed to cause cascading effects on organismic C cycling. Future changes in CO2 levels or terrestrial nutrient discharges in marine ecosystems can lead to increased phytoplankton C:N ratios relative to consumer C:N ratios, lowering the quality of the food source. In this study, we compared the single and interactive effects of diet quality and temperature on the feeding behavior and C and N intake and release of a common and abundant intertidal mixotrophic protist, the foraminifer Haynesina germanica. Two batches of artificially produced and dual isotope-labeled (13C/15N) chlorophyte detritus with different C:N ratios (5.6 and 7.1) were fed to the foraminifer at 3 different temperatures (15, 20, 25°C). We observed a strong interactive effect of temperature and diet. A very strong increase in feeding rates was observed at 20°C for the low-quality food source. Respiration rates of carbon derived from the low-quality diet (C:N ratio of 7.1) were lower than those of the high-quality diets and increased at 25°C. This indicates that a high C content of the diet might be of advantage in calcifying mixotrophs, since respired excess C could be advantageous for test calcification. Additionally, respired excess C could be a useful resource of CO2 for kleptoplast photosynthesis and functionality in the mixotrophic lifestyle of H. germanica. Further, the observed effects of diet and temperature could impact nutrient fluxes in the habitat of H. germanica, possibly leading to food-web shifts in the future.

Benthic foraminifera are abundant marine protists which play an important role in the transfer of energy in the form of organic matter and nutrients to higher trophic levels. Due to their aquatic lifestyle, factors such as water temperature, salinity and pH are key drivers controlling biomass turnover through foraminifera. In this study the influence of salinity on the feeding activity of foraminifera was tested. Two species, Ammonia tepida and Haynesina germanica, were collected from a mudflat in northern Germany (Friedrichskoog) and cultured in the laboratory at 20 ∘C and a light–dark cycle of 16:08 h. A lyophilized algal powder from Dunaliella tertiolecta, which was isotopically enriched with 13C and 15N, was used as a food source. The feeding experiments were carried out at salinity levels of 11, 24 and 37 practical salinity units (PSU) and were terminated after 1, 5 and 14 d. The quantification of isotope incorporation was carried out by isotope ratio mass spectrometry. Ammonia tepida exhibited a 10-fold higher food uptake compared to H. germanica. Furthermore, in A. tepida the food uptake increased with increasing salinity but not in H. germanica. Over time (from 1–5 to 14 d) food C retention increased relative to food N in A. tepida while the opposite was observed for H. germanica. This shows that if the salinity in the German Wadden Sea increases, A. tepida is predicted to exhibit a higher C and N uptake and turnover than H. germanica, with accompanying changes in C and N cycling through the foraminiferal community. The results of this study show how complex and differently food C and N processing of foraminiferal species respond to time and to environmental conditions such as salinity.

Benthic foraminifera (marine protists) with a calcareous shell (test) show a variety of feeding strategies including detrivory, herbivory, bacterivory, carnivory or mixotrophic adaptations. However, little is known about the feeding habits of agglutinated foraminifera, which build there tests from sedimentparticles, embedded in organic cement secreted by the foraminifer. Liebusella goesi is a benthic foraminifer with an agglutinated test, which requires stable hydrographic conditions with low seasonal variation. In general, information on the biology or ecology of this species is very limited. Furthermore, there are no data available on foraminiferal metabolic rates and their feeding ecology (energy consumption in calories/time, food consumption rates) of such foraminifera by now.In this study, we compared the respiration rates of L. goesi at in situ conditions incubated in sterile seawater with or without 1.5 mM labelled glucose (33 atom% 13C/12C) as a potential food source. Additionally, we estimated the individual metabolic rates of L. goesi specimens from their rate of glucose uptake over time.Liebusella goesi individuals were collected in August 2021 with the R/V Oscar von Sydow at the deepest spot of the Swedish Gullmar Fjord (Alsbäck deep, 120m), by sampling the surface layer of sediments recovered from multiple box corer hauls. Subsequently, specimens from the 5000 µm - 125 µm sediment fraction were picked under a stereo microscope, in the laboratory at the Sven Lovén Centre in Kristineberg. Seventy specimens were pooled in six replicates per treatment and incubated within glass vials (1.5 mL) filled with either plain sterile seawater or with seawater amended with 13C-glucose, and sealed airtight. Finally, simultaneous non-invasive oxygen measurements were carried out over the course of 45 h. Foraminifera incubated with 13C-glucose were prepared for elemental analysis and isotope ratio mass spectrometry to evaluate the amount of 13C-glucose intake during the experimental period.The respiration rates of L. goesi were in the range of previously observed rates of other foraminifera. Liebusella goesi actively feeded on dissolved organic carbon in the form of glucose and a significant increase of the respiratory activity of the specimens incubated with the 13C-glucose was observed. Metabolic rates of L. goesi will be presented in comparison with those of other protists based on data synthesis from other available sources.The results of this experiment enhance the knowledge about the metabolism of agglutinated foraminifera. However, further experimentation with other species and eventually further variation of incubation factors will be needed, to get a more comprehensive picture of the feeding strategies and metabolic adaptations of agglutinated foraminifera.

Benthic foraminifera are highly abundant, ubiquitous marine protists, with many species feeding on microalgae or phytodetritus. Knowledge about carbon and nitrogen budgets and metabolic activities of benthic foraminifera can help to increase our understanding about their ecology and their role in aquatic biogeochemistry at the sediment-water interface. This can further increase their application as proxies for environmental changes. Shifts in the benthic foraminiferal communities of the Swedish Gullmars Fjord document the shift from well oxygenated bottom waters to seasonal hypoxia at its deepest location the Alsbäck Deep (125 m), during the last century.So far there are only investigations available relating foraminiferal community composition with increased primary productivity and resulting hypoxia in this Fjord. In contrast, studies about the species-specific feeding ecology or food derived foraminiferal carbon and nitrogen fluxes are scarce.Therefore, laboratory feeding experiments and respiration rate measurements were carried out with Bulimina marginata, Cassidulina laevigata and Globobulima turgida, abundant foraminifera in such environments, collected in August 2017.Experiments were conducted to evaluate the carbon and nitrogen intake and turnover of dual (13C and 15N) isotope labelled Phaeodactylum tricornutum detritus; detritus of a common diatom in the Gullmar Fjord. For the feeding experiments, foraminifera were incubated at 9.1°C in the dark, in sterile filtered seawater at ambient oxygen concentrations. The foraminifera were fed for a period of 24 hours and subsequently incubated without food for another 24 hours. After each incubation cycle, foraminiferal respiration rates were measured. The individuals were analyzed via Elemental Analyzer-Isotope Ratio Mass Spectroscopy to evaluate 13C/12C and 15N/14N ratios and their bulk content of organic carbon and nitrogen.Additionally, we present carbon and nitrogen to volume ratios for the foraminifera B. marginata, C. laevigata, G. turgida, G. auriculata and Nonionella turgida, as derived from elemental analysis and light microscopy imaging.The results show, that B. marginata, an opportunistic species associated with high fluxes of organic matter, had the highest rate of specific carbon and nitrogen intake and turnover. Cassidulina laevigata, a species that co-occurs with fresh phytodetritus and does not tolerate very low oxygen concentrations, showed lower carbon and nitrogen intake rates. Globobulima turgida, a denitrifying infaunal species that thrives under hypoxia, showed the lowest specific carbon and nitrogen intake and turnover rates. Respiration rates of all species did not depend on incubation with or without a food source. The foraminifera showed similar carbon and nitrogen densities per test volume across all species.Overall this study helps to improve the knowledge on the nutritional ecology of the investigated species, demonstrating the close relation between feeding/metabolic rates and their environmental niche and highlighting the need to introduce foraminiferal data in future marine carbon and nitrogen flux models.

Studies of carnivorous behaviour of benthic foraminifers are rare and mostly focused on laboratory experiments. Controlled experiments have shown that some agglutinated and intertidal species prey on meio- to macrofaunal metazoans. Here we present observations of the behaviour of specimens of the infaunal benthic foraminiferal species, Globobulimina auriculata and G. turgida, made within several hours of collection from ∼117 m depth in the Alsbäck Deep of the Gullmar Fjord, Sweden. We observed live nematodes within the tests of G. auriculata. Video observations recorded over a 17-hour period showed a G. auriculata specimen with a living nematode whose tail appeared to be entangled within the foraminifer's reticulopodial network. The nematode eventually coiled around the foraminifer's aperture and became much less active, though ingestion into the foraminifer's test was not documented. If these observations indicate feeding by G. auriculata, they differ from previous observations of predation by Ammonia tepida, which utilised external reticulopodial activity to extract the soft tissue of its prey. An alternative interpretation of the video observations, consistent with the observations of the live nematodes inside G. auriculata, was that the nematode was attempting to prey upon the foraminifer. The G. turgida specimens, in contrast, relatively quickly surrounded themselves in soft sediment spheres commonly seen in deposit-feeding foraminifers, and were never observed with nematodes within their tests. We speculate that these contrasting feeding strategies might reduce competition and facilitate the coexistence of these two globobuliminid species.

Benthic foraminifera play a major role as primary consumers and detrivores redistributing organic carbon and nitrogen in intertidal environments. Here we compared the differences of phytodetrital carbon and nitrogen intake and turnover of two dominant intertidal foraminifera, Ammonia tepida and Haynesina germanica. Their lifestyles in relation to feeding behavior (feeding preferences, intake and turnover of phytodetrital carbon and nitrogen) and temperature adaptations were compared to obtain a closer definition of their specific roles in intertidal organic matter processing. For this comparison, we carried out a series of short-term laboratory incubations with stable-isotope-labeled (13C and 15N) detritus as the food source. We compared the response of the two species to diatom detritus at three different temperatures (15, 20, 25 ∘C). Ammonia tepida showed a very high, temperature-influenced intake and turnover rates with more excessive carbon turnover, compared to nitrogen. The fairly low metabolic nitrogen turnover in H. germanica was not affected by temperature and was higher than the carbon turnover. This might be related with the chloroplast husbandry in H. germanica and its lower demands for food-derived nitrogen sources. Ammonia tepida prefers a soft chlorophyte food source over diatom detritus, which is harder to break down. In conclusion, A. tepida shows a generalist behavior that links with high fluxes of organic matter (OM). Due to its high rates of OM processing and abundances, we conclude that A. tepida is an important key player in intertidal carbon and nitrogen turnover, specifically in the short-term processing of OM and the mediation of dissolved nutrients to associated microbes and primary producers. In contrast, H. germanica is a highly specialized species with low rates of carbon and nitrogen budgeting.

Benthic foraminifera are highly abundant heterotrophic protists in marine sediments, but future environmental changes will challenge the tolerance limits of intertidal species. Metabolic rates and physiological processes in foraminifera are strongly dependent on environmental temperatures. Temperature-related stress could therefore impact foraminiferal food source processing efficiency and might result in altered nutrient fluxes through the intertidal food web. In this study, we performed a laboratory feeding experiment on Ammonia tepida and Haynesina germanica, two dominant foraminiferal species of the German Wadden Sea/Friedrichskoog, to test the effect of temperature on phytodetritus retention. The specimens were fed with 13C and 15N labelled freeze-dried Dunaliella tertiolecta (green algae) at the start of the experiment and were incubated at 20, 25 and 30 °C respectively. Dual labelling was applied to observe potential temperature effects on the relation of phytodetrital carbon and nitrogen retention. Samples were taken over a period of 2 weeks. Foraminiferal cytoplasm was isotopically analysed to investigate differences in carbon and nitrogen uptake derived from the food source. Both species showed a positive response to the provided food source, but carbon uptake rates of A. tepida were 10-fold higher compared to those of H. germanica. Increased temperatures had a far stronger impact on the carbon uptake of H. germanica than on A. tepida. A distinct increase in the levels of phytodetrital-derived nitrogen (compared to more steady carbon levels) could be observed over the course of the experiment in both species. The results suggest that higher temperatures have a significant negative effect on the carbon exploitation of H. germanica. For A. tepida, higher carbon uptake rates and the enhanced tolerance range for higher temperatures could outline an advantage in warmer periods if the main food source consists of chlorophyte phytodetritus. These conditions are likely to impact nutrient fluxes in A. tepida/H. germanica associations.

This study aims to emphasize the ecological importance of foraminifera contained within the > 1000 µm sediment fraction. Stones and gravel offer a habitat for diverse and rich epilithic foraminiferal communities. However, gravel-rich sediments > 1000 µm are usually not the subject of quantitative benthic foraminifera studies, because most foraminifera species used as proxies or bioindicators are sediment-dwelling and found within smaller sediment size fractions. Therefore, there is a current lack of knowledge about the biology and ecology of epilithic foraminifera, specifically about agglutinated species.During a cruise at the Gullmarfjord/Sweden in September 2018, we retrieved a core at a station at 7 m water depth, which contained organic-rich material and a relatively large portion of gravel and shell fragments, which were densely populated by monothalamus, agglutinated foraminifera.A faunal analysis of foraminifera in the > 1000 µm sediment fraction showed, that the most abundant species (> 85 %, 54 ind. 10 cm-3) in > 1000 µm consisted of Tholosina vesicularis, an unicellular agglutinated species that can reach up to 4 mm diameter. SEM-analysis revealed, that large quantities of partially decomposed diatom frustules were embedded within the protoplasm of T. vesicularis individuals, which were supposedly the remains of the foraminiferal diet. The sediment fraction of 125 - 1000 µm was dominated by Ammonia species (58 %, 190 ind. 10 cm-3), a genus known for its fast turnover of diatom-derived carbon and as key-players in benthic nutrient fluxes. Preliminary biovolume analysis of the two genera (T. vesicularis: n = 74, Ammonia spp.: n = 110) resulted in far higher values for T. vesicularis (853 ± 944 µm-3 10 cm-3, med. = 506 µm-3 10 cm-3), than for Ammonia spp. (117 ± 56 µm-3 10 cm-3, med. = 96 µm-3 10 cm-3), within the uppermost sediment layer of 0 - 1 cm. Therefore, T. vesicularis could be a main driver of benthic carbon turnover in gravel-dominated sediments. We hypothesize, that the epilithic fauna, when present is at least as relevant as sediment-dwelling species.Additionally, biodiversity indices (species diversity, Shannon-Index, Evenness), show a slightly higher diversity and a more even distribution within the softer substrate, compared to the stones.This is most likely due to the motility of the free-living forms of the smaller size fraction, which allows them a flexible response to spacial competition. However, niche separation between hard and soft-substrate allows the co-existence of the opportunistic species T. vesicularis and Ammonia spp..In conclusion, this study shows, that if present, the > 1000 µm fraction can contain important information for the interpretation of ecological studies on the communities of live foraminifera and their role in marine ecosystems.

The desmid Oocardium stratum is sporadically and exclusively found in active meteogene travertine habitats. The restriction to these peculiar biotopes is probably linked to specific carbon requirements. We studied inorganic carbon (Ci) uptake under controlled laboratory conditions. First, we developed a method for isolating and cultivating O. stratum. Colonies visible to the naked eye (green pinhead-shaped formations) were collected from a tufa spring in Lower Austria. We then grew unialgal cultures on solidified agar prepared with Wood's Hole culture medium, buffered with 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) to pH 7 and enriched with CaCO3. Benthic foraminifera are highly abundant heterotrophic protists in marine sediments, but intertidal communities are expected to undergo future changes. Environmental changes can exceed the tolerance limits of intertidal species causing a shift in species composition which might result in altered nutrient fluxes. Factors limiting the abundance of specific foraminiferal species can be temperature related stress tolerance or food source processing efficiency. In this study, we performed a laboratory feeding experiment on Ammonia tepida and Haynesina germanica, two dominant foraminiferal species of the German Wadden Sea/Friedrichskoog, to test the effect of temperature on phytodetritus ingestion. The specimens were fed with 13C and 15N labelled freeze dried Dunaliella tertiolecta (green algae) at the start of the experiment and were incubated at 20 °C, 25 °C, and 30 °C respectively. Foraminifera are an important faunal element of the benthos in oxygen-depleted settings such as Oxygen Minimum Zones (OMZs) where they can play a relevant role in the processing of phytodetritus. We investigated the uptake of phytodetritus (labeled with (13)C and (15)N) by calcareous foraminifera in the 0-1 cm sediment horizon under different oxygen concentrations within the OMZ in the eastern Arabian Sea. The in situ tracer experiments were carried out along a depth transect on the Indian margin over a period of 4 to 10 days. The uptake of phytodetrital carbon within 4 days by all investigated species shows that phytodetritus is a relevant food source for foraminifera in OMZ sediments. The decrease of total carbon uptake from 540 to 1100 m suggests a higher demand for carbon by species in the low-oxygen core region of the OMZ or less food competition with macrofauna. Especially Uvigerinids showed high uptake of phytodetrital carbon at the lowest oxygenated site. Variation in the ratio of phytodetrital carbon to nitrogen between species and sites indicates that foraminiferal carbon and nitrogen use can be decoupled and different nutritional demands are found between species. Lower ratio of phytodetrital carbon and nitrogen at 540 m could hint for greater demand or storage of food-based nitrogen, ingestion, or hosting of bacteria under almost anoxic conditions. Shifts in the foraminiferal assemblage structure (controlled by oxygen or food availability) and in the presence of other benthic organisms are likely to account for observed changes in the processing of phytodetritus in the different OMZ habitats. Foraminifera dominate the short-term processing of phytodetritus in the OMZ core but are less important in the lower OMZ boundary region of the Indian margin as biological interactions and species distribution of foraminifera change with depth and oxygen levels.
