Apera spica-venti, a prevalent weed in Czech winter wheat fields, has developed resistance to ALS-inhibiting herbicides due to their frequent use. This study reports a biotype of A. spica-venti resistant to pyroxsulam, with cross and multiple resistance to iodosulfuron, propoxycarbazone, pinoxaden, and chlortoluron. Dose–response experiments revealed high resistance of both R1 and R2 biotypes to pyroxsulam, with resistance factors (RF) of 6.69 and 141.65, respectively. Pre-treatment with malathion reduced RF by 2.40× and 1.25× in R1 and R2, indicating the potential involvement of cytochrome P450 (CytP450). NBD-Cl pre-treatment decreased RF only in R2, suggesting possible GST involvement. Gene analysis revealed no mutations (at previously reported sites) or overexpression in the acetolactate synthase (ALS) gene. However, a significant difference in ALS enzyme activity between resistant and susceptible biotypes points to target-site resistance mechanisms. Studies with 14C-labeled pyroxsulam showed that reduced absorption and translocation were not likely resistance mechanisms. In summary, herbicide resistance in A. spica-venti appears to result from multiple mechanisms. Possible causes include target-site resistance from an unidentified ALS mutation (within coding or regulatory regions). Enhanced herbicide metabolism via CytP450s and GSTs is also a contributing factor. Further experimental validation is needed to confirm these mechanisms and fully understand the resistance. This evolution underscores the adaptive capacity of weed populations under herbicide pressure, emphasizing the need for alternative control strategies.

Cuba is struggling with a growing environmental problem: the uncontrolled spread of the allochthonous weed species marabou (Dichrostachys cinerea) throughout the country. Over the last 70 years, marabou has become a formidable invasive species that poses a threat to Cuban biodiversity and agricultural productivity. In this paper, we present a free and affordable method for regularly mapping the spatial distribution of the marabou based on the Google Earth Engine platform and ecological surveys. To test its accuracy, we develop an 18-year remote sensing analysis (2000–2018) of marabou dynamics using the Valle de los Ingenios, a Cuban UNESCO World Heritage Site, as an experimental model. Our spatial analysis reveals clear patterns of marabou distribution and highlights areas of concentrated growth. Temporal trends illustrate the aggressive nature of the species, identifying periods of expansion and decline. In addition, our system is able to detect specific, large-scale human interventions against the marabou plague in the area. The results highlight the urgent need for remedial strategies to maintain the fragile ecological balance in the region.

The effective and regular remote monitoring of agricultural activity is not always possible in developing countries because the access to cloud-based geospatial analysis platforms or expensive high-resolution satellite images are not always available. Herein, using paid high-resolution satellite images first and then free medium-resolution satellite images, we aimed to develop a cost-free, affordable method for regularly mapping the spatial distribution of sicklebush (Dichrostachys cinerea), an archetypal allochthonous invasive plant in Cuba that is becoming impossible to control owing to its rapid growth in areas planted with sugar cane in the Trinidad-Valle de los Ingenios area (Cuba), a UNESCO World Heritage Site. Two types of images were used (WorldView-2 and Landsat-8); these were subjected to supervised classification, with accuracy values of 88.7% and 93.7%, respectively. Vegetation cover was first derived from the purchased WorldView-2 image, and this information was then used as the training field to obtain spectral signatures from the Landsat-8 free image so that Landsat images may be regularly used to monitor D. cinerea infestations. The results obtained in the spatial distribution map for sicklebush in the Landsat-8 images had an overall reliability of 93.7% and a Kappa coefficient reliability of 91.9%. These values indicate high confidence in the results, which suggests that sicklebush has invaded 52.7% of the total 46,807.26-ha area of the Trinidad-Valle de los Ingenios. This process proved extremely effective and demonstrated the benefits of using high-resolution spatial images from which information can be transferred to free satellite images with a larger pixel size.

Six Johnsongrass populations suspected of being glyphosate resistant were collected from railways and freeways near Cordoba (SW Spain), where glyphosate is the main weed control tool. The 50% reduction in shoot fresh weight (GR50) values obtained for these six populations ranged from 550.4 to 1169 g ae ha−1, which were 4.2 to 9 times greater than the value obtained for the susceptible population. Glyphosate was equally metabolized to the same extent in both resistant and susceptible populations, with no significant differences in either 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) inhibition or basal activity. No amino acid substitutions were observed in any of the resistant populations. Slight but significant differences in glyphosate penetration were observed among some but not all of the resistant populations and for the times of incubation assayed, although these differences were not considered further. The proposed primary mechanism of resistance in these six glyphosate-resistant Johnsongrass populations is reduced herbicide translocation, because the amount of glyphosate that translocated from treated leaves to shoots and roots in the susceptible population was double that observed in the resistant populations. As glyphosate multiple resistance due to more than one mechanism is not uncommon, this is the first time that glyphosate-resistant Johnsongrass populations have been fully described for all known mechanisms.

Resistance to acetolactate synthase (ALS) inhibiting herbicides has recently been reported in Glebionis coronaria from wheat fields in northern Tunisia, where the weed is widespread. However, potential resistance mechanisms conferring resistance in these populations are unknown. The aim of this research was to study target-site resistance (TSR) and non-target-site resistance (NTSR) mechanisms present in two putative resistant (R) populations. Dose–response experiments, ALS enzyme activity assays, ALS gene sequencing, absorption and translocation experiments with radiolabeled herbicides, and metabolism experiments were carried out for this purpose. Whole plant trials confirmed high resistance levels to tribenuron and cross-resistance to florasulam and imazamox. ALS enzyme activity further confirmed cross-resistance to these three herbicides and also to bispyribac, but not to flucarbazone. Sequence analysis revealed the presence of amino acid substitutions in positions 197, 376, and 574 of the target enzyme. Among the NTSR mechanisms investigated, absorption or translocation did not contribute to resistance, while evidences of the presence of enhanced metabolism were provided. A pretreatment with the cytochrome P450 monooxygenase (P450) inhibitor malathion partially synergized with imazamox in post-emergence but not with tribenuron in dose–response experiments. Additionally, an imazamox hydroxyl metabolite was detected in both R populations in metabolism experiments, which disappeared with the pretreatment with malathion. This study confirms the evolution of cross-resistance to ALS inhibiting herbicides in G. coronaria from Tunisia through TSR and NTSR mechanisms. The presence of enhanced metabolism involving P450 is threatening the chemical management of this weed in Tunisian wheat fields, since it might confer cross-resistance to other sites of action.
